Avec ces mandélamidines on peut préparer des sels d'addition d'acides non toxiques de ces composés et leurs isomères optiques.
On a trouvé que les composés répondant à la formule ci-dessus possèdent des activités sur le système cardio- vasculaire et sont particulièrement intéressants pour le traitement de l'hypertension artérielle.

On peut obtenir les mandélamidines de formule I dans lesquelles R4 représente un groupe -OH (mandél- amidoximes) en faisant réagir une mandélamidine de formule I dans laquelle R3 et R4 sont des atomes d'hy drogène, obtenue selon le procédé ci-dessus, avec l'hy- droxylamine.
Pour la préparation du 3,4-méthylènedioxy-mandélo- nitrile de départ, on fait réagir, en règle générale, le pipé ronal avec le cyanure de potassium en milieu aqueux, en présence de bisulfite de sodium comme catalyseur et d'acide chlorhydrique dilué jusqu'à pH 4,0 à 5,0 environ, en opérant dans un appareil de réaction étanche.

A une solution de 10 g de chlorhydrate de 3,4-mé- thylène-dioxy-mandélamidine (0,043 mole) dans 70 ml de méthanol, on ajoute lentement une solution d'hy- droxylamine, obtenue par addition de 3,4 g de méthy- late de sodium (0,063 mole) à 4,4 g de chlorhydrate d'hydroxylamine (0,063 mole) en milieu méthanolique puis filtration après 30 mn d'agitation.
Après 48 heures de repos, on évapore sous vide. Le résidu est repris à l'eau, filtré, essoré et desséché sous vide.
On obtient 7,3 g de 3,4-méthylènedioxy-mandélami- doxime (F: 1030 C) que l'on transforme en chlorhydrate en solution dans l'éthanol.
Le chlorhydrate est précipité par addition d'éther éthylique puis purifié par cristallisation dans un mélange éthanol-acétate d'éthyle.
Le chlorhydrate de la 3,4-méthylènedioxy-mandél- amidoxime obtenu avec un rendement de 80 0/0 (8,5 g) se présente sous l'aspect d'une poudre blanche; soluble dans l'eau (à 20 % environ), le méthanol, l'éthanol, et insoluble dans l'éther éthylique, l'éther de pétrole, le benzène, l'acétate d'éthyle. Le point de fusion instantané est de 165o C.
<I>Exemple 2:</I> <I>Chlorhydrate</I> de (-) et (-I-) (3,4-méthylènedioxy)-mandélamidine 29 g de chlorhydrate de 3,4-méthylènedioxy-mandél- amidine (0,125 mole), en suspension dans 30 ml d'eau, sont traités, à froid, par une solution de 6 g de soude (0,125 mole) dans 30 ml d'eau.
L'amidine base, libérée, insoluble, est rapidement iso lée par filtration, essorée et séchée sous vide.
La 3,4-méthylènedioxy-mandélamidine racémique, ob tenue avec un rendement de 80 % (19,5 g), a un point de fusion instantané de 1180 C.
On ajoute, à une solution de ce produit (0,1 mole) dans 80 ml d'éthanol, 15,2g d'acide L-mandélique (0,1 mole), a20D = -156,5 (c = 5 % dans l'eau). On chauffe à 500 C pour tout dissoudre, on laisse cristalliser et on filtre. On obtient<B>17,3</B> g d'un produit la dont le point de fusion instantané est de 160-20 C.
Le filtrat évaporé à sec laisse un résidu qui, repris par 100 ml d'un mélange, à volume égal d'acétone et d'éther éthylique, fournit après filtration 20 g d'un pro duit IIa dont le point de fusion instantané est de 132-4,) C.
Le produit la (17,3 g) recristallisé dans l'éthanol donne 13 g d'un produit<B>lb</B> (F: 168-90 C). Le filtrat éva poré laisse un résidu qui, traité par le mélange acétone- éther éthylique, fournit 3,2 g d'un produit IIb (F 132 C).
Le produit IIa traité par l'acétone à chaud laisse un insoluble<B>le,</B> 5 g (F : 165-6a C). Après cristallisation de la solution acétonique on obtient 14 g d'un composé IIc (F: 135-7 C).
Les produits lb (13 g) et Ic (5 g) recristallisés dans l'éthanol permettent d'obtenir 16,2 g de () mandélate de (-) (3,4-méthylènedioxy)-mandélamidine (composé I) dont le point de fusion instantané est de 168-90 C et le pouvoir rotatoire a20D = + 8 (c = 0,5 % dans le mé thanol).
<I>Chlorhydrate</I> de () (3,4-méthylènedioxy)-mandélamidine Le chlorhydrate de la () (3,4-méthylènedioxy)-man- délamidine est préparé à partir du composé I qui, en suspension dans 30 ml d'eau, est traité par un excès d'acide chlorhydrique 5 N.
L'acide (-)mandélique libéré est extrait avec trois fois 100 ml d'éther éthylique. La solution aqueuse, éva porée sous vide, laisse un résidu que l'on purifie par cristallisation dans un mélange éthanol-éther éthylique. On obtient, avec un rendement de 63 % , 9,2 g de chlor- hydrate de (-) (3,4-méthylènedioxy)-mandélamidine qui se présente sous l'aspect d'une poudre cristalline blan che, dont le point de fusion instantané est de 160o C et ayant un pouvoir rotatoire a20D = -71 (c = 0,5 % dans le méthanol).
<I>Chlorhydrate</I> de (-I-) (3,4-méthylènedioxy)-mandélamidine Le chlorhydrate de la (-i-) (3,4-méthylènedioxy)- mandélamidine est préparé à partir du composé II (12,5 g) traité comme décrit pour le composé I. On ob tient, avec un rendement de 50 %, 7,2 g de chlorhydrate de (-I-) (3,4-méthylènedioxy)-mandélamidine se présen tant sous l'aspect de cristaux prismatiques blancs, dont le point de fusion instantané est de 160o C et possédant un pouvoir rotatoire a20D = + 69 (c = 0,5 % dans le méthanol).
<I>Exemple 3:</I> <I>Chlorhydrate</I> de (-) (3,4-méthylènedioxy)-mandélamidoxime Une solution d'hydroxylamine, préparée à partir de 4,2 g de chlorhydrate d'hydroxylamine (0,06 mole) et de 3,25 g de méthylate de sodium (0,06 mole) dans 50 ml de méthanol, est ajoutée à une solution de 9,2 g de chlorhydrate de () (3,4-méthylènedioxy)-mandélamidine, préparé à l'exemple 2 dans 50 ml de méthanol.
Après 48 heures de repos, on acidifie avec une solu tion éthanolique d'acide chlorhydrique, on évapore sous vide, on reprend le résidu à l'eau et on précipite la base par addition de carbonate acide de sodium. On obtient après filtration 7,4 g de l'amidoxime base dont le point de fusion instantané est de 114o C.
Ce produit solubilisé dans l'éther éthylique est traité par une solution éthanolique d'acide chlorhydrique. Le chlorhydrate isolé est purifié par cristallisation dans un mélange éthanol-acétate d'éthyle.
Le chlorhydrate de (-) (3,4-méthylènedioxy)-mandél- amidoxime, obtenu avec un rendement de 83 %(8,2 g) se présente sous l'aspect de cristaux blancs, dont le point de fusion instantané est de 138-9 C, solubles dans l'eau, le méthanol, l'éthanol, et insolubles dans l'éther éthylique, l'éther de pétrole, le benzène, l'acétate d'éthyle. Il possède un pouvoir rotatoire a20D = -42- (c = 0,5 % dans le méthanol).
<I>Exemple 4:</I> <I>Chlorhydrate</I> de (-I-) (3,4-méthylènedioxy)-mandélamidoxime Le chlorhydrate de (-i-) (3,4-méthylènedioxy)-man- délamidoxime a été préparé, selon le mode opératoire décrit à l'exemple 3, en utilisant 7,7g de chlorhydrate de (-I-) (3,4-méthylènedioxy)-mandélamidine, préparé à l'exemple 2.
Le produit obtenu, avec un rendement de 76 0/0 (6,2 g) se présente sous l'aspect d'une poudre blanche, dont le point de fusion instantané est de 138-9 C, soluble dans l'eau, le méthanol, l'éthanol, et insoluble dans l'éther éthylique, l'éther de pétrole, le benzène, l'acétate d'éthyle. Il possède un pouvoir rotatoire a20D = + 46o (c = 0,5 0/0 dans le méthanol).
On a en outre préparé les produits suivants 4-hydroxy-3-méthoxy-mandélamidine, HCl (F : 2000 C) 4-méthoxy-mandélamidine, HCl (F : 246-C) N-diéthyl-mandélamidine, HCl (F : 167e C) N-phényl-mandélamidine, HCl (F: 128-130 C) N-isopropyl-3-hydroxy-mandélamidine, HCl (F: 212-3e C) N-isopropyl-3,4-dihydroxy-mandélamidine, HCl (F: 1730 C) 3-hydroxy-mandélamidoxime, HCl (F: 1840 C).
Les produits préparés par le procédé selon l'inven tion fournissent des sels d'addition avec les acides, et en particulier les chlorhydrates se présentent sous l'aspect de cristaux blancs solubles dans l'eau, le méthanol, l'éthanol, et insolubles dans l'éther éthylique, l'éther de pétrole, le benzène, l'acétate d'éthyle.
Les activités pharmacologiques des produits obtenus par le procédé selon l'invention sont les suivantes On a étudié l'activité du chlorhydrate de 3,4-méthy- lènedioxy-mandélamidoxime (exemple 1) quant à la toxi cité aiguë et des propriétés convulsivantes et les proprié tés cardiovasculaires, et on a constaté une - augmentation de 25 % fugace de la pression artérielle différentielle par élévation de la pression systolique. - très légère augmentation (+ 9 0/0) de la pression artérielle moyenne qui passe de 80 à 87 mm de mer cure.
- légère diminution (-14 0/0) de la pression artérielle dans le ventricule droit après 15 mn.
- diminution de la fréquence cardiaque, -32 % après 15 mn. Le rythme passe de 155 à 105 battements/mn. - augmentation du débit cardiaque et surtout du débit systolique (+ 78 % après 15 mn).
- augmentation de l'index cardiaque, + 20 (1/o après 15 mn.
- augmentation du stroke volume index de 20 0/0 en fin de perfusion et de 77 % après 15 mn.
- augmentation de 20 % du travail ventriculaire gau che pendant 15 mn.
- diminution de la résistance périphérique de 16 % après 15 mn.
On a étudié pharmacologiquement d'autres composés obtenus par le procédé selon l'invention et on donne ci- après le résumé des propriétés qui ont pu être mises en évidence. 4-hydroxy-3-méthoxy-mandélamidine Le produit en solution aqueuse présente un pH de 5. Par voie intraveineuse, chez la souris, la DL50 est 400 mg/kg. Les animaux qui meurent ont une respiration accélérée et leur mort intervient en quelques minutes. 200 mg/kg du produit injecté par voie intramuscu laire à des souris n'ont provoqué aucune action ni sur le système nerveux végétatif, ni tranquillisante, ni anal gésique.
4-méthoxy-mandélamidine Le produit est soluble dans l'eau à la concentration de 0,5 %. Par voie intraveineuse, chez la souris, la DL50 est 117 mg/kg. Les symptômes observés sont: convul sions, pâleur, temps de saignement allongé après l'in- jection, quand la mort a lieu elle est immédiate. Des souris ayant reçu 60 mg/kg du produit par voie intra musculaire ont eu une forte diarrhée et une légère hypo thermie.
L'étude des propriétés cardiovasculaires a donné les résultats suivants le)<I>Pression artérielle du</I> rat <I>éveillé</I> Le produit par voie intramusculaire à la dose de 60 mg/kg sur 5 rats anesthésiés, provoque une hypo tension maximale de 200/0 entre la 30e et la 120e mi nute. Le retour à la normale n'est pas observé en 6 heures. 211) <I>Pression artérielle du lapin anesthésié</I> Le produit par voie intramusculaire, à la dose de 60 mg/kg sur 10 animaux, abaisse la pression artérielle de 33 % après 2 heures. L'hypotension débute après 30 mn. Trois heures après l'injection, l'hypotension est encore de 29 %.
N-isopropyl-3-hydroxy-mandélamidine Par voie intraveineuse, chez la souris. la DL,-,, est 36 mg/kg.
Par voie intramusculaire, elle est 135 mg/kg. A de fortes doses, proches de la DL50 (125 mg/kg), on observe des convulsions chez les animaux traités.
L'étude des propriétés cardiovasculaires a donné les résultats suivants 1 ) Pression artérielle chez le rat éveillé Par voie intramusculaire, la dose de 70 mg/kg, chez 5 rats, abaisse la pression artérielle en moyenne de 13 0/0 sauf dans un cas, l'action hypotensive ne débute que tar divement après 1 h à 5 h, le maximum d'effet se situe entre 2 et 6 h, le retour à la normale a lieu entre _5 et 6 h 30. 20) Action cardiovasculaire chez le chat Par voie intraveineuse chez un chat à 20 mg/kg, le produit abaisse la pression carotidienne de 40 % . Il ne modifie pas l'action pressive de l'adrénaline, de l'acétyl choline, de la Tyramine. Il augmente l'hypertension pro duite par le diméthylphénylpipérazinium.
Sur la mem brane nictitante, il n'exerce aucun effet direct, ne modi fie pas les contractions produites par stimulations pré et post-ganglionnaires du nerf cervical supérieur.
Par voie intra-artérielle vers le ganglion aux doses de 166 et 330 mg/kg, le produit abaisse légèrement (12 0/0) la pression artérielle à la dose la plus forte. Il diminue de façon fugace les contractions produites par stimulations pré et post-ganglionnaires du nerf cervical supérieur.
Le produit a été utilisé chez l'homme à la dose de 25 mg par dose, 4 comprimés ou cachets par jour avec de bons résultats dans 70 0/0 des cas. N-isopropyl-3,4-dihydroxy-mandélamidine Le produit en solution aqueuse présente un pH de 5. Par voie intraveineuse, chez la souris, la DL50 est 62,5 mg/kg. Les symptômes observés sont : dyspnée, exophtahnie, vasoconstriction périphérique.
L'étude des propriétés cardiovasculaires a donné les résultats suivants 1 0) Cour isolé de lapin Sur 2 cours perfusés avec du liquide de Van Dyke Hastings sans chlorure de baryum, le produit à la dose de 100 g/ml # augmente légèrement le débit coronarien (-I- 31 ()/o) # exerce un effet inotrope positif (-f- 96 %) v ne modifie pas le rythme.
211) <I>Pression artérielle du lapin anesthésié</I> Sur 5 lapins, le produit à la dose de 31 mg/kg par voie intramusculaire abaisse la pression artérielle de 400/0.
3-hydroxy-mandélamidoxime Par voie intraveineuse, chez la souris, des doses al lant jusqu'à 1 g/kg n'ont provoqué aucune mortalité, les animaux manifestant seulement un phénomène de pilo- érection. Des souris ayant reçu 1 g/kg du produit par voie intramusculaire ont présenté les phénomènes sui vants: légère excitation, piloérection importante, légère hypothermie, pas d'action tranquillisante ni d'action analgésique.
Le produit injecté à la dose de 500 mg/kg par voie intramusculaire à 3 rats éveillés, a abaissé en moyenne leur pression artérielle de 20 % .

7.3 g of 3,4-methylenedioxy-mandelamidoxime (F: 1030 C) are obtained, which is converted to the hydrochloride in solution in ethanol.
The hydrochloride is precipitated by adding ethyl ether and then purified by crystallization from an ethanol-ethyl acetate mixture.
The hydrochloride of 3,4-methylenedioxy-mandel-amidoxime obtained in a yield of 80% (8.5 g) appears as a white powder; soluble in water (approximately 20%), methanol, ethanol, and insoluble in ethyl ether, petroleum ether, benzene, ethyl acetate. The instantaneous melting point is 165o C.
<I> Example 2: </I> <I> Hydrochloride </I> of (-) and (-I-) (3,4-methylenedioxy) -mandelamidine 29 g of 3,4-methylenedioxy-mandel hydrochloride amidine (0.125 mole), suspended in 30 ml of water, are treated, cold, with a solution of 6 g of sodium hydroxide (0.125 mole) in 30 ml of water.
The amidine base, liberated, insoluble, is rapidly isolated by filtration, drained and dried under vacuum.
Racemic 3,4-methylenedioxy-mandelamidine, obtained in 80% yield (19.5 g), has an instantaneous melting point of 1180 C.

The product IIa treated with hot acetone leaves an insoluble <B> le, </B> 5 g (F: 165-6a C). After crystallization of the acetone solution, 14 g of a compound IIc (F: 135-7 C) are obtained.
The products lb (13 g) and Ic (5 g) recrystallized from ethanol make it possible to obtain 16.2 g of (-) (3,4-methylenedioxy) -mandelamidine () mandelate (compound I) whose point instantaneous melting temperature is 168-90 C and the optical rotation a20D = + 8 (c = 0.5% in methanol).
<I> () (3,4-methylenedioxy) -mandelamidine hydrochloride </I> () (3,4-methylenedioxy) -man-delamidine hydrochloride is prepared from compound I which, suspended in 30 ml of water is treated with an excess of 5N hydrochloric acid.
The liberated mandelic acid (-) is extracted with three times 100 ml of ethyl ether. The aqueous solution, evaporated in vacuo, leaves a residue which is purified by crystallization from an ethanol-ethyl ether mixture. 9.2 g of (-) (3,4-methylenedioxy) -mandelamidine hydrochloride is obtained with a yield of 63%, which appears as a white crystalline powder, the point of which is instantaneous melting is 160o C and having a rotatory power a20D = -71 (c = 0.5% in methanol).
<I> (-I-) (3,4-methylenedioxy) -mandelamidine hydrochloride </I> (-i-) (3,4-methylenedioxy) - mandelamidine hydrochloride is prepared from compound II (12 , 5 g) treated as described for compound I. There is obtained, with a yield of 50%, 7.2 g of (-I-) (3,4-methylenedioxy) -mandelamidine hydrochloride present under the appearance of white prismatic crystals, with an instantaneous melting point of 160o C and possessing a rotatory power a20D = + 69 (c = 0.5% in methanol).
<I> Example 3: </I> <I> (-) (3,4-methylenedioxy) -mandelamidoxime hydrochloride </I> A hydroxylamine solution, prepared from 4.2 g of hydroxylamine hydrochloride (0.06 mole) and 3.25 g of sodium methoxide (0.06 mole) in 50 ml of methanol, is added to a solution of 9.2 g of () (3,4-methylenedioxy) hydrochloride -mandelamidine, prepared in Example 2 in 50 ml of methanol.
After standing for 48 hours, the mixture is acidified with an ethanolic solution of hydrochloric acid, evaporated in vacuo, the residue is taken up in water and the base is precipitated by adding sodium hydrogen carbonate. 7.4 g of amidoxime base are obtained after filtration, the instantaneous melting point of which is 114o C.
This product, solubilized in ethyl ether, is treated with an ethanolic solution of hydrochloric acid. The isolated hydrochloride is purified by crystallization from an ethanol-ethyl acetate mixture.
(-) (3,4-methylenedioxy) -mandel-amidoxime hydrochloride, obtained with a yield of 83% (8.2 g), appears as white crystals, the instantaneous melting point of which is 138 -9 C, soluble in water, methanol, ethanol, and insoluble in ethyl ether, petroleum ether, benzene, ethyl acetate. It has a rotatory power a20D = -42- (c = 0.5% in methanol).
<I> Example 4: </I> <I> (-I-) (3,4-methylenedioxy) -mandelamidoxime hydrochloride </I> (-i-) (3,4-methylenedioxy) -man hydrochloride - Delamidoxime was prepared, according to the procedure described in Example 3, using 7.7 g of (-I-) (3,4-methylenedioxy) -mandelamidine hydrochloride, prepared in Example 2.
