Abstract
凝集物が少ない粉末を形成でき、また、水系媒体への再分散性にも優れる粉末を形成することができ、しかも、酸性条件下にあっても再分散性に優れる粉末を形成することができる組成物を提供する。 本発明の組成物は、カンナビノイドと、ショ糖脂肪酸エステルと、水溶性高分子化合物とを含み、前記ショ糖脂肪酸エステルは、平均エステル化度が1.00~1.15である。
Description
本発明は、カンナビノイドを含む組成物に関する。
カンナビノイドは、アサの葉及び種子皮に含まれており、カンナビジオール(CBD)等の種々の成分を有することが知られている。カンナビジオールは、特異な効能を有することから、食品添加剤等の食品分野、及び、製剤等の医療分野に広く適用されており、利用価値の高い成分である。
例えば、特許文献1には、クルクミンと、少なくとも1つのカンナビノイドとを組み合わせた可溶化物が提案されており、斯かる可溶化物により、水と混合したときに濁りのない混合物を与えることを可能としている。
しかしながら、カンナビノイド、とりわけカンナビジオールは、粉末化することが難しく、例えば、粉末中に凝集物が混在しやすいという問題があった。また、一度粉末化されると、再度の水系媒体への分散性が悪いため、例えば、食品や製剤用途へのカンナビノイドを含む粉末を添加剤として使用するにも、再分散性能が低いことから、均一な混合が難しい場合もあった。
本発明は、上記に鑑みてなされたものであり、凝集物が少ない粉末を形成でき、また、水系媒体への再分散性にも優れる粉末を形成することができ、しかも、酸性条件下にあっても再分散性に優れる粉末を形成することができる組成物を提供することを目的とする。
本発明者らは、上記目的を達成すべく鋭意研究を重ねた結果、水溶性高分子化合物と共に特定のショ糖脂肪酸エステルを必須成分とすることにより上記目的を達成できることを見出し、本発明を完成するに至った。
すなわち、本発明は、例えば、以下の項に記載の主題を包含する。

本発明の組成物は、凝集物が少ない粉末を形成でき、また、水系媒体への再分散性にも優れる粉末を形成することができ、しかも、酸性条件下にあっても再分散性に優れる粉末を形成することができる。
以下、本発明の実施形態について詳細に説明する。なお、本明細書中において、「含有」及び「含む」なる表現については、「含有」、「含む」、「実質的にからなる」及び「のみからなる」という概念を含む。
1.組成物
本発明の組成物は、カンナビノイドと、ショ糖脂肪酸エステルと、水溶性高分子化合物とを含み、前記ショ糖脂肪酸エステルは、平均エステル化度が1.00~1.15である。本発明の組成物は、カンナビノイドを含む組成物において、特定の前記ショ糖脂肪酸エステルと、水溶性高分子化合物とを含むので、凝集物が少ない粉末を形成できる。
本発明の組成物は、カンナビノイドと、ショ糖脂肪酸エステルと、水溶性高分子化合物とを含み、前記ショ糖脂肪酸エステルは、平均エステル化度が1.00~1.15である。本発明の組成物は、カンナビノイドを含む組成物において、特定の前記ショ糖脂肪酸エステルと、水溶性高分子化合物とを含むので、凝集物が少ない粉末を形成できる。
また、本発明の組成物は、水系媒体への再分散性にも優れる粉末を形成することができ、しかも、酸性条件下にあっても再分散性に優れる粉末を形成することができる。本明細書において、「酸性条件下」とは、乳化組成物のpHが7以下、好ましくは4以下であることを意味する。
従って、本発明の組成物を粉末状の添加剤等として用いた場合に、良好な分散状態を維持することが可能となる。
(カンナビノイド)
本発明の組成物に含まれるカンナビノイドとしては、例えば、アサ由来のカンナビノイドに含まれる種々の化合物群(具体的には、カンナビジオール(CBD)、テトラヒドロカンナビノール等)の1種又は2種以上を使用することができる。
本発明の組成物に含まれるカンナビノイドとしては、例えば、アサ由来のカンナビノイドに含まれる種々の化合物群(具体的には、カンナビジオール(CBD)、テトラヒドロカンナビノール等)の1種又は2種以上を使用することができる。
カンナビノイドはカンナビジオールを含むことが好ましい。この場合、溶媒、特には水性溶媒への分散性に特に優れ、粉末化も容易である上、食品分野、化粧分野、飼料分野、又は、医薬分野に特段適用しやすい。カンナビノイドはカンナビジオールを50質量%以上含むことが好ましく、80質量%以上含むことがより好ましく、90質量%以上含むことがさらに好ましく、95質量%以上含むことが特に好ましい。カンナビノイドはカンナビジオール1種のみとすることもできる。
カンナビノイドは、例えば、公知の方法で製造することができる。あるいは、市販されているカンナビノイドを本発明の組成物に適用することができる。同様に、カンナビジオールも、例えば、公知の方法で製造することができ、あるいは、市販されているカンナビジオールを本発明の組成物に適用することができる。
(ショ糖脂肪酸エステル)
本発明の組成物に含まれるショ糖脂肪酸エステルは、前述のように、平均エステル化度が1.00~1.15の範囲である。つまり、ショ糖脂肪酸エステルの平均エステル化度は、1.00以上、1.15以下である。念のための注記に過ぎないが、本明細書において、「ショ糖脂肪酸エステル」とは、ショ糖と脂肪酸とがエステル化反応により形成される化合物であることを意味する。
本発明の組成物に含まれるショ糖脂肪酸エステルは、前述のように、平均エステル化度が1.00~1.15の範囲である。つまり、ショ糖脂肪酸エステルの平均エステル化度は、1.00以上、1.15以下である。念のための注記に過ぎないが、本明細書において、「ショ糖脂肪酸エステル」とは、ショ糖と脂肪酸とがエステル化反応により形成される化合物であることを意味する。
平均エステル化度が1.00~1.15であるショ糖脂肪酸エステルが組成物に含まれることで、組成物を溶媒(特に水性溶媒)に分散させたときに、カンナビノイドの溶媒への分散性に優れ、また、組成物が粉末であっても溶媒(特に水性溶媒)への分散性に優れ、たとえ溶媒(特に水性溶媒)が酸性条件下であっても分散性に優れるものである。
ショ糖脂肪酸エステルの平均エステル化度が1.15を超過すると、組成物が粉末化したときに凝集物が多くなり、また、粉末の溶媒への分散性も悪化する。
ショ糖脂肪酸エステルの平均エステル化度は1.01以上であることが好ましい。また、ショ糖脂肪酸エステルの平均エステル化度は1.12以下であることが好ましく、1.10以下であることがさらに好ましく、1.08以下であることが特に好ましい。
本発明において、ショ糖脂肪酸エステルの平均エステル化度は、下記(1)式によってショ糖脂肪酸エステルの平均エステル化度Xを算出することができる。

上記(1)式に、既知であるNs、Nx、Ny、OHV、MwSug及びMwFaを代入してXを算出することができる。なお、Ns、Nx、Ny、OHV、MwSug及びMwFaは、公知の方法で測定することができる。
上記平均エステル化度の算出方法からもわかるように、本発明におけるショ糖脂肪酸エステルは、エステル化度が異なるショ糖脂肪酸エステル分子の集合体であるといえる。
本発明の組成物において、ショ糖脂肪酸エステルとして市販品を使用する場合、ショ糖脂肪酸エステルのメーカー保証値あるいはカタログ値等、メーカーが開示する値を平均エステル化度の値として採用することができる。
ショ糖脂肪酸エステルは平均エステル化度が前記範囲である限り、その種類は特に限定されず、例えば、公知のショ糖脂肪酸エステルを広く本発明の組成物に適用することができる。
ショ糖脂肪酸エステルにおいて、ショ糖脂肪酸エステルの脂肪酸部位(RCOO-部位)の種類は特に限定されない。ショ糖脂肪酸エステルの脂肪酸部位は、飽和脂肪酸由来の部位であってもよいし、あるいは、不飽和脂肪酸由来の部位であってもよく、好ましくは飽和脂肪酸由来である。
ショ糖脂肪酸エステルにおいて、ショ糖脂肪酸エステルの脂肪酸部位の炭素数は特に限定されず、例えば、カンナビノイドの分散性を高めやすいという点で、脂肪酸部位の炭素数(エステル炭素も含む)は30以下であることが好ましく、25以下であることがより好まく、20以下であることがさらに好ましく、18以下であることが特に好ましい。また、カンナビノイドの分散性を高めやすいという点で、脂肪酸部位の炭素数(エステル炭素も含む)は8以上であることが好ましく、10以上であることがより好ましく、12以上であることがさらに好ましく、14以上であることが特に好ましい。ショ糖脂肪酸エステルにおける脂肪酸部位は、例えば、ラウリン酸、パルミチン酸、ステアリン酸等に由来するものが好ましい。
ショ糖脂肪酸エステルにはモノエステル体、ジエステル体及びトリエステル体が存在する。本発明の組成物に含まれるショ糖脂肪酸エステルは、それらのいずれのエステル体であってもよいし、1種のみならず2種又は全てのエステル体を含むこともできる。言い換えれば、ショ糖脂肪酸エステルは、モノエステル体、ジエステル体及びトリエステル体からなる群より選ばれる1種以上のショ糖脂肪酸エステルを含み得る。
ショ糖脂肪酸エステルの製造方法は特に限定されず、例えば、公知の製造方法を広く採用することができる。例えば、ショ糖と脂肪酸エステルとのエステル交換反応により、ショ糖脂肪酸エステルを合成することができる。脂肪酸エステルとしては、脂肪酸のメチルエステルを挙げることができる。
ショ糖脂肪酸エステルの製造方法で使用するショ糖は、例えば、公知の方法で合成してもよく、あるいは、市販品からショ糖を入手することができる。ショ糖は、ショ糖脂肪酸エステル製造時に未反応であったショ糖を回収したもの(いわゆる、「回収糖」)であってもよい。ショ糖の形態は、固体の状態であってもよく、溶媒に溶解した溶液状態であってもよい。ショ糖脂肪酸エステルの製造方法で使用する脂肪酸は、例えば、公知の方法で合成してもよく、あるいは、市販品から脂肪酸を入手することができる。
ショ糖脂肪酸エステルの平均エステル化度を調節する方法も特に限定されず、例えば、公知の方法を広く採用することができる。例えば、ショ糖脂肪酸エステルの製造で使用するショ糖及び脂肪酸又はそのエステルの使用割合、脂肪酸の種類、エステル化反応条件等の種々の合成条件を調製することで、得られるショ糖脂肪酸エステルの平均エステル化度を所望の範囲に制御することができる。
その他、平均エステル化度が既知であって、異なる平均エステル化度のショ糖脂肪酸エステルを2種以上混合することで、所望の範囲に制御されたショ糖脂肪酸エステルを得ることもできる。この場合、各ショ糖脂肪酸エステルのエステル化度及び混合質量比に基づいて、平均エステル化度を容易に所望の範囲に調整することができる。
本発明の組成物において、ショ糖脂肪酸エステルの含有割合は特に限定されない。例えば、組成物が粉末であるときの溶媒への分散性が高まりやすい点で、組成物中のカンナビノイドの全質量を100質量部としたときのショ糖脂肪酸エステルの含有量が100質量部以上であることが好ましく、300質量部以上であることがより好ましく、500質量部以上であることがさらに好ましく、600質量部以上であることが特に好ましく、また、5000質量部以下であることが好ましく、3000質量部以下であることがより好ましく、2000質量部以下であることがさらに好ましく、1500質量部以下であることが特に好ましい。
本発明の組成物は1種又は2種以上のショ糖脂肪酸エステルを含むことができる。
(水溶性高分子化合物)
本発明の組成物に含まれる水溶性高分子化合物は、水に溶解することができる高分子化合物であることを意味する。例えば、水100質量部に対して、0.1質量部以上溶解する高分子化合物を水溶性高分子化合物とすることができる。
本発明の組成物に含まれる水溶性高分子化合物は、水に溶解することができる高分子化合物であることを意味する。例えば、水100質量部に対して、0.1質量部以上溶解する高分子化合物を水溶性高分子化合物とすることができる。
本発明の組成物が含むことができる水溶性高分子化合物としては、例えば、公知の水溶性高分子化合物を広く挙げることができ、セルロース誘導体、ポリビニルピロリドン等を挙げることができる他、ポリアクリル酸又はその塩、ポリメタクリル酸又はその塩、ポリビニルアルコール、ポリアルキレンオキシド、ポリスチレンスルホン酸又はその塩等を挙げることができる。
中でも、前記水溶性高分子化合物は、セルロース誘導体及びポリビニルピロリドンからなる群より選ばれる少なくとも1種であることが好ましい。この場合、組成物が粉末であっても凝集物が生じにくく、また、溶媒(特に水性溶媒)への分散性がより優れるものとなり、たとえ溶媒(特に水性溶媒)が酸性条件下であっても分散性がより優れるものになりやすい。
前記セルロース誘導体としては、例えば、ヒドロキシアルキルセルロースを挙げることができ、具体的には、ヒドロキシメチルセルロース、ヒドロキシエチルセルロース、ヒドロキシプロピルセルロース等を挙げることができる。
前記水溶性高分子化合物の重量平均分子量は特に限定されず、例えば、10000~100000の範囲とすることができる。
水溶性高分子化合物は、公知の方法で製造することができ、また、市販品等から入手することも可能である。
本発明の組成物において、水溶性高分子化合物の含有割合は特に限定されない。例えば、前記ショ糖脂肪酸エステルと前記水溶性高分子化合物との質量比が0.5~30であることが好ましい。すなわち、本発明の組成物は、「ショ糖脂肪酸エステルの質量/水溶性高分子化合物質量」の値が0.5~30であることが好ましい。この場合、組成物が粉末であっても凝集物が生じにくく、また、溶媒(特に水性溶媒)への分散性がより優れるものとなり、たとえ溶媒(特に水性溶媒)が酸性条件下であっても分散性がより優れるものになりやすい。
本発明の組成物は、前記ショ糖脂肪酸エステルと前記水溶性高分子化合物との質量比(ショ糖脂肪酸エステルの質量/水溶性高分子化合物質量)の値が2以上であることがより好ましく、4以上であることがさらに好ましく、5以上であることが特に好ましく、また、25以下であることがより好ましく、20以下であることがさらに好ましく、15以下であることが特に好ましい。
本発明の組成物は1種又は2種以上の水溶性高分子化合物を含むことができる。
(その他成分)
本発明の組成物は、本発明の効果が阻害されない限り、カンナビノイド及びショ糖脂肪酸エステル以外に他の成分を含むことができる。例えば、本発明の組成物は、前記ショ糖脂肪酸エステル以外の界面活性剤を含むこともできる。斯かる界面活性剤は、例えば、公知の界面活性剤を広く挙げることができ、例として、ソルビタン脂肪酸エステル、ポリグリセリン脂肪酸エステル等を挙げることができる。
本発明の組成物は、本発明の効果が阻害されない限り、カンナビノイド及びショ糖脂肪酸エステル以外に他の成分を含むことができる。例えば、本発明の組成物は、前記ショ糖脂肪酸エステル以外の界面活性剤を含むこともできる。斯かる界面活性剤は、例えば、公知の界面活性剤を広く挙げることができ、例として、ソルビタン脂肪酸エステル、ポリグリセリン脂肪酸エステル等を挙げることができる。
本発明の組成物は、油脂を含有することもできる。この場合、カンナビノイドの分散性が向上しやすくなるので、本発明の組成物は、食品分野、化粧分野、飼料分野、又は、医薬分野等の各種添加剤に適用しやすい。
油脂としては食用油脂を挙げることができ、具体的には、MCTオイル等に代表される中鎖脂肪酸トリグリセリド等の合成油脂が挙げられる他、綿実油、ひまし油、大豆油、菜種油、コーン油、オリーブ油、パーム油、サフラワー油、米油、ひまわり油、胡麻油等の植物性油脂;ラード、牛脂、乳脂、魚油等の動物性油脂が挙げられる。油脂は、例えば、市販品を広く使用することができる。組成物に含まれる油脂は1種単独又は2種以上とすることができる。
本発明の組成物が油脂を含む場合、その含有割合は、例えば、カンナビノイドの全質量を100質量部に対し、20質量部以上であることが好ましく、40質量部以上であることがより好ましく、60質量部以上であることがさらに好ましく、80質量部以上であることが特に好ましく、また、500質量部以下であることが好ましく、400質量部以下であることがより好ましく、300質量部以下であることがさらに好ましく、200質量部以下であることが特に好ましい。
本発明の組成物は、その他添加剤を含むことができ、例えば、食品用途で広く使用されている各種添加剤、医薬品用途で広く使用されている各種添加剤を挙げることができる。本発明の組成物が、その他添加剤を含む場合、その含有割合は、例えば、組成物全質量に対し、50質量%以下、好ましくは30質量%以下、より好ましくは20質量%以下、さらに好ましくは10質量%以下、特に好ましくは1質量%以下である。
(組成の形態)
本発明の組成物は、分散液等の液状の形態であってもよいし、粉末等の固体状の形態であってもよい。

本発明の組成物が分散液である場合、本発明の組成物はさらに溶媒を含むことができる。斯かる溶媒としては、カンナビノイド(とりわけカンナビジオール)の分散性が高まりやすく、また、乾燥させて粉末化させることが容易である点で、水性溶媒であることが好ましい。水性溶媒としては、水、アルコール分を挙げることができる。
アルコール分は、メタノール、エタノール、イソプロパノール等の低級アルコール化合物の他、グリセリン等の多価アルコール化合物も挙げることができる。
水性溶媒は、水と、前記低級アルコール及び/又は前記多価アルコール化合物との混合溶媒を挙げることができ、また、水性溶媒は、前記低級アルコール化合物及び前記多価アルコール化合物等のアルコール分のみであってもよい。水性溶媒が低級アルコール及び多価アルコールの混合物を含む場合、低級アルコールメタノール、エタノール、イソプロパノール等が挙げられ、多価アルコール化合物としてはグリセリンが挙げられる。
本発明の組成物が分散液である場合、水性溶媒は、少なくとも低級アルコール化合物及び多価アルコール化合物等のアルコール分を含むことがより好ましい。この場合、組成物を粉末化した後、斯かる粉末の溶媒(特に水性溶媒)への分散性がより優れるものとなり、たとえ溶媒(特に水性溶媒)が酸性条件下であっても分散性がより優れるものになりやすい。
水性溶媒が低級アルコール化合物及び多価アルコール化合物を含むアルコール分を含む場合、低級アルコール化合物及び多価アルコール化合物の割合は特に限定されず、例えば、低級アルコール化合物100質量部に対して、多価アルコール化合物の含有割合が10~50質量部である。
本発明の組成物が水性溶媒を含む分散液である場合において、水性溶媒が低級アルコール化合物及び多価アルコール化合物等のアルコール分を含む場合、アルコール分の含有割合は、例えば、カンナビノイドの全質量を100質量部に対し、100質量部以上であることが好ましく、500質量部以上であることがより好ましく、1000質量部以上であることがさらに好ましく、また、10000質量部以下であることが好ましく、5000質量部以下であることがより好ましく、3000質量部以下であることがさらに好ましく、2000質量部以下であることが特に好ましい。
本発明の組成物が溶媒を含む分散液である場合、溶媒の総含有量は特に限定されない。例えば、溶媒100質量部に対するカンナビノイド及びショ糖脂肪酸エステルの含有量は0.01~20質量部であることが好ましく、0.05~10質量部であることがより好ましく、0.1~5質量部であることがさらに好ましく、0.2~3質量部であることが特に好ましい。
本発明の組成物が溶媒を含む分散液である場合、斯かる分散液のメディアン径(D50)は特に限定されない。例えば、分散安定性が良好になりやすく、また、後記する粉末化が容易に行えるという点で、分散液のメディアン径(D50)は1~150nmであることが好ましく、1~100nmであることがより好ましく、1~50nmであることがさらに好ましく、1~20nmであることが特に好ましい。分散液のメディアン径(D50)は、動的光散乱法により、粒子径測定装置(商品名:nanoSAQLA、大塚電子社製)を用いて測定することができる。メディアン径(D50)とは、散乱強度分布において、頻度の累計が50%になる粒子径D50を意味する。
本発明の組成物は粉末であってもよい。この場合、粉末の平均粒子径は特に限定されない。例えば、各種添加剤として好適に使用できるという点で、粉末の体積平均粒子径は、0.1~500μmであることが好ましく、1~150μmであることがさらに好ましく、10~100μmであることが特に好ましい。粉末の体積平均粒子径は、粒子径分布測定装置(商品名:MT3000II、マイクロトラックベル社製)を用いて測定することができる。本発明の組成物が粉末である場合、斯かる粉末は、例えば、凝集粒子で構成される。
本発明の組成物が粉末である場合であっても前述の食用油を前述の量で含むことができ、また、食用油以外の添加剤を含むこともできる。ただし、前述の水性溶媒(例えば、水、並びに、低級アルコール化合物及び多価アルコール化合物等のアルコール分)は粉末化する過程で一部または全部が揮発し得る。本発明の組成物が粉末である場合、粉末中に含まれる低級アルコール化合物及び多価アルコール化合物の含有割合は、10~25質量%程度である。
本発明の組成物が粉末である場合、その製造方法は特に限定されず、例えば、分散液である本発明の組成物を、適宜の方法で溶媒を乾燥させることで、粉末化することができる。溶媒の乾燥方法は特に限定されず、公知の方法、例えば、加熱乾燥、噴霧乾燥、凍結乾燥等の種々の方法を挙げることができる。従って、本発明の一態様として、前記分散液の乾燥物たる粉末を挙げることができる。斯かる方法で得られる粉末は、例えば、油脂に溶解したカンナビノイドの水性分散液が乾燥することで形成される粉末である。
前記粉末は、カンナビノイド、特定のショ糖脂肪酸エステル及び水溶性高分子化合物を含有するので、溶媒(特に水性溶媒)への分散性がより優れるものとなり、たとえ溶媒(特に水性溶媒)が酸性条件下であっても分散性がより優れるものになりやすい。すなわち、本発明の組成物が粉末である場合は、水系媒体への再分散性に優れ、しかも、酸性条件下にあっても再分散性に優れる。従って、本発明の組成物を粉末状の添加剤等として用いた場合に、良好な分散状態を維持することが可能となる。また、再分散性に優れるという性質を有することで、粉末の添加量を多くすることができ、すなわち、従来よりもカンナビノイドの割合(濃度)を高くすることができ、高濃度のカンナビノイドを含む組成物を提供することも可能となる。
本発明の組成物は種々の用途の適用することができ、とりわけ、従来からカンナビジオールを含む組成物が適用されている用途に広く適用することができる。具体的に本発明の組成物は、食品用、化粧用、飼料用、又は、医薬用等に適用することができる。本発明の組成物が分散液及び粉末のいずれの形態であっても、前記各種用途に使用することができる。本明細書において、食品用とは、飲料、サプリメント、健康食品、機能性食品、美容を目的とする食品、ペットなどの動物用の餌等を広く包含する。また、本明細書において、飼料用とは、各種畜産分野で用いることができる飼料を広く含み、また、養殖に用いられる飼料も含む。本明細書において、医薬用とは、ヒトへの服用又は投与のための医薬を含む他、動物への服用又は投与のための医薬を含む。本発明の組成物が食品用、化粧用又は医薬用に適用される場合、いずれの用途においても美容を目的とするものを含む。
以上より、本発明の組成物は、例えば、食品用添加剤、飼料用添加剤、医薬品用添加剤及び化粧品用添加剤として特に好適に使用することができる。
(組成物の調製方法)
本発明の組成物の製造方法は特に限定されず、例えば、カンナビノイド、ショ糖脂肪酸エステル及び水溶性高分子化合物を用いて、種々の方法で製造することができる。例えば、カンナビノイドを含む原料液と、ショ糖脂肪酸エステル及び水溶性高分子化合物を含む原料液とをそれぞれ順不同に混合させる方法が挙げられる。カンナビノイドを含む原料液は、各種オイルを含むことができ、例えば、前記食用油脂が挙げられ、中でもMCTオイル等に代表される中鎖脂肪酸トリグリセリド、その他、綿実油、ひまし油等を好ましく使用できる。MCTオイルは、例えば、市販品を広く使用することができる。
本発明の組成物の製造方法は特に限定されず、例えば、カンナビノイド、ショ糖脂肪酸エステル及び水溶性高分子化合物を用いて、種々の方法で製造することができる。例えば、カンナビノイドを含む原料液と、ショ糖脂肪酸エステル及び水溶性高分子化合物を含む原料液とをそれぞれ順不同に混合させる方法が挙げられる。カンナビノイドを含む原料液は、各種オイルを含むことができ、例えば、前記食用油脂が挙げられ、中でもMCTオイル等に代表される中鎖脂肪酸トリグリセリド、その他、綿実油、ひまし油等を好ましく使用できる。MCTオイルは、例えば、市販品を広く使用することができる。
カンナビノイドと、溶媒との使用割合は特に限定されず、例えば、カンナビノイド100質量部あたり、溶媒の使用量を50~200質量部とすることができ、80~150質量部とすることが好ましい。
ショ糖脂肪酸エステル及び水溶性高分子化合物を含む原料液は、溶媒を含むことができ、例えば、前述の水性溶媒(例えば、水、低級アルコール化合物及び多価アルコール化合物等)を挙げることができる。多価アルコール化合物は、カンナビノイドを含む原料液と、ショ糖脂肪酸エステル及び水溶性高分子化合物を含む原料液とを混合してから加えることもできる。ショ糖脂肪酸エステルと、溶媒との使用割合は特に限定されず、例えば、ショ糖脂肪酸エステル100質量部あたり、溶媒の使用量を50~200質量部とすることができ、80~150質量部とすることが好ましい。また、水溶性高分子化合物と、溶媒との使用割合は特に限定されず、例えば、水溶性高分子化合物100質量部あたり、溶媒の使用量を50~200質量部とすることができ、80~150質量部とすることが好ましい。
例えば、カンナビノイドと溶媒とを含む原料液、ショ糖脂肪酸エステルと溶媒とを含む原料液、及び、水溶性高分子化合物と溶媒とを含む原料液を混合させる場合、混合時の温度も特に限定されず、例えば、20~100℃、好ましくは30~90℃、より好ましくは40~80℃である。混合方法も特に限定されず、例えば、公知の混合機を広く使用することができる。上記のように得られる組成物にさらに水等を加えて、濃度を調節することもできる。
カンナビノイドを含む原料液と、ショ糖脂肪酸エステル及び水溶性高分子化合物を含む原料液とを混合して得られる組成物は、前述のように、必要に応じて適宜の乾燥処理を行って粉末化することができる。
以下、実施例により本発明をより具体的に説明するが、本発明はこれら実施例の態様に限定されるものではない。
(製造例1)
攪拌機、温度計、減圧装置、およびジメチルスルホキシド(DMSO)の還流装置を備えた1Lフラスコに、DMSO400gとショ糖34.2gを混合し、95℃で溶解した。ここに、炭酸カリウム2.4gおよび脂肪酸エステルとして、ステアリン酸メチル15.0gを加え、2kPaの減圧下、95℃で3時間反応させた。続いて、乳酸を加えて中和した後、DMSOを減圧留去した。得られた粗生成物をメチルエチルケトン200gに溶解し、10%飽和食塩水200gで3回洗浄し、溶媒を減圧留去することにより、ショ糖脂肪酸エステルAを得た。ショ糖脂肪酸エステルAの平均エステル化度は1.01であった。
攪拌機、温度計、減圧装置、およびジメチルスルホキシド(DMSO)の還流装置を備えた1Lフラスコに、DMSO400gとショ糖34.2gを混合し、95℃で溶解した。ここに、炭酸カリウム2.4gおよび脂肪酸エステルとして、ステアリン酸メチル15.0gを加え、2kPaの減圧下、95℃で3時間反応させた。続いて、乳酸を加えて中和した後、DMSOを減圧留去した。得られた粗生成物をメチルエチルケトン200gに溶解し、10%飽和食塩水200gで3回洗浄し、溶媒を減圧留去することにより、ショ糖脂肪酸エステルAを得た。ショ糖脂肪酸エステルAの平均エステル化度は1.01であった。
(製造例2)
ステアリン酸メチルの使用量を15.5gに変更したこと以外は製造例1と同様の方法で脂肪酸エステルBを得た。ショ糖脂肪酸エステルBの平均エステル化度は1.04であった。

(製造例3)

(実施例1)
カンナビジオール(CBD、Sigma-Aldrichs社製)0.1gと、油脂としてMCTオイル(商品名:アクターM-1、理研ビタミン社製)0.1gとを70℃で混合して原料液1を得た。別の容器に、界面活性剤として、製造例1で得られたショ糖脂肪酸エステルAを1.2gと、エタノールを1gと、水溶性高分子化合物であるヒドロキシプロピルセルロース(商品名:CELNY-SSL(重量平均分子量40000)、日本曹達社製)0.2gとを70℃で混合することで原料液2を得た。原料液1及び2、並びに、グリセリン0.3gを70℃で混合することにより、混合物を得た。この混合物に水24gを加え、-50℃のエタノール低温槽で凍結させた後、凍結乾燥機を用いて乾燥処理をすることで、粉末を得た。

(実施例2)
水溶性高分子化合物(HPC-SSL)の使用量を0.1gに変更し、グリセリンの使用量を0.4gに変更したこと以外は実施例1と同様の方法で組成物を得た。

<粉末の再分散性評価>
各実施例及び比較例で得られた粉末の水分散液を調製した。具体的には、スターラーで撹拌しているクエン酸水溶液(pH3.4)に、CBD濃度が0.1質量%となるように粉末を加え、3分間撹拌することでサンプルを調製した。得られたサンプルを目視し、下記判定基準に基づき、粉末の再分散性を評価した。
A:沈殿物が見られず、非常に良好な再分散性を有していた。
B:沈殿物が見られ、再分散性は劣るものであった。
各実施例及び比較例で得られた粉末の水分散液を調製した。具体的には、スターラーで撹拌しているクエン酸水溶液(pH3.4)に、CBD濃度が0.1質量%となるように粉末を加え、3分間撹拌することでサンプルを調製した。得られたサンプルを目視し、下記判定基準に基づき、粉末の再分散性を評価した。
A:沈殿物が見られず、非常に良好な再分散性を有していた。
B:沈殿物が見られ、再分散性は劣るものであった。
<全光線透過率>
粉末の再分散性評価で調製したサンプルを25℃の状態で全光線透過率をヘーズメーター NDH4000(日本電色工業社製)により測定し、下記判定基準に基づき、評価した。
A:全光線透過率95%以上であった。
B:全光線透過率90%以上95%未満であった。
C:全光線透過率85%以上90%未満であった。
D:全光線透過率80%以上85%未満であった。
E:全光線透過率80%未満であった。

A:全光線透過率95%以上であった。
B:全光線透過率90%以上95%未満であった。
C:全光線透過率85%以上90%未満であった。
D:全光線透過率80%以上85%未満であった。
E:全光線透過率80%未満であった。
各実施例及び比較例で得られた粉末をpH3.4のクエン酸水溶液に再分散させてサンプルを調製し、25℃の状態で全光線透過率をヘーズメーター NDH4000(日本電色工業社製)により測定し、下記判定基準に基づき、評価した。
A:全光線透過率95%以上であった。
B:全光線透過率90%以上95%未満であった。
C:全光線透過率85%以上90%未満であった。
D:全光線透過率80%以上85%未満であった。
E:全光線透過率80%未満であった。
表1は、各実施例及び比較例の組成物の配合条件、並びに、評価結果を示している。なお、表1中の空欄は、その原料を使用していないことを意味する。
表1の結果から、カンナビノイドと、特定のショ糖脂肪酸エステルと、水溶性高分子化合物とを少なくとも含む組成物(粉末)は、凝集物が少なく、また、水系媒体への再分散性にも優れ、酸性条件下にあっても再分散性に優れるものであることもわかった。
Claims (8)
- カンナビノイドと、ショ糖脂肪酸エステルと、水溶性高分子化合物とを含み、前記ショ糖脂肪酸エステルは、平均エステル化度が1.00~1.15である、組成物。
- 前記水溶性高分子化合物は、セルロース誘導体及びポリビニルピロリドンからなる群より選ばれる少なくとも1種である、請求項1に記載の組成物。
- 前記ショ糖脂肪酸エステルと前記水溶性高分子化合物との質量比(ショ糖脂肪酸エステル/水溶性高分子化合物)が0.5~30である、請求項1又は2に記載の組成物。
- さらに油脂を含有する、請求項1又は2に記載の組成物。
- さらに溶媒を含む、請求項1又は2に記載の組成物。
- 前記溶媒が水性溶媒である、請求項5に記載の組成物。
- 粉末状である、請求項1又は2に記載の組成物。
- 食品用、化粧用、飼料用、又は、医薬用である、請求項1又は2に記載の組成物。
