Verfahren zur Darstellung eines 2,4-Diogo-3,3-dialhyl-tetra- hydropyridin-6-carbonsäureesters. Nach dem Verfahren der schweizerischen Patentschrift Nr. 187826 und Zusätze kann man die durch Kondensation von Dialkyl- acetessigestern mit Ameisensäureestern und nachherige Einwirkung von Ammoniak gewonnenen Aminomethylendialkylacetessig- ester durch alkalische Kondensationsmittel leicht in Pyridinabkömmlinge überführen.
Es wurde nun gefunden, dass man zu 2,4- Dioxo-3,3-diakyltetrahydropyridin-6 - carbon- säureestern bezw. den freien Säuren ge langen kann, indem man 2,2 - Dialkyl - 3,5 - dioxo-hexan-disäure-(1,6)-dialkylester mit Ammoniak umsetzt und auf den so erhaltenen 2,2-Dialkyl-3-oxy-5-amino-hexen- (4)-disäure- (1,6)-dialkylester Alkali-Alkohollate einwir ken lässt und die. zunächst entstehenden Ester gegebenenfalls verseift.
Vertreter der durch Einwirkung von Oxal- ester auf Dialkylacetessigester erhältlichen Ausgangsstoffe, das heisst der 2,2 - Dialkyl- 3,5-dioxo-hexan-disäure- (1,6) - dialkylester, sind bis jetzt nicht bekannt. Sie bilden farb lose, im luftverdünnten Raum unzersetzt, aber ziemlich hoch siedende Öle.
Bei der Ein wirkung von Ammoniak bleiben beide Carb- oxylestergruppen unverändert; es entstehen hierbei zunächst die Ammoniumsalze der Diogoester, die man, wenigstens in der Salz form, als 2,2-Dialkyl-3-oxo-hexen-(4)-ol-(5)- disäure - (1,6) - dialkylester ansprechen muss.
Durch gelindes Erwärmen werden diese Am moniumsalze unter Wasserabspaltung in 2,2- Dialkyl - 3 -oxo- 5 -amino -hexen- (4) - disäure- (J,6)-diester umgewandelt. Die Aminohexen- säureester sind ebenfalls hoch, aber im luft verdünnten Raum unzersetzt siedende Öle.
Lässt man auf sie Alkalialkoholate einwirken, so entstehen in glatter Umsetzung 2,4-Dioxo- 3, 3 - dialkyltetrahydropyridlin- 6 - carbonsäure- ester. Die vorhin erwähnten, Umsetzungen lassen sich wie folgt darstellen; dabei bedeuten R = Alkyl, R' und R" = Alkyle, die gleich oder verschieden sein können:

Die neuen 2,4 - Dioxo - 3,3 - dialky ltetra- hydropyridin - 6 - carbonsäureester sind nie drig schmelzende, destillierbare Verbindun gen, die bei Einwirkung von Alkalien leicht verseift werden. Die entsprechenden freien Säuren kristallisieren gut und bilden neu tral reagierende, in Wasser leicht lösliche Alkalisalze. Sie stellen Zwischenprodukte für die Herstellung von Arzneimitteln dar.
So kann man ausgehend von den freien Säu ren durch Decarboxylierung zu den aus der schweizerischen Patentschrift Nr.187826 und Zusätze bekannten Schlafmitteln der Pyridin- reihe gelangen.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung von 2,4-Di oxo-3,3-di-n-propyl-tetrahydropyridin-6-ca.r- bonsäureäthylester, -welches dadurch gekenn zeichnet ist, dass man 2,2-Di-n-propyl-3,5- dioxo - heran - disäure - (1,6) - diäthylester mit Ammoniak umsetzt und auf den so erhal tenen 2,2-Di-n-propyl-3-oxo-5-amino-hexen- .1)-disäure-(1,6)-diätbvlester Natriumäthvlat einwirken lässt.
Die neue Verbindung besteht aus gelbe liehen Kristallen vom Schmelzpunkt 64 bis 65 C, welche unter 14 mm bei 905 bis 206 C sieden. Sie ist in organischen Lö sungsmitteln leicht löslich und kann aus einer Lösung in hochsiedendem Petroläther (1 : 1) umkristallisiert werden. Sie dient als Zwi- schenprodukt für die Synthese von Arznei mitteln der Pyridinreihe.
Beispiel: Aus Di-n-prop@#1-acetessigester wird mit- telst Oxalester und _Natriummetall der 2,2- Di-n-propyl-3.5-di.oxo-hexan-disäure, (1,6)-di- äthylester, welcher unter 14 mm bei 195 bis 197 C siedet, gewonnen. Dieser wird durch Einwirkung von Ammoniak in den unter 14 mm Druck bei 202 bis 204 C siedenden 2.2-Di-n-propyl-3-oxo-:3-amino -hexen - (4) - d i säure-(1.6)-diäthylester übergeführt.
313 Gewichtsteile dieses Esters werden mit 290 Gewichtsteilen Natriumäthylat- lösung, enthaltend 23 Gewichtsteile Natrium, -während 30 'Minuten am Rückfloss gekocht. Die braune Lösung wird abgekühlt und mit rund 90 Gewichtsteilen konzentrierter Salz säure angesäuert, wobei starke Aufhellung bis zu schwach gelber Farbe eintritt.
Der vom Äthylalkohol befreite 2,4-Dioxo-3,3-di- n - propyl - tetrahydropyridin-6-carbonsäure- äthylester wird in Äther aufgenommen, die Ätherlösung mit 5 ö iger Natriumcarbonat- lösung gewaschen und der nach dem Ver jagen des Äthers verbleibende Rückstand unter vermindertem Druck destilliert. Er siedet unter 14 mm bei 205 bis 206 C und schmilzt bei 64 bis 65 C.
Er löst sich sehr leicht in organischen Lösungsmitteln und kann aus der gleichen Menge hochsiedendem Petroläther umkristallisiert werden.
Die daraus erhältliche 2,4-Dioxo-3,3-di- n - propyl - tetrahydropyridin - 6 - carbonsäure kristallisiert aus verdünntem Alkohol mit Kristallwasser und schmilzt bei 110 bis 111 C.
Man kann auch ohne vorherige Abtren nung des Esters unmittelbar zu der freien Säure gelangen, wenn man nach dem Kochen mit Natriumäthylat die alkoholische Lösung des Natriumsalzes des Esters mit etwa der gleichen Menge Wasser versetzt und nach Abdestillieren des Alkohols schwach kongo sauer stellt.
