Chinese Herbal Medicine for Treating Gout and Preparation Method thereof
Technical Field
The present application discloses a Chinese herbal medicine for treating gout, and meanwhile provides a method of preparing the Chinese herbal medicine, belonging to the technical field of traditional Chinese medicine pharmacy.
Background Art
Gout is caused when purine metabolism disorders occurs in a body, specifically, excessive purine is oxidized to generate uric acid, which cannot be excreted through kidney and then through urine, but flows to connective tissues again through blood, and is deposited therein in a form of amino salt crystals, and when there is an inducement, the uric acid crystals deposited in soft tissues and articular membrane will be caused to release, leading to sensitive response of a body’s immune system, causing inflammation, and resulting in gout.
At present, there are a lot of medicines and methods for treating the gout. For example, Chinese herbal medicine decoction, Chinese patent medicines, western medicines, medicinal tea, instrument treatments and other methods and medicines. However, the number of treated and new-found patients is not reduced, but increases year by year.
According to clinical researches and analysis, it is considered that current dosage forms and methods have the following problems: I. There still lacks of radical cure:
All of the existing medicines and methods neglect to enhance an anti-oxidation capacity of the body, and to use an intervention anti-oxidation function to prevent or decrease purine oxidation, thus it cannot solve, from the source, the problem of slow acting and long treatment period when curing gout radically. II. Use of incorrect medicines and methods for the treatment will cause drug-induced injuries.
Since the uric acid is incorporated in the blood, neutralizing the uric acid in the blood is an effective measure for retarding and preventing the uric acid deposition. Among the existing western medicines, for example, colchicine and other western medicines have a function of neutralizing the uric acid, but bring great drug-induced injuries. Chinese herbal medicines also can neutralize the uric acid, but it acts slowly, and the efficacy thereof is unnoticeable. Only a fast, 1
P01651314BJ
P01651314BJ 2016102079 05 Dec 2016 effective, safe and reliable acidic substance neutralizer can solve the problem of slow acting, unnoticeable efficacy and other problems. III. The existing medicines and methods cannot radically cure gout
The gout is featured by long duration, repeated attack, no radical cure etc. The current medicines and methods lack the conceptual foundation of “great care is better than cure”, and it is insufficient to merely take measures of improving the immunologic function. Only to stimulate the function of a “self-healing rehabilitation system” of the human body is a primary measure for fundamentally curing the gout. IV. There lacks of comprehensiveness, systematicness, harmony, and synchronicity
The gout involves bones, nerves, blood circulation, urination, endocrine secretion and other systems, and spreads to heart, lungs, liver, spleen, kidneys and other internal organs. For example, the gout will cause osteoporosis, and complications will cause cardiovascular diseases and then induce kidney failure, sudden death etc. Simple protection is insufficient. Some Chinese herbal medicines have the efficacy of dissolving the uric acid crystals, but will cause increased uric acid concentration in the blood, and spread to heart and kidney sudden illnesses, and produces hazards. Therefore, there must be a measure for recuperating depleted yang and rescuing a patient from collapse while protecting the heart and kidneys, which is an important prevention measure.
Disclosure of the Innovation
The present application provides a Chinese herbal medicine for treating gout, wherein the purine oxidation and the purine synthetase activity are inhibited from the source comprehensively, systematically, harmoniously, and synchronously, and measures for dissolving urate crystals, neutralizing the uric acid in the blood, preventing and treating sudden change of disease state and so on are used; both yin and yang are replenished to improve the positive energy of the body, reinforce the immunologic function of the body, and stimulate the capacity of the “self-healing rehabilitation system” of the body itself, so as to achieve the noticeable effect of curing the gout radically.
The present application further discloses a method of preparing the Chinese herbal medicine for treating gout, applicable to industrial production.

Clinical analysis of the present application: an effective ingredient flavone in the used cudweed herb dissolves urate crystals and flows back into the blood with the blood circulation. According to this characteristic, the complex calcium 3
P01651314BJ
P01651314BJ 2016102079 05 Dec 2016 is used as a uric acid neutralizer, to neutralize the uric acid in the blood and the acid environment of the body, to form acid-base balance, achieving comprehensiveness of dissolution and neutralization.
When the urate crystals are dissolved, amount of the uric acid in the blood will increase. In order to prevent occurrence of coronary heart diseases, diabetes, hypertension, kidney failure complications, effective ingredient of the Aconitum Carmichaelii is used to prevent sudden change of disease state, and a plurality of natural Chinese herbal medicine materials for protecting the functions of the heart, liver, spleen, lungs, and kidneys are used, so as to strengthen yang energies of the body, and prevent and relieve occurrence of complications.
Since the illness results in loss of enormous energies of the body, a lot of nutrient substances should be supplemented while enhancing the body function. The materials of the prescription comprise polysaccharides, amino acids, vitamins, microelements and so on, so as to compensate for consumption, nourish yin and replenish deficiency. Replenishing both yin and yang at same time improves the positive energies of the body.
Therefore, only replenishing of both yin and yang at same time can radically cure gout comprehensively, so as to solve the problem of slow acting and drug-induced hazards caused by the current methods and medicines.
In the prescription of the present application, using Aconitum Carmichaelii for recuperating the depleted yang and rescuing a patient from collapse is an effective and scientific method used according to the pathological characteristics of the gout. The usage of complex calcium to neutralize the uric acid is also unique and novel about the present application.
By the present application, the functions of spleens and stomach are enhanced, and the capacities of absorbing, using, transporting and converting effective ingredients are improved; the functions of the endocrine system and urinary system are strengthened and the metabolic capacity of the body is improved; the artificial tiger bone is used to strength and invigorate bones, and by cooperating with the amino sugar, to repair bone joints and cartilage tissues; the complex calcium is used to nourish the bones and treat osteoporosis caused by decalcification of the bone system. The function of the respiratory system is improved by nourishing and replenishing the lungs, to increase a perfusion volume of the blood circulation. The sudden change of disease state is prevented by strengthening the heart, reinforcing the kidneys, and protecting the liver. The gout is radically cured systematically, and the problems of slow acting, long period of treatment and so on of the current methods and medicines are solved. If the disease state suddenly changes and the degree of neutralizing the uric acid is insufficient, the proportion may be adjusted by increasing the amount of the complex calcium, which follows the principle of combining actual treatment effect with actual disease situation, and achieves 4
P01651314BJ
P01651314BJ 2016102079 05 Dec 2016 harmony of combining dissolution, neutralization and system functions of the body. The problems of slow acting, uncomprehensive treatment, and difficulty in radical cure of the current methods and medicines are solved. In the process of radically curing the gout, not only synchronicity of dissolution and neutralization should be taken into consideration, but also the synthetic reaction of the purine in the liver should be inhibited and the anti-oxidation function of the body should be enhanced synchronously. Such technology of radically curing gout at the source is unique and inventive, and is incomparable relative to the current similar products for treating the gout.
The advantages of the present application further lie in: repairing and treatment are synchronous; and synchronous proliferation of bone synovial fluid is synchronously increased while repairing bone joint cells and cartilage tissue cells, to reduce pains and wear of the bone joints. A prominent advantage of the present application further lie in: according to the principle of “great care is better than cure” of the Chinese medical science, a relatively large amount of the complex calcium is used to enhance the capacity of the “self-healing rehabilitation system” of the body itself in synchronization while improving the immunologic function; and the superior feature of giving consideration to both treatment and rehabilitation contributes to inventiveness and novelty of the present application.
To sum up, the present application is a novel form of product structure, with scientific prescription compatibility, reasonable combination of nutrient elements, and reasonable and feasible process technology, and combination of comprehensive treatment at the source, complete prevention, fast repairing, yang recuperation without dryness, yin recuperation without greasiness, nourishment of internal organs, and integrated body system, under the guidance of the new concept of treatment, prevention, repairing, recuperation, and nourishment. Radical cure of the gout at the source on the whole without relapse is reflected in the aspects of noticeable effects of fast acting, short treatment period, and quick healing. The used Chinese herbal medicine materials are purely natural, safe and reliable with strong affinity.
Prescription analysis of the present application: a unique, scientific, reliable, correct, and dialectical method for radically curing the gout is to combine the solution of making the purine anti-oxidantive, dissolving the urate crystals, and neutralizing uric acid in the blood from the source; to reduce swelling and to alleviate pains; to combine internal therapy and external treatment; to combine treatment and prevention of complications and sudden lesions; to combine yang reinforcement and yin replenishment; to combine treatment and repairing; to combine improvement of immunologic function and enhancement of the capacity of the “self-healing recovery system” of the body itself, under the principle of comprehensiveness, systematicness, harmony, and synchronicity. 5

In the prescription: the dried rehmannia has the functions of strengthening the heart, lowering the blood pressure, and lowering the blood glucose. The dried rehmannia has the efficacies of improving and enhancing kidney functions and reducing fatality rate. The dried rehmannia has anti-inflammatory and antibacterial functions against acute inflammation, and has sedation function which increases sleep and relieves pain. The dried rehmannia can promote transformation of lymphoblasts of the body to increase the number of T lymphocytes and enhance the function of phagocytes, and especially has more noticeable effect for immunocompromised patients. Aconitum Carmichaelii has a relatively good heart strengthening function; has the function of tonifying fire and helping yang, to recuperate depleted yang and rescue a patient from collapse for kidney failure and prevent occurrence of uremia, thus alleviating critical illness. The gout is a kind of gouty arthritis. The Chinese medical science considers that wind and cold cause inflammation. The Aconitum Carmichaelii has the functions of eliminating cold and dispelling wind, and mainly used to treat symptoms of wind-cold-dampness arthralgia, and can be used to treat bone joint deformity caused by the gout. The efficacies of the aconitum carmichaelii for recuperating depleted yang and rescuing a patient from collapse, resisting inflammation, and alleviating pains cannot be compared by any medicines. This is the reason for using Aconitum Carmichaelii in the prescription. The toxicity of Aconitum Carmichaelii is neutralized by the liquorice. The ingredients flavone and luteolin, rich in the cudweed herb, can well dissolve the urate crystals deposited on the joints and cartilage tissues which are then gently returned back to the blood and neutralized in the blood. The cudweed herb contains flavone substances, and promotes blood circulation to remove blood stasis, reduces the blood fat, lowers the blood viscosity, eliminates fat deposition on the vascular wall and scavenges urate deposits in the kidney, so that the kidney is in a clean state. The cudweed herb also has the function of firming and repairing the bone joints. The complex calcium, generated through enzymolysis of calcium carbonate, is easy to be absorbed and used by human body. Calcium deficiency not only will cause diseases of the bone, but also will cause diseases of the body in various aspects. Using complex calcium to neutralize the uric acid to radically cure the gout is fundamental to the solution with fast acting and noticeable therapeutic effect, and is a safe, harmonious and scientific method without side effects.
To sum up, the dried rehmannia is used for comprehensive replenishment and conditioning; the artificial tiger bone meal is used to quickly reduce swelling and alleviate pains, and systematically repair the injured joints and synovia; the Aconitum Carmichaelii is used to recuperate depleted yang and rescue a patient from collapse to prevent or relieve accidental critical illness caused by complications resulted from the gout; the cudweed herb is used to dissolve the urate crystals, and promotes the blood circulation to remove the blood stasis, increase the blood flow volume, and eliminate the deposits of the 6
P01651314BJ
P01651314BJ 2016102079 05 Dec 2016 kidney and dirt on big vascular walls; the complex calcium is used to neutralize the acidic substances in the blood and the body, balance the acidity and alkalinity of the body, and treat osteoporosis. The above five materials achieve the noticeable effect of radically curing the gout comprehensively and synchronously with their own functions, according to the characteristics of pathologies, symptoms and harmfulness of the gout. Therefore, the five materials act at monarch drugs.
The purine, existing in the blood in a form of purine nucleotide crystals, is a fat soluble substance, with a molecular formula of C5H4N4. After being esterified, the purine becomes to exist in the blood in a form of liquid, and is oxidized to generate the uric acid, with a molecular formula of C5H4N4O3. But in fact, it is a kind of nitrogen-containing heterocyclic organic substance, with a molecular formula of C5(HN4)03. Therefore, after oxidation, the purine nucleotide is transformed from a liquid to solid crystals having amino groups existing in the blood, and will be deposited when confronted with obstacles (such as joints, soft tissues and so on) during the process of blood circulation. According to the above analysis of mechanism characteristics, it is one of the keys for radically curing gout at the source to enhance the anti-oxidation capacity of the body, reduce the contents of triglyceride and cholesterol in the blood and lower the blood viscosity. In the prescription: the angelica has the functions of promoting circulation of blood, nourishing the blood, strengthening the blood circulation, and accelerating the acid dissolution and excretion, has a noticeable function of alleviating pains, and noticeablely scavenges free radicals, resists lipid peroxidation reaction and inhibits oxidation reaction. The Chinese yam has noticeable functions of tonifying the spleen, nourishing the lung, and reinforcing the kidney, promotes the blood circulation, and accelerates the uric acid neutralization and metabolism, and meanwhile has a relatively good immunoregulation function and resistance to hypoxia. The Fructus Corni has a relatively good immunologic function, reduces the blood glucose, improves the survival rate of cardiac muscle cells, and prevents occurrence of coronary heart diseases and diabetes caused by the gout, and meanwhile has an anti-oxidation function and prevents the blood stored in the livers from being damaged by viruses. The Radix Glehniae has the functions of relieving arteriosclerosis and resisting lipid oxidation. The above four materials not only have the functions of resisting inflammation, relieving pains, preventing and treating coronary heart diseases and diabetes and improving the immunologic function, but also have the anti-hypoxia and anti-oxidation functions, realizing the noticeable effect of reducing purine oxidation, achieving the effect of radically curing the gout at the source, and cooperating with the monarch drugs to have unique efficacies, therefore, they are known as minister drugs.
The liver is an important organ for synthesizing purine from the beginning. The site for synthesis is cytosol, and the required substances are ribose 7
P01651314BJ
P01651314BJ 2016102079 05 Dec 2016 phosphate, glutamine and so on. The purine is gradually synthesized on the basis of ribose phosphate molecules. The key enzyme for the synthesizing reaction is a transfer synthetase.
Researches suggested that inhibiting and reducing the activity and number of the transfer synthetase, controlling the reaction medium of the purine synthesis, improving the functions of various amino acids for synthesizing protein in the liver, and inhibiting and reducing the material needs of the purine synthetic reaction are also important measures for radically curing the gout at the source.
In the prescription, the Ophiopogon Japonicas nourishes yin, generates body fluid, moistens the lungs, clears away the heart fire, improves the anti-hypoxia capacity, reduces the occurrence rate of arrhythmia, protects myocardial ischemia, improves the dynamic effect of the heart’s blood, increases the myocardium energy supply, and restricts the range of myocardial infarction. Among the effective ingredients of the monarch drug cudweed herb, the flavone has the function of inhibiting the activity of enzyme for synthesis of purine with ribose phosphate. The minister drug angelica contains 12 amino acids essential to the human body, and also has the function of protecting the liver against injury, and noticeable effect of promoting the blood circulation to remove the blood stasis. The Ophiopogon Japonicas nourishes the stomach, replenishes yin and protects the heart, and the mucin contained in the Ophiopogon Japonicas provides raw materials for the angelica to repair liver cell membranes. The Ophiopogon Japonicas has a good effect of assisting the monarch drug cudweed herb and the minister drug angelica, and acts as an adjuvant drug. The Chinese medical science considers that “kidneys are the origin of congenital constitution” and “the root of life”, and that “yang of all internal organs is supplemented by the kidney yang, and yin of all internal organs is nourished by the kidney yin”. Thus, the kidney plays a very important role in the body health and life.
Since the uric acid is excreted with urine through the kidneys, when the concentration of the uric acid increases continuously, the kidney functions cannot meet the requirements of excretion, therefore, excess uric acid crystals will deposit in the kidneys, and long-term deposition will cause kidney failure, leading to uremia and resulting in death. Hence, it is indispensable to protect the kidneys, reinforce the kidneys, and nourish the kidneys so as to enhance the excretory function of the kidneys while radically curing the gout.
In the prescription, the cordyceps is able to tonify the kidneys, clear away the lung heat, invigorate the bone marrows, and support healthy energies. Among the monarch drugs, the effective ingredients of the Aconitum Carmichaelii supplement and nourishe the kidney yang, recuperate the depleted yang and rescue a patient from collapse; the effective ingredients of the dried rehmannia, Chinese yam, and Fructus Corni nourish the kidney yin, 8
P01651314BJ
P01651314BJ 2016102079 05 Dec 2016 and replenish both yin and yang, which, on one hand, supplies sufficient nutrient substances, and on the other hand, reinforces the kidney functions, achieving the noticeable effects of protecting the kidneys, reinforcing the kidneys, and nourishing the kidneys. The cordyceps supports the healthy energies and acts as an adjuvant drug. To enhance the ability of excreting the purine and the oxidized uric acid is one of the important measures for radically curing the gout.
In the prescription, the Rhizoma Alismatis is used to treat gonorrhea and hematuria and has a noticeable diuretic effect. The Cortex Moutan has the function of preventing apoenzyme from inducing injury and a good diuretic effect. The poria has a noticeable diuretic effect for renal and cardiac patients, immunomodulatory effect, anti-cirrhosis effect, and functions of improving the gastrointestinal function and resisting ulcer. The Rhizoma Dioscorea Hypoglauca activates the blood, stimulates the menstrual flow, dispels the wind and dampness, eases joint movement, induces diuresis for treating stranguria, and noticeably reduces serum cholesterol and the occurrence rate of aortic atherosclerosis. The effective ingredients of the above four materials coordinate with each other, to achieve the noticeable effects of replenishing the kidneys, supporting yang, nourishing the depleted yin, reinforcing the heart, protecting the liver, and improving the transporting and metabolic capacities of the spleens and the stomach. In the prescription, all of the Rhizoma Alismatis, the cortex moutan, the poria, and the Rhizoma Dioscorea Hypoglauca are adjuvant drugs.
Improving the immunologic function of the body itself and resisting internal invasion of exogenous evils are an important measure for treating diseases, and are also a powerful measure for radically curing the gout.
Reinforcing the capacity of the “self-healing rehabilitation system” of the body itself is an important measure for radically curing the gout.
Only by reinforcing the capacity of the “self-healing rehabilitation system” on the basis of improving the immunologic function, and integrating treatment and rehabilitation can the noticeable effect of radically curing the gout be achieved.
In the prescription, the effective ingredients of the monarch drug dried rehmannia noticeably increases the T lymphocytes in the peripheral blood, and promotes generation of antibody—hemolysis. The effective ingredient in the Chinese yam improves the transformation rate of lymphocytes and promotes generation of serum hemolysin. The effective ingredient polysaccharide in the Fructus Corni has noticeable immune enhancement effect, and can activate the activity of natural killing and phagocyte system. The effective ingredient in the Radix Glehniae has the function of enhancing the mononuclear phagocyte. The effective ingredient in the angelica has the noticeable effect of activating the phagocyte system, and the function of regulating immunity and recovering the immunologic competence for the immunocompromised patients. 9

P01651314BJ 2016102079 05 Dec 2016
To sum up, all of the monarch drugs, the minister drugs, and the adjuvant drugs in the prescription have the noticeable functions of improving the immunologic function.
The “self-healing rehabilitation system” of the body itself is formed by the skeletal system, the meridian system, and the nervous system, and it serves the function of self-healing during the treatment of diseases and rehabilitation after the treatment. Therefore, enhancing the capacity of the “self-healing rehabilitation system” is an extremely critical measure for radically curing the gout, and is also a key means for making the gout free of relapse.
The self-healing rehabilitation system cannot do without calcium. Otherwise, the “self-healing rehabilitation system” cannot play its role, because the calcium is an important ingredient of the skeletal system, a second messenger of the meridian system, and also neurotransmitter for transmitting information of the nervous system. Adding a large amount of complex calcium to the prescription just aims at neutralizing the uric acid in the blood and repairing and stimulating the system capacity of the “self-healing rehabilitation system” lost due to decalcification. In this way, the purpose of fast recovery and radical cure can be achieved.
With long-standing gout, since the urate deposits in the bone joints and the cartilage tissues, abnormality of cellular metabolism of the bone joints and the cartilage tissues is caused, and the self-synthesizing capacity, self-secreting capacity, and the capacity of resisting oxidation reaction of the raw material—glucosamine—in the bone joints for generating cartilage tissue proteoglycan will be decreased, leading to loss of source of cartilage synovial fluid, thereby cause huge pains, and injuries and severe deformities of the joints and soft tissues.
Therefore, Repairing the injuries of the bone joints and the cartilage tissues, enhancing the cellular metabolism, and improving the synovial fluid secretion capacity is an indispensable measure for radically curing the gout.
In the prescription, the amino sugar is a pure natural substance extracted from marine organisms, and has the noticeable effects of activating connective tissue cells of the bone joints, cartilage tissue cells, synthesizing polysaccharide and collagenous fibers to generate cartilage matrix, repair the bone joints and the cartilage tissues, and induce joint synovial fluid. Due to the properties and effects possessed by the amino sugar, the amino sugar is an indispensable substance for recovering the normal physiological status of the bone joints and the cartilage tissues during the process of radically curing gout. The liquorice has the functions of replenishing the spleens, tonifying the vital energies (qi), clearing away heat and toxic materials, and moderating properties of herbs. In the prescription of the present application, in addition to clearing away heat and toxic materials, the liquorice is mainly used for moderating the properties of herbs, so as to make the prescription become a 10
P01651314BJ
P01651314BJ 2016102079 05 Dec 2016 non-toxic and hormone-free product. The poria can enter into the heart, liver, spleen, stomach, lung, and kidney channels, and can take various herbs into the channels and act as a conductant drug.
The positive effects of the present application lie in:
The purine oxidation and the purine synthetase activity are inhibited at the source comprehensively, systematically, harmoniously, and synchronously, and measures for dissolving the urate crystals, neutralizing the uric acid in the blood, preventing and treating sudden change of disease state and so on are used; both yin and yang are replenished to improve the positive energy of the body and reinforce the immunologic function of the body, and the capacity of the “self-healing rehabilitation system” of the body itself is stimulated, so as to achieve the noticeable effect of radically curing the gout. On a pure natural, safe and reliable basis, the prescription has scientific prescription compatibility, and reasonable combination of nutrient elements and materials. The comprehensive, systematic, harmonious, and synchronous, scientific, dialectical, and novel method is used to radically cure the gout, and has the noticeable efficacies of improving the immunologic function and enhancing the self-healing rehabilitation of the body.
