Verfahren zur Darstellung eines 2,4-1)loxo-3,3-dialkyl-tetrahydropyridin- 6-ear bonsäureester s. Nach dem Verfahren der schweizerischen Patentschrift Nr. 187826 und Zusätze kann man die durch Kondensation von Dialkyl- acetessigestern mit Ameisensäureestern und nachherige Einwirkung von Ammoniak gewonnenen Aminomethylendialkylacetessig- eater durch alkalische Kondensationsmittel leicht in Pyridinabkömmlinge überführen.
Es wurde nun gefunden, dass man zu 2,4-Dioxo-3,3-dialkyl-tetrahydropyridin-6-car- bonsäureestern bezw. den freien Säuren ge langen kann, indem man 2,2-Dialkyl-3,5- dioxo-hexan - disäure - (1,6) - dialkylester mit Ammoniak umsetzt und auf den erhaltenen 2,2 - Dialkyl-3-oxy-5-aminohexen-(4)-disäure- (1,6)-dialkylester, Alkali-Alkoholate einwirken lässt und die zunächst entstehenden Ester gegebenenfalls verseift.
Vertreter der durch Einwirkung von Oxaleater auf Dialkylaceteasigester erhält- liehen Ausgangsstoffe, dass heisst der 2,2 Dialkyl-3,5-dioxo-hexan-disäure-(1,6)-dialkyl- ester, sind bis jetzt nicht bekannt. Sie bilden farblose, im luftverdünnten Raum un- zersetzt, aber ziemlich hoch siedende Öle. Bei der Einwirkung von Ammoniak bleiben beide Carboxylestergruppen unverändert; es entstehen hierbei zunächst die Ammonium.
salze der Dioxoester, die man, wenigstens in der Salzform, als 2,2-Dialkyl-3-oxo-hexen- (4)-ol-(5)-disäure-(1,6)-dialkylester ansprechen muss. Durch gelindes Erwärmen werden diese Ammoniumsalze unter Wasserabspaltung in 2, 2-Dialkyl- 3 -oxo- 5-amitro-hexen- (4)-d i säure- (1,6)-diester umgewandelt. Die Aminohexen- säureester sind ebenfalls hoch, aber im luft verdünnten Raum unzersetzt siedende Öle.
Lässt man auf sie Alkalialkoholate einwirken, so entstehen in glatter Umsetzung, 2,4-Dioxo- 3,3- dialkyl-teträhydropyxidin- 6 - carbonsäure- ester. Die vorhin erwähnten Umsetzungen lassen sich wie folgt darstellen;

Die neuen 2,4-Dioxo-3,3-dialkyltetz-ahy- dropyr,idiri-6-cai-bonsäureestei, sind niedrig schmelzende, destillierbare Verbindungen, die bei Einwirkung von Alkalien leicht verseift werden.
Die entsprechenden freien Säuren kristallisieren gut und bilden neutral reagie rende, in Wasser leicht löslielie Alkalisalze. Sie stellen Zwischenprodukte für die Her stellung von Arzneimitteln dar. So kann man ausgehend von den freien Säuren durch Decarboxylierung zu den aus der schweize rischen Patentschrift Nr. 187826 und Zusätze bekannten Schlafmitteln der Pyridinreilie gelangen.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung des 2,4-Dioxo- 3,3-diäthy 1-tetraliydropyridiri-6-carbonsäure- methylesters. Erfindungsgemäss setzt man 2,2-Diäthyl - 3,5-dioxo-hexari-disäur@e-(1,6)-di- äthylester mit Ammoniak um und lässt auf den erhaltenen 2,2-Diäthyl-3-oxo-5-amirio- hexen-(4)-disäui#e-(1,6)-diäthylestei-, Alkali methylat einwirken. Die auf diese Weise gebildete treue Verbindung besteht aus gelb lichen Kristallen, welche bei<B>80-810</B> C schmelzen.
Sie ist in Wasser ziemlich schwer, in organischer) Lösungsmitteln dagegen leicht löslich; durch Einwirkung vor) Alkalien tritt leicht Verseifung ein. Beispiel: 24 Gewichtsteile pulverisiertes Natrium werden in 450 Gewichtsteilen trockenem Toluol unter Rülii-en verteilt. 31arr lässt etwa Vlo einer Mischung von<B>186</B> Gewichtsteilen Di,itli3rlacetessigester und 146 Gewichtsteilen Ox:tls;
iriredüitliylestei-, das heilät 35 Gewichts teile, zulaufen lind erwärmt nun das Ganze unter fortgesetztem Rühren. Zwischen 50 und<B>600</B> beginnt die Umsetzung. Die äussere Wärmezufuhr wird sofort unterbrochen und die Temperatur dureli schwaches Kühlen immer auf ungefähr derselben Höhe gehalten.
Hat die Hauptumsetzung nachgelassen, so lässt man die weiteren 9Jio des Gemisches so zutropfen, dass die Temperatur ohne äussere Wärmezufuhr immer zwischen 50 und 55(1 bleibt. Wenn alles zugetropft ist, das heisst nach etwa einer Stunde, wird noch 30 Minu ten bei 50 verrübrt. Das Natrium ist nun vollstündig verschwunden. Die Lösung ist braungelb und klar.
Unter weiterem Rühren wird darauf abgekühlt, und darin werden 100 Gewichtsteile Eis und 340 Raumteile 3 ri-Schwefelsäure zugefügt. Nach rund ein- halbstündigein Rühren ist das Ganze fast farblos geworden. Die Toluolschicbt enthält nun den freien 2,2-Dititliyl-3,5-dioxo-hexandi- säure-(1,6)-diäthylestei-. Dieser wird nach dem Verjagen des Toluols durch Destillation ge reinigt. Er siedet unter 14 mm bei 17ä-176 .
Zur Weiterverarbeitung werden 286 Ge wichtsteile des so erhaltenen Esters mit 100 Gewichtsteilen Methanol verdünnt und unter guter Eiskühlung und Rühren zu 200 Gewichtsteilen 15%igem methylalkobolischem Ammoniak nach und nach so zugefügt, dass die Temperatur nie über 200 steigt.
Das erhaltene Ammoniumsalz des 2,2-Diäthyl- 3,5 - dioxo - heran - disäu re - (1,6) - diäthylesters wird unter vermindertem Druck bei etwa 400 von Methanol und überschüssigem Am moniak befreit und das feste Ammoniumsalz im Wasserbad unter vermindertem Druck langsam bis zum Sieden des Wassers erwärmt, wobei unter Wasserabspaltung das Ammo- niumsalz in den 2,2-Diäthyl-3-oxo-5-amino- hexen-(4)-disäure-(1,6)-diäthylester übergeht.
Die vollständige Wasserabspaltung ist etwa nach einer Stunde erreicht. Das nun zurück bleibende dünnflüssige Ö1 wird im Vakuum destilliert. Der 2,2-Diäthyl-3-oxo-5-amino- hexen-(4)-disäure-(1,6)-diäthylester siedet unter 14 mm bei 181-1830.
Zur Durchführung des Ringschlusses werden 285 Gewichtsteile dieses Esters mit einer Natriummethylatlösung, die aus 200 Gewichtsteilen Methanol und 23 Gewichts teilen Natrium bereitet worden ist, eine halbe Stunde am Rückfluss gekocht. Die ab gekühlte Lösung wird mit rund 90 Gewichts teilen konzentrierter Salzsäure schwach kon gosauer gestellt. Nun wird das Methanol verjagt. Der zurückbleibende Kristallbrei wird in Äther aufgenommen und die Äther- lösung mit Wasser gewaschen.
Nach dem Abdampfen des Äthers und Umkristallisieren des Rückstandes aus Dibutyläther erhält man den unter Ringschluss und gleichzeitiger Umesterung entstandenen 2,4-Dioxo-3,3-di- äthyltetrahydropy ridin- 6-carbonsäuremethyl- ester vom Schmelzpunkt 80-81 in schwach gelben Kristallen.
Löst man diesen Ester in der vierfachen Menge 3 n-Natronlauge, so tritt bald Er wärmung unter Verseifung des Esters ein. Säuert man nach 20-30 Minuten an, so fällt die 2,4-Dioxo-3,3-diäthyl-tetrahydropy- ridin-6-carbonsäure in derben Kristallen aus. Die Säure kristallisiert mit Kristallwasser und schmilzt wasserfrei bei 145-1460.

Representatives of the starting materials obtained by the action of oxaleater on dialkyl aceteate ester, that is to say the 2,2 dialkyl-3,5-dioxo-hexane-diacid (1,6) -dialkyl ester, are not yet known. The new 2,4-dioxo-3,3-dialkyltetz-ahy- dropyr, idiri-6-cai-bonsäureestei, are low-melting, distillable compounds that are easily saponified when exposed to alkalis.
The corresponding free acids crystallize well and form neutrally reactive alkali salts that are easily soluble in water. They are intermediate products for the manufacture of pharmaceuticals. So you can proceed from the free acids by decarboxylation to the sleeping pills known from Swiss patent No. 187826 and additives of the pyridine line.
The subject of the present patent is a process for the preparation of the 2,4-dioxo-3,3-diethy 1-tetraliydropyridiri-6-carboxylic acid methyl ester. According to the invention, 2,2-diethyl-3,5-dioxo-hexari-diacid @ e- (1,6) -di-ethyl ester is reacted with ammonia and the resulting 2,2-diethyl-3-oxo-5- amiriohexen- (4) -disäui # e- (1,6) -diäthylestei-, alkali methylate act. For further processing, 286 parts by weight of the ester obtained in this way are diluted with 100 parts by weight of methanol and gradually added to 200 parts by weight of 15% methyl alcoholic ammonia with good ice cooling and stirring so that the temperature never rises above 200.
The resulting ammonium salt of 2,2-diethyl- 3,5 - dioxo - heran - diacid - (1.6) - diethyl ester is freed from methanol and excess ammonia under reduced pressure at about 400 and the solid ammonium salt in a water bath under reduced pressure The pressure is slowly heated to the boiling point of the water, the ammonium salt being converted into the 2,2-diethyl-3-oxo-5-aminohexen (4) diacid (1,6) diethyl ester with elimination of water.
Complete elimination of water is achieved after about an hour. The now remaining thin liquid oil is distilled in a vacuum. The 2,2-diethyl-3-oxo-5-amino-hexen (4) -diacid- (1,6) -diethyl ester boils below 14 mm at 181-1830.
To carry out the ring closure, 285 parts by weight of this ester are refluxed for half an hour with a sodium methylate solution which has been prepared from 200 parts by weight of methanol and 23 parts by weight of sodium. The cooled solution is made slightly con gosauer with around 90 parts by weight of concentrated hydrochloric acid. Now the methanol is chased away. The remaining crystal pulp is taken up in ether and the ether solution is washed with water.
After evaporation of the ether and recrystallization of the residue from dibutyl ether, the 2,4-dioxo-3,3-diethyltetrahydropyridine-6-carboxylic acid methyl ester of melting point 80-81, formed with ring closure and simultaneous transesterification, is obtained in pale yellow crystals.
If this ester is dissolved in four times the amount of 3N sodium hydroxide solution, warming soon occurs with saponification of the ester. If it is acidified after 20-30 minutes, the 2,4-dioxo-3,3-diethyl-tetrahydropyridine-6-carboxylic acid precipitates in coarse crystals. The acid crystallizes with crystal water and melts anhydrous at 145-1460.
