Abstract
De huidge uitvinding heeft betrekking op een hippuurzuurderivaat en een analysemethode bestemd om de eventuële aanwezigheid van groeistimulatoren in het bijzonder van het type beta-agonisten, alsook beta-antagonisten in een (gespikt) urine-, vlees- of bloedmonster van een dier of mens op te sporen. Het betreft ook een vulmateriaal om de analysemethode toe te passen bij vleeskontroles en anti-dopping-testen. Het vulmateriaal is een hippuurzuurderivaat dat de volgende formule bezit : (zie de fig) waarin R6 = NH2, OH, CONH2,SO2NH2;R7 en/of R8 = C1, Br, OH, CH2OH, CF3, NO2, F;R9 = alkyl in C2-C6 or alkylaryl in C7-C9, CH2OH, CH2SH, (CH2)nSMe, (CH2)nCONH2, (CH2)nCOOH, (CH)nNH2, (CH2)nNHC(NH)NH2, CH2-O-OH en (CH2)nNH2, Y = CO of SO2.
Description
Analysemethode geschikt om bêta-aoonisten en bêta-antaaonisten on te sporen
De huidge uitvinding heeft betrekking op een verbinding en een analysemethode bestemd om de eventuële aanwezigheid van /?-agonisten, alsook /3-antagonisten in een (gespikt) urine-, vlees- of bloedmonster van een dier of mens op te sporen.
Het betreft ook een vulmateriaal om de analysemethode toe te passen bij vleeskontroles en anti-doping-testen.
Bij de vetmesting van rundvee wordt al jaren gebruik gemaakt van groeistimulatoren. De meest bekende zijn anabole steroïden waarvan het gebruik sinds 01.01.1988 in alle E.E.G. lidstaten verboden is.
Ook worden andere groeibevorderaars aangetroffen in de vetmesting, in het bijzonder jS-agonisten en /3-mimetica met Clenbuterol als best gekende vertegenwoordiger.
Deze stoffen en soortelijke verbindingen werden oorspronkelijk toegediend bij de behandeling van astmatische aandoeningen, als bronchospasmolyticum voor het verwijderen van de bronchitis bij varkens en vee en als uterusrelaxans bij dreigende abortus.
Maar omdat deze stoffen ook de eigenschap bezitten het vet en spiermetabolisme te beïnvloeden en een betere karkaskwaliteit mee te brengen, hebben zij onmiddellijk de interesse en de winstzucht van de veehouders gewekt.
Omdat de biochemische werking schijnbaar via hetzelfde mechanisme verloopt als voor twee welgekende hormonen adrenaline en noradrenaline het geval is, kunnen jS-agonisten beschouwd worden als stoffen met hormonale werking.
Het gebruik van (3-agonisten ter verbetering van de vleeskwaliteit is nu illegaal binnen de E.E.G.
Aangezien de variëteit binnen de groep van de ß-agonisten relatief groot is (functionele groepen, zuur-base eigenschappen), is men er tot nu toe nog niet in geslaagd een snelle monsteropzuiveringsmethode te ontwikkelen die in staat is stoffen van de ß-agonisten groep die allen aanverwante eigenschappen bezitten voldoende snel te concentreren en te identificeren. Een stap in de goede richting zijn de recent beschreven immuno-affiniteitskolommen, maar deze zijn enkel specifiek voor de N-tertiair-butyl, isopropyl en 2-pentyl groep. Toch schijnt een gemeenschappelijke basisstructuur te bestaan in alle ß-agonisten, namelijk de fenylethanol amine structuur die de volgende formule bezit :

De gemeenschappelijke fenylethanolamine-groep, die steeds aanwezig is, is tevens het farmacologisch actief fragment van de ß-agonisten.
Naarmate steeds meer ß-agonisten (tabel 1) gesynthetiseerd werden is de gelijkenis met de oorspronkelijke cathecholamines steeds geringer.

Het technisch probleem bestaat erin een methode te ontwikkelen die geschikt is om selectief fenylethanol-amines, die de basis vormen van elke /3-agonist, op een snelle manier op te sporen en op te zuiveren.
/3-agonisten zijn potentieel aanwezig in urine van slachtdieren.
De huidige uitvinding stelt een affiniteits-methode voor, welke in staat is fenylethanoamines op een snelle manier te scheiden en te detecteren.
Volgens een bijzonderheid van de uitvinding wordt in de affiniteitschromatografische methode gebruik gemaakt van een chemisch ligand dat in staat is ß-agonisten te binden.
Als mogelijk ligand wordt 4-amino-3,5-dichloor-benzoylglycine (A.D.B.G.) vooropgesteld voor de opzui-vering van /3-agonisten.
In een voorkeursvorm, wordt A.D.B.G. door middel van resorcinol als spacer aan een klassiek Epoxy-activated Sepharose kolommateriaal gekoppeld. Het materiaal bezit een zeer goede affiniteit voor de geteste ß-agonisten, met uitzondering van salbutamol.
De uitvinding betreft ook een werkwijze om 4-amino-3,5-dichloorbenzoylglycine te synthetiseren en te koppelen met spacermoleculen om een efficiënte koppeling aan een affiniteitsmatrix tot stand te brengen.
Als mogelijke liganden voor affiniteitschromatograf ie van /3-agonisten, worden volgende verbindingen vooropgesteld :

en (CH2) nNH2.
Controles van gespikte urine en urinemonsters met behulp van affiniteitschromatografie geven uitstekende resultaten voor de verdere toepassing van dit ligand bij de toepassing van de methode voor de residu analyse van zowel bekende als onbekende j8-agonisten.
Uit initiële testen van affiniteitschromatograf ie is gebleken dat CNBr-Activated Sepharose 4B, Epoxygeactiveerde silica of Sepharose en Glutaraldehyde-P-Silica kolommaterialen wanneer zij rechtstreeks met A.D.B.G. gekoppeld zijn geen affiniteit voor de geteste /3-agonisten vertonen. Mogelijke oorzaken kunnen de afscherming van de ligand door de drager zijn.
De affiniteit wordt verbeterd door een extra spacermolecule in te voeren met een functionele groep (NH2 of OH) welke makkelijk te koppelen is aan de kolom-matrix.
Goede resultaten worden bekomen met een aromatische spacer. Fenol, resorcinol of aniline wordt vooraf aan een geschikte kolommatrix gekoppeld. Daarna worden de gediazoteerde A.D.B.G. moleculen op deze gekoppelde aromaten gebonden. Bijkomend voordeel van deze methode is dat er bij de vorming van de azobenzeenverbinding een rode kleur ontstaat, zodat men ogenblikkelijk kan zien of er ligand gekoppeld is.
Gemiddeld werd met het kolommateriaal volgens de uitvinding met fenol- of resorcinol spacers en het 4-amino-3,5-dichloorbenzoylglycine een bezettingsgraad van 10 à 15 μιηοΐ/ml gel behaald.
A. SYNTHESE LIGAND : 1. Synthese 4-amino-3.5-dichloorbenzoëzuur en het overeenkomstig ethvlester 1.1. Bereiding van ethyl-4-amino-benzöaat
In een kolf van 1,5 1 voorzien van een magnetische roerstaaf wordt (0,73 mol) 100 g para amino-benzoëzuur (1) opgelost in 500 ml ethanol.
Bij het toedruppelen van (0,73 mol) 87 g thionylchloride wordt de oplossing gekoeld met een waterbad, waarna we het mengsel 10 uur refluxen op een oliebad.
Bij het afkoelen ontstaat een neerslag welke na toevoegen van 500 ml water terug oplost. Daarna wordt in kleine hoeveelheden 81 g natriumcarbonaat toegevoegd. Na uitkristallisatie wordt de neerslag afgefiltreerd en uit het filtraat na indampen een tweede worp gehaald.
MW * 165 g rendement = 118,0 g = 98% 1.2. Bereiding van ethyl-4-amino-3,5-dichloorbenzöaat
In een 3 1 kolf met 900 ml tetrachloormethaan wordt (0.7 mol) 115.5 g ethyl-4-amino-benzöaat (2.) opgelost.
Na toevoegen van (1,4 mol) 198 g sulfuryl-chloride wordt gedurende 15 uur gerefluxt op een oliebad.
Na de reactie wordt in een scheitrechter 500 ml water toegevoegd. Na afscheiden van de tetralaag wordt deze gedroogd met magnesiumsulfaat en ingedampt tot ongeveer 150 ml. Hieraan wordt 100 ml hexaan toegevoegd en gekoeld bij -10°C zodat het ethyl-4-amino-3,5-dichloorbenzöaat (2) uitkristalliseert.
MW = 233,9 g rendement = 95,0 g = 58% 1.3. Verzepen van ethyl-4-amino-3,5-dichloorbenzöaat
Van het bekomen ethylester (2) wordt (0.36 mol) 85 g opgelost in een 1 1 kolf met een mengsel van 480 ml ethanol en 120 ml water met 34 g natriumhydroxide.
Het mengsel wordt gedurende drie dagen gere-fluxt op een oliebad.
Na de reactie wordt een mengsel van 110 ml water met 90 ml geconcentreerd zoutzuur (11M) toegevoegd. Hierbij ontstaat een witte neerslag. Dit wordt een weinig opgewarmd tot bijna alles opnieuw oplost, daarna laten we het mengsel overnacht bij kamertemperatuur uitkristalliseren.
Na affiltreren wordt zuiver ethyl-4-amino-3,5-dichloorbenzöaat gedroogd in de droogstoof.
MW = 205,9 g rendement = 75,1 g = 96% 2 . Synthese van 4-amino-3.5-dichloorbenzovlalvcine en het overeenkomstige methvlester 2.1. Bereiding van 4-amino-dichloorbenzoylchloride
In een 1 liter kolf, voorzien van een bolkoeler met Cad,- buis, wordt (0,1 mol) 2 0,6 g 4-amino-3,5-dichloorbenzoëzuur (4) afgewogen en 10 minuten lichtjes gerefluxt in 300 ml dichloormethaan zodat alles goed oplost.
Daarna wordt (0,3 mol) 36 g thionylchloride toegevoegd en gedurende 5 uur gerefluxt, totdat een heldere oplossing bekomen wordt.
Watervrij milieu is noodzakelijk om hydrolysen van het zuurchloride (5) tot het carbonzuur (4.) te voorkomen .
Bij het gebruik van thionylchloride wordt door een bijreactie ook 4-thionylimino-3,5-dichloorbenzoyl-chloride gevormd. Ongeveer 50% van de aminofunctie wordt in de thionyliminovorm omgezet. Dit percentage wordt groter naarmate langer gerefluxt wordt. In waterig milieu ontstaat terug het vrije amine (5.) .
Het reaciemengsel wordt volledig drooggedampt met behulp van een rotavapor en als dusdanig verder gebruikt.
2.2. Bereiding van 4-amino-3,5-dichloorbenzoylglycine
Het ruw zuurchloride (£) , (0,1 mol) uit de vorige stap wordt opgelost in 300 ml dichloormethaan.
Hieraan wordt een oplossing, van (0,45 mol) 33,75 g glycine en (0,45 mol) 18 g natriumhydroxide in 150 ml water, toegedruppeld. De vrije aminogroep van glycine reageert met het zuurchloride via een nucleofiele additie en vormt 4-amino-3,5-dichloorbenzoylglycine (6).
Het reactiemengsel wordt, in een kolf met luchtkoeler, gedurende een ganse nacht bij kamertemperatuur geroerd.
Na reactie wordt de waterlaag met geconcentreerd zoutzuur op pH = 3 gebracht. Na affiltreren van de neerslag bekomen we 20,72 g ruw product.

Als nevenreactie, wordt door de additie van water ongeveer 10% van het oorspronkelijk zuur (4.) gevormd.
Door omkristallisatie in 45 ml opgewarmd azijnzuur kan 4-amino-3,5-dichloorbenzoylglycine (6) opgezuiverd worden. Het filtraat laat men langzaam uitkristalliseren, na affiltreren en wassen met een kleine hoeveelheid azijnzuur bekomen we een 100% zuivere verbinding.
MW = 263 g rendement (2 stappen) = 15,6 g * 58% 2.3. Bereiding van 4-amino-3,5-dichloorbenzoylglycine methylester
Analoog als voor de bereiding van 4-amino-3,5-dichloorbenzoylglycine wordt het ruwe zuurchloride (.5) , bekomen uit 0,1 mol 4-amino-3,5-dichlorobenzoëzuur (4.) opgelost in een 1 1 kolf met 200 ml dichloormethaan.
Een oplossing, van (0,2 mol) 25,1 g glycinehy-drochloride methylester in 150 ml dichloormethaan en (0,3 mol) 30,3 g triethylamine, wordt aan het zuur-chloride toegedruppeld en overnacht geroerd bij kamertemperatuur .

Triëthylamine wordt toegevoegd om het zout zuur van het glycinehydrochloridemethylester te binden, zodat de vrije aminofunctie een nucleofiele additie kan uitvoeren op de carbonylgroep van het zuurchloride.
Na reactie wordt het triëthylaminehydrochloride en de overgebleven vrije base met 120 ml water geëxtraheerd .
De afgescheiden organische laag wordt gedroogd met magnesiumsulfaat. Na uitkristallisatie wordt de dichloor- methaanlaag afgefiltreerd. Het filtraat wordt ingedampt terwijl 80 ml tetrachloormethaan toegevoegd wordt, hieruit wordt een tweede worp gehaald.
MW - 277 g rendement (2 stappen) * 19,7 g = 69% B. KOPPELINGSREACTIES LIGAND MET SPACER : 1. Synthese van 6-trifluoracetvlamine-l-hexanoylchloride
Het uiteindelijke doel is een chemisch ligand te vormen door 6-aminohexaanzuur (8) te koppelen met het aromatisch aminesubstituent van 4-amino-3,5-dichloor-benzoylglycine methylester (7).
Daar de koppeling van de aminofunctie van 4-amino-3,5-dichloorbenzoylglycine met de kolommatrix moeilijk verloopt worden spacers met vrije actieve groepen vooraf aan de aromatische aminofunctie gekoppeld. Om ringsluiting, met vorming van caprolactam tot gevolg, te vermijden wordt de aminofunctie van de alifatische keten (8.) vooraf beschermd met een later terug af te splitsen trif luoracetylgroep. (13.) 1.1. Bereiding van 6-trifluoracetylamine-hexaanzuur
In een 500 ml kolf wordt (0,15 mol) 19,8 g 6-aminohexaanzuur (8) opgelost in 300 ml dichloormethaan, samen met (0,16 mol) 13,43 g pyridine.
Hieraan wordt (0,20 mol) 42 g trifluor-azijnzuuranhydride toegedruppeld en gedurende 5 uur geroerd bij kamertemperatuur.
Deze oplossing wordt bij kamertemperatuur geroerd tot een heldere oplossing bekomen wordt, die als dusdanig verder gebruikt wordt.
Als nevenreactie wordt een deel van het carbonzuur (9) omgezet tot het gemengd anhydride, wat later zeer snel in het zuurchloride (i0) omgezet wordt.
1.2. Bereiding van 6-trifluoracetylamine-hexanoylchloride
Aan de hierboven bekomen oplossing van hexa-anzuur met beschermde aminogroep (9) wordt (0,25 mol) 19,8 g thionylchloride toegevoegd en gedurende 30 minuten lichtjes verwarmd in oliebad van 50 °C.
De kolf wordt hiervoor voorzien van een bol-koeler en een CaCl2-buis.
Het mengsel wordt overnacht verder geroerd bij kamertemperatuur en daarna volledig drooggedampt met de rotavapor.
Gebaseerd op 1JC-NMR vinden we dat ongeveer 85% van het carbonzuur in het zuurchloride werd omgezet.
2. Synthese van 4-N-fe-amino-l-carbonvlpentvl)-3,5-dichloorbenzovlalvcine 2.1. Bereiding van 4-N-(l-carbonylpentyl-6-trifluor- acetylamine)-3, 5-dichloorbenzoylglycinemethylester
In een 500 ml kolf wordt (0,05 mol) 13,8 g 4-araino-3,5-dichloorbenzoylglycine methylester (2) en (0,17 mol) 13,4 g pyridine opgelost in 250 ml dichloormethaan, waarbij gedurende 30 minuten lichtjes opgewarmd wordt.
Het ingedampte ruw zuurchloride (20), bekomen uit 0,15 mol 6-aminohexaanzuur, wordt opgelost in 100 ml dichloormethaan en aan de methylester (2) oplossing toegedruppeld. Het mengsel wordt gedurende 5 dagen geroerd bij kamertemperatuur, na 24 uur verschijnen de eerste fijne witte kristallen (.11) .
De bekomen oplossing wordt in een scheitrechter gewassen met 200 ml 0,5 N zoutzuur. Hierbij ontstaat een kleverige emulsie welke afgefiltreerd wordt en goed gewassen met dichloormethaan.
MW = 476 g rendement = 17,4 g = 73% 2.2. Bereiding van 4-N-(6-amino-l-carbonylpentyl)-3,5-dichloorbenzoylglycine
In een 250 ml kolf wordt (0,075 mol) 3 g natriumhydroxide opgelost in 90 ml methanol.
Hieraan wordt (0,03 mol) 14,28 g 4-N-(l-carbo-nylpentyl-6-trifluoracetylamine)-3,5-dichloorbenzoyl-glycine methylester (11) toegevoegd en gedurende 30 minuten verwarmd in een oliebad van 60°C. Daarna wordt gedurende twee dagen geroerd bij kamertemperatuur.
Na reactie wordt 50 ml water toegevoegd en wordt de pH van het mengsel op 5,5 gebracht.
Met de rotavapor wordt een deel van de methanol uitgedampt tot de eerst fijne kristallen (12.) verschijnen. We laten de oplossing overnacht verder uitkristalliseren en verwezenlijkt de koppeling van 4- amino-3,5-dicloorbenzoylglycine met een alifatische C6-spacer.
MW = 366 g rendement = 9,1 g = 83% 3. Synthese van 2.6-dichloor-4-carbonvlaziinzuur-M^hvdroxv of 2 *.4'-dihvdroxv)-azobenzeen
Op analoge wijze wordt een diazokoppeling van 4-amino-3,5-dichloorbenzoylglycine met een aromatische spacer verwezenlijkt.
In 100 ml erlenmeyer wordt (0,48 mmol) 125 mg 4-amino-3,5-dichlorobenzoylglycine afgewogen en opgelost in 5 ml koud demi-water (pH > 7). Daarna wordt de pH met geconcentreerd zoutzuur op 1 à 1,5 gebracht, waarbij een fijne suspensie ontstaat.
De oplossing wordt afgesloten van het licht in een ijsbad geplaatst. Terwijl geroerd wordt voegen we 4 ml (1,5 N) natriumnitriet toe, de pH wordt regelmatig gecontroleerd en op 1 à 1,5 gehouden.
Na 30 minuten wordt de oplossing gefiltreerd, aan het gele filtraat wordt voorzichtig 2 g ammonium-sulfamaat, opgelost in 2 ml demi-water, toegdruppeld. Dit totdat er geen belletjes meer gevormd worden. (1 uur, 4°C, donker)
Als test worden enkele druppels van de actieve diazoverbinding gemengd met enkele druppels dimethyl-N-aniline, er moet onmiddellijk een diep rode kleur verschijnen.
De hier bekomen diazo-oplossing wordt gebruikt voor de koppeling aan een kolommatrix welke vooraf met benzeenringen bezet werd. (zie deel IV)
Na de koppeling aan de kolommatrix wordt de 5 resterende actieve diazoverbinding afgef iltreerd. Aan het bekomen filtraat wordt, (1 mmol) 94 mg fenol of (lmmol) 110 mg resorcinol opgelost in 5 ml koud demi-water pH = 1,5 toe- gevoegd en gedurende 15 uur in de ijskast geplaatst.
) De ontstane rode neerslag wordt na affiltreren en wassen met demiwater pH = 1,5 gebruikt voor de karakterisatie.
C. KOPPELING LIGAND AAN KOLOMMATRIX
Voor het koppelen van de aminofunctie van het ligand wordt volgens de uitvinding een kolommatrix op agarose basis nl. sepharose en een silicamatrix gekozen.
De sepharose affiniteitsmatrixen hebben een open poriën structuur, zodat binnenin de matrix plaats vrij is voor het vasthechten van (grote) ligandmoleculen.
Even belangrijk zijn de extreem lage non-specifieke interacties van de matrix zelf en de korrelvormige structuur die zorgt voor de goede floweigenschappen.
Sepharose-matrixen zijn in de handel verkrijgbaar met verschillende soorten actieve groepen, al dan niet met spacer, waaraan aminogroepen onder milde condities kunnen gekoppeld worden.
1. Koppeling aan Epoxv-activated Sepharose 6B
1.1. Reactiemechanisme
Deze kolommatrix is standaard voorzien van een lange (12 atomen) spacer eindigend met een epoxygroep en is uiterst geschikt voor affiniteitschromatografie met kleine ligandmoleculen.
Door nucleofiele additie van een hydroxylgroep of een aminofunctie aan de reactieve epoxygroep wordt een zeer stabiel ringopeningsprodukt gevormd.
1.2. Koppelingscondities
Nadat 1 g gevriesdroogde kolommatrix gewassen werd met 100 ml demi-water, moet de koppelingsbuffer ommiddellijk aan het gel toegevoegd worden. Anders worden de epoxygroepen vooraf gehydrolyseerd.
Voor de koppeling bij 55°C werd het te reageren mengsel in een erlenmeyer in een droogstof geplaatst en regelmatig omgeschud.
Na de koppelingsreactie wordt alternerend gewassen met in totaal 600 ml 0,1 M natriumhydroxide buffer (pH=ll) en 0,1 M acetaat buffer (pH=3) welke beide 0,5 M NaCl bevatten.
1.3. Titratiecurve
Zoals afgebeeld in figuur 1, tonen de vier titratiecurves met Epoxy-activated Sepharose 6B aan dot de rechtstreekse koppeling via het aromatisch amine is niet mogelijk is. Ook de koppeling via een fenol spacer is weinig efficiënt en bindt ongeveer 5 μπιοί gebonden A.D.B.G mole- culen per ml gel.
Via een alifatische spacer is het mogelijk 10 Aimol/ml gel van het A.D.B.G. te koppelen.
Het beste resultaat wordt bekomen met resorcinol of aniline als spacer, waarbij 15-20 /zmol/ml gekoppeld werd.
1.4. NMR-spectroscopie
Ongeveer 1 ml van het, via de resorcinol spacer met gediazoteerd 4-amino-3,5-dichloorbenzoylglycine A.D.B.G., gekoppelde epoxy-sepharosegel wordt in een NMR buis gebracht. (90% H20, 10% d20, pH=10)
Het ‘H-NMR spectrum van dit gel ziet u in addendum 3.
Als we dit spectrum vergelijken met het vroeger opgenomen 2,6-dichloro-4-carbonyl-azijnzuur-2', 4'-dihydroxy-azobenzeen kunnen we aan de hand van de aromatische waterstoffen van A.D.B.G. en de 4 zichtbare (beweeglijke) alifatische waterstoffen van de spacer berekenen dat ongeveer 30% van de spacers (12 atomen) met A.D.B.G. bezet is.
D. VOORBEELDEN
1. Voorbeeld 1 : elutiepatroon van standaardoplossing (H20) met als ß-agonisten (salbutamol, fenoterol, cimbuterol, Clenbuterol, mabuterol, clenpenterol) en als ß-antagonist (propanolol) voor de Epoxy-Activated sepharose met 4-amino-3,5-dichloorbenzoylglycine als chemisch ligand gekoppeld via resorcinol-spacer.
De beste resultaten worden bekomen met resorci-nol als aromatische spacer (fig. 2). Hierbij kunnen alle teststoffen op de kolom vastgehouden worden bij spoelen met de spoelbuffer, met uitzondering van salbutamol dat een te zwakke affiniteit vertoont.
Cimbuterol en 2,4-aminophenylethanolamnine (2.4.A.P.E.A.) vinden we voor 100% terug in de 55mM zout fractie, ook voor mabuterol wordt de affiniteit bij deze zoutconcentratie verbroken. Voor Clenbuterol gebeurt dit pas bij een spoelen met 110 mM zout. Clenpenterol vinden we slechts voor 25% in deze fractie terug, wat op een sterkere affiniteit wijst. De sterkste interacties treden op voor fenoterol en propanolol, die voor het grootste deel in de 300mM zoutfractie terug te vinden zijn.
Deze kolom werd tevens getest met een 1 mM acetaat spoelbuffer (pH=5,2), waarbij de OH groep van de aromatische spacer gedeprotoneerd is. Ook hier werd hetzelfde verloop teruggevonden.
2. Voorbeeld 2 : het zelfde kolommateriaal als in voorbeeld 1 wordt ditmaal getest met gespikte kalverurine.
Hiervoor wordt 1 ml blanco kalverurine gemengd met lml van het standaard mengsel beschreven in voorbeeld 1 en off-line op de lange Epoxy-resor-A.D.B.G. kolom gebracht.
Daarna wordt gespoeld met 20 ml spoelbuffer (ImM fosfaat, pH=7), gevolgd door 20 ml elutiebuffer met traag stijgende zoutconcentratie (11 mM, 33 mM, 55 mM, 110 mM, 330 mM en 550 mM).
In de grafiek (fig.4.) zien we dezelfde elutievolgorde terug als voor een waterig mengsel. De teststoffen worden reeds na toevoegen van lagere zoutconcentraties geëlueerd.
Dit kan verklaard worden door het reeds aanwezige zout in de urine. Tijdens de spoelstap van 20 ml elueren histamine, 2.4.A.P.E.A., salbutamol en een deel van de toegevoegde cimbuterol van de kolom. Clenbuterol, fenoterol, mabuterol, clenpenterol en propanol blijft op de kolom.
Claims (9)
2. Toepassing van het hippuurzuurderivaat volgens conclusie 1, in een analysemethode bestemd om stoffen in het bijzonder van het type ß-agonisten en ß-antagonisten in urine-, vlees- of bloedmonsters op te sporen.
3. Analysemethode bestemd om groeistimulatoren in het bijzonder van het type ß-agonisten alsook ß-antagonisten in urine-, vlees- of bloedmonsters op te sporen, met het kenmerk dat men het monster op een affi-niteitsschromatografiekolom brengt met de aromatische meermaals gesubstitueerd hippuurzuurderivaat volgens conclusie 1 als chemisch ligand.
4. Analysemethode volgens conclusie 1, met het kenmerk dat het hippuurzuurderivaat uit 4-amino-3,5-dichloorbenzoylglycine bestaat.
5. Vulmateriaal voor affiniteitschromato-grafie-kolom bestemd voor de analysemethode volgens conclusie 1, met het kenmerk dat het uit een hippuur-zuurderivaat volgens conclusie 1 bestaat als chemisch ligand dat op een klassieke kolommatrix met behulp van een aromatische of alifatische verbinding als spacer wordt gebonden.
6. Vulmateriaal volgens conclusie 4, met het kenmerk dat het hippuurzuurderivaat uit 4-amino-3,5-dichloorbenzoylglycine bestaat.
7. Vulmateriaal volgens conclusie 2, met het kenmerk dat 4-amino-3,5-dichloorbenzoylglycine op een Epoxygeactiveerde silica of Sepharose drager met behulp van resorcinol of fenol als spacer wordt gebonden.
8. Werkwijze om een vulmateriaal te vervaardigen volgens een der conclusies 5 tot en met 7, met het kenmerk dat van fenol of resorcinol gediazoteerd 4-amino-3,5-dichloorbenzoylglycine op deze gekoppelde aromaten bindt.
9. Werkwijze om 4-amino-3,5-dichloorbenzoylglycine te synthetiseren uit ethyl-4-aminobenzöaat dat met sulfurylchloride gedurende 15 uur met reflux opgewarmd wordt op een oliebad en uit hexaan uitgekristalli-zeerd en het bekomen ethyl-4-amino-3,5-dichloorbenzöaat met thionylchloride gedurende 5 uur met reflux opwarmt.
