Verfahren zur Herstellung von 3ss,11a,21-Triacetoxy-pregnanon-(20). Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von 3ss,11u, 21-Triacetoxy -pregnan-on-(20), einem wert vollen Zwisehenprodukt zur Synthese von Hormonen der 1Vebennierenrinde, ausgehend von Sarmentogenin.
Das Ausgangsmaterial kann beispielsweise aus Sarmentocymarin gewonnen werden. Das erfindungsgemässe Verfahren ist nachstehend durch die Formeln I bis VI schematisch dar- L-estellt. . Das Sarmentogenin (Formel I) wird zu nächst in das 3,11-Diacetat der Formel II übergeführt und dann die C,C-Doppelbin- dung im Ring E durch oxydierende Mittel geöffnet, z.

Ozonid wird vorzugsweise unter Verwendung eines Reduktionsmittels zersetzt und liefert ein Glyoxylsäure-derivat, welches durch Versei- fang in das freie Ketol der Formel III über geführt wird.
Das erhaltene c'ss,11a,14,21- Tetroxy-14-isopregnanon - (20 )- diaeetat - (3,11) wird dann in das Acetat der Formel IV über geführt und die Hydroxylgruppe am Kohlen- stoffatom 14 unter Bildung einer C,C-Doppel- bindung beseitigt, z. B. durch Einwirkung von POC13 in einer Pyridinlösung als wasserab spaltendes :Mittel, wodurch die Verbindung der Formel V erhalten wird.
Die erwähnte Acetylierung der Hydroxyl- gruppe am Kohlenstoffatom 21 und die Ab spaltung der Hydroxylgruppe am Kohlenstoff atom 14 kann auch in umgekehrter Reihen folge stattfinden.
Die Verbindung der Formel V wird einer Hydrierung unter Sättigung der C,C-Doppel- bindung unterworfen, beispielsweise unter Verwendung eines Platinkatalysators, und das Reaktionsprodukt sodann oxydiert, beispiels weise durch Behandlung mit Chromsäure in essigsaurer Lösung, wodurch das gewünschte Produkt gemäss Formel VI erhalten wird.

in beliebiger Reihenfolge die Hydroxylgruppe ain Kohlen stoffatom 21 acetyliert und die Hydroxyl- gruppe am Kohlenstoffatom 14 unter Bildung einer C,C-Doppelbindung durch Einwirkung eines wasserabspaltenden Mittels beseitigt.

2,07 g Sarmentocymarin vom Schmp. 212 bis 214 C -irden in 50 eni3 Methanol auf gelöst, die Lösung mit. 50 eni3 0,1 n-Schwefel- säLire versetzt und 1/z Stunde auf dem Wasser bad erhitzt. Das Methanol wurde im Vakuum entfernt und das ausgefallene Sarmentogenin abgenutseht, mit Wasser gewaschen und im Vakuum getrocknet. Ausbeute l,63 g kristallines Produkt vom Sehmp. 250 bis 270 C.
Das Fil trat wurde mit Chloroform und Essigester aus geschüttelt und lieferte eine zusätzliche Menge von 4.0m-- rohes Sarmentogenin.

IIerstellit7i.g <I>des</I> Acetats der Formel II. 1,3 g Sarmentogenin vom Schmelzpunkt 250 bis 270 C wurden in 6 em3 Pvridin auf aelöst und mit. 4 cm3 Aeetaiiliydrid versetzt. Die lIischung blieb über Nacht bei 18 C und eitere 2 Stunden bei 60 C stehen. Die übliche Aufarbeitung lieferte 1,64 g Rohprodukt.
Durch Umkristallisieren ans Aceton-Äther wurde ein kristallines Produkt vom Schmelz punkt 145 bis 165 C erhalten.
Hei,stellitiig von 3ss,11a,11,?I-Tetr,oxy-1l-iso- pregyican-on-(ND)-dia,cetat-(3,11) <I>der Formel</I> III. 1,540 g rohes Acetat der Formel II wurden in 50 em3 neutralem Essigester aufgelöst und bei einer Temperatur von -80 C während einer halben Stunde mit trockenem ozonhalti gem Sauerstoff (etwa<B>100</B> eill@@hn. mit un gefähr 4'/'0 Ozongehalt) behandelt.
Das Ge misch blieb weitere 20 Min. bei -80 C stehen, wurde dann im Vakuum eingedampft und der Rückstand in 6 cm3 Eisessig aufgelöst und unter Zusatz von Zinkstaub in kleinen Por tionen etwa. eine Stunde bei 20 bis 30 C ge schüttelt. Dieser wurde in 1.00 em3 Methanol aufgelöst, mit 1,2 g Kaliumbikarbonat in 35 cm3 Wasser versetzt und die Mischung 16 Stunden stehengelassen. Herstellung <I>des</I> Acetates <I>der</I> Formel <I>IV.</I> 228 mg des erhaltenen RohprodrLktes wur den in 1,5 em3 Pyridin und 1 em3 Aeetanhy- drid 4 Stunden bei 18 C und weitere 3 Stun den bei 60 C stehengelassen. Die übliche Aufarbeitung lieferte 252 mg Rohprodukt.
