Abstract
"ENZIMA DE XILANASE, XILANASE MODIFICADA E PROCESSOS PARA MELHORAR A CAPACIDADE DE BRANQUEAMENTO DE PASTA DE MADEIRA E PARA MODIFICAR UMA ENZIMA DE XILANASE". Produção de uma enzima de xilanase de desempenho superior no branqueamento da pasta. Mais especificamente, uma xilanase modificada da Família 11 que mostra melhoradas termofilicidade, alcalofilicidade e termoestabilidade quando comparada à xilanase natural. As xilanases modificadas contêm qualquer um de três tipos de modificações: (1) mudar aminoácidos 10, 27 e 29 da xilanase de Trichoderma reesei II ou os correspondentes aminoácidos de outra xilanase da Família 11, em que estes aminoácidos são mudados para histidina, metionina e leucina, respectivamente; (2) substituição de aminoácidos na região N-terminal com aminoácidos de outra enzima de xilanase. Em uma forma de realização preferida, a substituição da xilanase de Bacillus circulans ou Trichoderma reesei natural com uma sequência curta de aminoácidos da xilanase de Thermomonospora fusca deu xilanases quiméricas com termofilicidade e alcalofilicidade mais altas; (3) uma extensão a montante do N-término de até 10 aminoácidos. Em uma forma de realização preferida, a extensão do N-término da xilanase com o tripeptídeo glicina-arginina-arginina melhorou seu desempenho.
