Abstract

본 발명은 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치에 관한 것으로서, 보다 상세하게는 세포외 소포체를 대용량의 배양액으로부터 분리하고 추출하기 위해 세포외 소포체가 양이온 물질로 표면이 개질된 다공성 매질로 구성된 필터를 통과하면서 흡착 및 추출되도록 하는 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치에 관한 것이다. 본 발명에 따르면, 엑소좀 또는 세포외 소포체를 고순도로 경제적이고 효율적으로 분리 및 정제할 수 있으며, 대량으로 세포외 소포체의 분리 및 정제가 가능해져서 GMP에도 적합해지며, 고순도이면서 입자크기 분포가 균일한 엑소좀 또는 세포외 소포체를 높은 수율로 경제적이면서 효율적으로 분리 및 정제할 수 있는 효과가 있다.

Description

생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치
본 발명은 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치에 관한 것으로서, 보다 상세하게는 세포외 소포체를 대용량의 배양액으로부터 분리하고 추출하기 위해 세포외 소포체가 양이온 물질로 표면이 개질된 다공성 매질로 구성된 필터와 접선유동여과장치를 직렬로 연결하여 통과시키는 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치에 관한 것이다.
최근 세포 분비물(secretome)에 세포의 행동을 제어하는 다양한 생체활성인자가 포함되어 있다는 연구가 보고되고 있으며, 특히 세포 분비물 내에는 세포 간 신호전달 기능을 갖는 나노 소포체인 '엑소좀(exosome)' 또는 '세포외 소포체(extracellular vesicles; EV)'가 포함되어 있어 그 성분과 기능에 대한 연구가 활발히 진행 중에 있다.
세포는 세포외 환경에 다양한 막(membrane) 유형의 소포체를 방출하는데, 통상 이러한 방출 소포체들을 세포외 소포체라고 부르고 있다. 세포외 소포체는 세포막 유래 소포체, 엑토좀(ectosomes), 쉐딩 소포체(shedding vesicles), 마이크로파티클(microparticles), 엑소좀 등으로 불려지기도 하며, 경우에 따라서는 엑소좀과는 구별되어 사용되기도 한다.
본 발명에서는 미세 소포체와 엑소좀을 합쳐서 세포외 소포체라고 정의한다.
엑소좀은 세포막의 구조와 동일한 이중 인지질막으로 이루어진 수십 내지 수백 나노미터 크기의 소포체로서, 내부에는 엑소좀 카고(cargo)라고 불리는 단백질, mRNA, miRNA 등이 포함되어 있다. 엑소좀 카고에는 광범위한 신호 전달 요소들(signaling factors)이 포함되며, 이들 신호전달 요소들은 세포 타입에 특이적이고 분비세포의 환경에 따라 상이하게 조절되는 것으로 알려져 있다.
엑소좀은 세포가 분비하는 세포 간 신호전달 매개체로서 이를 통해 전달된 다양한 세포 신호는 표적 세포의 활성화, 성장, 이동, 분화, 탈분화, 사멸(apoptosis), 괴사(necrosis)를 포함한 세포 행동을 조절한다고 알려져 있다.
엑소좀은 유래된 세포의 성질 및 상태에 따라 특이적인 유전물질과 생체활성 인자들이 포함되어 있다. 증식하는 줄기세포 유래 엑소좀의 경우 세포의 이동, 증식 및 분화와 같은 세포 행동을 조절하고, 조직 재생과 관련된 줄기세포의 특성이 반영되어 있다(Nature Review Immunology 2002 (2) 569-579).
이와 같은 엑소좀 또는 세포외 소포체를 분리하는 종래 기술로는 초원심분리법(ultracentrifugation), 밀도구배원심법(density gradient centrifugation), 초미세여과법(ultrafiltration), 사이즈 배제 크로마토그래피(size exclusion chromatography), 이온교환 크로마토그래피(ion exchange chromatography), 면역친화성 분리법(immunoaffinity capture), 미세유체기술 분리법(microfluidics-based isolation), 침전법(exosome precipitation), 총엑소좀 추출 키트(total exosome isolation kit), 또는 폴리머 기반 침전법(polymer based precipitation), 접선 유동 여과법 (Tangential Flow Filtration) 등이 있다.
초원심분리법(ultracentrifugation)은 엑소좀 또는 세포외 소포체를 분리하는데 지금까지 가장 널리 사용되었던 방법이나, 수율이 낮고, 분리하는데 시간이 많이 소요되며 노동집약적이고, 비싼 기기가 필요하다는 단점이 있다. 또한, 초원심분리법은 분리과정에서 엑소좀 또는 세포외 소포체에 손상을 줄 수 있어 후속 분석 과정이나 응용에 지장을 줄 수 있는 단점이 있다.
초미세여과법(ultrafiltration)은 초원심분리법과 함께 사용되어, 엑소좀 또는 세포외 소포체의 순도를 높일 수 있지만, 엑소좀 또는 세포외 소포체가 필터에 달라붙어 분리 후의 수율이 낮다는 문제점이 있다.
면역친화성 분리법은 항체를 엑소좀 또는 세포외 소포체에 붙여서 분리하는 방법으로 특이도가 높은 장점이 있지만, 항체를 만드는 과정과 분리 후 항체를 제거하는 과정이 필요하고 비용이 많이 드는 단점이 있으며 스케일-업(scale-up)에 부적합한 방식이다.
한편, 최근에는 엑소좀을 분리하는 방법으로 침전법(exosome precipitation), 총엑소좀 추출 키트(total exosome isolation kit), 또는 폴리머 기반 침전법(polymer based precipitation) 등의 다양한 엑소좀 분리 키트가 상업적으로 판매되고 있으나, 사용이 간편한 대신 시약값이 고가여서 실험실 수준에서 엑소좀 또는 세포외 소포체를 분리하기 위해 사용될 수는 있어도 대량으로 엑소좀 또는 세포외 소포체를 분리 및 정제하기에는 적합하지 않은 문제가 있다.
접선유동여과법(Tangential Flow Filtration; TFF)은 멤브레인 필터 표면과 평행하게 유체를 이동시키며 타겟물질을 여과시키는 방식으로서 연속 공정으로 대용량의 시료를 처리할 수 있는 장점을 가지고 있으나, 회수율은 앞선 사용기술인 침전법 대비 매우 낮은 것으로 알려져 있다.
무엇보다도 엑소좀 또는 세포외 소포체의 분리과정에서 문제가 되는 것은 단백질과 같은 불순물이 섞여 함께 분리되면서 순도나 수율이 낮다는 점이다.
또한, 엑소좀 또는 세포외 소포체의 분리 및 정제에는 시간이 많이 소모되고 번거롭고 비용이 많이 소모된다.
또한, 순도를 높이기 위해 개발된 기존의 분리 방법은 스케일-업(scale-up)을 어렵게 하고, GMP(Good Manufacturing Practice)에 부적합한 문제가 있다.
따라서, 본 발명이 속한 기술분야에서는 엑소좀 또는 세포외 소포체를 고순도로 경제적이고 효율적으로 분리 및 정제할 수 있는 동시에 스케일-업(scale-up)이 가능하고 GMP(Good Manufacturing Practice)에도 적합한 기술에 대한 요구가 꾸준히 계속되고 있다. 즉, 고순도이면서 입자크기 분포가 균일한 엑소좀 또는 세포외 소포체를 높은 수율로 경제적이면서 효율적으로 분리 및 정제하는 새로운 기술이 필요한 실정이다.
전술한 문제점을 해결하기 위한 본 발명은 대용량의 세포외 소포체 배양액에서 생성된 세포외 소포체 및 엑소좀이 음의 하전 특성 및 30 ㎚ 내지 500 ㎚의 크기를 갖는 것을 이용하여 미세입자나 멤브레인 등의 포획표면에 양이온 물질을 부착시켜 분리하는 기술과 나노미터 이하의 물질만을 배출해버리는 기술을 융합함으로써 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치를 제공하는 것을 목적으로 한다.
또한, 세포외 소포체가 부착될 표면적을 증대시키기 위하여 미세 구조의 다공성 매질로 여과부를 구성하여 세포외 소포체의 포획 효율을 획기적으로 증대시키도록 하는 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치를 제공하는 것을 목적으로 한다.
또한, 다공성 매질로 구성된 장치가 필터 형태로 구성되도록 함으로써 연속 공정으로 대용량의 배양액을 처리하여 세포외 소포체의 추출 및 정제 효율을 향상시키도록 하는 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치를 제공하는 것을 목적으로 한다.
또한, 배양액의 이동 방향에 대해 수직으로 배치되는 필터를 이용하여 세포외 소포체를 여과하고, 배양액의 이동 방향에 대해 수평으로 배치되는 접선유동여과장치를 이용하여 입자의 크기 또는 분자량이 작은 단백질 등의 잔류물질을 제거함으로써 고순도의 세포외 소포체를 추출할 수 있도록 하는 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치를 제공하는 것을 목적으로 한다.
전술한 문제점을 해결하기 위해 안출된 본 발명은 세포외 소포체 용액으로부터 세포외 소포체를 추출하는 방법으로서, 상기 세포외 소포체 용액을 예비로 여과하는 제1단계와; 상기 여과된 세포외 소포체 용액을 양의 하전물질로 표면개질화된 미세 다공성 구조체로 이루어진 미세필터구조체에 투입하여 상기 세포외 소포체를 흡착하는 제2단계와; 상기 제2단계 이후에 통과된 시료액을 접선유동여과장치에 투입하여 일정 크기 이하의 이물질을 제거하는 제3단계와; 상기 제2단계와 상기 제3단계를 소정의 회수만큼 반복하는 제4단계와; 용리액을 상기 미세필터구조체에 주입하여 세포외 소포체를 분리시키는 제5단계;를 포함한다.
상기 미세필터구조체는 미세입자, 멤브레인 구조, 메쉬 구조, 구형 비드(beads)의 적층 구조 중 어느 하나의 미세 다공성 구조체로 형성되는 것을 특징으로 한다.
상기 미세필터구조체는 상기 세포외 소포체 용액을 저장한 시린지 내부에 설치되는 것을 특징으로 한다.
상기 미세 다공성 구조체의 표면은 다가 양이온 물질에 의해 개질화된 것을 특징으로 한다.
상기 다가 양이온 물질은 폴리라이신, 프로타민, 폴리아르기닌, 폴리히스티딘, 양이온성 덱스트란, 양이온성 덴드리머, 양이온성 폴리사카라이드, 폴리아미도아민, 폴리에틸렌이민, 폴리쿼터늄, 폴리-2-다이메틸아미노에틸 메타크릴레이트(PDMAEMA), 폴리-2-다이메틸아미노메틸 스티렌(PDMAMS), 폴리-1-비닐피롤리돈(p1-VP), 폴리 다이에틸아미노에틸 아크릴레이트(pDEAEA), 폴리 다이메틸아미노에틸 아크릴레이트(pDMAEA), 폴리 다이에틸아미노에틸 메타크릴레이트(pDEAMA), 리포폴리아민(lipopolyamines), 4급 암모늄(quaternary ammoniums), 구아니딘(guanidine), 이미다졸(imidazole), 폴리아닐린(polyaniline), 폴리피롤(polypyrrol) 및 키토산(chitosan) 중 어느 하나 이상인 것을 특징으로 한다.
상기 미세 다공성 구조체에 포함된 미세입자는 폴리아크릴레이트, 폴리아크릴아미드, 폴리메타크릴레이트, 폴리에틸렌글리콜, 폴리스틸렌비닐벤젠, 폴리스티렌, 하이드로겔, 아가로오스, 세라믹, 실리카, 라텍스, 금속 입자, 유리 입자 및 자성 입자 중 어느 하나 이상으로 이루어지는 것을 특징으로 한다.
상기 용리액을 투입하여 상기 세포외 소포체를 분리하는 제5단계는 제1시약을 상기 미세필터구조체에 투입하고, 제1조건으로 상기 미세필터구조체를 세척하는 제5-1단계를 포함하고, 상기 제1시약은 Cl-, CH3COO-, SO4 2-, HCO-, SiO- 및 OH- 중 어느 하나 이상을 포함하는 물질이며, 상기 제1조건은 pH 5.5 내지 6.5, pH 7.5 내지 9.5 중 어느 하나의 pH 범위 내에서 1회 이상 수행되는 것으로 설정되는 것을 특징으로 한다.
상기 제5-1단계 이후에 제2시약을 상기 미세필터구조체에 투입하고, 제2조건으로 상기 미세필터구조체를 세척하는 제5-2단계를 더 포함하고, 상기 제2시약은 Cl-, CH3COO-, SO4 2-, HCO-, SiO- 및 OH- 중 어느 하나 이상을 포함하는 것으로서 0.2 M 내지 3 M의 농도의 물질이며, 상기 제2조건은 1분 내지 60분의 시간 동안 500rpm 내지 2000rpm으로 혼합하면서 1회 이상 수행되는 것으로 선택 사항으로 설정되는 것을 특징으로 한다.
상기 제5-2단계 이후에 추가적인 버퍼 치환을 하는 제5-3단계를 더 포함하고, 상기 제5-3단계에서 치환되는 버퍼는 생리식염수, 탈이온수, 증류수 중 어느 하나 이상인 것을 특징으로 한다.
다른 실시예에 따른 본 발명은 세포외 소포체 용액으로부터 세포외 소포체를 추출하는 장치로서, 제1시린지와; 제2시린지와; 양측 말단이 상기 제1시린지 및 상기 제2시린지의 출구에 각각 연결되는 접선유동여과장치와; 상기 접선유동여과장치의 일측에 형성되는 배출구와; 상기 배출구에 연결되는 배출용기와; 상기 제1시린지 또는 상기 제2시린지 중 어느 하나 이상의 내부에 설치되는 미세필터구조체;를 포함한다.
상기 제1시린지에 투입된 세포외 소포체 용액은 상기 접선유동여과장치를 통과한 후 상기 제2시린지에 유입되는 것을 특징으로 한다.
상기 세포외 소포체 용액은 상기 제1시린지 내부의 양압 또는 상기 제2시린지 내부의 음압에 의해 이동하는 것을 특징으로 한다.
상기 미세필터구조체는 상기 세포외 소포체 용액에 포함된 세포외 소포체를 양의 하전물질로 표면개질화된 미세 다공성 구조체의 표면에 흡착하여 여과하는 것을 특징으로 한다.
상기 접선유동여과장치는 상기 세포외 소포체 용액이 흘러가는 방향에 대해 수평방향이거나 90° 보다 작은 각도만큼 벌어진 방향으로 중공섬유막이 형성된 구조를 가지며, 상기 중공섬유막에 형성된 미세 크기의 공극을 통해 이물질이 배출되도록 하는 것을 특징으로 한다.
상기 중공섬유막에 형성된 공극의 크기는 50 ㎚ 이하인 것을 특징으로 한다.
상기 미세 다공성 구조체의 내부에 형성되는 공극의 크기는 800 ㎚ 이상인 것을 특징으로 한다.
또 다른 실시예에 따른 본 발명은 세포외 소포체 용액으로부터 세포외 소포체를 추출하는 장치로서, 분리 대상이 되는 세포외 소포체를 포함한 세포외 소포체 용액을 저장하며, 저장된 용액을 공급하는 공급용기와; 상기 세포외 소포체 용액에 포함된 세포외 소포체를 양의 하전물질로 표면개질화된 미세 다공성 구조체의 표면에 흡착하여 여과하는 미세필터구조체와; 상기 세포외 소포체 용액이 흘러가는 방향에 대해 측면 방향에 형성된 중공섬유막에 형성된 미세 크기의 공극을 통해 이물질이 배출되도록 하는 접선유동여과장치;를 포함한다.
상기 공급용기와 상기 미세필터구조체를 연결하는 제1배관과; 상기 제1배관에 설치되는 제1펌프와; 상기 미세필터구조체와 상기 접선유동여과장치를 연결하는 제2배관과; 상기 제2배관에 설치되는 제2펌프;를 더 포함한다.

본 발명에 따르면, 엑소좀 또는 세포외 소포체를 기존 기술 대비하여 매우 높은 순도로 경제적이고 효율적으로 분리 및 정제할 수 있으며, 대량으로 세포외 소포체의 분리 및 정제가 가능해져서 GMP에도 적합해지며, 고순도이면서 입자크기 분포가 균일한 엑소좀 또는 세포외 소포체를 높은 수율로 경제적이면서 효율적으로 분리 및 정제할 수 있는 효과가 있다.

이하에서 도면을 참조하여 본 발명의 실시예에 따른 "생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법 및 장치"를 설명한다.

본 발명은 생물학적 샘플에 포함된 세포나 바이러스 등으로부터 세포외 소포체 또는 엑소좀을 대량으로 추출 및 정제하기 위한 기술로서, 연속적으로 대량의 배양액을 투입하면서 2단계에 걸친 여과 과정과 세포외 소포체 분리 과정을 적용하는 것을 특징으로 한다.
이를 위해 도 1 및 도 2에 도시된 바와 같은 "생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치"(이하, '추출장치'라 한다)를 추출 및 정제 공정에 적용한다.
본 발명의 추출장치는 세포외 소포체 용액(세포외 소포체 또는 엑소좀이 배양된 상태의 용액)을 공급하는 수단과, 세포외 소포체 용액에서 세포외 소포체를 여과하는 수단과, 세포외 소포체 용액에 포함된 미세 크기의 이물질을 외부로 배출시키는 수단과, 여과 후 필터에서 세포외 소포체를 분리하는 수단으로 구성될 수 있다. 그리고, 세포외 소포체를 여과하는 수단은 용액의 유동방향에 수직방향으로 배치하며, 이물질을 배출시키는 수단은 용액의 유동방향에 수평방향으로 배치할 수 있다.
도 1의 제1 실시예에 따른 추출장치(100)와 도 2의 제2 실시예에 따른 추출장치(200)는 기본 구조는 동일하며, 미세필터구조체의 수가 다르다.
본 발명의 추출장치(100, 200)는 두 개의 시린지 사이에 접선유동여과장치를 직렬로 결합하고, 두 개의 시린지 중 어느 하나에 세포외 소포체 용액을 투입한 후, 양쪽의 시린지를 번갈아 동작시키면서 세포외 소포체 용액이 가운데에 있는 접선유동여과장치의 내부를 왕복이동하면서 여과 및 정제 과정이 진행되도록 구성된다.
접선유동여과장치에서 배출된 이물질은 별도의 배출용기에 저장된다.
또한 한 개의 시린지 또는 두 개의 시린지 내부에 미세필터구조체를 설치하여 시린지의 동작에 의해 용액이 이동하는 과정에서 세포외 소포체의 여과 작용이 이루어지도록 구성된다. 이러한 구조를 통해 시린지에서는 세포외 소포체의 여과(흡착)가 이루어지고, 접선유동여과장치에서는 이물질의 배출이 이루어지면서 세포외 소포체의 여과 효율이 증대되면서 순도가 높아질 수 있다.
제1 실시예
제1 실시예에 따른 추출장치(100)는 제1시린지(102)와 제2시린지(104)를 포함하며, 제1시린지(102)와 제2시린지(104)의 출구는 접선유동여과장치(106)의 양측 말단에 연결된다. 이를 통해 제1시린지(102), 제2시린지(104), 접선유동여과장치(106)에 수용된 용액은 시린지의 동작에 의해 이동할 수 있다.
접선유동여과장치(106)의 일측에는 배출구(110)가 형성되며, 배출구(110)에는 배출용기(108)가 연결된다. 배출구(110)를 통해서 세포외 소포체에 포함되어 있던 이물질이 배출되어 배출용기(108)에 저장된다. 이물질은 접선유동여과장치(106)에 의해 배출된 미세 크기의 단백질 등 이물질로서, 본 발명에서 여과 대상이 되는 세포외 소포체를 제외한 다른 물질을 의미한다.
제1시린지(102)의 내부에는 미세필터구조체(112)가 설치된다. 도 4에 도시된 바와 같이, 미세필터구조체(112)의 내부에는 멤브레인이나 다공성 입자 등으로 이루어진 필터가 설치되며, 필터의 표면에는 하전입자가 결합된다. 세포외 소포체는 하전입자에 전기력에 의해 결합되면서 여과 작용이 이루어진다. 필터에 결합된 세포외 소포체는 용리액 등의 투입을 통해 분리되는데, 미세필터구조체(112)의 여과 작용에 대해서는 후술한다.
제2시린지(104)에는 별도의 여과장치 또는 분리장치가 설치되지 않으며, 세포외 소포체 용액의 이동을 위한 압력을 인가하는 기능을 수행하도록 구성된다.
제1시린지(102)에는 분리 대상이 되는 세포외 소포체가 포함된 세포외 소포체 용액이 저장된 상태에서 분리 과정을 진행한다. 그리고 초기에 제2시린지(104)는 비어있는 상태로서 피스톤이 실린더 내부에 들어가 있다.
먼저, 세포외 소포체 용액을 준비하여 제1시린지(102)에 투입한다.(S102) 제1시린지(102)에 투입되는 세포외 소포체 용액에 포함된 불순물을 제거하기 위해 예비여과필터를 이용한 예비 여과 과정이 추가될 수 있다.
그리고, 제1시린지(102)의 피스톤을 눌러서 세포외 소포체 용액을 배출시키고, 제2시린지(104)의 피스톤을 빼면서 세포외 소포체 용액이 유입되도록 한다. 경우에 따라서는 세포외 소포체 용액을 배출하는 제1시린지(102)의 피스톤만 누르도록 구성될 수 있다. 이와 같이 세포외 소포체 용액을 배출하는 제1시린지(102)에서 발생한 양압과, 세포외 소포체 용액이 유입되는 제2시린지(104)에서 발생한 음압으로 인해 용액이 이동할 수 있다.
세포외 소포체가 분리되지 않은 상태의 세포외 소포체 용액은 제1시린지(102)의 배출구를 빠져나오게 되는데, 이 과정에서 제1시린지(102)의 내부에 장착된 미세필터구조체(112)에 세포외 소포체가 흡착되면서 1차 여과된다.(S104)
그리고 세포외 소포체 용액은 접선유동여과장치(106)의 일측으로 유입되고, 타측으로 이동하면서 미세 크기의 물질이 측면으로 배출되면서 2차 여과가 이루어진다.(S106)
접선유동여과장치(106)는 도 7에 도시된 바와 같이, 세포외 소포체 용액이 흘러가는 방향에 대해 수평방향으로 중공섬유막이 형성된 구조를 가진다. 경우에 따라서는 용액의 이동방향과 수평방향이거나, 90° 보다 작은 각도만큼 벌어진 방향으로 이동하도록 구성할 수도 있을 것이다.
접선유동여과장치(106)는 길이방향으로 세포외 소포체 용액의 이동통로가 형성되며, 이동통로의 일측면에는 중공섬유막으로 된 배출구간이 형성된다. 중공섬유막에는 미세 크기의 공극이 형성된다.
중공섬유막에 형성되는 공극의 크기는 100 ㎚ 이하, 80 ㎚ 이하, 60 ㎚ 이하, 40 ㎚ 이하일 수 있는데, 통상적인 세포외 소포체의 크기가 50 ㎚ 이상인 특성을 고려하면 공극의 크기는 50 ㎚ 이하인 것이 바람직하다. 현재 시중에서 쉽게 구할 수 있는 중공섬유막의 공극 크기는 10 ㎚ 내지 40 ㎚ 크기이다.
이와 같은 크기의 공극을 갖는 접선유동여과장치(106)를 사용하면 크기가 50 ㎚ 이하인 입자 또는 분자량이 100 kDa 또는 500 kDa 이하의 단백질 등의 이물질을 제거할 수 있다. 본 발명에서는 접선유동여과장치(106)에 의해 분리, 배출되는 세포 조각(debris), 단백질(protein)과 기타 입자를 "이물질"이라고 정의한다.
또한 접선유동여과장치(106)에 사용되는 중공섬유막은 중공섬유(Hollow fibe) 외에 통상적인 여과막(Membrane), 메쉬, 미세입자층, 다공성물질일 수 있다.
본 발명에서와 같이 미세필터구조체(112)와 접선유동여과장치(106)를 직렬로 연결하게 사용하면, 미세필터구조체(112)는 음의 하전을 갖는 나노 크기의 입자만을 포획하게 되고, 50 ㎚ 이하의 이물질은 접선유동여과장치(106)를 통해서 제거할 수 있다.
접선유동여과장치(106)를 통과하면서 1차 여과와 2차 여과가 이루어진 세포외 소포체 용액은 제2시린지(104)에 유입된다. 이때 제2시린지(104)의 피스톤은 실린더 밖으로 밀려나면서 용액이 내부로 들어올 수 있다. 접선유동여과장치(106)의 중공섬유막을 통해 배출된 이물질은 일측에 형성된 배출구(110)를 통해 배출용기(108)에 저장된다.
이 상태에서 제2시린지(104)의 피스톤을 눌러서 세포외 소포체 용액이 반대로 이동하도록 하면, 용액은 접선유동여과장치(106)를 지나면서 이물질의 배출이 이루어지고, 제1시린지(104)의 내부로 들어가면서 미세필터구조체(112)에 의해 세포외 소포체의 여과가 반복적으로 이루어진다.(S108) 이와 같은 1차 여과 및 2차 여과를 용액의 이동 방향을 바꾸면서 반복한다.
설정된 만큼의 반복을 통해 여과가 완료되면, 제1시린지(104)를 분리하고, 제1시린지(104)에 용리액을 투입하여 분리 과정을 진행한다.(S110) 경우에 따라서는 제1시린지(104)에서 미세필터구조체(112)를 분리하고, 분리된 미세필터구조체(112)에 용리액을 투입하는 방법을 적용할 수도 있을 것이다.
용리액의 투입으로 인해 미세필터구조체(112)에 부착된 음하전의 나노입자가 필터 표면에서 이탈되면서 세포외 소포체가 분리된다.(S112) 이와 같은 미세필터구조체(112)와 접선유동여과장치(106)의 두 단계 분리수단을 직렬연결한 하이브리드 공정을 적용할 경우, 최종적으로 음으로 하전된 50 ㎚ 이상의 세포외 소포체 또는 단백질만이 존재하도록 분리 및 정제할 수 있게 된다.
도 4에 도시된 바와 같이, 미세 필터 구조체(112)는 다수의 통공이 형성된 미세 다공성 구조체로 형성되며, 미세 다공성 구조체를 세포외 소포체 용액이 통과하면서 세포외 소포체가 미세 다공성 구조체에 잔류하는 방식으로 여과가 이루어진다. 미세 다공성 구조체의 표면에는 특정 전하를 갖는 물질이 부착된 형태로 제작되며, 세포외 소포체가 필터의 물질의 표면에 전하에 의해 결합된다. 통상적으로 세포외 소포체 용액에 포함된 세포외 소포체나 엑소좀은 음의 하전 특성을 가지므로, 미세필터구조체(112) 내부의 미세 다공성 구조체의 물질 표면에는 양의 하전 특성을 가지는 다가 양이온 물질로 개질화하는 것이 바람직하다.
미세필터구조체(112)의 표면 개질화에 사용되는 다가 양이온 물질은 폴리라이신, 프로타민, 폴리아르기닌, 폴리히스티딘, 양이온성 덱스트란, 양이온성 덴드리머, 양이온성 폴리사카라이드, 폴리아미도아민, 폴리에틸렌이민, 폴리쿼터늄, 폴리-2-다이메틸아미노에틸 메타크릴레이트(PDMAEMA), 폴리-2-다이메틸아미노메틸 스티렌(PDMAMS), 폴리-1-비닐피롤리돈(p1-VP), 폴리 다이에틸아미노에틸 아크릴레이트(pDEAEA), 폴리 다이메틸아미노에틸 아크릴레이트(pDMAEA), 폴리 다이에틸아미노에틸 메타크릴레이트(pDEAMA), 리포폴리아민(lipopolyamines), 4급 암모늄(quaternary ammoniums), 구아니딘(guanidine), 이미다졸(imidazole), 폴리아닐린(polyaniline), 폴리피롤(polypyrrol) 및 키토산(chitosan) 중 어느 하나 이상일 수 있다.
미세필터구조체(112)에 적용되는 미세 다공성 구조체는 구형 비드(beads)의 적층 구조를 포함하는 미세입자 구조, 멤브레인 구조, 메쉬 구조 중 어느 하나의 구조로 형성될 수 있다.
미세입자 구조인 경우에는 적층 구조 내부에 공극이 형성되어 세포외 소포체 용액이 통과하면서 내부에 포함된 세포외 소포체가 미세입자의 양이온층 표면에 하전 결합되도록 구성된다. 미세입자의 표면에 부착된 양이온층은 음의 하전을 갖는 세포외 소포체를 전기적인 인력으로 끌어당겨 결합하므로, 미세입자 사이의 공극의 크기가 세포외 소포체의 크기에 비해 작을 필요가 없다. 오히려 세포외 소포체 용액의 원활한 통과가 가능할 정도의 크기를 갖는 것이 바람직하다.
미세입자와 공극의 크기는 소재에 따라 달라질 수 있으나, 구형의 미세입자를 기준으로 세포외 소포체와 미세입자의 직경의 비율은 1:5 내지 1:10인 것이 바람직하다. 이러한 크기의 미세입자를 사용할 경우, 내부에 형성되는 공극의 크기는 세포외 소포체의 직경에 비해서 2배 내지 5배 정도가 될 수 있으며, 바람직하게는 공극의 크기가 800 ㎚ 이상이 되도록 구성한다.
본 발명에 사용되는 미세입자는 폴리아크릴레이트, 폴리아크릴아미드, 폴리메타크릴레이트, 폴리에틸렌글리콜, 폴리스틸렌비닐벤젠, 폴리스티렌, 하이드로겔, 아가로오스, 세라믹, 실리카, 라텍스, 금속 입자, 유리 입자 및 자성 입자 중 어느 하나 이상일 수 있다.
미세필터구조체(112)는 멤브레인 구조 또는 메쉬 구조로 형성될 수도 있으며, 이 경우에는 다가 양이온 물질이 흡착 가능한 소재가 사용된다. 멤브레인이나 메쉬의 소재로는 나일론, 폴리에틸렌, 폴리프로필렌, 폴리에틸렌 테레프탈레이트(Polyethylene terephthalate; PET), 폴리락틴산 (Polylactic acid; PLA), Polyethersulfone(PES), Cellulose acetate(CA), Polyvinylidene fluoride(PVDF), Polycarbonate(PC) 중 어느 하나 이상이 사용될 수 있다. 전술한 미세입자 구조와 동일하게 멤브레인이나 메쉬에도 다가 양이온 물질이 표면에 흡착된 상태로 제작된다.
멤브레인 구조나 메쉬 구조는 단일층일 수 있으며, 경우에 따라서는 여과 효 율을 높이기 위해 다중층일 수도 있다. 다중층으로 구성하는 경우에는 10개 이상의 단일층을 적층한 구조로 형성하는 것이 바람직하다.
미세필터구조체(112)에 흡착된 세포외 소포체를 분리하기 위해 용리액을 투입한다.
용리액을 사용한 용리과정은 단일 과정으로 구성될 수 있으나, 용리 효율을 높이기 위해 다중 과정으로 구성될 수도 있다. 본 발명에서는 두 종류의 용리액을 순차적으로 투입하여 용리 효율을 향상시킬 수 있다.
첫 번째는 제1시약을 이용하여 제1조건으로 미세필터구조체(112)를 1차 세척하도록 구성한다. 제1시약은 Cl-, CH3COO-, SO4 2-, HCO-, SiO- 및 OH- 중 어느 하나 이상을 포함하는 물질이 사용될 수 있다. 그리고, 제1조건은 pH 5.5 내지 6.5, pH 7.5 내지 9.5 중 어느 하나의 pH 범위 내에서 1회 이상 수행되는 것으로 설정할 수 있다.
두 번째는 제2시약을 이용하여 제2조건으로 미세필터구조체(112)를 2차 세척하도록 구성한다. 제2시약은 Cl-, CH3COO-, SO4 2-, HCO-, SiO- 및 OH- 중 어느 하나 이상을 포함하는 물질이 사용될 수 있는데, 0.2 M 내지 3 M의 농도로 사용하는 것이 바람직하다. 그리고 제2조건은 1분 내지 60분의 시간 동안 500rpm 내지 2000rpm으로 혼합하면서 1회 이상 수행되는 것으로 설정할 수 있다.
용리 과정은 제1시약을 제1조건으로 투입하는 단계와, 제2시약을 제2조건으로 투입하는 단계로 진행될 수 있다.
제1조건과 제2조건으로 용리 과정이 끝난 후에는 추가적인 버퍼 치환 단계를 추가할 수 있는데, 이 단계에서 치환되는 버퍼는 생리식염수, 탈이온수, 증류수 중 어느 하나 이상을 사용할 수 있다.
이와 같이 세포외 소포체가 분리된 상태로 저장된 제1시린지(102)의 분리 용액을 별도의 공정을 거쳐 처리함으로써 세포외 소포체의 추출 및 정제 과정이 완료된다.
제2 실시예
제2 실시예에 따른 추출장치(200)는 제1 실시예에 따른 추출장치(100)와 기본 구성은 동일하다. 즉, 제1시린지(202)와 제2시린지(204)를 포함하며, 제1시린지(202)와 제2시린지(204)의 출구는 접선유동여과장치(206)의 양측 말단에 연결된다. 이를 통해 제1시린지(202), 제2시린지(204), 접선유동여과장치(206)에 수용된 용액은 시린지의 동작에 의해 이동할 수 있다.
또한 접선유동여과장치(206)에 의해 분리된 이물질은 배출구(210)를 통해 배출용기(208)에 저장된다.
제2 실시예에서는 제1시린지(202)의 내부에 제1미세필터구조체(212)를 설치하고, 제2시린지(204)에는 제2미세필터구조체(214)를 설치하도록 구성한다. 즉, 양쪽의 시린지에 모두 미세필터구조체를 설치하여 분리 효율을 향상시킬 수 있다.
이 상태에서 제1시린지(202)와 제2시린지(204) 중 어느 한 곳에, 또는 제1시린지(202)와 제2시린지(204)의 양쪽 모두에 세포외 소포체 용액을 투입하고, 좌우측으로 용액을 이동시키면서 세포외 소포체 여과 및 이물질 분리 과정을 진행한다.
반복 여과가 종료되면, 제1시린지(202) 및 제2시린지(204)를 분리하여 용리액을 투입하면서 세포외 소포체를 분리할 수 있다. 제1 실시예에서와 동일하게, 제1미세필터구조체(212) 및 제2미세필터구조체(214)를 시린지로부터 분리하여 별도의 장치에서 용리 과정을 진행할 수도 있을 것이다.
한편, 도 5는 미세 필터 구조체의 구조를 나타낸 단면도이며, 도 6은 다른 실시예에 따른 미세 필터 구조체를 나타낸 모식도이다.
전술한 바와 같이, 여과막(61)이 미세입자(63) 구조로 이루어지는 경우에는 미세입자(63) 사이의 공극(64)의 크기가 세포외 소포체(EV)의 크기 보다 상대적으로 더 크게 형성한다. 미세입자(63)는 여과막(61)과 측벽(62)의 사이에 적층된다. 그리고 여과막(61)은 다중층으로 형성될 수 있다.
세포외 소포체는 미세입자(63)의 표면에 흡착된 양이온 물질에 의해 하전 결합된다. 세포외 소포체의 크기가 공극(64)의 크기에 비해 상대적으로 더 작기 때문에 세포외 소포체가 충분히 흡착된 상태에서도 세포외 소포체 용액은 원활하게 통과할 수 있다.
멤브레인 구조 또는 메쉬 구조인 경우에도 격자 형태의 원사(65)에 의해 형성되는 공극의 크기가 여전히 세포외 소포체의 크기에 비해 상당히 크기 때문에 원사(65)에 세포외 소포체가 충분히 흡착된 상태에서도 세포외 소포체의 용액은 원활하게 이동할 수 있다.
도 8은 여과방법에 따른 분리 입자의 크기를 비교하여 나타낸 그래프이며, 도 9는 여과방법에 따른 분리 입자의 농축도를 비교하여 나타낸 그래프, 도 10은 여과방법에 따른 프로틴 농축도를 비교하여 나타낸 그래프이다.
도 8에 도시된 바와 같이, 미세필터구조체(ExoFilter) 또는 접선유동여과장치(TFF)를 적용하지 않은 비교예(UC)의 결과에서는 입자의 크기가 상대적으로 가장 컸으며, 접선유동여과장치, 미세필터구조체를 적용했을 때 순차적으로 함유된 입자의 크기가 작았음을 확인하였다.
또한, 도 9에서와 같이, 분리된 세포외 소포체의 농축도는 미세필터구조체를 적용했을 때 가장 크고, 접선유동여과장치를 적용했을 때에는 상대적으로 작다는 점을 확인할 수 있다.
반대로, 도 10에서와 같이, 여과를 거친 용액 내부에 포함된 단백질 입자의 농축도는 비교예에서 가장 높고, 그 다음으로 접선유동여과장치, 미세필터구조체의 순으로 낮아진다.
이와 같은 결과를 통해 미세필터구조체를 단독으로 적용하여 여과했을 때 여과 효율이 가장 높고, 그 다음이 접선유동여과장치를 적용했을 때라는 점을 확인할 수 있다.
한편, 도 11은 미세필터구조체의 여과 후에 접선유동여과장치를 적용하는 방법1의 조건을 나타낸 표이며, 도 12는 접선유동여과장치의 여과 후에 미세필터구조체를 적용하는 방법2의 조건을 나타낸 표, 도 13은 방법1과 방법2에 따른 프로틴농축도를 비교하여 나타낸 그래프, 도 14 내지 도 17은 단위부피당 입자의 농축도를 비교하여 나타낸 그래프, 도 18은 방법1과 방법2에 따른 단위부피당 입자의 농축도를 비교하여 나타낸 그래프이다.
본 발명에 적용된 미세필터구조체와 접선유동여과장치는 각각 다른 방식으로 세포외 소포체를 분리할 수 있다. 미세필터구조체는 하전 결합에 의해 세포외 소포체를 흡착하여 분리하며, 접선유동여과장치는 크기가 상대적으로 작은 이물질을 제거함으로써 세포외 소포체를 분리할 수 있다. 두 가지 여과 장치를 결합하여 사용함으로써 더 큰 효과를 낼 수 있다는 것이 본 발명의 특징이다.
그런데, 두 가지 여과장치를 단순히 직렬로 연결하는 경우, 어떤 방식을 먼저 적용하느냐에 따라서 분리 효율에 차이가 있음을 확인하였다.
도 11에는 방법1에 의한 여과 조건이 기재되어 있는데, 방법1에서는 세포외 소포체 용액을 미세필터구조체에 먼저 통과시켜 1차 여과하고, 1차 여과된 용액을 접선유동여과장치에 통과시켜 2차 여과하였다.
그리고 도 12에 기재된 방법2에 의한 여과 조건은 접선유동여과장치에서 1차 여과를 하고, 미세필터구조체에서 2차 여과하는 방법을 적용하였다.
두 개의 실험에 사용된 미세필터구조체와 접선유동여과장치의 규격과 소재, 샘플 용액의 농도는 모두 동일하며, 두 개의 여과장치의 여과 순서만을 변경하여 실험하였다.
도 13에 도시된 결과는 접선유동여과장치(TFF)와 미세필터구조체(ExoFilter)를 각각 단독으로 사용한 경우와, 방법1(ExoFilter+TFF)을 적용한 경우와, 방법2(TFF+ExoFilter)를 적용한 경우의 단백질 입자의 농축도를 나타내고 있다.
접선유동여과장치(TFF)와 미세필터구조체(ExoFilter)를 각각 단독으로 사용한 경우의 농축도는 411과 516이며, 방법1을 적용한 경우에는 농축도가 53이며, 방법2를 적용한 경우에는 농축도가 228임을 확인할 수 있다.
따라서 두 개의 여과 장치를 순차적으로 적용할 경우, 각각의 여과장치를 단독으로 사용하는 경우에 비해서 효율이 크게 향상됨을 알 수 있다. 또한, 두 개의 여과장치를 연결 및 사용하는 순서에서도 미세필터구조체를 통한 1차 여과 후에 접선유동여과장치를 사용하여 2차 여과하는 것이 상대적으로 효율이 더 크다는 점을 알 수 있다.
도 14 내지 도 18의 그래프를 통해서 네 가지 경우의 여과에 따른 결과가 동일한 순서로 나타남을 확인할 수 있다.
제3 실시예
한편, 도 19는 본 발명의 제3 실시예에 따른 세포외 소포체 추출장치의 구조를 나타낸 개념도이다.
제3 실시예에 따른 추출장치(300)는 미세필터구조체와 접선유동여과장치를 통과하는 세포외 소포체 용액의 이동을 위한 압력제공을 펌프를 이용하도록 하여 자동화된 장치로 구성하는 것을 특징으로 한다.
공급용기(302)는 분리 대상이 되는 세포외 소포체를 포함한 세포외 소포체 용액을 저장하며, 저장된 용액을 필터와 여과장치로 공급한다.
미세필터구조체(306)는 앞서 설명한 바와 동일한 원리로 세포외 소포체를 하전 결합 방식으로 흡착하여 여과한다.
접선유동여과장치(310)는 미세필터구조체(306)를 통과한 세포외 소포체 용액에 포함된 단백질 등 이물질을 분리하여 배출한다.
접선유동여과장치(310)에서 분리된 이물질은 배출용기(316)에 저장된다.
공급용기(302)와 미세필터구조체(306)는 제1배관(318)으로 연결되며, 제1배관(318)에는 제1펌프(304)가 설치된다. 제1배관(318)에는 여과되기 전의 세포외 소포체 용액이 제1펌프(304)에서 발생한 압력에 의해 흐르게 된다.
미세필터구조체(306)와 접선유동여과장치(310)는 제2배관(320)에 의해 연결되며, 제2배관(320)에는 제2펌프(308)가 설치된다. 제2배관(320)에는 미세필터구조체(306)에 의해 세포외 소포체가 분리되고 접선유동여과장치(310)에 의해 이물질이 분리된 용액이 제2펌프(308)에서 발생한 압력에 의해 흐르게 된다.
접선유동여과장치(310)와 배출용기(316)는 제3배관(322)에 의해 연결되며, 제3배관(322)에는 제3펌프(314)가 설치된다. 제3배관(322)에는 접선유동여과장치(310)에 의해 분리된 이물질이 포함된 용액이 제3펌프(314)에서 발생한 압력에 의해 흐르게 된다.
접선유동여과장치(310)와 제3배관(322)의 연결부에는 클램프(312)가 추가될 수 있다. 클램프(312)는 제3배관(322)을 열거나 닫는 기능을 하며, 추출장치(300)의 동작 상태에 따라서 닫히거나 열리도록 제어된다.
이와 같은 구성을 갖는 추출장치(300)에서는 제1 실시예 및 제2 실시예에 설명된 내용과 동일한 방식으로 세포외 소포체가 분리된다.
공급용기(302)에 저장된 세포외 소포체 용액은 제1배관(318)을 따라 미세필터구조체(306)에 투입된다. 미세필터구조체(306)는 세포외 소포체를 양이온으로 하전된 막의 표면에 흡착하는 방식으로 여과한다.
미세필터구조체(306)를 통과한 세포외 소포체 용액은 접선유동여과장치(310)에 투입되고, 이동 방향에 대해 평행 방향으로 배치된 중공섬유막의 공극을 통해 이물질을 배출한다. 배출된 이물질은 제3배관(322)을 통해 배출용기(316)에 저장된다.
접선유동여과장치(310)를 통과한 세포외 소포체 용액은 제2배관(320)을 통해 순환하여 다시 미세필터구조체(306)에 투입되고, 미세필터구조체(306)와 접선유동여과장치(310)를 반복 통과하면서 여과와 분리 작용이 이루어진다.
제어부(도면 미도시)는 정해진 회수만큼 순환을 하면서 여과가 완료된 후에는 펌프의 동작을 중단하며, 작업자는 미세필터구조체(306)를 분리하여 용리액을 투입하는 등 세포외 소포체 분리 과정을 진행한다.
이상 첨부된 도면을 참조하여 본 발명의 바람직한 실시예를 설명하였지만, 상술한 본 발명의 기술적 구성은 본 발명이 속하는 기술 분야의 당업자가 본 발명의 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로서 이해되어야 하고, 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허청구범위에 의하여 나타내어지며, 특허청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.

Claims (19)

  1. 세포외 소포체 용액으로부터 세포외 소포체를 추출하는 방법으로서,
    상기 세포외 소포체 용액을 예비로 여과하는 제1단계와;
    상기 여과된 세포외 소포체 용액을 양의 하전물질로 표면개질화된 미세 다공성 구조체로 이루어진 미세필터구조체에 투입하여 상기 세포외 소포체를 흡착하는 제2단계와;
    상기 제2단계 이후에 통과된 시료액을 접선유동여과장치에 투입하여 일정 크기 이하의 이물질을 제거하는 제3단계와;
    상기 제2단계와 상기 제3단계를 소정의 회수만큼 반복하는 제4단계와;
    용리액을 상기 미세필터구조체에 주입하여 세포외 소포체를 분리시키는 제5단계;를 포함하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  2. 제1항에 있어서,
    상기 미세필터구조체는 미세입자, 멤브레인 구조, 메쉬 구조, 구형 비드(beads)의 적층 구조 중 어느 하나의 미세 다공성 구조체로 형성되는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  3. 제1항에 있어서,
    상기 미세필터구조체는 상기 세포외 소포체 용액을 저장한 시린지 내부에 설치되는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  4. 제1항에 있어서,
    상기 미세 다공성 구조체의 표면은 다가 양이온 물질에 의해 개질화된 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  5. 제4항에 있어서,
    상기 다가 양이온 물질은 폴리라이신, 프로타민, 폴리아르기닌, 폴리히스티딘, 양이온성 덱스트란, 양이온성 덴드리머, 양이온성 폴리사카라이드, 폴리아미도아민, 폴리에틸렌이민, 폴리쿼터늄, 폴리-2-다이메틸아미노에틸 메타크릴레이트(PDMAEMA), 폴리-2-다이메틸아미노메틸 스티렌(PDMAMS), 폴리-1-비닐피롤리돈(p1-VP), 폴리 다이에틸아미노에틸 아크릴레이트(pDEAEA), 폴리 다이메틸아미노에틸 아크릴레이트(pDMAEA), 폴리 다이에틸아미노에틸 메타크릴레이트(pDEAMA), 리포폴리아민(lipopolyamines), 4급 암모늄(quaternary ammoniums), 구아니딘(guanidine), 이미다졸(imidazole), 폴리아닐린(polyaniline), 폴리피롤(polypyrrol) 및 키토산(chitosan) 중 어느 하나 이상인 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  6. 제2항에 있어서,
    상기 미세 다공성 구조체에 포함된 미세입자는
    폴리아크릴레이트, 폴리아크릴아미드, 폴리메타크릴레이트, 폴리에틸렌글리콜, 폴리스틸렌비닐벤젠, 폴리스티렌, 하이드로겔, 아가로오스, 세라믹, 실리카, 라텍스, 금속 입자, 유리 입자 및 자성 입자 중 어느 하나 이상으로 이루어지는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  7. 제1항에 있어서,
    상기 용리액을 투입하여 상기 세포외 소포체를 분리하는 제5단계는 제1시약을 상기 미세필터구조체에 투입하고, 제1조건으로 상기 미세필터구조체를 세척하는 제5-1단계를 포함하고,
    상기 제1시약은 Cl-, CH3COO-, SO4 2-, HCO-, SiO- 및 OH- 중 어느 하나 이상을 포함하는 물질이며,
    상기 제1조건은 pH 5.5 내지 6.5, pH 7.5 내지 9.5 중 어느 하나의 pH 범위 내에서 1회 이상 수행되는 것으로 설정되는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  8. 제7항에 있어서,
    상기 제5-1단계 이후에 제2시약을 상기 미세필터구조체에 투입하고, 제2조건으로 상기 미세필터구조체를 세척하는 제5-2단계를 더 포함하고,
    상기 제2시약은 Cl-, CH3COO-, SO4 2-, HCO-, SiO- 및 OH- 중 어느 하나 이상을 포함하는 것으로서 0.2 M 내지 3 M의 농도의 물질이며,
    상기 제2조건은 1분 내지 60분의 시간 동안 500rpm 내지 2000rpm으로 혼합하면서 1회 이상 수행되는 것으로 설정되는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  9. 제8항에 있어서,
    상기 제5-2단계 이후에 추가적인 버퍼 치환을 하는 제5-3단계를 더 포함하고,
    상기 제5-3단계에서 치환되는 버퍼는 생리식염수, 탈이온수, 증류수 중 어느 하나 이상인 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 방법.
  10. 세포외 소포체 용액으로부터 세포외 소포체를 추출하는 장치로서,
    제1시린지와;
    제2시린지와;
    양측 말단이 상기 제1시린지 및 상기 제2시린지의 출구에 각각 연결되는 접선유동여과장치와;
    상기 접선유동여과장치의 일측에 형성되는 배출구와;
    상기 배출구에 연결되는 배출용기와;
    상기 제1시린지 또는 상기 제2시린지 중 어느 하나 이상의 내부에 설치되는 미세필터구조체;를 포함하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  11. 제10항에 있어서,
    상기 제1시린지에 투입된 세포외 소포체 용액은 상기 접선유동여과장치를 통과한 후 상기 제2시린지에 유입되는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  12. 제11항에 있어서,
    상기 세포외 소포체 용액은 상기 제1시린지 내부의 양압 또는 상기 제2시린지 내부의 음압에 의해 이동하는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  13. 제10항에 있어서,
    상기 미세필터구조체는 상기 세포외 소포체 용액에 포함된 세포외 소포체를 양의 하전물질로 표면개질화된 미세 다공성 구조체의 표면에 흡착하여 여과하는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  14. 제10항에 있어서,
    상기 접선유동여과장치는 상기 세포외 소포체 용액이 흘러가는 방향에 대해 수평방향이거나 90° 보다 작은 각도만큼 벌어진 방향으로 중공섬유막이 형성된 구조를 가지며, 상기 중공섬유막에 형성된 미세 크기의 공극을 통해 이물질이 배출되도록 하는 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  15. 제14항에 있어서,
    상기 중공섬유막에 형성된 공극의 크기는 50 ㎚ 이하인 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  16. 제13항에 있어서,
    상기 미세 다공성 구조체의 내부에 형성되는 공극의 크기는 800 ㎚ 이상인 것을 특징으로 하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  17. 세포외 소포체 용액으로부터 세포외 소포체를 추출하는 장치로서,
    분리 대상이 되는 세포외 소포체를 포함한 세포외 소포체 용액을 저장하며, 저장된 용액을 공급하는 공급용기와;
    상기 세포외 소포체 용액에 포함된 세포외 소포체를 양의 하전물질로 표면개질화된 미세 다공성 구조체의 표면에 흡착하여 여과하는 미세필터구조체와;
    상기 세포외 소포체 용액이 흘러가는 방향에 대해 측면 방향에 형성된 중공섬유막에 형성된 미세 크기의 공극을 통해 이물질이 배출되도록 하는 접선유동여과장치;를 포함하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  18. 제17항에 있어서,
    상기 공급용기와 상기 미세필터구조체를 연결하는 제1배관과;
    상기 제1배관에 설치되는 제1펌프와;
    상기 미세필터구조체와 상기 접선유동여과장치를 연결하는 제2배관과;
    상기 제2배관에 설치되는 제2펌프;를 더 포함하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
  19. 제18항에 있어서,
    상기 접선유동여과장치와 배출용기를 연결하는 제3배관과;
    상기 제3배관에 설치되는 제3펌프와;
    상기 접선유동여과장치와 상기 제3배관의 연결부에 설치되어 상기 제3배관을 열거나 닫도록 제어하는 클램프;를 더 포함하는, 생물학적 샘플로부터 세포외 소포체를 대량으로 추출하고 고순도로 정제하는 장치.
