Verfahren zur Herstellung von Bisnoreholadien.(4,6)-on-(3)-al-(22). Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung eines neuen Stoffes, nämlich von Bisnorcholadien-(4,6)-on- (3)-al-(22).

Sie eignet sich als Zwischenprodukt für die Herstellung von Steroidhormonen; das Ver fahren ermöglicht, ausgehend von Isoergo- steron, zu Progesteron, den Androgenen und Oestrogenen zu gelangen.

unter Verwendung von 1,0 bis 1,25 Mol Ozon pro Mol Isoergosteron bei einer Temperatur zwischen -80 und + 30 C ozonisiert, das so erhaltene Ozonid unter reduzierenden Bedin gungen zersetzt und das Bisnor-choladien- (4,6)-on-(3)-al-(22) aus dem Reaktionspro- dukt isoliert wird.
Der Erfolg des erfindungs gemässen Verfahrens ist überraschend, da es bisher als unmöglich erachtet wurde, einen derart stark ungesättigten Stoff selektiv zu ozonisieren.
Man löst z. B. das Isoergosteron in einem Lösungsmittel, kühlt die Lösung auf eine zwi schen -80 und ;- 30 C liegende Temperatur und leitet Ozon oder ozonisierte Luft oder ozonisierten Sauerstoff in die Lösung ein, bis 1,0 bis 1,25 Mol, vorzugsweise 1,0 bis 1,1 Mol, Ozon pro Mol Isoergosteron absorbiert worden sind. Die Addition des Ozons an die 22:23-Doppelbindung erfolgt derart rasch, dass nur eine kleine Menge Ozon aus dem Re aktionsgemisch entweicht.
Die gewöhnlich er forderliche Ozonmenge entspricht deshalb ziemlich genau der theoretischen Menge. Falls das Lösungsmittel selbst auch mit dem Ozon reagiert, so müssen die dadurch bedingten Ver luste bei der Berechnung der einzuführenden Ozonmenge berücksichtigt werden. Die Tem peratur der Lösung wird während der Ein führung des Ozons vorzugsweise zwischen -30 und -70 C gehalten.
Die niederen Tem peraturen des angegebenen Bereiches (gegen --80(1 C ) können leicht erhalten werden, in- dem die Lösung des Isoergosterons mit einem Bad von festem Kohlendioxyd in Aceton oder dergleichen gekühlt wird. Viele der bei Ozonisierungen gewöhnlich verwendeten Lö- si-ungsmittel, wie z. B. Chloroform, Essigsäure, Tetrachlorkohlenstoff, Äthylenchlorid, Me- thylenchlorid usw., können verwendet werden.
Das Ozonid wird hierauf unter reduzie renden Bedingungen reduziert, das heisst in Abwesenheit von' oxydierenden Substanzen. Man muss also verhindern, dass der durch Zer setzung des Ozonids gebildete überschüssige Sauerstoff durch Reaktion mit etwa vorhan dener Feuchtigkeit Wasserstoffperoxyd bildet und den gebildeten Aldehyd oxydiert. Man kann dies zweckmässigerweise dadurch errei chen,
dass man das Ozonid in Eisessig durch Zugabe von feingepulvertem Zink oder mit tels einer katalytischen Menge eines kolloi dalen Metalles, wie z. B. Silber, Platin oder Palladium, in einem Lösungsmittel, wie z. B. Eisessig, Alkohol oder Äthylessigester, zer setzt.
Die Zersetzung von Ozoniden unter reduzierenden Bedingungen ist an sich gut bekannt [Hill und Kelly, Organic Chemistry , Seite 63, Blackiston Company, Philadelphia (1943); Church et a1, J.
Am. Chem. Soe. 56, 176-184 (1934) ; Gilman Organic Chemistry , Zweite Auflage, Seite 63, John Wiley and Sons, New York (1943); Long, Chem. Reviews 27, 452---454 (1940)1.
Es ist bei Ozonisierimgen, die in andern Lösungsmitteln als Eisessig durchgeführt wer den, üblich, das für die Ozonisierung ver wendete Lösungsmittel nach beendeter Ozoni- sierimg durch Zugabe von Eisessig und Ent fernung des niedrigsiedenden Lösungsmittels durch fraktionierte Destillation zu ersetzen. Dies ist auch im vorliegenden Fall zweck mässig.
Die Entfernung des Lösungsmittels kann auch so erfolgen, dass man das Ozonisie- rungsgemisch vor. der Zugabe des Eisessigs unter vermindertem Druck vorsichtig erwärmt.
Nach erfolgter Zersetzung des Ozonids und Entfernung z. B. des Zinks oder des Kataly- sators kann der Aldehyd durch Verdünnen der Essigsäure mit Wasser oder in anderer üblicher Weise, beispielsweise durch Bildung eines Aldehydderivates, z. B. des Dinitro- phenylhydrazons, isoliert werden.
<I>Beispiel:</I> Eine Lösung von 0,79 g (2,0 Millimol) Iso- ergosteron in 60 cm3 Methylenchlorid wurde in einem Bad aus Trockeneis und Aceton auf einer Temperatur von etwa -70 C gehalten, während ozonisierter Sauerstoff mit einem Gehalt von 1,06 Äquivalenten Ozon in die Lö sung eingeleitet wurde.
Das Reaktionsgemisch wurde hierauf auf Raumtemperatur erwärmt und mit 15 cm3 Eisessig versetzt, worauf die Lösung im Vakuum auf etwa 10 cm3 ein geengt wurde, um das Methylenchlorid zu ent fernen. Nach Zugabe von Essigsäure zwecks Erhöhung des Volumens auf 20 em3 wurden im Verlaufe von 20 Minuten bei einer Tem peratur von 25 bis 31 C 2,3 g Zinkstaub por- tionenweise zugesetzt. Das Zink wurde durch Filtrieren entfernt, worauf die essigsaure Lö sung in Wasser gegossen wurde.
Es wurde 0,59 g eines festen Produktes erhalten, das in einem Gemisch von Äther und Alkohol gelöst und dann mit 36 % igem wässrigem Natrium- bisulfit extrahiert wurde. Die Bisulfitlösung wurde mit Natriumcarbonat zersetzt und mit Äther extrahiert.
Beim Abdampfen des Äthers blieben etwa 200 mg einer gummiartigen Masse zurück, die sich bei der Analyse als Bisnor- choladien- (4,6) -on- (3) -a1- (22), C22H3002, Molekulargewicht 326,46, erwies. Aus dem In frarotspektrum war ersichtlich, dass eine Al dehyd- und eine konjugierte Ketogruppe vor handen waren.
Die Ultraviolettanalyse ergab ein Absorptionsmaximum bei 285 m,ca und einen Extinktionskoeffizienten von 19 900, woraus geschlossen werden konnte, dass die 3-Keto- gruppe und die Doppelbindungen in 4- und 6-Stellung immer noch vorhanden waren (für Isöergosteron betragen das Absorptionsmaxi mum 280 m@c und der Extinktionskoeffizient 33100 (Heilbron, J. Chem. Soc. 1938, 869).
Das Bisnorcholadien-(4,6)-on-(3)-al-(22) wies nach Umkristallisation aus verdünnter Essig säure einen Schmelzpiuikt von etwa 120 C auf.
Das aus dem rohen Aldehyd und 2,4-Di- nitro-phenylhydrazin hergestellte Dinitro- phenylhydrazon wies nach Ohromatographie und Umkristallisation einen Schmelzpunkt von 215 bis 22311 C auf..
Das Bisnorcholadien-(4,6)-on-(3)-al-(22) eignet sich unter anderem als Ausgangs material für die Herstellung von Bisnorcholen- 3-on-22-al der Formel

ozonized using 1.0 to 1.25 moles of ozone per mole of isoergosterone at a temperature between -80 and + 30 C, the ozonide thus obtained decomposes under reducing conditions and the bisnorcholadiene (4,6) -on- (3) -al- (22) is isolated from the reaction product.
The success of the method according to the invention is surprising, since it has hitherto been considered impossible to selectively ozonize such a highly unsaturated substance.
One solves z. B. isoergosterone in a solvent, cools the solution to a temperature between -80 and -30 C and introduces ozone or ozonated air or ozonated oxygen into the solution, up to 1.0 to 1.25 mol, preferably 1 .0 to 1.1 moles of ozone per mole of isoergosterone have been absorbed. The addition of the ozone to the 22:23 double bond takes place so rapidly that only a small amount of ozone escapes from the reaction mixture.
The amount of ozone usually required therefore corresponds almost exactly to the theoretical amount. It is common for ozonization in solvents other than glacial acetic acid to replace the solvent used for ozonization by adding glacial acetic acid and removing the low-boiling solvent by fractional distillation after ozonization is complete. This is also useful in the present case.
The solvent can also be removed in such a way that the ozonization mixture is first. the addition of glacial acetic acid was carefully heated under reduced pressure.

The reaction mixture was then warmed to room temperature and admixed with 15 cm3 of glacial acetic acid, whereupon the solution was concentrated in vacuo to about 10 cm3 in order to remove the methylene chloride. After adding acetic acid to increase the volume to 20 cubic meters, 2.3 g of zinc dust were added in portions over the course of 20 minutes at a temperature of 25 to 31 C. The zinc was removed by filtration and the acetic acid solution was poured into water.
0.59 g of a solid product was obtained which was dissolved in a mixture of ether and alcohol and then extracted with 36% strength aqueous sodium bisulfite. The bisulfite solution was decomposed with sodium carbonate and extracted with ether.
When the ether evaporated, about 200 mg of a gummy mass remained, which was found to be bisnorcholadien- (4,6) -one- (3) -a1- (22), C22H3002, molecular weight 326.46, upon analysis. The infrared spectrum showed that an aldehyde and a conjugated keto group were present.
The ultraviolet analysis showed an absorption maximum at 285 m, approx and an extinction coefficient of 19,900, from which it could be concluded that the 3-keto group and the double bonds in the 4- and 6-positions were still present (for isoergosterone the absorption maximum is 280 m @ c and the extinction coefficient 33100 (Heilbron, J. Chem. Soc. 1938, 869).
The bisnorcholadien- (4,6) -one- (3) -al- (22) had a melting point of about 120 ° C. after recrystallization from dilute acetic acid.
The dinitrophenylhydrazone produced from the crude aldehyde and 2,4-di-nitro-phenylhydrazine had a melting point of 215 to 22 311 C after chromatography and recrystallization.
