Abstract
本発明は、光老化細胞を標的とする免疫の誘導のための手段を提供すること、又は、光老化細胞を標的とする抗体若しくはその結合性断片を提供する。具体的には、本発明は、免疫原性GPR17ポリペプチド、免疫原性CD34ポリペプチド、免疫原性GABRR1ポリペプチド、免疫原性OR2AG2ポリペプチド、免疫原性CMKLR1ポリペプチド、免疫原性CDH19ポリペプチド、免疫原性CD93ポリペプチド、免疫原性AVPR2ポリペプチド、免疫原性CCR7ポリペプチド及び免疫原性OXGR1ポリペプチドからなる群より選ばれる少なくとも1種のポリペプチドを含有し、該少なくとも1種のポリペプチドが、キャリアタンパク質に結合された、又はキャリアタンパク質に結合されていない、対象中の光老化細胞に対する免疫を誘導するための、抗原組成物を含む。
Description
本発明は、光老化細胞に対する免疫を誘導するための抗原組成物及び抗原発現用組成物、並びに、光老化細胞に対して結合性を有する抗体組成物に関する。
細胞老化は、紫外線照射による光老化と、加齢などによる自然老化の2種類に大別され、その特徴が異なっている。例えば、皮膚の光老化は、顔、首、手の甲などの太陽光が当たる部分に好発し、シミ、シワ(特に深いシワ)、たるみ、皮膚の肥厚、皮膚の黄色や褐色の着色などの原因となる。一方、皮膚の自然老化は太陽光の照射に関わらず進行する。自然老化では、シミの形成はほとんどなく、シワも細かく浅いものが形成される。また、自然老化では皮膚が委縮する傾向にあり、皮膚の着色もない。
しかし、これまでに、細胞の自然老化のマーカーとしては老化関連酸性β-ガラクトシダーゼ(SA-β-Gal)やサイクリン依存性キナーゼ阻害因子(p16)などが知られていたが(非特許文献1)、光老化に特有の細胞マーカーは知られていなかった。

光老化を予防又は改善するうえで、正常細胞に対して光老化細胞に対して選択的に薬理活性や免疫応答を及ぼすための手段が必要とされている。
しかし、既知の細胞老化マーカーであるSA-β-Galやサイクリン依存性キナーゼ阻害因子(p16)は光老化細胞に対して特異的なものではなく、細胞表面にもないことから、生きた光老化細胞を標的とする薬物療法に利用することができない。
そこで本発明は、光老化細胞を標的とする免疫の誘導のための手段を提供すること、又は光老化細胞を標的とする抗体若しくはその結合性断片を提供することを課題とする。
本発明者らは、鋭意検討した結果、まず光老化細胞に特異的に発現し、自然老化細胞に発現していない遺伝子を探索し、GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1の10遺伝子を光老化細胞表面マーカーとして同定した。
そして、本発明者らは、以下の知見をもとに本発明を完成させた。
(1)GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1タンパク質からなる群より選ばれる少なくとも1種のポリペプチドを含む抗原組成物が、光老化細胞に対する免疫を誘導すること。
(2)前記免疫の誘導に基づいて、前記抗原組成物が光老化細胞の減少又は増加抑制の用途に使用できること。
(3)GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1タンパク質からなる群より選ばれる少なくとも1種のポリペプチドに対して結合性を有する抗体又はその結合性断片を含有する抗体組成物により、光老化細胞を特異的に標的化できること。
(4)前記抗体又はその結合性断片が光老化細胞を標的とすることにより、前記抗体組成物が光老化細胞への薬物の特異的な送達又は光老化細胞の減少若しくは増加抑制の用途に使用できること。
(1)GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1タンパク質からなる群より選ばれる少なくとも1種のポリペプチドを含む抗原組成物が、光老化細胞に対する免疫を誘導すること。
(2)前記免疫の誘導に基づいて、前記抗原組成物が光老化細胞の減少又は増加抑制の用途に使用できること。
(3)GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1タンパク質からなる群より選ばれる少なくとも1種のポリペプチドに対して結合性を有する抗体又はその結合性断片を含有する抗体組成物により、光老化細胞を特異的に標的化できること。
(4)前記抗体又はその結合性断片が光老化細胞を標的とすることにより、前記抗体組成物が光老化細胞への薬物の特異的な送達又は光老化細胞の減少若しくは増加抑制の用途に使用できること。

本発明の抗原発現用組成物は、光老化細胞の細胞表面マーカーに由来するポリペプチドをコードするポリヌクレオチドを含有し、対象に投与することにより、対象内で当該ポリペプチドが発現する。その結果、本発明の抗原発現用組成物は、光老化細胞に対する免疫を誘導することができる。
本発明の抗体組成物は、光老化細胞に結合性を有する抗体又はその結合性断片を含有し、光老化細胞を標的化することができる。
本明細書において、「光老化細胞」とは、紫外線照射によって老化状態となった生細胞であって、癌化していない細胞全般を表す。光老化細胞は、特に限定されないが、例えば以下の(1)~(3)の少なくとも1つ、好ましくは(1)~(3)すべての特徴を有する:
(1)β-ガラクトシダーゼ(SA-β-Gal)又はサイクリン依存性キナーゼ阻害因子(p16)陽性である;
(2)細胞分裂の停止又は著しい低下が見られる;
(3)細胞が肥大化している。

前記光老化細胞の由来としては、例えば皮膚の細胞が挙げられる。皮膚の細胞の具体例としては、例えば、ケラチノサイト、色素細胞(メラノサイト)、ランゲルハンス細胞、メルケル細胞などの表皮系細胞;線維芽細胞などの真皮系細胞;脂肪細胞などの脂肪組織系細胞;毛乳頭細胞、毛母細胞、内毛根鞘細胞、外毛根鞘細胞などの毛包系細胞などが挙げられる。中でも前記光老化細胞の由来としては、表皮系細胞又は真皮系細胞が好ましく、ケラチノサイト又は線維芽細胞がより好ましく、ケラチノサイトがさらに好ましい。
本明細書において、特定の物質(抗原)に対する「免疫を誘導する」又は「免疫誘導」とは、特定の物質(抗原)に対して検出可能な免疫応答を引き起こす、増強する、迅速化する又は延長することを表す。前記免疫応答としては、例えば、体液性免疫系の応答(例えば、B細胞による応答、より具体的には、特定の物質に対する抗体の産生及び分泌);細胞性免疫系の応答(例えば、T細胞、NK細胞の活性化、より具体的には、特定の物質に対するT細胞又はNK細胞の活性化);自然免疫系の応答(例えば、Toll-like受容体シグナル系の活性化、リンホカイン(Th1、Th2、Th17等のサイトカイン又はケモカイン)の分泌、マクロファージの活性化、樹状細胞の活性化等)、免疫系細胞の増殖促進(例えば、前記特定の物質に対する抗体を産生するB細胞、又は前記特定の物質に対するT細胞若しくはNK細胞の増殖促進)等が挙げられる。
本明細書において、「免疫原性」は、ある物質が、必要に応じてキャリアタンパク質との結合及び/又はアジュバントの併用を行って対象に投与された場合に、対象中で当該物質に対する免疫を誘導することができる能力を表す。

前記抗原ポリペプチドのアミノ酸残基数は、それぞれ独立して、例えば3以上であり、本発明の効果をより顕著に奏する観点から、好ましくは5以上、より好ましくは8以上、さらに好ましくは10以上とすることができ、そして200以下、100以下、50以下、40以下、30以下又は20以下とすることができる。
前記抗原ポリペプチドは、対応する野生型ポリペプチドのアミノ酸配列に対するアミノ酸配列の同一性が、本発明の効果をより顕著に奏する観点から、好ましくは80%以上、より好ましくは90%以上、さらに好ましくは95%以上、さらにより好ましくは98%以上、特に好ましくは99%以上の同一性を有する。

前記抗原ポリペプチドの調製法は、そのアミノ酸残基の数に応じて適宜選択し得るが、例えば、固相合成法等により化学的に合成されてもよいし、ポリペプチドをコードするmRNAを細胞又は無細胞翻訳系で翻訳させて生合成してもよい。得られたポリペプチドは、適宜クロマトグラフィー等の既知の精製方法により精製して使用することができる。
前記抗原ポリペプチドは、それぞれ翻訳後修飾されていてもよいし、又は未修飾のポリペプチドであってもよい。
前記抗原ポリペプチドは、本発明の効果をより顕著に奏する観点から、任意で、免疫誘導を促進するために使用されるキャリアタンパク質に結合されてもよい。ここで、キャリアタンパク質への結合は、例えば、当該抗原ポリペプチド、キャリアタンパク質及び任意で架橋剤等の化学物質の存在下で、化学的処理によって共有結合的又は非共有結合的な連結によってなされる。あるいは、キャリアタンパク質への結合は、当該抗原ポリペプチドとキャリアタンパク質が一体のポリペプチドとして翻訳されることによってなされてもよい。キャリアタンパク質としては、例えば、キーホールリンペットヘモシアニン(KLH)、ウシ血清アルブミン(BSA)、チキンガンマグロブリン(CGG)、オボアルブミン等が挙げられる。
前記抗原ポリペプチドは、主要組織適合複合体(MHC)との結合性を増強するため、あるいは細胞内又は特定の組織へのペプチド送達を促進するための付加的なアミノ酸残基又はアミノ酸残基の変異を有してもよい。
(抗原発現用組成物、抗原発現用ポリヌクレオチド)
本発明の抗原発現用組成物は、前記抗原ポリペプチドの少なくとも1種をコードするポリヌクレオチド(本明細書中「抗原発現用ポリヌクレオチド」と呼ぶ)を1種又は2種以上含有する。1種のポリヌクレオチドに複数のポリペプチドをコードする場合は、2種以上のポリペプチドがポリシストロニックに発現するように構成されてもよい。
本発明の抗原発現用組成物は、前記抗原ポリペプチドの少なくとも1種をコードするポリヌクレオチド(本明細書中「抗原発現用ポリヌクレオチド」と呼ぶ)を1種又は2種以上含有する。1種のポリヌクレオチドに複数のポリペプチドをコードする場合は、2種以上のポリペプチドがポリシストロニックに発現するように構成されてもよい。
前記抗原発現用ポリヌクレオチドは、対象中で前記抗原ポリペプチドを発現し得るものであれば特に限定されない。前記抗原発現用ポリヌクレオチドは、例えばDNA又はRNAであり、ヌクレオチド修飾(例えば、ホスホロチオエート(PS)修飾、シュードウリジン修飾等のヌクレオチド安定性や翻訳効率の向上に関する修飾)を一部又は全体的に含んでいてもよい。中でも前記抗原発現用ポリヌクレオチドは、DNA又はmRNAが好ましく、保存安定性の観点からDNAがより好ましい。
前記抗原発現用ポリヌクレオチドは、発現のために必要な制御エレメント(例えば、プロモーター、エンハンサー、ターミネーター、サイレンサー、ポリシストロニック発現因子(IRES、2Aペプチド配列等))の少なくとも1種を作動可能に連結していることが好ましく、少なくとも1種のプロモーターを作動可能に連結していることがより好ましい。
本明細書において、ある構成要素が「作動可能に連結する」とは、当該構成要素がその意図する様式で機能することを可能にするように配置することを表す。例えば、ある制御エレメントをある遺伝子に対して作動可能に連結するとは、遺伝子の発現を当該制御エレメントの意図する様式で制御できるように、当該制御エレメントが遺伝子をコードするポリヌクレオチドにライゲーションされることを表す。
一実施形態では、前記抗原発現用ポリヌクレオチドは、発現ベクターであり得る。前記発現ベクターには、前記制御エレメントを少なくとも1種含んでもよく、少なくとも1種のプロモーターを含むことがより好ましい。また、前記発現ベクターには、分泌シグナル配列、タグ配列、複製起点、ポリA配列、マルチクローニングサイト、選別マーカーなどをさらに含むことができる。
前記発現ベクターは、例えば、プラスミドベクター、コスミドベクター又はウイルスベクターであり得る。
前記ウイルスベクターとしては、例えば、アデノウイルス、アデノ随伴ウイルス、単純ヘルペスウイルス、サイトメガロウイルス、ワクシニアウイルス、Epstein-Barrウイルス、レトロウイルス(レンチウイルス等)等のウイルスベクターが挙げられる。

前記抗原組成物及び前記抗原発現用組成物の具体的な態様は、それぞれ、特に限定されないが、例えば、医薬組成物(医薬品、医薬部外品を含む)、化粧料組成物、食品(特定保健用食品、機能性表示食品、栄養機能食品、サプリメント等の機能性食品を含む)又は飼料が挙げられ、好ましくは医薬組成物又は化粧料組成物である。
前記抗原組成物及び前記抗原発現用組成物の具体的な態様は、それぞれ、特に限定されないが、例えば、医薬組成物(医薬品、医薬部外品を含む)、化粧料組成物、食品(特定保健用食品、機能性表示食品、栄養機能食品、サプリメント等の機能性食品を含む)又は飼料が挙げられ、好ましくは医薬組成物又は化粧料組成物である。

(機能、用途)
本明細書の実施例に示されるように、GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1の各遺伝子は、光老化細胞において、対応する正常細胞に比べて発現量が増加している。そのため、これらの遺伝子にコードされたタンパク質は、光老化細胞特異的なマーカーとして機能し得る。
本明細書の実施例に示されるように、GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1の各遺伝子は、光老化細胞において、対応する正常細胞に比べて発現量が増加している。そのため、これらの遺伝子にコードされたタンパク質は、光老化細胞特異的なマーカーとして機能し得る。
前記抗原組成物は、これらマーカーの全長又は一部を含む。そのため、前記抗原組成物を対象に投与すると、これらタンパク質に対する免疫を誘導し、その結果光老化細胞に対する免疫を誘導することができる。そして、前記抗原組成物は、誘導された免疫により、対象中の光老化細胞を減少させ、又は増加抑制することができ、さらには光老化状態を予防又は改善することができる。
前記抗原発現用組成物は、対象に投与すると、それに含まれるポリヌクレオチドにコードされた光老化細胞特異的なマーカーの全長又は一部が発現する。その結果、前記抗原組成物と同様に、対象中の光老化細胞に対する免疫を誘導することができる。そして、前記抗原発現用組成物は、誘導された免疫により、対象中の光老化細胞を減少させ、又は増加抑制することができ、さらには光老化状態を予防又は改善することができる。
以上によれば、前記抗原組成物又は前記抗原発現用組成物は、それぞれ、対象に投与して、対象中の光老化細胞に対する免疫を誘導するために用いることができる。
また、前記抗原組成物又は前記抗原発現用組成物は、それぞれ、対象に投与して、対象中の光老化細胞を減少させる、又は増加抑制するために用いることができる。

さらに、前記抗原ポリペプチド、抗原発現用ポリヌクレオチド又はそれらの組み合わせは、対象に投与して、(I)対象中の光老化細胞に対する免疫を誘導するため、(II)その有効量を投与することにより、対象中の光老化細胞を減少させ、若しくは増加抑制するため、又は、(III)対象中の光老化状態を予防若しくは改善するための方法に使用することができる。

前記抗原組成物、前記抗原発現用組成物、前記抗原ポリペプチド又は前記抗原発現用ポリヌクレオチドは、さらに別のアジュバントと併用して使用することができる。当該アジュバントは、上記のアジュバントを使用することができる。

[抗体組成物]
本発明の抗体組成物は、GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1タンパク質からなる群より選ばれる少なくとも1種のポリペプチドに対して結合性を有する抗体又はその抗原結合性断片を含有する。

前記抗体の可変領域のフレームワーク領域及び定常領域の配列は、前記抗原に対する結合性が失われない限りにおいて特に限定されず、例えばヒト化抗体、ヒトキメラ抗体、マウスキメラ抗体等の、複数種の遺伝子に由来する抗体の断片を遺伝子工学的手法によって組み合わせたものであってもよい。
本明細書において、「キメラ抗体」とは、非ヒト動物抗体由来の可変領域とヒト抗体の定常領域とを組み合わせてなる抗体を表す。

前記抗体又はその抗原結合性断片は、その用途に応じて、薬物を1種単独又は2種以上を組み合わせて連結した、薬物コンジュゲートであり得る。ここで、前記連結は、共有結合、又は、配位結合、イオン結合、疎水性相互作用、ファンデルワールス相互作用若しくはその他の非共有結合であり得る。
前記薬物は、例えば、細胞傷害剤、抗老化成分、抗酸化成分、ビタミン類、抗炎症剤、細胞賦活化剤等が挙げられる。
前記細胞傷害剤は、化学療法剤、放射性同位体、放射線増感剤又は光応答性プローブ〔IRDye(登録商標)700DX(Nature Medicine, 2011, Vol.17, pp.1685-1691)などのフタロシアニン系プローブ、マラカイトグリーン等〕などが挙げられる。細胞傷害剤を連結した前記連結体は、光老化細胞へ送達されることにより光老化細胞の増加の抑制又は減少をもたらし得ることから、光老化状態の予防又は改善に好適に使用される。
前記抗老化成分、抗酸化成分、ビタミン類、抗炎症剤又は細胞賦活化剤は、光老化細胞の老化状態自体やそれに伴う異常を改善し得ることから、光老化状態の予防又は改善に好適に使用される。
本発明の抗体又はその抗原結合性断片は、タグを付加することができる。このようなタグとしては、例えば、FLAG、HA(ヘマグルチニン)、GST(グルタチオン-S-トランスフェラーゼ)、Myc、ポリヒスチジン(6×Hisタグ等)等が挙げられる。これらのタグは抗体の固相化、精製、検出等に用いることができる。タグが付加された抗体又は抗原結合性断片は、例えば、本発明の抗体又はその抗原結合性断片をコードするポリヌクレオチドに、タグをコードするポリヌクレオチドを付加したものを作製し、動物細胞等のin vivo又は無細胞翻訳系等のin vitroのタンパク質発現システムに導入することにより製造することができる。
前記抗体は、例えば、抗体を産生させる対象に、上記の免疫の誘導方法を行って血漿に分泌される抗体を分離し、適宜精製して得ることができる。あるいは、前記抗体は、免疫誘導した個体から得られた抗体産生細胞を用いてハイブリドーマを作製して生合成することもできる。
また、前記抗体又はその抗原結合性断片は、それらのタンパク質をコードするポリヌクレオチドを、例えば、動物細胞(チャイニーズハムスター卵巣(CHO)細胞、サル腎臓細胞(COS)、HEK 293細胞、NSO細胞、HeLa細胞、ベビーハムスター腎臓(BHK)細胞、ヒト肝細胞ガン細胞(例えば、Hep G2)等)、非動物真核細胞(昆虫細胞等)、細菌(大腸菌等)、酵母、昆虫、植物、トランスジェニック動物(ウシ、ニワトリ等)又はin vitro翻訳系(真核生物由来、大腸菌由来無細胞翻訳系等)に導入し、目的のタンパク質を発現させる方法によっても産生することができる。ウサギ由来抗体、マウスキメラ抗体、ヒトキメラ抗体、ヒト化抗体又はこれらの抗原結合性断片の製造においては、このようなポリヌクレオチド導入によって抗体を製造することが好ましい。ポリヌクレオチドの導入は、例えば、プラスミド、コスミド、バクテリオファージ等をベクターとして用いて行うことができる。ポリヌクレオチドの導入は、慣用の遺伝子工学的手法を用いて行うことができる。
得られた前記抗体又はその抗原結合性断片は、適宜断片化(パパイン消化、ペプシン消化等)、分離、精製等を行ってもよい。断片化、分離、精製の方法としては、タンパク質、抗体に使用される公知の方法を適宜組み合わせて使用することができる。
さらに、前記抗体又はその抗原結合性断片の製造方法は、前記連結体を調製するために種々の連結物質を連結する工程を含んでもよい。これらの物質は、抗体又はその抗原結合性断片上の任意の部位に連結され得るが、抗原結合性を損なわない観点から、例えば定常部位が好ましい。当該連結においては、適宜適切なリンカーを介在させてもよい。

実施例にも示されるように、前記抗体又はその抗原結合性断片は、光老化細胞表面に結合でき、対象中の生きた光老化細胞に対しても結合性を有する。これにより、前記抗体組成物は、対象に投与すると、含まれる抗体若しくはその抗原結合性断片が、光老化細胞に結合する。結合した抗体又はその結合性断片を介した免疫応答によって、光老化細胞の減少又は増加抑制がもたらされ、ひいては光老化状態を予防又は改善することができる。
実施例にも示されるように、前記抗体又はその抗原結合性断片は、光老化細胞表面に結合でき、対象中の生きた光老化細胞に対しても結合性を有する。これにより、前記抗体組成物は、対象に投与すると、含まれる抗体若しくはその抗原結合性断片が、光老化細胞に結合する。結合した抗体又はその結合性断片を介した免疫応答によって、光老化細胞の減少又は増加抑制がもたらされ、ひいては光老化状態を予防又は改善することができる。
また、前記抗体又はその結合性断片は、それらに連結した連結物質を光老化細胞に局在させ、送達させることもできる。あるいは、例えば、1つ以上の抗体又は抗原結合性断片をドラッグデリバリー(DDS)キャリア(例えば、ベシクル)と会合させることにより、前記薬剤を含むDDSキャリアを光老化細胞に送達させることもできる。これにより、前記抗体組成物は、対象に投与することにより、前記薬剤を光老化細胞に選択的(好ましくは特異的)に送達することができる。そして、前記薬剤の作用を介して光老化細胞の減少又は増加抑制をもたらし、ひいては光老化状態を予防又は改善することができる。
以上によれば、前記抗体組成物は、対象に投与して、対象中の光老化細胞に薬剤を送達するために用いることができる。

以下に、実施例を用いて本発明をさらに詳しく説明する。ただし、これらの例は本発明を制限するものではない。
[試験例1.光老化細胞の作製方法の検証]
ヒト皮膚細胞の一種である正常ヒト表皮ケラチノサイト(NHEK)に対して、以下の手順でピーク波長が308nmの紫外線を照射して光老化細胞を作製した。図1に手順の模式図を示した。培養及びインキュベートはCO2インキュベーターを使用して37℃で行った。紫外線光源には、セラビーム(登録商標)UV308 mini(ウシオ電機製)を使用した。即ち、照射した紫外線は、XeClエキシマ放電ランプを光源として、280nm付近の波長をカットする硼珪酸ガラス(厚さ1mm)を通した光であり、そのスペクトルは、特開2007-267936号公報の図8b及び図9bで表され、297nm未満の領域の成分を有しないUVBである。
(1)コラーゲンコートディッシュ(Corning社製)に、ケラチノサイト専用培地(LONZA社製)で常法によりNHEKをサブコンフルエントになるまで培養した。
(2)培地を除去して、リン酸緩衝生理食塩水(PBS)で2回細胞を洗浄してから、細胞が十分浸るようにPBSを加えた。
(3)0mJ/cm2、20mJ/cm2、40mJ/cm2、80mJ/cm2又は100mJ/cm2の紫外線を照射した。ここで、各照射量は、照射時間を0、1、3、5又は6秒間とすることで得られた。
(4)ディッシュのPBSをケラチノサイト専用培地(LONZA社製)に置換して、3日間培養した。
(5)培地を吸引除去し、SA-β-Gal前処理液(CELL BIOLABS社製)を培地に2mL加えて、2時間インキュベートした。
(6)SA-β-Gal前処理液を吸引除去し、SA-β-Gal基質溶液(CELL BIOLABS社製)を培地に10μL加えて、さらに1日培養した。
ヒト皮膚細胞の一種である正常ヒト表皮ケラチノサイト(NHEK)に対して、以下の手順でピーク波長が308nmの紫外線を照射して光老化細胞を作製した。図1に手順の模式図を示した。培養及びインキュベートはCO2インキュベーターを使用して37℃で行った。紫外線光源には、セラビーム(登録商標)UV308 mini(ウシオ電機製)を使用した。即ち、照射した紫外線は、XeClエキシマ放電ランプを光源として、280nm付近の波長をカットする硼珪酸ガラス(厚さ1mm)を通した光であり、そのスペクトルは、特開2007-267936号公報の図8b及び図9bで表され、297nm未満の領域の成分を有しないUVBである。

(6)SA-β-Gal前処理液を吸引除去し、SA-β-Gal基質溶液(CELL BIOLABS社製)を培地に10μL加えて、さらに1日培養した。
図2は、各照射量で作製した光老化細胞の顕微鏡画像を比較した図である。照射量の増加につれて、細胞の数が少なくなったが、剥離した細胞(死細胞)はあまり観察されなかった。これは、照射量を増加させると、老化細胞の特徴である「細胞分裂の停止」を示す細胞が増加したことを表す。すなわち、照射量が40mJ/cm2以上では、細胞分裂を停止した細胞が顕著に増加し、80mJ/cm2以上で十分に細胞分裂が停止した老化細胞が得られることがわかった。
また、照射量を増加させると、細胞の大きさが大きくなった。これは老化細胞の特徴である「細胞の肥大化」を表す。
なお、これらの細胞分裂の停止又は細胞の肥大化を示した細胞は、いずれもSA-β-Gal染色されていたことから、老化細胞であることが確かめられた。
また、照射量を増加させると、細胞の大きさが大きくなった。これは老化細胞の特徴である「細胞の肥大化」を表す。
なお、これらの細胞分裂の停止又は細胞の肥大化を示した細胞は、いずれもSA-β-Gal染色されていたことから、老化細胞であることが確かめられた。
よって、ピーク波長が308nmの紫外線を照射することにより、生きた光老化細胞を効率よく作製することができることが示された。
[試験例2.光老化ケラチノサイトのmRNA発現解析]
試験例1の(1)~(5)と同じ手順で、0mJ/cm2の照射量(光照射なし)のケラチノサイト(UN群)、80mJ/cm2の照射量で紫外線照射処理したケラチノサイト(U80群)を作製した。また、50日間ケラチノサイト専用培地で継代して、自然老化したヒト正常ケラチノサイト(P50群)を作製した。各群n=10である。各群の細胞をディッシュから回収し、TRIzol(登録商標、Thermo Fisher Scientific社製)を用いて細胞からmRNAを抽出して、RNA-Seq解析を行った。解析された全遺伝子のリードカウント数について正規化して群間比較を行った。RNA-Seqの実施及びデータの解析は、株式会社DNAチップ研究所に委託して行った。
試験例1の(1)~(5)と同じ手順で、0mJ/cm2の照射量(光照射なし)のケラチノサイト(UN群)、80mJ/cm2の照射量で紫外線照射処理したケラチノサイト(U80群)を作製した。また、50日間ケラチノサイト専用培地で継代して、自然老化したヒト正常ケラチノサイト(P50群)を作製した。各群n=10である。各群の細胞をディッシュから回収し、TRIzol(登録商標、Thermo Fisher Scientific社製)を用いて細胞からmRNAを抽出して、RNA-Seq解析を行った。解析された全遺伝子のリードカウント数について正規化して群間比較を行った。RNA-Seqの実施及びデータの解析は、株式会社DNAチップ研究所に委託して行った。
図3にリードカウントの比較の結果を示した。GPR17、CD34、GABRR1、OR2AG2、CMKLR1、CDH19、CD93、AVPR2、CCR7及びOXGR1が、紫外線照射によって作製された光老化細胞で特異的に増加していることが見出された。
以上から、前記の10遺伝子及びそれらタンパク質は、光老化細胞を検出又は定量するためのマーカーとして使用可能であることが示された。
中でも、GPR17、GABRR1、CMKLR1、AVPR2、CCR7及びOXGR1は、U80群以外にはmRNA発現が観察されなかった。これらの遺伝子は光老化細胞に顕著に特異的なマーカーであることが示された。
中でも、GPR17、GABRR1、CMKLR1、AVPR2、CCR7及びOXGR1は、U80群以外にはmRNA発現が観察されなかった。これらの遺伝子は光老化細胞に顕著に特異的なマーカーであることが示された。
[試験例3.抗GPR17モノクローナル抗体の調製]
以下の方法により、抗GPR17モノクローナル抗体を単離した。
(1)ヒトGPR17ロングアイソフォームの1~36位のアミノ酸配列(配列番号5)のC末端にシステイン残基を付加した抗原ペプチド(GPR17LFペプチド)を、ペプチド合成により調製した。ペプチド合成は、株式会社細胞工学研究所に委託した。抗原ペプチドのC末端のシステイン残基に対して、キャリアタンパク質としてキーホールリンペットヘモシアニン(KLH)又はウシ血清アルブミン(BSA)と架橋剤であるMBS(m-マレイミドベンゾイル-N-ヒドロキシスルホスクシンイミドエステル)を反応させ、結合させた。
(2)前記KLH結合抗原ペプチドをウサギ(Slc:JW/CSK、13週齢)1匹に、開始時(0日後)2.5mg投与した。さらに追加免疫抗原として、開始時と同じ抗原ペプチドをアジュバント(TiterMax Gold;フナコシ(株);カタログ番号CYT G-1)と体積比1:1で混合したものを14日後、28日後、42日後にそれぞれ2.5mgずつ投与し、免疫した。46日後に、抗体価が上昇していることを確認するため、ウサギから血液を採取し、血清及びリンパ球を回収した。リンパ球については培養し、培養上清を分離した。得られた血清及び培養上清に対して、GPR17LFペプチドを固相化したプレートを用いて、ELISA法で抗体価を測定した。
(3)抗体価の上昇が確認されたウサギの脾臓由来リンパ球を選抜した。GPR17LFペプチドを固相化Single-cell ELISAプレートに、RPMI培地(10% FBS)で希釈したリンパ球を1ウェルに1細胞のみが含まれるように加え、45分インキュベートした。プレートの抗原と結合した抗GPR17抗体を標識された抗ウサギIgG抗体で検出することによって陽性クローンを特定した。その結果、2×105個のリンパ球をスクリーニングし、32個の陽性クローンを得た。得られた陽性クローンから、PCR法を用いて各細胞のIgG重鎖及び軽鎖遺伝子を増幅した。
(4)得られた各細胞の重鎖及び軽鎖遺伝子に、PCR法を用いてCAGプロモーターを付加した。得られた重鎖及び軽鎖遺伝子を含むポリヌクレオチドを、HEK293細胞に導入し、RPMI培地で3日間培養した。そして、モノクローナル抗体を含む培養上清を回収した。
(5)得られた各クローンの培地上清中の抗GPR17抗体価をELISAにより測定した。プレートにはGPR17LFペプチドでコーティングしたELISAプレートを、二次抗体には抗ウサギIgG抗体を使用した。
以下の方法により、抗GPR17モノクローナル抗体を単離した。

(4)得られた各細胞の重鎖及び軽鎖遺伝子に、PCR法を用いてCAGプロモーターを付加した。得られた重鎖及び軽鎖遺伝子を含むポリヌクレオチドを、HEK293細胞に導入し、RPMI培地で3日間培養した。そして、モノクローナル抗体を含む培養上清を回収した。
(5)得られた各クローンの培地上清中の抗GPR17抗体価をELISAにより測定した。プレートにはGPR17LFペプチドでコーティングしたELISAプレートを、二次抗体には抗ウサギIgG抗体を使用した。
その結果、GPR17LFペプチドに対して結合性を有するモノクローナル抗体が、15クローンも得られた。したがって、GPR17由来のペプチドにより、対象の免疫が誘導されること、及び抗体スクリーニングにより抗GPR17抗体が再現良く得られることが示された。
[試験例4.抗ヒトGPR17モノクローナル抗体を用いた光老化細胞の検出]
(光老化細胞の準備)
細胞培養基材としてコラーゲンコートしたガラスプレートを用いたこと以外は試験例1と同じ方法により、紫外線を照射した細胞を作製した。照射量は0mJ/cm2(UV(-)群)又は80mJ/cm2(UV(+)群)とした。UV照射は5秒間行った。コラーゲンコートしたガラスプレートとして、セルマトリックス Type I-C(新田ゼラチン社製)0.1mLを希釈液(希塩酸pH3.0)0.9mLに加えたものを、Multitest slide, 8-well(MP Biomedical社製)に20μL/well加え、30分室温にて静置後、ケラチノサイト専用培地20μL/wellで2回洗浄して得られたものを使用した。
(光老化細胞の準備)
細胞培養基材としてコラーゲンコートしたガラスプレートを用いたこと以外は試験例1と同じ方法により、紫外線を照射した細胞を作製した。照射量は0mJ/cm2(UV(-)群)又は80mJ/cm2(UV(+)群)とした。UV照射は5秒間行った。コラーゲンコートしたガラスプレートとして、セルマトリックス Type I-C(新田ゼラチン社製)0.1mLを希釈液(希塩酸pH3.0)0.9mLに加えたものを、Multitest slide, 8-well(MP Biomedical社製)に20μL/well加え、30分室温にて静置後、ケラチノサイト専用培地20μL/wellで2回洗浄して得られたものを使用した。
次に、得られた培養細胞を以下の手順で染色及び観察した。
(1)培地を吸引除去し、一次抗体として試験例3で得られた抗ヒトGPR17モノクローナル抗体のうちの1クローンを約1μg/mL含む溶液0.1mLを加え、4℃で1時間インキュベートした。核染色にはHoechstを使用した。
(2)0.2%(w/v)BSA含有リン酸緩衝生理食塩水(PBS)を用いて、細胞を氷上で3回洗浄した。
(3)二次抗体としてAnti-Rabbit IgG Cy3 Conjugate (Jacson ImmunoReserch Laboratry社製)を0.2%(w/v)BSA含有PBSで500倍希釈したものを添加し、4℃で30分インキュベートした。
(4)0.2%(w/v)BSA含有PBSを用いて3回、さらにPBSで1回、細胞を氷上で洗浄した。
(5)SlowFadeTMGold antifade reagent(Thermo Fisher Scientific社製)で得られたケラチノサイトをスライドガラス上に包埋し、蛍光顕微鏡観察した。
(1)培地を吸引除去し、一次抗体として試験例3で得られた抗ヒトGPR17モノクローナル抗体のうちの1クローンを約1μg/mL含む溶液0.1mLを加え、4℃で1時間インキュベートした。核染色にはHoechstを使用した。
(2)0.2%(w/v)BSA含有リン酸緩衝生理食塩水(PBS)を用いて、細胞を氷上で3回洗浄した。
(3)二次抗体としてAnti-Rabbit IgG Cy3 Conjugate (Jacson ImmunoReserch Laboratry社製)を0.2%(w/v)BSA含有PBSで500倍希釈したものを添加し、4℃で30分インキュベートした。
(4)0.2%(w/v)BSA含有PBSを用いて3回、さらにPBSで1回、細胞を氷上で洗浄した。
(5)SlowFadeTMGold antifade reagent(Thermo Fisher Scientific社製)で得られたケラチノサイトをスライドガラス上に包埋し、蛍光顕微鏡観察した。
得られた染色像を図4に示した。UV(+)群の細胞は抗ヒトGPR17モノクローナル抗体で染色されたが、UV(-)群の細胞は染色されなかった。以上から、得られた抗GPR17モノクローナル抗体が光老化細胞に結合性を有することが示された。

- 免疫原性GPR17ポリペプチド、免疫原性CD34ポリペプチド、免疫原性GABRR1ポリペプチド、免疫原性OR2AG2ポリペプチド、免疫原性CMKLR1ポリペプチド、免疫原性CDH19ポリペプチド、免疫原性CD93ポリペプチド、免疫原性AVPR2ポリペプチド、免疫原性CCR7ポリペプチド及び免疫原性OXGR1ポリペプチドからなる群より選ばれる少なくとも1種のポリペプチドを含有し、
該少なくとも1種のポリペプチドが、キャリアタンパク質に結合された、又はキャリアタンパク質に結合されていない、対象中の光老化細胞に対する免疫を誘導するための、抗原組成物。 - 免疫原性GPR17ポリペプチド、免疫原性CD34ポリペプチド、免疫原性GABRR1ポリペプチド、免疫原性OR2AG2ポリペプチド、免疫原性CMKLR1ポリペプチド、免疫原性CDH19ポリペプチド、免疫原性CD93ポリペプチド、免疫原性AVPR2ポリペプチド、免疫原性CCR7ポリペプチド及び免疫原性OXGR1ポリペプチドからなる群より選ばれる少なくとも1種のポリペプチドを含有し、
