Abstract
従来研究されてきた土耕栽培や水耕栽培における施肥などの園芸管理技術、培土の選択、生育環境における気温、園芸照明の強度と質、光周期、植物の生殖成長期間における干ばつ処理の工夫では花穂乾燥重量や花穂中の二次代謝産物含量の増大は限定されていた。 植物の根の少なくとも一部が親水性フィルム表面に密着し、該植物の根が密着する該親水性フィルムの反対面に養液が接するように養液が配置されるようなシステムを用い、該植物の栄養成長期間と生殖成長期間の切り替えを日長時間の変更によって行う。 本発明の栽培システム及び栽培方法は、二次代謝産物を集積する花穂の成長を促進して栽培することができるので、農業や医薬品製造分野で利用できる。
Description
本発明は、光周性を持つ短日植物または長日植物の栽培に適した植物栽培システムに関する。より詳しくは医療用途などに有用なカンナビノイドを産生する大麻草などの栽培に適した栽培システムに関する。
光周性に依存する植物の現象としては花芽形成、茎の伸長、休眠、落葉などが知られているが、とりわけ花芽形成についてよく研究されている。植物には日長が短くなると花芽形成をし、花成に至る短日植物、日長が長くなると花成する長日植物、日長とは無関係に花成する中性植物などがある。
菊などは日長がある特定の時間よりも短くなった場合にだけ花芽を形成(短日植物)し、キンギョソウ、ストックなどは日長がある特定の時間よりも長くなった場合にだけ花芽を形成(長日植物)する。短日植物の花芽形成(すなわち生殖成長)を抑えて栄養成長を続けさせるために、あるいは長日植物の開花を促進させるために、夜中に数時間電灯照明をして植物を栽培する電照栽培が行われている。また、短日植物の開花を促進するために、あるいは長日植物の花芽形成を抑えて栄養成長を続けさせるために、太陽光を数時間遮断して植物を栽培する遮光栽培が行われている。
大麻草(カンナビス・サティバ・エル)の花穂のエッセンシャルオイルは、長年にわたり種々の疾患及び障害の治療に有用な医薬品として使用されてきた。一方、その精神活性や依存性が問題視され、大麻草の栽培が禁止されてきた歴史がある。そのため、栽培者は逸話的な情報に基づいて栽培せざるを得なかった。過去10年間で大麻草の栽培と使用、特に薬用目的に関する規制は、北米とヨーロッパの一部で自由化され、この分野の研究が進展してきた。
大麻草雌花の花穂には、フィトカンナビノイドとして知られる多様なメロテルペノイド化合物が二次代謝産物として集積される。テトラヒドロカンナビノール (THC) およびカンナビジオール (CBD) を含むいくつかのカンナビノイドは、その精神活性および薬効について広く研究されている (非特許文献1)。生きた植物では、カンナビノイドは主にテトラヒドロカンナビノール酸 (THCA) およびカンナビジオール酸 (CBDA)などのカルボン酸として存在する (非特許文献2) 。
大麻草の花穂乾燥重量や花穂中のカンナビノイド含量の増大は、これまで主に育種や化学表現型の選択に依存している。大麻草の花穂乾燥重量や花穂中のカンナビノイド含量の増大を目的として、施肥などの園芸管理技術 (非特許文献3) 、培土の選択 (非特許文献4) 、生育環境における気温 (非特許文献5) 、園芸照明の強度と質 (非特許文献6) 、光周期 (非特許文献7) 、植物の生殖成長期間における干ばつ処理 (非特許文献8) などの工夫が試みられている。
従来より、本発明者らは、親水性フィルムを使用した養液栽培について研究を重ねており、以下の植物栽培システムや植物栽培方法について開示している:養液と接触する無孔性親水性フィルム上で、該フィルムと植物の根を一体化させて植物を栽培する植物栽培用器具および植物栽培方法(特許文献1)、上記フィルム上部にも灌水する植物栽培用器具および植物栽培方法(特許文献2)、上記フィルムが養液上を連続的に移動する植物栽培システム(特許文献3)、上記フィルムとその上部に配置される蒸発抑制部材の間に空気層を設ける植物栽培システム(特許文献4)、上記フィルムの下面側に養液を連続的に供給する手段を用いる植物栽培システム(特許文献5)、上記フィルムとして特定のポリビニルアルコール(PVA)系を用いる植物栽培システム(特許文献6)。

しかしながら、上記の特許文献1~6に記載の植物栽培システムを用いて光周性を持つ短日植物または長日植物の栽培をしても、花穂乾燥重量や花穂中の二次代謝産物含量を増大させることは困難であると考えられていた。理由は以下の通りである。まず、上記のいずれかのシステムによる植物栽培では、生殖成長期間だけでなく栄養成長期間においても植物に水分ストレスを与えるため、植物の栄養成長を阻害すると考えられる。さらに、光周性を持つ短日植物または長日植物の栽培においては、生殖成長期間または栄養成長期間において日長時間が短くなるため、光合成が制限され、当該短日植物または長日植物の成長が抑制されると考えられる。このため、花穂乾燥重量や花穂中の二次代謝産物含量の増大を目的とした短日植物または長日植物の栽培を、上記のいずれかのシステムを用いて行なっても、目的は達成されない、と考えられていた。
また、非特許文献1~8に記載されるような従来研究されてきた土耕栽培や水耕栽培における施肥などの園芸管理技術、培土の選択、生育環境における気温、園芸照明の強度と質、光周期、生殖成長期間における干ばつ処理の工夫では大麻草などの花穂乾燥重量や花穂中の二次代謝産物含量の増大は限定されていた。
そこで、本発明者らは、かかる現況に鑑みて鋭意研究を重ねた結果、意外にも、親水性フィルムの上で大麻草などの光周性を持つ短日植物または長日植物を栽培し、かつ日長時間の調節によって栄養成長期間と生殖成長期間を切り替えて栽培することにより、大麻草などの花穂乾燥重量や花穂中のカンナビノイド含量の増大を達成できることを見出し、本発明を完成した。
すなわち、本発明は、以下の諸態様を含む。
1.親水性フィルムおよび養液保持手段を構成要素とする植物栽培システムであって、使用時には、植物の根の少なくとも一部が親水性フィルム表面に密着し、該植物の根が密着する該親水性フィルムの反対面に養液が接するように養液が配置され、該植物の栄養成長期間と生殖成長期間の切り替えを日長時間の変更によって行うことを特徴とする植物栽培システム。
2.前記親水性フィルムが、該フィルムを介して水と塩水とを対向して接触させた際に、測定開始後4日目(96時間)の水/塩水の電気伝導度(EC)の差が4.5dS/m以下のフィルムであることを特徴とする前項1に記載の植物栽培システム。
3.該フィルムの水中平衡膨潤度が130%以上300%未満かつ乾燥時の厚みが20μm以上150μm未満であることを特徴とする前項1または2に記載の植物栽培システム。
4.養液保持手段が水耕栽培用水槽であり、該親水性フィルムの下面に接触するように配置された養液が水耕栽培用水槽に収容されてなることを特徴とする、前項1~3のいずれかに記載の植物栽培システム。
5.該養液保持手段が水不透過性表面を有し、その上に該親水性フィルムが敷設されてなり、親水性フィルムと養液保持手段との間に該養液を連続的または間歇的に供給する養液供給手段をさらに含むことを特徴とする、前項1~3のいずれかに記載の植物栽培システム。
6.遮光手段および照明手段からなる群より選ばれる少なくとも1種をさらに含むことを特徴とする、前項1~5のいずれかに記載の植物栽培システム。
7.短日植物または長日植物の栽培に用いるための前項1~6のいずれかに記載の植物栽培システム。
8.(1)植物をその上で栽培するための親水性フィルム、および該親水性フィルムの植物の反対面に接触するように配置された養液保持手段を含む植物栽培システムを提供し、
(2)該システム内の親水性フィルムの上に短日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する短日植物の栽培方法。
(2)該システム内の親水性フィルムの上に短日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する短日植物の栽培方法。
9.(1)植物をその上で栽培するための親水性フィルム、および該親水性フィルムの植物の反対面に接触するように配置された養液保持手段を含む植物栽培システムを提供し、
(2)該システム内の親水性フィルムの上に長日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は毎日13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する長日植物の栽培方法。

(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は毎日13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する長日植物の栽培方法。
本発明の植物栽培システムの使用時においては、植物の根の少なくとも一部が親水性フィルム表面に密着し、該植物の根が密着する該親水性フィルムの反対面に養液が接するように養液が配置される。親水性フィルムの形状やサイズには特に限定はなく、使用時における親水性フィルム、養液保持手段、植物、養液の位置関係にも特に限定はないが、通常の態様としては、平坦な親水性フィルムを水平に配置し、親水性フィルムの上面に植物を配置するので、以下の説明においては、この通常の態様の場合を想定して「フィルムの上(上面)」、「フィルムの下(下面)」などという表現をする。もちろん、親水性フィルムをチューブ状あるいは袋状として植物栽培システムを構成することも出来、その場合は親水性フィルムの上面は外面、下面は内面と言い換えることができる。さらに、親水性フィルムを壁面に垂直に配置して植物を栽培することもできる。当業者は、通常の態様でない場合であっても、使用時における親水性フィルム、養液保持手段、植物、養液の位置関係を適宜設定することができる。
本発明によれば、光周性を持つ短日植物または長日植物の栽培期間中に、日長時間の変更を行うことにより植物の成長モードを栄養成長期間から生殖成長期間に切り替えることができ、花芽が形成され、植物の二次代謝産物が集中する花穂の形成を促進する。さらに、本発明によれば、植物が親水性フィルムを介して養液を吸収するため、適度な水分ストレスを植物に付加することができ、植物がストレスに対抗するために産生する二次代謝産物の生成を促進する。その相乗効果の結果、本発明によれば、植物の花穂乾燥重量や花穂中の二次代謝産物含量の増大が達成される。
本発明によれば、植物が親水性フィルムを介して養液を吸収することにより、水分ストレスが植物に付加されることは、相対的に葉や茎を成長させる栄養成長を抑え、子孫を残そうとする生殖成長を促進する。その結果、花などの生殖器官が相対的に葉、茎よりも成長し易くなる。葉が大きく茂ることは、光合成量を増大させるものの、日陰を作ってしまうので植物の定植密度を下げる必要がある。本発明によれば、上記水分ストレスで相対的に葉の面積を小さくすることにより、植物の定植密度を高めることができる。本発明では日長時間の変更を行うことにより花芽形成時期を任意にコントロールすることとの相乗効果により、想定以上の単位面積当たり植物の花穂乾燥重量や花穂中の二次代謝産物含量の増大が達成される。
また本発明に用いられる親水性フィルムは雑菌やウイルスを通過させず、養液は該親水性フィルムを介して植物の根と接するため、植物の病気を誘発する雑菌などからの感染を回避して、また根ぐされなどの原因となる根の酸素欠乏状態を招くことなく、効率的且つ安定的に、長期間に亘り十分な量の養分を植物の根から吸収させることができ、それにより長期間に亘り持続的に植物の生長を著しく促進させることが可能となる。
以下、本発明について具体的に説明する。
<植物栽培システム>
本発明の植物栽培システムの構成要素として、本発明の親水性フィルムおよび養液保持手段は必須であるが、養液保持手段の違いによって、大きく2種に分けることができる。第1のタイプは、養液保持手段が水耕栽培用水槽であり、本発明の親水性フィルムの下面に接触するように配置された養液が水耕栽培用水槽に収容されてなることを特徴とする、植物栽培システムである。このようなシステムについては、たとえば特許文献1を参照することができる。
<植物栽培システム>
本発明の植物栽培システムの構成要素として、本発明の親水性フィルムおよび養液保持手段は必須であるが、養液保持手段の違いによって、大きく2種に分けることができる。第1のタイプは、養液保持手段が水耕栽培用水槽であり、本発明の親水性フィルムの下面に接触するように配置された養液が水耕栽培用水槽に収容されてなることを特徴とする、植物栽培システムである。このようなシステムについては、たとえば特許文献1を参照することができる。
第2のタイプは、養液保持手段が水不透過性表面を有し、その上に本発明の親水性フィルムが敷設されてなり、本発明の親水性フィルムと養液保持手段との間に養液を連続的または間歇的に供給する養液供給手段をさらに含むことを特徴とする植物栽培用システムであり、養液供給手段の代表的なものが本発明の親水性フィルムと養液保持手段との間に設置された点滴灌水チューブである。即ち、この第2のタイプの栽培システムは、養液保持手段を基材層とし、その上に直接的または間接的に本発明の親水性フィルムが積層されてなる多層構造を有するシステムである。このようなシステムについては、たとえば特許文献5を参照することができる。
図1は、第1のタイプの植物栽培システムの基本的な一態様を示す模式断面図である。図1の例においては、本発明の親水性フィルム(1)の下に水槽(2)が設置され、水槽内に肥料成分を含む養液(3)が収容される。該養液(3)は、本発明の親水性フィルム(1)に吸収され、植物(4)の根(5)は、本発明の親水性フィルム(1)の上面に密着し、本発明の親水性フィルム(1)に含まれる水、肥料成分を吸収する。植物体の上部には光源(8)を設置する。また光源を任意の時間にON-OFFするためのタイマーコントローラー(9)を光源と電源の間に介在させる。
必要に応じて、親水性フィルム(1)の上に土壌などの植物栽培用支持体(6)、および/または、水蒸気を通さないか、または低透過性の蒸発抑制部材(例えば、後述するマルチング材)あるいは定植板(7)を配置することができる。親水性フィルム(1)の上に植物栽培用支持体(6)を配置すると、植物体の根を保護する効果が得られる。また、蒸発抑制部材あるいは定植板(7)を配置することにより親水性フィルム(1)から大気中に蒸散する水蒸気を蒸発抑制部材表面あるいは植物栽培用支持体(6)中に凝結させると、凝結した水蒸気を水として植物が利用できる。
本発明の植物栽培システムによれば、肥料成分を含む養液(3)は本発明の親水性フィルム(1)を介して植物に供給される。これに対して、植物の根が水(または養液)に浸かっている従来の水耕栽培方法においては、水や養液の表面が空気層と接しているため、空気中の細菌や菌類が容易に混入してしまい、細菌や菌類が植物の根に繁殖して著しい生育障害や植物の病気を誘発する。
また、植物の根が水(または養液)に浸かっている従来の水耕栽培方法においては、根は水に溶存した酸素を吸収するため、栽培に使用する水の溶存酸素量を一定以上に保つ必要があった。これに対して、本発明の植物栽培システムにおいては、植物の根は本発明の親水性フィルム(1)の上の空気層にあるので、酸素は空気中から十分に、自由に吸収することができる。
さらに、必要に応じて、フィルム(1)の上部に細霧噴霧用手段(例えば、バルブ)を配置し、間歇的に水、養液または農薬希釈液を噴霧することができる。このような細霧噴霧用手段を配置することにより、水の間歇的噴霧による特に夏季の冷却と、養液の噴霧による環境の冷却と葉面散布による肥料成分の供給、農薬の配合された水または養液の噴霧による農薬の散布などの自動化が可能となるというメリットを得ることができる。
親水性フィルム(1)は、植物の根の少なくとも一部が親水性フィルム表面に密着するという性質を有する。このため、使用時には、供給される水や肥料の分子が親水性フィルム(1)を介して拡散して植物の根に移行することが可能であると考えられる。後述するように、この密着が強いときには、植物の根と親水性フィルム(1)が実質的に一体化する。
本発明の植物栽培システムの好ましい態様によれば、本発明の親水性フィルム(1)上で栽培される植物の根が本発明の親水性フィルム(1)を介して養液を吸収しようとして、植物の根と本発明の親水性フィルム(1)が実質的に一体化する。フィルムと根の「一体化」を促進させるためには、該フィルム(1)の下からは養液を供給することが好ましい。
本発明の親水性フィルム(1)の下面から水のみを供給した場合と比較して、養液を使用した方が、植物の生育のみならず、根とフィルムの接着強度が著しく向上する。これは、植物がフィルムを介して、水のみならず肥料成分をも吸収していることを示している。さらに、フィルムを介して水および肥料成分を効率良く吸収するためには、根がフィルム表面に強く密着することが必須であり、その結果として根とフィルムが一体化することになるものと考えられる。
本発明の親水性フィルム(1)と根の「一体化」が完成する前に、該フィルム上から水分を加え過ぎると、植物はフィルム上の取り易い水分を吸収して、該フィルム下からの水分を取る必要が減じ、その結果、根が該フィルムと一体化し難くなる傾向がある。従って、根が該フィルムと一体化するまでは、該フィルム上からは、過剰の水分を加えることは好ましくない。他方、根が本発明の親水性フィルム(1)と一体化した後であれば、適宜、該フィルム上から水分/養分を与えても良い。
<植物栽培用システムの構成>
以下、本発明の植物栽培システムにおける各部の構成について詳細に説明する。このような構成(ないしは機能)に関しては、必要に応じて、本発明者らによる文献(特許文献1~6)の「発明の詳細な説明」、「実施例」等を参照することができる。

(本発明の親水性フィルム)
本発明の植物栽培システムにおいては、植物をその上で栽培するための本発明の親水性フィルムが必須である。本発明において、使用可能なフィルム材料は、特に制限されず、公知の材料から適宜選択して使用することが可能である。このような材料は、通常フィルムないし膜の形態で用いることができる。

より具体的には、このようなフィルム材料としては、例えば、ポリビニルアルコール(PVA)、セロファン、酢酸セルロース、硝酸セルロース、エチルセルロース、ポリエステル等の親水性材料が使用可能である。
上記フィルムの厚さ(厚さは、乾燥時の厚さを意味する。以下同じ。)も特に制限されないが、通常は、300μm以下程度、更には200~5μm程度、特に100~20μm程度であることが好ましい。
フィルムの厚さが好ましい範囲を超えると、養液の肥料成分の透過が遅くなり植物の成長が阻害される。一方フィルムの厚さが好ましい範囲を下回る場合は、フィルムの強度が不足し、植物の根がフィルムを貫通し易くなるので好ましくない。
本発明で使用される親水性フィルムは、水中で水を吸収して膨潤する性質を有することが好ましい。本発明で用いる親水性フィルムの水中(30℃)における平衡膨潤度は、130%以上300%以下の範囲であることが好ましく、150%以上250%以下であることがより好ましい。
親水性フィルムの該平衡膨潤度がこの範囲の下限を下回ると、水と肥料成分の透過性が不十分となり、植物の成長速度が遅くなる。一方、親水性フィルムの該平衡膨潤度がこの範囲の上限を上回ると、水中における親水性フィルムの強度が低下し、植物の根の貫通に耐え難くなる。
ここで、親水性フィルムの水中(30℃)における平衡膨潤度は以下のように測定する。まず、乾燥状態の親水性フィルムを20cmx20cmの正方形に切り出し、その重量(a)gを測定する。次いで、切り出した親水性フィルムを30℃の水に浸漬し、30分間静置する。該フィルムを水中から取り出し、フィルム表面に付着した余剰の水分を素早く拭き取り、該フィルムの重量(b)gを測定する。平衡膨潤度はb/a x 100%で算出する。
親水性フィルムは、無孔性であってもよいし、無孔性でなくてもよいが、無孔性であることが好ましい。
(一体化試験)
本発明の親水性フィルムは、栽培している「植物体の根と実質的に一体化し得る」フィルムであることが好ましい。「植物体の根と実質的に一体化し得る」フィルムとは、本発明の植物栽培用システムの本発明の親水性フィルムの上で植物を35日間栽培した際に、本発明の親水性フィルムを栽培した植物の根から剥離するための剥離強度が10g以上となるフィルムである。根とフィルムの一体化を測定するための「一体化試験」は、次のようにして実施することができる。
本発明の親水性フィルムは、栽培している「植物体の根と実質的に一体化し得る」フィルムであることが好ましい。「植物体の根と実質的に一体化し得る」フィルムとは、本発明の植物栽培用システムの本発明の親水性フィルムの上で植物を35日間栽培した際に、本発明の親水性フィルムを栽培した植物の根から剥離するための剥離強度が10g以上となるフィルムである。根とフィルムの一体化を測定するための「一体化試験」は、次のようにして実施することができる。
「ざるボウルセット」を使い、ざる上に試験すべきフィルム(200×200mm)を乗せ、フィルムの上にバーミキュライト150g(水分73%、乾燥重量40g)を載せ、サニーレタスの幼苗(本葉1枚強)を2本植え付ける。このざるを、240~300gの養液が張られたボウル中に設置し、該フィルムを該養液と接触させ、幼苗を栽培する。栽培はハウス内で行い、自然光(太陽光)を使用し、気温は0~25℃、湿度は50~90%RHの条件下で35日間行う。次に、栽培した植物の根元で茎葉を切断し、根の密着したフィルムの茎がほぼ中心になるように、該フィルムを巾5cm(長さ:約20cm)に切断して試験片とする。
ばね式手秤に市販のクリップを付け、上記で得た試験片の一方をクリップで固定して、ばね式手秤の示す重量(試験片の自重に対応=Aグラム)を記録する。次いで試験片の中心にある茎を手で持ち、下方に緩やかに引き下げて、根とフィルムが離れる(または切断される)際の重量(荷重=Bグラム)をばね式手秤の目盛りから読み取る。この値から初期の重量を差し引いた(B-A)グラムを巾5cmの引き剥がし荷重とし、この引き剥がし荷重を剥離強度とする。
本発明において使用する本発明の親水性フィルムの剥離強度は、10g以上であることが好ましく、30g以上であることがさらに好ましく、100g以上であることが最も好ましい。
(イオン透過性試験)
さらに本発明においては、本発明の親水性フィルムが「植物体の根と実質的に一体化し得る」か否かを判断するための指標の1つとして、イオン透過性のバランスが挙げられる。

本発明の植物栽培システムを使用して植物を栽培すると、植物はフィルムを通して肥料をイオンとして吸収する。従って、使用するフィルムの塩類(イオン)透過性が、植物に与えられる肥料成分の量に影響する。本発明においては、本発明の親水性フィルムを介して水と0.5質量%塩水とを対向して4日間(96時間)接触させた際に、水と塩水の栽培温度において測定した電気伝導度(EC)の差が4.5dS/m以下となるフィルムが好ましい。
水と塩水の電気伝導度の差は、3.5dS/m以下であることがさらに好ましく、2.0dS/m以下であることが最も好ましい。このようなフィルムを用いた際には、根に対する好適な水あるいは肥料溶液を供給し、該フィルムと根との一体化を促進することが容易となる。

電気伝導度(EC、イーシー)は、液中に溶けている塩類(あるいはイオン)量の指標であり、比導電率とも言う。ECとしては、断面積1cm2の電極2枚を1cmの距離に離したときの電気伝導度の値を使用し、単位はシーメンス(S)であり、S/cmで表す。しかし、養液のECは小さいので、1/1000の単位となるmS/cmを使う(国際単位系ではdS/m(dはデシ)と表示する)。
フィルムのイオン透過性は、以下のようにして測定することができる。市販の食塩10gを水2000mlに溶解して、0.5%塩水を作製する(EC:約9dS/m)。「ざるボウルセット」を使い、ざる上に試験すべきフィルム(サイズ:200~260×200~260mm)を乗せ、該フィルム上に水150gを加える。他方、ボウル側に上記の塩水150gを加え、得られた系全体を食品用ラップ(ポリ塩化ビニリデンフィルム、商品名:サランラップ(登録商標)、旭化成社製)で包んで、水分の蒸発を防ぐ。この状態で、常温で放置して、24時間毎に水側、塩水側のECを測定する。具体的には、電気伝導度計の測定部位(センサー部)にスポイトを用いて試料(即ち、水側または塩水側の溶液)を少量乗せ、導電率を測定する。
(水分透過性/グルコース溶液透過性試験)
本発明においては、本発明の親水性フィルムを介した植物の根の養分(有機物)吸収を容易とする点から、本発明の親水性フィルムは、所定のグルコース透過性を示すことが好ましい。このグルコース透過性の優れたフィルムは、本発明の親水性フィルムを介して水と5%グルコース水溶液とを対向して3日間(72時間)接触させた際に、水とグルコース溶液の栽培温度において測定した濃度(Brix%)の差が4以下、さらに好ましくは3以下、より好ましくは2以下、最も好ましくは1.5以下となるフィルムである。
本発明においては、本発明の親水性フィルムを介した植物の根の養分(有機物)吸収を容易とする点から、本発明の親水性フィルムは、所定のグルコース透過性を示すことが好ましい。このグルコース透過性の優れたフィルムは、本発明の親水性フィルムを介して水と5%グルコース水溶液とを対向して3日間(72時間)接触させた際に、水とグルコース溶液の栽培温度において測定した濃度(Brix%)の差が4以下、さらに好ましくは3以下、より好ましくは2以下、最も好ましくは1.5以下となるフィルムである。
フィルムのグルコース透過性は、以下のようにして測定することができる。
市販のグルコース(ブドウ糖)を用いて5%グルコース溶液を作製する。上記イオン透過性試験と同様の「ざるボウルセット」を使い、ざる上に試験すべき本発明の親水性フィルム(サイズ:200~260×200~260mm)を乗せ、該フィルム上に水150gを加える。他方、ボウル側に上記のグルコース溶液150gを加え、得られた系全体を食品用ラップ(ポリ塩化ビニリデンフィルム、商品名:サランラップ(登録商標)、旭化成社製)で包んで、水分の蒸発を防ぐ。この状態で、常温で放置して、24hrs毎に水側、グルコース溶液側の糖度(Brix%)を糖度計で測定する。
市販のグルコース(ブドウ糖)を用いて5%グルコース溶液を作製する。上記イオン透過性試験と同様の「ざるボウルセット」を使い、ざる上に試験すべき本発明の親水性フィルム(サイズ:200~260×200~260mm)を乗せ、該フィルム上に水150gを加える。他方、ボウル側に上記のグルコース溶液150gを加え、得られた系全体を食品用ラップ(ポリ塩化ビニリデンフィルム、商品名:サランラップ(登録商標)、旭化成社製)で包んで、水分の蒸発を防ぐ。この状態で、常温で放置して、24hrs毎に水側、グルコース溶液側の糖度(Brix%)を糖度計で測定する。
(耐水圧)
さらに本発明においては、本発明の親水性フィルムが耐水圧として10cm以上の水不透性を有することが好ましい。このような本発明の親水性フィルムを用いた際には、根とフィルムの一体化を促進することができる。また、根に対する好適な酸素供給および本発明の親水性フィルムを介しての病原菌汚染を防止することが容易となる。
さらに本発明においては、本発明の親水性フィルムが耐水圧として10cm以上の水不透性を有することが好ましい。このような本発明の親水性フィルムを用いた際には、根とフィルムの一体化を促進することができる。また、根に対する好適な酸素供給および本発明の親水性フィルムを介しての病原菌汚染を防止することが容易となる。
耐水圧はJIS L1092(B法)に準じた方法によって測定することができる。本発明で使用する本発明の親水性フィルムの耐水圧は10cm以上であることが好ましく、より好ましくは20cm以上、さらに好ましくは30cm以上であり、特に好ましくは200cm以上である。
<植物栽培用支持体>
本発明の植物栽培システムにおいては、植物体の根を保護するために、本発明の親水性フィルムの上に土壌などの植物栽培用支持体を配置することができる。使用する植物栽培用支持体に特に限定はなく、通常使用される土壌ないし培地が使用可能である。このような土壌ないし培地としては、例えば、土耕栽培に用いられる土壌、および水耕栽培に用いられる培地が挙げられる。
本発明の植物栽培システムにおいては、植物体の根を保護するために、本発明の親水性フィルムの上に土壌などの植物栽培用支持体を配置することができる。使用する植物栽培用支持体に特に限定はなく、通常使用される土壌ないし培地が使用可能である。このような土壌ないし培地としては、例えば、土耕栽培に用いられる土壌、および水耕栽培に用いられる培地が挙げられる。
無機系の植物栽培用支持体としては、天然の砂、れき、パミスサンドなど、加工品(高温焼成等)では、ロックウール、バーミキュライト、パーライト、セラミック、籾殻くん炭などが挙げられ、有機系の植物栽培用支持体としては、天然のピートモス、ココヤシ繊維、樹皮培地、籾殻、ニータン、ソータンなどや、合成した粒状フェノール樹脂などが挙げられ、これらは単独でも、複数種を適宜混合して使用することもできる。また、合成繊維の布あるいは不織布も植物栽培用支持体として使用可能である。
必要最小限の肥料および微量要素を上記植物栽培用支持体に加えてもよい。本発明者らの知見によれば、植物の根が、親水性フィルムを介して接触する水/養液から水または養分を吸収可能な程度に伸びるまで、言い換えると根と本発明の親水性フィルムが一体化するまでは、ここで言う「必要最小限の肥料および微量要素」として、養分を本発明の親水性フィルム上の植物栽培用支持体に加えておくことが望ましい。
<養液保持手段>
本発明の植物栽培システムは、養液を本発明の親水性フィルムの下に保持するための養液保持手段を含んでいる。本発明の植物栽培システムにおいては、養液を収容する容器状の養液保持手段、あるいは水不透過性表面を有する基材層として機能する養液保持層の何れかが使用可能である。

養液を収容する容器状の養液保持手段としては、必要な量の養液を保持することのできる容器である限り特に限定はなく、その材質としては、軽量化、易成形性および低コスト化の点からはポリスチレン、ポリプロピレン、ポリ塩化ビニル、ポリエチレン、ポリアクリレート等の汎用プラスチックが好適に使用可能である。例えば、従来使用されてきた水耕栽培用水槽を使用することができる。
養液保持層の水不透過性表面は水を通さない材質からなるものであれば特に限定はなく、合成樹脂、木材、金属あるいはセラミックなどが挙げられる。その養液保持層の形状にも特に限定はなく、フィルム状、シート状、板状、または箱状などが挙げられる。
養液供給手段は、従来から水あるいは養液の連続的あるいは間歇的な供給に使用されている手段であれば特に限定はない。本発明においては、養液を少量ずつ供給することが可能な点滴灌水チューブ(「ドリップチューブ」とも称される)の使用が好ましい。点滴灌水チューブを使用した点滴潅水によって、作物の生育に必要な水および肥料をできるだけ少量供給することができる。
さらに、養液保持層と養液供給手段とを含む態様においては、本発明の親水性フィルムへの養液の供給を補助するために、さらに吸水性材料を本発明の親水性フィルムと水不透過性表面との間に設置することができる。吸水性材料は、基本的には水を吸収して保持する材料であれば特に限定はない。一例としては、合成樹脂から作られたスポンジや不織布、織物からなる布、植物性の繊維状、チップ状、粉末状、または、ピートモスや水苔をはじめ一般的に植物支持体として使用される材料も使用可能である。
<遮光手段および照明手段>
本発明の植物栽培システムは、遮光手段および照明手段からなる群より選ばれる少なくとも1種を含んでいてもよい。
遮光手段は、植物に照射される光を遮断することができるものである限り限定はない。遮光手段として遮光カーテンなどを用いることができる。また、暗室自体を遮光手段として用いてもよい。
照明手段にも特に限定はない。照明手段として、太陽光または人工光を光源として用いる照明手段を用いることができる。照明手段は、同時に複数のものを用いてもよい。
本発明の植物栽培システムは、遮光手段および照明手段からなる群より選ばれる少なくとも1種を含んでいてもよい。
遮光手段は、植物に照射される光を遮断することができるものである限り限定はない。遮光手段として遮光カーテンなどを用いることができる。また、暗室自体を遮光手段として用いてもよい。
照明手段にも特に限定はない。照明手段として、太陽光または人工光を光源として用いる照明手段を用いることができる。照明手段は、同時に複数のものを用いてもよい。
本発明の植物栽培システムは、光周性を持つ短日植物または長日植物の栽培に用いることができる。後述するように、当該植物への光照射は、人工光もしくは太陽光またはそれらの併用による光照射である。
<光源>
本発明の栽培システムにおいて、光周性を持つ短日植物のまたは長日植物の栄養成長期間に用いるための光源の種類に制限は無く、人工光と太陽光のいずれも使用可能であり、人工光と太陽光とを併用してもよい。人工光としては、白熱灯、蛍光灯、ナトリウムランプ、ハロゲンランプ、LED、有機ELなどの光源を利用できる。太陽光を用いる場合、太陽光を光ファイバーケーブルなどにより取り込んだものを植物に照射してもよい。
本発明の栽培システムにおいて、光周性を持つ短日植物のまたは長日植物の栄養成長期間に用いるための光源の種類に制限は無く、人工光と太陽光のいずれも使用可能であり、人工光と太陽光とを併用してもよい。人工光としては、白熱灯、蛍光灯、ナトリウムランプ、ハロゲンランプ、LED、有機ELなどの光源を利用できる。太陽光を用いる場合、太陽光を光ファイバーケーブルなどにより取り込んだものを植物に照射してもよい。
本発明の栽培システムにおいて、光周性を持つ短日植物または長日植物の生殖成長期間に用いるための光源は、上記の栄養成長期間に用いるための光源(人工光もしくは太陽光またはそれらの併用)を用いても良いし、栄養成長期間に用いるための光源とは別に設置しても良い。栄養成長期間に用いるための光源とは別に、光周性を持つ短日植物または長日植物の生殖成長期間に用いるための光源を設置する場合は、人工光が好ましく、白熱灯、蛍光灯、ナトリウムランプ、ハロゲンランプ、LED、有機ELなどの光源を利用できる。
本発明の栽培システムにおいては、光周性を持つ短日植物または長日植物の栄養成長と生殖成長の切り替えを日長時間の変更によって行う。本発明において「日長時間の変更」とは、「光源からの光照射時間サイクルの変更」とも言い、広い意味で用いるものであって、1日における光照射、明時間、暗時間(特に、連続した暗時間)の長さを変更することを意味する。「暗時間」とは、後述の光量が4μmol/m2/sec未満である時間を意味する。「明時間」とは「暗時間」ではない時間を意味する。「明時間」を「明期」とも言い、「暗時間」を「暗期」とも言う。「明時間」と「光照射時間」とは同じ意味を有する。
後述するように、短日植物の場合は、連続した暗時間の長さが限界暗期より長くなるような光周条件下で、花芽の形成が起こるか、あるいはそれが促進される。そこで、短日植物の場合は、連続した暗時間の長さが限界暗期より長くなった時点以降を生殖成長期間とし、それより前を栄養成長期間とする。また、後述するように、長日植物の場合は、連続した暗期の長さが限界暗期より短くなるような光周条件下で、花芽の形成が起こるか、あるいはそれが促進される。そこで、長日植物の場合は、連続した暗時間の長さが限界暗期より短くなった時点以降を生殖成長期間とし、それより前を栄養成長期間とする。
短日植物や長日植物の栽培を、太陽光を用いて実施しようとする場合、必要に応じて、遮光手段を用いて、または人工光を併用して、光照射時間の長さを調節することにより、任意の季節に短日植物や長日植物の栽培をすることができるようになる。たとえば、短日植物の場合には、必要に応じて遮光手段を用いることによって連続した光照射時間を長時間から短時間に変更することにより(これによって、連続した暗時間が短時間から長時間に変更される)、花成が促進される。他方、長日植物の場合には、必要に応じて人工光を併用することによって連続した光照射時間を短時間から長時間に変更することにより(これによって、連続した暗時間が長時間から短時間に変更される)、花成が促進される。
光照射時間サイクルは1日(24時間)を単位とするが、この1日は午前0時から午後12時までの24時間である必要はなく、任意の時刻から開始される24時間でよい。
本発明の栽培システムにおいて、植物に光合成を十分行わせるという観点からは、栄養成長期間も生殖成長期間も、光照射時間は出来るだけ長くすることが望ましい。しかしながら、光周性を持つ短日植物または長日植物の場合、その光周性に適合させることが優先される。
短日植物の場合、連続した暗期の長さが、ある一定の時間(限界暗期)より長くなるような光周条件下で、花芽の形成が起こるか、あるいはそれが促進される。明期より暗期の時間が長いというだけでは効果がなく、一定以上の連続した暗期が必要である。従って、暗期が十分長くても、途中で光を照射して中断してしまうと無効になる。
短日植物の花成が促進される連続した暗期の長さは通常11時間以上であるが、植物によってはオナモミのように8.5時間以上の連続した暗期が与えられたときに花成が誘導される場合もある。短日植物でも限界暗期は植物の種類によりそれぞれ異なるので、光照射時間サイクルは植物の限界暗期に合わせて適宜設定することが望ましい。
短日植物の例としては、大麻草、菊、イネ、大豆、コスモス、しそ、アサガオ、アカザ、オナモミ、アオウキクサなどを挙げることができる。日本では夏から秋にかけて開花するものが多い。
例えば、大麻草の場合、一例として、栽培初期に長時間(連続18時間)の光照射栽培と短時間(連続6時間)の暗所栽培という光サイクル期間では栄養成長となるが、この光サイクルを短時間(連続12時間)の光照射栽培と長時間(連続12時間)の暗所栽培という光サイクルに切り替えると生殖成長(開花期間)への切り替えが促進される。
本発明で対象とする大麻草(Cannabis sativa)には特に制限なく、様々な品種の大麻草を用いることができる。品種の例として、サティバ(Cannabis sativa var. sativa)、インディカ(Cannabis sativa var. indica)、ルデラリス(Cannabis sativa var. ruderalis)を挙げることができる。これらの品種の混合(hybrid)を用いてもよい。二次代謝産物の化学表現型からの分類では、テトラヒドロカンナビノール(THC)の産生が優位なタイプI、カンナビジオール(CBD)とTHCの産生が同等なタイプII、CBDの産生が優位なタイプIII、カンナビゲロール(CBG)の産生が優位なタイプIVがあるが、いずれのタイプも制限なく栽培することができる。
本発明における大麻草などの短日植物や長日植物の栽培開始状態は、種子、挿し木、苗のいずれでも良い。種子や挿し木から予め苗になるまで成長させておいたり、組織培養体から苗になるまで成長させておいたりしてから、フィルムに移植しても良い。
長日植物の場合、連続した暗期の長さが、ある一定の時間(限界暗期)より短くなるような光周条件下で、花芽の形成が起こるか、あるいはそれが促進される。連続した暗期の長さが十分に短ければ、明期の長さは関係がない。長い暗期の場合、光中断を行うと花芽形成がみられる。
長日植物は一般に、自然界では日長が長くなると花芽が形成される。長日植物の多くは、光周条件(長日)が整う以前に、一定期間、寒冷期を経ることを必要とすることが多い。
長日植物の例としては、ペチュア、キンギョソウ、ストック、トルコギキョウ、フクシア、アブラナ、ダイコンなどを挙げることができる。日本では冬から春にかけて開花するものが多い。全ゲノム配列が解読され、モデル植物として世界中で広く研究に利用されているシロイヌナズナも長日植物として知られている。
太陽光を光源に用いる場合、季節や天候により太陽光の光量は異なるが、光量については特に限定はない。晴天時の光量は約2000μmol/m2/sec、曇天時の光量は約50~約100μmol/m2/secであるが、たとえば曇天時には必要に応じて人工光を同時に用い、太陽光と人工光との合計光量が、後述の人工光のみを光源として用いる場合の光量の通常の範囲、好ましい範囲、さらに好ましい範囲となるように設定してもよい。なお、本発明において光量とは、植物に照射される光の葉面における強度を光合成光量子束密度(PPFD)で表示したものを表す。
人工光のみを光源として用いる場合、光量は、栄養成長期間においても生殖成長期間においても、通常100~2000μmol/m2/secの範囲、好ましくは200~1000μmol/m2/secの範囲、さらに好ましくは300~500μmol/m2/secの範囲である。
上述の光照射サイクルの変更は光源が人工光の場合、光源電源の入切をタイマーによって容易に行うことができる。太陽光の場合は遮光カーテンなどを使用して暗時間を任意に作ることができる。太陽光を光源とする場合、季節によって長時間の光照射が困難な時は人工光との組み合わせで長時間の光照射を行うことができる。この時、人工光による光量は人工光のみを光源として用いる場合の上記の光量と違って、低いもので良く、通常4μmol/m2/sec以上、好ましくは10μmol/m2/sec以上、さらに好ましくは100μmol/m2/sec以上である。
<栽培方法>
本発明の短日植物栽培方法の1つは、以下の通りである。

(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する短日植物の栽培方法。
(2)該システム内の親水性フィルムの上に短日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、

短日植物の栽培において、上記工程(4)の栄養成長期間における連続した暗時間の長さは、通常1時間~10時間、好ましくは3時間~9時間、さらに好ましくは5時間~7時間である。
短日植物の栽培において、上記工程(5)の生殖成長期間における連続した暗時間の長さは、通常11時間~18時間、好ましくは12時間~16時間、さらに好ましくは13時間~15時間である。
短日植物の栽培において、上記工程(4)の栄養成長期間における光照射時間は、通常13時間~23時間、好ましくは15時間~20時間、さらに好ましくは16時間~18時間である。
短日植物の栽培において、上記工程(5)の生殖成長期間における光照射時間は、通常12時間~4時間、好ましくは11時間~6時間、さらに好ましくは10時間~8時間である。
短日植物の栽培において、上記工程(4)の栄養成長期間の長さ(日数)は、植物の種類、フィルム栽培開始時の植物の状態(種子、苗など)などに応じて適宜定めることができる。たとえば、大麻草を後述の実施例1におけるような苗の状態でフィルム栽培を開始するという条件の下においては、栄養成長期間の長さ(日数)は、通常18~60日間、好ましくは20~50日間、さらに好ましくは25~40日間である。
短日植物の栽培において、上記工程(5)の生殖成長期間の長さ(日数)は、植物の種類、フィルム栽培開始時の植物の状態(種子、苗など)などに応じて適宜定めることができる。たとえば、大麻草を後述の実施例1におけるような苗の状態でフィルム栽培を開始するという条件の下においては、生殖成長期間の長さ(日数)は、通常45~80日間、好ましくは50~65日間、さらに好ましくは55~60日間である。
本発明の長日植物栽培方法の1つは、以下の通りである。
(1)植物をその上で栽培するための親水性フィルム、および該親水性フィルムの植物の反対面に接触するように配置された養液保持手段を含む植物栽培システムを提供し、
(2)該システム内の親水性フィルムの上に長日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は毎日13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する長日植物の栽培方法。
(2)該システム内の親水性フィルムの上に長日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は毎日13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行い、そして

ことによって、該親水性フィルムの上で植物を栽培することを包含する長日植物の栽培方法。
長日植物の栽培において、上記工程(4)の栄養成長期間における連続した暗時間の長さは、通常11時間~23時間、好ましくは15時間~20時間、さらに好ましくは16時間~18時間である。
長日植物の栽培において、上記工程(5)の生殖成長期間における連続した暗時間の長さは、通常1時間~10時間、好ましくは3時間~9時間、さらに好ましくは5時間~7時間である。
長日植物の栽培において、上記工程(4)の栄養成長期間における光照射時間は、通常12時間~4時間、好ましくは11時間~6時間、さらに好ましくは10時間~8時間である。
長日植物の栽培において、上記工程(5)の生殖成長期間における光照射時間は、通常13時間~18時間、好ましくは14時間~17時間、さらに好ましくは15時間~16時間である。
長日植物の栽培において、上記工程(4)の栄養成長期間の長さ(日数)は、植物の種類、フィルム栽培開始時の植物の状態(種子、苗など)などに応じて適宜定めることができる。たとえば、シロイヌナズナを後述の実施例3におけるような種子の状態でフィルム栽培を開始するという条件の下においては、栄養成長期間の長さ(日数)は、通常9~20日間、好ましくは11~17日間、さらに好ましくは13~15日間である。
長日植物の栽培において、上記工程(5)の生殖成長期間の長さ(日数)は、植物の種類、フィルム栽培開始時の植物の状態(種子、苗など)などに応じて適宜定めることができる。たとえば、シロイヌナズナを後述の実施例3におけるような種子の状態でフィルム栽培を開始するという条件の下においては、生殖成長期間の長さ(日数)は、通常18~42日間、好ましくは21~35日間、さらに好ましくは25~30日間である。
なお、短日植物の栽培の場合も長日植物の栽培の場合も、工程(4)及び(5)における光照射栽培の時間とは、光照射栽培の合計(つまり、断続的に光照射をしたときは、それらの合計)を意味する。
栽培を開始するに際し、植物は種子あるいは苗の状態で、養液を吸収した本発明の親水性フィルム上に配置することができる。該フィルム上に種子の状態で播種された場合は、まず発芽、発根させる必要があり、種子の発芽、発根に必要な少量の灌水を行う。ここでフィルム上に多量の水分が存在すると、植物の根とフィルムの一体化を妨げるので、種子の発芽、発根に必要な最小限の灌水にとどめる必要がある。必要な最小限の灌水は、通常0.01~10μL/cm2、好ましくは0.5~5μL/cm2、より好ましくは0.1~1μL/cm2である。
植物が本発明の親水性フィルムに苗の状態で配置された場合は、発芽、発根のための灌水は必ずしも必要ないが、苗の根が伸張してフィルムと一体化し、フィルムから水と養分を吸収できるようになるまでは、根が乾燥しないように根の周囲を湿潤状態に保つ必要がある。
保水性の高い植物栽培用支持体をフィルム上に配置する態様によれば、上述のようなフィルム上に多量の水分が存在することを回避しつつ、根の周囲の湿潤状態を維持し易いので好ましい。
以下、実施例により本発明をさらに具体的に説明する。
実施例1
長さ32cm、幅24cm、深さ4cmの黒色ポリプロピレン製トレーに1.5Lの養液を入れ、長さ40cm、幅29cm、厚さ90μmのポリビニルアルコール製フィルム(メビオール社製、日本)を載せた。該フィルムの30℃水中における平衡膨潤度は194%であった。養液は、液体有機肥料Nutri Plus Organic Grow (4.0N-1.3P-1.7K;EZ-GRO Inc., カナダ)にカルシウム-マグネシウムサプリメント(3.0Ca-1.6Mg;EZ-GRO Inc., カナダ)を2mL/L加え、純水で窒素濃度389mg/Lの濃度に希釈することにより得られたものである。養液中の微量元素の濃度(mg/L)は、14.5Zn、12.0B、2.6Mo、2.1Cu、8.5Feであった。

上記親水性フィルムに24℃で養液を吸収させて30分間膨潤させた後、該フィルム上にpH調整済ピートモス(pH約5~6)を約500mL載せ、厚さ約1cmに拡げた。14日間栄養成長した発根挿し木大麻草苗(化学表現II型、高さ約10cm、本葉6枚)を上記膨潤フィルム上に根が接触するように移植し、ピートモスが湿潤状態になるよう上記養液をスプレーした。
温度24℃、相対湿度73%の室内で植物用LEDを葉面光強度400μmol/m2/secで連続18時間照射、連続6時間暗期のサイクルで4週間栽培した(栄養成長期間)。フィルム下面の養液は栽培期間中無くならないように随時追加した。室内には二酸化炭素を導入し、二酸化炭素濃度を約700ppmで維持し、光合成を促進させた。
4週間上記条件で栽培した後、温度22℃、相対湿度60%、二酸化炭素濃度を約700ppmの室内で植物用LEDを葉面光強度400μmol/m2/secで連続12時間照射、連続12時間暗期のサイクルに切り替えて8週間栽培した(開花期間)。本発明の栽培システムでは葉面積が小さいので開花期間中の大麻草栽培密度は、12株/m2(平方メートル)で栽培することができた。
上記栽培後、茎を培土(ピートモス)直上で切断した。茎から大き目の葉を取り除き、植物体を18℃、相対湿度45%で2日間乾燥させ、その後18℃、相対湿度57%で12日間熟成させた。次いで、花穂を枝 (茎頂および腋窩枝の両方) から切断し、花穂から突出していた葉を、花穂乾燥重量測定前にTwister T4機械トリミング機 (Keirton Inc., Surrey, BC, Canada) を用いてトリミングした。
中性カンナビノイドTHC、CBD、およびカンナビゲロール(CBG)、ならびに酸型THCA、CBDA、およびカンナビゲロール酸(CBGA)の分析は、高速液体クロマトグラフィー(Nexera X3、日本国株式会社島津製作所製)によって実施された。乾燥花穂水分含量は、米国薬局方条約第921条、方法3 (米国薬局方および全国処方箋、2017) に記載の方法を用いて決定し、カンナビノイド濃度を0%含水率(以下、含水率の単位はすべて重量%である)に補正した。カンナビノイド収量は、カンナビノイド濃度に花穂燥重量を掛けた値として算出し、単位面積当たりに表した (g/m2) を含水率0%に補正した。
乾燥花穂重量は300g/m2、含水率は9%であった。従って含水率0%の時、乾燥花穂重量は273g/m2と算出された。乾燥花穂中のTHC, CBD, CBG, THCA, CBDA, CBGAの含有率はそれぞれ0.5%, 0.3%, 0.1%, 5.5%, 10.5%, 0.5%であった。従って単位面積当たりのカンナビノイドTHC, CBD, CBG, THCA, CBDA, CBGAの収量はそれぞれ1.4g/m2、0.8g/m2、0.3g/m2、15g/m2、29g/m2、1.4g/m2であった。
実施例2
実施例1において、30℃水中における平衡膨潤度が160%厚さ60μmのポリビニルアルコール製フィルム(メビオール社製、日本)を用いる以外は実施例1と同様の栽培試験をおこなった。開花期間中の大麻草栽培密度は、実施例1と同様12株/m2(平方メートル)で栽培することができた。
実施例1において、30℃水中における平衡膨潤度が160%厚さ60μmのポリビニルアルコール製フィルム(メビオール社製、日本)を用いる以外は実施例1と同様の栽培試験をおこなった。開花期間中の大麻草栽培密度は、実施例1と同様12株/m2(平方メートル)で栽培することができた。
乾燥花穂重量は255g/m2、含水率は10%であった。従って含水率0%の時、乾燥花穂重量は230g/m2と算出された。乾燥花穂中のTHC, CBD, CBG, THCA, CBDA, CBGAの含有率はそれぞれ0.6%, 0.4%, 0.2%, 5.9%, 10.8%, 0.6%であった。従って単位面積当たりのカンナビノイドTHC, CBD, CBG, THCA, CBDA, CBGAの収量はそれぞれ1.4g/m2、0.9g/m2、0.4g/m2、14g/m2、25g/m2、1.4g/m2であった。
比較例1
容量11Lの黒色鉢(直径28cmx高さ24cm)に実施例1と同様のピートモス約10Lを入れ、実施例1と同じ14日間栄養成長した発根挿し木大麻草苗(化学表現II型、高さ約10cm、本葉6枚)を移植した。

実施例1と同じ養液を、植物が萎れないように随時手灌水し、温度24℃、相対湿度73%の室内で植物用LEDを葉面光強度400μmol/m2/secで連続18時間照射、連続6時間暗期のサイクルで4週間栽培した(栄養成長期間)。室内には二酸化炭素を導入し、二酸化炭素濃度を約700ppmで維持し、光合成を促進させた。
4週間上記条件で栽培した後、温度22℃、相対湿度60%、二酸化炭素濃度を約700ppmの室内で植物用LEDを葉面光強度400μmol/m2/secで連続12時間照射、連続12時間暗期のサイクルに切り替えて8週間栽培した(開花期間)。親水性フィルムを使わない栽培では葉面積が大きく育つため、隣接する株と葉が重ならないよう配置するためには大麻草栽培密度を、6.4株/m2とせざるをえなかった。
実施例1と同様に収穫し、分析した結果、乾燥花穂重量は160g/m2、含水率は8%であった。従って含水率0%の時、乾燥花穂重量は147g/m2と算出された。乾燥花穂中のTHC, CBD, CBG, THCA, CBDA, CBGAの含有率はそれぞれ0.3%, 0.2%, 0.1%, 4.7%, 9.0%, 0.4%であった。従って単位面積当たりのカンナビノイドTHC, CBD, CBG, THCA, CBDA, CBGAの収量はそれぞれ0.4g/m2、0.3g/m2、0.1g/m2、6.9g/m2、13g/m2、0.6g/m2であった。
比較例2
実施例1と同様の大麻草栽培を行い、連続18時間照射、連続6時間暗期のサイクルで4週間栽培した(栄養成長期間)後、連続12時間照射、連続12時間暗期のサイクルに切り替えずに温度22℃、相対湿度60%、二酸化炭素濃度約700ppm で8週間栽培したところ、花芽分化が誘導されず、花穂の十分な形成が認められなかった。
実施例1と同様の大麻草栽培を行い、連続18時間照射、連続6時間暗期のサイクルで4週間栽培した(栄養成長期間)後、連続12時間照射、連続12時間暗期のサイクルに切り替えずに温度22℃、相対湿度60%、二酸化炭素濃度約700ppm で8週間栽培したところ、花芽分化が誘導されず、花穂の十分な形成が認められなかった。
実施例3
長さ32cm、幅24cm、深さ4cmの黒色ポリプロピレン製トレーに1.5Lの養液を入れ、長さ40cm、幅29cm、厚さ50μmのポリビニルアルコール製フィルム(メビオール社製、日本)を載せた。該フィルムの30℃水中における平衡膨潤度は250%であった。養液はムラシゲ-スクーグ培地(MS培地)(日本国和光純薬株式会社製)を使用した。
長さ32cm、幅24cm、深さ4cmの黒色ポリプロピレン製トレーに1.5Lの養液を入れ、長さ40cm、幅29cm、厚さ50μmのポリビニルアルコール製フィルム(メビオール社製、日本)を載せた。該フィルムの30℃水中における平衡膨潤度は250%であった。養液はムラシゲ-スクーグ培地(MS培地)(日本国和光純薬株式会社製)を使用した。
上記親水性フィルムに22℃で養液を吸収させて30分間膨潤させた後、長日植物として知られるシロイヌナズナの種子を播種し、種子上に水をスプレーした。発芽するまでは種子を載せたフィルム上の湿度を100%に保つため、トレー上を食品用ラップ(ポリ塩化ビニリデンフィルム、商品名:サランラップ(登録商標)、旭化成社製)で完全に覆った。植物用LEDを種子面光強度400μmol/m2/secで連続8時間照射、連続16時間暗期のサイクルで、フィルムに播種してから2週間栽培した(栄養成長期間)。トレー上のラップは種子が発根、発芽後、徐々に外して栽培室内の環境に馴化させた。栽培室内の相対湿度は約60%、二酸化炭素濃度は約400ppmであった。
上記2週間短日条件で栽培後、植物用LEDを種子面光強度400μmol/m2/secで連続16時間照射、連続8時間暗期のサイクルに切り替えて栽培を続けた(長日処理)ところ、種子を播種してから4週間でシロイヌナズナが開花した。
比較例3
実施例3と全く同様の条件でシロイヌナズナ栽培を行い、長日処理を行わず連続8時間照射、連続16時間暗期のサイクルで栽培を続けたところ、種子を播種してから4週間では開花せず、開花までに8週間を要した。

水または養液を吸収し、適度な水分ストレスを付加して植物を栽培できる親水性フィルムを用いる本発明の植物栽培システムを使用して植物の栽培を行うと、植物の病気を誘発する雑菌などからの感染を回避することができると同時に、植物の生殖成長を促進できる。また、相対的に葉の面積拡大を抑制でき、花穂の葉に対する収量を増大させることが出来る。
また植物の光周性を利用する本発明の植物栽培システムを使用すると、花芽分化を促すことができ、医薬品などとして有用な物質が二次代謝産物として集積される花穂の花成を促進できる。
本発明によれば根ぐされなどの原因となる根の酸素欠乏状態を招くことなく、効率的且つ安定的に、長期間に亘り十分な量の養分を植物の根から吸収させることができ、それにより長期間に亘り持続的に植物の生長を著しく促進させることが可能となる。従って本発明は、植物の栽培が関わる産業、例えば農業や園芸、医薬品製造業など、幅広い分野で利用できる。

Claims (9)
- 親水性フィルムおよび養液保持手段を構成要素とする植物栽培システムであって、使用時には、植物の根の少なくとも一部が親水性フィルム表面に密着し、該植物の根が密着する該親水性フィルムの反対面に養液が接するように養液が配置され、該植物の栄養成長期間と生殖成長期間の切り替えを日長時間の変更によって行うことを特徴とする植物栽培システム。
- 前記親水性フィルムが、該フィルムを介して水と塩水とを対向して接触させた際に、測定開始後4日目(96時間)の水/塩水の電気伝導度(EC)の差が4.5dS/m以下のフィルムであることを特徴とする請求項1に記載の植物栽培システム。
- 該フィルムの水中平衡膨潤度が130%以上300%未満かつ乾燥時の厚みが20μm以上150μm未満であることを特徴とする請求項1または2に記載の植物栽培システム。
- 養液保持手段が水耕栽培用水槽であり、該親水性フィルムの下面に接触するように配置された養液が水耕栽培用水槽に収容されてなることを特徴とする、請求項1~3のいずれかに記載の植物栽培システム。
- 該養液保持手段が水不透過性表面を有し、その上に該親水性フィルムが敷設されてなり、親水性フィルムと養液保持手段との間に該養液を連続的または間歇的に供給する養液供給手段をさらに含むことを特徴とする、請求項1~3のいずれかに記載の植物栽培システム。
- 遮光手段および照明手段からなる群より選ばれる少なくとも1種をさらに含むことを特徴とする、請求項1~5のいずれかに記載の植物栽培システム。
- 短日植物または長日植物の栽培に用いるための請求項1~6のいずれかに記載の植物栽培システム。
- (1)植物をその上で栽培するための親水性フィルム、および該親水性フィルムの植物の反対面に接触するように配置された養液保持手段を含む植物栽培システムを提供し、
(2)該システム内の親水性フィルムの上に短日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する短日植物の栽培方法。 - (1)植物をその上で栽培するための親水性フィルム、および該親水性フィルムの植物の反対面に接触するように配置された養液保持手段を含む植物栽培システムを提供し、
(2)該システム内の親水性フィルムの上に長日植物を配置し、
(3)養液を、該親水性フィルムを介して該植物に接触させ、
(4)初期の栄養成長期間は毎日13時間未満の短時間光照射栽培および連続11時間以上の暗時間栽培を行い、そして
(5)引き続き生殖成長期間は13時間以上の長時間光照射栽培および連続11時間未満の暗時間栽培を行う
ことによって、該親水性フィルムの上で植物を栽培することを包含する長日植物の栽培方法。
