Abstract
L'invention concerne un système binaire d'expression virale régulée dans des plantes. Il est composé de deux éléments intégrés dans les chromosomes. Un élément est un proréplicon contenant des séquences virales à action cis nécessaires pour la réplication dont une séquence contient un gène ciblé. L'autre élément est un gène chimère de réplication à action trans comprenant un promoteur relié à la région codante pour la réplication de la protéine virale. Le proréplicon est exempt du gène de réplication essentiel pour la réplication du réplicon et, de ce fait, ne peut pas subir de réplication épisomique autonome. Cependant, l'expression régulée de la protéine de réplication à action trans dans des cellules de la plante contenant également le proréplicon déclenchera la libération d'un réplicon libre depuis le proréplicon intégré, ce qui produira sa réplication épisomique in trans, ainsi que l'expression du gène ciblé, s'il est présent, par l'intermédiaire de l'amplification génique. Ce système d'expression est utile à la fois pour la production de protéines étrangères et la mise en sommeil de gènes endogènes et de transgènes dans le tissu de la plante. L'expression spécifique au tissu est commandée par le choix du promoteur régulant la transcription du gène de réplication à action trans.
