Verfahren zur Herstellung von 11a -Oxypregnan-3,20-dion. Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von lla-Oxy- ljregnan-3,20-dion, einer neuen Verbindung, welche als Zwischenprodukt bei der präpara- tiven Darstellung biologisch aktiver Verbin= dungen wie Cortison Verwendung finden kann, aber auch an sich physiologisch wirk sam ist.
lla-Oxypregnan-3,20-dion kann leicht zu dem bekannten Pregnan-3,11,20-trion dadurch oxydiert werden, dass man es mit Chromsäure in Essigsäure zur Reaktion bringt. Es kann auf bekannten Wegen in Cortison über geführt werden, zum Beispiel durch Einfüh rung von Oxygruppen in die Siebzehn- und l,'inundzwanzigstellung und Einführung einer Doppelbindung am C-Atom 4. Die besondere Verwendungsmöglichkeit der neuen Verbin dung ist durch die a-Konfiguration der Oxy- gruppe am C-Atom 11 gegeben.
Im Gegensatz zu den Verbindungen, bei welchen sich diese Oxygruppe in ss-Konfiguration befindet, kann die erfindungsgemäss zu erhaltende Verbin- dung leicht verestert werden. Solche Ester können zur Maskierung der Oxygrzppe bei naehfolgenden chemischen Reaktionen dienen. Sie können ferner bei der Herstellung von biologisch wirksamen Verbindungen mit Estergruppen am 11 C-Atom von Nutzen sein.
Das Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dass lla-Oxyproge- steron (zum Beispiel hergestellt aus Proge- si eron durch Fermentation, wie im Beispiel besehrieben) in Gegenwart eines Wasserstoff- überträgers mit Wasserstoff hydrie,@eTd,, Als Wasserstoffüberträger kann Palladium oder Palladiumkohle verwendet werden.
Bei spielsweise kann die Verwendung von 30 o/oiger Palladiumaktivkohle vorteilhaft sein. Die Ausgangsverbindung kann üblicher weise in einem organischen Lösungsmittel ge löst werden, wie z. B. in Methanol, Äthanol, Azeton, Benzol, Chloroform und ähulichen,wo- bei Äthanol bevorzugt wird. Der Katalysator wird sodann zugesetzt. Die Lösung kann, wenn gewünscht,<I>auch</I> durch Zusatz einiger Tropfen eines Amins, zum Beispiel des Tri- äthanolamins, basisch gemacht werden. Zur Hydrierung wird zweckmässig mit Wasser stoff so lange geschüttelt, bis die theoretische Menge an Wasserstoff absorbiert ist.
Im all gemeinen kann die Reaktion bei Raumtempe ratur und etwa 0,4 kg/cm2 (10 Pfund) Wasserstoffdruck zu Ende geführt werden. Man kann aber auch bei andern Tempera turen, beispielsweise zwischen etwa 0 und etwa 60 C arbeiten. Man kann natürlich auch bei höheren oder niedrigeren Wasser stoffdrucken arbeiten.- Das Reduktionspro dukt kann auf irgendeinem konventionellen Wege, zum Beispiel durch Abdampfen des Lösungsmittels lind Kristallisieren des Rück standes, auf chromatographischem Wege oder anderswie isoliert werden.
<I>Beispiel:</I> a) 4 Liter einer 32-48 Stunden ent wickelten Kultur von Rhizopus arrhizus wur- den mit 1 g Progesteron versetzt, welches, um eine Suspendierung des Steroids in der wässrigen Kultur zu gewährleisten, in 50 cin3 Azeton gelöst war. Die Kultur wurde sodann bei Raumtemperatur 48 Stunden bebrütet. Darnach hatte sich ein p],-Wert von 3,5 ein gestellt. Fermentationsflüssigkeit und lUycel wurden nun nacheinander je dreimal mit 1 Liter, einmal mit 2 Liter und einmal mit 1 Liter Methylenchlorid extrahiert.
Die Me- thylenchloridauszüge wurden vereint, zweimal mit je 400 em3 2%iger wässriger Natrium- bicarbonatlösung und dreimal mit je 500 cm 3 Wasser gewaschen.
Der Methylenchloridaus- zug wurde im Vakuum zur Trockene abge- dunstet und der Rückstand in 50 cm3 Methy- lenchlorid aufgenommen. Diese Lösung wurde in einem 100 cms fassenden Becherglas durch einen Luftstrom abgedunstet. Der 1,585 g wiegende Rückstand wurde in 5 ems heissem Methanol gelöst und langsam bis auf Raum temperatur abkühlen gelassen, wobei sich 75 mg Kristalle ausschieden.
Der Rückstand aus der Mutterlauge, durch Verdunsten an der Luft vom Lösungsmittel befreit, wurde in 50 cm3 Benzol gelöst und über A1003 ehro- matographiert. Zu diesem Zweck wurden 50 ä mit. Säure gewaschenes A1203, das bei 45 C getrocknet war, als Adsorbens verwendet. Die Säule wurde mit 100 em3-Portionen der Lösungsmittel entwickelt.
Die Lösungsmittel und ihre Reihenfolge waren dabei: Benzol, Benzol Benzol + 5% Äther, Benzol + 5% Äther, Benzol + 10% Äther, Benzol + 50% Äther,

Azeton + 50% Methanol. Die Chloroform und Chloroform + 5% Azeton-Fraktionen wurden vereinigt und zur Trockene ab- gedunstet. Der Rückstand wurde in 2 cm-' heissem Methanol gelöst und filtriert.
Nach Abkühlen über Nacht wurden 171 mg kristallisiertes 11a-Oxyprogesteron mit einem Sehmp. 166-167 C erhalten. Eine rekristal- lisierte Probe zeigte die folgenden Konstan ten: Schmp. 166-167 C;

b) 250 mg (0,75 Millimol) 11a-Oxypro- gesteron, gelöst in 100 cm3 Äthanol und mit 6 Tropfen Triäthanolamin versetzt, wurden bei Raumtemperatur und unter etwa 5 kg/cm2 Wasserstoffdruck in Gegenwart von 45mg 300%iger Palladium-Kohle als Katalysator in einem HydriQrapparat-(Parr apparatus) mit Quecksilbermanometer hydriert.
Die Zeit, welche zur Hydrierung nöti- war, betrar 20 Minuten. Nach Filtrieren und Abdampfen wurden 265 nig Rückstand mit. einem Schmelzintervall hon 145 bis 185 C erhalten. Dieses Produkt wurde mit. einem Gemiseli von 1 em3 Äther und 9 em3 Petroläther der Siede grenzen 60-70 ( Skelly Solve B ), extra hiert.
Aus diesem Extrakt. schieden sich beim Stehen 8 mg (32%) 1.la-@xypregnan-3,20- dion als gefiedeiee Nadeln mit einem Selimp. von etwa 85-90 C aus. Der Sehmp. zeigte ausserordentlich schwankende Werte. Sie lagen zwischen etwa.<B>55</B> und 130 C. Dennoch :@r-aben die verschiedenen Chargen bei der Weiterverarbeitung in verschiedenen Arbeits gängen die gleichen Resultate.
Analyse: Berechnet. für: C21113003 : C = 75,86 %; H = 9,70 %. Gefunden: C = 76,13 %;
H = 9,63 %. Die Separierung wurde auch durch Chromatographieren ergänzt. Aus einer in nigen Mischung von 16,7 g Diatomeenerde ( C'elite 545 ) mit Aktiv-Kohle ( Darco (;-60 ) wurde mit etwa 150 cm3 Methanol eine 30 cm dicke Säule feucht gepackt.
Es wa r etwa 1 kg; cmz Druckluft nötig, um das 1.@isun,:,mittel zum vollständigen Eindringen in das Adsorbens zu bringen. Es wurde ein Wat.tebauseh am Kopf der Säule angebracht und diese nacheinander einmal mit. 200 em3 Methanol und zweimal mit 200 ein-' Azeton behandelt..
Nun wurde der Hydrierungsrüek- stand in Methanol (siehe oben) einer Hydrie rung von 500 mg lla-Oxyprogesteron in 200 em3 Azeton gelöst aufgegeben. Nach dem Passieren der Säule wurde diese Fraktion als die erste bezeichnet. Anschliessend wurden achtmal 200 em3 Azeton und viermal 200 em3 Methylenehlorid aufgegeben.
Die Zugabe er folgte jeweils, wenn der Rest der vorher- gehenden Charge den -\)passierte. Aus den vereinten Fraktionen 2 bis 6 ein- schliesslich erhielt. man nach dem Abdun:st.en des Lösungsmittels 11a-Oxypregilan-2,20-dion.
Process for the preparation of 11a -oxypregnane-3,20-dione.
