On obtient.

Process for preparing threo-2-dichloromethyl-4- (p-nitrophenyl) -hydroxymethyl-oxazoline- (2). In the main patent a process for the preparation of the isomer has been described; eometric allo of? -Clieliloromethvl-4- (p-tiitro- l) Iietivl) -li, # - droxvtnétlivl-oxazoline- (') of formula
EMI0001.0012
The present patent relates to a process (the preparation of the geometric isomer threo (the same substance.
Due to the presence of two asymmetric carbon atonies in the a and p positions of the phenolic noj-au, the preceding formula corresponds to two epic geometric shapes depending on the relative position of the substituents carried. by these two asymmetric carbon atoms.
By analogy with the nomenclature adopted by Rebstock and others (Ain. Soc. 71, pages 2458-2473 [19-191), with regard to (it concerns the diastereoisomers of 1-p- n itropliénvl - 2- dichloroacétamido-pt # opanediol- 1,) of which one of the isomers is known as tio iii (chloramphenicol, these two geometric isomers (structural isomers)
are de-iso-n signed ièreallo by is the prefixes also designated a llo and.
thr isomer om ervthro o .L 'or regular or cis, and the threo isomer is often referred to as pseudo or trans. Either of the two geometric isomers allo and threo can exist in its racemic form (DL) and in two optically active forms (D and L)
giving a total of six different isomers. The preceding structural formula therefore denotes both the complete mixture of all six forms, the racemates of. allo series and threo series, as well as the four individual isomers L-allo, D-allo, L-threo and D-threo. It should be noted that the.
The configurational designation of isomers such as D and L has no relation to the actual sense of rotational power, but refers only to the configuration at the carbon atom in the a position of the phenyl ring.
The D-threo isomer a. the. same setup regarding this. carbon atom in position a that the therapeutically active isomer of chloramphenicol which has been designated as D-threo-1-p-nitrophenyl =? - dichloroacetamido - pro-panediol-1,3 and whose rotatory power is negative in the 'ethyl acetate.
The process according to the invention for the preparation of tlireo-2-dichloromethvl-4- (p-nitrophenvl) -hvdroxvmethyl-oxazoline- (2) is. characterized in that it is subjected to cyclization in a medium. alkaline threo-1-p- nitrophénv 1 = 2-dichlora.cétamido - 3 - chloro - pro panol- (1) of formula
EMI0002.0001
The threo-oxazoline thus obtained is not a new substance.
All the optical forms (DL, L and D) of this threo-oxazoline have interesting antibiotic properties and can also serve as a starting product for the. preparation. substances with antibiotic activity.
Preferably, the. evelisation using a hydroxide of an alkali metal, such as NaOH or IZOH, in a dilute aqueous solution. This cyclization can be carried out in the presence of an organic solvent miscible with water, such as ethanol. Usually, the starting material is dissolved in an organic solvent miscible with. water and then added to this solution a dilute aqueous solution of NaOH or KOH. The. Temperature of the. cyclization is preferably close to room temperature.
The racemic form or one of the optically active forms of threo-1-p-nitrophenyl \? -Dichloracetamido-3-chloro-propanol-1 can be used as starting material.
The starting material for the process according to the invention can be prepared as follows. Heated under reflux for 2 hours 50 @> DL-a-dicliloracétanüdo-p) -hvdroxr-p-nitro-propiophenone and 65 em3 of thionyl chloride. The mixture is left to crystallize in the cold, filtered, washed with anhydrous ether and the product separated off in vacuo over 1% OH. We obtain. 42.7 g of DL-a-dichlorac6t-amido-N-chloro-p-nitro-propiophenone, melting at 137-140 C.
A mixture of 10.5 g of the latter product is heated with 16 g of aluminum isopropylate and 250 (msec of anhydrous isopropyl alcohol. During this heating, 135 em3 of a mixture of acetone and isopropanol are removed by distillation After cooling, the reaction mixture is taken up in 90 em3 of 2N sulfuric acid.
