Abstract

Patente de Invenção: <B>''MOLéCULAS DE áCIDO NUCLéICO CORRELACIONADAS COM O FENóTIPO RHESUS D FRACO''<D>. A presente invenção refere-se a novas moléculas de ácido nucléico codificando um antígeno Rhesus D contribuindo para o fenótipo D fraco que são caracterizadas por uma ou uma combinação de mutações de sentido errado ou por uma conversão de gene envolvendo exons 6 a 9 dos genes RHD e RHCE. A presente invenção ainda refere-se a vetores compreendendo as moléculas de ácido nucléico da invenção, a hospedeiros transformados com referidos vetores, a proteínas codificadas por referidas moléculas de ácido nucléico e a processo para produzir estes polipeptídeos. O fato que as mutações de sentido errado mudam e a conversão referida acima podem estar diretamente correlacionados com o fenótipo D fraco que tem um impacto significante sobre o teste de rotina de amostras de sangue. Por exemplo, oligonucleotídeos e anticorpos podem agora ser projetados que geralmente permite a detecção de fenótipos D fraco em uma amostra. Estes oligonucleotídeos, anticorpos assim como uma variedade de processos diagnósticos estão todos no escopo da presente invenção. Os antígenos RHD codificados pelas novas moléculas de ácido nucléico podem ser usados para a caracterização, padronização e controle de qualidade de anti-soro anti-D monoclonal e policlonal. Finalmente, a invenção refere-se a um kit utilizável para testar a presença de fenótipos D fraco.
