Abstract
본 발명은 두룸아마이드 A의 제조 방법에 관한 것이다. 본 발명의 일 구체예에 따른 코리네박테리움 두룸을 이용한 추출 방법 및 트립타민을 이용한 합성 방법을 이용함으로써 고수율의 두룸아마이드 A를 제조할 수 있다.
Description
본 발명은 두룸아마이드 A(durumamide A)의 제조 방법에 관한 것이다.
천연물로부터 신물질 및 유용물질을 개발하는 연구는 정밀화학과 생명공학의 핵심분야로서 의약품, 건강보조제 등 고부가가치 산업의 필수 기반기술이다. 현재 시판되는 의약품의 1/3 이상이 천연물이거나 그 유도체라는 점을 고려해 보면 천연물의 산업적 중요성을 알 수 있다.
한편, 방선균(Actinomycetes)은 그람-양성 세균에 속하는 미생물로 자연계에 널리 분포하며, 아미노산, 비타민, 효소, 항암물질, 항생 물질 등의 이로운 물질을 생산하기 때문에 생물공학 분야에서 중요한 역할을 하고 있다. 또한, 방선균이 생산하는 유용한 화합물 중 130~140 종류는 인간의 의약품으로, 더 나아가 일부 15~20 종류의 화합물은 식물 보호 인자 및 살충제, 식품 첨가제, 의약품 등으로 널리 이용되고 있다. 대한민국 공개공보 제10-2020-0143262호에는 트레오닌을 생산하는 코리네박테리움(Corynebacterium) 속 균주 및 이의 배양물을 위장질환 예방 또는 치료를 위한 의약품, 식품, 사료 등으로 활용할 수 있음이 개시되어 있다.
따라서 미생물로부터 의약품, 식품 등 다양한 산업 분야에서 유용하게 사용될 수 있는 화합물을 발굴하고 이를 추출 또는 합성하는 방법에 대해 많은 연구가 요구되고 있다.
본 발명은 코리네박테리움 두룸을 이용한 두룸아마이드 A의 제조 방법을 제공하는 것을 목적으로 한다.
또한, 본 발명은 3-인돌글리옥실릴 클로라이드와 4-다이메틸아미노피리딘 및 트립타민을 이용한 두룸아마이드 A의 제조 방법을 제공하는 것을 목적으로 한다.
또한, 본 발명은 DMSO에 트립타민을 용해한 용해물을 이용한 두룸아마이드 A의 제조 방법을 제공하는 것을 목적으로 한다.
두룸아마이드 A는 하기 화학식 1로 표시되는 2-(1H-indol-3-yl)-N-[2-(1H-indol-3-yl)ethyl]-2-oxoacetamide로서 코리네박테리움 두룸(Corynebacterium durum)에서 추출되거나 트립타민(tryptamine)으로부터 합성된 화합물이다. 종래에는 코리네박테리움 두룸이 두룸아마이드 A를 생산하는 것이 확인된 바 없으며, 인공적으로 합성하더라도 인돌(indole)로부터 합성된 방법만이 알려져 있었다. 이에, 본 발명자들은 코리네박테리움 두룸을 이용한 추출 방법 및 트립타민을 이용한 합성 방법을 개발하여 고수율의 두룸아마이드 A를 제조함으로써 본 발명을 완성하였다.

본 발명의 일 양상은 a) 코리네박테리움 두룸을 배양하여 배양물을 수득하는 단계; b) 상기 배양물을 아세톤으로 추출하여 아세톤 추출물을 수득하는 단계; c) 상기 아세톤 추출물을 유기용매로 분획하여 분획물을 수득하는 단계; 및 d) 상기 분획물에 크로마토그래피를 수행하여 두룸아마이드 A를 수득하는 단계를 포함하는 두룸아마이드 A의 제조 방법을 제공한다.
상기 a) 단계는 두룸아마이드 A를 코리네박테리움 두룸으로부터 분리하기 위하여 코리네박테리움 두룸을 배양하여 준비하는 단계이다.
상기 a) 단계의 배양은 당업자에게 통상적인 방법으로 시행될 수 있으며, 바람직한 예로는 BHI(brain heart infusion) 배지에서 30 내지 40℃, 바람직하게는 37℃에서 2주 내지 6주, 또는 3주 내지 5주, 바람직하게는 4주간 배양할 수 있다.
상기 b) 단계는 상기 a) 단계의 결과물인 배양물에서 두룸아마이드 A를 추출하기 위해 아세톤 추출을 시행하는 단계이다.
본 발명의 일 실시예에 따르면, 코리네박테리움 두룸이 배양된 배양물에 다공성 수지를 투입하고 흔든 후, 유기화합물이 흡착된 수지는 여과하여 거른 다음 아세톤으로 추출하는 단계이다.
본 발명의 바람직한 일 실시예에 따르면, 상기 다공성 수지는 XAD-7-HP일 수 있으나 이에 제한되지 않는다.
상기 b) 단계의 아세톤은 1 내지 100% (v/v)일 수 있다. 본 발명의 일 실시예에 따르면, 상기 추출용매 중 아세톤은 20 내지 100% (v/v), 30 내지 100% (v/v)일 수 있으며, 예를 들어 30%, 50%, 70%, 바람직하게는 99.5%일 수 있다.
상기 c) 단계는 상기 b) 단계의 결과물인 아세톤 추출물을 유기용매로 분획하는 단계이다.
본 발명의 일 구체예에 따르면, 상기 유기용매는 메탄올, 에탄올, 부탄올, 아세톤, 에틸아세테이트, 에테르, 클로로포름, 펜탄, 헥산, 헵탄, 노난, 데칸, 벤젠, 톨루엔 및 자일렌으로 이루어진 군에서 선택되는 어느 하나일 수 있다.
본 발명의 일 구체예에 따르면, 상기 c) 단계는 c-1) 상기 아세톤 추출물을 클로로포름을 포함한 용매로 분획하여 클로로포름 분획물을 수득하는 단계; 및 c-2) 상기 클로로포름 분획물을 아세토니트릴을 포함한 용매로 분획하여 아세토니트릴 분획물을 수득하는 단계를 포함하는 것일 수 있다.
상기 c-1) 단계는 클로로포름 분획물을 수득하는 단계이다.
본 명세서에서 사용된 "분획물"은 여러 다양한 구성 성분들을 포함하는 혼합물로부터 특정 성분 또는 특정 성분 그룹을 분리하는 분획방법을 수행하여 얻어진 결과물을 의미한다.
상기 클로로포름 분획물은 클로로포름을 포함한 유기용매로 분획한 결과물을 의미하며, 본 발명에서 상기 분획물을 얻는 분획 방법은 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 수행될 수 있다. 본 발명의 바람직한 일 실시예에 따르면, 상기 유기용매는 클로로포름과 탈염 증류수의 혼합용매일 수 있다. 본 발명에서는 상기의 분획방법을 단독으로 사용하거나, 2 종 이상 혼합하여 사용할 수 있고, 각각의 분획방법을 반복하여 사용할 수도 있다.
본 발명의 일 구체예에 따르면, 상기 c-1) 단계의 클로로포름을 포함한 용매는 클로로포름과 물을 포함하는 것일 수 있다.
상기 c-2) 단계는 아세토니트릴 분획물을 수득하는 단계이다.
상기 c-2) 단계는 c-1) 단계의 결과물인 클로로포름 분획물의 유기용매 층에 용해된 유기물질의 지방성분의 제거를 위하여 아세토니트릴을 포함한 용매로 분획하는 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 c-2) 단계는 아세토니트릴과 n-핵산을 포함하는 용매로 분획하는 단계일 수 있다. 본 발명에서 상기 분획물을 얻는 분획 방법은 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 수행될 수 있다. 본 발명에서는 상기의 분획방법을 단독으로 사용하거나 2 종 이상 혼합하여 사용할 수 있고, 각각의 분획방법을 반복하여 사용할 수도 있다.
상기 d) 단계는 두룸아마이드 A를 수득하는 단계이다.
본 발명의 일 구체예에 따르면, 상기 d) 단계의 크로마토그래피는 아세토니트릴/물을 이동상으로 하여 아세토니트릴 분획물로부터 두룸아마이드 A를 추출하는 것일 수 있다.
상기 크로마토그래피는 고정상과 이동상을 이용하여 여러 가지 물질들이 섞여 있는 혼합물을 이동속도 차이에 따라 분리하는 방법이다. 크로마토그래피는 정지상의 특성에 따라 평면, 종이, 얇은막 크로마토그래피로 나뉘고, 이동상의 특성에 따라 기체 또는 액체 크로마토그래피로 나뉠 수 있다. 기타 친화성, 이온 교환, 크기 배제 또는 이차원 크로마토그래피 기법이 존재하며 본 발명에서의 크로마토그래피는 어느 한 기법에 제한되지 않는다.
상기 두룸아마이드 A 수득은 5% 이내, 바람직하게는 0.1%의 포름산이 포함된 1 내지 10%, 바람직하게는 5% 아세토니트릴/물을 이동상으로 하여 5분 동안 용리시킨 후, 기울기 용리로 10분 내지 20분 동안, 바람직하게는 15분간 100%까지 증가시켜 얻는 것일 수 있다.
본 발명의 일 실시예에 따르면, 상기 d) 단계는 아세토니트릴층에 용해된 분획물로부터 C18 HPLC (4.6 Х 250 mm, 1.0 mL/min)를 이용하여 0.1% 포름산이 포함된 5% 아세토니트릴/H2O 이동상으로 5분간 용리시킨 후, 기울기 용리로 15분간 100%까지 증가시켜 두룸아마이드 A (t
R 17.2 min)를 얻을 수 있다.
본 발명의 다른 양상은, i) 3-인돌글리옥실릴 클로라이드(3-indoleglyoxylyl chloride)와 4-다이메틸아미노피리딘(4-dimethylaminopyridine)을 혼합하여 혼합 용액을 준비하는 단계; 및 ii) 상기 혼합 용액에 트립타민을 첨가하여 반응액을 제조하는 단계를 포함하는, 두룸아마이드 A의 제조 방법을 제공한다.
상기 i) 단계는 트립타민에 제공될 치환기로서 인돌-3-글리옥실레이트(indole-3-glyoxylate)를 합성하기 위해 3-인돌글리옥실릴 클로라이드와 4-다이메틸아미노피리딘을 이용하여 혼합용액을 준비하는 단계이다.
본 발명의 일 실시예에 따르면, 3-인돌글리옥실릴 클로라이드와 4-다이메틸아미노피리딘을 건조 피리딘에 용해하여 혼합용액을 준비하는 단계일 수 있다.
상기 ii) 단계는 상기 혼합용액에 트립타민을 첨가하여 반응액을 제조한 후 분획하여 두룸아마이드 A를 수득하는 단계이다.
상기 반응액은 트립타민과 인돌-3-글리옥실레이트가 반응하여 합성된 두룸아마이드 A를 포함하며, 미반응된 화합물 또한 포함한다.
본 발명의 일 구체예에 따르면, 상기 ii) 단계 이후에, iii) 상기 반응액을 디클로로메탄을 포함한 용매로 분획하여 디클로로메탄 분획물을 수득하는 단계를 더 포함하는 것일 수 있다.
상기 디클로로메탄을 포함한 용매는 디클로로메탄과 탈염 증류수를 포함하는 것일 수 있으나 이에 제한되지 않는다. 전술한 바와 같이, 본 발명에서 상기 분획물을 얻는 분획 방법은 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 수행될 수 있다. 본 발명에서는 상기의 분획방법을 단독으로 사용하거나, 2 종 이상 혼합하여 사용할 수 있고, 각각의 분획방법을 반복하여 사용할 수도 있다.
본 발명의 일 구체예에 따르면, 상기 iii) 단계 이후에, iv) 상기 디클로로메탄 분획물에 크로마토그래피를 수행하여 n-핵산/에틸아세테이트 (7:3, 5:5, 3:7) 및 순수 에틸아세테이트 순으로 계단식 용리하는 단계를 더 포함하는 것일 수 있다.
상기 크로마토그래피는 본 발명의 바람직한 일 실시예에 따르면 플래시 칼럼 크로마토그래피일 수 있다.
본 발명의 또 다른 양상은, i) DMSO에 트립타민을 용해하여 용해물을 준비하는 단계; 및 ii) 상기 용해물을 환류시켜 환류 추출물을 수득하는 단계를 포함하는, 두룸아마이드 A의 제조 방법을 제공한다.
상기 i) 단계는 트립타민 용해물을 준비하는 단계이다
상기 DMSO는 40 내지 90%, 50 내지 80% 또는 60 내지 70%, 바람직하게는 67% DMSO/물일 수 있다.
상기 ii) 단계는 환류 추출물을 수득하는 단계이다.
본 발명의 바람직한 일 실시예에 따르면, 상기 환류는 오일 배스를 이용한 것일 수 있다. 상기 환류는 80℃ 이상 또는 90℃ 이상, 바람직하게는 100℃에서 10시간 이상 또는 15시간 이상, 바람직하게는 20시간 수행될 수 있다.
본 발명의 일 구체예에 따르면, 상기 ii) 단계 이후에, iii) 상기 환류 추출물을 클로로포름으로 추출하여 클로로포름 추출물을 수득하는 단계를 더 포함하는 것일 수 있다.
상기 iii) 단계는 클로로포름 추출물을 수득하는 단계이다.
상기 클로로포름 추출물은 클로로포름을 포함한 유기용매로 추출한 결과물을 의미하며, 본 발명에서 상기 추출물을 얻는 방법은 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 수행될 수 있다. 본 발명의 바람직한 일 실시예에 따르면, 상기 환류 추출물을 클로로포름 (350 mL × 3)으로 추출하여 클로로포름 추출물 (1.1 g)을 제조한 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 iii) 단계 이후에 iv) 상기 클로로포름 추출물에 크로마토그래피를 수행하여 아세토니트릴로 용리하는 단계를 더 포함하는 것일 수 있다.
상기 iv) 단계는 크로마토그래피를 수행하여 두룸아마이드 A를 수득하는 단계이다. 상기 크로마토그래피는 고정상과 이동상을 이용하여 여러 가지 물질들이 섞여 있는 혼합물을 이동속도 차이에 따라 분리하는 방법이다. 크로마토그래피는 정지상의 특성에 따라 평면, 종이, 얇은막 크로마토그래피로 나뉘고, 이동상의 특성에 따라 기체 또는 액체 크로마토그래피로 나뉠 수 있다. 기타 친화성, 이온 교환, 크기 배제 또는 이차원 크로마토그래피 기법이 존재하며 본 발명에서의 크로마토그래피는 어느 한 기법에 제한되지 않는다.

이하 하나 이상의 구체예를 실시예를 통해 보다 상세하게 설명한다. 그러나, 이들 실시예는 하나 이상의 구체예를 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 코리네박테리움 두룸(
Corynebacterium durum
) 아세톤 추출물로부터 두룸아마이드 A(durumamide A) 분리
1-1. 코리네박테리움 두룸 배양
코리네박테리움 두룸을 BHI(brain heart infusion) 배지 (9 × 1 L)에서 4주간 37℃로 진탕 배양하여 배양물을 수득하였으며, 다공성 수지 XAD-7-HP (20 g/L)를 투입하고 상온에서 2시간 동안 흔들었다.
1-2. 아세톤 추출
상기 유기화합물들이 흡착된 수지는 여과하여 거른 다음 99.5% 아세톤으로 아세톤 추출물을 수득하였다 (14.7 g).
1-3. 유기용매로 분획
상기 아세톤 추출물은 클로로포름 (270 mL × 3)과 탈염 증류수 (270 mL)로 분획하였고, 유기용매 층에 용해된 유기물질 (1.6 g)은 지방성분 제거를 위하여 n-핵산 (15 mL × 3)과 아세토니트릴 (15 mL)로 다시 한 번 분획하여 분획물을 수득하였다.
1-4. 크로마토그래피 수행 및 두룸아마이드 A의 정제
상기 아세토니트릴층에 용해된 분획물로부터 C18 HPLC (4.6 × 250 mm, 1.0 mL/min)를 이용하여 크로마토그래피를 수행한 후, 0.1% 포름산이 포함된 5% 아세토니트릴/물을 이동상으로 하여 5분 동안 용리시킨 후, 기울기 용리로 15분간 100%까지 증가시켜 두룸아마이드 A (t
R 17.2 min)를 얻었다 (도 2).
실시예 2. 두룸아마이드 A의 합성 1
2-1. 3-인돌글리옥실릴 클로라이드(3-indoleglyoxylyl chloride)와 4-다이메틸아미노피리딘(4-dimethylaminopyridine, DMAP)의 혼합용액 준비
3-인돌글리옥실릴 클로라이드 (777 mg, 1.2 equiv)와 4-다이메틸아미노피리딘 (457 mg, 1.2 equiv)을 건조 피리딘(dry pyridine) (64 mL)에 용해하여 혼합용액을 준비하였다.
2-2. 트립타민(tryptamine) 첨가
상기 혼합용액에 트립타민 (500 mg, 1.0 equiv)을 투입하여 반응시켰다. 반응액을 상온에서 22시간 동안 교반시킨 반응물을 디클로로메탄(dichloromethane) (100 mL)과 탈염 증류수 (100 mL × 3)로 분획하여 디클로로메탄 분획물을 수득하였다.
2-3. 크로마토그래피 수행 및 두룸아마이드 A 정제
디클로로메탄 분획물 (902 mg)로부터 플래시 컬럼 크로마토그래피(flash column chromatography) (20 × 80 mm)를 이용하여 n-핵산/에틸아세테이트 (800 mL each of 7:3, 5:5, 3:7), 순수 에틸아세테이트 (800 mL) 순으로 계단식 용리하여 두룸아마이드 A (618 mg, 60%)를 정제하였다.
실시예 3. 두룸아마이드 A의 합성 2
3-1. 트립타민 용해물 준비
트립타민 (1.9 g)을 67% DMSO/물 (360 mL)에 용해하여 용해물을 준비하였다.
3-2. 환류 추출물 제조
상기 용해물을 오일 배스(oil bath)를 이용하여 100℃에서 교반하면서 20시간 환류하여 환류 추출물을 얻었다.
3-3. 클로로포름 추출물 제조
상기 환류 추출물을 클로로포름 (350 mL × 3)으로 추출하여 클로로포름 추출물을 제조하였다 (1.1 g).
3-4. 크로마토그래피 수행 및 두룸아마이드 A 정제
상기 클로로포름 추출물을 분취용 C18 HPLC (21.2 × 250 mm, 8 mL/min)를 이용하여 크로마토그래피를 수행한 후 45% 아세토니트릴/물을 이동상으로 하여 33분간 용리하여 두룸아마이드 A (t
R 27.0 min)를 얻었다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.

- a) 코리네박테리움 두룸(Corynebacterium durum)을 배양하여 배양물을 수득하는 단계;b) 상기 배양물을 아세톤으로 추출하여 아세톤 추출물을 수득하는 단계;c) 상기 아세톤 추출물을 유기용매로 분획하여 분획물을 수득하는 단계; 및d) 상기 분획물에 크로마토그래피를 수행하여 두룸아마이드 A를 수득하는 단계를 포함하는두룸아마이드 A의 제조 방법.
- 청구항 1에 있어서, 상기 유기용매는 메탄올, 에탄올, 부탄올, 아세톤, 에틸아세테이트, 에테르, 클로로포름, 펜탄, 헥산, 헵탄, 노난, 데칸, 벤젠, 톨루엔 및 자일렌으로 이루어진 군에서 선택되는 어느 하나인 것인,두룸아마이드 A의 제조 방법.
- 청구항 1에 있어서, 상기 c) 단계는c-1) 상기 아세톤 추출물을 클로로포름을 포함한 용매로 분획하여 클로로포름 분획물을 수득하는 단계; 및c-2) 상기 클로로포름 분획물을 아세토니트릴을 포함한 용매로 분획하여 아세토니트릴 분획물을 수득하는 단계를 포함하는 것인,두룸아마이드 A의 제조 방법.
- 청구항 3에 있어서, 상기 c-1) 단계의 클로로포름을 포함한 용매는 클로로포름과 물을 포함하는 것인,두룸아마이드 A의 제조 방법.
- 청구항 3에 있어서, 상기 c-2) 단계의 아세토니트릴을 포함한 용매는 아세토니트릴과 n-핵산을 포함하는 것인,두룸아마이드 A의 제조 방법.
- 청구항 1에 있어서, 상기 d) 단계의 크로마토그래피는 아세토니트릴/물을 이동상으로 하여 아세토니트릴 분획물로부터 두룸아마이드 A를 추출하는 것인,두룸아마이드 A의 제조 방법.
- i) 3-인돌글리옥실릴 클로라이드(3-indoleglyoxylyl chloride)와 4-다이메틸아미노피리딘(4-dimethylaminopyridine)을 혼합하여 혼합 용액을 준비하는 단계; 및ii) 상기 혼합 용액에 트립타민을 첨가하여 반응액을 제조하는 단계를 포함하는 두룸아마이드 A의 제조 방법.
- 청구항 7에 있어서,상기 ii) 단계 이후에,iii) 상기 반응액을 디클로로메탄을 포함한 용매로 분획하여 디클로로메탄 분획물을 수득하는 단계를 더 포함하는,두룸아마이드 A의 제조 방법.
- 청구항 8에 있어서,상기 iii) 단계 이후에,iv) 상기 디클로로메탄 분획물에 크로마토그래피를 수행하여 n-핵산/에틸아세테이트 (7:3, 5:5, 3:7) 및 순수 에틸아세테이트 순으로 계단식 용리하는 단계를 더 포함하는,두룸아마이드 A의 제조 방법.
- i) DMSO에 트립타민을 용해하여 용해물을 준비하는 단계; 및ii) 상기 용해물을 환류시켜 환류 추출물을 수득하는 단계를 포함하는,두룸아마이드 A의 제조 방법.
- 청구항 10에 있어서,상기 ii) 단계 이후에,iii) 상기 환류 추출물을 클로로포름으로 추출하여 클로로포름 추출물을 수득하는 단계를 더 포함하는,두룸아마이드 A의 제조 방법.
- 청구항 11에 있어서,상기 iii) 단계 이후에,iv) 상기 클로로포름 추출물에 크로마토그래피를 수행하여 아세토니트릴로 용리하는 단계를 더 포함하는,두룸아마이드 A의 제조 방법.
