Verfahren zur Herstellung von 3-niedermolekular- Alkoxy-19-nor-d 2,5 (10)_androstadien-17-on-Verbindungen Die vorliegende Erfindung betrifft ein neues Ver fahren zur Herstellung von 3-niedermolekular- Alkoxy-19 -nor-J2.5 (1o) -androstadien-17-on-Verbin- dungen, insbesondere der entsprechenden 3-Methoxy- Verbindung.
3-Methoxy-19-nor-d2.5 (io)-androstadien-17-on ist eine bekannte Zwischenverbindung, die leicht in das wertvolle progestationale Hormon 44-19-nor-17- Äthinyl - androsten - 17ss - o1 - 3 - an (19-nor-Äthinyl- testosteron) umgewandelt werden kann, wie es in der amerikanischen Patentschrift Nr. 2 744 122 an gegeben worden ist.
Obgleich in dieser amerikani schen Patentschrift ein Verfahren für die Herstellung eines 19-nor-Äthinyltestosterons offenbart worden ist, so wurde gemäss vorliegender Erfindung jedoch ein wirksameres Verfahren gefunden, nach welchem als Ausgangsmaterial Oestron benutzt wird, und dieses umfasst die Herstellung des neuen 17-Diacetals des Oestrons mittels Äthyl-orthoformiat, die Um wandlung des Oestron-17-diacetals in die entspre chende 3-niedermolekulare Alkoxy-Verbindung,
die Reduktion des aromatischen Systems im Ring A zu einem Dien-System durch Reaktion mit einem Alkali metall in flüssigem Ammoniak und schliesslich die selektive Hydrolyse des neuen 17-Diacetals des 3 - niedermolekular - Alkoxy - 19 - nor-d 2.5 (io)-andro- stadien-17-ons zwecks Herstellung der bekannten Zwischenverbindungen, nämlich der 3-niedermoleku- laren Alkoxy-Verbindungen von 19-nor-42,
5 cio>- Androstadien-17-on. Diese letzteren Verbindungen, und insbesondere die 3-Methoxy-Verbindung, kön nen, wie bekannt, mit einer 17a-Äthinylgruppe ver sehen werden, i ndem sie mit Acetylen in Gegenwart von Kalium-t-amylat umgesetzt werden, und sie können dann der Wirkung einer verdünnten Säure unterworfen werden, um die Methoxylgruppe bei C-3 zu hydrolysieren und gleichzeitig eine Verschiebung der Doppelbindung zu bewirken,
um das d4-3-Keto- system von 19-nor-17-Athinyltestosteron zu bilden.
Die folgende Gleichung veranschaulicht das er findungsgemässe Verfahren und die Herstellung der vorstehend genannten neuen Zwischenverbindungen.

In der obigen Gleichung steht R für eine nieder molekulare Alkylgruppe.
Die Durchführung des erfindungsgemässen Ver fahrens wird im folgenden beispielsweise beschrie ben: Oestron wird in einem organischen Lösungs mittel, wie abs. Äthanol, suspendiert und mit Äthylorthoformiat behandelt. Es wird dann eine kleine Menge Mineralsäure, wie Schwefelsäure, zu gesetzt und das Gemisch kurze Zeit auf 40 C er hitzt. Darauf wird weiteres Äthylorthoformiat zu gesetzt und das Erhitzen eine kurze Zeit bei einer etwas höheren Temperatur fortgesetzt, bis das ge samte Oestron in Lösung gegangen ist.
Die Lösung wird dann abgekühlt, und es wird langsam ein Alkali, wie Kaliumhydroxyd, und dann Wasser zugesetzt. Die Lösung enthält nun das Diacetal des Oestrons, das ohne Abscheidung für die weitere Reaktion be nutzt werden kann.
Zur Herstellung des entsprechenden Methyl- äthers wird der Lösung tropfenweise Dimethylsulfat zusammen mit Kaliumhydroxyd oder einem andern Alkali in Perlform zugesetzt, wobei der pH-Wert über 11 gehalten wird. Nach dem Zusatz des Methyl- sulfats wird Wasser und eine weitere Menge Alkali zugesetzt. Das Durchrühren wird dann eine kurze Zeit fortgesetzt, um die Hydrolyse des überschüssigen Methylsulfats zu Ende zu führen.
Das erhaltene kri stalline Produkt wird abfiltriert, gewaschen und in einem organischen Lösungsmittel, wie Hexan, das Pyridin enthält, gelöst und die Lösung konzentriert. Es wird ein Öl erhalten, das aus dem 17-Diacetal des Oestron-3-methyl-äthers besteht, das ohne wei tere Reinigung für die nächste Verfahrensstufe be nutzt oder unter Kühlung aufbewahrt werden kann.
Für die Reduktion wird das 17 Diacetal des Oestron- 3-methyläthers in einem organischen Lösungsmittel, wie Dowanol 33 , (Markenprodukt), ein Glykol- monoäther, gelöst und die Lösung auf eine wesentlich unter Zimmertemperatur liegende Temperatur, wie beispielsweise auf etwa 3 C, abgekühlt. Es wird dann wasserfreies flüssiges Ammoniak und dann ein Alkalimetall in kleinen Stücken, wie Lithium- metall, zugesetzt. Die Reaktionslösung wird etwa 1 Stunde durchgerührt, worauf Wasser, das eine kleine Menge Hydrochinon enthält, zugesetzt wird.
Das Pro dukt wird abfiltriert und beispielsweise mit Wasser, das eine kleine Menge Hydrochinon enthält, neutral gewaschen. Das erhaltene Produkt ist das 3-Methyl- äther-17-diacetal von 19-nor-J',5 (1 -Androstadien- 3ss-ol-17-on. Das feuchte Produkt dieser Stufe wird unter Rühren in einem organischen Lösungsmittel, wie Benzol, gelöst. Es wird dann Äthylalkohol zu gesetzt und die Temperatur der Lösung auf Zimmer temperatur oder etwa 20 C eingestellt.
Darauf wird unter kräftigem Rühren gerade nur so viel Säure, vorzugsweise eine verdünnte Mineralsäure, wie Schwefelsäure, in Alkohol zugesetzt, dass eine leicht saure Reaktion erzeugt wird, worauf man die Um setzung sich eine kurze Zeit, wie beispielsweise 16 Sekunden bis zu 1 Minute fortsetzen lässt. Es wurde gefunden, dass bei dieser kurzen Hydrolysezeit, die durch den Zusatz einer Base, wie Kaliumhydroxyd, in Wasser und Alkohol beendet wird, eine quantita tive selektive Hydrolyse der schützenden Diacetal- gruppierung in Stellung 17 erfolgt, ohne dass die Äthergruppe in der C 3-Stellung beeinflusst wird.
Das erhaltene Produkt besteht beispielsweise aus dem 3 -Methoxy-19-nor-J2,5 (io _androstadien-17-on, das nach bekannten Verfahren in 19-nor-Äthinyl- testosteron umgewandelt werden kann. Das vorliegende Verfahren hat den wesentlichen Vorteil, dass die Reduktion des aromatischen Systems zu dem d 2.5 (io)-Diensystem unter dem Schutz der Ketogruppe in C 17-Stellung in Form des Diacetals durchgeführt wird.
Auf diese Weise wird die Not wendigkeit der Regenerierung dieser Gruppe durch nachträgliche Oxydation vermieden, die praktisch nur schwierig und mit Gefahr durchzuführen ist. <I>Beispiel</I> 100 g vollkommen trockenes, in 400 cm3 abs. Athanol suspendiertes Oestron werden mit 100 cm3 wasserfreiem Äthylorthoformiat behandelt.
Unter leichtem Rühren wird dann 1 cmG chemisch reine Schwefelsäure zugesetzt und das Gemisch 33 Mi nuten auf 40 C erwärmt. Es werden dann weitere 25 cm3 Äthylorthoformiat zugesetzt und das Er hitzen 20-30 Minuten auf 55 C fortgesetzt, bis das gesamte Oestron in Lösung gegangen ist. Unter äusserem Kühlen werden dann im Laufe von 15 Mi nuten 50 g Kaliumhydroxyd in Schuppen- oder Perl form zugesetzt, die Temperatur auf 10 C erniedrigt und dann 400 ein- Wasser zugesetzt.
Unter kräftigem Rühren werden dann gleichzei tig a) 55 cm3 Dimethylsulfat mit, einer Geschwin digkeit von etwa 2-3 Tropfen/Sek. und b) Kalium hydroxyd in Perlform in Mengen von jeweils 10 g zugesetzt, so dass der pH-Wert unter 11 zu fallen neigt. Nach dem Zusatz des gesamten Methylsulfats werden allmählich 400 cm3 Wasser zugesetzt und die Menge Kaliumhydroxyd einschliesslich des bereits zugesetzten auf 50 g erhöht. Das Durchrühren wird weitere 30 Minuten fortgesetzt, um die Hydrolyse des überschüssigen Methylsulfats zu Ende zu führen.
Das kristalline Produkt wird abfiltriert (aus dem Fil trat können etwa 501o nicht umgesetztes Oestron durch Ansäuern zurückgewonnen werden), mit leicht alkalischem Wasser gut ausgewaschen und dann in 2 1 Hexan, die 10 em3 Pyridin enthielten, gelöst. Das etwa vorhandene Wasser kann in einem Scheide trichter abgetrennt werden. Die Lösung wird über Natriumsulfat getrocknet und schnell bei der Tempe ratur des Dampfbades konzentriert. Kurz vor Ende wird die Temperatur erniedrigt und die Konzentra tion unter verringertem Druck durchgeführt.
Auf diese Weise wird ein Öl im Gewicht von etwa 135 g erhalten, das aus dem 17-Diacetal des Oestron-3- methyläthers besteht, das ohne weitere Reinigung in der folgenden Verfahrensstufe benutzt werden kann. Wenn die nächste Verfahrensmassnahme nicht sofort durchgeführt wird, so empfiehlt es sich, das Produkt in einem Kühlschrank aufzubewahren.
<B>135</B> g des 17-Diacetals des Oestron-3-methyl- äthers werden in 4 1 wasserfreiem Dowanol <B>33 </B> gelöst und die Lösung auf etwa 3 C abgekühlt. Unter kräftigem Rühren unter wasserfreien Bedingungen werden dann 6 1 wasserfreies flüssiges Ammoniak und dann nach etwa 4-8 Minuten 100g Lithium- metall in kleinen Stücken zugesetzt. Das Durch- rühren wird 60 Minuten fortgesetzt, und es werden dann 10 1 mit Eis versetztes destilliertes Wasser, das 10/0o Hydrochinon enthält, zugesetzt.
Das Pro dukt wird abfiltriert, wobei dafür gesorgt wird, dass es nicht trocken in den Trichter gelangt, und mit einem grossen LUberschuss Wasser, das 10/0o HYdro- chinon enthält, vollkommen neutralgewaschen. Auf diese Weise werden 120 g des 3-Methyläther-17-d'i- a.cetals von 19-nor-d2.5 (io)-Androstadien-3ss-ol-17-on erhalten.
Das feuchte Produkt wird unter leichtem Rühren in 550 cm3 Benzol gelöst, und, falls notwendig, wird Wasser zugesetzt, so dass das Reaktionsgemisch ein Gewicht von 900 g besitzt. Falls das Gemisch ein höheres Gewicht besitzt, wird eine gewisse Menge des Wassers durch Dekantieren entfernt, bis das ge rannte Gewicht erreicht ist. Es werden dann 2 1 96%iger' Alkohol zugesetzt, und die Temperatur der Lösung wird auf 20 C eingestellt.
Unter kräfti gem Rühren wird eine Lösung von 4,4 eins chemisch reiner Schwefelsäure in 50 em3 Alkohol zugesetzt. Nach 16 Sekunden wird eine Lösung von 15g Ka- liumhydroxyd in 10 cm3 Wasser und 50 cm3 Alkohol zugesetzt. Das Gemisch soll leicht alkalisch bleiben, und mit dem Durchrühren wird dann aufgehört, um eine Belüftung der Lösung zu vermeiden. Das Ge misch wird dann unter verringertem Druck bei der Dampfbadtemperatur zur Trockne eingedampft, wo bei ein Stickstoffstrom langsam durch die Lösung geschickt wird.
Nach der Beendigung wird der Stick stoffstrom aufrechterhalten, bis das Produkt abge kühlt ist, worauf das Produkt unter einer Stickstoff atmosphäre gehalten wird, bis es für die folgende Massnahme benutzt wird.
