Verfahren zur Herstellung eines Oxoacylamins der Cyclopentanopolyhydrophenanthrenreihe. Die vorliegende Erfindung betrifft neue Verbindungen der Steroidreihe und das Ver fahren zur Herstellung derselben.
Bei den neuen Verbindungen handelt es sieh um solche Verbindungen der Cyclopen- tanopolyliydrophenanthrenreihe, die in 3-Stel- lung eine Oxogruppe und in 17-Stellung eine Aeylaminogruppe tragen.
Diese neuen Verbindungen können gewon nen werden, indem man 3-Oxy-17-acylamino- verbindungen. der CyclopentanopolyhyArophen- anthrenreihe in an sich, bekannter Weise mit Oxydationsmitteln behandelt.
Als Ausgangsmaterial kommen beispiels weise folgende Verbindungen in Frage: 3- Oxy-17-aeylamino-45-androsten, 3-Oxy-17-acyl- amino-androstan, 3-Oxy-17-acylaminoätioeho- lan. Als Aeylreste kommen vornehmlich nie dere aliphatisehe Acylreste wie beispielsweise Aeetyl, Propionyl, Butyryl, Isobutyryl und Valeryl in Betracht.
Als Oxydationsmittel kommen beispiels weise in Frage: Bromaeetamid, Chromsäure, ferner Mischungen aus Metallalkoholaten, z. B. Aluininiutnisopropylat, und einem Keton in indifferenten Lösungsmitteln.
Die 0xy dation mit. Chromsäure wird zweckiriässigerweise so ausgeführt, dass man die 3-Oxyverbindung in Eisessig mit. Chrom säure eini;e Zeit. bei Temperaturen unterhalb Zimmertemperatur stehenlässt. Bei ungesät tigten Verbindungen ist. es zweckmässig, die Doppelbindungen, z. B. durch Anlagerung von Brom, intermediär zu schützen.
Die Verwendung von Mischungen aus Me- tallalkoholaten und einem Keton in indiffe renten Lösungsmitteln als Oxydationsmittel geschieht in üblicher Weise so, dass man das Reakt.ionsgemiseh einige Stunden am Rüek- fluss kocht.
Bromacetamid kann als Oxydationsmittel. vor allem bei gesättigten Verbindungen ver wendet werden. Es wird' zweckmässig in Lö- sungsmitteln wie beispielsweise Butanol oder Pyridin gebraucht. Auch N-Bromsuecinimid kommt als Oxydationsmittel in Betracht.
Die als Ausgangsmaterial dienenden Oxyver- bindungen können nach verschiedenen Verfah ren gewonnenwerden, z. B. kann man 3-Oxy-20- ketoxime der Cyclopentanopolyhydrophenan- threnreihe der Beckmannsehen Umlagerung unterwerfen gemäss dem Verfahren der Pa tentschrift Nr. 303659. Man erhält so beispiels weise aus 45-Pregnen-3-ss-ol-20-on-oxim das 3- ss-Oxy-17-aeetylamino-45-androst.en.
Weiterhin sind die 3-Oxy-17-ac@>laminover- bindungen der Steroidreihe erhältlich durch partielle Aeylierung der entsprechenden 3- Oxy-17-aminoverbindungen.
Da die 17-Aeylaminoverbindungen der Cyelopent.anopolyhydrophenanthrenreihe sich nur schwer zu freien 17-Aminoverbindüngen verseifen lassen, wie aus der Schweizer Pa tentschrift Nr. 303659 bekannt ist, kann man leicht 3-Oxy-17-acylaminoverbindungen der Ste- roidreihe durch partielle Verseifung der 3- Aevloxy-17-aevlaminoverbindun,-en gewinnen. Die partielle Verseifung kann in üblicher Weise mit Säure oder Alkali erfolgen. llan erhält so z.

Die Oxoacylamine sollen als Therapeutiea und als Zwischenprodukte für weitere Uni- setzungen Verwendung finden.
Gegenstand des vorliegenden Patentes ist. ein Verfahren zur Herstellung von 3-Oxo-17- aeetvla.nrino-androstan, welches dadurch ge kennzeichnet ist, dass man 3-Oxi--17-aeetyl- ainino-androstan mit Oxydationsmitteln, ins besondere mit Chromsiiureanhydrid in Gegen wart von Eisessig, behandelt.
Die so erhaltene Verbindung kristallisiert in Forin von Blättchen, die einen Sehnielz- punkt von 927 aufweisen. Sie ist in Wasser unlöslich, in Aceton und Äther schwer lös lich, dagegen in Methanol und Eisessig lös lich. Das Produkt kann als Therapeutikum oder als Zwischenprodukt zur Herstellung wertvoller Arzneimittel verwendet. werden.
<I>Beispiel:</I> <B>500</B> mg 3-Oxy-17-aeetvlaniino-androstari werden in 10 ema Eisessig gelöst, die Lösung wird auf 0 abgekühlt und unter Eiskühhuig eine Lösung von 150 mg Chromsäureanhydrid in 7,5 em3 Eisessig zugesetzt. Nach 16stündi- gem Stehen bei Zimmertemperatur wird die Reaktionslösung in Wasser gegossen und der ausgefallene Niederschlag abfiltriert, mit Wasser gewaschen und getrocknet.
Man er hält 335 mg Rohprodukt vom Schmelzpunkt 21.0-220 . Nach mehrmaligem Umkristallisie- ren wird das 3-Oxo-17-aeetylamino-androstan rein in Blättchen vom Schmelzpunkt 227 er halten.
Das als Ausgangsstoff verwendete 3-Oxy- 17-acet.ylainino-androstan wurde in einer der nachstehend beschriebenen Weisen hergestellt: a.) 1.,8 g 3-Acetoxy -17-acetylamino-45- androsten erden in 30 em3 Eisessig gelöst und mit 100 mg vorhydriertem Platinoxydkata- lyBator in einer Wasserstoffatmosphäre bis zur Aufnahme von etwas mehr als der berechneten Menge Wasserstoff geschüttelt.
Nach Abfil- trieren des Platins und Einengen der Lösung wird das 3-Acetoxy-17-acetylamino-androstan vom Schmelzpunkt 180 erhalten, das nach Umkristallisieren aus Alkohol bei 106 schmilzt und identisch ist. mit dem im Patent Nr. 303650 beschriebenen Produkt, das durch Beckmann- sche Umlagerung desallo-Pregnan-3ss-ol-20-on- < lcetat-oxims erhalten wird.
500 mg des erhaltenen 3-Aeetoxy-17-acetyl- i amino -androstans werden in einer Mischung von 20 em3 Alkohol und 3 cm3 40%iger Na tronlauge 25 Minuten gekocht.
Die Reaktions- -SI[>l #@IUi luasso & a2 aassnm ui paim .uullaslul essig angesäuert und das ausgefallene Reak tionsprodukt abfiltriert und mit Wasser ge waschen. -Man erhält 360 mg Rohprodukt vom Schmelzpunkt 245-255 . Nach Umkristalli- sieren aus Alkohol wird reines 3-Oxy-17-acetyl- amino-androstan vorn Schmelzpunkt 262 er halten.
h) 200 mg essigsaures Salz des 3-Oxy-17- ainino-45-androstens werden in einer Mischung von \? em3 Pyridin und 0,1 cm3 Essigsäure anhydrid h/2 Stunde bei Zimmertemperatur aufbewahrt. Die Reaktionslösung wird in Was ser @,@e@@ossen, das Reaktionsprodukt abfiltriert und mit Wasser gewaschen. Nach dem Trock nen auf dem Wasserbad erhält man 120 mg Rohprodukt vom Schmelzpunkt 242 .
Nach mehrfachem Umkristallisieren aus verdünntem Methanol erhält man reines 3-Oxy-17-acetyl- amino-d5-androsten, das identisch ist mit dem durch Beckmannsche Umlagerung des Pre- gnenolonoxims hergestellten Produkt, das in der obengenannten Patentschrift beschrieben ist.
1 g 3 - Oxy -17 - acetylamino -d 5 - androsten wird in 30 cm3 Eisessig gelöst und in einer Wasserstoffatmosphäre mit 100 mg vorhydrier- tem Platinoxyd geschüttelt, bis die nötige Menge Wasserstoff aufgenommen ist. Der Katalysator wird abfiltriert und die Eisessig- lösung auf etwa 15 ems eingeengt. Beile Ab kühlen kristallisiert das Hydrierungsprodukt in einer Ausbeute von 720 mg vom Schmelz punkt 257 aus.
Aus der Mutterlauge werden nach Zusatz von Wasser noch 180 mg gewon nen. Nach Umkristallisieren aus Alkohol er hält man 3-Oxy-17-acetylamino-androstan in schönen, zu Drusen vereinigten Prismen vom Schmelzpunkt 262 .

These new compounds can be obtained by adding 3-oxy-17-acylamino compounds. of the CyclopentanopolyhyArophenanthene series treated in a manner known per se with oxidizing agents.
The following compounds, for example, can be used as starting material: 3-oxy-17-aylamino-45-androstene, 3-oxy-17-acylamino-androstane, 3-oxy-17-acylaminoethanol. Particularly suitable alkyl radicals are aliphatic acyl radicals such as, for example, acetyl, propionyl, butyryl, isobutyryl and valeryl.
As an oxidizing agent, for example, are possible: Bromaeetamid, chromic acid, also mixtures of metal alcoholates, eg. B. Aluininiutnisopropylat, and a ketone in inert solvents.
