本発明は、比較的温和な条件で、グライム配位アルカリ金属イオンと塩を形成して安定化されたSF5 -を脱酸素フッ素化剤として用いて、ケトン基、アルデヒド基、ヒドロキシ基等を有する多種多様な酸素含有化合物に対してフッ素原子を導入して、フッ素含有化合物を製造する方法を提供する。本発明は、一般式(E1)[式中、Mは、Cs+、K+、又はRb+を表し;G4は、テトラエチレングリコールジメチルエーテルを表し;nは、1~4の整数を表す]で表される脱酸素フッ素化剤を用いて、DMPU、G4、DMA、HMPA、及びMeCNからなる群より選択される1種以上の溶媒中で、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する酸素含有化合物を脱酸素フッ素化して、フッ素含有化合物を製造する、フッ素含有化合物の製造方法である。[M(G4)n][SF5] (E1)
Description
本発明は、脱酸素フッ素化剤を用いて、ケトン基、アルデヒド基、ヒドロキシ基等を有する酸素含有化合物にフッ素原子を導入して、フッ素含有化合物を製造する方法に関する。
本願は、2022年10月25日に、日本に出願された特願2022-170509号に基づき優先権を主張し、その内容をここに援用する。

フッ素原子は、電気陰性度が高く、かつ水素原子と同様に小さい。この特徴により、フッ素原子は多くの原子と安定して結合することができ、フッ素が導入された有機化合物は、導入前よりも耐熱性、耐薬品性、耐光性、耐水性等が向上する傾向にある。特に、炭素-フッ素結合は、結合距離が短く、剛直であり、分極率も低い。この炭素-フッ素結合の特徴のため、炭素-フッ素結合が導入された有機化合物は、反応性が低下し、化合物としての安定性が向上する。有機化合物をフッ素化することで、有用な有機化合物を合成することができるため、様々なフッ素化剤が開発されている。
フッ素化剤としては、例えば、N-ジエチルアミノスルフルトリフルオリド(DAST)が知られている。例えば、非特許文献1には、アシルピロール化合物は、ピロール環中のNHを保護することにより、DASTによって脱酸素フッ素化反応が生じ、ジフルオロアルキルピロリック化合物が合成されることが報告されている。また、DASTは、カルボン酸及びアルデヒドに対しても脱酸素フッ素化剤として機能する。例えば、特許文献1には、DASTによって、レチノイン酸からフッ化レチノイルが、レチナールから15,15-ジフルオロアキセロフテンが、それぞれ生成されることが報告されている。しかしながら、アシルピロール化合物のようなフリーのNHを有する化合物は、NHを保護せずに脱酸素フッ素化することは困難である。
その他、SF4も脱酸素フッ素化剤として有用であるが、SF4での脱酸素フッ素化は、HFを共存させる必要がある。そこで、非特許文献2には、SF4のより安定で使用しやすい錯体として、グライム配位アルカリ金属イオンとSF5
-の塩([M(G4)n][SF5])の脱酸素フッ素化剤としての利用が報告されている。当該文献では、THF中で、3-フェニル-1-プロパノールを[Cs(G4)2][SF5]と反応させることにより、脱酸素フッ素化物が得られたことが報告されているが、収率は低く、さらなる改善が必要である。

本発明者らは、グライム配位アルカリ金属イオンとSF5
-の塩を脱酸素フッ素化剤として行う脱酸素フッ素化を、特定の溶媒中で行うことにより、比較的温和な条件で、多種多様な酸素含有化合物に対して脱酸素フッ素化できることを見出し、本発明を完成させた。
すなわち、本発明は以下の通りである。

本発明に係るフッ素含有化合物の製造方法により、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する多種多様な酸素含有化合物を、比較的温和な条件で脱酸素フッ素化することができ、これにより様々なフッ素含有化合物を得ることができる。
本発明及び本願明細書において、「Cp1-p2」(p1及びp2は、p1<p2を満たす正の整数である)は、炭素数がp1~p2の基であることを意味する。
本発明及び本願明細書において、「置換基を有していてもよい基」とは、置換基を有していない基と、置換基を有している基の両方を含む。
また、以降において、「化合物(n)」は式(n)で表される化合物を意味する。
本発明及び本願明細書において、「C1-30脂肪族炭化水素基」は、置換基を有していてもよいC1-30アルキル基、置換基を有していてもよいC2-30アルケニル基、置換基を有していてもよいC2-30アルキニル基、の全てを含む。「C1-30脂肪族炭化水素基」は、直鎖であってもよく、分岐鎖であってもよく、環式基であってもよい。
本発明及び本願明細書において、「置換基を有しているCp1-p2脂肪族炭化水素基」は、Cp1-p2脂肪族炭化水素基の炭素原子に結合している水素原子の1又は複数個、好ましくは1~3個が、他の官能基に置換されている基である。2個以上の置換基を有する場合、置換基同士は互いに同種であってもよく、異種であってよい。
本発明及び本願明細書において、「C1-30アルキル基」は、炭素数1~30のアルキル基であり、直鎖であっても分岐鎖であってもよく、環状構造を含む基であってもよい。C1-30アルキル基の例としては、メチル基、エチル基、プロピル基、イソプロピル基、ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、ペンチル基、イソペンチル基、ネオペンチル基、tert-ペンチル基、ヘキシル基、ヘプチル基、オクチル基、ノニル基、デシル基、ウンデシル基、ドデシル基、トリデシル基、テトラデシル基、ペンタデシル基、ヘキサデシル基、ヘプタデシル基、オクタデシル基、ノナデシル基、エイコシル基、ヘンエイコシル基、ドコシル基、トリコシル基、テトラコシル基、ペンタコシル基、ヘキサコシル基、ヘプタコシル基、オクタコシル基、ノナコシル基、トリアコンチル基、シクロプロピル基、シクロブチル基、シクロペンチル基、シクロヘキシル基、シクロヘプチル基、シクロオクチル基等が挙げられる。

本発明及び本願明細書において、「脂肪族ヘテロ環式基」とは、脂肪族ヘテロ環を含む基、すなわち、環状構造を含む脂肪族炭化水素基の環状構造を形成する炭素原子の1~3個が、炭素原子以外の原子に置換されている基である。当該脂肪族ヘテロ環式基としては、窒素原子を含む基(含窒素脂肪族ヘテロ環式基)であってもよく、酸素原子を含む基(含酸素脂肪族ヘテロ環式基)であってもよく、硫黄原子を含む基(含硫脂肪族ヘテロ環式基)であってもよい。また、脂肪族ヘテロ環式基を構成する炭素原子以外の原子が2種以上であってもよい。当該脂肪族ヘテロ環式基としては、例えば、ピロリジル基、ピペリジル基、ピペラジル基、テトラヒドロフラニル基、テトラヒドロピラニル基、テトラヒドロチオフェニル基、チアニル基、モルホリル基、チオモルホリル基、ジオキサニル基、ジチアニル基等が挙げられる。

本発明及び本願明細書において、「アリール基」は、芳香族性を備える環式基であり、当該環が炭素原子のみによって構成されている基である。
本発明及び本願明細書において、「C6-14アリール基」は、炭素数6~14の芳香族炭化水素基であり、C6-12アリール基が特に好ましい。C6-14アリール基の例としては、フェニル基、ナフチル基、アントリル基、9-フルオレニル基等が挙げられ、フェニル基が特に好ましい。
本発明及び本願明細書において、「C6-14アリール基」は、炭素数6~14の芳香族炭化水素基であり、C6-12アリール基が特に好ましい。C6-14アリール基の例としては、フェニル基、ナフチル基、アントリル基、9-フルオレニル基等が挙げられ、フェニル基が特に好ましい。
本発明及び本願明細書において、「置換基を有しているアリール基」は、アリール基の炭素原子に結合している水素原子の1又は複数個、好ましくは1~3個が、他の官能基に置換されている基である。2個以上の置換基を有する場合、置換基同士は互いに同種であってもよく、異種であってよい。当該置換基としては、C1-6アルキル基、Cs-6アルケニル基、C1-6アルコキシ基、C1-6アルキルチオ基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、ニトロ基、アルデヒド基、C1-10アシル基、置換基を有していてもよいアミノ基、置換基を有していてもよいC6-14アリール基、及び置換基を有していてもよいC6-14ヘテロアリール基等が挙げられる。「置換基を有していてもよいC6-14アリール基」の例としては、フェニル基、ナフチル基、アントリル基、2-メチルフェニル基、4-メチルフェニル基、3,5-ジメチルフェニル基、2,6-ジメチルフェニル基、2,4-ジメチルフェニル基、2-トリフルオロメチルフェニル基、2-メトキシフェニル基、4-メトキシフェニル基、2,4-ジメトキシフェニル基、3,5-ジメトキシフェニル基、2-メチルチオフェニル基、4-メチルチオフェニル基、2,4-ジメチルチオフェニル基、3,5-ジメチルチオフェニル基、4-クロロフェニル基、4-ブロモフェニル基、4-ニトロフェニル基、4-アミノフェニル基、4-(メチルアミノ)フェニル基、4-(ジメチルアミノ)フェニル基、4-(アセチルアミノ)フェニル基等が挙げられる。
本発明及び本願明細書において、「ヘテロアリール基」は、芳香族性を備える環式基であり、当該環が炭素原子と炭素原子以外の原子によって構成されている基である。ヘテロアリール基としては、窒素原子を含む基(含窒素ヘテロアリール基)であってもよく、酸素原子を含む基(含酸素ヘテロアリール基)であってもよく、硫黄原子を含む基(含硫ヘテロアリール基)であってもよい。また、芳香環を構成する炭素原子以外の原子が2種以上であってもよい。
C5-14含窒素ヘテロアリール基としては、例えば、ピロリル基、イミダゾリル基、ピラゾリル基、ピリジル基、ピラジニル基、ピリミジニル基、ピリダジニル基、インドリル基、イソインドリル基、インドリニル基、ベンゾイミダゾリル基、ベンゾトリアゾリル基、キノリル基、イソキノリル基、キナゾリル基、カルバゾリル基等が挙げられる。C5-14含酸素ヘテロアリール基としては、例えば、フラニル基、ピラニル基、ベンゾピラニル基、キサンテニル基等が挙げられる。C5-14含硫ヘテロアリール基としては、例えば、チエニル基等が挙げられる。2種以上のヘテロ原子を含むC5-14ヘテロアリール基としては、例えば、オキサゾリル基、イソオキサゾリル基、チアゾリル基、イソチアゾリル基等が挙げられる。
本発明及び本願明細書において、「置換基を有しているヘテロアリール基」は、ヘテロアリール基の炭素原子に結合している水素原子の1又は複数個、好ましくは1~3個が、他の官能基に置換されている基である。2個以上の置換基を有する場合、置換基同士は互いに同種であってもよく、異種であってよい。当該置換基としては、C1-6アルキル基、Cs-6アルケニル基、C1-6アルコキシ基、C1-6アルキルチオ基、C1-10アシル基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、トリハロメチル基、ニトロ基、アルデヒド基、置換基を有していてもよいアミノ基、置換基を有していてもよいC6-14アリール基、及び置換基を有していてもよいC6-14ヘテロアリール基等が挙げられる。
本発明及び本願明細書において、「芳香族基」は、置換基を有していてもよいアリール基(芳香族炭化水素基)と、置換基を有していてもよいヘテロアリール基(複素環式基)の両方を含む。
<脱酸素フッ素化剤>
本発明において用いられる脱酸素フッ素化剤(以下、「本発明の脱酸素フッ素化剤」ということがある。)は、下記一般式(E1)で表される化合物である。当該脱酸素フッ素化剤は、SF5 -イオンが、ケトン基の炭素原子や、ヒドロキシ基と結合する炭素原子に対して求核性試薬として機能することにより、これらの炭素原子に結合している酸素原子に代えてフッ素原子を結合させる。本発明の脱酸素フッ素化剤は、このSF5 -イオンが、グライム配位アルカリ金属イオンと塩を形成しているため、熱安定性や低圧耐性が向上している。具体的には、本発明の脱酸素フッ素化剤は、低圧環境下でもSF4が脱離し難く、また100℃程度での熱分解も抑制されている。このため、比較的穏やかな条件で脱酸素フッ素化反応を進行させられることに加えて、脱酸素フッ素化させる基質の構造的制限が緩和されている。本発明の脱酸素フッ素化剤は、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する多種多様な酸素含有化合物を基質とすることができる。
本発明において用いられる脱酸素フッ素化剤(以下、「本発明の脱酸素フッ素化剤」ということがある。)は、下記一般式(E1)で表される化合物である。当該脱酸素フッ素化剤は、SF5 -イオンが、ケトン基の炭素原子や、ヒドロキシ基と結合する炭素原子に対して求核性試薬として機能することにより、これらの炭素原子に結合している酸素原子に代えてフッ素原子を結合させる。本発明の脱酸素フッ素化剤は、このSF5 -イオンが、グライム配位アルカリ金属イオンと塩を形成しているため、熱安定性や低圧耐性が向上している。具体的には、本発明の脱酸素フッ素化剤は、低圧環境下でもSF4が脱離し難く、また100℃程度での熱分解も抑制されている。このため、比較的穏やかな条件で脱酸素フッ素化反応を進行させられることに加えて、脱酸素フッ素化させる基質の構造的制限が緩和されている。本発明の脱酸素フッ素化剤は、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する多種多様な酸素含有化合物を基質とすることができる。
一般式(E1)中、Mは、セシウムイオン、カリウムイオン、又はルビジウムイオンを表す。また、G4は、テトラエチレングリコールジメチルエーテル(テトラグライム)(CAS No.: 143-24-8)を表し、nは、1~4の整数を表す。本発明において用いられる脱酸素フッ素化剤としては、一般式(E1)中、Mがセシウムイオンである化合物([Cs(G4)n][SF5])が好ましく、Mがセシウムイオンであり、nが2である化合物([Cs(G4)2][SF5])が特に好ましい。
<フッ素含有化合物の製造方法>
本発明に係るフッ素含有化合物の製造方法は、本発明の脱酸素フッ素化剤を用いて、1,3-ジメチル-3,4,5,6-テトラヒドロ-2(1H)-ピリミジノン(DMPU)、テトラエチレングリコールジメチルエーテル(テトラグライム)(G4)、ジメチルアセトアミド(DMA)、ヘキサメチル亜リン酸トリアミド(HMPA)、及びアセトニトリル(MeCN)からなる群より選択される1種以上の溶媒中で、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する酸素含有化合物を脱酸素フッ素化して、フッ素含有化合物を製造する方法である。本発明の脱酸素フッ素化剤は、室温では安定であるが、溶媒中で温度を上げると分解されやすい。特にテトラヒドロフラン(THF)やジメチルエーテル(DME)中では60℃程度でも分解されてしまうが、MeCN中では80℃程度まで安定であり、DMPU、G4、DMA、及びHMPA中では、室温~120℃においてほとんど分解しない。本発明においては、脱酸素フッ素化反応を行う際の溶媒として、DMPU、G4、DMA、HMPA、及びMeCNからなる群より選択される1種以上を用いることにより、効率よくフッ素含有化合物を製造できる。本発明に係るフッ素含有化合物の製造方法で用いる溶媒としては、1種類の溶媒であってもよく、2種以上の溶媒の混合溶媒であってもよい。
本発明に係るフッ素含有化合物の製造方法は、本発明の脱酸素フッ素化剤を用いて、1,3-ジメチル-3,4,5,6-テトラヒドロ-2(1H)-ピリミジノン(DMPU)、テトラエチレングリコールジメチルエーテル(テトラグライム)(G4)、ジメチルアセトアミド(DMA)、ヘキサメチル亜リン酸トリアミド(HMPA)、及びアセトニトリル(MeCN)からなる群より選択される1種以上の溶媒中で、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する酸素含有化合物を脱酸素フッ素化して、フッ素含有化合物を製造する方法である。本発明の脱酸素フッ素化剤は、室温では安定であるが、溶媒中で温度を上げると分解されやすい。特にテトラヒドロフラン(THF)やジメチルエーテル(DME)中では60℃程度でも分解されてしまうが、MeCN中では80℃程度まで安定であり、DMPU、G4、DMA、及びHMPA中では、室温~120℃においてほとんど分解しない。本発明においては、脱酸素フッ素化反応を行う際の溶媒として、DMPU、G4、DMA、HMPA、及びMeCNからなる群より選択される1種以上を用いることにより、効率よくフッ素含有化合物を製造できる。本発明に係るフッ素含有化合物の製造方法で用いる溶媒としては、1種類の溶媒であってもよく、2種以上の溶媒の混合溶媒であってもよい。
本発明に係るフッ素含有化合物の製造方法において、脱酸素フッ素化反応は、DMPU、G4、DMA及びHMPAからなる群より選択される1種以上からなる溶媒に、基質となる化合物と本発明の脱酸素フッ素化剤とを添加して、室温~120℃で所定時間保持することによって進行させることができる。反応中、反応系の温度は、室温~120℃の範囲内で段階的に変化させてもよい。反応系に添加する基質化合物と本発明の脱酸素フッ素化剤の量は、基質化合物1モルに対して、本発明の脱酸素フッ素化剤が0.5~100モル当量が好ましく、0.5~50モル当量がより好ましく、0.5~10モル当量がさらに好ましい。
本発明に係るフッ素含有化合物の製造方法において用いられる基質化合物は、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する酸素含有化合物であれば特に限定されるものではない。当該酸素含有化合物としては、少なくとも、アルデヒド基、ケトン基、カルボキシ基、及びヒドロキシ基のうちの1個以上を有していればよく、アルデヒド基、ケトン基、カルボキシ基、及びヒドロキシ基を2個以上有していてもよい。アルデヒド基、ケトン基、カルボキシ基、及びヒドロキシ基を複数個有している場合、同種の官能基を有していてもよく、アルデヒド基、ケトン基、カルボキシ基、及びヒドロキシ基の中の2種以上を組み合わせて有していてもよい。
本発明に係るフッ素含有化合物の製造方法は、例えば、基質とする酸素含有化合物が、下記一般式(S1)で表される化合物である場合に、フッ素含有化合物として、下記一般式(P1)で表される化合物を製造することができる。
一般式(S1)及び(P1)中、R1は、水素原子又はC1-6アルキル基である。R1がC1-6アルキル基である場合、置換基を有していないC1-6アルキル基であってもよく、置換基を有しているC1-6アルキル基であってもよい。当該置換基としては、本発明の脱酸素フッ素化剤による脱酸素フッ素化を阻害しない基であれば特に限定されるものではない。当該置換基としては、例えば、C1-6アルコキシ基、C1-6アルキルチオ基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、トリハロメチル基、ニトロ基、置換基を有していてもよいアミノ基等が挙げられる。置換基を有していてもよいアミノ基としては、アミノ基の1個又は2個の水素原子が、C1-6アルキル基、C1-10アシル基等で置換されている基であり、具体的には、ジメチルアミノ基、ジエチルアミノ基、メチルアミノ基、アセチルアミノ基、アセチルメチルアミノ基等が挙げられる。本発明に係るフッ素含有化合物の製造方法における基質としては、一般式(E1)中、R1は、水素原子、又は、置換基を有していないC1-6アルキル基であることが好ましく、水素原子、メチル基、エチル基、プロピル基、又はイソプロピル基であることがより好ましく、水素原子又はメチル基であることがさらに好ましい。
一般式(S1)及び(P1)中、R11は、置換基を有していてもよいC1-30脂肪族炭化水素基、置換基を有していてもよいアルコキシ基、置換基を有していてもよい脂肪族ヘテロ環式基、置換基を有していてもよいアリール基、又は置換基を有していてもよいヘテロアリール基である。
R11が置換基を有していてもよいC1-30脂肪族炭化水素基である場合、当該置換基としては、本発明の脱酸素フッ素化剤による脱酸素フッ素化を阻害しない基であれば特に限定されるものではない。当該置換基としては、例えば、C1-6アルコキシ基、C1-6アルキルチオ基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、トリハロメチル基、ニトロ基、置換基を有していてもよいアミノ基、置換基を有していてもよいアリール基、及び置換基を有していてもよいヘテロアリール基等が挙げられる。置換基を有していてもよいアミノ基としては、前記R1の置換基として挙げられた基と同様の基を用いることができる。
R11が置換基を有していてもよいC1-30脂肪族炭化水素基である場合、当該置換基を有していてもよいC1-30脂肪族炭化水素基としては、置換基を有していてもよいC1-10アルキル基又は置換基を有していてもよいC2-10アルケニル基が好ましく、置換基を有していてもよいアリール基若しくは置換基を有していてもよいヘテロアリール基を置換基として有していてもよいC1-10アルキル基、又は、置換基を有していてもよいアリール基若しくは置換基を有していてもよいヘテロアリール基を置換基として有していてもよいC2-10アルケニル基がより好ましく、置換基を有していてもよいフェニル基を置換基として有していてもよいC1-10アルキル基、又は、置換基を有していてもよいフェニル基を置換基として有していてもよいC2-10アルケニル基がさらに好ましい。なかでも、置換基を有していてもよいフェニル基を置換基として有していてもよい、メチル基、エチル基、プロピル基、又はビニル基が特に好ましい。当該置換基を有していてもよいフェニル基としては、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、ニトロ基、アミノ基、ジメチルアミノ基、メチルアミノ基、アセチルアミノ基、メトキシ基、ビニル基、プロペニル基、1-メチルビニル基、及びヒドロキシ基からなる群より選択される1種以上の基を置換基として有するフェニル基、置換基を有していないフェニル基が挙げられる。
R11が置換基を有していてもよいアルコキシ基である場合、当該アルコキシ基としては、C1-10アルコキシ基が好ましく、アルキル基部分が直鎖状又は分岐鎖状のC1-10アルコキシ基がより好ましく、アルキル基部分が直鎖状のC1-6アルコキシ基がさらに好ましい。
R11が置換基を有していてもよいアルコキシ基である場合、当該置換基としては、本発明の脱酸素フッ素化剤による脱酸素フッ素化を阻害しない基であれば特に限定されるものではない。当該置換基としては、例えば、C1-6アルコキシ基、C1-6アルキルチオ基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、ニトロ基、置換基を有していてもよいアリール基、及び置換基を有していてもよいヘテロアリール基等が挙げられる。
R11が置換基を有していてもよいアルコキシ基である場合、当該置換基を有していてもよいアルコキシ基としては、置換基を有していてもよいC1-10アルコキシ基が好ましく、アルキル基部分が直鎖状又は分岐鎖状であり、かつ置換基を有していてもよいC1-10アルコキシ基がより好ましく、アルキル基部分が直鎖状であり、かつ置換基を有していてもよいC1-6アルコキシ基がさらに好ましい。なかでも、アルキル基部分が直鎖状であり、かつ、C1-6アルコキシ基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、及びニトロ基、置換基を有していてもよいフェニル基からなる群より選択される1種以上の置換基を有していてもよいC1-6アルコキシ基が好ましく、置換基を有していてもよいフェニル基を置換基として有するC1-6アルコキシ基、置換基を有していないフェニル基を置換基として有するC1-6アルコキシ基、又は置換基を有していないC1-6アルコキシ基がより好ましく、置換基を有していないフェニル基を置換基として有するC1-6アルコキシ基がさらに好ましい。
R11が置換基を有していてもよい脂肪族ヘテロ環式基である場合、当該脂肪族ヘテロ環式基としては、含窒素脂肪族ヘテロ環式基が好ましく、ピロリジル基、ピペリジル基、ピペラジル基、又は、モルホリル基がより好ましく、ピロリジル基又はピペリジル基がさらに好ましい。これらの含窒素脂肪族ヘテロ環式基は、ヘテロ環を形成する窒素原子が保護基で保護されていてもよい。当該保護基としては、tert-ブトキシカルボニル(Boc)基、ベンジルオキシカルボニル基(Cbz基)、9-フルオレニルメチルオキシカルボニル基(Fmoc基)、アリルオキシカルボニル基(Alloc基)、2,2,2ートリエトキシカルボニル基(Troc基)等のアミンの保護基として用いられているものを適宜用いることができる。
R11が置換基を有していてもよい脂肪族ヘテロ環式基である場合、当該置換基としては、C1-6アルキル基、Cs-6アルケニル基、C1-6アルコキシ基、C1-6アルキルチオ基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、トリハロメチル基、ニトロ基、置換基を有していてもよいアミノ基、置換基を有していてもよいアリール基、及び置換基を有していてもよいヘテロアリール基等が挙げられる。置換基を有していてもよいアミノ基としては、前記R1の置換基として挙げられた基と同様の基を用いることができる。
R11が置換基を有していてもよい脂肪族ヘテロ環式基である場合、当該置換基を有していてもよい脂肪族ヘテロ環式基としては、置換基を有していてもよく、かつ窒素原子が保護基によって保護されていてもよい含窒素脂肪族ヘテロ環式基が好ましく、置換基を有していてもよく、かつ窒素原子が保護基によって保護されていてもよいピロリジル基又はピペリジル基がより好ましく、置換基を有していてもよく、かつ窒素原子がBoc基若しくはCbz基によって保護されていてもよいピロリジル基又はピペリジル基がさらに好ましく、置換基を有していてもよく、かつ窒素原子がBoc基若しくはCbz基によって保護されていてもよいピロリジル基がよりさらに好ましい。
R11が置換基を有していてもよいアリール基である場合、当該アリール基としては、置換基を有していてもよいフェニル基、又は、置換基を有していてもよいナフチル基が好ましく、置換基を有していてもよいフェニル基がより好ましい。
R11が置換基を有していてもよいアリール基である場合、当該置換基としては、C1-6アルキル基、C2-6アルケニル基、C1-6アルコキシ基、C2-6アシル基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、ニトロ基、置換基を有していてもよいアミノ基、ヒドロキシ基、アリール基、ヘテロアリール基等が挙げられる。置換基を有していてもよいアミノ基としては、前記R1の置換基として挙げられた基と同様の基を用いることができる。
R11が置換基を有していてもよいアリール基である場合、当該置換基を有していてもよいアリール基としては、置換基を有していてもよいフェニル基が好ましく、C1-6アルキル基、C2-6アルケニル基、C1-6アルコキシ基、C2-6アシル基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、ニトロ基、アミノ基、ジメチルアミノ基、メチルアミノ基、アセチルアミノ基、フェニル基、ハロゲン化フェニル基(ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)で置換されたフェニル基)、ピロリル基、ピラゾリル基、ピリジル基、ピラジニル基、ピリミジニル基、インドリル基、イソインドリル基、キノリル基、イソキノリル基、フラニル基、及びチエニル基からなる群より選択される1種以上の置換基を有していてもよいフェニル基がより好ましい。
R11が置換基を有していてもよいヘテロアリール基である場合、当該ヘテロアリール基としては、ピロリル基、ピラゾリル基、ピリジル基、ピラジニル基、ピリミジニル基、ピリダジニル基、オキサゾリル基、チアゾリル基、イミダゾリル基、インドリル基、イソインドリル基、キノリル基、イソキノリル基、フラニル基、又はチエニル基が好ましく、ピロリル基、ピラゾリル基、イミダゾリル基、ピリジル基、インドリル基、キノリル基、フラニル基、又はチエニル基がより好ましい。
R11が置換基を有していてもよいヘテロアリール基である場合、当該置換基としては、C1-6アルキル基、C2-6アルケニル基、C1-6アルコキシ基、C2-6アシル基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、ニトロ基、置換基を有していてもよいアミノ基、ヒドロキシ基、アリール基、ヘテロアリール基等が挙げられ、なかでも、C1-6アルキル基、C2-6アルケニル基、C1-6アルコキシ基、C2-6アシル基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、置換基を有していてもよいアミノ基、ヒドロキシ基、フェニル基、及びハロゲン化フェニル基からなる群より選択される1種以上の置換基が好ましい。置換基を有していてもよいアミノ基としては、前記R1の置換基として挙げられた基と同様の基を用いることができる。
R11が置換基を有していてもよいヘテロアリール基である場合、当該置換基を有していてもよいヘテロアリール基としては、ピロリル基、ピラゾリル基、ピリジル基、インドリル基、キノリル基、フラニル基、又はチエニル基が好ましく、これらのヘテロアリール基は、置換基を有していてもよい。化合物(S1)としては、C1-6アルキル基、C2-6アルケニル基、C1-6アルコキシ基、C2-6アシル基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、又はヨウ素原子)、アミノ基、ジメチルアミノ基、メチルアミノ基、アセチルアミノ基、フェニル基、及びハロゲン化フェニル基からなる群より選択される1種以上の置換基を有していてもよい、ピロリル基、ピラゾリル基、イミダゾリル基、ピリジル基、インドリル基、キノリル基、フラニル基、又はチエニル基がより好ましく、ハロゲン原子、フェニル基、及びハロゲン化フェニル基からなる群より選択される1種以上の置換基を有していてもよい、ピロリル基、ピラゾリル基、イミダゾリル基、ピリジル基、インドリル基、キノリル基、フラニル基、又はチエニル基がさらに好ましい。
本発明に係るフッ素含有化合物の製造方法は、例えば、基質とする酸素含有化合物が、下記一般式(S2)で表される化合物である場合に、フッ素含有化合物として、下記一般式(P2)で表される化合物を製造することができる。
一般式(S2)及び(P2)中、R12は、置換基を有していてもよいC1-30脂肪族炭化水素基、置換基を有していてもよいアルコキシ基、置換基を有していてもよい脂肪族ヘテロ環式基、置換基を有していてもよいアリール基、又は置換基を有していてもよいヘテロアリール基である。具体的には、置換基を有していてもよいC1-30脂肪族炭化水素基、置換基を有していてもよいアルコキシ基、置換基を有していてもよい脂肪族ヘテロ環式基、置換基を有していてもよいアリール基、及び置換基を有していてもよいヘテロアリール基としては、それぞれ、前記R11で挙げられた基と同じ基を用いることができ、前記R11で好ましいとされた基が好ましく用いられる。
本発明に係るフッ素含有化合物の製造方法は、例えば、基質とする酸素含有化合物が、下記一般式(S3)で表される化合物である場合に、フッ素含有化合物として、下記一般式(P3)で表される化合物を製造することができる。
一般式(S3)及び(P3)中、R13は、置換基を有していてもよいC1-30脂肪族炭化水素基である。具体的には、置換基を有していてもよいC1-30脂肪族炭化水素基としては、前記R11で挙げられた基と同じ基を用いることができ、前記R11で好ましいとされた基が好ましく用いられる。
化合物(S3)中のヒドロキシ基は、保護基で保護されていてもよい。当該保護基としては、アルコールの保護基として用いられている公知の保護基の中から適宜選択して用いることができる。当該保護基としては、具体的には、トリアルキルシリル基、テトラヒドロピラニル基(THP基)、メトキシメチルエーテル基(MOM基)、トリチル基(Tr基)、tert-ブチル基等が挙げられる。トリアルキルシリル基としては、例えば、トリメチルシリル基(TMS基)、トリエチルシリル基(TES基)、トリイソプロピルシリル基(TIPS基)、トリtert-ブチルシリル基(TBS基)等が挙げられる。
以下、本発明を実施例により説明するが、本発明はこれらの実施例に限定されない。
実施例等の分析に使用したNMR装置は、日本電子製JNM-ECS400(400MHz)又はECZ500R(500MHz)である。19F NMRではC6F6を-162PPMの基準値とした。実施例中に記載する収率(%)の単位は、モル%である。
[製造例1]
以降の実験において、脱酸素フッ素化剤として用いた[Cs(G4)2][SF5]は、Matsumotoらの方法(非特許文献2)にしたがって合成した。

[実施例1]
シリル保護アルコールを基質として、[Cs(G4)2][SF5](以下、「Cs(G4)2SF5」ということがある)による脱酸素フッ素化反応における溶媒の影響を調べた。

シリル保護アルコールとして、下記化合物(1)を用いた。
溶媒として、G4、THF、DME、ジエチレングリコールジメチルエーテル(CAS No.: 111-96-6)(G2)、アセトニトリル(MeCN)、ジメチルホルムアミド(DMF)、N-メチルピロリドン(NMP)、DMPU、DMA、HMPA、及びジメチルスルホキシド(DMSO)を用いた。
溶媒として、G4、THF、DME、ジエチレングリコールジメチルエーテル(CAS No.: 111-96-6)(G2)、アセトニトリル(MeCN)、ジメチルホルムアミド(DMF)、N-メチルピロリドン(NMP)、DMPU、DMA、HMPA、及びジメチルスルホキシド(DMSO)を用いた。
アルゴンガスを満たしたグローブボックス中で、出発物質(基質)として化合物(1)(0.1mmol)と、内標としての1,4-ジフルオロベンゼン(0.5mmol、5μL)とを、ガラスバイアル中で溶媒(0.5mL)に溶解させて、基質溶液を調製した。別のガラスバイアル中に、固体のCs(G4)2SF5(0.15mmol、基質に対して、1.5等量)を仕込み、前記基質溶液をシリンジを用いて加え、よく混合することによって、均一な溶液である反応用混合物を調製した。得られた反応用混合物は、FEP-inletを内包するNMRチューブに移し、PTFE plugを用いてシールした。続いて、NMRチューブを60℃に加熱して、反応を実施した。反応の進行は、19F NMRによって追跡した。
各反応の反応時間、基質化合物の変換率(フッ素化率)(%)、及びNMR測定データから算出された収率(%)を表1に示す。表1に示すように、DMPU、G4、DMA、及びHMPAを溶媒とした反応系では、72時間の反応で70%以上の高い収率で、フッ素含有化合物が得られた。また、MeCNを溶媒とした場合には、22時間の反応で低転化率ではあるが、Cs(G4)2SF5の分解が進行せず、副生物が少なかった。特に、DMPUは、副産物が少なく、かつ収率が非常に高かったことから、Cs(G4)2SF5による脱酸素フッ素化反応の反応溶媒として最適であった。一方で、Cs(G4)2SF5が分解されやすいTHFやDME中では、フッ素含有化合物は得られなかった。
[実施例2]
各種の基質化合物に対して、Cs(G4)2SF5を脱酸素フッ素化剤、DMPUを反応溶媒として用いて脱酸素フッ素化反応を行い、フッ素含有化合物を合成した。

アルゴンガスを満たしたグローブボックス中で、基質化合物(0.05mmol)と内標としての1,4-ジフルオロベンゼン(0.5mmol、5μL)とを、ガラスバイアル中でDMPU(0.4mL)に溶解させて、基質溶液を調製した。別のガラスバイアル中に、固体のCs(G4)2SF5(0.1~0.2mmol、基質に対して、2~4等量)を仕込み、前記基質溶液をシリンジを用いて加え、よく混合することによって、均一な溶液である反応用混合物を調製した。得られた反応用混合物は、FEP-inletを内包するNMRチューブに移し、PTFE plugを用いてシールした。続いて、NMRチューブを所定の温度に加熱して、反応を実施した。反応の進行は、19F NMRによって追跡した。
反応生成物の単離は、以下の通りにして行った。
アルゴンガスを満たしたグローブボックス中で攪拌子を入れたPTFE製丸底試験管に、出発物質(0.2mmol)を仕込んだ。ガラスバイアル中で、Cs(G4)2SF5(0.4~0.8mmol、基質に対して、2~4等量)をDMPU(1.5mL)に溶解した溶液を調製し、前記丸底試験管に仕込んだ。当該PTFE製丸底試験管をスクリューキャップを用いてシールし、所定の温度で所定時間反応させた。続いて、反応粗液を室温まで冷却した後、水(10mL)を加え、ジクロロメタン溶媒を用いて生成物を抽出した。得られた有機層は、水と飽和食塩水にて洗浄し、硫酸マグネシウムにて脱水した。さらに、溶媒を留去することによって粗生成物を得た。得られた粗生成物は、シリカゲルカラムクロマトグラフィーにて精製した。
アルゴンガスを満たしたグローブボックス中で攪拌子を入れたPTFE製丸底試験管に、出発物質(0.2mmol)を仕込んだ。ガラスバイアル中で、Cs(G4)2SF5(0.4~0.8mmol、基質に対して、2~4等量)をDMPU(1.5mL)に溶解した溶液を調製し、前記丸底試験管に仕込んだ。当該PTFE製丸底試験管をスクリューキャップを用いてシールし、所定の温度で所定時間反応させた。続いて、反応粗液を室温まで冷却した後、水(10mL)を加え、ジクロロメタン溶媒を用いて生成物を抽出した。得られた有機層は、水と飽和食塩水にて洗浄し、硫酸マグネシウムにて脱水した。さらに、溶媒を留去することによって粗生成物を得た。得られた粗生成物は、シリカゲルカラムクロマトグラフィーにて精製した。
本発明に係るフッ素含有化合物の製造方法では、グライム配位アルカリ金属イオンと塩を形成して安定化されたSF5
-を脱酸素フッ素化剤として用いているため、ケトン基、アルデヒド基、ヒドロキシ基等を有する多種多様な酸素含有化合物に対して、比較的温和な条件で脱酸素フッ素化を行うことができる。このため、本発明は、多種多様なフッ素含有化合物の合成に有用である。
Claims (4)
- 下記一般式(E1)
で表される脱酸素フッ素化剤を用いて、1,3-ジメチル-3,4,5,6-テトラヒドロ-2(1H)-ピリミジノン、テトラエチレングリコールジメチルエーテル、ジメチルアセトアミド、ヘキサメチル亜リン酸トリアミド、及びアセトニトリルからなる群より選択される1種以上の溶媒中で、アルデヒド基、ケトン基、カルボキシ基、又はヒドロキシ基を有する酸素含有化合物を脱酸素フッ素化して、フッ素含有化合物を製造する、フッ素含有化合物の製造方法。
