EMI1.1

<Desc/Clms Page number 2>

<Desc/Clms Page number 3>
<Desc/Clms Page number 4>
<Desc/Clms Page number 5>

"Process for the preparation of oxyhydrophenanthrene carboxylic acids or their derivatives." The first three patent applications filed in SWITZERLAND, the
January 10, 1944, April 4, 1944 and May 19, 1944.
The applicant has found that one can obtain

oxyhydrophenanthrenecarboxylic acids or their derivatives, when reacting the 1-ketohydrophenanthrene% enl¯.ques compounds which carry in position 2 a hydrocarbon residue as well as a functional derivative of a carboxylic group and in position 7 a free phenolic hydroxyl group or a substituent capable of being transformed into a group of this kind, with an appropriate organometallic compound allowing the introduction of a hydrocarbon residue in position 1, which the new tertiary hydroxyl group is removed directly or indirectly,

converting, where appropriate, the functional derivative of the carboxylic group in position 2 to a free carboxylic group and / or the substituent in position 7 to a free hydroxyl group and hydrogenating the multiple, non-aromatic bonds between atoms of carbon, in any phase of the process, after the reaction with the organ-metallic compound.
As initial products, we will use compounds

derived from 1.2.3.4tetrahydrophenanthrene, 1.2.3.4.9.10hexahydrophenanthrene or octohydrophenanthrene. These compounds contain in position 1 a ketone group and in position 2 a hydrocarbon residue, for example a residue

or with an inorganic acid, a group: T02 '};} {2 or a halogen atom. These initial compounds can have any sterile configuration and carry still other substituents.
In order to introduce hydrocarbon residues, for example methyl, ethyl, allyl, propyl, benzyl or ethylene in position 1, the initial compounds are first reacted with suitable organ-metallic compounds, for example with organo-metal halides. magnesians

or organo-zinc, with alkali metal alkyl or alkaline compounds or analogous compounds. In the tertiary alcohols thus obtained, the tertiary hydroxyl groups formed are then cleaved directly or indirectly, where appropriate after having hydrogenated the multiple bonds between carbon atoms of side chains. This cleavage can be carried out with the formation of a double bond between carbon atoms.
It is caused by splitting water directly or by replacing the hydroxyl group first of all with a halogen atom or with an ether-salt or etheroxide residue. A hydroxyl group thus transformed can then be removed, for example by heat cleavage or by treatment with agents capable of cleaving acids or alcohols from these compounds. The cleavage of the tertiary hydroxyl groups formed by the treatment according to Grignard mentioned above can also, where appropriate, take place directly by causing the Grignard agent to act at high temperature.
Removal of the hydroxyl group or the halogen atom can also be carried out by reduction, for example with hydrogen, in the presence of a catalyst containing a noble metal or also by reduction with hydriodic acid in presence of glacial acetic acid. The conversion of the functional derivatives of the carboxylic group in position 2 into a free carboxylic group and / or of the substituents in position 7 into a free hydroxyl group takes place, in particular for the ether-salt and ether-oxide groups, by the action of agents hydrolysers. When dealing with a benzyl ether oxide, reducing agents can, for example, also be employed.
When these two kinds of substituents in position 2 and 7 are converted into a free carboxylic or hydroxyl group,
<Desc / Clms Page number 4>
one can also carry out this transformation at different stages and in any order.
To saturate multiple bonds between non-aromatic carbon atoms, formed in particular by scission of a tertiary hydroxyl group or by reaction of unsaturated organometallic compounds having double or triple bonds between carbon atoms, the compounds formed can be hydrogenated, after the reaction with organometallic compounds, in any phase of the process. For this purpose, for example, chemical processes, such as catalytic or electrolytic hydrogenation and nascent hydrogen, or also biochemical processes will be employed.
When an unsaturated residue has been introduced, the multiple bond between carbon atoms of this residue, as well as the double bond between carbon atoms, formed by scission of the tertiary hydroxyl group, can also be hydrogenated in two different reaction stages. It is in particular possible to saturate with hydrogen first of all the multiple bond introduced with the unsaturated residue, then to split the tertiary hydroxyl group, which forms a double bond, and then to hydrogenate this double bond.
The compounds obtained having a free carboxylic group can be esterified, for example, directly with diazomethane or diazoethane, or indirectly through the acid chloride or through the alkali metal salts of the carboxylic acid. Finally, it is possible to esterify or etherify the phenolic hydroxyl groups in a known manner.
Mention will be made here of esters with organic acids, in particular aliphatic and aromatic acids, for example with
<Desc / Clms Page number 5>
acetic, propionic, butyric, palmitic, stearic and benzdic acids, or with sulfonic, polycarboxylic, or sulfocarboxylic acids, and finally with inorganic acids, for example with sulfuric, phosphoric and carbonic acids and their derivatives Among the ethers-oxides , mention will in particular be made of glycosides.
The free carboxylic acids can be converted into salts, for example alkali, alkaline earth, or ammonium salts. When the ether-salt residue or the etheroxide residue in position 7 contains basic groups, the corresponding salts can also be prepared with inorganic acids, for example also the salts of betain ether-salts. These salts make it possible to use these compounds in particular in aqueous solutions or for therapy with formation of deposits.
The products of the present invention have surprisingly very high estrogenic action both when administered parenterally and orally.
The following examples illustrate the present invention without however limiting it. Unless otherwise specified, the parts indicated are by weight.
