Definitions
- the present invention relates to the field of pharmaceutical chemistry, and in particular to a class of 5-amino-1-alkyl-1H-1,2,3-triazole-4-carboxamide compounds and a preparation method and application thereof.
- Ion channels are an important class of drug targets. As a class of transmembrane proteins, ion channels contain water pores formed by multiple subunits, which can regulate the influx or efflux of different ions across the cell membrane when ligands bind or voltage changes. Ion channels function as basic excitatory or inhibitory units in the cell membranes of many tissues, including nerves, muscles, and glands. When the channel is open, the influx or efflux of ions generates various electrical signals, which promote a large number of cellular activities and basic physiological processes. Ion channels not only affect neurotransmitter release, muscle contraction, and glandular secretion, but also play a key role in learning and memory, cell volume maintenance, and homeostasis.
- Ion channel dysfunction or ion channelopathy is associated with impaired pain perception, inflammatory response, diabetes, and tumor cell proliferation and migration.
- abnormal ion channels are associated with many neurological diseases and arrhythmias, including anxiety, insomnia, epilepsy, schizophrenia, Parkinson's disease and Alzheimer's disease, as well as long/short QT syndrome. Therefore, ion channels are an important class of drug targets.
- the patent specification with publication number CN113717133A discloses a class of 3-amido-N-arylbenzamide compounds and their use in the preparation of anti-epileptic therapeutic drugs.
- the patent specification with publication number CN114044757A discloses a class of triazole-4-(N-substituted carboxamide)-5-triazene compounds and their preparation methods and their use in the preparation of anti-epileptic drugs. Neither of these two prior arts reports the drug effects in the lamotrigine resistance model, and does not involve the solution of the drug resistance problem.
- triazole-4-(N-substituted carboxamide)-5-triazene compounds disclosed in the patent specification with publication number CN114044757A contain triazene groups, which makes these compounds have biological toxicity and difficult to pass in terms of drug safety.
- the present invention changes the necessary triazene-containing core structure in CN114044757A, discards the triazene structure with potential biological toxicity, and replaces it with an amino group, designs and synthesizes a class of non-biologically toxic 5-amino-1-alkyl-1H-1,2,3-triazole-4-carboxamide compounds, and unexpectedly finds that this class of new structural compounds still has obvious electrophysiological activity and can show inhibitory activity on sodium and other ion channels.
- PTZ pentylenetetrazol acute epilepsy attack model
- MES maximum electric shock epilepsy attack model
