Verfahren zur Darstellung eines Ogy-aldehyds der Cyclopentanopolyhydro- phenanthr en-Reihe. Es wurde gefunden, dass man zu einem neuen Oxy-aldehyd der Cyclopentanopoly- hydrophenanthren-Reihe gelangen kann, wenn man in 17-Trioxypropyl-testosteron, dessen 17-ständige Seitenkette der Formel

entspricht, die Hydroxylgruppen 1 und 2 mit Carbonylverbindungen in Gegenwart wasser entziehender Mittel kondensiert,
die Hydro- xylgruppe 3 verestert, anschliessend mit sau ren Mitteln die Hydroxylgruppen 1 und 2 wieder in Freiheit setzt, das entstandene Gly- kol mit Oxydationsmitteln zum Ester des d4-Pregnen-3-on-17,20-diol-21-als aufspaltet und diesen zum freien Oxy-aldehyd verseift.
Der Ausgangsstoff für das neue Verfah ren lässt sich nach an sich bekannten Me thoden darstellen, beispielsweise aus 17-Allyl- testosteron durch Wasserabspaltung und an schliessende Hydroxylierung. In erster Stufe wird der Ausgangsstoff mit Carbonylverbindungen, wie Aceton, Me- thyläthylketon, Cyclohexanon oder Benzalde- hyd, in Gegenwart von wasserentziehenden Mitteln, beispielsweise von Säuren oder von wasserfreien Salzen, wie Kupfersulfat, Na triumsulfat oder Zinkchlorid, behandelt.
Auf diese Art lassen sich die beiden aussen stehenden Hydroxylgruppen in der 17-stän- digen Seitenkette durch Umwandlung in die entsprechenden Derivate, beispielsweise die getonide, intermediär schützen. Es folgt in zweiter Stufe eine, zum Beispiel in üblicher Weise und insbesondere in Gegenwart basi scher Mittel durchgeführte Veresterung .der noch freien dritten Hydroxylgruppe der Sei tenkette.
Hierbei können zum Beispiel alipha- tische, hydroaromatische oder aromatische Carbonsäure- oder Sulfonsäurereste einge führt werden, wie die Reste der Ameisen-, Essig-, Propion-, Butter-, Benzoe- oder Toluolsulf onsäuren. Anschliessend spaltet man die früher ein geführte Carbonylverbindung mit sauren Mitteln, zum Beispiel mit organischen oder anorganischen Säuren, wie Essig-,
Schwefel- oder Halogen-Wasserstoffsäuren, wieder ab, und setzt so die intermediär geschützt ge- wesenen Hydroxylgruppen 1 und 2 wieder in Freiheit. Diese Abspaltung lässt sich über raschenderweise so durchführen, dass die an der dritten Hydroxylgruppe sitzenden Säure reste, selbst wenn es sich um Reste niederer aliphatischer Säuren handelt, nicht an die frei werdende gegebenenfalls primäre Car- binolgruppe wandern, wie dies in der Zucker reihe sonst meist beobachtet wird.
Die in Freiheit gesetzte Glykolgruppie- rung spaltet man nun in an sich bekannter Weise, zum Beispiel mit Bleitetraacylaten oder mit Perjodsäure, zum Aldehyd auf, man gewinnt dabei Ester, die anschliessend durch hydrolysierende Mittel, beispielsweise durch Bicarbonate, in den freien Oxy-aldehyd über geführt werden.
Nach dem erfindungsgemässen Verfahren gewinnt man das d4-Pregnen-3-on-17,20-diol- 21-al in Form farbloser Kristalle. Es soll therapeutische Verwendung finden oder als Zwischenprodukt zur Herstellung therapeu tisch verwendbarer Stoffe dienen.
<I>Beispiel:</I> 2 Teile 17-Trioxypropyl-testosteron, dar stellbar durch Wasserabspaltung aus 17- Allyl-testosteron und anschliessender Hy- droxylierung mit Osmiumtetroxyd, werden in 4000 Teilen Aceton gelöst, mit 30 Teilen wasserfreiem Kupfersulfat versetzt und 15 Stunden auf der Maschine geschüttelt. Es wird filtriert, mit Aceton nachgewaschen, die klare Lösung 10 Minuten mit etwas ge hulverter Pottasche geschüttelt und erneut filtriert. Das klare Filtrat dampft man ein, löst den Rückstand in absolutem Äther, fil triert nochmals und dampft die Lösung wie derum ein.
Durch Umkristallisieren des Rückstandes aus Äther erhält man 1,85 Ge wichtsteile der Monoaceton-Verbindung vom Smp. 220 bis 235 . Bei weiterer Umkristalli- sation aus Aceton gewinnt man feine, farb lose Nadeln vom Smp. 235 bis 236,5 .
Die Aceton-Verbindung vom Smp. 220 bis 235 wird in 20 Teilen absolutem Pyri- din gelöst, mit 15 Teilen Essigsäureanhy- drid versetzt und 2 Stunden auf 60 erwärmt. Dann wird im Vakuum eingedampft, der Rückstand in Äther aufgenommen, die Lö sung mit verdünnter Salzsäure, Sodalösung und Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft. Den Rückstand kristallisiert man aus Äther um und erhält so 1,5 Gewichtsteile des Acetats vom Smp. 205 bis 217 .
Bei weiterem Umkristallisieren aus Aceton-Äther werden farblose Rhomben und langgestreckte Sechsecke erhalten, die bei 221 bis 223 schmelzen. Die spezifische Drehung beträgt [a] 17 = -(-107,4 2 (c = 2,01 in Aceton).
0,8 Teile der acetylierten Aceton-Verbin- dung werden in 10 Teilen reinstem Eisessig gelöst, mit 8 Teilen Wasser versetzt und hier auf 11 /_# Stunden auf 55 erwärmt. Dann dampft man im Vakuum bei 50 Badtem- peratur vollständig ein. Der amorphe Rück stand wird in wenig Aceton gelöst und mit der gleichen Menge Äther versetzt, worauf bald Kristallisation eintritt. Die Kristalle werden abgenutscht und mit Aceton und Äther gewaschen.
Ausbeute an Trioxypropyl-20- monoaeetat-Verbindung: 48 mg vom Smp. 198 bis 205 ; durch Umkristallisieren aus Aceton erhält man beidseitig zugespitzte Stäbchen vom Smp. 210 bis 211,5 . Die Mischprobe mit dem Ausgangsmaterial schmilzt bei 175 bis 195 .
0,3 Teile des beschriebenen Monoacetates werden in 30 Teilen reinstem und frisch über Natrium destilliertem Dioxan gelöst, mit der Lösung von 0,34 Teilen Perjodsäure in 6 Tei len Wasser versetzt und 16 Stunden bei Zim mertemperatur in Kohlendioxyd-Atmosphäre stehen gelassen. Hierauf engt man im Va kuum bei 30 Badtemperatur ein, versetzt nochmals mit 30 Teilen Wasser und engt wiederum im Vakuum stark ein. Die dadurch entstandene Fällung wird durch Ausschüt teln mit frisch destilliertem Äther in Lösung gebracht. Die Ätherauszüge werden mit Was ser und etwas galiumbicarbonatlösung ge waschen, mit Natriumsulfat getrocknet und eingedampft.
Man erhält 0,28 Teile eines Rohproduktes, das aus Aceton-Äther kristal lisiert. Die mit Äther und Pentan gewa schenen Kristalle schmelzen bei 201 bis 204 . Nochmaliges Umkristallisieren aus Aceton- Äther ergibt wollige Nädelchen, die unter leichter Zersetzung bei 206 bis 208 schmel zen. Aus den Mutterlaugen lassen sich wei tere Mengen derselben Verbindung vom Smp. 202 bis 205 erhalten.
Nach der Ana lyse erweist sie sich als 44-Pregnen-3-on- 17,20-diol-21-al-20-monoacetat. Die Substanz reduziert, in wenig Methanol gelöst, alkali sche Silber-diaminlösung bei Zimmertempera tur, jedoch etwas langsamer als Corticosteron und ähnliche Stoffe. Mit 1,4-Dioxynaph- thalin wird eine starke Rotfärbung erhalten. Diese Reaktion fällt mit Desoxy-corticosteron und seinem Acetat negativ aus.
Wird eine Probe der Substanz auf dem Objektträger mit wenig konzentrierter Schwefelsäure befeuch tet, so entsteht eine in der Durchsicht braun orange Lösung, die auf schwarzer Unterlage lebhaft grün fluoresziert. Der neue Acetoxy- aldehyd ist die erste synthetische Substanz, die einen positiven Ausfall dieser von ge- I wissen Nebennierenrinden-Hormonen her be kannten Reaktion ergibt.
In analoger Weise lassen sich auch an dere Ester, beispielsweise das Propionat, Bu- tyrat oder Benzoat gewinnen. Semicarbazon: 1 Teil des Acetoxy-alde- hyds@ wird in 300 Teilen Dioxan gelöst und mit der Lösung von 3 Teilen Semicarbazid- chlorhydrat und 4,5 Teilen Natriumacetat 2 Minuten gekocht. Es tritt keine feste Ab scheidung, sondern nur leichte Gelbfärbung ein. Die Lösung wird filtriert, im Vakuum stark eingeengt und mit Wasser versetzt.
Das flockig ausgefällte Produkt wird abge- nutscht, mit Wasser gewaschen, in Methanol gelöst und die erneut filtrierte Lösung nach Zusatz von Wasser stark eingeengt. Das als feines Pulver ausfallende Material wird ab- genutscht, mit Wasser und Äther gewaschen und im Vakuum getrocknet. Beim Erwär men färbt es sich von 190 an braun und zer setzt sich ohne zu schmelzen.
Der Acetogy-aldehyd wird durch wässrig- alkoholische Kaliumbicarbonatlösung bei Zimmertemperatur zum d4 - Pregnen - 3 - on- 17,20-diol-21-al verseift, das in Form farb loser Kristalle erhalten wird.

There is no solid separation, only a slight yellow color. The solution is filtered, concentrated strongly in vacuo and mixed with water.
The flocculent precipitated product is filtered off with suction, washed with water, dissolved in methanol and the solution, which has been filtered again, is strongly concentrated after adding water.
