Abstract
본 발명은 HSP90의 에피토프인 서열번호 1의 펩타이드 및 서열번호 2의 펩타이드를 포함하는 항암용 백신 조성물에 관한 것으로, 본 발명에 따른 백신 조성물은 종양 세포주 이식 동물모델에서 심각한 부작용 없이 종양의 성장을 효과적으로 억제할 수 있으므로, 암을 치료하거나 재발을 예방하는데 유용하게 사용될 수 있다.
Description
본 출원은 2021년 9월 29일자 한국 특허 출원 제10-2021-0128516호에 기초한 우선권의 이익을 주장하며, 해당 한국 특허 출원의 문헌에 개시된 모든 내용은 본 명세서의 일부로서 포함한다.
본 발명은 암 치료 및 재발 방지를 위한 HSP90 항원성 펩타이드를 포함하는 항암용 백신 조성물 및 이의 용도에 관한 것이다.
현재 암은 심장질환 등과 더불어 인류의 최대 질병 중의 하나로 대한민국 국민의 사망원인 중 1위이지만, 아직 발병 원인을 명확하게 알지 못하고 있다. 암을 치료하기 위해 수술 요법, 방사선 요법 및 화학 요법 등 다양한 치료 방법이 고안되었고, 암의 종류에 따라 각각의 치료 방법을 단독 또는 복합적으로 적용하고 있으나, 항암제에 대한 내성 획득 등으로 완치되지 않는 환자가 존재할 뿐 아니라, 경우에 따라서는 전이 및 재발률이 높아 치료에 어려움이 있다.
백신은 신체가 유해한 성분 및 질병에 걸린 세포를 인지하고 파괴하도록 면역계를 훈련시킴으로써 질환과 싸우는 것을 돕는다. 백신은 크게 2가지 유형, 즉 예방용 및 치료용 백신으로 분류된다. 예방 백신은 전통적인 백신의 형태로서, 건강한 사람들에게 특정 질환의 발생을 예방하도록 제공되는 반면, 면역요법으로도 불리는 치료용 백신은 질환이 있는 것으로 진단된 사람에게 질환이 성장하고 발전하는 것을 억제하거나, 예방하는 방법으로 제공된다. 치료용 백신의 대표적인 형태는 항암용 백신(또는 암 백신으로 불리기도 한다)이다.
항암용 백신은 세포독성 T 세포(cytotoxic T cell)를 활성화하여, 활성화된 T 세포가 특정 유형의 암 세포를 인지하여 이에 맞서 작용하거나, 암 세포 표면에 존재하는 항원과 결합하는 항체의 생산을 유도하도록 디자인된다. 항암용 백신의 기본적인 치료 목표는 암 환자에게 이미 존재하는 암 조직의 크기를 줄이거나, 암 환자의 암 조직을 외과적 수술 또는 화학요법 등으로 제거한 후, 재발을 억제하는 것이다.
지난 수년간 암 치료를 위해 또는 재발을 막기 위해 백신 연구가 광범위하게 이루어졌지만, 전립선암 백신인 시플루셀-T(sipuleucel-T, Provenge®)를 제외하고는 아직 성공적인 사례가 존재하지 않는다. 이러한 낮은 성공률에 대해서는 특정 유형의 종양 세포에 항원이 특이적으로 발현되더라도, 각 개인에 따라서는 낮은 수준으로 항원을 발현하거나, 전이성 종양의 경우 최초 발생 종양과 전이성 종양 간에 항원 프로파일이 변화될 수 있고, 종양세포의 면역회피 기전도 일부 낮은 성공률의 원인으로 설명하고 있다.
바람직한 백신 항원은 종양세포에서 배타적으로 또는 적어도 증가된 수준으로 발현되어야 한다. HSP90(Heat Shock Protein 90)는 많은 암종에서 과발현한다는 것이 알려져 있고, 실제 HSP90의 특정 항원성 영역(에피토프)으로 백신 접종을 하는 경우, 동물 모델에서 항종양 효과를 보인다는 것이 알려져 있다.
애주번트는 백신용 조성물에 항원과 함께 포함되는 성분으로 항원에 반응하는 면역반응을 조절하거나 증강시키는 용도로 개발되어 왔다. 애주번트는 단독으로 투여되었을 때에는 그 자체로 면역성을 가지고 있지 않지만, 항원과 같이 투여되는 경우 항원에 대한 면역반응을 증진시키고 지속시킬 수 있다. 전형적인 애주번트의 예는 포타슘 알룸(potassium alum), 수산화알루미늄(aluminium hydroxide) 등의 알루미늄 계열의 무기물, 파라핀 오일 등의 오일류, 사포닌(saponin)의 계면활성제, 프로인드 보강제(Freund's complete adjuvant/Freund's incomplete adjuvant; CFA/IFA) 등이 있다.
최근에는 톨 유사 수용체(Toll-like receptor; TLR)가 활성화되는 경우 선천성 면역계(innate immune system)가 활성화된다는 것이 알려짐에 따라 상기 TLR을 활성화시키는 TLR 아고니스트(agonist)를 애주번트로 이용하려는 시도가 있어왔다. TLR은 세포막 통과 수용체(Transmembrane receptor) 단백질로서, 선천성 면역세포(innate immune cell)에서 주로 발현되는 것으로 알려져 있다. TLR의 서브타입으로 TLR1, TLR2, TRL4, TLR5, TLR6는 세포 표면에서 발현하고, TLR3, TLR7, TLR8, TLR9은 세포 내 엔도좀의 세포막(endosomal membrane)에서 발현하는 것으로 알려져 있다.
최근 최적화된 면역반응을 나타내는 백신 제조를 위해 항원과 알루미늄 기반 애주번트의 최적화된 조합을 도출하기 위한 연구가 시도되고 있다. 하지만, HSP90 항원성 펩타이드를 이용한 암백신에 있어서, 최적화된 애주번트에 대해서는 확인한 바 없다.

이에 본 발명자들은 상기 문제를 해결하고자 연구를 수행한 결과, HSP90 항원성 펩타이드 및 TLR 아고니스트를 애주번트로 포함하는 항암용 백신 조성물이 암세포를 이식한 종양 마우스 모델에서 종양 성장을 효과적으로 억제할 수 있음을 확인하여 본 발명을 완성하였다.
본 발명은 서열번호 1의 아미노산 서열을 포함하는 펩타이드 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드로 구성된 군에서 선택된 하나 이상의 펩타이드; 및 TLR2 아고니스트(agonist) 및 TLR3 아고니스트(agonist)를 애주번트로 포함하는, 항암용 백신 조성물을 제공하는 것을 목적으로 한다.
상기 TLR2 아고니스트(agonist)는 Pam3Cys-SKKKK, PHC-SKKKK, Ole2PamCys-SKKKK, Pam2Cys-SKKKK, PamCys(Pam)-SKKKK, Ole2Cys-SKKKK, Myr2Cys-SKKKK, PamDhc-SKKKK, PamCSKKKK, Dhc-SKKKK 중에서 선택되는 하나 이상인 것일 수 있다.
상기 TLR2 아고니스트(agonist)는 Pam3Cys-SKKKK일 수 있다.

본원에서, 어떤 부분이 어떤 구성 요소를 “포함”한다고 할 때, 이는 특별히 반대되는 기재가 없는 한 다른 구성 요소를 제외하는 것이 아니라 다른 구성 요소를 더 포함할 수 있는 것을 의미한다.

용어 "환자" 또는 "대상체"란, 본 발명의 백신 조성물의 투여에 의해 질환이 예방 또는 치료될 수 있는 병태, 예컨대 암, 구체적으로 고형암을 앓고 있거나, 치료되었거나, 그러한 병태에 걸리기 쉬운, 살아있는 유기체를 지칭하며, 인간과 동물을 둘 다 포함한다. 대상체로서는 포유동물(예컨대, 마우스, 원숭이, 말, 소, 돼지, 개, 고양이 등)을 포함하나, 이에 한정되는 것은 아니며, 바람직하게는 인간이다. 또한, 본 발명에서의 "환자" 또는 "대상체"는 구분하지 않고 암 환자, 고형암 환자, 유방암 환자 또는 HER2를 발현하는 유방암 환자를 포함한다.
용어 "투여하는 것"이란, 이 발명이 속하는 기술분야에서 공지된 다양한 방법과 전달 시스템 중 임의의 것을 사용하여 백신 조성물을 대상체에 도입하는 것을 지칭한다. 예시적 투여 경로는 정맥내, 근육내, 피하, 복강내, 척수 또는 기타 다른 비경구 투여 경로, 예컨대 주사 또는 주입에 의한 투여 경로를 포함한다. 본 발명의 명세서에 사용된 "투여"란, 일반적으로 주사에 의한 피하(subcutaneous) 또는 피내(intradermal) 투여 방식을 의미하고, 정맥내, 근육내, 동맥내, 척수강내, 또는 림프내 주사 및 주입을 포함하나, 이에 한정되는 것은 아니다. 기타 다른 비경구 경로는 국소, 상피 또는 점막 투여 경로, 예컨대 비내, 질, 직장, 또는 설하 경로를 포함한다. 본 명세서에서는 대상체에 백신을 투여하는 행위인 "백신 투여(vaccine injection)", 처치(treatment)"와 "접종(vaccination)"은 구분되지 않고 사용된다.
용어 "항원성 펩타이드(antigenic peptide)"는 특정 항체에 의해 인식되는 항원결합부위의 아미노산 잔기 세트, 또는 T 세포에서의 T 세포 수용체(T cell receptor; TCR) 및/또는 주요 조직적합성 복합체(Major Histocompatibility Complex; MHC) 수용체에 의해 인식되는 에피토프(epitope)를 의미한다.
본 발명에서, 용어 "면역 항암제"는 암 자체를 공격하는 기존 항암제와는 달리 인공면역 단백질을 체내에 주입하여 면역체계를 자극함으로써 면역세포가 선택적으로 암세포만을 공격하도록 유도하는 치료약제로서, 암세포가 획득한 면역억제 또는 면역회피 기전을 극복하기 위하여 면역체계의 종양 인지능력 또는 파괴능력을 회복 또는 강화시키는 기전의 항암제이다. 상기 면역항암제는 면역관문 억제제, 면역세포 치료제 및 면역바이러스 치료제를 포함하지만 이에 제한되지는 않는다.
본 발명에서, 용어 "면역관문 억제제"는 일부 암세포가 체내 면역세포인 T 세포의 면역체크포인트를 활용하면 서 면역을 회피하는 경우에, T 세포 억제에 관여하는 면역체크포인트 단백질(Immune Checkpoint Protein)의 활성화를 차단하여 T 세포를 활성화시켜 암세포를 공격하는 작용을 하는 면역항암제의 일종으로서, CTLA-4 억제제, PD-1 억제제, LAG-3 억제제, TIM-3 억제제, TIGIT 억제제 및 VISTA 억제제를 포함하지만 이에 제한되지는 않는다. 또한, 상기 면역관문 억제제는 길항제로서, 길항제 항체 또는 소분자 화합물일 수 있다.
열 충격 단백질 90(heat shock protein 90: HSP90)은 세포 내에서 합성되는 모든 펩타이드 사슬이 온전한 구조 형성을 위해 단백질 접힘, 성숙 및 입체 형태 안정화를 위해 작용하는 샤페론(chaperone) 단백질의 일종이다. HSP90 단백질 또는 유전자의 발현양은 악성 종양 세포(malignant cell)에서 정상세포에 비해 2~10배 정도 높아, HSP90는 암 세포의 생존 및 성장에 주요한 역할을 할 것으로 예측되어 효과적인 약물 타겟으로 대두되고 있다.
또한, 기존 연구에 의해서도 유방암 환자군의 serum에서 대조군(normal individual + 양성 유방암 환자)에 비해 HSP90에 대한 자가 항체(auto-antibody, 항-HSP 항체)의 농도가 높다는 것이 보고된 바 있다(Lancet, 1995: 345, 126).
본 발명의 항암용 백신 조성물은 서열번호 1의 아미노산 서열을 포함하는 펩타이드(제1 펩타이드) 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드(제2 펩타이드)로 구성된 군에서 선택된 하나 이상의 펩타이드; 및 TLR2 아고니스트(agonist) 및 TLR3 아고니스트(agonist)를 애주번트로 포함하는 것일 수 있다.
일 구현예에 따르면, 상기 항암용 백신 조성물은 서열번호 1의 아미노산 서열을 포함하는 펩타이드(제1 펩타이드) 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드(제2 펩타이드); 및 TLR2 아고니스트(agonist) 및 TLR3 아고니스트(agonist)를 애주번트로 포함하는 것일 수 있다.
상기 제1 펩타이드는 서열번호 1의 아미노산 서열을 포함하는 펩타이드일 수 있으며, 구체적으로 서열번호 1의 아미노산 서열로 구성된 펩타이드 일 수 있다. 또한, 상기 제2 펩타이드는 서열번호 2의 아미노산 서열을 포함하는 펩타이드일 수 있으며, 구체적으로 서열번호 2의 아미노산 서열로 구성된 펩타이드 일 수 있다.
본 발명의 서열번호 1의 아미노산 서열로 표시되는 펩타이드 및 서열번호 2의 아미노산 서열로 표시되는 펩타이드의 HSP 90 항원성 펩타이드는 HSP90 풀 시퀀스를 5개의 알고리즘 프로그램을 이용하여 MHC class II 대립유전자(allele)에 결합친화도(binding affinity)가 좋을 것으로 예상되는 15-mer 펩타이드 시퀀스 12개 중 선정된 것이다(대한민국 등록특허 제10-2256267호).
본 발명에의 명세서 기재된 "서열번호 X의 아미노산 서열로 표시되는 펩타이드", "서열번호 X의 아미노산 서열로 구성된 펩타이드" 및 "서열번호 X의 펩타이드"는 같은 의미로서 구분되지 않고 사용된다.
본 발명의 일부 실시예에서는 서열번호 1의 아미노산 서열로 표시되는 펩타이드 (LYVRRVFIMDNCEEL) 및 서열번호 2의 아미노산 서열로 표시되는 펩타이드(PyroGlu-SKILKVIRKNLVKK) 각각 화학적으로 합성하여 사용할 수 있다.
본 발명의 백신 조성물은 상기 서열번호 1의 아미노산 서열을 포함하는 펩타이드(제1 펩타이드) 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드(제2 펩타이드)를 각각 별개로 분리된 펩타이드의 형태로 포함하거나 또는 서로 직접적 또는 간접적으로 연결된 형태로 포함하는 것일 수 있다. 상기 제1 펩타이드 및 제2 펩타이드가 서로 직접적 또는 간접적으로 연결된 형태로 포함되는 경우, 상기 제2 펩타이드는 제1 펩타이드의 N-말단 또는 C-말단에 직접적으로 또는 링커를 통해 간접적으로 연결될 수 있다. 상기 링커는 상기 펩타이드가 항암 백신 활성을 나타나게 하는 한 특별히 이에 제한되지 않으며, 예를 들어 1 내지 50개의 아미노산으로 구성된 것일 수 있다.
본 명세서에 개시된 백신 조성물은 펩타이드 백신, DNA 백신 또는 mRNA 백신으로 제조될 수 있다.
본 발명의 일 실시예에서, 상기 항암용 백신 조성물은 서열번호 1의 아미노산 서열을 포함하는 펩타이드(제1 펩타이드)의 암호화 서열을 포함하는 핵산 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드(제2 펩타이드)의 암호화 서열을 포함하는 핵산으로 구성된 군에서 선택된 하나 이상의 핵산; 및 TLR2 아고니스트(agonist) 및 TLR3 아고니스트(agonist)를 애주번트로 포함하는 것일 수 있다.
일 구현예에 따르면, 상기 항암용 백신 조성물은 서열번호 1의 아미노산 서열을 포함하는 펩타이드(제1 펩타이드)의 암호화 서열을 포함하는 핵산 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드(제2 펩타이드)의 암호화 서열을 포함하는 핵산; 및 TLR2 아고니스트(agonist) 및 TLR3 아고니스트(agonist)를 애주번트로 포함하는 것일 수 있다.
상기 제1 펩타이드의 암호화 서열을 포함하는 핵산은 제1 핵산으로 명명할 수 있고, 상기 제1 핵산은 제1 펩타이드의 암호화 서열을 포함하는 핵산일 수 있으며, 구체적으로 제1 펩타이드의 암호화 서열로 구성된 핵산일 수 있다. 또한, 상기 제2 펩타이드의 암호화 서열을 포함하는 핵산은 제2 핵산으로 명명할 수 있고, 상기 제2 핵산은 제2 펩타이드의 암호화 서열을 포함하는 핵산일 수 있으며, 구체적으로 제2 펩타이드의 암호화 서열로 구성된 핵산일 수 있다.
상기 핵산은 DNA 또는 RNA(mRNA)일 수 있다.
본 발명의 일부 실시예에서, 상기 항암용 백신 조성물이 DNA 백신의 형태로 제조될 경우, 예를 들어 상기 DNA 백신은 프로모터(promoter), 적절한 전사 및 번역 제어 요소 및 본 발명의 서열번호 1의 아미노산 서열을 포함하는 펩타이드의 암호화 서열을 포함하는 핵산 서열(제1 핵산) 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드의 암호화 서열을 포함하는 핵산 서열(제2 핵산) 중 하나 이상을 갖는 하나 이상의 DNA 분자(예를 들어, 플라스미드)를 포함할 수 있으며, 상기 플라스미드는 추가적으로 증강(enhance) 서열, 예를 들어, 발현 수준, 세포 내 표적화 등을 증강하는 서열을 포함할 수 있다.
또한, 상기 DNA 백신에서 제1 핵산 및 제2 핵산은 각각 별개로 분리된 플라스미드에 포함되는 형태이거나, 서로 직접적 또는 간접적으로 연결되어 하나의 플라스미드에 포함되는 형태일 수 있다. 상기 제1 핵산 및 제2 핵산이 서로 연결되어 하나의 플라스미드에 포함되는 형태인 경우, 상기 제2 핵산은 제1 핵산의 5'-말단 또는 3'-말단에 직접적으로 또는 링커를 통해 간접적으로 연결될 수 있다. 상기 링커는 상기 핵산이 암호화하는 펩타이드가 항암 백신 활성을 나타나게 하는 한 특별히 이에 제한되지 않으며, 예를 들어 1 내지 150개의 뉴클레오타이드로 구성된 것일 수 있다. 또한, 제1 핵산 및 제2 핵산이 하나의 플라스미드에 포함되는 형태일 경우, 이로부터 번역되는 제1 펩타이드 및 제2 펩타이드는 각각 별개의 분리된 펩타이드로 번역되거나, 제1 펩타이드 및 제2 펩타이드가 서로 직접적 또는 간접적으로 연결된 형태로 번역될 수 있다.
본 발명의 일부 실시예에서, 상기 항암용 백신 조성물이 mRNA 백신의 형태로 제조될 경우, 예를 들어 상기 mRNA 백신은 본 발명의 서열번호 1의 아미노산 서열을 포함하는 펩타이드의 암호화 서열을 포함하는 핵산 서열(제1 핵산) 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드의 암호화 서열을 포함하는 핵산 서열(제2 핵산) 중 하나 이상을 갖는 하나 이상의 mRNA 분자를 포함할 수 있으며, 상기 mRNA 분자는 5'-캡(cap), 5'-비번역 부위(untranslated region, UTR), 3'-비번역 부위 및 폴리(A) 꼬리(Poly(A) tail)로 구성된 군에서 선택된 하나 이상을 추가로 포함할 수 있다.

본 발명의 백신 조성물이 DNA 백신 또는 mRNA 백신일 경우, 상기 백신은 A(Adenine), T (Thymine), G(Guanine), C(Cytosine), U(Uridine) 및/또는 이들의 변형된 형태를 포함하는 핵산으로 구성된 것일 수 있다. 예를 들어 상기 변형된 형태는 5-메틸시티딘(5mC), N6-메틸아데노신(m6A), 3,2'-O-디메틸우리딘(m4U), 2-티오우리딘(s2U), 2' 플루오로우리딘, 슈도우리딘, 2'-O-메틸우리딘(Um), 2' 데옥시 우리딘(2' dU), 4-티오우리딘(s4U), 5-메틸우리딘(m5U), 2'-O-메틸아데노신(m6A), N6,2'-O-디메틸아데노신(m6Am), N6,N6,2'-O-트리메틸아데노신(m62Am), 2'-O-메틸시티딘(Cm), 7-메틸구아노신(m7G), 2'-O-메틸구아노신(Gm), N2,7-디메틸구아노신(m-2,7G), N2,N2,7-트리메틸구아노신(m-2,2,7G) 및 N1-메틸-슈도우리딘으로 구성된 군에서 선택된 하나 이상을 포함하는 것일 수 있다.
본 발명의 백신 조성물은 전술한 항원성 펩타이드(서열번호 1 및 서열번호 2의 아미노산 서열을 포함하는 펩타이드)뿐 아니라, TLR2 아고니스트 및 TLR3 아고니스트를 포함하는 애주번트(대한민국 등록특허 제10-1501583호)를 포함한다.
애주번트는 일반적으로 백신 항원과 조합되었을 때, 항원만으로 유도된 면역반응에 비해 백신 항원에 대한 면역반응을 증가시키는 임의의 화합물 또는 물질이다. 애주번트는 체액성 면역반응 또는 세포성 면역반응 또는 이들 두 가지 면역반응의 수위를 증가시키는 기능을 하면서, 추가적으로 이들 반응을 지속시키는 기능을 한다.
일반적으로 사용되는 애주번트는 포타슘 알룸(potassium alum), 수산화알루미늄(aluminium hydroxide) 등의 알루미늄 계열의 무기물, 파라핀 오일 등의 오일류, 사포닌(saponin) 계열의 계면활성제류, 프로인드 보강제(Freund's complete adjuvant/Freund's incomplete adjuvant) 등이 있고, 최근 연구가 진행중인 톨 유사 수용체(Toll like receptor; TLR)를 활성화시키는 리간드인 TLR 아고니스트가 이용되고 있다.
TLR2 아고니스트는 박테리아와 마이코플라즈마에서 유래한 리포펩티드 합성 유사체로서, am3Cys-SKKKK, PHC-SKKKK, Ole2PamCys-SKKKK, Pam2Cys-SKKKK, PamCys(Pam)-SKKKK, Ole2Cys-SKKKK, Myr2Cys-SKKKK, PamDhc-SKKKK, PamCSKKKK, Dhc-SKKKK 등 다양한 리포펩티드 유사체들이 합성되었다. 또한, 상기 리포펩티드들의 지방산 및 아미노산은 화학적으로 변형될 수 있다. 본 발명에서는 "TLR2 아고니스트" 및 "TLR2L"는 구분되지 않고 혼용되어 사용된다.
본 발명의 일실시예에서는 백신 조성물에 상기 TLR2 아고니스트로 Pam3Cys-SKKKK를 포함하지만, 이에 제한되지 않는다.
TLR3 아고니스트는 폴리 (I:C) (폴리 Polyinosinic-polycytidylic acid)는 이중가닥 RNA의 합성 유사체로서, 시험관 내(in vitro) 또는 생체 내(in vivo) 연구에서 Th1 면역반응을 유도하는 것으로 알려져 있다. 더욱이 폴리 (I:C)는 항원 제시 세포(Antigen Presenting Cell; APC)인 수지상세포(Dendritic Cell; DC)를 안정적으로 성숙시키는 것으로 알려져 있다. 본 발명에서는 "TLR3 아고니스트" 및 "TLR3L"는 구분되지 않고 혼용되어 사용된다.
본 발명의 일실시예에서는 백신 조성물에 상기 TLR3 아고니스트로 그 길이가 50 내지 2,000bp인 폴리 (I:C)를 포함하지만, 이에 제한되지 않는다.
종래 연구에서는 리포펩티드(lipopeptide) 유래인 TLR2 아고니스트와 리포좀을 혼합한 후 TLR3 아고니스트를 혼합한 애주번트가 Th1 면역반응을 증진시킨다는 결과가 보고된 바 있다(대한민국 등록특허 제10-2098097호).
리포좀은 인지질 이중 층의 matrix 형태를 갖추고 있는 세포막과 가장 유사한 형태의 단층 혹은 다층의 지질-이중 막 구조를 가진다. 상기 지질은 양이온, 음이온 또는 중성 지질일 수 있다. 일례로, 상기 지질은 DOTAP(1,2-Dioleoyl-3-Trimethylammonium-Propane), DOPE(1,2-dioleoyl-sn-glycero-3- phosphoethanolamine), DDA(Dimethyldioctadecylammonium), DC-chol(3β-[N-(N',N'-Dimethylaminoethane)- carbamoyl]cholesterol), DOPG(1,2-Dioleoyl-sn-Glycero-3-[Phospho-rac-(1-glycerol)]), DPPC(1,2- dipalmitoyl-sn-glycero-3-phosphocholine), DOPC(1,2-dioleoyl-sn-glycero-3-phosphocholine) 및/또는 콜레스테롤을 포함한다.
리포좀은 구조적 특성으로 친수성이나 소수성 물질 모두를 전달하는데 효과적이며, 선택적으로 사용할 수 있다는 장점이 있다. 따라서, 천천히 분해되는 동안 서서히 약물이 방출되므로, 백신의 약물 전달체로서 이용되고 있고, 약물의 양을 줄일 수 있다는 장점이 있으므로, 백신 조성물 제조시 필수적인 고려사항 중 하나이다.
하지만, 본 발명의 백신 조성물은 전술한 HSP90 항원성 펩타이드에 애주번트로 TLR2 아고니스트 및 TLR3 아고니스트를 모두 포함하는 경우 TLR2 아고니스트(TLR2L) 또는 TLR3 아고니스트(TLR3L) 단독으로 포함하는 경우보다 더 높은 수준의 Th1 면역반응이 일어나는 것을 확인하였을 뿐 아니라, 놀랍게도 애주번트로 리포좀을 더 포함하는 경우 증가한 Th1 면역반응은 거의 없어지는 것을 확인할 수 있었다.
따라서, 종래 TLR2/3 아고니스트(TLR2 아고니스트 및 TLR3 아고니스트) 및 리포좀을 혼합하여 애주번트로 사용하는 경우, TLR2/3 아고니스트에 비해 면역 효과를 증가된다고 알려져 있었으나, 본 발명의 백신 조성물에서 리포좀은 상승효과가 아닌 부정적인 역할을 한다는 것을 알 수 있다.
위 결과를 감안하면, 본 발명의 일실시예의 백신 조성물은 리포좀을 포함하지 않는 것이 바람직하다.
본 발명에서는 "TLR2 아고니스트 및 TLR3 아고니스트"및 "TLR2/3L"는 구분되지 않고 혼용되어 사용된다.
일 실시예에 따르면, 본 발명의 HSP90 항원성 펩타이드인 서열번호 1 및 서열번호 2의 펩타이드는 다른 종류의 TLR 아고니스트인 TLR4 아고니스트(TLR4) 또는 TLR7 아고니스트 및 TLR8 아고니스트(TLR7/8L)를 애주번트로 사용하는 경우 Th1 면역반응을 거의 유도하지 못하는 반면, 앞서 상승효과를 확인한 TLR2 아고니스트 및 TRL3 아고니트스(TLR2/3L)에서 유의하게 Th1 면역반응을 유도하는 것을 확인하였다. 이러한 면역반응은 마우스 종과는 관계없는 것으로 확인하였다.
또한, 본 발명의 백신 조성물은 종양 이식 모델에서 종양의 성장을 효과적으로 억제하는 결과를 보였다. 이러한 결과는 본 발명의 백신 조성물을 투여받은 대상체(마우스) 내에서 이식된 암 세포에 대한 세포독성 T 세포 반응을 유도 또는 유지하여 이식된 암 세포의 증식을 억제한 것으로 볼 수 있다.
따라서, 본 발명의 백신 조성물은 종양의 성장 또는 재발을 효과적으로 억제할 수 있는 항암용 백신으로 사용될 수 있다.
본 발명의 백신 조성물은 암 성장 또는 재발에 대해 치료 효과를 유도 및/또는 유지하는 것이 목적이므로, 본 발명에서는 HSP90에 대한 세포독성 T 세포 반응을 유도 및/또는 유지하기에 적합한 임의의 스케줄로 본 발명의 백신 조성물을 환자에게 투여할 수 있다. 예를 들어, 일차 접종으로서 본 명세서에서 기술되고 예시되는 바와 같은 백신 조성물을 일정 스케줄로 환자에게 투여한 후, 면역을 유도 및/또는 유지하기 위해 부스터를 투여할 수 있다.
본 발명의 일부 실시예에서, 백신 조성물은 1개월 당 1회, 2회 또는 그 이상의 횟수로 환자에게 투여될 수 있다. 부스터는 접종 스케줄을 마친 후 일정 간격으로, 예를 들어 1개월의 간격으로 단수 또는 복수 회 투여될 수 있고, 6개월 이상의 기간을 가지고 단수 또는 복수 회 투여될 수 있다. 본 발명의 일부 실시예에서 일차 접종으로 매달 1회씩 3회 백신 조성물을 투여할 수 있고, 6개월 또는 1년 후 등의 임의의 기간이 경과 후에 추가적인 부스터를 접종할 수 있다.
이때 부스터는 일차 접종시 사용한 백신 조성물과 동일한 용량 및 구성일 수도 있고, 용량 및/또는 구성이 변경될 수도 있다.
본 발명의 명세서에 기재된 백신 조성물은 HSP90 항원성 펩타이드 및 TLR2/3 아고니스트 외에 약학적으로 허용되는 부형제(excipient), 담체, 희석제, 버퍼(buffer), 안정제(stabilizer), 보존제, 또는 이 발명이 속하는 기술분야에 잘 알려진 기타 물질을 포함할 수 있다. 하지만, 별도로 리포좀은 포함하지 않는다. 이러한 물질은 비독성이고 유효 성분(들)의 약학적 활성을 방해하지 않는다. 약제학적 담체 또는 희석제는 예를 들어 수용액일 수 있다. 본 발명의 백신 조성물에 포함될 수 있는 담체 또는 다른 물질은 투여 경로(예를 들어 경구, 정맥, 피부 또는 피하, 코, 근육, 피내 및 복강 투여)에 따라 다르게 선택될 수 있다.
본 발명의 백신 조성물은 하나 또는 그 이상의 "약제학적으로 허용되는 담체"를 포함할 수 있다. 이들은 전형적으로 단백질, 당류(saccharides), 폴리락트산(polylactic acids), 폴리글리콜산(polyglycolic acids), 폴 리머릭(polymeric) 아미노산, 아미노산 공중합체, 수크로오스(sucrose), 트레할로스(trehalose) 등이 속한다. 이러한 담체는 이 발명이 속하는 기술분야에 잘 알려져 있다. 백신 조성물은 또한 물, 식염수, 글리세롤 등과 같은 희석제를 함유할 수 있다. 또한, 습윤제 또는 유화제, pH 버퍼 물질 등과 같은 보조 물질이 존재할 수 있다. 무균 발열원이 없는 인산염 버퍼 생리식염수(PBS), 트로메타민(트리스)이 대표적인 담체이다.
본 발명의 백신 조성물에는 면역 조절제가 추가로 포함될 수 있다. 상기 면역조절제로 사용하기 적합한 사이토카인은 인터루킨(interleukin)(예를 들어, IL-1, IL-2, IL-4, IL-5, IL-6, IL-7, IL-12, 등), 대식세포 콜로니 자극 인자(macrophage colony stimulating factor, M-CSF), 종양 괴사 인자(tumour necrosis factor, TNF), 과립구 대식세포 콜로니 자극 인자(granulocyte, macrophage colony stimulating factor, GM-CSF) 등에서 선택될 수 있지만, 이에 제한되지 않는다.
본 발명의 백신 조성물은 화학 항암제, 표적 항암제 및 면역 항암제 중 선택되는 하나 이상의 항암제와 병용투여할 수 있다. 본 발명의 백신 조성물과 병용투여될 수 있는 화학 항암제, 표적 항암제 및 면역 항암제 중 선택되는 하나 이상의 항암제는 상호 동시에 또는 시간 차를 두고 순차적으로 투여될 수 있으며, 적절한 시간 및 주기에 따라 선택될 수 있다.
본 발명에 있어서, 화학 항암제는 세포독성항암제, 화학약물항암제로도 불린다. 상기 화학항암제는 예를 들면, 젬시타빈(Gemcitabine)(제시타빈주), 사이타라빈(Cytarabine), 카르보플라틴(Carboplatin)(파라플라틴), 시스플라틴(Cisplatin)(플라티놀, 플라티놀-AQ), 크리조티닙(Crizotinib)(잘코리), 시클로포스파미드(Cyclophosphamide)(시톡산, 네오사르), 도세탁셀(Docetaxel)(탁소티어), 독소루비신(Doxorubicin)(아드리아마이신), 에토포사이드(Etoposide)(베페시드), 플루오로우라실(5-fluorouracil, 5-FU), 이리노테칸(irinotecan)(캄토사르), 리포좀-캡슐화된 독소루비신(독실), 메토트렉세이트(Methotrexate)(폴렉스, 멕세이트, 아메토프테린), 파클리탁셀(Paclitaxel)(탁솔, 아브락산), 토포테칸(Topotecan)(하이캄틴), 트라벡티딘(Trabectedin)(욘델리스), 빈크리스틴(Vincristine)(온코빈, 빈카사르 PFS) 또는 빈블라스틴(Vinbrastine)(벨반), 카페시타빈(capecitabine) 또는 옥살리플라틴(oxaliplatin) 등이 있지만, 이에 제한되지 않는다.
본 발명에 있어서, 표적 항암제는 소분자 표적 항암제, 항체 표적 항암제, 및 항체-약물 결합물질인 ADC(Antibody-drug conjugate) 중 선택되는 것일 수 있다.
상기 소분자 표적 항암제는 트라메티닙(Trametinib), 베무라페닙(vemurafenib), 알페리십(alpelisib), 닥톨리십(dactolisib), 게피티닙(Gefitinib), 엘로티닙(Erlotinib), 라파티닙(Lapatinib), 수니티닙(Sunitinib), 소라페닙(Sorafenib), 크리조티닙(Crizotinib), 다브라페닙(Dabrafenib), 트라스투주맙(Trastuzumab), 세툭시맙(Cetuximab), 베바시주맙(Bevacizumab), 파니투무맙(Panitumumab), 이필리무맙(Ipilimumab), 퍼투주맙(Pertuzumab), 토파시티닙(Tofacitinib), 이마티닙(Imatinib), 및 보르테조밉(Bortezomib)으로 이루어진 군으로부터 선택되는 하나 이상일 수 있지만, 이에 제한되지 않는다.

본 발명에 있어서, 상기 면역 항암제는 CTLA-4(cytotoxic T-lymphocyte-associated protein 4), PD-1(Programmed cell death protein 1), PD-L1(Programmed death-ligand 1), LAG-3(Lymphocyte Activation Gene-3), TIM-3(T-cell Immunoglobulin and Mucin-domain containing-3), TIGIT(T-cell Immunoreptor with IG and ITIM domain) 및 VISTA(V-domain Ig Suppressor of T cell Activation)로 구성된 군으로부터 선택된 어느 하나에 대한 면역관문 억제제(immune checkpoint inhibitor)일 수 있으나, 이에 제한되지 않는다.
본 발명에 있어서, 상기 조성물은 항암 보조제를 추가로 포함할 수 있고, 보다 바람직하게는 상기 항암 보조제는 STING(STimulator of InterferoN Gene) 아고니스트일 수 있으나 이에 제한되지는 않는다.

본 발명에 있어서, 상기 암은 예컨대 편평세포 암(squamous cell cancer, 예를 들어, 상피의 편평세포 암), 소형 세포 폐암, 비-소형 세포 폐암, 폐암, 복막암, 결장암, 담도 종양, 비인두암, 후두암, 기관지암, 구강암, 골 육종, 담낭암, 신장암, 백혈병, 방광암, 흑색종, 뇌암, 신경 교종, 뇌종양, 피부암, 췌장암, 유방암, 간암, 골수암, 식도암, 대장암, 위암, 자궁경부암, 전립선암, 난소암, 두경부암 및 직장암으로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 할 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 백신 조성물은 동결건조되거나 수성 형태(aqueous form)일 수 있다. 동결건조된 백신 조성물의 경우 환자 투여 전 모든 성분을 적절한 수성 매질(aqueous medium)에서 재구성(reconstitution)한다. 수성 형태의 백신 조성물은 수용액 또는 현탁액일 수 있다. 이러한 유형의 액체 제제(Liquid formulations)는 조성물이 수성 매질에서 재구성할 필요없이 포장된 형태로 직접 투여할 수 있으므로 주사에 이상적이다. 백신조성물은 바이알(vials)로 제공되거나, 미리 충진된 주사기로 제공될 수 있다. 주사기는 바늘과 함께 또는 바늘 없이 공급될 수 있다. 주사기는 단일 용량을 포함하는 반면, 바이알은 단일 용량 또는 다수의(multiple) 용량을 포함할 수 있다. 또한, 본 발명의 백신 조성물은 마이크로니들을 이용하여 투여될 수 있다.
본 발명의 백신 조성물은 다수의(multiple) 투여 포맷(multiple dose format)으로 포장될 때 항균제(antimicrobial)를 포함할 수 있다. 항균제로는 2-페녹시에탄올(2-phenoxyethanol) 또는 파라벤(parabens)(메틸, 에틸, 프로필 파라벤) 등에서 선택될 수 있다. 임의의 보존제는 바람직하게는 낮은 수준으로 존재한다. 보존제는 외인성으로 첨가될 수 있고 및/또는 조성물을 형성하기 위해 혼합되는 벌크 항원의 성분일 수 있다.
본 발명의 백신 조성물은 설명서 등을 포함하는 키트 형태로 제공될 수 있다.
본 발명에서 유효성분과 결합하여 사용된 "치료학적으로 유효한 양"이란 용어는 대상 질환을 예방 또는 치료하 는데 유효한 양을 의미하며, 본 발명의 조성물의 치료적으로 유효한 양은 여러 요소, 예를 들면 투여방법, 목적 부위, 환자의 상태 등에 따라 달라질 수 있다. 따라서, 인체에 사용 시 투여량은 안전성 및 효율성을 함께 고려하여 적정량으로 결정되어야 한다. 동물실험을 통해 결정한 유효량으로부터 인간에 사용되는 양을 추정하는 것도 가능하다.
달리 나타내지 않는 한, 본 명세서 및 청구범위에 사용된 모든 숫자는, 언급되었든지 아니든지, 모든 경우에 용어 "약"에 의해 수식될 수 있는 것으로 이해되어야 한다. 또한, 본 명세서 및 특허청구범위에 사용된 정밀한 수치는 본 개시내용의 추가적인 실시양태를 형성하는 것으로 이해되어야 한다. 실시예에 개시된 수치의 정확성을 보장하기 위해 노력을 기울였다. 그러나, 측정된 모든 수치는 내재적으로 이의 각각의 측정 기법에서 실측된 표준 편차로부터 생성된 특정 오차값을 함유할 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
HSP90 항원성 펩타이드의 면역원성 측정 및 항암 효과를 분석하기 위한 동물은 C57BL/6 마우스 (암컷, 7주령), FVB 마우스 (암컷, 6주령) 및 MMTVneu 형질전환 마우스(암컷, 6주령)을 실험의 목적에 따라 선택하였고, 각 군당 4마리씩 사용하였다.
실시예 1의 동물시험은 차의과학대학교 실험동물운영위원회 (IACUC, 승인번호: #200081) 승인하에, 실시예 2, 3의 동물실험은 고려대학교 실험동물운영위원회 (IACUC, 승인번호: KOREA-2019-129) 승인하에 윤리적으로 실시하였다.
실험에서 사용한 HSP90 항원성 펩타이드의 서열정보는 표 1에 기재되어 있다.

실시예 1
HSP90에서 선별한 항원성 펩타이드(표 1 참조)와 다양한 애주번트 및 liposome에 의한 효능을 분석하기 위해 표 2 및 표 3의 조성으로 시험 백신을 제조하였다. G6와 G7 제형의 경우 리포좀을 포함하고 있기 때문에 약물의 방출 속도가 느려지므로, 항원성 펩타이드의 양을 20%로 줄여서 시험 백신을 제조하였다. 제조된 시험 백신 G1 내지 G7 제형은 C57BL/B6 마우스(암컷, 7주령)을 사용하여 각 군당 4마리씩 면역화하였다. CFA/IFA는 플루인드 애주번트(Complete Freund's adjuvant/Incomplete Freund's adjuvant)를 사용하였고, TLR2 아고니스트(TLR2L)로는 Pam3Cys-SKKKK 리포폴리펩타이드를 사용하였고, TLR3 아고니스트(TLR3L)로는 폴리(I:C)를 사용하였다. 리포좀은 양이온성 리포좀 사용하였다.
시험 백신은 기 출원된 등록 특허 제 10-0900837호 또는 제 10-2098097에서 기술된 바와 같이, 본 발명의 항원성 펩타이드를 Pam3Cys-SKKKK (G3) 또는 폴리(I:C) (G4)와 각각 상온에서 섞어주거나, Pam3Cys-SKKKK와 폴리(I:C)를 함께 포함하는 L-pampo 제형으로 (G5, G6, G7) 제조하였다. TLR2 아고니스트 및/또는 TLR3 아고니스트를 포함하는 G3, G4, G5, G6, G7의 시험 백신은 표 2 또는 표 3의 함량이 전체 volume인 200 ㎕에 포함되도록 제조하였다. 또한, G3, G4, G5, G6, G7의 시험백신은 150 mM NaCl과 10 mM sodium phosphate가 포함되고 pH는 7.0으로 맞춰진 수용액 상태의 제형으로 제조하였다.
시험 백신은 각 군당 4마리씩 10일마다 3회씩 피하주사로 각 백신 제형을 접종하고, 마지막 접종으로부터 10일 후 희생한 마우스의 비장에서 비장세포(splenocytes)를 분리한 후 이전에 공지된 Disis등의 방법(J Clin Oncol. 2009 Oct 1; 27(28): 4685-92)에 따라 서열번호 1 또는 서열번호 2의 HSP90 항원성 펩타이드(자극제)에 의해 자극된 IFN-γ 또는 IL-10 엘리스팟법을 통해 각각의 HSP90 항원성 펩타이드에 대한 특이적인 반응을 측정하였다.
시험 백신 접종 후 엘리스팟법으로 측정된 비장세포에서HSP90 항원성 펩타이드 특이적인 IFN-γ 생산량이 증가한 경우 Th1 면역반응이 활성화된 것으로 간주하였고, HSP90 항원성 펩타이드 특이적인 IL-10 생산량이 증가한 경우 Th2 면역반응이 활성화된 것으로 간주하였다.

서열번호 1 및 2의 각 항원성 펩타이드를 포함하는 G1 내지 G7의 시험 백신에 대한 엘리스팟 결과는 도 1 및 도 2에 나타내었다.
도 1a에서는 서열번호 1의 HSP90 항원성 펩타이드를 항원으로 사용하는 경우 G3 제형(TLR2 아고니스트 단독 포함)에서는 Th1 면역반응을 거의 보이지 않았고, G4 제형(TLR3 아고니스트 단독 포함)에서는 대조군인 CFA/IFA와 유사한 정도의 HSP90 특이적인 Th1 면역반응을 보였다. 하지만, TLR2 아고니스트 및 TLR3 아고니스트를 모두 포함하는 G5 제형에서 G3 및 G4 제형에 비해 상승효과(시너지 효과)를 보이는 것을 확인할 수 있었고, G5 제형에 리포좀을 추가하는 경우 HSP90 특이적인 Th1 면역반응이 급격하게 감소하는 경향을 보였다. TLR3 아고니스트를 더 많은 양으로 포함하는 G7 제형의 경우 HSP90 특이적인 Th1 면역반응이 G6 제형보다 더 감소하는 경향을 보였다.
서열번호 2의 HSP90 항원성 펩타이드를 항원으로 사용하는 경우 애주번트의 종류 및 리포좀 포함 여부와는 상관없이 HSP90 특이적인 Th1 면역반응이 거의 관찰되지 않았다(도 1b).
도 2a에서는 서열번호 1의 HSP90 항원성 펩타이드를 사용하는 경우 G2 제형(CFA/IFA)의 경우 Th2 면역반응이 가장 높았고, TLR2 아고니스트 및/또는 TLR3 아고니스트를 사용하는 경우 Th2 면역반응은 거의 관찰되지 않았다(G3~G5 제형). 하지만, TLR2 아고니스트 및 TLR3 아고니스트를 포함하면서 리포좀을 포함하는 경우 그 자체로 Th2 면역반응성이 증가하는 경향은 보였으나, 통계적으로 유의하지는 않았다.
서열번호 2의 HSP90 항원성 펩타이드를 사용하는 경우, Th1 면역반응보다는 Th2 면역반응 양상을 관찰할 수 있었지만, 각 제형 간에 통계적으로 유의한 결과는 얻지 못하였다(도 2b).
종합해보면, 본 실험에서는 서열번호 1의 HSP90 항원성 펩타이드가 암 백신으로서 요구되는 효과인 HSP90 특이적인 T 세포 면역반응을 효과적으로 유도하는 것을 알 수 있고, 이때 애주번트로 TLR2 아고니스트, TRL3 아고니스트의 혼합 사용이 상승효과(synergic effect)를 가지는 것을 알 수 있다(G5 제형). 하지만, 이러한 상승효과는 제형에 리포좀을 첨가하는 경우 완전히 사라지는 경향을 확인하였다. 따라서, HSP90 항원성 펩타이드를 포함하는 항암용 백신 조성물에는 애주번트로 TLR2 아고니스트 및 TLR3 아고니스트를 포함하고, 리포좀은 별도로 포함하지 않는 것이 HSP90 특이적인 T 세포 면역반응을 위해서는 바람직한 제형이라는 것을 알 수 있다.
실시예 2
마우스 모델에서 다양한 TLR 아고니스트의 애주번트로서의 활성을 분석하기 위해 서열번호 1 및 2의 HSP90 항원성 펩타이드 혼합물(이하 'HSP90 펩타이드'또는 'HSP90 peptide"라 함)과 애주번트로 사용된다고 알려진 TLR4 아고니스트(TLR4L), TLR2/3 아고니스트(TLR2 아고니스트 및 TRL3 아고니스트의 혼합; TLR2/3L) 및 TLR7/8 아고니스트(TLR7 아고니스트 및 TLR8 아고니스트의 혼합; TLR7/8L)을 제형화하여 FVB 마우스(암컷, 6주령)를 각 군당 4마리씩 10일마다 3회씩 피하주사로 각 백신 제형을 접종하고, 마지막 접종으로부터 10일 후 마우스의 비장세포를 분리한 후 IFN-γ 엘리스팟법을 이용하여 HSP90 특이적인 T 세포 반응을 측정하였다.
본 실험에서 사용한 CFA/IFA는 시그마사에서 구입하였고, TLR4 아고니스트는 Quratis사의 GLA-SE를 사용하였다. TLR2/3 아고니스트는 차백신 연구소에서 제공받았으며, TLR7/8 아고니스트 3M-052는 3M사에서 구입하였다. 음성 대조군으로 사용한 TT 펩타이드는 파상풍 독소(tetanus toxin) 펩타이드이고, 콘카나발린 A는 양성 대조군으로 사용하였다. 백신 제형에 포함되는 각 성분의 양으로 CFA/IFA는 100㎍, TLR2 아고니스트는 125 ㎍, TLR3 아고니스트는 100 ㎍, HSP90 펩타이드는 서열번호 1로 표시되는 펩타이드 및 서열번호 2로 표시되는 펩타이드 각각 50 ㎍씩 조합한 100㎍을 사용하였고 잔량은 PBS로 전체 부피를 200㎕로 조절하였다.
도 3에서 나타난 바와 같이, HSP90 항원성 펩타이드와 애주번트로 TLR4 아고니스트 또는 TLR7/8 아고니스트를 포함하는 제형은 모두 HSP90 특이 T 세포 반응을 유도하지 못했다. HSP90 항원성 펩타이드와 애주번트로 CFA/IFA를 사용하는 경우와 TLR2/3 아고니스트를 사용하는 경우 모두 HSP90 특이 T 세포 반응을 유도하였지만, TLR2/3 아고니스트를 사용하는 경우 일반적으로 항암백신에서 강력한 면역반응을 유도하는 것으로 알려진 CFA/IFA보다도 2배 이상 더 높은 HSP90 특이 T 세포 반응을 유도하였다. 이로부터 TLR2/3 아고니스트가 HSP90 항원성 펩타이드에 더욱 적합한 애주번트임을 확인하였다.
따라서, 서열번호 1 및 2의 HSP90 항원성 펩타이드를 면역원성 항원으로 사용하는 경우, 다양한 TLR 아고니스트 중에서 TLR2/3 아고니스트가 특이적으로 HSP90 특이 T 세포 반응을 유도하는 것을 확인하였고, 이러한 결과는 HSP90 항원성 펩타이드를 면역원으로 하는 경우 애주번트로 TLR2/3 아고니스트를 사용하는 경우, 암 백신에서 목적하는 면역원성 특이적인 T 세포 반응이 유도될 것이라는 것을 반영한다.
실시예 3
종양 이식 모델은 마우스 유방암 세포주를 마우스에 이식하는 모델을 사용하였다. 종양 생성을 위한 마우스 유방암 세포주는 mouse mammary cancer cell-FVB/N-Tg (MMTVneu)-202Mul 마우스 유래 HER-2 과발현 유방암 세포주를 사용하였고, 상기 유방암 세포주를 이식할 마우스는 MMTVneu 형질전환 마우스 암컷 6주령을 사용하였다.
각 군당 4마리씩 생리식염수(PBS)와 애주번트(TLR2/3 아고니스트)군(이하 "대조군"이라 함) 또는 서열번호 1 및 서열번호 2의 HSP90 항원성 펩타이드와 애주번트(TLR2/3 아고니스트)군(이하 "시험군"이라 함)으로 나누었다. 10일 마다 3회씩 피하주사를 대조군 또는 시험군의 백신을 접종하고 마지막 접종으로부터 10일 후 생리식염수와 matrigel을 1:1 로 섞은 gel에 한 마리 당 5 X 105개의 마우스 유방암 세포주를 마우스 옆구리에 피하 주사하여 종양의 성장을 3~4일 간격으로 관찰하여 종양의 크기를 측정하여, HSP90 항원성 펩타이드 백신의 종양 형성을 억제하는 효과를 확인하였다. 백신 접종 및 종양 세포 이식의 동일한 기간 동안 각 마우스의 체중은 대조군 및 시험군 사이에 차이가 없었다.
암 세포 이식 후 35일이 되는 날에 각 마우스를 희생하여, 암 조직 및 비장을 적출하여 크기를 측정하였다.
도 4에서 나타난 바와 같이 시험군(HSP90 항원성 펩타이드와 TLR2/3 아고니트스 백신 제형 투여군)의 마우스는 대조군에 비해 통계학적으로 유의하게 종양 성장이 억제되었다. 구체적으로는 대조군의 평균 평균 종양 부피가 1031 ± 450 mm3 인 반면에, 시험군의 평균 종양 부피는 170 ± 108 mm3로서, 시험군의 평균 종양 부피는 대조군의 17%에 불과하여, TLR2/3 아고니스트 백신제형이 항원 특이적인 종양 모델에서 효과적으로 종양 생성을 억제할 수 있음을 확인할 수 있었다. 이때 각 군의 체중 차이는 없어서, 독성반응에 의한 차이가 아닌 것으로 확인하였다.
이때 희생시킨 대조군 및 시험군에서 적출한 종양 및 비장의 크기는 도 5에 나타내었다. 시험군은 대조군에 비해 비장의 크기가 확연하게 비대해져 있었고, 이로부터 시험군에서 면역반응이 활성화되었음을 확인할 수 있었다.
