본 발명은 제한된 현미경 배율에서 더 높은 배율로 세포를 관찰하기 위한 세포 시료의 준비와 관련된 기법으로 세포를 하이드로겔화하고 하이드로겔화된 세포를 최소 5~5.5배 최대 22~24배까지 확대 가능한 하이드로겔 세포 필름 제조 방법에 관한 기술이다.

현재의 세포 이미징 분석하기 위한 방법은 현미경의 렌즈 배율(4~100 배율)에 의존하고 있으며, 그 이상의 배율로 관찰하기 위해서는 수 억원 상당의 고가 이미징 장비에 의존하거나, 이 또한 배율의 한계점을 지니고 있다. 따라서, 배율의 한계점을 지닌 현미경에서 더 큰 이미지 분석을 위해서는, 시료의 크기를 더 크게 만들면 현미경 분석에서의 배율의 한계를 극복할 수 있다.
MAP (magnified analysis of proteome) 기법은 조직 및 세포를 최대 4배 확대하는 기술로, 8 mm 둥근 커버슬립 위에 세포 배양 후, MAP 용액을 도포하여 하이드로겔을 제작하여 이루어진다. 다만, dH2O에서 4배 커진 결과물은 볼록렌즈 형태의 하이드로겔로 이미징 분석을 위한 하이드로겔을 고정하기 어려우며, 또한 하이드로겔 두께가 3mm 이상으로 부피가 상당히 컸다.
이후, 조직 및 세포를 최대 8배 확대 가능한 ZOOM 기법이 개발되었으며, 2가지의 ZOOM 용액을 사용하여 하이드로겔 배양을 총 4번의 round cycle을 걸쳐 최대 8배 커지게 하였다.
최근, 얇은 조직을 10배 확대 가능한 Epitope-preserving magnified analysis of proteome (eMAP) 기법이 보고되었으며, 총 3번의 round cycle을 걸쳐 약 10배 커지게 하였다. eMAP은 오직 조직을 대상으로 평가하였으며 세포를 대상으로 적용하지 않았다.
따라서, 세포의 크기를 흡수성이 있는 하이드로겔로 만들어 세포를 더 커지게 하고, 세포가 포함된 시료가 커지더라도 두께를 얇게하여 현미경에서 이미지 분석이 용이한 기술의 개발이 요구되었다.
따라서, 본 발명의 목적은 증류수에 용해된 아크릴아마이드(acrylamide), 비스-아크릴아마이드(bis-acrylamide), 소듐아크릴레이트(sodium acrylate) 및 TEMED를 포함하는 세포 필름 제조용 조성물을 제공하는 것이다.
본 발명의 또다른 목적은 세포에 제1용액과 제2용액을 도포하여 응고반응을 진행하는 단계를 포함하는 세포 필름 제조 방법으로서, 상기 제1용액은 증류수에 용해된 아크릴아마이드, 비스-아크릴아마이드, 소듐아크릴레이트 및 TEMED를 포함하는 조성물이고, 상기 제2용액은 증류수에 용해된 5 내지 20% APS인, 세포 필름 제조 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 항의 상기 세포 필름 제조 방법으로 제조된 세포 필름을 제공하는 것이다.
본 발명은 또한, (a) 상기 세포 필름을 제1용액에서 배양하는 단계; (b) 상기 (a) 단계에서 배양된 세포 필름에 제3용액을 도포하여 응고반응을 진행하는 단계;를 포함하는 세포 확대 방법으로서, 상기 제1용액은 증류수에 용해된 아크릴아마이드, 비스-아크릴아마이드, 소듐아크릴레이트 및 TEMED를 포함하는 조성물이고, 상기 제3용액은 증류수에 용해된 0.5 내지 2% APS인, 세포 확대 방법을 제공한다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
본 명세서에서 사용한 용어는 단지 설명을 목적으로 사용된 것으로, 한정하려는 의도로 해석되어서는 안 된다. 단수의 표현은 문맥상 명백하게 다르게 뜻하지 않는 한, 복수의 표현을 포함한다. 본 명세서에서, "포함하다" 또는 "가지다" 등의 용어는 명세서 상에 기재된 특징, 숫자, 단계, 동작, 구성요소, 부품 또는 이들을 조합한 것이 존재함을 지정하려는 것이지, 하나 또는 그 이상의 다른 특징들이나 숫자, 단계, 동작, 구성요소, 부품 또는 이들을 조합한 것들의 존재 또는 부가 가능성을 미리 배제하지 않는 것으로 이해되어야 한다. 다르게 정의되지 않는 한, 기술적이거나 과학적인 용어를 포함해서 여기서 사용되는 모든 용어들은 실시예가 속 하는 기술 분야에서 통상의 지식을 가진 자에 의해 일반적으로 이해되는 것과 동일한 의미를 가지고 있다. 일반적으로 사용되는 사전에 정의되어 있는 것과 같은 용어들은 관련 기술의 문맥 상 가지는 의미와 일치하는 의미를 가지는 것으로 해석되어야 하며, 본 출원에서 명백하게 정의하지 않는 한, 이상적이거나 과도하게 형식적인 의미로 해석되지 않는다.
본 발명은 하이드로겔 조성을 최적화하여 기본 베이스 용액을 증류수 (dH2O)로 바꿔주어 세포 이미징에 활용할 수 있는 세포 필름 제조에 관한 것이다. 이러한 dH2O 기반 조성으로 0.17mm 두께의 커버슬립들을 이용하여 0.17mm 두께의 5배 확대 가능한 얇은 하이드로겔 세포 필름을 제작하였다(도 2). 또한, dH2O 기반 조성의 하이드로겔화 용액(이하 "필름"액)과 1% APS (응고액-2)를 이용한 배양 및 응고 과정을 총 5회에 걸쳐서 순차적으로 최대 약 22배 이상 커질 수 있게 하였음을 확인하였으며, 필름용액에서의 배양 횟수에 따른 세포의 크기들을 비교 분석하여 제시하였다(1회: 6.2~6.8배, 2회: 8.2~9배, 3회: 10.6~11.8배, 4회: 16.7~18.5배, 5회: 22~24배)([표 3] 및 도 3 내지 6 참조). 파라필름으로 밀봉하여 가위로 절단하기 쉽고, 간편하게 비닐 지퍼백에 넣어 상온에서 보관하기 용이하다. 따라서, 실험자가 세포 관찰 실험을 위하여 하이드로겔 세포 필름을 절단하고 원하는 세포 확대 비율에 따라 세포를 확장하여 현미경을 활용한 고해상도 이미지 분석을 진행하기에 용이하다. 또한, 이를 바탕으로 다양한 세포주들과 줄기세포 플라그, 인체 유래물, 박테리아 등을 대상으로 하이드겔 세포 필름들을 선택적으로 제작할 수 있으며, 초고해상도 이미징 분석용 하이드로겔 세포 필름으로 이용가능하다.
따라서, 본 발명은 증류수에 용해된 아크릴아마이드(acrylamide), 비스-아크릴아마이드(bis-acrylamide), 소듐아크릴레이트(sodium acrylate) 및 TEMED를 포함하는 세포 필름 제조용 조성물을 제공한다.
본 명세서에서 용어 "세포 필름"은 세포를 4배 내지 22배 확대/확장시켜 현미경으로 세포의 고해상도 이미징 분석을 수행하기 위한 하이드로겔화 된 고정된 세포 시료를 의미한다. 하이드로겔 "세포 필름"은 액체 흡수성 특성을 지니며 세포 필름 제조용 조성물에 배양 및 응고 반응을 반복 시행을 통해 세포 형태 및 세포내 소기관들을 확장시키는 특성을 지닌다. 기존 현미경의 렌즈 배율에만 의존하던 부분을 관찰 시료의 확대로 배율을 증가시켜 극복할 수 있으며, 세포의 구조를 고해상도의 이미지로 관찰할 수 있다. 또한, 세포, 박테리아, 곰팡이, 효모 등 다양한 세포 및 미생물을 대상으로 얇은 세포 필름으로 제작이 가능하며, 보관이 편리하고 사용이 매우 간편한 장점을 지닌다. 본 명세서에 있어서 "세포 필름"은 "하이드로겔 세포 필름"과 동일한 의미로 사용된다.
상기 조성물의 아크릴아마이드와 비스-아크릴아마이드는 크로스 링킹 구조를 이루어 하이드로겔화 하는 성분이고, 소듐아크릴레이트는 수분을 흡수하여 확장하는 특성을 지닌다.
본 발명의 조성물은 PBS가 아닌 증류수에 각각의 성분을 용해시켜 이미징 분석에 이용하기 위한 조성물이며, 구체적인 일실시예에서는 각각의 성분을 PBS와 증류수에 용해시켰을 때의 배율을 비교하였고, 이 때 증류수에 용해시킨 하이드로겔 조성물을 이용한 경우 확대 배율이 확실히 더 큰 것을 확인하였다(도 1 및 표 2). 0.1M PBS 용액 기반으로 0-round 경우 PBS에서 4배 확대 가능하지만, 본 발명을 이용한 dH2O 기반 조성물을 사용한 경우 0-round 경우 5~5.5배 확대가 가능한 것을 확인하였으며, 배양 및 응고 반응을 순차적으로 5회 round cycle까지 진행해주었을 경우 최대 22배 세포 확대가 가능함을 확인하였다.
본 발명의 세포 필름 제조용 조성물에 허용되는 부형제는 특정한 처리 형태 목적에 적합한 임의의 모든 용매, 분산 매질, 희석제, 또는 다른 액체 비히클, 분산액 또는 현탁 보조제, 표면 활성제, 등장화제, 증점제 또는 유화제, 보존제, 고체 결합제, 윤활제 등을 포함한다.
세포 필름 제조용 조성물에 포함되는 예시적인 보존제는 항산화제, 킬레이트화제, 항미생물 보존제, 항진균 보존제, 알콜 보존제, 산성 보존제, 및 다른 보존제를 포함할 수도 있다. 예시적인 항산화제는 알파 토코페롤, 아스코르브산, 아스코르빌 팔미테이트, 부틸화 히드록시아니솔, 부틸화 히드록시톨루엔, 모노티오글리세롤, 메타중아황산칼륨, 프로피온산, 프로필 갈레이트, 아스코르브산나트륨, 중아황산나트륨, 메타중아황산나트륨, 및 아황산나트륨을 포함하지만 이에 제한되지 않는다. 예시적인 킬레이트화제는 에틸렌디아민테트라아세트산 (EDTA), 시트르산 1수화물, 이나트륨 에데테이트, 이칼륨 에데테이트, 에데트산, 푸마르산, 말산, 인산, 소듐 에데테이트, 타르타르산, 및 트리소듐 에데테이트를 포함한다. 예시적인 항미생물 보존제는 벤즈알코늄 클로라이드, 벤제토늄 클로라이드, 벤질 알콜, 브로노폴, 세트리미드, 세틸피리디늄 클로라이드, 클로르헥시딘, 클로로부탄올, 클로로크레졸, 클로로크실레놀, 크레졸, 에틸 알콜, 글리세린, 헥세티딘, 이미드우레아, 페놀, 페녹시에탄올, 페닐에틸 알콜, 페닐질산수은 (phenylmercuric nitrate), 프로필렌 글리콜, 및 티메로살을 포함하지만 이에 제한되지 않는다. 예시적인 항진균 보존제는 부틸 파라벤, 메틸 파라벤, 에틸 파라벤, 프로필 파라벤, 벤조산, 히드록시벤조산, 칼륨 벤조에이트, 소르브산칼륨, 벤조산나트륨, 프로피온산나트륨, 및 소르브산을 포함하지만 이에 제한되지 않는다. 예시적인 알콜 보존제는 에탄올, 폴리에틸렌 글리콜, 페놀, 페놀계 화합물, 비스페놀, 클로로부탄올, 히드록시벤조에이트, 및 페닐에틸 알콜을 포함하지만 이에 제한되지 않는다. 예시적인 산성 보존제는 비타민 A, 비타민 C, 비타민 E, 베타-카로틴, 시트르산, 아세트산, 데히드로아세트산, 아스코르브산, 소르브산, 및 피트산을 포함하지만 이에 제한되지 않는다. 다른 보존제는 토코페롤, 토코페롤 아세테이트, 데테록시메 메실레이트, 세트리미드, 부틸화 히드록시아니솔 (BHA), 부틸화 히드록시톨루엔드 (BHT), 에틸렌디아민, 소듐 라우릴 술페이트 (SLS), 소듐 라우릴 에테르 술페이트 (SLES), 중아황산나트륨, 메타중아황산나트륨, 칼륨 술파이트, 메타중아황산칼륨, 글리단트 플러스, 페노닙, 메틸파라벤, 저몰 115, 게르마벤 II, 네올론, 카톤, 및 에욱실을 포함하지만 이에 제한되지 않는다.
예시적인 완충제는 시트레이트 완충 용액, 아세테이트 완충 용액, 포스페이트 완충제 용액, 염화암모늄, 탄산칼슘, 염화칼슘, 칼슘 시트레이트, 칼슘 글루비오네이트, 칼슘 글루셉테이트, 칼슘 글루코네이트, D-글루콘산, 글리세로인산칼슘, 락트산칼슘, 프로판산, 칼슘 레불리네이트, 펜탄산, 이염기성 인산칼슘, 인산, 삼염기성 인산칼슘, 수산화칼슘 포스페이트, 아세트산칼륨, 염화칼륨, 글루콘산칼륨, 칼륨 혼합물, 이염기성 인산칼륨, 일염기성 인산칼륨, 인산칼륨 혼합물, 아세트산나트륨, 중탄산나트륨, 염화나트륨, 시트르산나트륨, 락트산나트륨, 이염기성 인산나트륨, 일염기성 인산나트륨, 인산나트륨 혼합물, 트로메타민, 수산화마그네슘, 수산화알루미늄, 알긴산, 피로겐-자유수, 등장성 염수, 링거(Ringer's) 용액, 에틸 알콜, 및 이들의 조합을 포함하지만 이에 제한되지 않는다.
예시적인 윤활제는 스테아르산마그네슘, 스테아르산칼슘, 스테아르산, 실리카, 활석, 맥아, 글리세릴 베하네이트, 수소화 식물성 오일, 폴리에틸렌 글리콜, 벤조산나트륨, 아세트산나트륨, 염화나트륨, 류신, 마그네슘 라우릴 술페이트, 소듐 라우릴 술페이트, 및 이들의 조합을 포함하지만 이에 제한되지 않는다.
일양태에 있어서, 상기 아크릴아마이드의 농도는 20 내지 40 % (w/v)이고, 비스아크릴아마이드는 0.01 내지 0.1 % (v/v), 소듐아크릴레이트의 농도는 5 내지 20% (w/v), TEMED의 농도는 0.5 내지 0.8 % (v/v)일 수 있다. 더욱 바람직하게는, 아크릴아마이드의 농도는 30%, 비스아크릴아마이드의 농도는 0.02%, 소듐아크릴레이트의 농도는 10%일 수 있으며, TEMED의 농도는 0.65 %일 수 있다. 상기 범위를 벗어나는 경우, 세포 확대가 원하는 만큼 이루어지지 않을 수 있다(도 1 참조).
본 발명의 세포 필름 제조에 이용될 수 있는 상기 세포는 동물, 원생동물, 갑각류, 파충류, 식물, 곤충 유래 세포, 박테리아, 효모 및 곰팡이로 이루어진 군에서 선택되는 어느 1종으로부터 유래된 세포일 수 있다. 상기 동물은 마우스, 랫트, 주머니쥐, 메추라기, 햄스터, 토끼, 개, 돼지, 기니피그, 양, 소, 고양이, 밍크, 오리, 닭 등일 수 있으며, 이에 제한되지 않는다.
상기 세포는 피부, 혈액, 뼈, 골수, 배아, 대식세포, 폐, 혈관, 뇌, 심장, 췌장, 정소, 난소, 근육, 연골, 간, 비장, 자궁경관,유방, 신경모세포종, 대장, 뇌하수체, 각막, 망막, 이자, 멜라노마, 감상선, 방광, 점액, 전립선, 성상세포, 신경교세포, 복막, 지방세포 및 흉선으로 이루어진 군에서 선택되는 어느 1종으로부터 유래된 세포일 수 있으나, 이에 제한되지 않는다.
또한, 상기 세포는 부착성(adherent) 세포, 부유성(suspension) 세포, 지방 줄기세포, 유도만능 줄기세포, 줄기세포로부터 유도 분화된 세포, 293T, HeLa, 섬유아세포(fibroblast), 바이러스 및 박테리아에 감염된 세포일 수 있으나, 이에 제한되지 않는다.
여기서, 상기 부착성 세포는 1차적으로 세포배양 된 피부 섬유아세포(skin fibroblast), 배아 섬유아세포(embryonic fibroblast), HEK293T 세포(human embryonic kidney cell), A549 폐암세포(lung carcinoma cell), Hela 자궁경부암세포(cervix adenocarcinoma cell), Raw 264.7 대식세포, U87 교모세포종세포(glioblastoma cell) 등을 포함한 하기 표 1의 세포일 수 있으나, 이에 제한되지 않는다.
또한, 상기 부유성 세포는 HL-60 전골수성 백혈병 세포(peripheral blood acute promyelocytic leukemia cell), U-937 단핵구 백혈별 세포(pleural effusion histiocytic lymphoma cell) 등을 포함한 하기 표 1의 세포일 수 있으나, 이에 제한되지 않는다.
일실시예에 있어서, 상기 세포는 하기 표 1에서 선택되는 어느 1종 이상일 수 있으나, 이에 제한되지 않는다.

구체적인 일 양태에서, 상기 조성물에 아자이드화나트륨(sodium azide)을 추가로 포함할 수 있다. 하이드로겔 세포 필름은 보관 기간에 따라 단기 보관용 및 장기 보관용으로 나뉘며 장기보관용은 방부제 특성의 0.01% 아자이드화나트륨이 추가로 포함될 수 있다.
본 발명은 또한, 세포에 제1용액과 제2용액을 도포하여 응고반응을 진행하는 단계를 포함하는 세포 필름 제조 방법으로서, 상기 제1용액은 증류수에 용해된 아크릴아마이드, 비스-아크릴아마이드, 소듐아크릴레이트 및 TEMED를 포함하는 조성물이고, 상기 제2용액은 증류수에 용해된 5 내지 20% APS인, 세포 필름 제조 방법을 제공한다.
본 명세서에 있어서 상기 "제1용액"은 본 명세서에서 "필름액"과 동일한 의미로 사용되며, 상기 "제2용액"은 실시예상 "응고액-1"과 같은 의미로 사용될 수 있고, 제1용액인 "필름액"과 혼합시 화학 반응으로 폴리-하이드로겔화 반응을 진행시킬 수 있다.
구체적인 세포 필름 제조 방법은 도 2a 및 도 2b에 나타난 순서도와 같으며, 본발명의 세포 필름 제조 방법에 있어서, 상기 아크릴아마이드의 농도는 20 내지 40 % (w/v)이고, 비스아크릴아마이드는 0.01 내지 0.1 % (v/v), 소듐아크릴레이트의 농도는 5 내지 20% (w/v), TEMED의 농도는 0.5 내지 0.8 % (v/v)일 수 있다. 더욱 바람직하게는, 아크릴아마이드의 농도는 30%, 비스아크릴아마이드의 농도는 0.02%, 소듐아크릴레이트의 농도는 10%일 수 있으며, TEMED의 농도는 0.65 %일 수 있다. 상기 범위를 벗어나는 경우, 세포 확대가 원하는 만큼 이루어지지 않을 수 있다(도 1 참조).
상기 세포 필름 제조 방법에 이용될 수 있는 세포는 동물, 원생동물, 갑각류, 파충류, 식물, 곤충 유래 세포, 박테리아, 효모 및 곰팡이로 이루어진 군에서 선택되는 어느 1종으로부터 유래된 세포일 수 있으며, 본 명세서의 표 1에서 선택되는 어느 1종 이상으로부터 유래한 세포일 수 있다. 이외에도 가능한 세포에 대해서는 상기 세포 필름 제조용 조성물에서 전술한 바와 같으므로, 기재를 생략한다.
세포 필름 제조 방법에 이용되는 제1용액에 아자이드화나트륨을 추가로 포함할 수 있다. 이는 상기 세포 필름 제조용 조성물에서 전술한 바와 같으므로, 기재를 생략한다. 단기 및 장기 보관용 필름액으로 제작된 세포 필름을 비닐 지퍼백으로 밀봉하여 상온에서 간편하게 보관이 용이하며, 형광 항체 및 dye가 염색된 세포의 하이드로겔 세포 필름은 차광 비닐 지퍼백을 이용하여 보관이 가능하다.
상기 세포 필름 제조 방법에서 응고반응은 5분 내지 60분간 수행될 수 있으며, 바람직하게는 10분 내지 40분간 수행될 수 있고, 더욱 바람직하게는 15분 내지 30분간 수행될 수 있다.
상기 응고된 세포 필름을 파라필름으로 밀봉하는 단계를 추가로 포함할 수 있다. 구체적인 일실시예에서 커버슬립 내에서 하이드로겔 세포 필름의 정형화가 가능하였으며, 하이드로겔이 흡착되지 않는 파라필름을 부착하였고, 가위를 이용하여 사용자가 원하는 크기로 건조된 하이드로겔 세포 필름을 절단하여 사용 가능하도록 제조가능하다(도 2c). 이와 같은 방법으로 제조된 세포 필름은 세포 확대 이미징 분석이 필요한 사용자에게 번거러운 세포 배양 과정이 없이 일회성으로 사용이 가능하며 또한 세포 필름의 보관 또한 용이하다.
본 발명은 상기 세포 필름 제조 방법으로 제조된 세포 필름을 제공한다.
본 발명은 또한, (a) 전술한 세포 필름을 제1용액에서 배양하는 단계; 및
(b) 상기 (a) 단계에서 배양된 세포 필름에 제3용액을 도포하여 응고반응을 진행하는 단계; 를 포함하는 세포 확대 방법으로서, 상기 제1용액은 증류수에 용해된 아크릴아마이드, 비스-아크릴아마이드, 소듐아크릴레이트 및 TEMED를 포함하는 조성물이고, 상기 제3용액은 증류수에 용해된 0.5 내지 2% APS인, 세포 확대 방법을 제공한다.
세포 확대 방법은 전술된 방법으로 제조된 세포 필름을 추가적으로 필름액 또는 응고액에서 하이드로겔화 및 재응고화를 진행하여 세포를 확대시키를 방법을 의미하며, 구체적인 과정은 도 4에 나타난 바와 같다.
본 명세서에 있어서 상기 "제3용액"은 실시예상 "응고액-2"와 같은 의미로 사용될 수 있고, 제1용액인 "필름액"과 혼합시 화학 반응으로 폴리-하이드로겔화 반응을 진행시킬 수 있다.
세포 필름을 제1용액에 배양 및 제3용액으로 하이드로겔화 및 재응고 과정(1~5 회)을 반복하여 확대되는 비율을 확인한 결과는 [표 3]과 같고, 이를 이미징화 한 결과는 도 5a 내지 도 5e와 같다.
상기 세포 확대 방법에서 (a) 단계의 제1용액의 아크릴아마이드의 농도는 30 내지 40 %이고, 비스아크릴아마이드는 0.01 내지 0.1 %, 소듐아크릴레이트의 농도는 5 내지 20%, TEMED의 농도는 0.5 내지 0.8 %일 수 있다. 더욱 바람직하게는, 아크릴아마이드의 농도는 30%, 비스아크릴아마이드의 농도는 0.02%, 소듐아크릴레이트의 농도는 10%일 수 있으며, TEMED의 농도는 0.65 %일 수 있다.
구체적인 일 양태에서, 상기 (a) 단계의 배양은 2℃ 내지 10℃에서 4시간 내지 12시간 동안 진행될 수 있으며, 바람직하게는 5시간 내지 8시간동안 진행될 수 있고, 통상의 기술자가 적절히 조절할 수 있다.
구체적인 일 양태에서, 상기 (b) 단계의 응고반응은 5분 내지 60분간 수행될 수 있으며, 바람직하게는 5분 내지 30분간 수행될 수 있다.
일실시예에 있어서, 상기 (a) 단계 내지 (b) 단계를 1 내지 3회 반복할 수 있다. 상기 (a) 단계 내지 (b) 단계를 1 내지 3회 반복하여 제조된 세포 필름을 제1용액에서 2℃ 내지 10℃에서 6시간 내지 24시간 동안 배양하는 단계를 추가로 포함할 수 있다.
전술한 세포 확대 방법에서 (b) 단계 이후, (c) 상기 (b) 단계에서 응고된 세포 필름을 PBS에서 배양하는 단계; 및 (d) 상기 (c) 단계에서 배양된 세포 필름을 증류수에서 배양하는 단계;를 추가로 포함할 수 있다.
상기 하이드로겔 세포 필름을 PBS에서 배양하는 단계에서 PBS의 농도는 0.05 내지 0.2M 일 수 있으며, 바람직하게는 0.1M 일 수 있다.
일 양태에 있어서, 상기 (c) 단계 또는 (d) 단계에서 형광 이미징 분석을 위한 항체 또는 염색 물질을 처리하는 단계를 추가로 포함할 수 있다. 세포 이미징 분석 기술 분야에 있어서. 이용될 수 있는 항체 또는 염색 물질이라면 제한없이 사용가능하다. 항체 염색을 진행하여 초고해상도 이미징 분석이 가능하여여 최대 22~24배 확대가 가능하다. 본 발명에 의한 확대 방법을 이용하는 경우, 얇은 두께의 하이드로겔 세포 필름으로 최대 확장되어도 이미징 분석 가능한 두께이다. 0.17mm (170um)의 두께의 얇은 하이드로겔 세포 필름을 5round의 반복적인 하이드로겔화를 진행시 최대 20배 확대가 되며, 확대된 하이드로겔 두께는 약 2.5mm로서 현미경의 작동거리 (보통 2mm)내에 하이드로겔 내의 확대된 세포 이미징 분석이 가능하다.
본 발명의 하이드로겔 세포 필름을 이용하는 경우, 최대 약 22~24배 확대가 가능하며, 세포가 포함된 시료가 커지더라도 두께가 얇아 초고해상도 이미징 분석에 이용될 수 있다. 또한, 단기 및 장기 보관이 가능하며, 추가적인 세포 배양 없이도 언제든지 사용이 가능하다.
도 1은 PBS 기반의 용액과 본 발명의 dH2O 기반 용액의 하이드로겔 조성을 달리하여 세포 확대를 비교한 결과이다.
도 2a 및 도 2b는 하이드로겔 세포 필름 제작 과정, 도 2c는 하이드로겔 세포 필름의 보관 방법을 나타낸다.
도 3a 내지 3c는 본 발명에 따라 제작된 하이드로겔 세포 필름을 이용한 세포 확대 결과를 나타낸다.
도 4는 하이드로겔화 및 재응고 과정을 반복하여 확대시키는 과정을 나타낸다.
도 5a 내지 도 5e는 하이드로겔화 및 재응고과정을 반복하여 확대되는 비율을 확인하여 이미징화 한 결과이다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[준비예]
필요 시약 및 용품
50 mL conical tube, 유리 커버슬립 (24 x 24-mm), 유리 커버슬립 (24 x 60-mm), PALAFILM, Gel Releasers, 아크릴아마이드(acrylamide), 2% 비스-아크릴아마이드 용액(bis-acrylamide solution), 소듐아크릴레이트(sodium acrylate), 소듐 아자이드(sodium azide), 2~4% 파라포름알데히드(paraformaldehyde, (PFA)), 암모늄 펄설페이트(ammonium persulfate (APS)), 테트라메틸에틸렌다이아민(tetramethylethylenediamine, (TEMED)), 10X PBS (phosphate-Buffered Saline).
사용된 하이드로겔 필름액 및 응고액 제조법은 다음과 같다.
(1) 필름액 : 증류수에 용해된 30% 아크릴아마이드(acrylamide), 0.02% 비스-아크릴아마이드(bis-acrylamide), 10% 소듐아크릴레이트, 0.01% 소듐아자이드, 0.65% TEMED
(2) 응고액-1 (초기 필름 제작용) : 증류수에 용해된 10% APS
(3) 응고액-2 (Round 응고용) : 증류수에 용해된 1% APS
조성의 아크릴아마이드(acrylamide)와 비스-아크릴아마이드(bis-acrylamide)는 cross-linking 구조를 이루어 하이드로겔화 하는 성분이고 소듐아크릴레이트(sodium acrylate)는 수분을 흡수하여 확장하는 특성을 지닌다. 하이드로겔 세포 필름은 보관 기간에 따라 단기 보관용 및 장기 보관용으로 나뉘며 장기보관용은 방부제 특성의 0.01% sodium azide (아자이드화나트륨)가 추가 첨가된다.
<실시예 1>
세포 필름 제조용 조성물의 최적화
다양한 필름 용액의 조성물에 의한 하이드로겔의 확장된 크기를 비교하였다.
하기 표 2에서는 아크릴아마이드의 농도, 비스-아크릴아마이드의 농도를 각각 다르게 하고, PBS 및 dH2O에 용해시켰을 때의 확대 배율을 나타냈다.

(AA : Acrylamide, BA : Bis-acrylamide, SA : Sodium Acrylate) 도 1 및 표 2에 나타난 바와 같이, PBS 기반의 하이드로겔보다 본원발명의 dH2O 기반 하이드로겔이 더 큰 겔 확대를 이루는 것을 확인하였다. 또한, 30% 아크릴아마이드 및 0.02% 비스-아크릴아마이드가 첨가된 dH2O기반의 필름 용액은 0 round에서 5배 이상의 크기로 확장이 가능한 특징을 확인하였다. 따라서, 본 발명의 하이드로겔 세포 필름을 제작 및 최대 24배 확장을 위한 배양 용액은 (30% 아크릴아마이드, 0.02% 비스-아크릴아마이드, 10% 소듐 아크릴레이트 in dH2O) 조성으로 하여, 추후 실험을 진행하였다.
<실시예 2>
하이드로겔 세포 필름의 제작 과정
하이드로겔 세포 필름의 제작 과정은 다음과 같다(도 2a 및 도 2b)
1) 세포의 준비
6 웰 플레이트에 24 x 24 mm 커버슬립을 넣고 각 웰당 세포가 포함된 세포 배양 배지 (DMEM or DMEM/F12 + 10% FBS + 50 μg/ml penicillin/streptomycin)을 2~3 mL 넣어준 뒤 24시간 CO2 인큐베이터에서 배양한다. 배지를 제거하고 0.1M PBS로 세척한다. 각 웰에 2~4% 파라포름알데히드(paraformaldehyde, PFA)를 1 mL씩 넣어 20-30분간 고정한다. PFA를 제거해준 후, 0.1M PBS로 3회에 걸쳐 세척해준다.
2) 하이드로겔 세포 필름 제작을 위한 유리틀 준비
하이드로겔 필름은 두께 0.17 mm (170μm)의 유리 커버슬립 8개 (24 x 60mm)를 이용하여 틀에서 만들어지며, 하단부 받침 (3개), 기둥 (각 2개씩 2세트), 덮개 (1개)로 이루어져 있고, 중심부에 배양된 세포가 부착된 커버슬립 (22 x 22mm 혹은 24 x 24 mm)을 배치한다.
3) 배양된 세포가 키워진 커버슬립을 유리틀 내에 배치
하나의 하이드로겔 세포 필름을 제작시 필름액은 APS와 혼합시 화학반응으로 폴리-하이드로겔화 진행이 되며, 각 세포 필름을 제작할 시에는 유리틀 안으로 세포가 포함된 커버슬립을 배치한다.
4) 필름액과 응고액의 혼합액 도포 후 하이드로겔화 진행
필름액 285 μL 및 응고액-1 (10% APS) 15 μL을 혼합 후 빠르게 세포를 도포하여 필름액이 하이드로겔화 응고되기 전에 덮개용 커버슬립으로 덮어준다. 20분간 하이드로겔화를 진행한다.
5) 유리틀 제거 후 세포 필름 분리
응고 반응 10~20분 후 틀에 사용된 커버슬립들을 조심스럽게 제거하여 하이드로겔 세포 필름을 파라핀 성분으로 이루어진 파라필름 (4 x 10mm) 위에 올린다.
6) 파라필름에 배치
이후, 다른 파라필름 (4 x 10mm)으로 덮어 2개의 파라필름 사이에 배치토록하여 추가 건조시킨다. 가위를 이용하여 하이드로겔 잔여물을 제거한다.
7) 보관
다듬어진 하이드로겔 세포 필름을 새로운 파라필름으로 옮기고, 파라필름 표면에 세포의 윗면 및 아랫면 위치를 라벨링해준 뒤, 지퍼백에 밀봉하여 실온에서 보관한다. 파라필름과 샌드위치된 하이드로겔 세포 필름은 가위를 이용하여 사용자의 원하는 사양에 따른 크기로 절단하여 사용이 가능하다.
하이드로겔 세포 필름은 하이드로겔 특성상 접착력이 강하여 안전하게 도포하여 보관하기가 매우 어렵다. 파라필름은 하이드로겔 세포 필름을 도포시 밀봉 및 제거가 용이하며, 하이드로겔 세포 필름에 데미지를 주지 않는다. 또한, 파라필름은 하이드로겔 세포 필름을 밀봉 도포한 상태에서 가위를 이용하여 절단하기가 쉽다.
하이드로겔 세포 필름의 보관
[도 2c]에 나타난 바와 같이, 하이드로겔 세포 필름의 보관은 파라필름에 보존된 하이드로겔 세포 필름을 2가지의 지퍼백에 밀봉된 상태에서 별도의 보관함에 보관하며, 별도의 방부제와 함께 실온에서 장기간 보관이 용이하다.
- 일반적인 세포 필름 : 투명비닐 지퍼백 (예: 5 x 7 cm) 보관 및 스티커 라벨링
- 염색된 세포 필름 : 차광비닐 지퍼백 (예: 5 x 7 cm) 보관 및 스티커 라벨링
<실시예 3>
하이드로겔 세포 필름을 이용한 세포 확대
제작된 하이드로겔 세포 필름은 0.1M PBS에 옮겨 배양시 약 2.7~3배 커지고, 순차적으로 dH2O에 옮겨 배양하면 최종적으로 5~5.5배 커지며, 본 단계를 0 round라고 지칭한다. 이 때에, 0.1M PBS에서 배양 후 dH2O에 옮겨 배양하면 겔이 급격하게 커질 수 잇으므로, 겔을 안정적으로 커지게 하기 위해, PBS의 농도를 낮추어 0.01M PBS에서 배양하는 단계를 추가로 포함할 수 있다.
각각의 라운드 사이클을 반복할수록 확대 배율은 올라가고, 두께에는 큰 변화가 없음을 도 3a에서 확인할 수 있다.
5~5.5배 커진 하이드로겔을 0.1M PBS에 재배양시 하이드로겔은 크기가 작아져 이전 단계 약 2.7~3배의 크기로 되돌아간다. 이러한 하이드로겔의 팽창과 수축은 반복적으로 진행이 가능하며, 0.1M PBS에 배양 단계에서 형광 이미징 분석을 위한 항체 및 dye를 염색이 가능하다.
본 발명자는 사전에 배양된 세포를 DAPI dye를 이용하여 세포를 사전 염색하였고, 이를 이용하여 하이드로겔 세포 필름을 제작하였다. 하이드로겔 세포 필름을 가위로 이용화여 가로 세포 2.7mm x 2.7mm 혹은 3mm x 3mm 크기로 절단하여, 이후 세포 확대화를 진행하였다(도 3b).
도 3c에 나타난 바와 같이, 약 3mm의 하이드로겔 세포 필름 및 세포는 순차적으로 0.1M PBS에서 2.7~3배 커지며, 최종적으로 dH2O에서 5~5.5배 커지는 것을 확인하였다.
구체적으로, 하이드로겔화 및 재응고 과정(1~5 회)을 반복하여 확대시키는 과정은 도 4와 같다.
또한, 세포 필름을 필름액에 배양 및 응고액-2로 하이드로겔화 및 재응고 과정(1~5 회)을 반복하여 확대되는 비율을 확인한 결과는 하기 [표 3]와 같고, 이를 이미징화 한 결과는 도 5a 내지 도 5e와 같다.

[표 3]에 나타난 바와 같이, 최종적으로 dH2O에서 세포를 6.2~24배까지 확대시켜 줄 수 있다. 추가적으로 6 round 이상의 과정을 반복하여 세포를 더 크게 확장할 수 있으나, 현재의 보편적인 이미지 분석용 현미경 시스템에서는 작동거리 (Working distance)의 한계로 불가능하다. 하지만, 향후 작동거리의 한계가 극복되고 미세 형광 신호를 감지하는 현미경의 경우 시각적인 이미징 분석이 가능할 수 있다. 따라서, 이미징 분석 시스템의 한계로 인해 본 발명의 일반적인 현미경을 통한 관찰시 최대 적용 세포 크기는 4-5 round의 16.7배~22배 (18.5~24배)까지 가능하다. 이상, 본 발명의 내용의 특정한 부분을 상세히 기술하였지만, 이러한 구체적인 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
Claims (20)
- 증류수에 용해된 아크릴아마이드(acrylamide), 비스-아크릴아마이드(bis-acrylamide), 소듐아크릴레이트(sodium acrylate) 및 TEMED를 포함하는 세포 필름 제조용 조성물.
- 제1항에 있어서,상기 아크릴아마이드의 농도는 20 내지 40 % (w/v)이고, 비스아크릴아마이드는 0.01 내지 0.1 % (v/v), 소듐아크릴레이트의 농도는 5 내지 20% (w/v), TEMED의 농도는 0.5 내지 0.8 % (v/v)인, 세포 필름 제조용 조성물.
- 제1항에 있어서,상기 세포는 명세서 내 표 1에서 선택되는 어느 1종 이상인, 세포 필름 제조용 조성물.
- 제1항에 있어서,상기 조성물에 아자이드화나트륨(sodium azide)을 추가로 포함하는, 세포 필름 제조용 조성물.
- 세포에 제1용액과 제2용액을 도포하여 응고반응을 진행하는 단계를 포함하는 세포 필름 제조 방법으로서,상기 제1용액은 증류수에 용해된 아크릴아마이드, 비스-아크릴아마이드, 소듐아크릴레이트 및 TEMED를 포함하는 조성물이고,상기 제2용액은 증류수에 용해된 5 내지 20% APS인, 세포 필름 제조 방법.
- 제5항에 있어서,상기 아크릴아마이드의 농도는 30 내지 40 %이고, 비스아크릴아마이드는 0.01 내지 0.1 %, 소듐아크릴레이트의 농도는 5 내지 20%, TEMED의 농도는 0.5 내지 0.8 %인, 세포 필름 제조 방법.
- 제5항에 있어서,상기 세포는 명세서 내 표 1에서 선택되는 어느 1종 이상인, 세포 필름 제조 방법.
- 제5항에 있어서,상기 제1용액에 아자이드화나트륨을 추가로 포함하는, 세포 필름 제조 방법.
- 제5항에 있어서,상기 응고반응은 5분 내지 60분간 수행되는, 세포 필름 제조 방법.
- 제5항에 있어서,상기 응고된 세포 필름을 파라필름으로 밀봉하는 단계를 추가로 포함하는, 세포 필름 제조 방법.
- 제5항 내지 제10항 중 어느 한 항의 세포 필름 제조 방법으로 제조된 세포 필름.
- (a) 제11항의 세포 필름을 제1용액에서 배양하는 단계; 및(b) 상기 (a) 단계에서 배양된 세포 필름에 제3용액을 도포하여 응고반응을 진행하는 단계;를 포함하는 세포 확대 방법으로서,상기 제1용액은 증류수에 용해된 아크릴아마이드, 비스-아크릴아마이드, 소듐아크릴레이트 및 TEMED를 포함하는 조성물이고,상기 제3용액은 증류수에 용해된 0.5 내지 2% APS인, 세포 확대 방법.
- 제12항에 있어서,상기 (a) 단계의 제1용액의 아크릴아마이드의 농도는 30 내지 40 %이고, 비스아크릴아마이드는 0.01 내지 0.1 %, 소듐아크릴레이트의 농도는 5 내지 20%, TEMED의 농도는 0.5 내지 0.8 %인, 세포 확대 방법.
- 제12항에 있어서,상기 (a) 단계의 배양은 2℃ 내지 10℃에서 4시간 내지 12시간 동안 배양되는, 세포 확대 방법.
- 제12항에 있어서,상기 (b) 단계의 응고반응은 5분 내지 60분간 수행되는, 세포 확대 방법.
- 제12항에 있어서,상기 (a) 단계 내지 (b) 단계를 1 내지 3회 반복하는, 세포 확대 방법.
- 제12항에 있어서,상기 (a) 단계 내지 (b) 단계를 1 내지 3회 반복하여 제조된 세포 필름을 제1용액에서 2℃ 내지 10℃에서 6시간 내지 24시간 동안 배양하는 단계를 추가로 포함하는, 세포 확대 방법.
- 제12항 내지 제17항 중 어느 한 항의 (b) 단계 이후,(c) 상기 (b) 단계에서 응고된 세포 필름을 PBS에서 배양하는 단계; 및(d) 상기 (c) 단계에서 배양된 세포 필름을 증류수에서 배양하는 단계;를 추가로 포함하는, 세포 확대 방법.
- 제18항에 있어서,상기 PBS의 농도는 0.05 내지 0.2M 인, 세포 확대 방법.
- 제18항에 있어서,상기 (c) 단계 또는 (d) 단계에서 형광 이미징 분석을 위한 항체 또는 염색 물질을 처리하는 단계를 추가로 포함하는, 세포 확대 방법.
