be- zeiehnet.

Chem. Soe.

Process for the preparation of an addition product of a 3-acyloxy-5,7,9-pregnatrien-20-one. The invention relates to a method for the preparation of a new group of connec tions for the preparation of steroids with a.
Oxygen atom on the carbon atom 11. can be used and can be represented by the following structural formula:
EMI0001.0012
in which B is a hydroxyl group or an acyloxy group and A denotes an addition bridge which is derived from maleic acid, maleic acid diester or maleic acid diester. 1 to S carbon atoms in the ester group.
These new compounds are of particular interest in the field of steroid research. As is well known, the steroids containing an oxygen atom at carbon 11 (adrenal cortex hormones and derivatives) show significantly different biological effects than those in 11 = Stelluno @ unsubstituted.
Particularly interesting compounds of this kind are those in which B is an acylated oxy group, the acyl radical of which is derived from a carboxylic acid with up to 10 carbon atoms, such as formic acid, acetic acid, propionic acid, butyric acid, valeric acid, caproic acid, oenanthic acid, Caprylsäiu @ e,
Succinic acid, glutaric acid, cyclopentanoic acid, benzoic acid, toluene carboxylic acid, and also from. Naphthoic acid etc. These acids can also contain substituents such as halogens, alkyl and methoxy groups.
The bridge A of these compounds can for example correspond to the following formula
EMI0001.0075
in which R is hydrogen or an alkyl radical, for example the methyl, ethyl, propyl, isopropyl, butyl, isobutyl, lauryl, oetyl, cyclohexyl or benzyl radical, etc.,
denoted. The remainder. R. can also contain substituents such as halogens, methoxy or hydroxyl groups.
The new compounds are generally colorless crystalline solid substances which are insoluble in water and soluble in ether, halogenated hydrocarbons, acetone, ethyl acetate and benzene.
The acid anhydride addition products can easily be converted into the free 3-oxy acids by hydrolysis. These acid addition products can for their part be esterified with. conventional reagents, such as diazoalkanes, can easily be converted into the diester addition products (Wilds et al., J.
Org. Chem. 13, 76'3 [19481). Conversely, the diesters can be hydrolyzed to the two basic acids, which in turn can be converted into the corresponding anhydrides either by means of heat alone or preferably by means of a mild dehydrating agent, such as, for example, acetic anhydride or the like.
The new addition products are obtained by selective oxidation of enol esters of the addition products of 3-acyloxy = bisnor-5,7,9-eholatrien-22-al of the formula
EMI0002.0031
in which A and B has the meaning defined above, in which case the free hydroxyl group for B is excluded.
For this purpose, ozone is added to the 20:22 double bond of the enol ester. The 20.22 ozonide obtained in this way is then decomposed in the usual way for ozonides, for example by means of hydrogen peroxide, by hydrolysis, by means of zinc in glacial acetic acid or by means of a catalytic amount of a colloidal metal such as silver, platinum or palladium in one Solvents, such as glacial vinegar,
Alcohol or ethyl acetate. Suitable methods are described, for example, by Hill and Kelly in Organic Chemistry, page 63, Blaekiston Company, Philadelphia (19.13), Clnircli and coworkers in J. 9m. Chem. Soe.
56, 176-18.1 (193-1), Gilman in Organie Chemistry, 2nd Edition, page 63, John Wiley and Sons, New York (1943), and Long in Cliem. Reviews 27, 452-J54 (1940). Naeli the decomposition of the 20th:

The status of the present patent is. now a process for the preparation of the hla- leinsä.urea.nhydrid addition product of 3ss-acetoxy-5,7,9-pi # e, # - sodium-20-oii d by formula

Glacial acetic acid was then added, whereupon the meliyiene chloride was removed under reduced pressure. A further 30 cubic meters of glacial acetic acid were added, after which the ozonide was decomposed by adding 7 g of powdered zinc. practically uniform speed. were added while the reaction temperature:

When crystallization began, the rate of precipitation was increased by adding 2 volumes. Water increased. In this way 4.0 g of the maleic anhydride addition product of 3ss-leetoxy-5,7,9-pregnat.rien-20-one, melting at 240 to 2.64.5 ° C., were obtained. When the crude product was recrystallized several times from sea clay, the melting point rose to 263.5 to 64.7 C.
