Abstract
Patente de invenção: "PROMOTOR QUE PODE SER INDUZIDO PELO CALOR". A invenção refere-se a moléculas de ácido nucléico que compreendem um promotor que pode ser induzido pelo calor e a vetores de expressão e a células hospedeiras que contêm pelo menos uma molécula de ácido nucléico de acordo com a invenção. A presente invenção refere-se ainda a kits e a processos para a produção de uma ou mais proteínas utilizando as moléculas de ácido nucléico de acordo com a invenção e a vários usos das mesmas. O objetivo da invenção é fornecer um promotor cuja característica induzível por calor seja a mais seletiva possível, em particular um promotor que seja ativo em leveduras e que seja adequado para a expressão de proteínas a altas temperaturas. Este objetivo é alcançado por uma molécula de ácido nucléico que compreende um promotor que pode ser induzido pelo calor e que é selecionado dos ácidos nucléicos a seguir: (a) um ácido nucléico cuja seq³ência compreende a seq³ência promotora de um gene de Hansenula polymorpha que codifica uma proteína com atividade trealose-6-fosfato sintase; (b) um ácido nucléico com a seq³ência indicada na SEQ ID N<0>:1;(c) um ácido nucléico com uma seq³ência que exibe pelo menos 40% de identidade em relação a um comprimento de 300 pb com uma das seq³ências indicadas em (a) ou (b); (d) um ácido nucléico que se hibridiza à fita complementar de um dos ácidos nucléicos indicados em (a), (b) ou (c); (e) um derivado de um dos ácidos nucléicos indicados em (a), (b) ou (c) obtido pela substituição, adição e/ou deleção de um ou mais nucleotídeos; (f) um fragmento de um dos ácidos nucléicos indicados em (a) até (e) que mantém a função do promotor que pode ser induzido por calor; (g) uma combinação de vários dos ácidos nucléicos indicados em (a) até (f), em que as seq³ências dos ácidos nucléicos podem ser diferentes ou iguais; ou através de uma molécula de ácido nucléico cuja seq³ência é complementar à seq³ência de um dos ácidos nucléicos indicados em (a) até (g).
