Abstract
Un proceso para la preparación de 5,6-dietil-2,3-dihidro-1H-inden-2-amina, y sales de adición de ácido de la misma, a partir de 2-aminoindano. El proceso no utiliza los prejudiciales reactivos de Grignard o nitritos, tales como nitrito de isoamilo, y proporciona una alta regioselectividad y un alto rendimiento de la 5,6-dietil-2,3-dihidro-1H-inden-2-amina. En adición, se puede utilizar haluro de acetilo como un reactivo y también como un solvente en los Pasos (ii) y (iv). Reivindicación 1: Un proceso para la preparación de 5,6-dietil-2,3-dihidro-1H-inden-2-amina, el cual comprende: (i) proteger el grupo amino de 2-aminoindano con un grupo acilo en la presencia de un solvente orgánico para formar un compuesto de la fórmula (1): (ii) hacer reaccionar el compuesto de la fórmula (1) con haluro de acetilo en la presencia de un de un ácido de Lewis para formar un compuesto de la fórmula (2): (iii) tratar el compuesto de la fórmula (2) con un agente reductor en la presencia de un catalizador y un solvente orgánico para formar un compuesto de la fórmula (3): (iv) hacer reaccionar el compuesto de la fórmula (3) con haluro de acetilo en la presencia de un ácido de lewis para formar un compuesto de la fórmula (4): (v) tratar el compuesto de la fórmula (4) con un agente reductor en la presencia de un catalizador y un solvente orgánico para formar un compuesto de la fórmula (5): (vi) hidrolizar el compuesto de la fórmula (5) en la presencia de una base para formar la 5,6-dietil-2,3-dihidro-1H-inden-2-amina de la fórmula (6): en donde R se selecciona a partir del grupo que consiste en alquilo, arilo, alcoxilo, alquenilo, cicloalquilo, benzocicloalquilo, cicloalquilalquilo, aralquilo, heterocíclico, heteroaralquilo, y haloalquilo.
