탄력섬유인 엘라스틴을 분해하는 효소로 알려진 엘라스타아제(elastase)는 건강한 피부에서 낮은 농도로 존재하며, 피부 염증반응 시에는 엘라스타아제 농도가 급격히 증가하기도 한다. 이러한 엘라스타아제는 탄성 섬유단백질인 엘라스틴을 분해시켜 피부의 진피조직의 그물망을 끊어 줌으로서 주름생성의 주원인 효소로 알려져 있으며, 엘라스타아제 활성을 저하시킴으로써 피부노화를 억제할 수 있다. 따라서 엘라스타아제 활성 저해능력을 실험하여 추출물의 피부 주름개선 물질의 효력을 뒷받침할 수 있다. 엘라스타아제 저해 활성 측정은 기질로서 N-succinyl-Ala-Ala-Ala-p-nitroanilide (sigma)를 사용하여 생성된 p-nitoanilide의 생성량을 측정하였다.Elastase, known as an enzyme that decomposes elastin, an elastic fiber, exists in low concentrations in healthy skin, and the concentration of elastase increases rapidly during skin inflammatory reactions. 멜라닌 합성과정에서의 핵심 효소인 티로시나아제(tyrosinase)의 저해활성을 평가하여 미백효과의 가능성을 유추할 수 있다. 티로신(Tyrosine)은 효소인 티로시나아제에 의하여 멜라닌 생성의 전구체가 되는 DOPA 와 dopaquinone으로 대사된다. 따라서 티로시나아제의 억제는 피부의 멜라닌 색소의 조절을 통하여 피부 미백 효과를 기대할 수 있다. dopachrome법을 이용하여 티로시나아제 억제 효과를 측정하였다. mushroom Tyrosinase -300 unit 50 uL와 1.5 mM L-tyrosine 25 uL, 또는 시료 75 uL를 혼합한 후 배양 전, 15분간 배양 후의 각각의 흡광도를 490 nm의 파장에서 측정하였고, 결과는 하기 표 4와 도 3에 나타내었다. 대조군은 mushroom Tyrosinase -300 unit 50 uL와 1.5 mM L-tyrosine 25 uL, 정제수 75 uL를 혼합하여 처리하였다.The possibility of a whitening effect can be inferred by evaluating the inhibitory activity of tyrosinase, a key enzyme in the melanin synthesis process.
