Description
本発明は、三次元造形物の製造方法および三次元造形物作製用インクセットに関し、特に、再生医療の分野などで、ソフトマテリアルからなる複雑な形状の三次元造形物を製造する場合に適した三次元造形物の製造方法に関する。
三次元プリンタと呼ばれる造形装置を使用して三次元造形物を製造する技術が、種々の分野で実用化を目指して研究されている。例えば再生医療の分野では、人工臓器などとして使用するために、ソフトマテリアルからなる、ブリッジ部位や穴がある複雑な形状の三次元造形物を製造することが求められている。三次元プリンタを用いて、バイオインクからこのような三次元造形物を製造しようとすると、バイオインクが流出(液だれ)してゲル化前に崩壊したり、重力による変形が起こったりするため、造形精度の向上が求められている。
特許文献1には、再生医療などの分野への応用を目指し、天然高分子および/または合成高分子を含む3Dプリンティングシステム用インクにシルクナノファイバーを添加することで、ノズルからの吐出時には粘度が低く、吐出時の圧力を低くすることができ、且つ、プリント後の基板上では粘度が高く、造形物の変形を充分に防止できることが開示されている。一方、材料範囲の更なる拡大、新たな三次元造形物の製法の開発が望まれるところである。
本発明は、例えばソフトマテリアルからなる複雑な形状の三次元造形物を製造する場合であっても、良好な造形精度で三次元造形物を製造できる三次元造形物の製造方法および三次元造形物作製用インクセットを提供することを目的とする。
本発明者らは、種々検討したところ、光架橋性高分子を含むモデル材用インク、および、サポート材と架橋因子を含むサポート材用インクを、それぞれ吐出した後に、吐出された光架橋性高分子を光硬化させ、サポート材用インクの造形物を除去して三次元造形物を造形すると、例えばソフトマテリアルからなる複雑な形状の三次元造形物を作製する場合であっても、良好な造形精度で三次元造形物を製造できることを見出した。
すなわち、本発明(1)は、(a)光架橋性高分子を含むモデル材用インク、および、(b)サポート材と第1の架橋因子を含むサポート材用インクを、それぞれ吐出する工程、

本発明によれば、例えばソフトマテリアルからなる複雑な形状の三次元造形物を製造する場合であっても、光照射により三次元造形物を良好な造形精度で製造することができる。
<<三次元造形物の製造方法>>
本発明の、三次元造形物の製造方法は、(a)光架橋性高分子を含むモデル材用インク、および、(b)サポート材と第1の架橋因子を含むサポート材用インクを、それぞれ吐出する工程、吐出された光架橋性高分子を光硬化させる工程、ならびに、サポート材用インクの造形物を除去する工程を含むことを特徴とする。
本発明の、三次元造形物の製造方法は、(a)光架橋性高分子を含むモデル材用インク、および、(b)サポート材と第1の架橋因子を含むサポート材用インクを、それぞれ吐出する工程、吐出された光架橋性高分子を光硬化させる工程、ならびに、サポート材用インクの造形物を除去する工程を含むことを特徴とする。
以下に、図1を参照しながら、本発明の三次元造形物の製造方法を簡単に説明する。図1は、本発明の三次元造形物の製造方法によって、ブリッジ部位を有する三次元造形物を作製する工程を説明するための模式図である。図1(a)は、三次元プリンタのノズル3aからサポート材用インク1を吐出する様子を模式的に示す。図1(b)は、三次元プリンタのノズル3bからモデル材用インク2を吐出する様子を模式的に示す。なお、ノズル3aとノズル3bは、同一のノズルであってもよいが、異なるものであることが好ましい。ここで、サポート材用インク1を吐出し、次いで、サポート材用インク1で囲まれる位置にモデル材用インク2を吐出して1つの層を形成することができ、これを繰り返して積層構造とする。
図1(a)および図1(b)に示すように、吐出されたサポート材用インク1と吐出されたモデル材用インク2は接している。例えば、サポート材用インク1がサポート材とともに第1の架橋因子を含む場合、吐出されたサポート材用インク1から吐出されたモデル材用インク2に第1の架橋因子が供給され、吐出されたモデル材用インク2中の光架橋性高分子を光により架橋し、モデル材用インク2をゲル化する。図1(c)では、モデル材用インク2のゲル化が完了した後、得られた造形物を、サポート材用インク1の造形物の除去溶液5に浸漬し、サポート材用インク1の造形物を除去する工程を示している。サポート材用インク1の造形物を除去することで、ブリッジ部位2aや穴2bがある複雑な形状の三次元造形物2A(モデル材用インクの造形物)の作製も可能となる。
本発明の三次元造形物の製造方法ではビーカー、シャーレ等の円筒形状の容器、角筒形状の容器、その他の各種形状の容器中にモデル材用インク及びサポート材用インクを吐出して三次元造形物を作製してもよい。容器の内壁がモデル材用インク及び/又はサポート材用インクの支持体となるため、より高さの高い造形物を作製することができる。なお、容器の材質は、モデル材用インク及び/又はサポート材用インクの支持体となるものである限り特に限定されず、ガラス、樹脂等からなるものを使用できる。また、容器は三次元造形により製造されたものでもよい。
<(a)光架橋性高分子を含むモデル材用インク、および、(b)サポート材と第1の架橋因子を含むサポート材用インクを、それぞれ吐出する工程>
吐出する工程では、例えば、三次元プリンタのノズルから、基板上などの目的箇所に、モデル材用インク(a)および(b)サポート材と第1の架橋因子を含むサポート材用インクを吐出する。インクを吐出する工程で三次元プリンタを用いる場合、三次元プリンタは、特に限定されず、インクジェット式、レーザー光式、押し出し式などのいずれであってもよいが、押し出し式の三次元プリンタが好ましい。モデル材用インク(a)を吐出するノズルは、サポート材用インク(b)を吐出するノズルと異なることが好ましい。
吐出する工程では、例えば、三次元プリンタのノズルから、基板上などの目的箇所に、モデル材用インク(a)および(b)サポート材と第1の架橋因子を含むサポート材用インクを吐出する。インクを吐出する工程で三次元プリンタを用いる場合、三次元プリンタは、特に限定されず、インクジェット式、レーザー光式、押し出し式などのいずれであってもよいが、押し出し式の三次元プリンタが好ましい。モデル材用インク(a)を吐出するノズルは、サポート材用インク(b)を吐出するノズルと異なることが好ましい。
((a)光架橋性高分子を含むモデル材用インク)
モデル材用インク(a)は、光架橋性高分子を含み、サポート材用インクに含まれる第1の架橋因子によって光架橋性高分子が光により架橋することで、ゲル状となるものである。光架橋性高分子としては、特に限定されないが、例えば、高分子基材と、フェノール性水酸基を有する化合物とを結合させて得られる高分子、ゼラチンメタクリレート等の(メタ)アクリレート基を有する高分子、アミノ基含有多糖類、ポリフェノール類などが挙げられる。高分子基材が光架橋性官能基を有する場合は、そのまま光架橋性高分子として使用してもよいし、さらにフェノール性水酸基を有する化合物などと結合させて光架橋性官能基を導入して使用してもよい。
モデル材用インク(a)は、光架橋性高分子を含み、サポート材用インクに含まれる第1の架橋因子によって光架橋性高分子が光により架橋することで、ゲル状となるものである。光架橋性高分子としては、特に限定されないが、例えば、高分子基材と、フェノール性水酸基を有する化合物とを結合させて得られる高分子、ゼラチンメタクリレート等の(メタ)アクリレート基を有する高分子、アミノ基含有多糖類、ポリフェノール類などが挙げられる。高分子基材が光架橋性官能基を有する場合は、そのまま光架橋性高分子として使用してもよいし、さらにフェノール性水酸基を有する化合物などと結合させて光架橋性官能基を導入して使用してもよい。
高分子基材としては、特に限定されず、例えば、多糖類、核酸、炭水化物、タンパク質、ポリペプチド、ポリ(α-ヒドロキシ酸)、ポリラクトン、ポリアミノ酸、ポリ無水物、ポリオルトエステル、ポリ(無水物-co-イミド)、ポリオルトカーボネート、ポリ(α-ヒドロキシアルカノエート)、ポリジオキサノン、ポリホスホエステル、ポリ乳酸、ポリ(L-ラクチド)(PLLA)、ポリ(D,L-ラクチド)(PDLLA)、ポリグリコール酸、ポリグリコリド(PGA)、ポリ(ラクチド-co-グリコリド)(PLGA)、ポリ(L-ラクチド-co-D,L-ラクチド)、ポリ(D,L-ラクチド-co-トリメチレンカーボネート)、ポリヒドロキシブチレート(PHB)、ポリ(ε-カプロラクトン)、ポリ(δ-バレロラクトン)、ポリ(γ-ブチロラクトン)、ポリカプロラクトン、ポリ(メタ)アクリル酸、ポリカルボン酸、ポリアリルアミン塩酸塩、ポリジアリルジメチルアンモニウムクロリド、ポリエチレンイミンなどのポリアルキレンイミン、ポリプロピレンフマレート、ポリビニルアルコール、ポリビニルピロリドン、ポリエチレンなどのポリアルキレン、ポリメチルメタクリレート、炭素繊維、ポリエチレングリコールなどのポリアルキレングリコール、ポリエチレンオキシドなどのポリアルキレンオキシド、ポリビニルアルコール、ポリビニルピロリドン、ポリエチルオキサゾリン、ポリ(エチレンオキシド)-co-ポリ(プロピレンオキシド)ブロック共重合体、ポリ(エチレンテレフタレート)ポリアミド、これら高分子基材の塩などが挙げられる。
多糖類としては、例えば、セルロース、ヘミセルロース、デキストラン、ヒアルロン酸、キチン、キトサン、アルギン酸、コンドロイチン硫酸、澱粉、プルラン、カラギーナン、カードラン、フコイダン、アミロース、アミロペクチン、ペクチン、これらのアルカリ金属塩等の塩、誘導体などが挙げられる。なお、多糖類の誘導体としては、例えば、カルボキシメチルセルロース、メチルセルロース、脱アセチル化キチン類、脱アセチル化キトサン類などが挙げられる。
タンパク質としては、アルブミン、グロブリン、プロラミン、グルテリン、ヒストン、プロタミン、フィブロイン等の硬タンパク質などの単純タンパク質、核タンパク質、糖タンパク質、色素タンパク質、リンタンパク質などの複合タンパク質、ゼラチン、プロテオース、ペプトンなどの誘導タンパク質などが挙げられる。
高分子基材の塩における塩は、特に限定されず、酸付加塩、金属塩、アンモニウム塩、有機アミン塩などが挙げられる。酸付加塩としては、例えば、塩酸塩、硫酸塩、リン酸塩などの無機酸塩;酢酸塩、マレイン酸塩、フマル酸塩、酒石酸塩、クエン酸塩などの有機酸塩が挙げられる。金属塩としては、例えば、ナトリウム塩、カリウム塩などのアルカリ金属塩、カルシウム塩などのアルカリ土類金属塩、マグネシウム塩、アルミニウム塩、亜鉛塩などが挙げられる。有機アミン塩としては、例えば、モルホリン、ピペリジンなどの塩が挙げられる。
高分子基材としては、中でも、多糖類、タンパク質が、容易に供給されること、得られる三次元造形物の特性が良好なことから好ましく、多糖類がより好ましい。上記高分子基材は、上述したものの1種のみを用いてもよく、2種以上を併用してもよい。
フェノール性水酸基を有する化合物としては、特に限定されないが、下記一般式(1):
(式中、X1は、水酸基、アミノ基、または、カルボン酸(塩)基を表す。X2~X6のうち1つまたは2つは、水酸基を表し、残りは水素原子または炭素数1~6のアルコキシ基を表す。X2~X6のうち2つが水酸基を表す場合、2つの水酸基はオルトまたはパラ位の位置関係にあることが好ましい。Rは、置換基を有していてもよい炭素数1~10の二価の炭化水素基を表す。)で表される化合物が好ましい。高分子基材にフェノール性水酸基を導入した架橋性高分子は、光レドックス触媒および重合開始剤、または光重合開始剤の存在下で露光することにより容易に架橋する。X1は、フェノール性水酸基を高分子基材へ導入しやすいことから、カルボン酸(塩)基またはアミノ基が好ましく、アミノ基がより好ましい。なお、上記アミノ基は、第1級アミノ基又は第2級アミノ基が好ましい。上記カルボン酸(塩)基としては、カルボン酸基、カルボン酸基のアルカリ金属塩、アルカリ土類金属塩が好ましい。また、Rが表す二価の炭化水素基の炭素数は、1~8が好ましく、1~6がより好ましい。Rが表す二価の炭化水素基としては、特に限定されないが、アルキレン基、アルキルアルキレン基などが挙げられる。また、Rが有してもよい置換基としては、例えばアミド基、エステル基、エーテル基などが挙げられる。なお、X1は、水酸基である場合、アルコール性水酸基である。

フェノール性水酸基を有する化合物として、例えば、チラミン、ホモバニリン酸、これらの誘導体類などのフェノール性水酸基を1つ有する化合物、ドーパミン、ノルアドレナリン、アドレナリンなどのカテコールアミン類などのフェノール性水酸基を2つ有する化合物が挙げられる。これらの中でも、得られる架橋性高分子の架橋反応性の観点から、フェノール性水酸基を1つ有する化合物が好ましく、チラミン、チラミン誘導体類がより好ましい。
フェノール性水酸基を有する化合物としては、市販品を使用してもよい。市販品としては、例えば、チラミン塩酸塩(4-(2-アミノエチル)フェノール・塩酸塩、製品番号:T2879、シグマアルドリッチ社製)などが挙げられる。
高分子基材とフェノール性水酸基を有する化合物との結合方法は、特に限定されないが、フェノール性水酸基を有する化合物が有していてもよいカルボキシル基、アミノ基またはアルコール性水酸基などの官能基と、高分子基材が有していてもよいアミノ基、カルボキシル基またはチオール基などの官能基とを縮合剤を用いて縮合する方法、グルタミルトランスフェラーゼを用いて縮合する方法などが挙げられる。
縮合剤としては、特に限定されないが、例えば、1,1-カルボニルジイミダゾール、ジシクロヘキシルカルボジイミド、3-(3-ジメチルアミノプロピル)-1-エチルカルボジイミド・塩酸塩(水溶性カルボジイミド:WSCD・HCl)などが挙げられる。高分子基材およびフェノール性水酸基を有する化合物を、縮合剤を用いて縮合する方法、グルタミルトランスフェラーゼを用いて縮合する方法においては、使用する高分子基材、フェノール性水酸基を有する化合物、縮合剤やグルタミルトランスフェラーゼの性質に合わせて適宜、pH、高分子基材の濃度、反応温度、溶媒などを選択できる。
高分子基材に対するフェノール性水酸基を有する化合物の導入率(修飾率)は、高分子基材が有する縮合反応基の全体における、縮合反応した(修飾された)縮合反応基の割合である。ゲル化速度と硬度(硬すぎないこと)をバランス良く両立する観点からは、高分子基材が有する縮合反応基100mol%に対して0.3~20mol%が好ましく、1~10mol%がより好ましい。
光架橋性高分子としては、アミノ基含有多糖類、ポリフェノール類も使用できる。アミノ基含有多糖類としては、例えば、アセチル化キチン、キトサンなどが挙げられる。また、ポリフェノール類としては、例えば、カテキン類化合物、アントシアニン類化合物、ルチン、天然色素などが挙げられる。
ゼラチンメタクリレート(ゼラチンメタクリロイル)とは、ゼラチンの側鎖にメタクリロイル基を導入したものであり、その水溶液は光重合開始剤存在下で、その光重合開始剤に適した波長の光を照射するとゲルを形成する。
モデル材用インク(a)中の光架橋性高分子の濃度は、通常0.1~30重量%であり、0.5~20重量%が好ましく、0.75~10重量%がより好ましい。光架橋性高分子の濃度が、0.1重量%未満であると、三次元造形物の強度が充分なものでなくなるおそれがあり、30重量%を超えると、モデル材用インクの粘度が高くなり過ぎ、ノズルからの吐出時の圧力が高くなり、モデル材用インク(a)の材質に影響を及ぼすおそれがある。また、モデル材用インク(a)中の架橋性高分子の濃度は、通常0.1~30w/v%(g/100mL)であり、0.5~20w/v%(g/100mL)が好ましく、0.75~10w/v%(g/100mL)がより好ましい。
本発明の製造方法では、モデル材用インク(a)は、さらに、第2の架橋因子を含むことが好ましい。第2の架橋因子とは、サポート材用インクに含まれる第1の架橋因子とともに光架橋性高分子の架橋に係わるが、それ単独では光架橋性高分子を架橋しない成分であり、例えば光レドックス触媒などである。具体的には、たとえばトリス(2,2’-ビピリジル)ジクロロルテニウム(II)などのルテニウム錯体、トリス(2-フェニルピリジン)イリジウム(III)などのイリジウム錯体が挙げられる。
モデル材用インク(a)中の第2の架橋因子の含有量は、光架橋性高分子100質量部に対して0.1~50重量部が好ましく、1~30重量部がより好ましい。また、モデル材用インク(a)中の第2の架橋因子の濃度は、特に限定されないが、0.01mM以上、1M以下であることが好ましい。該濃度は、0.1mM以上であることがより好ましく、1mM以上であることがさらに好ましく、5mM以上であることが特に好ましい。
モデル材用インク(a)は、サポート材用インク(b)の非存在下で光架橋性高分子の光架橋が進まないように、実質的に光重合開始剤を含まないことが好ましい。光重合開始剤の含有量は、モデル材用インク(a)中、1質量%以下が好ましく、0.5質量%以下がより好ましく、0質量%がさらに好ましい。
モデル材用インク(a)は、光架橋性高分子、第2の架橋因子の他に、その他の成分を含有していてもよい。その他の成分としては、各種細胞、溶媒、連鎖移動剤、可塑剤、pH調整剤、緩衝剤、保存料、溶媒、紫外線吸収剤、色材、界面活性剤などが挙げられる。溶媒としては、例えば、水、リン酸緩衝生理食塩水(PBS)、水と有機溶剤との混合物などが挙げられる。有機溶剤としては特に限定されず、例えば、エタノール、グリセリン、ジメチルスルホキシドなどが挙げられる。
細胞の由来動物としては、哺乳類、鳥類、爬虫類などが挙げられ、哺乳類が好ましい。哺乳類としては、人間、ブタ、ウシ、ヒツジ、ヤギ、ウサギ、マウス、ラット、イヌ、ネコ、ニワトリなどが挙げられ、人間が好ましい。細胞種としては、上皮細胞、線維芽細胞、軟骨細胞、骨芽細胞、平滑筋細胞、神経細胞、幹細胞などが挙げられる。
モデル材用インクにおける細胞の濃度は特に限定されないが、1×103~1×109cells/mLが好ましく、1×104~1×107cells/mLがより好ましい。
モデル材用インク(a)は、室温で液状であることが好ましい。室温で液状であることで、3Dプリンタなどを用いて容易に吐出することができる。モデル材用インク(a)の20℃における粘度は、特に限定されないが、10mPa・s以上が好ましく、20mPa・s以上がより好ましく、50mPa・s以上がさらに好ましい。また、上記粘度は、100000mPa・s以下が好ましく、10000mPa・s以下がより好ましく、2000mPa・s以下がさらに好ましい。本発明の製造方法により、ゲル化前のモデル材用インク(a)の流出(液だれ)を充分に防止できるため、低粘度のモデル材用インク(a)を好適に使用できる。なお、上記粘度は、コーン・プレート型のE型粘度計を用いて、20rpmの回転速度で測定されるものである。
モデル材用インク(a)は、上述した光架橋性高分子、第2の架橋因子、必要に応じてその他の成分を混合することによって、製造することができる。
((b)サポート材と第1の架橋因子を含むサポート材用インク)
サポート材用インク(b)は、吐出された後、架橋により形態を保持し、造形物となるものであってもよいが、それ自体で形態を保持でき、そのまま造形物となるものが簡便で好ましい。サポート材用インク(b)の造形物は、モデル材用インク(a)が充分にゲル化するまでモデル材用インク(a)の形状を保持し、モデル材用インク(a)の流出(液だれ)や、重力によるモデル材用インク(a)の変形を防ぐ。モデル材用インク(a)がゲル化した後は、サポート材用インク(b)の造形物は、後述する方法で除去される。
サポート材用インク(b)は、吐出された後、架橋により形態を保持し、造形物となるものであってもよいが、それ自体で形態を保持でき、そのまま造形物となるものが簡便で好ましい。サポート材用インク(b)の造形物は、モデル材用インク(a)が充分にゲル化するまでモデル材用インク(a)の形状を保持し、モデル材用インク(a)の流出(液だれ)や、重力によるモデル材用インク(a)の変形を防ぐ。モデル材用インク(a)がゲル化した後は、サポート材用インク(b)の造形物は、後述する方法で除去される。
サポート材用インク(b)が含むサポート材は、天然高分子や合成高分子などの高分子であり、高分子としては、例えば、多糖類、核酸、炭水化物、タンパク質、ポリペプチド、ポリアミノ酸、ポリ乳酸、ポリエチレンオキシドなどのポリアルキレンオキシド、ポリエチレンイミンなどのポリアルキレンイミン、ポリビニルピロリドン、ポリエチレングリコールなどのポリアルキレングリコール、ポリアクリル酸、ポリメタクリル酸、ポリグルタミン酸、カルボキシビニルポリマ―、カルボキシメチルセルロース、メチルセルロース、ヒドロキシエチルセルロース、アルブミン、ゼラチン、コラーゲン、シルクフィブロイン、ポリビニルアルコール、これら高分子の塩などが挙げられる。上記高分子としては、多糖類、核酸、炭水化物、タンパク質、ポリペプチド、ポリアミノ酸、ポリ乳酸、ポリエチレンオキシド、ポリエチレンイミン、ポリビニルピロリドン、ポリエチレングリコール、ポリアクリル酸、ポリグルタミン酸、アルブミン、ゼラチン、コラーゲン、ポリビニルアルコール、これら高分子の塩が好ましい。多糖類、タンパク質、高分子の塩における塩は、高分子基材において前述した多糖類、タンパク質、高分子基材の塩における塩と同様である。また上記サポート材は、前述した高分子を架橋剤で架橋したものであってもよい。サポート材は、光照射で硬化しないものが好ましい。具体的には、光架橋性高分子や光架橋性モノマーを含まないことが好ましい。
多糖類としては、例えば、セルロース、ヘミセルロース、デキストラン、ヒアルロン酸、キチン、キトサン、アルギン酸、コンドロイチン硫酸、澱粉、プルラン、カラギーナン、カードラン、フコイダン、アミロース、アミロペクチン、ペクチン、これらのアルカリ金属塩等の塩、誘導体などが挙げられる。
中でも、サポート材は、水溶性高分子又は水分散性高分子であることが好ましい。これにより、後述するように、サポート材用インク(b)の造形物を除去する工程において、水又は水溶液を用いて温和な条件で簡便に造形物を除去できる。ここで、本明細書中、水溶性高分子の、20℃の水100gに対する溶解度は特に限定されないが、1g以上が好ましく、5g以上がより好ましい。
水溶性高分子又は水分散性高分子は、高分子水性ゲルを形成し、形成された高分子水性ゲルは、イオン性化合物と作用して凝集または粘度低下するものであることが好ましい。イオン性化合物と作用して凝集または粘度低下する高分子水性ゲルを形成する水溶性高分子又は水分散性高分子は、カルボン酸基などの酸基を有する炭化水素鎖(例えば、ポリアクリル酸)を主鎖として、架橋剤で架橋したものが挙げられる。架橋剤としては、特に限定されないが、アリルショ糖、ペンタエリスリトールなどが挙げられる。イオン性化合物については、後述する。
サポート材用インク(b)中の高分子の濃度は、サポート材用インク(b)の擬塑性をより一層向上する観点からは、0.1重量%以上が好ましく、0.3重量%以上がより好ましく、0.5重量%以上がさらに好ましく、1.0重量%以上が特に好ましい。また、高分子の濃度は、30.0重量%以下が好ましく、15.0重量%以下がより好ましく、10.0重量%以下がさらに好ましい。なお、サポート材用インク(b)中に高分子が2種以上含まれる場合、上記の濃度は、サポート材用インク中に含まれる全ての高分子の合計の濃度を意味する。
サポート材用インク(b)は、さらに、第1の架橋因子を含む。第1の架橋因子とは、単独で、もしくは、モデル材用インク及び/又はサポート材用インクに含まれる第2の架橋因子とともに、光架橋性高分子の架橋に係わる成分であり、例えば重合開始剤などである。具体的には、たとえば、ペルオキソ二硫酸ナトリウム(SPS)、リチウムフェニル-2,4,6-トリメチルベンゾイルホスフィネート(LAP)、ジフェニル-2,4,6-トリメチルベンゾイルホスフィンオキシド、2-ヒドロキシ-4-(2-ヒドロキシエトキシ)-2-メチルプロピオフェノンなどの重合開始剤が挙げられる。
第1の架橋因子の含有量は、光架橋性高分子100質量部に対して0.1~50重量部が好ましく、0.5~30重量部がより好ましい。また、サポート材用インク(b)中の第1の架橋因子の濃度は、特に限定されないが、0.01mM以上、1M以下であることが好ましい。該濃度は、0.1mM以上であることがより好ましく、1mM以上であることがさらに好ましく、5mM以上であることが特に好ましい。
サポート材用インク(b)は、さらに、第2の架橋因子を含むことが好ましい。第2の架橋因子の具体例は、前述したとおりである。
サポート材用インク(b)中の第2の架橋因子の含有量は、光架橋性高分子100質量部に対して 0.1~50重量部が好ましく、1~30重量部がより好ましい。また、サポート材用インク(b)中の第2の架橋因子の濃度は、特に限定されないが、0.01mM以上、1M以下であることが好ましい。該濃度は、0.1mM以上であることがより好ましく、1mM以上であることがさらに好ましく、5mM以上であることが特に好ましい。
サポート材用インク(b)は、サポート材用インクの造形物を容易に除去するために、サポート材用インク単独で光架橋しないものとできるように、実質的に光架橋性高分子または光架橋性モノマーを含まないことが好ましい。光架橋性高分子または光架橋性モノマーの含有量は、サポート材用インク(b)中、1質量%以下が好ましく、0.1質量%以下がより好ましく、0質量%がさらに好まししい。
サポート材用インク(b)は、サポート材としての高分子、第1の架橋因子の他に、その他の成分を含有していてもよい。その他の成分としては、グリセロールなどの保湿剤、アルカリ金属水酸化物、有機アミンなどのpH調整剤、保存料、色材、溶媒、可塑剤、紫外線吸収剤、界面活性剤などが挙げられる。溶媒としては、例えば、水、リン酸緩衝生理食塩水(PBS)、水と有機溶剤との混合物などが挙げられる。有機溶剤としては特に限定されず、例えば、エタノール、グリセリンなどが挙げられる。
サポート材用インク(b)は、室温で液状またはゲル状であることが好ましく、ゲル状であることがより好ましい。室温でゲル状であることで、3Dプリンタなどを用いて容易に造形することができる。サポート材用インク(b)の、20℃での、せん断速度0.01(1/s)における粘度は、10000mPa・s以上が好ましく、それ自体で形態を保持できるようにする観点からは、100000mPa・s以上がより好ましく、500000mPa・s以上がさらに好ましい。また、該粘度は、その上限は特に限定されないが、通常は10000000mPa・s以下である。
サポート材用インク(b)の、20℃での、せん断速度10(1/s)における粘度は、10mPa・s以上が好ましく、100mPa・s以上がより好ましく、1000mPa・s以上がさらに好ましい。また、該粘度は、その上限は特に限定されないが、通常は10000mPa・s未満である。なお、上記粘度は、サーモフィッシャーサイエンティフィック社製、粘度・粘弾性測定装置を用いて測定される値である。
サポート材用インク(b)の、20℃での、せん断速度10(1/s)における粘度に対するせん断速度0.01(1/s)における粘度の比は、5以上が好ましく、10以上がより好ましく、20以上がさらに好ましく、50以上が特に好ましい。これにより、サポート材用インク(b)の擬塑性がより優れるものとなる。
サポート材用インク(b)は、高分子と、第1の架橋因子と、必要に応じてその他の成分を混合することによって、製造することができる。混合時の条件は、特に限定されない。
上述したように、本発明の製造方法では、通常、三次元プリンタを用いて、モデル材用インク(a)とサポート材用インク(b)をそれぞれ吐出し、層を形成し、これを繰り返して積層する。吐出されたモデル材用インク(a)と吐出されたサポート材用インク(b)が接することにより、吐出された第1の架橋因子が単独で、または、第2の架橋因子とともに、光架橋性高分子を架橋し、モデル材用インク(a)の造形物を造形する。ここで、吐出されたサポート材用インク(b)が含む第1の架橋因子、または、第1の架橋因子および第2の架橋因子は、吐出されたモデル材用インク(a)に供給される。なお、本発明の製造方法では、第1の架橋因子、または、第1の架橋因子および第2の架橋因子による光架橋性高分子の架橋とともに、イオン架橋、光架橋、酵素架橋、温度変化などが架橋性高分子の架橋に寄与していても構わない。
(第二のサポート材を含む第二のサポート材用インク)
本発明の製造方法では、モデル材用インク(a)とサポート材用インク(b)(以下では、第一のサポート材用インクともいう)とともに、モデル材用インク及び第一のサポート材用インクの支持材として第二のサポート材用インクを吐出して製造してもよい。第二のサポート材用インクはあらかじめ吐出して三次元造形物(容器または外枠)を作製してもよいし、三次元造形物(容器または外枠)を一部分だけ作製して残りの部分を、モデル材用インク及び第一のサポート材用インクとともに吐出して作製してもよい。また全てを同時に作製してもよい。第二のサポート材用インクで作製される三次元造形物(容器または外枠)が、モデル材用インク及び第一のサポート材用インクの支持体となるため、モデル材用インク及び第一のサポート材用インクの崩壊をより充分に防止でき、また、所望により、より高さの高い三次元造形物を作製することができる。また、このような第二のサポート材用インクは、第1の架橋因子を含まなくてもよく、このような第二のサポート材用インクに第一のサポート材用インクを置き換えて第一のサポート材用インクの吐出量を減らすことで、第一のサポート材用インクに含まれる第1の架橋因子の使用量を節約することもできる。なお、第二のサポート材用インクが含む第二のサポート材は、モデル材用インク及び/又はサポート材用インクの支持体となるものである限り特に限定されず、上記サポート材用インク(b)に使用するサポート材やガラス、樹脂等を使用できる。また、後述するように、第二のサポート材用インクの造形物は適当な材料を選択することにより、第一のサポート材用インクの造形物と同様に取り除くことができる。モデル材用インクはその一部または全部が第二のサポート材で構成される造形物と接してもよい。
本発明の製造方法では、モデル材用インク(a)とサポート材用インク(b)(以下では、第一のサポート材用インクともいう)とともに、モデル材用インク及び第一のサポート材用インクの支持材として第二のサポート材用インクを吐出して製造してもよい。第二のサポート材用インクはあらかじめ吐出して三次元造形物(容器または外枠)を作製してもよいし、三次元造形物(容器または外枠)を一部分だけ作製して残りの部分を、モデル材用インク及び第一のサポート材用インクとともに吐出して作製してもよい。また全てを同時に作製してもよい。第二のサポート材用インクで作製される三次元造形物(容器または外枠)が、モデル材用インク及び第一のサポート材用インクの支持体となるため、モデル材用インク及び第一のサポート材用インクの崩壊をより充分に防止でき、また、所望により、より高さの高い三次元造形物を作製することができる。また、このような第二のサポート材用インクは、第1の架橋因子を含まなくてもよく、このような第二のサポート材用インクに第一のサポート材用インクを置き換えて第一のサポート材用インクの吐出量を減らすことで、第一のサポート材用インクに含まれる第1の架橋因子の使用量を節約することもできる。なお、第二のサポート材用インクが含む第二のサポート材は、モデル材用インク及び/又はサポート材用インクの支持体となるものである限り特に限定されず、上記サポート材用インク(b)に使用するサポート材やガラス、樹脂等を使用できる。また、後述するように、第二のサポート材用インクの造形物は適当な材料を選択することにより、第一のサポート材用インクの造形物と同様に取り除くことができる。モデル材用インクはその一部または全部が第二のサポート材で構成される造形物と接してもよい。
図11は、本発明の三次元造形物の製造方法において、第二のサポート材用インクをさらに用いて三次元造形物を作製する工程を説明するための模式図である。以下に、図1と異なる部分のみ説明する。図11(a)は、三次元プリンタのノズル13aから第二のサポート材用インク11sを吐出する様子を模式的に示す。図11(b)は、三次元プリンタのノズル13bから第一のサポート材用インク11を吐出する様子を模式的に示す。図11(c)は、三次元プリンタのノズル13cからモデル材用インク12を吐出する様子を模式的に示す。なお、ノズル13a、ノズル13b、および、ノズル13cは、同一のノズルであってもよいが、それぞれ異なるものであることが好ましい。図11では、第二のサポート材用インク11sを吐出し、第二のサポート材用インク11sで囲まれる位置に第一のサポート材用インク11を吐出し、第一のサポート材用インク11で囲まれる位置にモデル材用インク12を吐出することで1つの層を形成し、これを繰り返して積層構造とする。
図11(a)~図11(c)に示すように、吐出された第二のサポート材用インク11sは、内周部および外周部がともに平らな薄い壁状であり、吐出された第一のサポート材用インク11と接してこれを支持し、吐出されたモデル材用インク12も間接的に支持する。第二のサポート材用インク11sとして硬質な造形物を形成できるものを用いることで、モデル材用インク12および第一のサポート材用インク11がゲル化の途中などで崩れることをより充分に防止できる。また、所望により、より高さの高い三次元造形物を作製することができる。
図11(d)は、図1(c)と同様に、モデル材用インク12のゲル化が完了した後、得られた造形物を、除去溶液15に浸漬し、第一のサポート材用インク11の造形物及び第二のサポート材用インク11sの造形物を除去する工程を示している。この工程については、後述する。
図12は、第一のサポート材用インク21及び第二のサポート材用インク21sの吐出範囲を図8に示したように変更した以外は、図11に示した模式図と同様である。以下に、図11と異なる部分のみ説明する。図12では、第二のサポート材用インクを、内周部が下方へ拡がる段状となるように吐出し、図11の場合に対して、第一のサポート材用インクの一部を第二のサポート材用インクに置き換えている。第二のサポート材用インク21sとして、硬質な造形物を形成でき、第1の架橋因子を含まないものを用いることで、モデル材用インク22および第一のサポート材用インク21がゲル化の途中などで崩れることをより充分に防止しながら、第一のサポート材用インク21の吐出量を減らすことができ、第1の架橋因子の使用量を節約することができる。
図13は、第二のサポート材用インク31sの吐出範囲を図13に示したように変更した以外は、図12に示した模式図と同様である。以下に、図12と異なる部分のみ説明する。図13では、第二のサポート材用インクを、吐出されたモデル材用インク32および吐出された第一のサポート材用インク31の形状に合わせて、外周部が上方へ拡がる段状となるように吐出し、図12と比べて、第二のサポート材用インクの吐出量をも減らすことができる。
<吐出された光架橋性高分子を光硬化させる工程>
光硬化させる工程では、サポート材用インクからモデル材用インク層に移動した第1の架橋因子、または、第1の架橋因子および第2の架橋因子と光架橋性高分子が光の照射により反応して硬化する。光としては、可視光、紫外光、X線および電子線などが挙げられる。

<サポート材用インクの造形物を除去する工程>
除去する工程は、サポート材用インク(b)の造形物を除去し、モデル材用インク(a)の造形物が得られる限り特に限定されないが、例えば、サポート材用インク(b)の造形物を、サポート材用インク(b)の造形物の除去溶液に浸漬および/または洗浄することでおこなうことが好ましい。除去する工程は、モデル材用インク(a)の造形物(三次元造形物)に及ぼす影響を小さくする観点から、温和な条件下でおこなうことが好ましく、例えば、10~40℃の温度条件下でおこなうことが好ましい。また、除去溶液の20℃でのpHは、酵素活性の点から、2~10が好ましく、5~9がより好ましい。なお、サポート材用インクの造形物は、サポート材用インクからモデル材用インクに第1の架橋因子が供給された後の、サポート材(高分子)などの残りの成分からなるものである。
除去する工程は、サポート材用インク(b)の造形物を除去し、モデル材用インク(a)の造形物が得られる限り特に限定されないが、例えば、サポート材用インク(b)の造形物を、サポート材用インク(b)の造形物の除去溶液に浸漬および/または洗浄することでおこなうことが好ましい。除去する工程は、モデル材用インク(a)の造形物(三次元造形物)に及ぼす影響を小さくする観点から、温和な条件下でおこなうことが好ましく、例えば、10~40℃の温度条件下でおこなうことが好ましい。また、除去溶液の20℃でのpHは、酵素活性の点から、2~10が好ましく、5~9がより好ましい。なお、サポート材用インクの造形物は、サポート材用インクからモデル材用インクに第1の架橋因子が供給された後の、サポート材(高分子)などの残りの成分からなるものである。
サポート材用インク(b)とともに上述した第二のサポート材用インクをさらに用いる場合は、サポート材用インク(b)の造形物と第二のサポート材用インクの造形物とを、それぞれ異なる方法で除去してもよいが、上記除去する工程によりサポート材用インク(b)の造形物とともに第二のサポート材用インクの造形物も除去することが好ましい。
サポート材が、水溶性高分子又は水分散性高分子である場合、除去溶液として、水や水溶液を使用できる。また水溶性高分子又は水分散性高分子が、高分子水性ゲルを形成し、形成された高分子水性ゲルは、イオン性化合物と作用して凝集または粘度低下するものである場合は、除去溶液として、イオン性化合物の水溶液を特に好適に使用できる。水溶性高分子又は水分散性高分子が、イオン性化合物と作用するとは、水溶性高分子又は水分散性高分子とイオン性化合物とが接触した結果、高分子水性ゲルが凝集または粘度低下する限り特に限定されないが、例えば、イオン性化合物と反応(化学反応)するものが挙げられる。高分子水性ゲルが、酵素処理によって分解されるものである場合は、分解酵素を使用でき、酵素処理後に培養液、緩衝液、生理食塩水および水などを使用することで、サポート材用インクの造形物を除去できる。サポート材が、温度によって相転移するものである場合、加熱や冷却を行うことで相転移させることができ、相転移後に水や水溶液を使用することで、サポート材用インクの造形物を除去できる。
イオン性化合物としては、カルシウム、亜鉛、マグネシウム、バリウム、ストロンチウム、銅、鉄、アルミニウム、コバルト、等の多価金属塩が挙げられる。それらの内、塩化カルシウム、水酸化カルシウム、炭酸カルシウム、乳酸カルシウムが好ましく、塩化カルシウムがより好ましい。
除去溶液中のイオン性化合物の濃度は、1mM以上であることが好ましく、10mM以上であることがより好ましい。該濃度は、2M以下が好ましく、0.2M以下がより好ましい。
分解酵素としては、セルラーゼ、アミラーゼ、デキストラナーゼ、カラギナーゼ、アルギン酸リアーゼ、ヒアルロニダーゼ、キチナーゼ及びキトサナーゼ等が挙げられるが、これらに限定されない。
除去溶液中の分解酵素の濃度は、0.1~5重量%が好ましく、0.1~1重量%がより好ましい。
<その他の工程>
本発明の製造方法は、上述したモデル材用インクおよびサポート材用インクをそれぞれ吐出する工程の後、サポート材用インクの造形物を除去する工程の前に、モデル材を充分にゲル化させるために、吐出したモデル材用インクおよびサポート材用インクを静置する工程を含んでいてもよい。
本発明の製造方法は、上述したモデル材用インクおよびサポート材用インクをそれぞれ吐出する工程の後、サポート材用インクの造形物を除去する工程の前に、モデル材を充分にゲル化させるために、吐出したモデル材用インクおよびサポート材用インクを静置する工程を含んでいてもよい。
本発明の製造方法を用いて得られる三次元造形物は、例えばソフトマテリアルからなる複雑な形状の三次元造形物とすることが可能であり、特に、再生医療の分野で、人工臓器などとして好適に使用できるものである。なお、三次元造形物の形状は、特に限定されないが、ブリッジ部位および/または穴があるものにも適している。三次元造形物の寸法は、適宜設定できる。
<<三次元造形物作製用インクセット>>
本発明は、(a)光架橋性高分子を含むモデル材用インク、および、(b)サポート材と第1の架橋因子を含むサポート材用インクからなることを特徴とする。光架橋性高分子、サポート材、第1の架橋因子や、その他の成分については、上述した通りである。本発明の三次元造形物作製用インクセットを用いて、本発明の三次元造形物を好適に製造することができる。
本発明は、(a)光架橋性高分子を含むモデル材用インク、および、(b)サポート材と第1の架橋因子を含むサポート材用インクからなることを特徴とする。光架橋性高分子、サポート材、第1の架橋因子や、その他の成分については、上述した通りである。本発明の三次元造形物作製用インクセットを用いて、本発明の三次元造形物を好適に製造することができる。
以下、実施例を挙げて本発明を説明するが、本発明は以下の実施例に限定されない。以下、「部」または「%」は特記ない限り、それぞれ「重量部」または「重量%」を意味する。
製造例1(モデル材用インク1の作製)
0.1Mの2-モルフォリノエタンスルホン酸一水和物(2-Morpholinoethanesulfolic acid,monohydrate、MES)緩衝液(pH5.8)1Lにヒアルロン酸ナトリウム(株式会社富士メディシエ製)10gを溶解させた。さらにチラミン塩酸塩0.56g、水溶性カルボジイミド(WSCD・HCl)1.43gおよびN-ヒドロキシコハク酸イミド(NHS)0.43gを10mLの精製水に溶解させた溶液を加えた後、室温で22時間攪拌した。得られた反応混合物を限外ろ過にてろ過し、凍結真空乾燥を行うことによってHYA-Ph(フェノール性水酸基の修飾率:2.9mol%、Mw:300,000)を作製した。
0.1Mの2-モルフォリノエタンスルホン酸一水和物(2-Morpholinoethanesulfolic acid,monohydrate、MES)緩衝液(pH5.8)1Lにヒアルロン酸ナトリウム(株式会社富士メディシエ製)10gを溶解させた。さらにチラミン塩酸塩0.56g、水溶性カルボジイミド(WSCD・HCl)1.43gおよびN-ヒドロキシコハク酸イミド(NHS)0.43gを10mLの精製水に溶解させた溶液を加えた後、室温で22時間攪拌した。得られた反応混合物を限外ろ過にてろ過し、凍結真空乾燥を行うことによってHYA-Ph(フェノール性水酸基の修飾率:2.9mol%、Mw:300,000)を作製した。
作製したHYA-Phを20mg秤量し、リン酸緩衝生理食塩水PBS2.0mLを加えて溶解した。トリス(2,2’-ビピリジル)ジクロロルテニウム(II)六水和物(分子量640.53(無水)、748.62(六水和物))を3.0mg秤量し、HYA-Phの溶液に加えてRu2+濃度2.0mmol/L、HYA-Ph濃度1.0w/v%のモデル材用インク1を作製した。
製造例2(サポート材用インク1の作製)
プロゼリー(ジェクス株式会社製 高粘度タイプ)を4mL分注し、ペルオキソ二硫酸ナトリウム(SPS,MW238.10)を9.524mg加えて10mMのSPSを含有するサポート材用インク1を作製した。

製造例3(モデル材用インク2の作製)
製造例1で作製したHYA-Phを20mg秤量し、リン酸緩衝生理食塩水PBS2.0mLを加えて溶解した。トリス(2,2’-ビピリジル)ジクロロルテニウム(II)六水和物(分子量640.53(無水)、748.62(六水和物))を3.0mgとペルオキソ二硫酸ナトリウム(SPS,MW238.10)を4.8 mgをそれぞれ秤量し、HYA-Phの溶液に加えてRu2+濃度2.0mmol/L、SPS濃度10.0mmol/L、HYA-Ph濃度1.0w/v%のモデル材用インク2を作製した。
製造例1で作製したHYA-Phを20mg秤量し、リン酸緩衝生理食塩水PBS2.0mLを加えて溶解した。トリス(2,2’-ビピリジル)ジクロロルテニウム(II)六水和物(分子量640.53(無水)、748.62(六水和物))を3.0mgとペルオキソ二硫酸ナトリウム(SPS,MW238.10)を4.8 mgをそれぞれ秤量し、HYA-Phの溶液に加えてRu2+濃度2.0mmol/L、SPS濃度10.0mmol/L、HYA-Ph濃度1.0w/v%のモデル材用インク2を作製した。
製造例4(モデル材用インク3の作製)
リン酸緩衝生理整理食塩水に10(w/v)%となるようにゼラチン(Sigma)を分散させた溶液を50℃に加温して、ゼラチンを溶解させた後、無水メタクリル酸をゼラチンの質量の60%の質量となるように添加し、1時間撹拌した。その後、透析膜(分画分子量10,000~20,000)に入れて、40℃の精製水で3日間透析を行った。透析終了後に、凍結真空乾燥することによりゼラチンメタクリレートを得た。
リン酸緩衝生理整理食塩水に10(w/v)%となるようにゼラチン(Sigma)を分散させた溶液を50℃に加温して、ゼラチンを溶解させた後、無水メタクリル酸をゼラチンの質量の60%の質量となるように添加し、1時間撹拌した。その後、透析膜(分画分子量10,000~20,000)に入れて、40℃の精製水で3日間透析を行った。透析終了後に、凍結真空乾燥することによりゼラチンメタクリレートを得た。
ゼラチンメタクリレートを100mg秤量し、蛍光粒子(蛍光パウダー、品番:MB-8502,松野工業株式会社製)5mgとリン酸緩衝生理食塩水PBS2.0mLを加えて溶解し、ゼラチンメタクリレート濃度5w/v%のモデル材用インク3を作製した。
製造例5(サポート材用インク2の作製)
プロゼリー(高粘度タイプ、ジェクス株式会社製)を4mL分注し、LAP(リチウムフェニル-2,4,6-トリメチルベンゾイルホスフィネート)を40mg加えて1w/v%のLAPを含有するサポート材用インク2を作製した。
プロゼリー(高粘度タイプ、ジェクス株式会社製)を4mL分注し、LAP(リチウムフェニル-2,4,6-トリメチルベンゾイルホスフィネート)を40mg加えて1w/v%のLAPを含有するサポート材用インク2を作製した。
製造例6(サポート材用インク3の作製)
製造例5において、LAPを加えなかったこと以外は製造例5と同様にしてLAPを含有しないサポート材用インク3を作製した。
製造例5において、LAPを加えなかったこと以外は製造例5と同様にしてLAPを含有しないサポート材用インク3を作製した。
製造例7(モデル材用インク4の作製)
ゼラチンメタクリレートを100mg秤量し、蛍光粒子(蛍光パウダー、品番:MB-8502,松野工業株式会社製)5mgとリン酸緩衝生理食塩水PBS2.0mLを加えて溶解し、1w/v%となるようにLAPを加え、ゼラチンメタクリレート濃度5w/v%のモデル材用インク4を作製した。
ゼラチンメタクリレートを100mg秤量し、蛍光粒子(蛍光パウダー、品番:MB-8502,松野工業株式会社製)5mgとリン酸緩衝生理食塩水PBS2.0mLを加えて溶解し、1w/v%となるようにLAPを加え、ゼラチンメタクリレート濃度5w/v%のモデル材用インク4を作製した。
製造例8(モデル材用インク5の作製)
0.1Mの2-モルフォリノエタンスルホン酸一水和物(2-Morpholinoethanesulfolic acid,monohydrate、MES)緩衝液(pH6.0)に3.3(w/v)%となるように変性ポリビニルアルコール(日本酢ビ・ポバール株式会社製AF-17)を溶解させた溶液を120℃で15分加温した。室温に冷却した後、チラミン塩酸塩、水溶性カルボジイミド(WSCD・HCl)およびN-ヒドロキシコハク酸イミド(NHS)をそれぞれ3.3、3.3、1.0(w/v)%となるように加えた後、40℃で20時間攪拌した。得られた反応溶液をエタノール中に加えて沈殿させた後、沈殿生成物を90%エタノール水溶液およびエタノールで洗浄し、減圧乾燥を行うことによりPVA-Ph(フェノール性水酸基の修飾率:2.4×10-4mol/g)を作製した。作製したPVA-Phが濃度5.0w/v%(g/100mL)となるように、製造例1と同様にしてモデル材用インク5を作製した。
0.1Mの2-モルフォリノエタンスルホン酸一水和物(2-Morpholinoethanesulfolic acid,monohydrate、MES)緩衝液(pH6.0)に3.3(w/v)%となるように変性ポリビニルアルコール(日本酢ビ・ポバール株式会社製AF-17)を溶解させた溶液を120℃で15分加温した。室温に冷却した後、チラミン塩酸塩、水溶性カルボジイミド(WSCD・HCl)およびN-ヒドロキシコハク酸イミド(NHS)をそれぞれ3.3、3.3、1.0(w/v)%となるように加えた後、40℃で20時間攪拌した。得られた反応溶液をエタノール中に加えて沈殿させた後、沈殿生成物を90%エタノール水溶液およびエタノールで洗浄し、減圧乾燥を行うことによりPVA-Ph(フェノール性水酸基の修飾率:2.4×10-4mol/g)を作製した。作製したPVA-Phが濃度5.0w/v%(g/100mL)となるように、製造例1と同様にしてモデル材用インク5を作製した。
製造例9(モデル材用インク6の作製)
0.05Mの2-モルフォリノエタンスルホン酸一水和物(2-Morpholinoethanesulfolic acid,monohydrate、MES)緩衝液(pH6.0)500mLにゼラチン(ゼライス株式会社製)10gを溶解させた。さらにチラミン塩酸塩1.84g、水溶性カルボジイミド(WSCD・HCl)1.85gおよびN-ヒドロキシコハク酸イミド(NHS)0.57gを5mLの精製水に溶解させた溶液を加えた後、室温で20時間攪拌した。得られた反応混合物を限外ろ過にてろ過し、凍結真空乾燥を行うことによってGela-Ph(フェノール性水酸基の修飾率:2.7×10-4mol/g)を作製した。作製したGela-Phが濃度4.0w/v%(g/100mL)となるように、製造例1と同様にしてモデル材用インク6を作製した。
0.05Mの2-モルフォリノエタンスルホン酸一水和物(2-Morpholinoethanesulfolic acid,monohydrate、MES)緩衝液(pH6.0)500mLにゼラチン(ゼライス株式会社製)10gを溶解させた。さらにチラミン塩酸塩1.84g、水溶性カルボジイミド(WSCD・HCl)1.85gおよびN-ヒドロキシコハク酸イミド(NHS)0.57gを5mLの精製水に溶解させた溶液を加えた後、室温で20時間攪拌した。得られた反応混合物を限外ろ過にてろ過し、凍結真空乾燥を行うことによってGela-Ph(フェノール性水酸基の修飾率:2.7×10-4mol/g)を作製した。作製したGela-Phが濃度4.0w/v%(g/100mL)となるように、製造例1と同様にしてモデル材用インク6を作製した。

0.05Mの2-モルフォリノエタンスルホン酸一水和物(2-Morpholinoethanesulfolic acid,monohydrate、MES)緩衝液(pH6.0)600mLにアルギン酸ナトリウム(株式会社舞昆のこうはら製)10gを溶解させた。さらにチラミン塩酸塩0.57g、水溶性カルボジイミド(WSCD・HCl)2.90gおよびN-ヒドロキシコハク酸イミド(NHS)0.87gを10mLの精製水に溶解させた溶液を加えた後、室温で19時間攪拌した。得られた反応混合物を限外ろ過にてろ過し、凍結真空乾燥を行うことによってAlg-Ph(フェノール性水酸基の修飾率:3.1mol%)を作製した。作製したAlg-Phが濃度2.0w/v%(g/100mL)となるように、製造例1と同様にしてモデル材用インク7を作製した。

実施例1
三次元プリンタ(製品名:BIO X、Cellink社製)を用いて、製造例1で作製したモデル材用インク1と、製造例2で作製したサポート材用インク1を基板上に、光を露光しながらそれぞれ吐出して層を形成し、これを繰り返して積層し、三次元造形物として、厚み:1mm、高さ:5mm、1辺の長さ:6mmの六角筒を作製した。露光する光の波長は450nmで、光強度は約60,000ルクスである。

比較例1
製造例3で作製したモデル材用インク2を用い、サポート材用インクを使用しなかったこと以外は実施例1と同様にして三次元造形物を作製した。
製造例3で作製したモデル材用インク2を用い、サポート材用インクを使用しなかったこと以外は実施例1と同様にして三次元造形物を作製した。
図5に作製した三次元造形物の写真を示す。1w/v%という三次元造形物には適切ではない低濃度であっても、架橋因子を含むサポート材用インクを使用すると、実施例1に示すように元データに忠実な三次元造形物を作製することができた。一方で、第1の架橋因子を含むモデル材用インクを使用し、またサポート材を使用しなかった場合には、比較例1に示すように、構造物上面が凹凸になり、うまく着液することが出来ず、構造物は得られなかった。サポート材無しの条件の溶液はゲル化時間自体はかなり短く、線状の構造物が崩れたような結果になった。
実施例2
三次元プリンタ(製品名:BIO X、Cellink社製)を用いて、製造例4で作製したモデル材用インク3と、製造例5で作製したサポート材用インク2を基板上に、光を露光しながらそれぞれ吐出して層を形成し、これを繰り返して積層し、三次元造形物として、厚み:1mm、高さ:5mm、1辺の長さ:6mmの六角筒を作製した。露光する光の波長は450nmで、光強度は約60,000ルクスである。

実施例2において、サポート材用インクとして製造例6で作製したLAPを含有しないサポート材用インク3を使用した以外は実施例2と同様にして三次元造形物を作製した。

比較例3
実施例2において、サポート材用インクを使用せず、製造例7で作製したLAPを含有するモデル材用インク4を使用したこと以外は実施例2と同様にして三次元造形物を作製した。

図6に作製した三次元造形物の写真を示す。実施例2のように、5w/v%という三次元造形物には適切ではない低濃度であっても、架橋因子を含むサポート材用インクを使用すると、元データに忠実な三次元造形物を作製することができた。一方、いずれのインクにもLAPを含まない比較例2では、重合が開始せず、三次元構造物を得ることができなかった。モデル材用インクにLAPを含むもののサポート材用インクを使用していない比較例3では、粘度が低く構造物をうまく積層できず、吐出した溶液がノズル先に絡みつき、構造物に着液していくことができなかった。5w/v%ゼラチンメタクリレート溶液の粘度が低く、そのヒドロゲルが脆弱なため、構造物上面の凹凸が大きかったことと、溶液を吐出した直後に露光されてゲル化してしまったことが原因である。この問題は光量を小さくしても解決できなかった。
上記の結果から、材料が柔らかいもの(ソフトマテリアル)からなる複雑な形状の造形物を製造する場合に、本発明の製造方法を適用することで、良好な造形精度で三次元造形物を得ることができることが分かった。
実施例3
モデル材用インクとして製造例8で作製したインク5を使用し、製造例2で作成したサポート材用インク1のSPS濃度を8mMに変更した以外は、実施例1と同様にして三次元造形物を作製した。

実施例4
モデル材用インクとして製造例9で作製したインク6を使用し、製造例2で作成したサポート材用インク1のSPS濃度を8mMに変更した以外は、実施例1と同様にして三次元造形物を作製した。

実施例5
モデル材用インクとして製造例10で作製したインク7を使用し、製造例2で作成したサポート材用インク1のSPS濃度を8mMに変更した以外は、実施例1と同様にして三次元造形物を作製した。
モデル材用インクとして製造例10で作製したインク7を使用し、製造例2で作成したサポート材用インク1のSPS濃度を8mMに変更した以外は、実施例1と同様にして三次元造形物を作製した。
図7~9に示すように、変性ポリビニルアルコールを含むモデル材用インク5を使用した実施例3、ゼラチンを含むモデル材用インク6を使用した実施例4、アルギン酸を含むモデル材用インク7を使用した実施例5でも、ヒアルロン酸を含むモデル材用インクを使用した実施例1と同様に、造形精度が非常に良好な造形物を作製することができた。
実施例6
マウス線維芽細胞10T1/2細胞を製造例1で作製したモデル材用インク1に1×105cells/mLで分散させ、得られた細胞含有モデル材用インクと製造例2で作製したサポート材用インクを用いて実施例1と同様にして三次元造形物(厚み:1mm、高さ:1mm、1辺の長さ:6mmの六角筒)を作製した。
マウス線維芽細胞10T1/2細胞を製造例1で作製したモデル材用インク1に1×105cells/mLで分散させ、得られた細胞含有モデル材用インクと製造例2で作製したサポート材用インクを用いて実施例1と同様にして三次元造形物(厚み:1mm、高さ:1mm、1辺の長さ:6mmの六角筒)を作製した。
造形直後に細胞を含む造形物を生細胞染色試薬Calcein―AMと死細胞染色試薬PIをそれぞれ1μg/mLで含むリン酸緩衝生理食塩水(pH7.4)に10分間浸して染色を行った。その結果を撮影した写真を図10に示す。細胞の生存率は85%であり、高い値を維持していた。

Claims (9)
- (a)光架橋性高分子を含むモデル材用インク、および、(b)サポート材と第1の架橋因子を含むサポート材用インクを、それぞれ吐出する工程、
吐出された光架橋性高分子を光硬化させる工程、ならびに、
サポート材用インクの造形物を除去する工程
を含む三次元造形物の製造方法。 - 前記モデル材用インク(a)が、さらに第2の架橋因子を含む請求項1に記載の三次元造形物の製造方法。
- 前記モデル材用インク(a)が、実質的に光重合開始剤を含まない請求項1または2に記載の三次元造形物の製造方法。
- 前記サポート材用インク(b)が、実質的に光架橋性高分子または光架橋性モノマーを含まない請求項1~3のいずれか1項に記載の三次元造形物の製造方法。
- 前記サポート材用インク(b)が、さらに第2の架橋因子を含む請求項1~4のいずれか1項に記載の三次元造形物の製造方法。
- 前記サポート材が、水溶性高分子又は水分散性高分子である請求項1~5のいずれか1項に記載の三次元造形物の製造方法。
- 前記水溶性高分子又は水分散性高分子が、イオン性化合物と作用して凝集または粘度低下する水性ゲルを形成する水溶性高分子又は水分散性高分子であり、サポート材用インクの造形物を除去する工程において、イオン性化合物を作用させて前記サポート材用インク(b)の造形物を除去する請求項6に記載の三次元造形物の製造方法。
- (a)光架橋性高分子を含むモデル材用インク、および、
(b)サポート材と第1の架橋因子を含むサポート材用インク
からなる三次元造形物作製用インクセット。 - 前記サポート材用インク(b)が、イオン性化合物と作用して凝集または粘度低下する水性ゲルを形成する水溶性高分子又は水分散性高分子を含む請求項8に記載の三次元造形物作製用インクセット。
