Abstract
本発明は、P.アンタクティカなどの担子菌酵母において、炭素カタボライト抑制を低減させた変異体を提供することを目的としている。 本発明は、グルコキナーゼ遺伝子に機能低下型変異を含む、担子菌酵母に関している。また、本発明は、担子菌酵母の炭素カタボライト抑制を低減する方法であって、前記担子菌酵母のグルコキナーゼの機能を抑制する工程を含む方法にも関している。
Description
本発明は、担子菌酵母及び担子菌酵母の炭素カタボライト抑制を低減する方法に関する。
微生物はタンパク質を生産するために使用されている。一般的には、遺伝子の発現を促すプロモーター配列の下流に目的のタンパク質をコードする遺伝子を連結して作製した組換え遺伝子を、微生物に導入して培養することにより、当該目的のタンパク質を取得する。これまでに実用化されている微生物を宿主に用いたタンパク質発現系には、原核微生物を用いる系と、真核微生物を用いる系があり、前者においては、大腸菌、ブレビバチルス、及びコリネバクテリウムなど、多様な原核微生物が使用されている。一方、後者においては、出芽酵母又は麹菌、あるいはシュードザイマ・アンタクティカ(Pseudozyma antarctica)などの担子菌酵母(特許文献1及び2並びに非特許文献1)を、目的のタンパク質の生産のために使用することが知られている。
また、微生物の培地中にグルコースなどの代謝されやすい炭素源があるときに、他の代謝されにくい炭素源(キシロースなど)の分解系の遺伝子発現を抑制する機構を炭素カタボライト抑制(CCR)という。このCCRに関して、グルコースの構造アナログである2-デオキシグルコース(2DG)は、グルコースによる抑制に対して抵抗性の変異の選択のために用いられている(非特許文献2)。そして、非特許文献3には、出芽酵母においてはヘキソキナーゼの1種であるHxk2が、麹菌においてはグルコキナーゼが、それぞれCCRを制御している旨が記載されており、特許文献3には、CreA及びCreBが麹菌などの糸状菌のCCRを制御している旨が記載されている。他方、担子菌酵母のCCRを制御する酵素については、いずれの文献にも記載されていない。

P.アンタクティカ由来のキシロース誘導性プロモーター(キシラナーゼプロモーター)は、生分解性プラスチック分解酵素PaEなどのタンパク質を、P.アンタクティカによって生産するために使用されているが、安価な炭素源であるグルコースを含む培地ではCCRによりプロモーター機能が制限されてしまう。そこで、本発明は、P.アンタクティカなどの担子菌酵母において、CCRが緩和された変異体を提供することを目的としている。
本発明者らは、上記課題を解決すべく鋭意検討した結果、担子菌酵母のグルコキナーゼがCCRを制御していることを見出し、本発明を完成させた。すなわち、本発明は、以下に示す担子菌酵母及び担子菌酵母の炭素カタボライト抑制を低減する方法を提供するものである。

本発明に従えば、担子菌酵母のグルコキナーゼ遺伝子に機能低下型変異を導入することにより、当該担子菌酵母のCCRを緩和することができる。したがって、安価なグルコースを炭素源とすることが可能となり、組換えタンパク質の製造コストの低減を図ることができる。
以下、本発明をさらに詳細に説明する。
本発明の担子菌酵母は、グルコキナーゼ遺伝子に機能低下型変異を含んでいる。本明細書に記載の「グルコキナーゼ」とは、グルコースからグルコース-6-リン酸へのリン酸化を促進する酵素である。本明細書に記載の「機能低下型変異」とは、グルコキナーゼのタンパク質としての発現を低下させたり、グルコースのリン酸化促進活性が低下したグルコキナーゼ変異体を発現させたりすることにより、前記担子菌酵母においてグルコキナーゼとしての機能の低下を引き起こす変異のことをいう。ある態様では、前記機能低下型変異は、グルコキナーゼの機能を実質的に失う機能喪失型変異であってもよい。前記機能喪失型変異としては、特に限定されないが、例えば、グルコキナーゼのタンパク質の発現が失われるような変異(スプライシング異常、フレームシフト変異、又は遺伝子の一部若しくは全体の欠失、あるいは遺伝子破壊など)であってもいいし、タンパク質としては発現するがグルコキナーゼとしての機能が失われるような変異(活性部位でのアミノ酸置換を伴う変異など)であってもよい。
本発明の担子菌酵母は、グルコキナーゼ遺伝子に機能低下型変異を含んでいる。本明細書に記載の「グルコキナーゼ」とは、グルコースからグルコース-6-リン酸へのリン酸化を促進する酵素である。本明細書に記載の「機能低下型変異」とは、グルコキナーゼのタンパク質としての発現を低下させたり、グルコースのリン酸化促進活性が低下したグルコキナーゼ変異体を発現させたりすることにより、前記担子菌酵母においてグルコキナーゼとしての機能の低下を引き起こす変異のことをいう。ある態様では、前記機能低下型変異は、グルコキナーゼの機能を実質的に失う機能喪失型変異であってもよい。前記機能喪失型変異としては、特に限定されないが、例えば、グルコキナーゼのタンパク質の発現が失われるような変異(スプライシング異常、フレームシフト変異、又は遺伝子の一部若しくは全体の欠失、あるいは遺伝子破壊など)であってもいいし、タンパク質としては発現するがグルコキナーゼとしての機能が失われるような変異(活性部位でのアミノ酸置換を伴う変異など)であってもよい。
背景技術の項で上述したように、出芽酵母ではHxk2がCCRを制御していること、及び、麹菌ではグルコキナーゼ、又はCreA及びCreBがCCRを制御していることが知られており(非特許文献3及び特許文献3を参照)、CCRを制御しているタンパク質は、微生物の種類によって異なっている。すなわち、グルコキナーゼのCCRへの関与は、特定の微生物においてしか知られておらず、またグルコキナーゼ以外にもCCRを制御するタンパク質は知られていたので、出芽酵母とも麹菌とも異なる担子菌酵母においてCCRを制御しているタンパク質を特定することは困難であった。本発明では、意外なことに、担子菌酵母においてはグルコキナーゼがCCRに関与していること、及び、グルコキナーゼの機能を低下させることがCCRの緩和に十分であることが明らかになった。
グルコキナーゼ遺伝子は、担子菌酵母の種類に応じて適宜選択することができるが、例えば、担子菌酵母であるP.アンタクティカのグルコキナーゼ遺伝子(PaGLK1遺伝子)は、配列番号1で示される以下の配列を有している。すなわち、ある態様では、前記グルコキナーゼ遺伝子は、以下の配列番号1で示される塩基配列(小文字はイントロン)を含む遺伝子又はそのホモログであってもよい。
本発明の担子菌酵母の種類は、特に限定されないが、例えば、P.アンタクティカ、P.アフィディス、P.ルグローサ、P.パラアンタクティカ、P.グラミニコラ、P.ツクバエンシス、P.フロキュロッサ、P.ヒューピエンシス、及びP.シャンシエンシスなどのシュードザイマ属の菌であってもよい。なお、P.アンタクティカは、イネやムギなどのイネ科植物の葉面に生息し得るシュードザイマ属菌(酵母)の一種であり、生分解性プラスチック分解酵素(PaE)などを産生し得るものである。最近の分類変更により、P.アンタクティカは、モエスジオマイセス・アンタクティクス(Moesziomyces antarcticus)と呼ばれることもある。
特定の糖の存在により活性化するプロモーター、例えば、キシロースで活性化するキシラナーゼプロモーター及びPaEプロモーターなどのキシロース誘導性プロモーターや、セルロースで活性化するセルラーゼプロモーターなどは、目的のタンパク質の発現のために利用されているが、グルコースの存在下ではCCRにより活性が抑制される。本発明の担子菌酵母においては、CCRが緩和されているため、たとえ培養する菌の炭素源としてグルコースを使用していても、前記キシロース誘導性プロモーターなどの特定の糖の存在により活性化するプロモーターを利用して、効率的に目的のタンパク質を生産することができる。すなわち、ある態様では、前記担子菌酵母は、キシロース誘導性プロモーターをさらに含んでもよく、任意で、当該キシロース誘導性プロモーターの下流に目的のタンパク質をコードする塩基配列をさらに含んでもよい。前記目的のタンパク質は、特に限定されないが、例えば、PaEであってもよい。
本発明の担子菌酵母は、本発明の目的を損なわない限り、任意の変異をさらに含んでもよく、CCRを低減するための別の処理をさらに受けたものであってもよい。
別の態様では、本発明は、担子菌酵母のCCRを低減する方法にも関しており、当該方法は、前記担子菌酵母のグルコキナーゼの機能を抑制する工程を含んでいる。前記グルコキナーゼの機能を抑制する手段は、特に制限されないが、例えば、前記担子菌酵母のグルコキナーゼ遺伝子への機能低下型変異(例えば機能喪失型変異)の導入、前記担子菌酵母のグルコキナーゼ遺伝子の発現抑制(例えばRNAiなど)、又は、前記担子菌酵母の培地中へのグルコキナーゼ阻害剤(例えば抗体又はD-マンノヘプツロースなど)の添加などであってもよい。
本発明のCCRを低減する方法は、本発明の目的を損なわない限り、任意の操作工程をさらに含んでもよく、別の手段によりCCRを低減する工程をさらに含んでもよい。その余の点については、本発明の担子菌酵母について上述したとおりである。
また別の態様では、本発明は、目的のタンパク質の生産方法にも関しており、当該方法は、
(1-1)グルコキナーゼ遺伝子に機能低下型変異を含み、かつ、キシロース誘導性プロモーター及びその下流に目的のタンパク質をコードする塩基配列を含む担子菌酵母を用意する工程、又は、
(1-2)キシロース誘導性プロモーター及びその下流に目的のタンパク質をコードする塩基配列を含む担子菌酵母を用意し、前記担子菌酵母のグルコキナーゼの機能を抑制する工程と、
(2)前記担子菌酵母を培養して、目的のタンパク質を発現させる工程と
を含んでいる。前記工程(2)で用いる担子菌酵母においては、CCRが緩和されているため、培養時の炭素源としてグルコースを採用することができる。すなわち、ある態様では、前記培養工程は、前記担子菌酵母をグルコース存在下で培養する工程を含んでもよい。
(1-1)グルコキナーゼ遺伝子に機能低下型変異を含み、かつ、キシロース誘導性プロモーター及びその下流に目的のタンパク質をコードする塩基配列を含む担子菌酵母を用意する工程、又は、
(1-2)キシロース誘導性プロモーター及びその下流に目的のタンパク質をコードする塩基配列を含む担子菌酵母を用意し、前記担子菌酵母のグルコキナーゼの機能を抑制する工程と、
(2)前記担子菌酵母を培養して、目的のタンパク質を発現させる工程と
を含んでいる。前記工程(2)で用いる担子菌酵母においては、CCRが緩和されているため、培養時の炭素源としてグルコースを採用することができる。すなわち、ある態様では、前記培養工程は、前記担子菌酵母をグルコース存在下で培養する工程を含んでもよい。
本発明の生産方法は、本発明の目的を損なわない限り、任意の操作工程をさらに含んでもよく、別の手段によりCCRを低減する工程をさらに含んでもよい。その余の点については、本発明の担子菌酵母及び本発明のCCRを低減する方法について上述したとおりである。
以下、実施例により本発明を具体的に説明するが、本発明の範囲はこれら実施例に限定されるものではない。
1.炭素カタボライト抑制が解除された変異株の取得
炭素カタボライト抑制(CCR)が解除される変異を見出すため、P.アンタクティカのウラシル要求性自然変異株(PGB371株)に紫外線を照射し、グルコースの構造アナログである2-デオキシグルコース(2DG)を含むキシロース培地で生育可能な菌株を選抜した。2DGは細胞内においてリン酸化されて2DG-6-リン酸を生じるが、これは炭素源として代謝されないだけでなく、CCRを引き起こすことで他の炭素源の代謝を阻害する。そのため、2DGを含むキシロース培地では、炭素カタボライト抑制が解除された株のみが生育可能である。具体的には、0.1%の2DGを添加したキシロース培地で生育可能なPGB371株の変異株を選抜し、CCRが緩和されているか否かを調べた。野生型P.アンタクティカでは、生分解性プラスチック分解酵素PaEの遺伝子(PaEプロモーターの制御下)の発現がCCRを受けることから、グルコースを含むPBSA含有平板寒天培地(3xFMM-0.5%PBSAエマルジョン、1%グルコース、及び1%キシロース寒天培地)に2DG耐性株を植菌し、PBSAの分解によって生じる透明の領域(クリアゾーン)の形成を比較した。なお、PBSA含有平板寒天培地については、要すれば特許文献2を参照されたい。
炭素カタボライト抑制(CCR)が解除される変異を見出すため、P.アンタクティカのウラシル要求性自然変異株(PGB371株)に紫外線を照射し、グルコースの構造アナログである2-デオキシグルコース(2DG)を含むキシロース培地で生育可能な菌株を選抜した。2DGは細胞内においてリン酸化されて2DG-6-リン酸を生じるが、これは炭素源として代謝されないだけでなく、CCRを引き起こすことで他の炭素源の代謝を阻害する。そのため、2DGを含むキシロース培地では、炭素カタボライト抑制が解除された株のみが生育可能である。具体的には、0.1%の2DGを添加したキシロース培地で生育可能なPGB371株の変異株を選抜し、CCRが緩和されているか否かを調べた。野生型P.アンタクティカでは、生分解性プラスチック分解酵素PaEの遺伝子(PaEプロモーターの制御下)の発現がCCRを受けることから、グルコースを含むPBSA含有平板寒天培地(3xFMM-0.5%PBSAエマルジョン、1%グルコース、及び1%キシロース寒天培地)に2DG耐性株を植菌し、PBSAの分解によって生じる透明の領域(クリアゾーン)の形成を比較した。なお、PBSA含有平板寒天培地については、要すれば特許文献2を参照されたい。
大きなクリアゾーンを形成した7株を選抜し、グルコース及びキシロースを炭素源とした液体培地で培養し、炭素カタボライト抑制を受けるキシラナーゼの生産量をSDS-PAGEにより比較した。その結果、グルコースが存在してもキシラナーゼの生産量が低下しない2DGr2株及び2DGr3株の2株をCCR解除株として選抜した。
2.CCR解除株におけるキシラナーゼプロモーターの働き
CCRの影響を受けることが知られているキシラナーゼプロモーターが、CCR解除株ではグルコース存在下でも機能するかどうか確認するため、P.アンタクティカのPaE遺伝子をレポーター遺伝子とする試験系を構築した。具体的には、キシラナーゼプロモーター、PaE遺伝子(PaCLE1)、その上流配列及び下流配列、遺伝子導入の選抜マーカーとしてP.アンタクティカのUra3遺伝子(ウラシルなどのピリミジン合成経路の遺伝子PaURA3)、その上流配列及び下流配列、並びに、キシラナーゼターミネーターを組み合わせて、次の二種類のPaE発現カセットを作製した。

CCRの影響を受けることが知られているキシラナーゼプロモーターが、CCR解除株ではグルコース存在下でも機能するかどうか確認するため、P.アンタクティカのPaE遺伝子をレポーター遺伝子とする試験系を構築した。具体的には、キシラナーゼプロモーター、PaE遺伝子(PaCLE1)、その上流配列及び下流配列、遺伝子導入の選抜マーカーとしてP.アンタクティカのUra3遺伝子(ウラシルなどのピリミジン合成経路の遺伝子PaURA3)、その上流配列及び下流配列、並びに、キシラナーゼターミネーターを組み合わせて、次の二種類のPaE発現カセットを作製した。
*PaE発現カセット1の構造
5’-[PaUra3遺伝子(マーカー)]-[キシラナーゼプロモーター]-[PaE遺伝子]-[キシラナーゼターミネーター]-3’
*PaE発現カセット2の構造
5’-[PaUra3遺伝子上流配列]-[キシラナーゼプロモーター]-[PaE遺伝子]-[キシラナーゼターミネーター]-[PaUra3遺伝子下流配列]-3’
PaE発現カセット1を、CCR解除株として選抜した2DGr2株及び2DGr3株に、ウラシル要求性相補を指標として常法によりランダムに導入した。これらを、グルコースを含むPBSA含有寒天培地に植菌し、最も大きなクリアゾーンを形成する形質転換体をそれぞれ1株ずつ選抜した(2DGr2-3株及び2DGr3-2株)。また、コントロール用に、PaE発現カセット2を1コピー有するCCRが解除されていないコントロール株(dnU株)を作製した。具体的には、野生型P.アンタクティカGB-4(0)株のPaE遺伝子上流配列、Ashbya gossypiiのTEF1プロモーターとターミネーターの間にStreptomyces nourseiのnat1を挿入したNourseothricin耐性遺伝子、PaE遺伝子下流配列を組合せたDNAカセットをGB-4(0)株に導入し、相同組換えにより染色体上のPaE遺伝子をNourseothricin耐性遺伝子と置換した株を、Nourseothricin耐性によって選抜を行った。次に、染色体上のURA3遺伝子をPaE発現カセット2で置換して1コピーのPaE発現カセットを組み込み、5’-FOA(5’-フルオロオロチン酸)耐性を指標としてウラシル要求性となった株を選抜し、PGB817株を得た。上記の手順で作製されたPGB817株を宿主として、染色体上のNourseothricin耐性遺伝子をPaUra3遺伝子と置換し、ウラシル要求性相補によって選抜を行い、得られた株をdnU株とした。そして、2DGr2-3株、2DGr3-2株、及びdnU株を、炭素源の異なる以下の3種類の液体培地でそれぞれ48時間培養した。
(1)8質量%キシロース(8X)
(2)4質量%キシロース+4質量%グルコース(4X4G)
(3)1質量%キシロース+7質量%グルコース(1X7G)
(1)8質量%キシロース(8X)
(2)4質量%キシロース+4質量%グルコース(4X4G)
(3)1質量%キシロース+7質量%グルコース(1X7G)
(PaE活性の測定方法)
PaE活性は、その生分解性プラスチック分解活性に基づき、非特許文献1に記載の方法を一部改良したPaE活性測定法によって評価した。具体的には、滅菌水で適宜希釈した培養上清10μLを、3%w/vに希釈されたポリブチレンサクシネートアジペート(PBSA)のエマルジョン(EM-301、昭和電工株式会社製)8~9μL、HEPES緩衝液(50mM、pH7.3)75μL、及び滅菌水56~57μLと、96穴マイクロプレート(プロテオセーブ(R)SS、住友ベークライト株式会社製)中で混合してOD660(濁度)が0.8となる150μLの反応液を調製し、ディープウェルプレートミキサーを用いて30℃、900rpmで振盪した。そして、反応後のOD660(濁度)を測定して単位時間当たりの濁度減少量を測定し、1分間に反応溶液のOD660を1減少させる酵素活性を1U/mLと定義し、PaE活性値を算出した。その結果を表1に示す。なお、括弧内は8質量%キシロースを含む液体培地で培養したときの培養上清の活性値を基準とした相対活性を示す。

CCRが解除されていないコントロールのdnU株は、8X培地ではPaEを産生することができるものの、4X4G培地では培地中のPaE活性(すなわちPaEの産生量)は低下し、1X7G培地では8X培地と比較して75%以上低下した。これに対して、2DGr2-3株及び2DGr3-2株は、炭素源が変わっても同じようにPaEを産生することができた。しかも、2DGr2-3株及び2DGr3-2株には上記PaE発現カセットが複数コピー導入されているため、培地中のPaE活性(すなわちPaEの産生量)が全体的に高かった。このことから、CCR解除株を用いれば、グルコースを高濃度に含む培地においてもキシラナーゼプロモーターにより、その下流に挿入した遺伝子を高い効率で発現させることができることが示された。
3.CCRの解除に寄与する変異遺伝子の同定
(1)変異体の遺伝子のシーケンシング
出芽酵母では、ヘキソキナーゼの1種であるHxk2遺伝子の変異体で炭素カタボライト抑制が解除されることが知られている(非特許文献3)。そこで、P.アンタクティカJCM10317株のゲノム上を探索すると、出芽酵母のHxk2のアミノ酸配列と相同性を示す配列が2つ存在していることが分かった。XP_014653676.1の推定アミノ酸配列は、Hxk2のアミノ酸配列に対して44%の同一性を示し、XP_014656502.1は36%の同一性を示す。XP_014656502.1の推定アミノ酸配列は、Hxk2よりも出芽酵母のグルコキナーゼ(Glk1)とより高い同一性(39%)を示すため、XP_014653676.1をPaHXK1、XP_014656502.1をPaGLK1と命名した。2DGr2株と2DGr3株におけるこれらの遺伝子配列を調べた結果、いずれもPaHxk1遺伝子には変異が存在せず、PaGlk1遺伝子に変異が存在していることが分かった。具体的な変異は以下のとおりだった。
(1)変異体の遺伝子のシーケンシング
出芽酵母では、ヘキソキナーゼの1種であるHxk2遺伝子の変異体で炭素カタボライト抑制が解除されることが知られている(非特許文献3)。そこで、P.アンタクティカJCM10317株のゲノム上を探索すると、出芽酵母のHxk2のアミノ酸配列と相同性を示す配列が2つ存在していることが分かった。XP_014653676.1の推定アミノ酸配列は、Hxk2のアミノ酸配列に対して44%の同一性を示し、XP_014656502.1は36%の同一性を示す。XP_014656502.1の推定アミノ酸配列は、Hxk2よりも出芽酵母のグルコキナーゼ(Glk1)とより高い同一性(39%)を示すため、XP_014653676.1をPaHXK1、XP_014656502.1をPaGLK1と命名した。2DGr2株と2DGr3株におけるこれらの遺伝子配列を調べた結果、いずれもPaHxk1遺伝子には変異が存在せず、PaGlk1遺伝子に変異が存在していることが分かった。具体的な変異は以下のとおりだった。
(2)PaGlk1遺伝子の破壊
PaGlk1のCCRへの関与を調べるため、PaE発現カセットが1コピー導入されているPGB817株のPaGlk1遺伝子を常法によりPaUra3遺伝子に置換することで、PaGlk1遺伝子破壊株を作製した。そして、コントロール株(dnU株)又はPaGlk1遺伝子破壊株を、8質量%のグルコースを含む培地で24時間培養した後、培地量に対して1質量%のキシロース及び3質量%のグルコースを新たに添加してさらに24時間培養し、その培養上清中のPaE活性を上記項目2と同様にして測定した。その結果、PaGlk1遺伝子破壊株では、コントロール株と比較してPaE活性が増加した(表3)。このことから、PaGLK1がCCRに関与していることが示された。
PaGlk1のCCRへの関与を調べるため、PaE発現カセットが1コピー導入されているPGB817株のPaGlk1遺伝子を常法によりPaUra3遺伝子に置換することで、PaGlk1遺伝子破壊株を作製した。そして、コントロール株(dnU株)又はPaGlk1遺伝子破壊株を、8質量%のグルコースを含む培地で24時間培養した後、培地量に対して1質量%のキシロース及び3質量%のグルコースを新たに添加してさらに24時間培養し、その培養上清中のPaE活性を上記項目2と同様にして測定した。その結果、PaGlk1遺伝子破壊株では、コントロール株と比較してPaE活性が増加した(表3)。このことから、PaGLK1がCCRに関与していることが示された。
(3)変異PaGlk1遺伝子の導入
PaGlk1遺伝子破壊株において、PaUra3遺伝子を、常法により2DGr2株の変異PaGlk1遺伝子と置換し、変異PaGLK1導入株を作製した。また、PaGlk1遺伝子破壊株のPaUra3遺伝子を常法により除去することでPaGLK1破壊―U株を作製した。PGB817株、PaGLK1破壊―U株、及び変異PaGLK1導入株は、PaUra3遺伝子を含まないウラシル要求性変異株であって、PaE発現カセットを1コピー有しているという点で共通の遺伝的バックグラウンドを有している一方、野生型PaGlk1遺伝子を有しているか、PaGlk1遺伝子を有していないか、又は変異PaGlk1遺伝子を有しているという点でのみ相違している。これらの3種類の菌株を、2質量%キシロースを含む(グルコースを含まない)PBSA含有平板寒天培地(2X)又は1%キシロース及び1質量%グルコースを含むPBSA含有平板寒天培地(1X1G)にそれぞれ移植して培養し、産生されたPaEの働きによってPBSAが分解されて生じる透明の領域(クリアゾーン)の外延の直径及びコロニーの直径をそれぞれ測定した。培養後の培地の写真を図1に示し、各直径の測定結果を表4に示す。
PaGlk1遺伝子破壊株において、PaUra3遺伝子を、常法により2DGr2株の変異PaGlk1遺伝子と置換し、変異PaGLK1導入株を作製した。また、PaGlk1遺伝子破壊株のPaUra3遺伝子を常法により除去することでPaGLK1破壊―U株を作製した。PGB817株、PaGLK1破壊―U株、及び変異PaGLK1導入株は、PaUra3遺伝子を含まないウラシル要求性変異株であって、PaE発現カセットを1コピー有しているという点で共通の遺伝的バックグラウンドを有している一方、野生型PaGlk1遺伝子を有しているか、PaGlk1遺伝子を有していないか、又は変異PaGlk1遺伝子を有しているという点でのみ相違している。これらの3種類の菌株を、2質量%キシロースを含む(グルコースを含まない)PBSA含有平板寒天培地(2X)又は1%キシロース及び1質量%グルコースを含むPBSA含有平板寒天培地(1X1G)にそれぞれ移植して培養し、産生されたPaEの働きによってPBSAが分解されて生じる透明の領域(クリアゾーン)の外延の直径及びコロニーの直径をそれぞれ測定した。培養後の培地の写真を図1に示し、各直径の測定結果を表4に示す。
野生型PaGlk1遺伝子を有しているPGB817株は、グルコースを含まない培地ではクリアゾーンを形成し、グルコースを含む培地ではクリアゾーンを形成しなかったのに対し、PaGLK1破壊―U株及び変異PaGLK1導入株は、グルコースの有無にかかわらず、いずれの培地でもクリアゾーンを形成した。このように、PaGlk1遺伝子の破壊によって解除されたCCRは変異PaGlk1遺伝子の導入によっては回復しなかったことから、変異PaGlk1遺伝子は機能を喪失していることが示された。
また、PGB817株、PaGLK1破壊―U株、及び変異PaGLK1導入株を、8質量%のグルコースを含む培地で24時間培養した後、遠心分離により培地を除き、各菌体を4質量%キシロースを含む液体培地(4X)又は1質量%キシロース及び3質量%グルコースを含む液体培地(1X3G)に移し替え、さらに24時間培養した。そして、上記項目2と同様にして、各培養上清のPaE活性を測定した。結果を表5に示す。なお、括弧内は4質量%キシロースの液体培地で培養したときの培養上清の活性値を基準とした相対活性を示す。
固体培養のときと同様に、液体培養においても、PaGlk1遺伝子の破壊によってCCRが解除され、この状態は変異PaGlk1遺伝子の導入によっては回復しないことが確認された。
(4)正常PaGlk1遺伝子の導入
2DGr2-3株及び2DGr3-2株に対して、常法により正常なPaGlk1遺伝子を導入して、正常PaGLK1再導入株を作製した。2DGr2-3株及び2DGr3-2株並びにこれらの正常PaGLK1再導入株を、4%キシロース及び4%グルコースを含むPBSA含有平板寒天培地にそれぞれ移植して培養し、産生されたPaEの働きによってPBSAが分解されて生じる透明の領域(クリアゾーン)の外延の直径及びコロニーの直径をそれぞれ測定した。培養後の培地の写真を図2に示す。
2DGr2-3株及び2DGr3-2株に対して、常法により正常なPaGlk1遺伝子を導入して、正常PaGLK1再導入株を作製した。2DGr2-3株及び2DGr3-2株並びにこれらの正常PaGLK1再導入株を、4%キシロース及び4%グルコースを含むPBSA含有平板寒天培地にそれぞれ移植して培養し、産生されたPaEの働きによってPBSAが分解されて生じる透明の領域(クリアゾーン)の外延の直径及びコロニーの直径をそれぞれ測定した。培養後の培地の写真を図2に示す。
2DGr2-3株及び2DGr3-2株(親株P)は、明確なクリアゾーンを形成したが、これらの正常PaGLK1再導入株(+PaGLK1)は、クリアゾーンをほとんど形成しなかった。すなわち、正常PaGLK1再導入株は、グルコース存在下ではPaEを産生することができず、PBSA分解活性が低下した。このことから、2DGr2-3株及び2DGr3-2株のPaE高生産能は、PaGlk1遺伝子の変異に起因することが示された。
以上より、担子菌酵母のグルコキナーゼ遺伝子に機能低下型変異を導入することにより、当該担子菌酵母のCCRを低減することができることが分かった。したがって、安価なグルコースを炭素源とすることが可能となり、組換えタンパク質の製造コストの低減を図ることができる。
Claims (12)
- グルコキナーゼ遺伝子に機能低下型変異を含む、担子菌酵母。
- シュードザイマ属の菌である、請求項1に記載の担子菌酵母。
- 前記グルコキナーゼ遺伝子が、配列番号1で示される塩基配列を含む遺伝子又はそのホモログである、請求項1又は2に記載の担子菌酵母。
- 前記機能低下型変異が、機能喪失型変異である、請求項1~3のいずれか1項に記載の担子菌酵母。
- 前記機能喪失型変異が、前記グルコキナーゼ遺伝子の破壊を含む、請求項4に記載の担子菌酵母。
- キシロース誘導性プロモーターと、その下流に続く目的のタンパク質をコードする塩基配列とをさらに含む、請求項1~5のいずれか1項に記載の担子菌酵母。
- 担子菌酵母の炭素カタボライト抑制を低減する方法であって、前記担子菌酵母のグルコキナーゼの機能を抑制する工程を含む、方法。
- 前記担子菌酵母が、シュードザイマ属の菌である、請求項7に記載の方法。
- 前記抑制工程が、グルコキナーゼ遺伝子に機能低下型変異を導入する工程を含む、請求項7又は8に記載の方法。
- 前記グルコキナーゼ遺伝子が、配列番号1で示される塩基配列を含む遺伝子又はそのホモログである、請求項9に記載の方法。
- 前記機能低下型変異が、機能喪失型変異である、請求項9又は10に記載の方法。
