On peut utiliser comme produit de départ soit la forme racémique, soit une des formes optiquement actives de ladite thréo-oxazoline. On préfère utiliser le racémique ou la forme dextrogyre (D) de cette oxazôline. Le thréo-1-p-nitrophényl-2-dichloracét- amido-3-chloro-propanol-1 obtenu par le pro cédé selon l'invention est une nouvelle sub stance cristallisée. Toutes ses formes optiques (DL, D et L) sont, des produits intermédiaires importants pour la préparation d'antibioti ques.
La. forme racémique fond à 135-l36 C, et les isomères optiquement actifs (D et L) fondent. à 130-l31 C. Le pouvoir rotatoire [a] D est de + 20 7 pour l'isomère D-thréo (c = 4 % dans l'acétate d'éthyle), et de - 21 8 pour l'isomère L-thréo (c = 4,6 0/0 dans l'acétate d'éthyle).
L'oxazoline de départ pour le procédé selon l'invention peut- être préparée de la ma nière suivante: On dissout 1,93 g de chlor- hydrate de DL-thréo-1-p-nitrophényl-2-amino- propanediol-1,3 dans 13 cm3 de pyridine an hydre. Puis on ajoute à. cette solution 1,8 g de chlorhydrate ,d'éther diehloracétiinino-éthy- lique (préparé par réaction entre du dichlor- acétonitrile et de l'éthanol en présence d'acide chlorhydrique).
On laisse la. solution au repos pendant deux heures à la température am biante. Il se forme un précipité de chlorhy- drate de pyridine, et- on verse le mélange dans 100 cms d'eau. De cette façon, la DL-thréo='- dichlorométhyl-4- (p-nitrophényl) -hydroxymé- thyl-oxazoline-2 cristallise rapidement et est ensuite séparée par filtration, lavée à l'eau et.
séchée à 100 C; point de fusion: 1.60-161 C.
Les exemples suivants montrent comment le procédé selon l'invention peut être mis en ceuvre. <I>Exemple 1: '</I> On dissout à température ordinaire 0,15 g de DL-thréo-2-diehlorométhyl-4-(p-nitrophé- nyl)-hydroxyméthyl-oxazoline-2 (point de fu sion: 160-161 C) dans 3 cm3 de dioxane an- hydre contenant 14 % d'acide chlorhydrique sec.
On laisse reposer 30 minutes, puis on verse dans 30 cm3 d'eau. Un produit cristal lise, qu'on essore, lave à l'eau et sèche dans le vide sur de l'acide sulfurique. On obtient ainsi 0,075 g de DL-thréo-1-p-nitrophényl-2-d'i- chloracétamido-3-chloro-propanol-l du point de fusion 135-136 C.
<I>Exemple 2:</I> On dissout à température ordinaire 3 g de L-thr éo - 2 - dichlorométhyl - 4 - (p-nitrophényl) - hydroxyniéthyl-oxazoline-'du point de fusion 141 C et [a] D = + 1"r3 (e - 0,57 "/o dan, l'acétate d'éthyle), dans 30 em3 de dioxane contenant. 13 0i0 d'acide chlorhydrique sec, en refroidissant. pour que la température ne dé passe pas 28 C.
On laisse reposer une demi heure à température ordinaire, puis on clia,se le dioxane ainsi que l'excès de 1-ICI dans le vide, à froid. On reprend le résidu par l'eau; après amorcage, le produit cristallise en peti tes aiguilles. On filtre ces aiguilles, les lave àll'eau et on les sèche sur de l'acide sulfurique dans le vide.
On obtient. ainsi 2,1 g de L-thréo- 1-p-nitropliényl- 2 -dielilora cét amido - 3 - ehloro- propanol-1 du point de fusion 130-1.31 C; [a] D = - 21 8 (c = 4,6% dans l'acétate d'éthyle).
<I>Exemple 3:</I> En opérant comme dans l'exemple 2, mais à partir de 3 g de D-thréo-2-dichlorométliyl- 4-(p-nitrophényl)-hy droxyméthyl-oxazoline-2 [point de fusion:
