PROCEDE DE FABRICATION D'ESTERS DE L'ACIDE CARBAMIQUE AVEC DES ACETYLENE-ALCOOLS
TERTIAIRES ET SECONDAIRES.
Jusqu'à présent, on ne connaît pas d'esters de l'acide carbamique avec des acétylène-alcools tertiaires et secondaires de la formule générale

dans laquelle R1, R2. R3 et R4 représentent'un atome d'hydrogène ou un reste d'hydrocarbure, et R un reste d'hydrocarbure.,
Or, selon l'invention, on a constaté que des composés de ce type peuvent être obtenus par les méthodes usuelles de préparation d'esters d'acide carbamique à partir d'alcools. Ainsi, par exemple, on peut partir d'halogénures de carbamyle substitués sur l'azote ou non substitués et les transformer en les esters désirés, par réaction avec les acétylène-carbinols tertiaires ou secondaires correspondants.
On peut cependant partir aussi des esters d'acide halogénoformique des acétylène-carbinols tertiaires ou secondaires, qui peuvent être obtenus par exemple par réaction de dihalogénures de carbonyle, tels que le phosgène, avec les acétylène-carbinols tertiaires ou seconr daires, et faire agir de l'ammoniaque ou des amines sur les esters d'acide halogénoformique.
Etant donné la puissance réactionnelle connue du groupe acétylène, qui laissait prévoir des réactions secondaires indésirables, par exemple des cyclisations, ou vu la sensibilité particulière connue des esters d'acide halogénoformique tertiaires (cf. le brevet allemand 254.471). il était dans une certaine mesure surprenant que les réactions selon l'invention s'accomplissent avec des rendements absolument utilisables techniquement.
Les produits obtenus par le procédé possèdent des propriétés thérapeutiques intéressantes, par exemple certains d'entre eux présentent un effet hypnotique prononcé avec simultanément de minimes phénomènes secondaires.

EXEMPLE 1.
----------
A 34 g = 430 millimoles de chlorure de carbamyle dans 250 cm3 d'é- ther absolu, on ajoute, en refroidissant à la glace, 42 g = 430 m moles de 3-méthylpentine-(1)-ol-(3). Après avoir laissé reposer pendant une nuit, on lave à l'eau et l'on achève le travail de la manière usuelle. La distillation dans le vide donne un distillat incolore, d'un p. d'éb.12: 120-121 . Cris- taux dans l'hexane, f. : 56-58 ., Le produit de la réaction forme un sel d'ar- gent, peut être transformé en ester d'acide carbamique du méthyldiéthylcarbi- nol par hydrogénation catalytique et se révèle ainsi être un ester d'acide carbamique du 3-métbyl-pentine-(1)-ol-(3).
EXEMPLE 2.
---------- On ajoute une solution de 51 cm3 = 750 millimoles de phosgène dals s 150 cm3 d'éther absolu, en refroidissant à la glace et au::-..-chlorure de sodium, à 67 cm3 = 720 m moles de 3-méthyl-butine(1)-ol-(3) dans 100 cm3 d'éther abso- lu; ensuite, on laisse couler lentement, goutte à goutte, 96 cm3 = 750 m moles de quinoléine dans 100 cm3 d'éther absolu, la température intérieure ne devant pas dépasser -5 o Après aboir laissé reposer une nuit à 0 , on sépare du chlor- hydrate de quinoléine à la trompe et on ajoute le filtrat, en refroidissant bien à la glace et au chlorure de sodium, à une solution éthérée d'ammoniaque, tout en introduisant en outre du gaz ammoniac.
Après avoir achevé le travail d'une manière analogue à l'exemple 1), on obtient 30 g a'ester d'acide carba- mique du 3-méthyl-butine(1)-ol-(3), p. d'éb.12;111 ; f.: 108 (cristalli- sation dans le cyclohexane).
EXEMPLE 3.
----------
Une solution de 34 cm3 = 500 millimoles de phosgène liquide dans 150 cm d'éther absolu, comme dans l'exemple 2, est mise d' abord en réaction avec 62 g = 500 m moles de 1-éthi ylcyclohexanol-(1) et 64 cm3 = 500 m moles de quinoléine et ensuite, après avoir continué de travailler selon les exem- ples 1 et 2, avec de l'ammoniaque dans l'éther. On obtient 45 g = 53% de la théorie. Ester d'acide carbamique du 1-éthinylcyclohexane-ol-(1); p.d'éb.3: 108-110 ; aiguilles incolores dans le cyclohexane.F.: 94-960.
EXEMPLE 4.
---------- Une solution de 34 cm3 = 500 millimoles de phosgène liquide dans 150 cm3 d'éther absolu est, comme dans l'exemple 2, mise en réaction avec 49 g = 500 m moles de 3-méthylpentine-(1)-ol-(3) et 64 cm3 de quinoléine et, après séparation du chlorhydrate de quinoléine, on introduit la solution à -10 dans une solution d'environ 40 g de méthylamine dans 100 cm3 d'éther ab- solu. On laisse le mélange arriver en l'espace de quelques heures à la tempé- rature ambiante, on le soumet ensuite à un lavage alcalin et à un lavage aci- de et l'on obtient finalement par distillation dans le vide l'ester d'acide N-méthylcarbamique du méthylpentinol avec un rendement de 45%. P. d'éb.14: 111-112 ; f.: 53-54 - aiguilles incolores par cristallisation dans l'hexane.
EXEMPLE 5.
----------
A une solution de 62 g d'éthinylcyclohexanol dans 200 cm3 de toluè- ne absolu et de 50-52 g de phosgène, on ajoute, goutte à goutte en agitant en l'espace d'environ 1 1/2 heure, 61 g de collidine dans 70 cm3 de toluène, la température étant maintenue à +5 - +10 par refroidissement à la glace.
Après avoir continué l'agitation pendant 1 à 2 heures, on laisse reposer pendant une nuit à environ + 10 . Le chlorhydrate de collidine pré- cipité est séparé à la trompe et lavé ensuite au toluène; les filtrats sont réunis. A travers 250 cm3 de toluène placé en avant, on fait passer un courant

continu de NH3 et on ajoute le filtrat au toluène précité, en refroidissant à la glace, l'addition se faisant assez lentement pour que la température ne dépasse pas +10 . Ensuite, on continue d'agiter encore pendant environ 15 minutes et l'on achève ensuite le travail comme dans les exemples précédents.
La solution toluénique séchée de l'ester d'acide carbamique est refroidie à une température de -5 à 0 , et la plus grande partie se cristallise. Après la séparation à la trompe, on peut concentrer le filtrat et de nouveau le soumettre à la cristallisation ou à la distillation dans le vide.
Rendement total 50,8 g = 60,7% de la théorie.
EXEMPLE 6.
----------
On dissout 6,2 g d'éthinylcyclohexanol dans 100 cm3 de pyridine absolue ; on y ajoute 5,5 cm3 d'isocyanate de phényle et l'on fait bouillir pendant 17 heures au reflux. L'achevement du travail en passant par l'éther fournit, outre de la diphénylurée, 4,2 g d'ester N-phényl-acide carbamique- 1-éthinyl-cyclohexylique-cristaux dans l'hexane. F. : 94-96 .
EXEMPLE 7.
----------
A 23 g = 0,3 mole de chlorure de carbanyle dans 150 cm3 d'éther sec, on ajoute, goutte à goutte, en agitant, à environ 0 , 32 g de propinyl- cyclohexanol dans 150 cm3 d'éther sec ; avoir laissé reposer pendant une nuit, on achève le travail avec de l'eau, on sèche la solution, on la concen- tre fortement et on la refroidit fortement; on obtient ainsi 10 g = 25% de la théorie, de cristaux incolores d'un point de fusion de 137-139 , qui repré- sentent l'ester d'acide carbamique du propinylcyclohexanol.
D'une manière analogue, il se forme à partir de phénéthinyleyclor- hexanol l'ester d'acide carbamique correspondant d'un point de fusion de 110- 111 , cristaux dans l'hexane.
EXEMPLE 8.
On dissout 28 g de chlorure de diéthylcarbanyle, 31 g d'éthinylcy- clohexanol et 31 g de 2,4,6-collidine dans 150 cme de toluène et on les fait bouillir au reflux pendant 5 heures ; sépare du chlorhydrate de collidine précipité, on lave la solution de toluène avec de l'acide dilué, de la lessive et de l'eau, on sèche et on concentre et on distille dans le vide. Fraction principal point d'éb.2: 94-96 ; f. : 30-33 ; rendement : 15 g (= 27% de la théorie). Ester diéthylcarbanylique d'éthinylcyclohexanol.
EXEMPLE 9.
----------
Une solution refroidie de 49,6 g d'éthinylcyclohexanol(correspon- dant à 1 mole) dans 164 cm3 de toluène (séché sur du sodium) est réunie, avec agitation, à environ 0 , avec une solution refroidie pareillement de 51,58 g de phosgène (correspondant à 1,3 mole) dans 157,6 cm3 de toluène. A cela on ajoute, en agitant, à +2 - +4 , à partir d'un entonnoir compte-gouttes, une solution de 27,2 g de triméthylamine (correspondant à 1,15 mole) dans 180 cm3 de toluène, en l'espace de 60-70 minutes. Après avoir encore agité pendant 1/2 heure, à environ 0 , le chlorhydrate de triméthylamine précipité est sé- paré à la trompe sur l'entonnoir-filtre et on lave quatre fois une quantité de toluène telle que le sel soit exactement couvert de solvants après chaque passage.
Le sel est séché à 60 à l'étuve. On obtient 41,45 g correspondant à 94,5% de la triméthylamine employée. On remet le filtrat et le toluène de lavage dans le ballon et on les amine en y introduisant du gaz ammoniac à environ + 3 . Lorsque l'amination est achevée (durée environ 1 1/2 heure), le mélange réactionnel est placé avec 100 cm3 dans un entonnoir à agitation et dilué avec 2 litres d'éther.
On fait écouler la solution de chlorure ammonique, on lave la cou- éthérée successivement à l'eau, plusieurs fois à l'acide sulfurique à 10% et

ensuite de nouveau à l'eau. On épuise ces eaux de lavage à l'éther conjoin- tement avec la solution de chlorure ammonique. Les eaux de lavage chargées d'acide sulfurique sont également épuisées plusieurs fois à l'éther en étant agitées et celui-ci est réuni avec la quantité principale. Après séchage sur du sulfate de sodium, on concentre toute la solution éthérée par évaporation (à la fin dans le vide).
Dès que la cristallisation s'amorce, on ajoute 100cm3 d'essence de pétrole (50-60 ) et on laisse reposer pendant plusieurs heures à 0 pour que la cristallisation s'achève., On sépare à la trompe sur l'enton- noir-filtre et on lave plusieurs fois à l'essence.
Après le séchage à 50 , on obtient 54,45 g d'éthinylcyclohexyluré- thane ayant un point de fusion de 93-95 , correspondant à 81,5% de la théorie.
Des solutions de latage à l'essence on récupère encore, après l'é- vaporation et la distillation aans le vide, un peu de la matière de départ.
Récupération de la triéthylamine :
Dans le ballon à distiller, on ajoute à 95 g (1 mole) du chlorhy- dra,te de triméthylamine obtenu dans les exemples ci-dessus, à l'aide d'un entonnoir compte-gouttes, une lessive de soude caustique à 50% avec 45 g de NaOH,la triméthylamine qui se dégage étant recueillie dans un récipient de condensation (récepteur) refroidi à très basse tempétature. Par chauffage du mélange réactionnel à environ 80 , on chasse les dernières fractions de triméthylamineo On obtient ainsi 58g de triméthylamine déjà très sec, que l'on sèche complètement avec de la potasse caustique et de l'oxyde de baryum, de préférence après dilution avec du toluène. Le rendement s'élève à 98-99% de la théorie. La solution toluénique peut être employée directement pour de nouvelles réactions.
EXEMPLE 10.

A une solution de 70 g de butine-(1)-01-(3) et de 99 g de phosgène dans 400 cm3 d'éther absolu, on a ajouté, à environ - 10 , en l'espace de 4 heures, en agitant, 129 g de quinoléine dans de l'éther absolu. Après un re- "pos prolongé à -6 , on a séparé à la trompe d'avec la chlorhydrate de quinoléi- ne et on a laissé la solution éthérée couler goutte à goutte, à -5 en agitant et en faisant passer de l'ammoniaque, dans de l'éther placé en avant..Lors- que la réaction a pris fin, on achève le travail de la manière habituelle et l'on épure la butine-(1)-01-(3)-uréthane par distillation.

EXEMPLE 11.

A une solution de 50 g d'éthinylcyclohexénylcarbinol et de 40 g de phosgène dans 200 cm3 de toluène, on ajoute lentement, à + 10 , en agitant,
56 cm3 de toluène, puis on agite encore pendant 2 heures et ensuite on conte- nue de travailler comme à l'exemple 1. Après concentration de la solution, on obtient 25,8 g = 40% de la théorie de l'ester d'acide carbamique correspon- dant; après deux recnistallisations dans l'éther, point de fusion : 117-118 .
EXEMPLE 12.

A 25 g de chlorure de carbamyle dans 50 cm3 d'éther sec, on ajoute goutte à goutte, à environ 0 , en agitant, 39 g d'endométhylène-cyclohexényl- éthinyl-carbinol dans 100 cm3 d'éther sec; après avoir laissé reposer pendant une nuit, on achève le travail avec de l'eau, on concentre fortement la so- lution et on la refroidit à basse température. 28 g = 53% de la théorie. P. de fusion : 114 - 116 .

Hitherto, no esters of carbamic acid with acetylene tertiary and secondary alcohols of the general formula are known.

where R1, R2. R3 and R4 represent a hydrogen atom or a hydrocarbon residue, and R a hydrocarbon residue.
However, according to the invention, it has been found that compounds of this type can be obtained by the usual methods of preparing carbamic acid esters from alcohols. Thus, for example, it is possible to start from nitrogen-substituted or unsubstituted carbamyl halides and transform them into the desired esters, by reaction with the corresponding tertiary or secondary acetylene carbinols.
However, it is also possible to start from the haloformic acid esters of tertiary or secondary acetylene carbinols, which can be obtained for example by reaction of carbonyl dihalides, such as phosgene, with tertiary or secondary acetylene carbinols, and to react ammonia or amines on haloformic acid esters.
In view of the known reaction power of the acetylene group, which foreshadowed undesirable side reactions, for example cyclizations, or in view of the particular known sensitivity of tertiary haloformic acid esters (cf. German patent 254,471). it was to some extent surprising that the reactions according to the invention were carried out with yields which were absolutely usable technically.
The products obtained by the process have interesting therapeutic properties, for example some of them exhibit a pronounced hypnotic effect with simultaneously minimal secondary phenomena.

EXAMPLE 1.

To 34 g = 430 millimoles of carbamyl chloride in 250 cm3 of absolute ether, are added, while cooling in ice, 42 g = 430 moles of 3-methylpentine- (1) -ol- (3). After leaving to stand overnight, it is washed with water and the work is completed in the usual manner. Vacuum distillation gives a colorless distillate, of 1%. from 12: 120-121. Crystals in hexane, f. : 56-58., The reaction product forms a silver salt, can be converted to the carbamic acid ester of methyldiethylcarbinol by catalytic hydrogenation and thus is found to be a carbamic acid ester of 3- metbyl-pentine- (1) -ol- (3).
EXAMPLE 2.
---------- A solution of 51 cm3 = 750 millimoles of phosgene dals 150 cm3 of absolute ether is added, while cooling with ice and :: -..- sodium chloride, to 67 cm3 = 720 moles of 3-methyl-butine (1) -ol- (3) in 100 cm3 of absolute ether; then, one lets flow slowly, drop by drop, 96 cm3 = 750 moles of quinoline in 100 cm3 of absolute ether, the internal temperature not having to exceed -5 o After having left to stand overnight at 0, the chlorine is separated - Quinoline hydrate with suction and the filtrate is added, cooling well with ice and sodium chloride, to an ethereal solution of ammonia, while additionally introducing ammonia gas.
After having completed the work in a manner analogous to Example 1), 30 g of 3-methyl-butine (1) -ol- (3) carbamic acid ester are obtained, e.g. from eb 12; 111; f .: 108 (crystallization from cyclohexane).
EXAMPLE 3.

A solution of 34 cm3 = 500 millimoles of liquid phosgene in 150 cm of absolute ether, as in Example 2, is first reacted with 62 g = 500 moles of 1-ethylcyclohexanol- (1) and 64 cm3 = 500 moles of quinoline and then, after continuing to work according to Examples 1 and 2, with ammonia in ether. We obtain 45 g = 53% of theory. 1-Ethinylcyclohexan-ol- carbamic acid ester (1); bb 3: 108-110; colorless needles in cyclohexane.F .: 94-960.
EXAMPLE 4.
---------- A solution of 34 cm3 = 500 millimoles of liquid phosgene in 150 cm3 of absolute ether is, as in Example 2, reacted with 49 g = 500 moles of 3- methylpentine- (1) -ol- (3) and 64 cm3 of quinoline and, after separation of the quinoline hydrochloride, the solution is introduced at -10 in a solution of about 40 g of methylamine in 100 cm3 of ab- ether. solu. The mixture is allowed to come to room temperature within a few hours, then subjected to an alkaline wash and an acid wash, and finally the ester of β-ester is obtained by vacuum distillation. N-methylcarbamic acid of methylpentinol with a yield of 45%. Bp 14: 111-112; f .: 53-54 - colorless needles by crystallization from hexane.
EXAMPLE 5.
----------
To a solution of 62 g of ethinylcyclohexanol in 200 cm3 of absolute toluene and 50-52 g of phosgene is added, dropwise with stirring over the course of about 1 1/2 hours, 61 g of collidine in 70 cm3 of toluene, the temperature being maintained at +5 - +10 by ice cooling.
After continuing stirring for 1 to 2 hours, it is left to stand overnight at about + 10. The precipitated collidine hydrochloride is removed with suction and then washed with toluene; the filtrates are combined. A current is passed through 250 cm3 of toluene placed in front

of NH3 and the aforementioned toluene filtrate is added with ice-cooling, the addition being slowly enough so that the temperature does not exceed +10. Then, stirring is continued for about 15 minutes more and then the work is completed as in the previous examples.
The dried toluene solution of the carbamic acid ester is cooled to a temperature of -5 to 0, and most of it crystallizes. After separation by suction, the filtrate can be concentrated and again subjected to crystallization or vacuum distillation.
Total yield 50.8 g = 60.7% of theory.
EXAMPLE 6.
----------
6.2 g of ethinylcyclohexanol are dissolved in 100 cm3 of absolute pyridine; 5.5 cm3 of phenyl isocyanate are added thereto and the mixture is boiled for 17 hours at reflux. Completion of the work through the ether provides, in addition to the diphenylurea, 4.2 g of N-phenyl-carbamic acid-1-ethinyl-cyclohexyl ester-crystals in hexane. F .: 94-96.
EXAMPLE 7.

To 23 g = 0.3 mole of carbanyl chloride in 150 cm3 of dry ether is added, dropwise, with stirring, to about 0.32 g of propinyl-cyclohexanol in 150 cm3 of dry ether; having left to stand overnight, the work is completed with water, the solution is dried, strongly concentrated and strongly cooled; 10 g = 25% of theory are thus obtained of colorless crystals with a melting point of 137-139, which represent the carbamic acid ester of propinylcyclohexanol.
Similarly, the corresponding carbamic acid ester with a melting point of 110-111 crystals in hexane is formed from phenethinyleyclorhexanol.
EXAMPLE 8.
28 g of diethylcarbanyl chloride, 31 g of ethinylcyclohexanol and 31 g of 2,4,6-collidine are dissolved in 150 cc of toluene and boiled under reflux for 5 hours; Separated from precipitated collidine hydrochloride, the toluene solution was washed with dilute acid, lye and water, dried and concentrated and distilled in vacuo. Fraction main rb 2: 94-96; f. : 30-33; yield: 15 g (= 27% of theory). Ethinylcyclohexanol diethylcarbanyl ester.
EXAMPLE 9.

A cooled solution of 49.6 g of ethinylcyclohexanol (corresponding to 1 mole) in 164 cm3 of toluene (dried over sodium) is combined, with stirring, to about 0, with a similarly cooled solution of 51.58 g. of phosgene (corresponding to 1.3 moles) in 157.6 cm3 of toluene. To this is added, while stirring, at +2 - +4, from a dropping funnel, a solution of 27.2 g of trimethylamine (corresponding to 1.15 mol) in 180 cm3 of toluene, in l 'within 60-70 minutes. After stirring for a further 1/2 hour, at about 0, the precipitated trimethylamine hydrochloride is pumped off on the filter funnel and an amount of toluene is washed four times such that the salt is exactly covered with solvents. after each pass.
The salt is dried at 60 in an oven. 41.45 g is obtained, corresponding to 94.5% of the trimethylamine used. The filtrate and the washing toluene are returned to the flask and they are aminated by introducing ammonia gas at about + 3. When the amination is complete (duration approximately 1 1/2 hours), the reaction mixture is placed with 100 cm3 in a stirred funnel and diluted with 2 liters of ether.
The ammonium chloride solution is run off, the cou- ethereal is washed successively with water, several times with 10% sulfuric acid and

then back to water. These ether washings are depleted together with the ammonium chloride solution. The washing waters loaded with sulfuric acid are also exhausted several times with ether while being stirred and the latter is combined with the main quantity. After drying over sodium sulfate, all of the ethereal solution is concentrated by evaporation (at the end in a vacuum).
As soon as crystallization begins, 100 cm3 of petroleum gasoline (50-60) are added and the mixture is left to stand for several hours at 0 so that the crystallization is completed., Separation with suction on the funnel -filter and wash several times with gasoline.
After drying at 50, 54.45 g of ethinylcyclohexylurethene having a melting point of 93-95 are obtained, corresponding to 81.5% of theory.
After evaporation and vacuum distillation, some of the starting material is still recovered from the gasoline latting solutions.
Recovery of triethylamine:
In the distillation flask, is added to 95 g (1 mol) of the hydrochloride, trimethylamine obtained in the above examples, using a dropping funnel, a solution of caustic soda at 50 % with 45 g of NaOH, the trimethylamine which evolves being collected in a condensing vessel (receiver) cooled to very low temperature. By heating the reaction mixture to about 80, the last fractions of trimethylamineo are removed.This gives 58 g of already very dry trimethylamine, which is completely dried with caustic potassium hydroxide and barium oxide, preferably after dilution with toluene. The yield is 98-99% of theory. The toluene solution can be used directly for new reactions.
EXAMPLE 10.

To a solution of 70 g of butine- (1) -01- (3) and 99 g of phosgene in 400 cm3 of absolute ether was added, at about -10, over the course of 4 hours, in stirring, 129 g of quinoline in absolute ether. After prolonged standing at -6, the quinoline hydrochloride was sucked off and the ethereal solution was allowed to drip, at -5 with stirring and passing through. ammonia, in ether placed in front .. When the reaction has ended, the work is completed in the usual manner and the butine- (1) -01- (3) -urethane is distilled off .
Eb 12: 108-109. Melting point: 48-50, crystals in hexane.
Yield: 50 g = 49% of theory.
EXAMPLE 11.

To a solution of 50 g of ethinylcyclohexenylcarbinol and 40 g of phosgene in 200 cm3 of toluene is added slowly, at + 10, with stirring,
56 cm3 of toluene, then stirring for a further 2 hours and then continuing to work as in Example 1. After concentration of the solution, 25.8 g = 40% of the theory of the ester d are obtained. the corresponding carbamic acid; after two recnistallizations in ether, melting point: 117-118.
EXAMPLE 12.

To 25 g of carbamyl chloride in 50 cm3 of dry ether is added dropwise, at approximately 0, with stirring, 39 g of endomethylene-cyclohexenyl-ethinyl-carbinol in 100 cm3 of dry ether; after allowing to stand overnight, the work is completed with water, the solution is strongly concentrated and cooled to low temperature. 28 g = 53% of theory.
