本開示は、一態様において、腫瘍が本開示におけるスプライシング制御化合物と接触することで産生するスプライス-ネオアンチゲンを有効成分として含有する、前記腫瘍の免疫原性を向上させるための医薬組成物に関する。
本開示によれば、一又は複数の実施形態において、腫瘍において新たなスプライス-ネオアンチゲンを誘導できる。
本開示によれば、一又は複数の実施形態において、免疫チェックポイント療法を増強できる。
本開示は、一態様において、SRSFsの活性を調節するスプライシング制御化合物が、免疫チェックポイント療法を増強できるという知見に基づく。
本開示におけるスプライシング制御化合物は、一又は複数の実施形態において、腫瘍細胞以外の細胞、例えば正常細胞、よりも腫瘍細胞に対して選択的にスプライス-ネオアンチゲンの産生を誘導できる。
本開示におけるスプライシング制御化合物は、一又は複数の実施形態において、がん種を超えて複数の腫瘍細胞に対してスプライス-ネオアンチゲンの産生を誘導できる。

本開示において、SRタンパク質とは、セリン(S)とアルギニン(R)の長いリピート配列を持つタンパク質ドメイン(RSドメイン)を含むタンパク質を意味し得る。SRタンパク質は、一又は複数の実施形態において、200~600アミノ酸のタンパク質であり、RNA認識モチーフ(RRM)領域とアルギニン/セリン・リピート(RSドメイン)の2つのドメインから構成される。

まず、SRSFを標的としたスプライシングモジュレーターである化合物1が抗腫瘍免疫を高めることで腫瘍の成長を抑制するかどうかを確認するため、免疫不全マウスを用いた担がん動物実験を実施した。化合物1投与により、CD8+T細胞及び腫瘍MHC I依存的に腫瘍増殖が抑制され、PD-1遮断効果も増強されることを確認した。
次に化合物1を用いて誘導可能なネオアンチゲンの探索を試みた。この課題に取り組むため、化合物1によって誘導又は増強される非注釈のスプライスバリアントを抽出し、これらの新規バリアントから翻訳される異常ペプチドを作成するためにRNA-seqを実施した。抽出されたネオアンチゲン候補の中から、IFN-γ ELISpotアッセイを用いて免疫原性ペプチドをスクリーニングし、同定した。
その後、in vivoワクチン接種試験により、化学的に誘導可能なスプライスネオアンチゲンを免疫すると、共培養条件下で腫瘍溶解のための内因性T細胞反応が誘発され、がん細胞を化合物1で感作すると腫瘍増殖が抑制されることが明らかになった。
以下詳述する。
悪性腫瘍の治療には、腫瘍細胞を直接死滅させる従来の細胞傷害性化学療法が用いられてきたが、重篤な副作用のためにその使用が制限されることが多い。低分子RNAスプライシングモジュレーターである化合物1を用い、RNAスプライシングイベントを操作することで、腫瘍の免疫原性を高め、直接的な細胞傷害作用を伴わない免疫チェックポイント療法への反応を増強できることを以下のように確認した。
まず、免疫不全マウスに皮内移植した同種MC38腫瘍を用いてin vivoでの活性評価を行った(図1-1A)。化合物1はin vitroでの細胞毒性や細胞増殖に関連しない濃度で(図1-1D及びE)、in vivoでの腫瘍増殖抑制と生存期間の延長をもたらした(図1-1B及びC)。
抽出した腫瘍を分析することにより、腫瘍浸潤CD8+T細胞の頻度が化合物1処理後に約1.5倍まで増加することを見出した(図1-1F及びG)。
23日目において、化合物1単体では28.5%の腫瘍増殖の抑制が確認され(アイソタイプIgG+担体vs.アイソタイプIgG+化合物1,p=1.1×10-3)、一方、αPD-L1では41.0%の腫瘍増殖の抑制が確認された(アイソタイプIgG+担体vs.αPD-L1+担体,p≦1.0×10-4)(図1-2 O及びP)。よって、化合物1とαPD-L1が相加的に機能する場合、約57.8%の腫瘍抑制が期待された。
しかし、化合物1とαPD-L1を共投与した場合67.8%の腫瘍抑制が観察され(アイソタイプIgG+担体vs.αPD-L1+化合物1,p≦1.0×10-4)、これらは相加的又は若干相乗的に機能することが示唆された(図1-2 O及びP)。
また、フローサイトメトリーにより、化合物1及びαPD-L1で共処理した群では、αPD-L1単独で処理した群と比較して腫瘍浸潤CD8+T細胞の数が増加したことが明らかになった。さらに、これらの実験において、採取した腫瘍量が測定した腫瘍体積とよく相関することが確認された。しかしながら、アイソタイプIgG、αPD-L1、又はαCD8で処置したマウスにおいて、化合物1の有無にかかわらず、体重の変化や明らかな副作用は見られなかった(図1-2Q)。
これらの結果は、化合物1によるSRSFの化学的活性化がCD8+T細胞依存的に抗腫瘍効果を発揮し、がん細胞に対する直接的な細胞障害性又は細胞増殖性効果なしにPD-1遮断の効力を向上させることを示した。
T細胞活性化におけるスプライスアイソフォームの産生の変化は、おそらくその後の機能に影響を及ぼすと考える。化合物1がCD8+T細胞依存的に抗腫瘍効果を発揮することを踏まえ、次にex vivo脾臓細胞培養アッセイを用いて化合物1が抗原非依存的にT細胞の活性化を誘導するかどうかを調べた。
まず正常マウスから脾臓細胞を単離し、細胞を化合物1で24時間処理しました(図2―1A)。次に、フローサイトメトリーを用いて、CD8+又はCD4+T細胞におけるCD44+エフェクターT細胞マーカー及びCD69+急性T細胞活性化マーカーの頻度を測定した(図2-1B及びC)。予想通り、コンカナバリンA(ConA)による処理はCD8+及びCD4+T細胞におけるCD44+及びCD69+マーカーの両方の発現を促進したが、化合物1による処理ではそのような効果はなかった(図2-1CからE)。また、化合物1投与群ではインターフェロンγ(IFN-γ)や腫瘍壊死因子-α(TNF-α)を含む炎症性サイトカインの分泌は観察さなかった(図2-1F及びG)。
これらの結果から、化合物1投与に伴うT細胞の抗原非依存的な活性化の関与は否定された。
体重変化は実験期間中、全群で同程度であることが確認された(図2-2I)。初回投与後7日目の急性反応に関して、αPD-L1の有無にかかわらず化合物1投与は脾臓重量、白血球数、脾臓及び末梢血におけるT細胞活性化マーカー(CD44high/CD8+、CD69+/CD8+、CD44high/CD4+、CD69+/CD4+)の発現に影響しないことが確認された(図2―2J及びK)。
さらに、28日目に得られた非悪性組織の組織学的解析により、正常マウスを化合物1に慢性的に曝露しても、αPD-L1との共処理にかかわらず、CD8+T細胞の蓄積や炎症性病変の出現は認められなかった(図2-3L及びM)。さらにまた、末梢血中の白血球数及び生化学的パラメータに変化は認められなかった(データ示さず)。
これらの結果は、化合物1が、有害な炎症反応や自己免疫反応なしに、がんの免疫原性を高めることを示した。
がん細胞に発現するMHCクラスIは、CD8+T細胞依存的な免疫監視と腫瘍殺傷に不可欠なネオアンチゲン提示に重要である。化合物1がネオアンチゲン提示経路を調節するかどうかを調べるために、まず、CD8+T細胞分泌IFN-γがMHCクラスI成分の主要な誘導因子であることから、MC38細胞におけるIFN-γシグナル並びにβ2ミクログロブリン(B2M)及びH2-Kb等のMHCクラスI成分の発現に対する化合物1の影響を調べた。IFN-γの投与はSTAT1及びSTAT3のリン酸化を強固に誘導し、B2Mタンパク質の発現を増加させたが、化合物1の投与はそのような影響を及ぼさなかった。また、H2-Kb発現のわずかな増加(0及び0.1ng/mLのIFN-γを72h投与で11.1%と12.4%)と、細胞表面のPD-L1発現の減少(0、0.1及び1ng/mLのIFN-γを72h投与で3.8%,3.2%,及び26.1%)が確認された。一方、宿主の樹状細胞やマクロファージでは、PD-L1の発現に変化は見られなかった。RNA-seqデータによると、H2-Kb並びにPD-L1のmRNA発現及びスプライシングパターンは、化合物1による処理後のMC38細胞において有意に変化しなかったことから、これらの変化は翻訳後調節に起因するものであると判断された。しかしながら、αPD-L1との共投与により化合物1の抗腫瘍効果の増強が認められたことから、これらの変化は、化合物1が抗腫瘍効果(図1-2 O及びP)を発揮する機能的メカニズムではないと考えられた。
次に、これらのB2m KO MC38クローンを免疫不全マウスに皮内接種し、化合物1の抗腫瘍効果を評価した(図3-2G)。化合物1を投与すると、陰性コントロールクローンでは一貫して腫瘍の成長が抑制されたが、B2m KOクローンではこの効果が完全に消失した(図3-2H)。
さらに、CD8+T細胞の数はB2m KO腫瘍で低く、T細胞増殖の促進におけるMHCクラスIの本質的役割と一致し、B2m KO由来腫瘍では化合物1による処理によってもはや誘導されなかった(図3-2I及びJ)。また、陰性コントロールクローンではαPD-L1の抗腫瘍効果が確認されたが、B2m KOクローンでは確認されなかった(図3-2K)。
これらの結果は、がん細胞において、MHCクラスIが化合物1の抗腫瘍効果を決定する上で重要な役割を担っていることを明確に示す。
腫瘍細胞における異常な選択的スプライシング事象は、潜在的なネオアンチゲンの源となり得る。腫瘍細胞のMHCクラスIが化合物1の抗腫瘍効果に不可欠であることを確認したので、化合物1が異常な選択的スプライシングイベントを変化させ、スプライス-ネオアンチゲンの産生を促進すると予想した(図4-1A)。この概念を検証するために、化合物1がMC38細胞に与えたRNAスプライシングパターンと、MHCクラスI結合可能な潜在的ネオアンチゲンをもたらすスプライス産物を評価するためにRNA-seq解析を行った(図4-1B)。まず、化合物1によって有意に変化したスプライシングイベントを抽出した。次に、潜在的なスプライス-ネオアンチゲンを翻訳するネオエピトープを同定するために、異常なコーディング配列を作成する化合物1によって誘発されたスプライシングイベントを抽出し、続いて、これらのイベントが異常なペプチドを作成するかどうかを参照ペプチドデータベースと比較して確認した。
最終的に、化合物1によって誘導された22のスプライスネオアンチゲン候補(8~11アミノ酸長)をその後の評価のために選別した(図4-2D及び表1)。
一例を挙げると、ニューロフィブロミン1(Nf1)であって、化合物1による処理により108bpのエクソン化が誘導され、その結果、これまで非注釈でありH2-Kbへの親和性が238.15nMと予測される「RNRSHIFPL」候補ネオエピトープをコードするスプライスが生成される(図4-2E)。RNA-seq解析はまた、化合物1による処理がMC38細胞で産生される従来のネオアンチゲンをコードする遺伝子のmRNA発現に影響を及ぼさないことを明らかにした(データ示さず)。
スプライス-ネオアンチゲン候補が得られたので、次に化合物1によるスプライス-ネオアンチゲンがMHCクラスIに結合し、CD8+T細胞の活性化を促進するかどうかを実験的に検証した。
22のスプライス-ネオアンチゲン候補と陽性対照としてメラノーマ抗原ペプチドtyrosinase related protein 2(TRP2)180-188を合成した。免疫不全マウスに,アジュバントとしてPoly(I:C)存在下,合成したペプチドを2回接種した。次に、ワクチン接種したマウスから脾臓細胞を調製し、ペプチドをパルスした骨髄由来樹状細胞(BMDC)と24時間共培養し、IFN-γELISpotアッセイを用いてCD8+T細胞の活性化を評価した(図5-1A)。
検討した22のペプチドのうち、INF-γELISpotアッセイにより、免疫原性を有する6つのペプチドが同定され、それらはKifc1、Nf1、Acbd4、Rfx7、Qpctl、及びNup153のスプライシング変化に由来していた(図5-1B及びC)。
次に、マウス肺がんLLC、膵管腺がんPan02、白血病WEHI3、大腸腺がんCT26、リンパ腫EG7、皮膚メラノーマB16細胞を用いて、化合物1により誘導されたスプライスネオアンチゲンががん種間で共通しているかどうかを検討した。興味深いことに、化合物1によって誘導されるこれらのスプライス変化は調べた細胞の大部分で共有されていることがわかり(図5-2F)、化合物1が幅広いスペクトルのがんに適用できることが示唆された。
さらに、化合物1投与マウスの結腸、肺、肝臓組織に対応するRNA-seqデータも調べ、スプライシング変化の程度を分析した。正常組織における化合物1誘発スプライシングイベントはMC38細胞におけるものと比較して50~60%少ないことが見出された。これらの組織において免疫応答が化合物1によって誘発されなかったことを考慮すると、正常組織におけるこれらの変化したスプライシングイベントは免疫原性に無関係である可能性が高いと考えられる(図2-2I~図2-3M)。実際、検証したスライス-ネオ抗原形態のいずれも正常組織では発現が著しく誘導されていないことが分かった(データ示さず)。
6種類の免疫原性ペプチドが由来する遺伝子と、該ペプチドのアミノ酸配列は以下の通り。

化合物1誘導性スプライス-ネオアンチゲンが免疫原性を発揮することを確認したので、次に、抗がん免疫応答への寄与を検討した。この目的のため、腫瘍の殺傷を確実にするために共培養アッセイを用いた(図6-1A)。
脾臓細胞は、6価のスプライスネオ抗原又は無関係のTpr2180-188抗原を接種したマウスから調製し、さらにIL-2の存在下で同じ抗原でパルスしたBMDCで刺激した。腫瘍殺傷活性は、抗原で誘導されたエフェクター細胞と生細胞蛍光染色色素CFSEで予備標識した標的細胞との共培養から24時間後のAnnexin V+/CFSE+の頻度に基づき定量化された。Tpr2免疫マウスのエフェクター細胞は、ペプチド負荷に関係なく何の反応も示さなかったが、6価のスプライスネオ抗原で刺激されたエフェクター細胞は、6つのスプライスネオピトープを提示するMC38細胞を選択的に殺傷した(図6-1B及びC)。その結果、B2m KO MC38細胞ではこの腫瘍殺傷活性が見られなかった(図6-1D)。このことは、この効果がMHCクラスIに依存することを示している。
さらに、個々のエピトープの寄与を共培養アッセイによる腫瘍殺傷から調べたところ、大多数のエピトープ(Kifc1、Nf1、Acbd4、Rfx7、Nup153)が個別に有意に腫瘍殺傷応答を引き起こすことが確認された(図7A)。さらに、化合物1処理により内因性スプライス-ネオアンチゲンの産生を誘導したがん細胞との関連で腫瘍殺傷応答を調べたところ、6価ワクチン免疫マウスからのエフェクター細胞は、調べた3つのがん細胞すべて、すなわちMC38(図6-2E)、B16(図6-2F)及びLLC(図6-2G)に対して細胞毒性活性を示しており、すべて化合物1処理に反応してスライスネオアンチゲンが発現した(図5-2)。
一方、これらのエフェクター細胞は、B6マウスの非がん性マウス胚性線維芽細胞(MEF)に対する細胞溶解活性を示さなかった(図6-2H)。スプライス-ネオアンチゲンの発現はMC38に比べて非常に低く、MEFはスプライス-ネオアンチゲンが十分に誘導されないためにCD8+T細胞による溶解を回避していることが示唆された。
6価ワクチン単独接種では腫瘍増殖の遅延が認められた(Poly(I:C)+担体vs.Poly(I:C)/6価ペプチド+担体、23日に24.7%抑制)(図6-2J)。これはおそらくがん細胞において基底発現したスプライス-ネオアンチゲンの免疫原性に起因するものであり(図5-2D及びE)、この効果は化合物1の同時投与によりさらに増強された(Poly(I:C)+担体vs.Poly(I:C)/6価ペプチド+化合物1、23日目に57.0%抑制)(図6-2J)。また、化合物1による個別ワクチン接種による各スプライス-ネオアンチゲンの効果を評価したところ、ワクチン非接種対照と比較して腫瘍増殖が6~23%抑制された(図7B)。個別接種の腫瘍抑制効果は、抑制率が30.7%であった6価ワクチン接種よりも弱く(図6-2J)、同定された6つのネオエピトープの同時接種により抗がん免疫応答が増強されることが示された。
下記化合物2は、セリン及びアルギニンリッチスプライシング因子(SRSFs)を標的としたスプライシングモジュレーターの1つである。また、化合物2は、Cdc2様キナーゼ(CLK)を調節する機能を有し、CLKの調節を介してSRSFsに影響を与える(Cell chem biol.,2020,vol.27,P1472-1482)。
ELISpotアッセイ
8週齢の雌性C57BL/6Nマウスに、0日目と7日目に、合成したペプチド(Genscript Japan)100μgとPBSに配合したポリ(I:C)50μg(5ペプチド/マウス)を皮下投与した。H2-Kb TRP-2180-188ペプチド(MBL International, Woburn, MA)を陽性対照として使用した。マウスは最初の注射から12日後に犠牲にし、脾臓細胞を単離した。刺激物として、骨髄由来樹状細胞(BMDC)を、いくつかの修正を加えて、以前に記載されたように生成した。簡単に言うと、骨髄細胞は、8週齢の雌のC57BL/6Nマウスの大腿骨から、2日目に遠心分離によって単離された。細胞を70μmのセルストレーナーに通し、10%FBS、100U/mL P/S、2mM L-グルタミン、50μM 2-ME、及び20ng/mL Gm-csf(Peprotech,Rocky Hill,NJ)を含むRPMI1640で10cmシャーレに播種した。培養は、2日又は3日ごとに培地の半分を交換することによって維持された。12日目に、非付着細胞のほとんどが典型的な樹状形態を獲得し、これらの細胞を2μg/mlのペプチドで37℃、2時間パルスした。ELISpotアッセイは、Mouse IFN-γ ELISpot BASICキット(Mabtech AB,Nacka Strand,Sweden)を用いて、製造者の説明書に従って実施した。新鮮な単離脾臓細胞(5×105細胞)を、抗IFN-γ抗体(クローンAN18)をコートしたELISpot PVDFホワイトプレートにおいて、5%CO2条件下、37℃で一晩、ペプチドパルスしたBMDC(4×104細胞)と共培養させた。その後、捕捉抗体(クローンR4-6A2)とBCIP/NBT-plus基質を用いてIFN-γの分泌を検出した。プレートを放置して乾燥させた後、解剖顕微鏡を用いてスポット数をカウントした。ImmunoSpot S6 Ultimate Analyzer(Cellular Technology Limited, Cleveland, OH)を用いて、スポット画像を取得した。
「ELISpotアッセイ」の項に記載したように、14日目にPoly(I:C)を接種したペプチドマウスから脾臓細胞を調製した。抗原特異的CD8+T細胞をさらに惹起するために、新たに単離した脾臓細胞を、10ng/mLのIL-2(Peprotech)の存在下でペプチド(s)-パルスした骨髄由来樹状細胞(BMDC)とさらに7日間共培養した。培養は、2日または3日ごとに培地の半分を交換することによって維持された。次に、BMDCsで刺激した脾臓細胞をエフェクター細胞として、標的細胞とエフェクターを、エフェクター:標的=5:1の比率で24時間共培養し、その溶菌活性を調べた。標的細胞の調製は、前日のエフェクター細胞との共培養において、細胞を2M carboxyfluorescein diacetate succinimidyl ester(CFSE;同仁堂)で10分間標識し、さらにFBSで10分間インキュベーションして標識反応を停止させた。培地で2回洗浄した後、細胞を10ng/mL IFN-γ存在下で一晩培養し、翌日、標的細胞を共培養前に10ng/mLペプチドで2時間パルスするか、エフェクター細胞の存在下で化合物1で処理した。24時間の共培養後、細胞をアネキシンV結合バッファー中、アネキシンV(Biolegend)で15分間染色し、フローサイトメトリー解析に供した。エフェクター細胞を介した殺傷活性は、アネキシンV+/CFSE+の頻度から算出した。
RNeasy Mini Kit(Qiagen)を用いて、製造元のプロトコールに従って細胞から総RNAを抽出し、High Capacity cDNA Reverse Transcription Kit(Applied Biosystems,Foster City,CA)を用いて逆転写を行った。遺伝子は、KOD One DNA polymerase(ToYoBo)と標的特異的プライマーセットで増幅した。RT-PCR産物は電気泳動で分離した。エチジウムブロマイドで染色した画像をChemiDoc MP Imaging Systemで撮影し、Image Jソフトウェアでデータを解析した。プライマーは以下のものを使用した。Kife1については5’-TTC CTG TGA GAA AGA GGT GGA G-3’(配列番号:23)及び5’-CAC AAA CAT AAG CCT TAT TGC TCA G-3’(配列番号:24)を使用した。Nflについては5’-CAC TTT AGT GAA GAG ACC AAG CAA G-3’(配列番号:25)及び5’-TCA TAC ATG CTT GTT GTG GCA GAG-3’(配列番号:26)を使用した。Acbd4については5’-CAT GGG TAC CGA GAA GGA AGA G-3’(配列番号:27)及び5’-TCC AAT GGG GTC CCA GAA C-3’(配列番号:28)を使用した。Rfx7については5’-AAA CTG GAG ATG GGG ACT TA-3’(配列番号:29)及び5’-TGA GGA TTC TCC TGG CAA TG-3’(配列番号:30)を使用した。Qpctlについては5’-CCA AGT GAG AAA GTT CCT GGA G-3’(配列番号:31)及び5’-GAG GAC AAA GAG CTC CTG CTG-3’(配列番号:32)を使用した。Nup153については5’-CCA GAG AAT GAG GAA GTG GAA G-3’(配列番号:33)及び5’-GTA AAT CCA ATC TTG TTT GG-3’(配列番号:34)を使用した。Actbについては5’-TCT TTG CAG CTC CTT CGT TG-3’(配列番号:35)及び5’-GGC CTC GTC ACC CAC ATA G-3’(配列番号:36)を使用した。Actbはローディングコントロールとして使用した。
RNA-SeqライブラリーはTruSeq Stranded mRNA Library Kit (Illumina, San Diego, CA)を用いて調製し、Illumina NovaSeq 6000プラットフォームでペアエンドシーケンスに使用した。RNA-seqデータは、HISAT2プログラムバージョン2.2.0で「--rna-strandness RF」オプションを使用してGRCm38(mm10)ゲノム配列にマッピングされた。この際、Ensembl gene annotation version 100から得られるスプライスジャンクションの情報を用いた。マップされた結果は、ペアリードのどちらかがゲノムにマップされていない場合や、マップ位置の関係が異常な場合は破棄された。
スプライシング解析には、rMATS program version 4.1.0 を用いた。rMATS解析に用いた参照転写産物モデルは、RNA-seqデータに基づく新しい転写産物セットをcufflinksプログラムバージョン2.2.1を使って構築した。rMATSプログラムでは、ソフトクリッピングリードやインデルを持つリードはアライメントでスキップされるため、ソフトクリッピングやインデルの情報はスプライスポジション情報を変更しないものとして、bamファイルを修正した。rMATS解析後、以下の基準に従って、変化したスプライスイベントを抽出した。FDR<0.01,dPSI≧5.0,平均スプライスリード数≧15,いずれかのサンプルに存在する。
スプライシングされた領域は、その領域にコードされるアミノ酸配列を記述するためのEnsembl遺伝子アノテーションと比較された。元のペプチドデータベースで観察されないアミノ酸配列は、既知の配列から7アミノ酸のマージンをとって検索・保存した。
スプライシング由来ペプチドのMHCクラスI(H2-Kb、H2-Db)に対する親和性の予測には、NetMHCpan version 4.1に「-1,8,9,10,11」のオプションを付けて実行した。DEG解析は、RSEMプログラムバージョン1.2.31を用いてTranscripts per million(TPM)値とraw reads countsを算出した。その後、DESeq2プログラムバージョン1.8.2を用いて、以下の基準を満たすDEGを検索した:いずれかのサンプルにおいて、adjusted-p<0.05,fold change≧1.3 or ≦1/1.3,average TPM≧1,及びaverage raw-read counts≧32,試料において。

- セリン及びアルギニンリッチスプライシング因子(SRSFs)の活性を調節するスプライシング制御化合物を有効成分として含有する、腫瘍の免疫原性を向上させるための医薬組成物。
- 前記化合物は、Cdc2様キナーゼ(CLK)を調節できる化合物である、請求項1に記載の医薬組成物。
- 前記化合物は、腫瘍の免疫原性を向上させるペプチドを腫瘍に産生亢進させる化合物である、請求項1又は2に記載の医薬組成物。
- 前記ペプチドは、下記ペプチド(1)~(6)及びこれらの2~6の組合せからなる群から選択される、請求項1から3のいずれかに記載の医薬組成物。

ペプチド(6):SQPLPNKIGF(配列番号:6)のアミノ配列を含むペプチド、又は該ペプチドにおいて配列番号6に相当するアミノ酸配列の1個~数個のアミノ酸が欠失、置換、及び/又は付加されたペプチドであって腫瘍の免疫原性の向上能を有するペプチド。 - 前記ペプチドが、ペプチド(1)及び(2)の少なくとも一方である、請求項4に記載の医薬組成物。
- 請求項4に規定されるペプチド(1)~(6)及びこれらの2~6の組合せからなる群から選択されるペプチドを腫瘍に産生亢進させるスプライシング制御化合物を有効成分として含有する、医薬組成物。
- 請求項4に規定されるペプチド(1)及び(2)の少なくとも一方を腫瘍に産生亢進させるスプライシング制御化合物を有効成分として含有する、医薬組成物。
- 前記スプライシング制御化合物は、スプライシング因子RBM39を調節するスプライシング制御化合物ではない、請求項1から7のいずれかに記載の医薬組成物。
- 前記スプライシング制御化合物は、下記式(I)~(II)で表される化合物及びその製薬上許容される塩からなる群から選択される少なくとも1つの化合物である、請求項1から8のいずれかに記載の医薬組成物。
R1及びR2は、それぞれ独立して、水素原子、置換若しくは無置換のC1-C6アルキル基、置換若しくは無置換のベンジル基、置換若しくは無置換のヘテロアリールメチル基、置換若しくは無置換のヘテロアリールエチル基、置換若しくは無置換のアリールオキシ基、置換若しくは無置換のヘテロアリールオキシ基、置換基若しくは無置換のアルコキシアミドアルキル基、置換若しくは無置換のアリール基、又は置換若しくは無置換のヘテロアリール基であり、或いは、R1とR2は結合してNとともに環を形成し、前記環が、置換若しくは無置換の単環式複素環、又は置換若しくは無置換の二環式複素環である;
R5は、水素原子、ハロゲン原子、置換若しくは無置換のC1-C6アルコキシ基、又はジアルキルアミノ基である;
X1は、N又は-CH-である;
X2は、-N(R3)-、S又はOである;
R3は、水素原子、C1-C6アルキル基、ベンジル若しくはヘテロアリールメチル基、置換若しくは無置換のアリール基、置換若しくは無置換のヘテロアリール基、又はCH2OC(O)R4-である;
R4は、C1-C6アルキル基、ベンジル若しくはヘテロアリ-ルメチル基、置換若しくは無置換のアリール基、又は置換若しくは無置換のヘテロアリール基である;
Xは、水素原子、ハロゲン原子、アミノ基、R1及びR2が置換したアミノ基、アジド基、シアノ基、ニトロ基、水酸基、C1-C6アルキルオキシ基、置換若しくは無置換のアリールオキシ基、置換若しくは無置換のヘテロアリールオキシ基、メルカプト基、C1-C6アルキルチオ基、置換若しくは無置換のアリールチオ基、置換若しくは無置換のヘテロアリールチオ基、ベンジル若しくはヘテロアリールメチル基、置換若しくは無置換のアリール基、又は置換若しくは無置換のヘテロアリール基である;
式(II)において、
X3及びX4は、それぞれ独立して、S又はNHである;
R6は、
R7は、水素原子、ハロゲン原子又はC1-C6アルキル基である。 - 前記スプライシング制御化合物は、下記式(III)~(VII)で表される化合物及びその製薬上許容される塩からなる群から選択される少なくとも1つの化合物である、請求項1から8のいずれかに記載の医薬組成物。
R8及びR9は、それぞれ独立して、水素原子、ハロゲン置換若しくは無置換のC1-C10アルキル基、又はC2-C6アルケニル基である;
R10は、水素原子、ハロゲン原子、ハロゲン置換若しくは無置換のC1-C10アルキル基、-OR11、-NHR11、又は-N(R11)2である;
R11は、水素原子、又はC1-C10アルキル基である;
式(IV)において、
R12及びR13は、それぞれ独立して、水素原子、又はC1-C6アルキル基である;
R14は、
R15は、水素原子、ハロゲン原子、又はC1-C6アルキル基である;
式(V)及び(VI)において、
R16及びR18は、それぞれ独立して、水素原子、C1-C6のアルキル基、ベンジル若しくはヘテロアリールメチル基、置換若しくは無置換のアリール基、又は置換若しくは無置換のヘテロアリール基である;
R17は、-R21、-C≡C-R21、-CH=CH-R21、又は-O-(CH2)n-R21であり、nは1~6であり、R21は、水素原子、水酸基、C1-C8アルキル基、-Si(R22)3、又は、置換若しくは無置換のフェニル基、単環式複素芳香環基又は環状脂肪族基である;
或いは、R16とR17は結合して環を形成し、-R16-R17-が、-(CH2)m-CH2-、-CH=CH-、-(CH2)m-O-、又はハロゲン原子で置換されたこれらのものであり、mは1~6であり、R22は、水素原子、C1-C6アルキル基、卜リハ口メチル基、又は水酸基であり、-Si(R22)3中の3つのR22はそれぞれ異なっていてもよい;
R19及びR20は、水素原子又はC1-C6アルキル基である;
式(VII)において、

R23は、
R24は、水素原子、ハロゲン原子、カルボキシル基、アミノ基、水酸基、ハロゲン置換若しくは無置換のC1-C4アルキル基である。 - 請求項4に規定されるペプチド(1)~(6)及びこれらの2~6の組合せ、並びに、それらの製薬上許容される塩、からなる群から選択される少なくとも1つを有効成分として含有する、医薬組成物。
- 腫瘍の免疫原性を向上させるための医薬組成物である、請求項6から11のいずれかに記載の医薬組成物。
- 免疫チェックポイント阻害剤と同時に、別々に、又は順次に併用投与されるものである、請求項1から12のいずれかに記載の医薬組成物。
- 免疫チェックポイント阻害剤が、(1)CTLA-4、(2)PD-1、(3)LAG-3、(4)BTLA、(5)KIR、(6)TIM-3、(7)PD-L1、(8)PD-L2、(9)B7-H3、(10)B7-H4、(11)HVEM、(12)GAL9、(13)CD160、(14)VISTA、(15)BTNL2、(16)TIGIT、(17)PVR、(18)BTN1A1、(19)BTN2A2、(20)BTN3A2、及び(21)CSF-1Rからなる群から選択される分子に対する1以上の薬剤である、請求項13に記載の医薬組成物。
- セリン及びアルギニンリッチスプライシング因子(SRSFs)の活性を調節するスプライシング制御化合物を腫瘍に接触させることを含む、腫瘍の免疫原性を向上させる方法。
- 請求項1から14のいずれかに記載の医薬組成物を腫瘍に接触させることを含む、腫瘍の免疫原性を向上させる方法。
- 免疫チェックポイント阻害剤と、請求項1から14のいずれかに記載の医薬組成物とを、同時に、別々に、又は順次に併用投与することを含む、免疫チェックポイント療法を増強する方法。
- 請求項1から14のいずれかに記載の医薬組成物と、免疫チェックポイント阻害剤とを含む、がん免疫療法のためのキット。
- 請求項1から14のいずれかに記載の医薬組成物と、免疫チェックポイント阻害剤の組合せの、がん免疫療法のための使用。
- セリン及びアルギニンリッチスプライシング因子(SRSFs)の活性を調節するスプライシング制御化合物の、腫瘍の免疫原性を向上させるための医薬組成物の製造のための使用。
- 腫瘍と、セリン及びアルギニンリッチスプライシング因子(SRSFs)の活性を調節するスプライシング制御化合物とを接触させることを含む、スプライス-ネオアンチゲンの製造方法
- 腫瘍が、セリン及びアルギニンリッチスプライシング因子(SRSFs)の活性を調節するスプライシング制御化合物と接触することで産生するスプライス-ネオアンチゲンを有効成分として含有する、前記腫瘍の免疫原性を向上させるための医薬組成物。
