Abstract
resumo patente de invenção: "padrão quantitativo para espectrometria de massa de proteínas". a presente invenção refere-se a um método de determinação da quantidade absoluta de um polipeptídeo alvo em uma amostra, o referido método compreendendo as seguintes etapas: (a) adicionar (aa) um polipeptídeo de fusão à referida amostra, o referido polipeptídeo de fusão compreendendo (i) pelo menos um sequência de marcação e (ii) uma subsequência do polipeptídeo alvo; e (ab) uma quantidade absoluta conhecida de um polipeptídeo de marcação que compreende ou consiste na referida sequência de marcação de acordo com (aa) à referida amostra, em que o referido polipeptídeo de fusão tem, por um lado, massa alterada quando comparado com o referido polipeptídeo alvo e o referido polipeptídeo de marcação, por outro lado, por exemplo, o referido polipeptídeo de fusão por um lado e o referido polipeptídeo alvo e o referido polipeptídeo de marcação por outro lado, são marcados isotopicamente de maneira diferente; (b) realizar uma digestão proteolítica da mistura obtida na etapa (a); (c) sujeitar o resultado da digestão proteolítica da etapa (b), opcionalmente após cromatografia, à análise por espectrometria de massa; e (d) determinar a quantidade absoluta do referido polipeptídeo alvo a partir (i) das intensidades de pico no espectro de massa adquirido na etapa (c) do referido polipeptídeo de fusão, do referido polipeptídeo de marcação e do referido polipeptídeo alvo e (ii) da referida quantidade absoluta conhecida do referido polipeptídeo de marcação.
