Verfahren zur Darstellung eines Sulfons. Die Erfindung betrifft ein Verfahren zur Darstellung des bekannten 4,4'-Diamino- diphenylsulfons, welches farblose Kristalle vom Schmelzpunkt 175 bildet und als Zwi schenprodukt zur Gewinnung von Heilmit teln verwendet wird.
Die bekannten Verfahren zur Darstellung dieses Sulfons sind komplizierte Synthesen, die über mehrere Zwischenstufen mit schlech ten Ausbeuten zum Endprodukt führen. So beschreibt z. B. Van Arendonk (Journal of the American Chemical Society 62, 3521) ein Verfahren, bei dem er vom Anilin ausgehend zunächst das 4,4'-Diamino-diphenylsulfid ge winnt, dessen Diacetylderivat zum 4,
4'-Di- acetylamino - diphenylsulfon oxydiert und daraus durch Verseifung das 4,4'-Diamino- diphenylsulfon erhält. Ein ähnliches, in der ungarischen Patentschrift Nr. 121910 be schriebenes Verfahren geht vom 4-Nitro-4'- acetylamino-diphenylsulfid aus.
Das durch Oxydation daraus erhaltene 4-Nitro-4'-acetyl- a.mino-diphenylsulfon wird z im 4-Amino-4'- acetylamino-diphenylsulfon reduziert; wel ches bei der Verseifung das 4,4'-Diamino- diphenylsulfon liefert. Roblin und Mitarbeiter geben in der USA.-Patentschrift Nr.
2227400 und im Journal of the American Chemical Society 63, 1930 ein Verfahren an, nach welchem p - Acetylamino - benzolsulfonsäure- chlorid zur entsprechenden Sulfinsäure redu ziert wird.
Deren Natriumsalz wird sodann mit einem p-halogensubstituierten Nitroben- zol zum 4-Acetylamino-4'-nitro-diphenylsul- fon umgesetzt, welches durch Reduktion der Nitrogruppe zur Aminogruppe und Abspal tung des Acetylrestes in das 4,4'-Diamino- diphenylsulfon übergeführt wird.
Der Versuch, das 4,4'-Diacetylamino-di- phenylsulfon aus Aeetanilid und p-Acetyl- amino-benzolsulfonsäurechlorid in Gegenwart der zur Ausführung der Friedel-Craftsschen Synthese üblichen Menge Aluminiumchlorid herzustellen, misslingt.
Es wurde nun ge funden, dass sich das Acetanilid und das p-Acetylamino-benzolsulfonsäurechlorid über- raschenderweise mit guter Ausbeute dann zum 4,4'-Diacetylamino-diphenylsulfon um setzen lassen, wenn man die Umsetzung in Gegenwart einer solchen Menge Aluminium chlorid vornimmt, dass auf 1 Molekül Acet- anilid mindestens 3 Moleküle Aluminium chlorid kommen.
Die Auffindung der Durch führbarkeit dieser Reaktion ist deshalb von grosser Bedeutung, weil sie die Möglichkeit schafft, das 4,4'-Diamino-diphenylsulfon aus den leicht erhältlichen Ausgangsstoffen Acet- anilid und p-Acetylamino-benzolsulfonsäure- chlorid und Abspaltung der Acetylgruppen aus dem daraus gewonnenen 4,4'-Diacetyl- amino-diphenylsulfon .darzustellen, also nach einem Verfahren, welches gegenüber den eingangs erwähnten, bekannten Verfahren zur Darstellung des 4,
4'-Diamino-diphenyl- sulfons als besonders einfach und aussichts reich bezeichnet werden kann.
Die Erfindung betrifft ein Verfahren zur Darstellung eines Sulfons, dadurch gekenn zeichnet, dass man ein Acylaminobenzol und ein p-Acylamino-benzolsulfonsäurehalogenid in Gegenwart einer mit Bezug auf das Acylaminobenzol mindestens dreifachen mo- laren Menge eines Kondensationsmittels, z. B.
Aluminiumchlorid, Ferrichlorid, Zinkchlorid, in der Hitze aufeinander einwirken lässt und das dabei unter Abspaltung von HCl erhal tene 4,4'- Diacylamino-diphenylsulfon zum 4,4'-Diamino-diphenylsulfon hydrolysiert.
Die Kondensation wird zweckmässig bei einer zwischen 120 und 200 C liegenden Temperatur vorgenommen. Es hat sich als besonders vorteilhaft erwiesen, auf 1 Mole kül Acylaminobenzol wenigstens annähernd 6 Moleküle Aluminiumchlorid zu verwenden.
Zur Darstellung des 4,4'-Diamino-diphenyl- sulfons nach dem neuen Verfahren verfährt man beispielsweise folgendermassen: Ein inniges Gemisch von 34 g Acetanilid und 200g wasserfreiem Aluminiumchlorid wird auf etwa 160 C erhitzt. In die Schmelze werden unter starkem Rühren 120 g trockenes p-Acetylamino-benzolsulfonsäurechlorid lang sam eingetragen. Nach dem Aufhören der HCl-Entwicklung lässt man erkalten und trägt das fein pulverisierte Reaktionsprodukt unter Rühren in 600 cm'' heisses Wasser ein, wobei nur das 4,4'-Diacetylamino-diphenyl- sulfon ungelöst bleibt.
Das gesammelte Diacetylderivat wird durch zweistündiges Kochen mit 200 cm' 5-n Salzsäure hydrolysiert. Beim Neutrali sieren der heissen, salzsauren Lösung auf px = 4 scheiden sich 40,1g (= 64,0 % d. Th.) reines, weisses 4,4'-Diamino-diphenylsulfon in kleinen Nadeln aus.
An Stelle des p-Acetylamino-benzolsulfon- säurechlorids kann auch ein anderes p-Acyl- amino-benzolsulfonsäurechlorid bezw. ein an deres p-Acetylamino-benzolsulfonsäurehalo- genid verwendet werden.
Process for the preparation of a sulfone. The invention relates to a method for the preparation of the known 4,4'-diamino diphenyl sulfone, which forms colorless crystals with a melting point of 175 and is used as an inter mediate product for the production of Heilmit items.
The known processes for the preparation of this sulfone are complicated syntheses which lead to the end product via several intermediate stages with poor yields. For example, B. Van Arendonk (Journal of the American Chemical Society 62, 3521) a process in which he initially wins the 4,4'-diamino-diphenyl sulfide starting from aniline, the diacetyl derivative of which is the 4th
4'-Di-acetylamino-diphenylsulfone is oxidized and the 4,4'-diaminodiphenylsulfone is obtained therefrom by saponification. A similar process described in Hungarian Patent No. 121910 starts with 4-nitro-4'-acetylamino-diphenyl sulfide.
The 4-nitro-4'-acetyl-a.mino-diphenylsulfone obtained therefrom by oxidation is reduced to 4-amino-4'-acetylamino-diphenylsulfone; which gives the 4,4'-diaminodiphenyl sulfone on saponification. Roblin et al., U.S. Patent No.
2227400 and in the Journal of the American Chemical Society 63, 1930 a process by which p - acetylamino - benzenesulfonic acid chloride is reduced to the corresponding sulfinic acid.
The sodium salt thereof is then reacted with a p-halogen-substituted nitrobenzene to give 4-acetylamino-4'-nitro-diphenylsulfone, which is converted into 4,4'-diaminodiphenylsulfone by reducing the nitro group to the amino group and cleaving off the acetyl radical becomes.
The attempt to prepare the 4,4'-diacetylamino-diphenylsulfone from ethanilide and p-acetylamino-benzenesulfonic acid chloride in the presence of the amount of aluminum chloride customary for carrying out the Friedel-Crafts synthesis fails.
It has now been found that the acetanilide and the p-acetylamino-benzenesulphonic acid chloride can, surprisingly, be converted into 4,4'-diacetylamino-diphenylsulphone with good yield if the reaction is carried out in the presence of such an amount of aluminum chloride as that for 1 molecule of acetanilide there are at least 3 molecules of aluminum chloride.
Finding the feasibility of this reaction is of great importance because it creates the possibility of the 4,4'-diamino-diphenyl sulfone from the readily available starting materials acetanilide and p-acetylamino-benzenesulfonic acid chloride and splitting off the acetyl groups from the 4,4'-diacetylamino-diphenylsulfone obtained therefrom. to represent, i.e. according to a process which, compared to the known processes mentioned at the beginning, for the representation of the 4,
4'-diamino-diphenyl sulfone can be described as particularly simple and promising.
The invention relates to a method for the preparation of a sulfone, characterized in that an acylaminobenzene and a p-acylamino-benzenesulfonic acid halide in the presence of at least three times the molar amount of a condensing agent with respect to the acylaminobenzene, eg. B.
Aluminum chloride, ferric chloride, zinc chloride, allowed to act on each other in the heat and hydrolyzed the 4,4'-diacylamino-diphenylsulfone obtained by splitting off HCl to give 4,4'-diamino-diphenylsulfone.
The condensation is expediently carried out at a temperature between 120 and 200.degree. It has been found to be particularly advantageous to use at least approximately 6 molecules of aluminum chloride per 1 mol of acylaminobenzene.
The procedure for preparing 4,4'-diamino-diphenylsulfone by the new process is as follows: An intimate mixture of 34 g of acetanilide and 200 g of anhydrous aluminum chloride is heated to about 160.degree. 120 g of dry p-acetylaminobenzenesulfonic acid chloride are slowly introduced into the melt with vigorous stirring. After the evolution of HCl has ceased, the mixture is allowed to cool and the finely powdered reaction product is introduced into 600 cm ″ of hot water while stirring, only the 4,4′-diacetylamino-diphenylsulfone remaining undissolved.
The collected diacetyl derivative is hydrolyzed by boiling with 200 cm 5N hydrochloric acid for two hours. When the hot, hydrochloric acid solution is neutralized to px = 4, 40.1 g (= 64.0% of theory) of pure, white 4,4'-diamino-diphenylsulfone separate out in small needles.
Instead of the p-acetylamino-benzenesulfonic acid chloride, another p-acylamino-benzenesulfonic acid chloride can also be used. Another p-acetylamino-benzenesulfonic acid halide can be used.
