PROCEDE DE PREPARATION DE 2-SULFANILAMIDO-THIODIAZOLS
En comparaison d'autres sulfamides les 2-sulfanilamido=1, 3, 4 thiodiazols sont remarquables par une toxicité réduite, peu d'effets acces- soires, degré d'acétylation faible et un effet bactériostatique éminant en regard d'un bon nombre des microbes pathogènes les plus communs.
Ces composés ont été produits jusqu'à présent selon la méthode ordinaire en faisant réagir de chlorure de l'acide acétylsulfanilique et d'un 2-amino-1,3 4-thiodiazol et en hydrolysant le composé acétylique ou disulfonylique obtenue
Cependant la préparation des 2-amino-1. 3, 4-thiodiazols est dif- ficile, parce qu'elle se compose de plusieurs réactions dont quelques-unes ne donnent que des rendements peu satisfaisants. De plus la réaction avec du chlorure de l'acide acéthlsulfanilique donne un rendement assez réduit et conséquemment on perd beaucoup d'amino-thiodiaol, qui ne peut pas être regagné.
On a trouvé, que les composés 2-amino-1, 3, 4-thiodiazols en ques- tion peuvent être préparés avec des rendements satisfaisants selon une mé- thode essentiellement nouvelle, qui a le grand avantage, que,. le nombre des procédés est notablement réduit comparativement à la méthode ci-dessus.
Suivant l'invention on fait réagir une benzène-sulfonamide para- substituée par un groupe susceptible d'être transformé en un groupe aminogè- ne ou en un sel alcalin d'un tel composé avec un surplus de thiophosgène, par quel moyen un des atomes hydrogènes du groupe aminogènes est remplacé par le groupe - CSC1 On fait réagir le composé ainsi obtenu avec un surplus de l'hydrazine en solution aqueuse, et on obtient un composé sulfonylique de thiosemicatbazide.
En faisant réagir ce composé avec des halogénures carboxyliques d'ordre moins élevé une chaîne fermée se forme, et on obtient des 2-ben- zénesulfamido-thiodiazols para-substitués.

La réaction finie, on enlève la majeure partie du toluène et le surplus du thiophosgène par distillation ; fait réagir le résidu avec une solution aqueuse de l'hydrazinehydrate froide (environ 10 pet) qui est pré- paré de 20 grammes du sulfate d'hydrazine; on agite vigoureusement la com- binaison et la réaction faite, on étend avec 100 cm3 d'eau. On enlève la ma- tière organique par l'addition d'un peu d'ether. La phase aqueuse enlevée, on fait sécher la solution toluène-éthérique et on enlève l'éther par dis- tillation.
On traite l'huile semi-cristalline restante avec 80 grammes de chlorure acétylique pur. Après un chauffage de peu de durée, une réaction active commence, par laquelle la combinaison se solidifieen une masse cris- talline en dégageant de l'acide chlorhydrique. On enlève le toluène en broyant avec de l'alcool froid, et on fait recristalliser le produit de l'alcool. On obtient du 2-acétylsulfanilamido-5-méthyl-l, 3, 4-thiodiazol sous forme d'une substance blanche et cristalline avec point' de fusion 235 C. La substance se hydrolyse par solution en l'hydroxyde de sodium aqueux 2-normal et par chauffage au point d'ébullition pendant vingt minu- tes environ.
Quand on ajoute de l'acide acétique jusqu'à pH 502 le 2-sulfanila- mido-5-méthyl-1, 3, 4-thiodiazol se dépose sous forme des cristals en écail- les sans couleur. La substance se fond à 207 C.

In comparison with other sulfonamides, 2-sulfanilamido = 1, 3, 4 thiodiazols are remarkable for their reduced toxicity, few side effects, low degree of acetylation and an outstanding bacteriostatic effect compared to a good number of most common pathogens.
These compounds have heretofore been produced by the ordinary method by reacting acetylsulfanilic acid chloride and a 2-amino-1,3 4-thiodiazol and hydrolyzing the acetyl or disulfonyl compound obtained.
However the preparation of 2-amino-1. 3, 4-thiodiazols is difficult, because it is composed of several reactions, some of which give only unsatisfactory yields. In addition, the reaction with acetylsulfanilic acid chloride gives a fairly reduced yield and consequently a lot of amino-thiodiaol is lost, which cannot be regained.
It has been found that the 2-amino-1, 3, 4-thiodiazol compounds in question can be prepared in satisfactory yields by an essentially new method, which has the great advantage, that. the number of methods is significantly reduced compared to the above method.
According to the invention, a benzene-sulfonamide para-substituted with a group capable of being converted into an aminogenic group or into an alkali salt of such a compound is reacted with a surplus of thiophosgene, by which means one of the atoms hydrogens of the aminogenic group is replaced by the group - CSC1. The compound thus obtained is reacted with a surplus of hydrazine in aqueous solution, and a thiosemicatbazide sulfonyl compound is obtained.
By reacting this compound with lower order carboxylic halides a closed chain is formed, and para-substituted 2-benzenesulfamido-thiodiazols are obtained.

This will be better understood by the following limiting examples: Example 1. - 23.6 grams of sodium acetylsulfide sodium are suspended in 100 cm3 of toluene and a solution of 15 grams thiophosgene is added in 60 cm3 toluene while cooling and stirring vigorously.
When the reaction is complete, most of the toluene and the excess thiophosgene are removed by distillation; reacts the residue with an aqueous solution of the cold hydrazinehydrate (about 10 µl) which is prepared from 20 grams of the hydrazine sulfate; the combination is stirred vigorously and the reaction is completed, diluted with 100 cm3 of water. Organic matter is removed by adding a little ether. With the aqueous phase removed, the toluene-ether solution is dried and the ether removed by distillation.
The remaining semi-crystalline oil is treated with 80 grams of pure acetyl chloride. After heating for a short time, an active reaction begins, in which the combination solidifies into a crystalline mass giving off hydrochloric acid. The toluene is removed by grinding with cold alcohol, and the product is recrystallized from the alcohol. 2-Acetylsulfanilamido-5-methyl-1,3,4-thiodiazol is obtained as a white, crystalline substance with a melting point of 235 C. The substance is hydrolyzed by solution to aqueous sodium hydroxide 2- normal and by heating to the boiling point for about twenty minutes.
When acetic acid is added up to pH 502, 2-sulfanilamido-5-methyl-1, 3, 4-thiodiazol is deposited as colorless crystal crystals. The substance melts at 207 C.
