Il convient de noter que la désignation D ou L n'indique pas néces sairement le signe réel du pouvoir rotatoire, mais se rapporte uniquement à la configu ration de l'atome de carbone en position a du noyau phénylique. La présente invention concerne la prépara tion des isomères thréo des dérivés repré sentés par la formule ci-dessus.

Ce procédé est, caractérisé en ce que l'on chauffe un l.-p-nitrophényl-2-dieliloracét- aniido-3-acétoxy-propanol-1 avec le chlorure de thionyle. Le thréo-1-p-nitrophényl-1-chloro- 2-dichloracétamido-3-acétoxy-propane ainsi obtenu est une nouvelle substance cristallisée dont la.
forme racémique fond à 1.20-121 C, tandis que la forme optiquement active D fond à 101-102 C et présente un pouvoir rotatoire [a] 20 =-8 (c = 6,401o dans l'acé tate d'éthyle). Toutes les formes optiques (D, L et DL) de cette nouvelle substance sont des produits intermédiaires importants pour la préparation d'antibiotiques du type du chlor amphénicol.
On peut utiliser pour le procédé selon l'invention soit la forme thréo soit la, forme allo du 3-acétoxy-propanol-1 de départ. Lorsqu'on part de la forme allo du corps de :départ, une épimérisation en la forme thréo a lieu lors de la substitution du groupe hydroxyle par l'atome de chlore.
Le D-allo- propanol-1 de départ donne le L-thréo-1- chloropropane et le L-allo-propanol-1 de départ donne l'isomère D-thréo du 1-chl.oro- propane désiré.
D'ordinaire, on chauffe le mélange réac tionnel à reflux jusqu'à achèvement de la réaction.
Le 1-p-nitrophényl-2-dichloracétamido-3- acétoxy-propanol-1 de départ. peut, par exem ple, être préparé par action du chlorure d'acétyle sur le 1-p-nitrophényl-2-dichloracét- amido-propanediol-1,3, en présence d'une base organique tertiaire.
Les exemples suivants montrent comment le procédé selon l'invention peut être mis en aeuvre <I>Exemple 1:</I> On chauffe à reflux, pendant 2 heures, 5,0 4 de DL-allo-l.-p-nitrophényl 2-dichloracétamido- 3-acétoxy-propanol-1 (P. F. =11 _2-115 ) dans 12,5 cm3 de chlorure de thionyle pur. On chasse l'excès de chlorure de thionyle sous pression réduite.
On reprend le résidu par 30 em3 d'éther; le produit cristallise, et on essore, lave à l'éther et sèche sous vide. On obtient 3,4g de DL-thréo-1-p-niti ophényl-1- chloro - 2 -,dichloracétamido - 3-acétoxy-propane de P. F. = 116-118 qui, après recristallisa- tion dans l'acétate d'éthyle, fond à 120 .
On peut obtenir le corps de départ par action de CH3COC1 sur le DL-allo-propane- cliol-1,3 correspondant, en présence de pyri- cline.
Exemple. <I>2:</I> En opérant comme dans ].'exemple 1, mais en utilisant 9,05 4 de L-allo-1-p-nitrophényl- 2-dichloracétamido-3-acétoxy-px-opanol-1 de P. F. = 112-11:

F. = 132-133 ) dans 20 cm3 de dioxane anhydre et 2,6 g de pyridin anhydre, on ajoute, en agitant et refroidissant., 4 g de chlorure de thionyle pur (rectifié sur huile de lin) ; on place le mélange réactionnel dans un bain d'huile à 80-90 pendant. 40 minutes, puis on distille à froid sous vide le dioxane. Le résidu est dissous dans 70 em3 de chloro forme; la solution ehloroformique est lavée à l'eau, séchée sur du sulfate de sodium et déco lorée à, l'aide de charbon de bois;
on chasse le chloroforme à froid sous vide jusqu'à siccité. Par recristallisation du résidu dans 45 em3 de benzène, on obtient 10,7 g de DL-thréo-1-p- nitr ophény l-1.-chl oro-2-diclilora cétamido-3-acét- oxy-propane: P.
