Description
本発明は、糸状菌固体培養物に関する。
トウモロコシからコーンスターチを工業的に製造する方法の一つにウェットミリング法がある。ウェットミリング法では、(1)トウモロコシの精選、(2)浸漬、(3)胚芽の分離、(4)磨砕及び繊維分離、(5)デンプンとグルテンの分離、という5つの基本的な工程により、トウモロコシをデンプン、油(胚芽)、繊維、タンパクの4つの成分に分離させる。これらの工程では、コーングルテンミール、コーンジャーム、コーングルテンフィード等の副産物を含むコーンウェットミリング由来物が生じる。
その内、(2)浸漬の工程では、トウモロコシの粒を柔らかくし、各成分を分離しやすくするために、0.1~0.3%程度の亜硫酸水が用いられる。コーンスターチにおいては最終的に洗浄することで亜硫酸類を除去することが可能であるが、コーングルテンミール等のコーンウェットミリング由来物には比較的多量の亜硫酸類が残存してしまうため、洗浄を行っても除去するのが難しいという問題がある。
亜硫酸類は酸化や変色の防止等を目的として食品に用いられることもあるが、日本では特別に定められたものを除き、食品中の二酸化硫黄の残存量として30ppm未満の使用に制限されている。そのため、現在のところ、コーングルテンミール等のコーンウェットミリング由来物は主に家畜用の飼料として使用されているが、二酸化硫黄の残存量を規定量以下に抑えることができれば、食用とすることも可能である。
このような問題に対して、例えば特許文献1にはコーングルテンミール粉末を105℃~150℃の温度で加熱することにより二酸化硫黄含量を30ppm未満に低減する方法が提案されている。
また、特許文献2には、コーングルテンミール等のトウモロコシタンパク製品を酸化剤(好ましくは過酸化水素)で処理することで、遊離亜硫酸塩濃度を150ppm未満に減少させる方法が提案されている。
しかしながら、上記加熱におる低減方法では、タンパク質が変性・変質するおそれや着色するおそれがあり、また、コーングルテンミール粉末中の水分量に応じて加熱の温度や時間等の加熱条件を調整しなければならないという問題がある。また、日本において食品としては、上述のとおり、150ppm未満よりも更に低減する必要がある。
本発明は、上記に鑑みて提案されたものであり、二酸化硫黄濃度が低減されたコーンウェットミリング由来物を提供することを目的とする。
本発明者らは上記目的について鋭意研究した結果、コーンウェットミリング由来物を糸状菌で培養することで、コーンウェットミリング由来物中の二酸化硫黄濃度が低減されることを見出し、本発明を完成するに至った。より具体的には、本発明は以下のものを提供する。
<1> コーンウェットミリング由来物の糸状菌固体培養物。
<2> 上記コーンウェットミリング由来物が、コーングルテンミール、コーングルテンフィード、及びコーンジャームからなる群から選択される一種以上である、<1>に記載の糸状菌固体培養物。
<3> 二酸化硫黄の含有量が30ppm以下である、<1>又は<2>に記載の糸状菌固体培養物。
<4> 二酸化硫黄の含有量が10ppm以下である、<1>~<3>のいずれか一項に記載の糸状菌固体培養物。

本発明によれば、二酸化硫黄濃度が低減されたコーンウェットミリング由来物を提供することができる。
以下、本発明の実施形態について説明する。ただし、本発明は以下の実施形態に限定されるものではない。
≪1.糸状菌固体培養物≫
本実施形態に係る糸状菌固体培養物は、基質となるコーンウェットミリング由来物に糸状菌を接種し、固体培養を行って得られる培養物である。本明細書において、「コーンウェットミリング由来物」とは、トウモロコシをウェットミリング法で処理した際に生成される生成物をいう。コーンウェットミリング由来物としては特に限定されず、例えば、コーングルテンミール、コーングルテンフィード、コーンジャーム等が挙げられる。
本実施形態に係る糸状菌固体培養物は、基質となるコーンウェットミリング由来物に糸状菌を接種し、固体培養を行って得られる培養物である。本明細書において、「コーンウェットミリング由来物」とは、トウモロコシをウェットミリング法で処理した際に生成される生成物をいう。コーンウェットミリング由来物としては特に限定されず、例えば、コーングルテンミール、コーングルテンフィード、コーンジャーム等が挙げられる。
中でも、本発明の奏する効果が得られやすいという観点から、コーンウェットミリング由来物はコーングルテンミールであることが好ましい。コーングルテンミールとは、トウモロコシ粒からウェットミリング法によって分離される、タンパク質と澱粉とを主成分とする材料をいう。コーングルテンミールは、グルテンミールともいわれる。
他方で、上述したとおり、コーンスターチは洗浄により二酸化硫黄を除去するのが容易であるため、コーンウェットミリング由来物は、コーンスターチを含まないものであってもよい。
本明細書において、コーンウェットミリング由来物を「基質」ということもある。
固体培養の方法は特に限定されず、公知の方法を使用することができる。例えば、糸状菌の種菌(胞子)を基質へ接種し、培養床に盛り込み、厳密に温湿度管理された空気を基質へ通風させて培養する方法が挙げられる。
糸状菌固体培養物は、その培養物に含まれる二酸化硫黄の含有量が、例えば30ppm以下、好ましくは30ppm未満、より好ましくは20ppm以下、更に好ましくは10ppm以下、更により好ましくは10ppm未満である。このように、本実施形態に係る糸状菌固体培養物は、二酸化硫黄の含有量が低減されており、食品として用いることも可能である。
糸状菌固体培養物中の二酸化硫黄の含有量は、「第2版 食品中の食品添加物分析法」別添3に定める二酸化硫黄の測定法のうち比色法(分析法B)によって測定する。
糸状菌固体培養物を得るための培養に用いる糸状菌は、摂取した動物に対して害が無く、基質を資化して増殖するものであれば特に限定されない。そのような糸状菌としては、例えば、アスペルギルス属、モナスカス属等が挙げられる。これらの内、食用として用いるという観点から、糸状菌はカビ毒非生産菌であることが好ましい。カビ毒非生産菌としては、例えば、カビ毒の生合成に関わる遺伝子の変異、欠損、又は転写抑制などの遺伝要因の蓄積によって、カビ毒の生産に関連する遺伝子が発現されず、カビ毒の生産能が喪失している菌を好適に使用することができる。具体的には、例えば、アスペルギルス・オリゼー(Aspergillus・oryzae)、アスペルギルス・ソーヤ(Aspergillus・sojae)、アスペルギルス・リューチューエンシス(Aspergillus luchuensis)、アスペルギルス・ニガー(Aspergillus・niger)、アスペルギルス・アワモリ(Aspergillus・awamori)、モナスカス・ピローサス(Monascus pilosus)等が挙げられる。これらの糸状菌は、発酵食品の醸造用の種菌が市販されているし、独立行政法人製品評価技術基盤機構バイオテクノロジーセンター(NBRC)で分譲を受けることもできる。上記の糸状菌は、遺伝子の改変を行っていない野生株でもよいし、例えば後述する酵素の生産を増加させるように、遺伝子工学的な手法により遺伝子の改変を行ったものであってもよい。
カビ毒としては、例えば、アフラトキシン、デオキシニバレノール、オクラトキシン、フモニシン、ゼアラレノン、パツリン、ステリグマトシスチン、フザリウム・トキシン等が挙げられる。
糸状菌固体培養物は、糸状菌の生菌を含んでいるものであってもよく、糸状菌の死菌を含んでいるものであってもよく、それらの両方を含んでいるものであってもよい。
糸状菌固体培養物は、糸状菌が生産する酵素を含むものであることが好ましい。糸状菌が生産する酵素としては特に限定されないが、例えば分解酵素が挙げられる。分解酵素の例としては、アミラーゼ、アルカリ性プロテアーゼ、酸性プロテアーゼ、中性プロテアーゼ、キシラナーゼ、β-グルカナーゼ、セルラーゼ、タンナーゼ、フィターゼ、ラクターゼ、リパーゼ、ポリガラクチュロナーゼ等のペクチナーゼ;キシラナーゼ・ペクチナーゼ複合酵素、セルラーゼ・プロテアーゼ・ペクチナーゼ複合酵素などが挙げられる。
このような酵素が含まれることで、糸状菌固体培養物には機能性が付与される。例えば、酵素がフィターゼの場合、フィターゼはフィチン酸からリン酸を切り離す化学反応を触媒する。フィチン酸は、カルシウムや亜鉛などのミネラルが摂取した動物の体内に吸収されるのを阻害するといわれている。このため、フィターゼでフィチン酸を分解することにより、ミネラルの吸収率を向上させることができる。また、フィチン酸が分解して生じたリンも摂取した動物の体内に吸収させることができる。
また、例えば、酵素がセルラーゼやペクチナーゼなどの場合、それらの分解酵素はセルロースやペクチンなどを分解する反応を触媒する。セルロースやペクチンなどの多糖は、植物の細胞壁を構成する成分の一種である。植物の細胞壁を構成する多糖には色々な種類が知られており、その形態は様々であるが、その構成は複雑である。複雑な構造の細胞壁多糖を効率よく分解するためには、複数の分解酵素を段階的に作用させることが好ましい。例えば、セルラーゼやペクチナーゼなど複数の酵素でセルロースやペクチンなどを分解することにより、植物性の原料の細胞壁の分解効率が向上し、消化されやすくなる。
また、例えば、酵素がタンナーゼの場合、タンナーゼはタンニンを分解する反応を触媒する。タンニンは、たんぱく質などの高分子と強く結合し複合体を形成するものがある。また、タンニンは、植物の細胞壁を構成する成分と複雑に絡み合った状態で存在し、細胞壁の分解を阻害する可能性がある。タンナーゼでタンニンを分解することにより、植物性の原料の細胞壁の分解効率が向上し、消化されやすくなる。
公知の遺伝子工学的な手法を利用して、糸状菌において上記の酵素が高発現するように形質転換してもよい。糸状菌においては、例えば、アミラーゼ(AmyB)のプロモーターやエノラーゼ(enoA)のプロモーターが、高い発現量を有することが知られている。これらのプロモーターに、公知の手法を利用して、上記の酵素のうち目的の酵素をコードする遺伝子とプロモーターに対応するターミネーター配列とを結合して、キメラ遺伝子を得る。このキメラ遺伝子を公知の方法により、糸状菌に導入すれば、目的の酵素を高発現させた糸状菌を得ることができる。糸状菌は、ゲノム配列の解析が既に終了しており、その配列がデータベースで公開されている。そのようなデータベースを利用して、高発現プロモーター、ターミネーター、及び目的の酵素をコードする遺伝子の配列を調べて、プライマーを設計する。設計したプライマーとcDNA、ゲノムDNAなどのテンプレートとを利用してPCRにより所望の遺伝子配列を増幅して、形質転換に利用することができる。形質転換に際しては、公知のゲノム編集の手法を用いて、ゲノムの目的の位置に上記のキメラ遺伝子が導入されるようにしてもよいし、キメラ遺伝子を糸状菌の細胞内に導入してゲノムの任意の位置に上記のキメラ遺伝子が導入されるようにしてもよい。形質転換された糸状菌を選択培養するには、例えば、niaD、ptrA等の公知のマーカー遺伝子を利用すればよい。
高発現させる遺伝子をクローニングする際には、形質転換する糸状菌と同一種の糸状菌のゲノムDNA、cDNAをテンプレートにすることが好ましい。形質転換する糸状菌に組み込む遺伝子が同一種に属する糸状菌に由来するようにすれば、外来遺伝子が組み込まれないので、糸状菌の安全性が担保される。このようなクローニング手法をセルフクローニングと呼ぶ。
糸状菌固体培養物は、糸状菌の菌体を構成する多糖類を含むものであることが好ましい。菌体を構成する細胞は細胞壁で覆われており、細胞壁の主成分は多糖類である。多糖類としては、例えばグルカン、キチン、キトサン等が挙げられる。これらの多糖類は免疫賦活効果に関与していることが知られているため、多糖類を含む糸状菌固体培養物を摂取した場合には、免疫力を向上させる効果が期待される。また、糸状菌の菌体に含まれるタンパク質や脂質なども、摂取した動物の栄養分として吸収される。
また、コーングルテンミール等の副産物はタンパク質の割合が非常に多く、糖質が少ないことで糸状菌の増殖効率が低い場合がある。したがって、コーンウェットミリング由来物にでんぷん質を添加して固体培養を行ってもよく、糸状菌固体培養物はでんぷん質原料を含むものであってもよい。でんぷん質原料としては特に限定されず、例えば米糠、フスマ、米粉、小麦粉等が挙げられる。また、でんぷん質原料は、加熱して糊化したものであってもよい。
糸状菌が生産する酵素の種類やバランスは、基質のpHによって変化することが知られている。また、基質のpHを酸性(pH2~4)にすると、目的の糸状菌以外の雑菌が繁殖するのを抑制することができる。したがって、コーンウェットミリング由来物にpH調整剤を添加して固体培養を行ってもよく、糸状菌固体培養物はpH調整剤を含むものであってもよい。pH調整剤としては特に限定されず、例えばクエン酸、炭酸カルシウム、炭酸ナトリウム、リン酸、コハク酸、酒石酸、乳酸等が挙げられる。
糸状菌固体培養物はそのまま使用することもできるが、他の材料と混合した組成物とすることもできる。糸状菌培養物を含む組成物に含まれる他の成分は特に限定されず、例えば一般的な食品や飼料などに通常用いられる他の原料や添加剤を適宜配合できる。なお、配合量は目的とする効果に応じて適宜調整できる。また、酸化防止剤、pH調整剤、香料、各種エステル類、有機酸類、有機酸塩類、無機酸類、無機酸塩類、無機塩類、色素類、乳化剤、保存料、調味料、品質安定剤等の各種添加剤を含んでいてもよい。
糸状菌固体培養物又は上記組成物は、そのまま動物に与えることもできるし、他の食品や動物用飼料に含まれるものであってもよい。また、糸状菌固体培養物又は上記組成物は、他の食品や動物用飼料に混合するための食品添加剤や動物用飼料添加剤に含まれるものであってもよい。
糸状菌固体培養物を与えることができる対象は、特に限定されず、ヒト、ウシ、ブタ、ヤギ、家禽等の家畜;エビ、カニ等の甲殻類;魚類(養殖魚を含む);などが挙げられる。家禽には、例えば、鶏、鴨、アヒル、ガチョウ等が含まれる。
上述の点から明らかなとおり、本実施形態には、本実施形態に係る糸状菌固体培養物又は本実施形態に係る組成物の、食品、動物用飼料、食品添加剤、又は動物用飼料添加剤の製造のための使用が包含される。また、本実施形態には、本実施形態に係る糸状菌固体培養物又は本実施形態に係る組成物を用いる工程を含む、食品、動物用飼料、食品添加剤、又は動物用飼料添加剤の製造方法が包含される。本実施形態に係る糸状菌固体培養物又は本実施形態に係る組成物を用いる工程としては、例えば、本実施形態に係る糸状菌固体培養物又は本実施形態に係る組成物を準備する工程、本実施形態に係る糸状菌固体培養物又は本実施形態に係る組成物を、食品、動物用飼料、食品添加剤、又は動物用飼料添加剤を構成する他の材料と混合する工程等が挙げられる。
≪2.糸状菌固体培養物の製造方法≫
糸状菌固体培養物の製造方法は、コーンウェットミリング由来物に糸状菌を接種する糸状菌接種工程と、上記糸状菌を接種したコーンウェットミリング由来物を固体培養する固体培養工程とを含む。糸状菌固体培養物の製造方法は、上記糸状菌接種工程の前に、コーンウェットミリング由来物に加水して蒸煮・冷却する原料処理工程をさらに含むものであってもよい。
糸状菌固体培養の方法自体は特に限定されず、公知の方法を使用することができる。例えば、コーンウェットミリング由来物に加水して蒸煮・冷却する原料処理工程と、上記原料処理をしたコーンウェットミリング由来物に糸状菌を接種する糸状菌接種工程と、上記糸状菌を接種したコーンウェットミリング由来物を固体培養する固体培養工程とを含む方法が挙げられる。
糸状菌固体培養物の製造方法は、コーンウェットミリング由来物に糸状菌を接種する糸状菌接種工程と、上記糸状菌を接種したコーンウェットミリング由来物を固体培養する固体培養工程とを含む。糸状菌固体培養物の製造方法は、上記糸状菌接種工程の前に、コーンウェットミリング由来物に加水して蒸煮・冷却する原料処理工程をさらに含むものであってもよい。
糸状菌固体培養の方法自体は特に限定されず、公知の方法を使用することができる。例えば、コーンウェットミリング由来物に加水して蒸煮・冷却する原料処理工程と、上記原料処理をしたコーンウェットミリング由来物に糸状菌を接種する糸状菌接種工程と、上記糸状菌を接種したコーンウェットミリング由来物を固体培養する固体培養工程とを含む方法が挙げられる。
培養前のコーンウェットミリング由来物は二酸化硫黄を含むものであってもよく、その二酸化硫黄濃度は100ppm以上であってもよく、200ppm以上であってもよく、500ppm以上であってもよい。このようなコーンウェットミリング由来物に糸状菌を接種し、固体培養することで、糸状菌固体培養物における二酸化硫黄の濃度は30ppm以下に低減される。
原料処理工程において、コーンウェットミリング由来物を原料処理する方法は特に限定されず、公知の方法を使用することができる。例えば、コーンウェットミリング由来物に加水して撹拌した後、蒸煮処理を行い、基質に糸状菌を接種できる温度になるまで冷却する。なお、コーンウェットミリング由来物の語句は、上述の糸状菌固体培養物に関する説明に記載したとおりである。
糸状菌接種工程において、コーンウェットミリング由来物に糸状菌を接種する方法は特に限定されず、公知の方法を使用することができる。例えば、上述した原料処理工程後の基質に糸状菌の種菌(胞子)を散布して植菌する。また、接種する糸状菌は特に限定されないが、例えば糸状菌固体培養物に関する説明に記載した糸状菌を用いることができる。
基質を蒸煮する方法は特に限定されず、例えば、コーンウェットミリング由来物に対して好ましくは0.5~1.5倍量、より好ましくは0.7~1.3倍量の水を加えて撹拌した後に、好ましくは60~160℃、より好ましくは90~130℃の温度で、好ましくは0.5~90分間、より好ましくは2~60分間、蒸煮処理する方法が挙げられる。
また、蒸煮した基質に糸状菌を接種する際の胞子数は特に限定されず、例えば、基質1g当たり1.0×103~1.0×109個とすることが好ましく、基質1g当たり1.0×104~1.0×108個とすることがより好ましい。
固体培養工程において、糸状菌を接種したコーンウェットミリング由来物を固体培養する方法は特に限定されず、公知の方法を使用することができる。例えば、植菌した基質を
固体培養装置の培養床に堆積し、基質の粒子の間を温度と湿度が管理された空気が通過するようにして培養を行う方法が挙げられる。

固体培養工程では、糸状菌が増殖し酵素を生産する過程で、基質の品温が培養経過に伴い変化する。そこで、所望の品温経過で培養が進むよう、基質に供給する空気の温度、湿度、及び風量のうち少なくとも一つを調整し、固体培養するのが好ましい。これにより、培養中の正確な温湿度の管理が可能となり、雑菌の増殖を抑えつつ糸状菌を効率的に増殖させることができる。
また、基質の温度によって糸状菌が生産する酵素の種類が変化することが知られている。そのため、品温をコントロールすることにより、目的の酵素の生産を高めることができる。
上記基質の温度は特に限定されないが、10~55℃であることが好ましく、15~50℃であることがより好ましい。基質の温度は培養開始から一定時間経過するまでの間と、該一定時間の経過後で異なる温度としてもよく、例えば、培養開始から培養24時間までは30℃、培養24時間以降は25℃となるように調整する方法等が挙げられる。
また、上記空気の湿度は特に限定されないが、相対湿度で50~99%であることが好ましく、70~99%であることがより好ましい。
糸状菌固体培養物の製造において、基質の含水率を調整することが好ましい。含水率を調整する方法としては、散水又は乾燥による方法が挙げられる。散水又は乾燥は、固体培養工程中に行ってもよいし、固体培養工程が終わった後に行ってもよい。固体培養の効率を高める観点から、固体培養工程中に散水して基質の含水率を調整する方法が知られている。固体培養工程中に散水することにより、糸状菌の生育に適した水分活性を維持し、糸状菌の増殖や酵素生産をより活発にすることができる。また、糸状菌固体培養物の保存性を高める観点から、固体培養工程後に乾燥して基質の含水率を調整することができる。糸状菌固体培養物や混合した他の材料の劣化を防ぎ、品質の安定化及び長期間の保存が可能となる。
固体培養工程における培養時間は特に限定されず、例えば、基質の表面を糸状菌の菌糸が覆うまで培養するようにしてもよい。具体的な培養時間としては、30~100時間であってもよく、40~90時間であってもよい。
また、上述したように、コーンウェットミリング由来物はタンパク質の割合が非常に多く、糖質が少ないことにより糸状菌の増殖効率が低い場合があるため、ウェットミリング由来物にでんぷん質原料を添加して固体培養を行ってもよい。添加してもよいでんぷん質原料の具体例は、上記糸状菌固体培養物に関する説明に記載したとおりである。でんぷん質原料の添加は、原料処理工程時に行ってもよく、糸状菌接種工程時に行ってもよく、固体培養工程時に行ってもよく、そのすべての工程で行ってもよい。
また、上述したように、糸状菌が生産する酵素の種類やバランスは、基質のpHによって変化する。また、基質のpHを酸性(pH2~4)にすると、目的の糸状菌以外の雑菌が繁殖するのを抑制することができる。そのため、コーンウェットミリング由来物にpH調整剤を添加して固体培養を行ってもよい。pHの調整は、例えばpH調整剤を添加するなどの公知の方法を使用して行うことができる。pH調整剤の例は、上記糸状菌固体培養物に関する説明に記載したとおりである。pH調整は、原料処理工程時に行ってもよく、糸状菌接種工程時に行ってもよく、固体培養工程時に行ってもよく、そのすべての工程で行ってもよい。
以下、実施例を示し、本発明を具体的に説明するが、本発明はこれらの実施例に限定されるものではない。
<実施例1>
実施例1では、コーンウェットミリング由来物であるコーングルテンミールを原料とし、アスペルギルス・オリゼー(Aspergillus oryzae)を接種して固体培養を行った。
実施例1では、コーンウェットミリング由来物であるコーングルテンミールを原料とし、アスペルギルス・オリゼー(Aspergillus oryzae)を接種して固体培養を行った。
含水率10.8%のコーングルテンミール60kgに対して、67.4Lの水を加えて撹拌した後、100℃で30分間蒸煮処理した。蒸煮後のコーングルテンミールを35℃前後になるまで冷却した後、醤油用の種菌として市販されているアスペルギルス・オリゼー(株式会社樋口松之助商店、スリーダイヤ無添加)を接種し、均一に種付けされるように混合した。なお、接種した胞子数は、コーングルテンミール1g当たり約2×106個に相当する。また、種菌のアスペルギルス・オリゼーは、カビ毒非生産菌である。この種菌を接種した基質を、固体培養装置の培養床に盛り込み、堆積された基質の厚みが均一になるよう均してから培養を開始した。培養中は、温度及び湿度が管理された空気を培養床の下部から供給し、堆積された基質へ通風することにより、基質の品温を厳密に制御した。基質の品温は、培養開始から培養24時間までは30℃、培養24時間以降は25℃となるよう、供給する空気の温度及び湿度を管理した。また、供給する空気の相対湿度は、常に90~99%の範囲となるよう管理した。培養中には、固体培養装置に備え付けられている撹拌機を用いて、適宜基質を撹拌し、培養状態が均一になるようにした。培養は45時間行い、基質のコーングルテンミールの粒体表面を糸状菌の菌糸が十分に覆っていることを確認した。さらに、基質の含水率を調整する目的で、一晩送風乾燥を行い、糸状菌固体培養物を得た。
培養前のコーングルテンミール及び糸状菌固体培養物の二酸化硫黄の含有量は、「第2版 食品中の食品添加物分析法」別添3に定める二酸化硫黄の測定法のうち比色法(分析法B)により測定した。培養前のコーングルテンミールの二酸化硫黄の含有量は380ppmであり、糸状菌固体培養物の二酸化硫黄の含有量は21ppmであった。
<実施例2>
実施例2では、コーンウェットミリング由来物であるコーングルテンミールを原料とし、モナスカス・ピローサス(Monascus・pilosus)を接種して固体培養を行った。

含水率10.6%のコーングルテンミール15kgに対して、11.8kgの水を加えて撹拌した後、121℃で3分間加圧蒸煮処理した。蒸煮後のコーングルテンミールを35℃前後になるまで冷却した後、NBRCより入手したモナスカス・ピローサス(NBRC4520)を接種し、均一に種付けされるように混合した。なお、接種した胞子数は、コーングルテンミール1g当たり約2×106個に相当する。また、種菌のモナスカス・ピローサスは、カビ毒非生産菌である。この種菌を接種した基質を、固体培養装置の培養床に盛り込み、堆積された基質の厚みが均一になるよう均してから培養を開始した。培養中は、温度及び湿度が管理された空気を培養床の下部から供給し、堆積された基質へ通風することにより、基質の品温を厳密に制御した。基質の品温は、培養開始から培養終了まで30℃となるよう、供給する空気の温度及び湿度を管理した。また、供給する空気の相対湿度は、常に90~99%の範囲となるよう管理した。培養中には、適宜基質を撹拌し、培養状態が均一になるようにした。培養は89時間行い、基質のコーングルテンミールの粒体表面を糸状菌の菌糸が十分に覆っていることを確認した。糸状菌固体培養物を得た。さらに、基質の含水率を調整する目的で、一晩送風乾燥を行い、糸状菌固体培養物を得た。
培養前のコーングルテンミール及び糸状菌固体培養物の二酸化硫黄の含有量は、実施例1と同様の方法で測定した。培養前のコーングルテンミールの二酸化硫黄の含有量は340ppmであり、糸状菌固体培養物の二酸化硫黄の含有量は14ppmであった。
Claims (13)
- コーンウェットミリング由来物の糸状菌固体培養物。
- 前記コーンウェットミリング由来物が、コーングルテンミール、コーングルテンフィード、及びコーンジャームからなる群から選択される一種以上である、請求項1に記載の糸状菌固体培養物。
- 二酸化硫黄の含有量が30ppm以下である、請求項1又は2に記載の糸状菌固体培養物。
- 二酸化硫黄の含有量が10ppm以下である、請求項1~3のいずれか一項に記載の糸状菌固体培養物。
- 前記糸状菌がカビ毒非生産菌である、請求項1~4のいずれか一項に記載の糸状菌固体培養物。
- 前記カビ毒非生産菌が、アスペルギルス・オリゼー(Aspergillus oryzae)、アスペルギルス・ソーヤ(Aspergillus sojae)、アスペルギルス・リューチューエンシス(Aspergillus luchuensis)、アスペルギルス・ニガー(Aspergillus niger)、アスペルギルス・アワモリ(Aspergillus awamori)、及びモナスカス・ピローサス(Monascus pilosus)からなる群から選択される一種以上である、請求項5に記載の糸状菌固体培養物。
- 前記糸状菌が生産する酵素を含む、請求項1~6のいずれか一項に記載の糸状菌固体培養物。
- 前記糸状菌の菌体を構成する多糖類を含む、請求項1~7のいずれか一項に記載の糸状菌固体培養物。
- コーンウェットミリング由来物に糸状菌を接種して固体培養することを含む方法により得られる、糸状菌固体培養物。
- 請求項1~9のいずれか一項に記載の糸状菌固体培養物を含む、組成物。
- 請求項1~9のいずれか一項に記載の糸状菌固体培養物又は請求項10に記載の組成物を含む、食品、動物用飼料、食品添加剤、又は動物用飼料添加剤。
- コーンウェットミリング由来物に糸状菌を接種する糸状菌接種工程と、
前記糸状菌を接種したコーンウェットミリング由来物を固体培養する固体培養工程と、を含む、
糸状菌固体培養物の製造方法。 - 前記コーンウェットミリング由来物が二酸化硫黄を含む、請求項12に記載の糸状菌固体培養物の製造方法。
