La présente invention a pour objet un nouveau procédé de préparation de 5,6,7,8-tétrahydronaphtol-2.

est une substance de départ de grande valeur pour la préparation de différents composés organiques, en particulier à action thérapeutique.
On connaît déjà plusieurs procédés de prépa-

ration de 5,6,7,8-tétrahydronaphtol-2 . Selon/[Annalen, 426,
119, (1922)] , on obtient ce composé sous la forme d'un produit brut difficile à purifier par fusion d'acide 5,6,7,8-tétrahydronaphtalènesulfonique-2 avec de l'hydroxyde de potassium et par ébullition ultérieure avec de l'acide chlorhydrique . Selon le brevet allemand 508.494, on peut préparer ce composé par
la réaction de 6-acétoxy-tétraline avec une solution aqueuse d'hydroxyde de sodium. Selon le procédé décrit dans le brevet français 664.408, on hydrogène à 100[deg.]C , sous une pression de
30 à 80 atmosphères , de l'acétate de p-naphtol en présence d'un catalyseur à base de nickel, et on traite l'acétate de 5,6,7,8-tétrahydronaphto - 2 . avec. du carbonate de
sodium. Cependant tous les procédés décrits jusqu'à présent , qui ne partent pas du p-naphtol , présentent l'inconvénient commun qu'ils sont d'un faible rendement.
Lors de l'hydrogénation catalytique également connue de p-naphtol, il se forme , à côté du 5,6,7,8-tétrahydronaphtol-2 différents autres dérivés hydrogénés de naphtol ou
de naphtalène , tels que du décahydronaphtalène (décaline), du décahydronaphtol-2 et du l,2,3,4-tétrahydronaphtol-2. Brown, Durand et Marvel [J.Am.Chem.Soc. 58 , 1594-1596 (1936)] ont préparé , par hydrogénation de 0-naphtol en présence d'un catalyseur à base de platine , un mélange de 5,6,7,8-tétrahydro-

communiqué le rapport en % de ces produits. Dans les mêmes conditions de réaction , HUckel [Annalen 451,117 (1925� a obtenu,comme produits principaux,du décahydronaphtalène et
du décahydronaphtol-2 , le 5,6,7,8-tétrahydronaphtol-2 n'étant obtenu que sous forme de produit secondaire. Sous une pression de 110 atmosphères,Baker et Schnetz ([J.Am.Chem.Soc.69;1251(1947) ont préparé , toujours dans les mêmes conditions de réaction, du décahydronaphtol-2, et on présume la formation de 1,2,3,4tétrahydronaphtol-2 comme produit intermédiaire.
HUckel [Annalen 441, 18 (1925)] s'est en premier lieu intéressé à l'hydrogénation de p-naphtol en présence d'un catalyseur à base de nickel; à 160[deg.]C et sous une pression élevée , il a obtenu un mélange de décaline, de tétraline , de 1,2,3,4-tétrahydronaphtol-2 et de 5,6,7,8-tétrahydronaphtol-2 , mais il n'a pas indiqué les proportions

hydrogéné du /3-naphtol en présence d'un catalyseur à base de nickel sous une pression de 230 atmosphères et à différentes valeurs de pH, et il a fait l'observation fondamentale que dans des milieux neutres ou acides il se forme,comme produit principal du 5,6,7,8-tétrahydronaphtol-2 , tandis que dans des milieux alcalins on obtient,comme produit principal,du 1,2,3,4tétrahydronaphtol-2 . Adkins et Kosek [J.Am.Chem.Soc.70,412
(1948)] ont hydrogéné du p-naphtol sous une pression de 100
à 200 atmosphères , en présence d'un catalyseur à base de nickel , et ils ont obtenus le 5,6,7,8-tétrahydronaphtol-2 avec un rendement de 80%; Odaira et Tsutsumi [Chem.Abstr. 53 ,
18920c-f (1959)] ont, à 100 atmosphères , obtenu d'une manière analogue le même produit avec un rendement de 67,6% et Matoyama

de 46%.
Suivant le brevet aux Etats-Unis d'Amérique
2.526.859 , on hydrogène le p-naphtcl sous une pression de
110 atmosphères,en présence d'un catalyseur à base de palladium
(5%) /charbon actif ; comme produit-principal on obtient du 3,4-dihydro-2-cétonaphtalène à côté d'un peu de 1,2,3,4-tétrahydronaphtol comme produit secondaire.
Dauben, Kusick et MUller [J.Am.Chem.Soc. 70,
4179-4131 (1948)] ont hydrogéné du 0-naphtol en présence d'un catalyseur à base de chromate de cuivre , sous une pression supérieure à 200 atmosphères; à côté de 7 à 8% de 5,6,7,8-tétrahydronaphtol-2 ils ont obtenu de manière prépondérante , comme produit principal, du 1,2,3,4-tétrahydronaphtol-2.
Les procédés connus partant de 9-naphtol ont donc l'inconvénient commun qu'on doit hydrogéner à des pressions supérieures à 100-200 atmosphères , pour obtenir le 5,6,7,8-tétrahydronaphtol-2 souhaité à des rendements acceptables.
La présente invention a pour objet de mettre
en oeuvre un procédé de préparation de 5,6,7,8-tétrahydronachtol2 , suivant lequel le produit souhaité peut être obtenu dans
des conditions industrielles et par l'application de pressions inférieures de l'ordre de quelques atmosphères , avec de bons rendements qui dépassent ceux que l'on a pu atteindre jusqu'à
présent. ,

alcool aliphatique ayant de 1 à 3 atomes de carbone , avec addition d'acides, en présence d'un catalyseur à base de palladium éventuellement traité avec un promoteur. Dans ces conditions , on obtient le 5,6,7,8-tétrahydronaphtol-2 avec des rendements de 80 à 85% , tandis qu'il se forme comme produits secondaires environ 3 à 5% de décahydronaphtol-2 et 10 et 15% de 1,2,3,4-tétrahydronaphtol-2.
En examinant les facteurs qui influencent la vitesse de la fixation d'hydrogène , comme la température, la
<EMI ID=8.1> lyseur , on a fait les observations suivantes:
si on augmente la température de 60 à 100[deg.]C au cours de l'hydrogénation sous une pression de 10 atmosphères , la vitesse de réaction s'élève à trois fois la valeur
de départ. Par l'augmentation de la quantité du catalyseur , la vitesse de réaction ne peut être augmentée que jusqu'à une valeur limite qui dépend également du volume du système. Si
on prend comme unité la vitesse de fixation d'hydrogène mesurée en présence d'un gramme de palladium (sous la forme d'un catalyseur à base de palladium précipité sur du charbon actif ) pour un volume total de 350 ml , la vitesse de réaction mesurée en présence de 2 g, 4g , 6 g , et respectivement 8 g
de palladium et de 14,30 , 51 et respectivement 53 unités. On

à base de palladium contenant des promoteurs à base de platine, de rhodium ou de ruthénium.
L'influence des différentes additions d'acide sur la vitesse de fixation de l'hydrogène est illustrée par les données suivantes:

Ainsi qu'il ressort des données ci-dessus , l'influence favorable souhaitée de l'addition d'acide sur la vitesse de réaction de l'hydrogénation r-�ut être plus ou moins atteinte avec les acides les plus différents , la quantité
de l'addition d'acide pouvant également jouer un rôle important; d'une manière générale , on a trouvé qu'on peut atteindre les résultats les plus favorables si on ajoute suffisamment d'acide pour que le pH-du mélange réactionnel soit abaissé à une valeur de 1 à 4 ,en particulier d'environ 3 à 4.
L'invention consiste donc en un procédé de préparation de 5,6,7,8-tétrahydronaphtol-2 par l'hydrogénation catalytique de �-naphtol , ce procédé étant caractérisé
en ce qu'on effectue l'hydrogénation dans un alcool ayant de 1 à 3 atomes de carbone , en milieu acide , en présence d'un catalyseur à base de palladium contenant éventuellement des promoteurs,- sous une pression de 1,2 à 20 atmosphères, de préférence de 4 à 6 atmosphères , et à des températures de l'ordre de 40 à 100[deg.]C , de préférence de 70 à 90[deg.]C.
Il est avantageux de mettre en oeuvre le procédé suivant l'invention de la manière suivante: on introduit du /3-naphtol dans un autoclave que l'on peut chauffer et refroidir , on ajoute un alcool ayant de 1 à 3 atomes de carbone , de préférence du méthanol , cet alcool étant anhydre ou contenant au maximum 10% d'eau, on ajoute ensuite un catalyseur à base de palladium, avantageusement du charbon actif palladié, contenant éventuellement également un promoteur , par exemple du platine, du rhodium ou du ruthénium, et on acidifie le mélange avec un acide organique ou inorganique , par exemple avec de l'acide chlorhydrique , sulfurique , formique, acétique, oxalique ou citrique, à une valeur de pH de 1 à 4, de préférence de 3 à 4.
On balaye alors l'autoclave avec de l'azote , et on hydrogène sous une pression de 1,5 à 20 atmosphères , de préférence à environ 5 atmosphères.Au début del'hydrogénation , le mélange réactionnel est chauffé à une température de 40 à 100[deg.]C, de préférence d'environ 80[deg.]C. La réaction d'hydrogénation se déclenche à 40[deg.]C, et elle est achevée approximativement en

l'acide présent et de la qualité du catalyseur. Comme la réaction se déroule de manière exothermique (18-20 kgcal/mole de

mélange réactionnel pendant la réaction. La réaction achevée , on élimine le catalyseur par filtration , on peut éventuellement régénérer le catalyseur , et on élimine le solvant du filtrat par distillation.
On ajoute ensuite au résidu de distillation une base , avantageusement une solution aqueuse à 6% d'hydroxyde de sodium, jusqu'à ce que l'atome d'hydrogène du groupe hydroxyle aromatique soit remplacé quantitativement par du

Le procédé suivant l'invention est décrit d'une manière plus détaillée à l'aide des exemples ci-après, sans être pour autant limité par ces derniers.
Exemple 1
Dans un autoclave à agitation que l'on peut chauffer et qui est d'une capacité de 350 ml , on introduit
30 g de /3-naphtol , 200 ml de méthanol , 6 g de catalyseur
à base de charbon actif palladié (10% ) et 2 ml d'acide acétique glacial. Après la fermeture , on balaye l'autoclave avec de l'azote, puis on met le contenu de l'autoclave sous une pression d'hydrogène de 5 atmosphère;. Sous cette pression , le

mélange réactionnel ne contient plus de p-naphtol qui n'ait pas réagi; le produit réactionnel est constitué de 75% de 5,6,7, 8-tétranaphtol-2, de 3% de décahydronaphtol -2 et de 22% de l,2,3,4-tétrahydronaphtol-2 . Après élimination du catalyseur par filtration , on sépare le méthanol par distillation , on ajoute au résidu 13 g d'hydroxyde de sodium sous la forme d'une solution aqueuse à 6% (216 ml), et on extrait ensuite du mélange obtenu (pH : 10) le décahydronaphtol-2 et le 1,2,3,4-tétrahydronaphtol-2 avec à deux reprises 50 ml de chloroforme. Le pH de la phase aqueuse est ensuite ajusté à une valeur de 2
à l'aide d'acide sulfurique concentré. On extrait le 5,6,7,8tétrahydronaphtol-2 séparé sous forme huileuse avec à deux reprises 25 ml de chloroforme; on isole le produit à partir de l'extrait par distillation du chloroforme. Production: 21,1 g
(rendement de 72,0% de la théorie); point de fusion: 60[deg.]C ; point d'ébullition : 148[deg.]C/10 mm de Hg.
Exemple 2
Dans l'autoclave à agitation décrit dans l'exem- <EMI ID=21.1>
5 g de catalyseur à base de palladium (10% ) /charbon actif, activé avec du rhodium , et 2 ml d'acide acétique glacial.
On effectue l'hydrogénation et l'isolement du produit de la manière décrite dans l'exemple 1. On obtient 2<2>g de 5,6,7,8-tétrahydronaphtol-2 (rendement 71,4% de la théorie); point de

5 atmosphères est achevée en 4 à 8 heures. L'isolement du produit est effectué de la manière déjà décrite. On obtient 86,5 kg de 5,6,7,8-tétrahydronaphtol-2 (rendement de 84,0% de la théorie); point de fusion : 60[deg.]C ; point d'ébullition : 148[deg.]C /10 mm de Hg.
Exemple 5
Dans un récipient d'hydrogénation que l'on peut chauffer et qui est d'une capacité de 250 ml , on introduit 15 g de /3-naphtol, 100 ml de métha�ol, 3 g de catalyseur à base de

concentré.On ferme l'appareil , on le balaye avec de l'azote et on effectue l'hydrogénation à 45[deg.]C et sous une pression d'hydrogène de 1,5 atmosphère. Après une période de réaction de
44 heures, le mélange réactionnel contient 97% de 5,6,7,8-tétrahydronaphtol-2 et 3% de j3-naphtol qui n'a pas réagi. Le produit est isolé de la manière courante . Production: 14,3 g
(rendement de 93,0% de la théorie): point de fusion: 60[deg.]C; point d'ébullition : 148[deg.]C/10 mm de Hg.
Exemple 6
Dans un'autoclave à agitation que l'on peut chauffer et qui est d'une capacité de 350 ml, on introduit
30 g de 0-naphtol, 200 ml de méthanol, 6 g d'un catalyseur
à base de palladium (10%)/charbon actif , et 2 ml d'acide acétique glacial . Après la fermeture , on balaye l'autoclave avec de l'azote et on l'amène sous une pression d'hydrogène de
20 à 30 atmosphères. Ensuite, on commence le secouage et simultanément on élève la température de l'autoclave à 80[deg.]C . L'hydrogénation dure 1,25 heure sous 20 atmosphères. L'isolement du produit s'effectue comme dans l'exemple 1.
On obtient 23,1 g de 5,6,7,8-tétrahydronaphtol-

Exemple 7
Dans un autoclave à agitation que l'on peut chauffer et qui est d'une capacité de 350 ml , on introduit 30 g de p-naphtol, 200 ml de méthanol, 6 g de catalyseur à base de palladium (10%)/charbon actif et 2 ml d'acide acétique glacial.
Après la fermeture,on balaye l'autoclave avec de l'azote et on 1 amène sous une pression d'hydrogène de 5 atmosphères. L'hydrogénation s'effectue à 50[deg.]C pendant 10 heures. Ensuite , on

Exemple 8
Dans l'autoclave à agitation décrit dans l'exemple 1, on introduit 144 g de [pound]-naphtol, 200 ml de méthanol,
7,2 g de catalyseur à base de palladium (5%) /charbon actif et
12 ml d'acide acétique glacial . L'hydrogénation et l'isolement du produit sont effectués de la manière décrite dans l'exemple 1, mais en une période de réaction de 16 heures. On obtient de cette manière 116,2 g de 5,6,7,8-tétrahydronaphtol-
2 .(rendement de 78,5% de la théorie); point de fusion: 60[deg.]C; point d'ébullition: 148[deg.]C/10 mm de Hg.
Exemple 9
Dans l'autoclave industriel décrit dans l'exemple 4, on introduit 200 kg de /3-naphtol, 500 litres de méthanol, 10 kg de catalyseur à base de palladium (10%)/charbon actif et 26 kg d'acide acétique glacial. On effectue l'hydrogénation à 80[deg.]C sous une pression d'hydrogène de 5 atmosphères, cette hydrogénation étant achevée en 6 à 10 heures. On isole
le produit suivant l'exemple 1. On obtient 166,2 kg de 5,6,7,8-tétrahydronaphtol-2 (rendement de 80,9% de la théorie); point de fusion : 60[deg.]C ; point d'ébullition : 148[deg.]C/10 mm de Hg.
Il doit être entendu que la présente invention n'est en aucune façon limitée aux modes de réalisation décrits ci-dessus et que bien des modifications peuvent y être apportées sans sortir du cadre du présent brevet.

The known processes starting from 9-naphthol therefore have the common drawback that it is necessary to hydrogenate at pressures above 100-200 atmospheres, in order to obtain the desired 5,6,7,8-tetrahydronaphthol-2 in acceptable yields.
The object of the present invention is to put
carrying out a process for the preparation of 5,6,7,8-tetrahydronachtol2, according to which the desired product can be obtained in
industrial conditions and by the application of lower pressures of the order of a few atmospheres, with good yields which exceed those which have been achieved up to
present. ,

It is advantageous to carry out the process according to the invention in the following way: / 3-naphthol is introduced into an autoclave which can be heated and cooled, an alcohol having 1 to 3 carbon atoms is added, preferably methanol, this alcohol being anhydrous or containing at most 10% water, a palladium-based catalyst is then added, advantageously palladium-based activated carbon, optionally also containing a promoter, for example platinum, rhodium or ruthenium, and the mixture is acidified with an organic or inorganic acid, for example with hydrochloric, sulfuric, formic, acetic, oxalic or citric acid, to a pH value of 1 to 4, preferably 3 to 4.
