Abstract

"OLIGONUCLEOTìDEOS PARA AMPLIAçãO E DETECçãO DE GENES DE HEMOCROMATOSE" A presente invenção providencia oligonucleotídeos e métodos para ampliação, detecção e identificação de variações de seq³ência associadas com hemocromatose. Descobriu-se que, a eficiência reduzida da extensão do primer por DNA polimerases, quando a extremidade 3' de um primer não hibridiza perfeitamente com o alvo, pode ser adaptada para uso como um meio em distinguir ou identificar o nucleotídeo no alvo, que está no sítio onde ocorre a combinação inadequada de diagnóstico entre o primer detector e o alvo. A eficiência da extensão do primer detector é vista como uma indicação da presença e/ou identidade de variação de seq³ência no alvo. Os métodos inventivos fazem uso de primers de ampliação específicos para gene de hemocromatose (HFE) e combinações inadequadas do nucleotídeo na ou próximo da extremidade 3' de um primer detector para ampliar o gene de HFE e discriminar entre alelos tipo selvagem e polimorfismos de nucleotídeo simples que podem ocorrer no exon 2 e exon 4 do gene de HFE.
