PROCESS FOR PREPARATION OF COMPOUNDS OF LONG-TERM PENICILLIN (PENICILLIN RETARD).
The main patent describes a process for the preparation of compounds of penicillin resulting from the combination of penicilin with various organic bases, these compounds making it possible to prepare forms with a prolonged effect (depot penicillin) due to their very low solubility. - content in water or blood serum.
The subject of the present addition is a process for the preparation of novel compounds of penicillin and of organic bases, which can be injected either in the form of oily suspensions, or in the form of aqueous suspensions, or finally in the form of solutions of higher viscosity than that. water, these compounds consisting of the combination of penicilin with organic amino bases.
Among these amino bases, we can mention
EMI1.1
amino-diphelylmethane (benzhydrylamine) of formula (C6H5) 2-CHoNH2 'amano-2-methyl-4-hexane, amino-2-meths + 1-6-heptaneo
The penicillinates of these amino bases can be obtained by double decomposition between a salt of penicillin (sodium, calcium, potassium salt) and a salt (hydrochloride, sulfate, etc.) of the amino base.
They can also be obtained by the action of penicillin in its free acid form and of the free amino base in an organic solvent, either by direct crystallization or by precipitation of the penicillinate by adding another organic solvent.
The following non-limiting examples illustrate an embodiment of the invention, namely obtaining the amino base penicillinate by double decomposition.
