本発明は、有機銅化合物、及び有機銅化合物の製造方法に関する。
非特許文献1は、9-フルオレニルメチルオキシカルボニル保護基(Fmoc)で保護した有機ヨウ素亜鉛化合物を開示している。
特許文献1は、tert-ブトキシカルボニル保護基(Boc)で保護した有機ヨウ素亜鉛化合物、及びBocで保護した有機臭素亜鉛化合物を開示している。
特許文献2は、Bocで保護した有機ヨウ素亜鉛化合物、及びベンジルオキシカルボニル保護基(Cbz)で保護した有機ヨウ素亜鉛化合物を開示している。

本発明は、新たに、特殊アミノ酸の合成における有用な有機銅化合物を提供する事を目的とする。
本発明者は、上記課題を解決すべく、鋭意研究を重ねた結果、銅剤を使用する事に依り、新たに、特殊アミノ酸の合成における有用な有機銅化合物を製造する事が出来た。
本発明者は、係る知見に基づき、更に研究を重ね、本発明を完成するに至った。
即ち、本発明は、以下の有機銅化合物、及び有機銅化合物の製造方法を包含する。

一般式(2)中、R1は、更に好ましくは、炭素数1~4の直鎖状又は分岐鎖状のアルコキシ基、置換基を有していてもよいフェノキシ基、置換基を有していてもよいベンジルオキシ基である。
一般式(2)中、R1は、特に好ましくは、メトキシ、tert-ブトキシ等である。

本発明の有機銅化合物の製造方法で使用する、一般式(2)で表される化合物は、一般式(3)で表される化合物に、亜鉛を、従来公知の方法、例えば、Org. Biomol. Chem., 1(23), 4254-4261 (2003)に記載の方法等に従い、反応させることで製造することができる。
Xは、塩素原子、臭素原子、シアノ基、又はトリフルオロメタンスルホナート基であり、
Protは、9-フルオレニルメチルオキシカルボニル保護基、tert-ブトキシカルボニル保護基、アリルオキシカルボニル保護基、又はベンジルオキシカルボニル保護基から選択される保護基であり、

一般式(3)で表される化合物のXが塩素原子、又は臭素原子である場合、有機溶媒中で、ハロゲン化合物の存在下、一般式(3)で表される化合物と亜鉛とを反応させることで一般式(2)で表される化合物を製造することができる。
前記有機溶媒は、好ましくは、非プロトン性極性溶媒、ハロゲン系溶媒、エーテル系溶媒、エステル系溶媒、炭化水素系溶媒、及び芳香族系溶媒から成る群から選択される少なくとも一種の有機溶媒である。
前記有機溶媒の使用量は、前記一般式(3)で表される化合物の濃度(M(mol/L))が、好ましくは、0.01M~5Mの濃度に、より好ましくは、0.1M~2Mの濃度に成る様に調整すればよい。
前記ハロゲン化合物は、好ましくは、ハロゲン、及びハロゲン化アルキルから成る群から選択される少なくとも一種のハロゲン化合物である。
ハロゲン化アルキルは、一般式Ra-Xa(Raはアルキル基を表し、Xaはハロゲン原子を表す)で表される有機化合物である。
一般式Ra-Xaにおいて、Raは、アルキル基を表し、好ましくは、直鎖状、分岐状又は環状のアルキル基が挙げられ、具体的には、例えば、メチル、エチル、n-プロピル、イソプロピル、n-ブチル、イソブチル、sec-ブチル、tert-ブチル、1-エチルプロピル等の炭素数1~4の直鎖状又は分岐鎖状のアルキル基;n-ペンチル、イソペンチル、ネオペンチル、n-ヘキシル、イソヘキシル、3-メチルペンチル、n-ヘプチル、n-オクチル、n-ノニル、n-デシル、n-ウンデシル、n-ドデシル、5-プロピルノニル、n-トリデシル、n-テトラデシル、n-ペンタデシル、ヘキサデシル、ヘプタデシル、オクタデシル等を加えた炭素数1~18の直鎖状又は分岐状アルキル基;シクロプロピル、シクロブチル、シクロペンチル、シクロヘキシル、シクロヘプチル、シクロオクチル等の炭素数3~8の環状アルキル基等である。
一般式Ra-Xaにおいて、Xaは、ハロゲン原子を表し、フッ素、塩素、臭素、及びヨウ素から成る群から選択される少なくとも一種のハロゲン原子であり、より好ましくは、塩素原子、又は臭素原子である。
ハロゲン化合物の使用量は、前記有機溶媒中の濃度(M(mol/L))が、好ましくは、0.01M~5Mの濃度に、より好ましくは、0.1M~2Mの濃度に成る様に調整すればよい。
前記亜鉛の使用量は、前記有機溶媒中の濃度(M(mol/L))が、好ましくは、0.01M~5Mの濃度に、より好ましくは、0.1M~2Mの濃度に成る様に調整すればよい。
一般式(2)で表される化合物の製造方法では、反応温度は、好ましくは、0℃~100℃であり、好ましくは、10℃~80℃であり、更に好ましくは、20℃~60℃とすればよい。
一般式(2)で表される化合物の製造方法では、反応時間は、好ましくは、0.1時間~100時間であり、好ましくは、0.1時間~50時間であり、更に好ましくは、0.1時間~10時間とすればよい。
この製造方法によれば、得られる一般式(2)で表される化合物のα位の立体配置は、使用する一般式(3)で表される化合物のα位の立体配置が維持されるため、一般式(2a)で表される化合物、もしくは一般式(2b)で表される化合物、又は一般式(2a)で表される化合物と一般式(2b)で表される化合物の混合物は、使用する一般式(3)で表される化合物を選択することによって製造することができる。

本発明の有機銅化合物の製造方法は、有機溶媒中で、前記一般式(2)で表される化合物(基質)と銅剤とを反応させる工程を含む。

前記有機溶媒は、より好ましくは、アセトン、アセトニトリル、DMF、DMSO等の非プロトン性極性溶媒、THF、ジオキサン、ジエチルエーテル等のエーテル系溶媒(極性溶媒)、ジクロロメタン、トリクロロメタン等のハロゲン系溶媒である。
前記有機溶媒の使用量は、前記一般式(2)で表される化合物(基質)の濃度(M(mol/L))が、好ましくは、0.01M~5Mの濃度に、より好ましくは、0.1M~2Mの濃度に成る様に調整する。

本発明の有機銅化合物の製造方法は、有機溶媒中で、前記一般式(2)で表される化合物と銅剤とを反応させる工程を含む。

本発明の有機銅化合物の製造方法では、前記一般式(1)中、前記Xが、塩素原子(Cl)、又は臭素原子(Br)である化合物を製造する時、前記銅剤は、好ましくは、塩化銅(I)、臭化銅(I)、及びヨウ化銅(I)から成る群から選択される少なくとも一種の化合物である。
本発明の有機銅化合物の製造方法では、前記一般式(1)中、前記Xが、塩素原子(Cl)、又は臭素原子(Br)である化合物を製造する時、好ましくは、前記銅剤は、塩化銅(I)、臭化銅(I)、及びヨウ化銅(I)から成る群から選択される少なくとも一種の化合物であり、塩化リチウム、臭化リチウム、及びヨウ化リチウムから成る群から選択される少なくとも一種の化合物の存在下(組み合わせ)で、反応を行う。
本発明の有機銅化合物の製造方法では、前記一般式(1)中、前記Xが、シアノ基である化合物を製造する時、前記銅剤は、好ましくは、シアン化銅(I)である。
本発明の有機銅化合物の製造方法では、前記一般式(1)中、前記Xが、シアノ基である化合物を製造する時、好ましくは、前記銅剤は、シアン化銅(I)(CuCN)であり、塩化リチウムの存在下(組み合わせ)で、反応を行う。
本発明の有機銅化合物の製造方法では、前記一般式(1)中、前記Xが、塩素原子、又は臭素原子である化合物を製造する時、前記銅剤は、より好ましくは、塩化銅(I)と塩化リチウムとの組み合わせである。
本発明の有機銅化合物の製造方法では、前記一般式(1)中、前記Xが、シアノ基である化合物を製造する時、前記銅剤は、より好ましくは、シアン化銅(I)と塩化リチウムとの組み合わせである。

本発明の有機銅化合物の製造方法において、銅剤中のリチウムハライド(LiCl, LiBr, LiI等)の使用量は、前記一般式(2)で表される化合物(基質)に対する当量として(基質を1当量とする時に)、モル比で、好ましくは、0.2当量~200当量であり、より好ましくは、0.8当量~20当量であり、更に好ましくは、0.9当量~10当量である。
本発明の有機銅化合物の製造方法において、銅剤の使用量は、前記一般式(2)で表される化合物(基質)に対する当量として(基質を1当量とする時に)、モル比で、特に、1当量の塩化銅(I)、臭化銅(I)、ヨウ化銅(I)及びシアン化銅(I)から成る群から選択される少なくとも一種の化合物対して、2当量の塩化リチウムであり、最大の収率で、目的化合物の一般式(1)で表される有機銅化合物を得ることができる。
本発明の有機銅化合物の製造方法では、銅剤を用いる事に依り、新たに、有機銅化合物を製造する事が出来、この有機銅化合物は、安定性に優れる。
本発明の有機銅化合物は、特殊アミノ酸の合成において、特に、アリール型特殊アミノ酸へ誘導化において、有用である。

本発明の有機銅化合物は、安定性に優れ、特殊アミノ酸の合成において、特に、アリル型特殊アミノ酸への誘導化において、有用である。
以上、本発明の実施形態について説明したが、本発明はこうした例に何ら限定されるものではない。本発明は、本発明の要旨を逸脱しない範囲において種々なる形態で実施し得ることは勿論である。
以下、実施例に基づき、本発明の実施形態をより具体的に説明する。
本発明がこれらに限定されるものではない。
なお、実施例中、MeOはメトキシ基、t-BuOはtert-ブトキシ基を示す。
参考例
(一般式(2)中、Prot=Fmoc, X=Br, R 1 =MeO, R 2 =Hである化合物の製造)
ジメチルスルホキシド(DMSO)(5mL)中に、亜鉛(3mmol、196.2mg)、ヨウ素(0.02mmol、5mg)を順に投入し、1分~5分間、撹拌した。
(一般式(2)中、Prot=Fmoc, X=Br, R 1 =MeO, R 2 =Hである化合物の製造)
ジメチルスルホキシド(DMSO)(5mL)中に、亜鉛(3mmol、196.2mg)、ヨウ素(0.02mmol、5mg)を順に投入し、1分~5分間、撹拌した。
保護された(l)-ブロモセリンメチルエステル(一般式(3)中、Prot=Fmoc, X=Br, R1=MeO, R2=Hである化合物)(2mmol、800mg)を加え、12時間撹拌し、一般式(2)で表される化合物(L体:Prot=Fmoc, X=Br, R1=MeO, R2=Hである化合物)を得た。
得られた有機亜鉛化合物を凍結乾燥した後、DMSO-d6中にて測定したNMRを記す。
1H NMR (300 MHz, d6-DMSO): 8.03 (d,J=8.4Hz,1H), 7.90 (d, J=7.2Hz, 2H), 7.73 (d, J=7.2Hz, 2H), 7.43(t, J=6.9Hz, 2H), 7.34(t, J=6.9Hz, 2H), 6.30(d, J=6.6Hz, 1H),3.96-3.89(m, 1H), 3.54(s, 3H), 0.27-0.18(m, 2H)
実施例の製造方法
実施例1
(一般式(1)中、Prot=Fmoc, X=Cl, R 1 =MeO,R 2 =Hである化合物の製造)
有機溶媒としてジメチルスルホキシド(DMSO)溶媒(1mL)中に、塩化銅(I)(CuCl)(2mmol、200mg)と塩化リチウム(LiCl)(4mmol、170mg)を投入し、30分間、室温下で撹拌した。
実施例1
(一般式(1)中、Prot=Fmoc, X=Cl, R 1 =MeO,R 2 =Hである化合物の製造)
有機溶媒としてジメチルスルホキシド(DMSO)溶媒(1mL)中に、塩化銅(I)(CuCl)(2mmol、200mg)と塩化リチウム(LiCl)(4mmol、170mg)を投入し、30分間、室温下で撹拌した。
その後、反応液を0℃まで冷却し、基質として有機亜鉛化合物(一般式(2a)(L体)中、Prot=Fmoc, X=Cl, R1=MeO,R2=Hである化合物)(850mg)を含むDMSO溶媒(5mL)を投入し、1時間~10時間撹拌した。
前記製造方法に依り、有機銅化合物(一般式(1a)(L体)中、Prot=Fmoc, X=Cl, R1=MeO,R2=Hである化合物)を得る事が出来た。
得られた有機銅化合物を凍結乾燥した後、DMSO-d6中にて測定したNMRを記す。
1H NMR (300 MHz, d6-DMSO): 8.02 (d,J=8.4Hz,1H), 7.90 (d, J=7.4Hz, 2H), 7.73 (d, J=7.4Hz, 2H), 7.44(t, J=6.9Hz, 2H), 7.34(t, J=6.9Hz, 2H), 6.30(d, J=6.6Hz, 1H),3.85-3.79(m, 1H), 3.54(s, 3H), 0.37-0.29(m, 2H)
実施例2
(一般式(1)中、Prot=Fmoc, X=Cl, R 1 =MeO,R 2 =Hである化合物の製造)
有機溶媒としてジメチルスルホキシド(DMSO)溶媒(1mL)中に、シアン化銅(I)(CuCN)(2mmol、180mg)と塩化リチウム(LiCl)(4mmol、170mg)を投入し、30分間、室温下で撹拌した。
(一般式(1)中、Prot=Fmoc, X=Cl, R 1 =MeO,R 2 =Hである化合物の製造)
有機溶媒としてジメチルスルホキシド(DMSO)溶媒(1mL)中に、シアン化銅(I)(CuCN)(2mmol、180mg)と塩化リチウム(LiCl)(4mmol、170mg)を投入し、30分間、室温下で撹拌した。
その後、反応液を0℃まで冷却し、基質として有機亜鉛化合物(一般式(2a)(L体)中、Prot=Fmoc, X=Cl, R1=MeO,R2=Hである化合物)(850mg)を含むDMSO溶媒(5mL)を投入し、1時間~10時間撹拌した。
前記製造方法に依り、有機銅化合物(一般式(1a)(L体)中、Prot=Fmoc, X=Cl, R1=MeO,R2=Hである化合物)を得る事が出来た。
得られた有機銅化合物を凍結乾燥した後、DMSO-d6中にて測定したNMRを記す。

実施例3

有機溶媒としてジメチルスルホキシド(DMSO)溶媒(1mL)中に、塩化銅(I)(CuCl)(2mmol、200mg)と塩化リチウム(LiCl)(4mmol、170mg)を投入し、30分間、室温下で撹拌した。
(一般式(1)中、Prot=Boc, X=Cl, R 1 =MeO,R 2 =Hである化合物の製造)

前記製造方法に依り、有機銅化合物(一般式(1a)(L体)中、Prot=Boc, X=Cl, R 1 =MeO,R2=Hである化合物)を得る事が出来た。
得られた有機銅化合物を凍結乾燥した後、DMSO-d6中にて測定したNMRを記す。
1H NMR (300 MHz, d6-DMSO): 6.11(d, J=6.6Hz, 1H),3.85-3.79(m, 1H), 3.53(s, 3H), 1.39 (s, 9H), 0.49-0.41(m, 2H)
実施例4
(一般式(1)中、Prot=Boc, X=Cl, R 1 =MeO,R 2 =Hである化合物の製造)
有機溶媒としてジメチルスルホキシド(DMSO)溶媒(1mL)中に、ヨウ化銅(I)(CuI)(2mmol、380mg)と塩化リチウム(LiCl)(4mmol、170mg)を投入し、30分間、室温下で撹拌した。
(一般式(1)中、Prot=Boc, X=Cl, R 1 =MeO,R 2 =Hである化合物の製造)
有機溶媒としてジメチルスルホキシド(DMSO)溶媒(1mL)中に、ヨウ化銅(I)(CuI)(2mmol、380mg)と塩化リチウム(LiCl)(4mmol、170mg)を投入し、30分間、室温下で撹拌した。
その後、反応液を0℃まで冷却し、基質として有機亜鉛化合物(一般式(2a)(L体)中、Prot=Boc, X=Cl, R1=MeO,R2=Hである化合物)(610mg)を含むDMSO溶媒(5mL)を投入し、1時間~10時間撹拌した。
前記製造方法に依り、有機銅化合物(一般式(1a)(L体)中、Prot=Boc, X=Cl, R1=MeO,R2=Hである化合物)を得る事が出来た。
得られた有機銅化合物を凍結乾燥した後、DMSO-d6中にて測定したNMRを記す。
1H NMR (300 MHz, d6-DMSO): 6.13(d, J=6.6Hz, 1H),3.85-3.79(m, 1H), 3.53(s, 3H), 1.30 (s, 9H), 0.22-0.11(m, 2H)
本発明の有機銅化合物の製造方法では、銅剤を用いる事に依り、新たに、有機銅化合物を製造する事が出来、この有機銅化合物は、安定性に優れる。
本発明の有機銅化合物は、特殊アミノ酸の合成において、特に、アリル型特殊アミノ酸への誘導化において、有用である。
Claims (4)
- 下記一般式(1):
Xは、塩素原子、臭素原子、シアノ基、又はトリフルオロメタンスルホナート基であり、
Protは、9-フルオレニルメチルオキシカルボニル保護基、tert-ブトキシカルボニル保護基、アリルオキシカルボニル保護基、又はベンジルオキシカルボニル保護基から選択される保護基であり、
R1は、水酸基、又はカルボン酸の保護基であり、
R2は、水素原子、又はアルキル基である。)
で表される有機銅化合物の製造方法であって、
有機溶媒中で、
下記一般式(2):
で表される化合物と銅剤とを反応させる工程、を含む、
有機銅化合物の製造方法。 - 前記一般式(1)中、前記Xが、塩素原子、又は臭素原子である化合物を製造する時、前記銅剤は、塩化銅(I)、臭化銅(I)、及びヨウ化銅(I)から成る群から選択される少なくとも一種の化合物であり、
前記一般式(1)中、前記Xが、シアノ基である化合物を製造する時、前記銅剤は、シアン化銅(I)である、
請求項2に記載の製造方法。 - 前記有機溶媒は、非プロトン性極性溶媒、ハロゲン系溶媒、エーテル系溶媒、エステル系溶媒、炭化水素系溶媒、及び芳香族系溶媒から成る群から選択される少なくとも一種の有機溶媒である、請求項2又は3に記載の製造方法。
