Abstract
La presente se relaciona con una nueva enzima relacionada con el citocromo P450 (11 alfa hidroxilasa de Aspergillus ochraceus) y con una oxidorreductasa (oxidorreductasa de Aspergillus ochraceus) aislada a partir de una biblioteca de ADNc generada a partir del ARNm de esporas de Aspergillus ochraceus. Cuando se coexpreso el ADNc que codificaba la 11 alfa hidroxilasa en células de Spodoptera frugiperda (Sf-9) con el ADNc que codificaba oxidorreductasa humana como donante de electrones, catalizo exitosamente la conversion del sustrato esteroide 4-androsten-3,17-diona (AD) en 11 alfa-hidroxi-AD, determinado por un análisis de HPLC. La presente se relaciona también con moléculas de ácidos nucleicos asociadas o derivadas de estos ADNc que incluyen complementos, homologos y fragmentos de los mismos, y métodos para usar estás moléculas de ácidos nucleicos para generar, por ejemplo, polipéptidos y fragmentos de los mismos. La presente se relaciona también con la generacion de anticuerpos que reconocen la 11 alfa hidroxilasa y la oxidorreductasa de Aspergillus Ochraceus, y métodos para usar estos anticuerpos para detectar la presencia de estos polipétidos nativos y recombinantes dentro de células huésped sin modificar y transformadas , respectivamente. La presente también provee métodos para expresar el gen de Aspergillus de 11 alfa hidroxilasa por separado o en combinacion con oxidorreductasa humana o de Aspergillus, en células huésped heterologas, para facilitar la bioconversion de sustratos esteroides en sus contrapartes de 11 alfa hidroxilo.
