磁性粒子から磁性被覆層が剥離しにくく、また磁性が強い磁性粒子を提供する。 コア粒子と前記コア粒子の表面に形成された磁性被覆層とを有する磁性粒子であって、前記コア粒子が排除限界分子量500以上の多孔質体である、磁性粒子。好ましい態様において、前記磁性被覆層は酸化鉄を含む。別の好ましい態様において、磁性粒子の粒径は0.1μm以上1000μm以下である。別の好ましい態様において、前記多孔質体は、セルロース等の多糖類又はその誘導体である。

Description

磁性粒子及びその製造方法
 本発明は、磁性粒子及びその製造方法に関する。
 生体試料等の分離・精製や、検体中の標的物質の検出を目的として、ナノサイズ~マイクロサイズの磁性粒子を担体として用いることが知られている。対象を磁性粒子表面のリガンド等に補足した後に磁石で容易に回収することができるため、診断・抽出の自動化などに応用されている(非特許文献1および2)。
 磁性粒子としては、マグネタイト等の粒子が用いられる他に、母核となるポリマー粒子の表面に磁性体微粒子を吸着させて被覆層を形成させた複合粒子も用いられる(特許文献1等)。この場合、磁性体微粒子の母核からの剥離・溶出を防ぐために、磁性粒子の表面をポリマーでさらに被覆することが提案されている。
 ところで、多孔性セルロース粒子などの多孔質体は、種々のクロマトグラフィー用充填剤としてその有用性が知られており、粒径や機械的強度等を制御して製造する方法や、様々なリガンドの導入等が研究されている(特許文献2等)。

 一般に、磁性粒子の使用場面において磁気分離による回収をより容易にする観点から、磁気応答性を高める、すなわち磁性を強くすることの要請がある。しかしながら、特許文献1のようにポリマー粒子を母核としてその表面に磁性体微粒子の被覆層が存する複合粒子では、ポリマー粒子から磁性体微粒子が剥離して溶出する場合があり、磁力による回収に十分な磁性が付与されなかったり耐久性に乏しかったりすることが問題となる。
 かかる状況に鑑みて、本発明は磁性粒子から磁性被覆層が剥離しにくく、また磁性が強い磁性粒子を提供することを課題とする。
 本発明者らは、特定の大きさの孔を有する多孔質体をコア粒子とし、その表面に磁性被覆層を形成させると、磁性被覆層が剥離しにくくまた磁性を強くできることを見出し、本発明を完成させた。
 すなわち、本発明は以下の通りである。
[1]コア粒子と前記コア粒子の表面に形成された磁性被覆層とを有する磁性粒子であって、
 前記コア粒子が排除限界分子量500以上の多孔質体である、磁性粒子。
[2]前記磁性被覆層が酸化鉄を含む、[1]に記載の磁性粒子。
[3]前記コア粒子の粒径が0.1μm以上1000μm以下である、[1]又は[2]に記載の磁性粒子。
[4]前記多孔質体が多糖類又はその誘導体、アクリルアミド系重合体、及びビニル系重合体から選択される、[1]~[3]の何れかに記載の磁性粒子。
[5]前記多糖類がセルロースである、[4]に記載の磁性粒子。
[6]コア粒子と水溶性鉄塩とを含有する水溶液にアルカリを添加する工程を含む、磁性粒子の製造方法であって、
 前記コア粒子が排除限界分子量500以上の多孔質体である、製造方法。
[7]前記コア粒子の粒径が、0.1μm以上1000μm以下である、[6]に記載の製造方法。
[8]前記多孔質体が多糖類又はその誘導体、アクリルアミド系重合体、及びビニル系重合体から選択される、[6]又は[7]に記載の製造方法。
[9]前記多糖類がセルロースである、[8]に記載の製造方法。
 本発明により、十分に強い磁性を有する磁性粒子が提供される。本発明の磁性粒子においては、コア粒子である多孔質体の表面は多数の孔が存するため表面積が大きく、磁性被覆層がコア粒子に強固に嵌合することになる。そのため、磁性被覆層の上にさらにコーティングをせずとも磁性被覆層が剥離しにくく、また磁性が強いものとなる。
 以下、本発明の実施の形態について詳細に説明する。
 本発明の磁性粒子は、コア粒子と前記コア粒子の表面に形成された磁性被覆層とを有し、前記コア粒子は排除限界分子量500以上の多孔質体である。
 前記多孔質体としては、特に限定されず、多糖類又はその誘導体、スチレン系重合体、アクリレート系重合体、メタクリレート系重合体、アリル系重合体、ビニル系重合体、アクリルアミド系重合体、シリカ等が挙げられるが、価格の面から多糖類又はその誘導体がより好ましい。
 多糖類は特に制限はないが、例えば、セルロース、グアーガム、スターチ、ヒドロキシエチルセルロース、ヒドロキシエチルグアーガム、ヒドロキシエチルスターチ、メチルセルロース、メチルグアーガム、メチルスターチ、エチルセルロース、エチルグアーガム、エチルスターチ、ヒドロキシプロピルセルロース、ヒドロキシプロピルグアーガム、ヒドロキシプロピルスターチ、ヒドロキシエチルメチルセルロース、ヒドロキシエチルメチルグアーガム、ヒドロキシエチルメチルスターチ、ヒドロキシプロピルメチルセルロース、ヒドロキシプロピルメチルグアーガム、ヒドロキシプロピルメチルスターチ等が挙げられ、これらの中では、セルロースがより好ましい。
 多糖類誘導体としては、前述したセルロース等の多糖類の水酸基の水素原子が、メチル基、エチル基、プロピル基等の低級アルキル基;ヒドロキシメチル基、ヒドロキシエチル基、ヒドロキシプロピル基等のヒドロキシ低級アルキル基;アセチル基、乳酸基等の低級アシル基等の官能基から選択される一種又は二種以上で置き換えられたものが包含される。具体的な化合物としては酢酸セルロース、乳酸セルロース等が挙げられる。
 本発明における多孔質体は、通常は連続孔または独立孔を多数有するものを指す。
 多孔質体の排除限界分子量は、一般に孔径の大きさと相関し、本発明においては一定以上の孔径を有する多孔質体をコア粒子とすることで、その表面の凹凸を覆う磁性被覆層が強固に嵌合し、また大きな表面積のために磁性被覆層が十分な磁性を付与することとなる。
 本発明における多孔質体の排除限界分子量は500以上であり、好ましくは1,000以上、より好ましくは5,000以上、さらに好ましくは100,000以上である。また、その上限は特に限定されないが、好ましくは500,000以下、より好ましくは350,000以下である。
 排除限界分子量は、分子量と標準物質の溶出体積またはカラム体積の関係から求めることができる。排除限界分子量の測定方法は、例えば、生物化学実験法11「ゲル濾過法」第2版(学会出版センター)や、Amaersham Biosciences(現GEヘルスケアバイオサイエンス株式会社)の資料「Gel Filtration Principles and Methods」の記載を参照することができる。
 本明細書における排除限界分子量は、以下の方法で測定した値とする。
 内径11mmのカラムに多孔質体をおよそ20cmの高さに充填し、純水を通液して平衡化し多孔質体のゲルベッドを安定化させる。安定化後の充填カラムを、株式会社島津製作所製のHPLCシステム(CLASS-VP)にセットして純水を流し平衡化する。なお、検出器として示差屈折計(RI)を使用する。25℃で流速0.4mL/分で、表1に示す分子量標準液10μLを注入してそれぞれの溶出時間を測定し、Kav値を計算する。分子量標準は、表1に示す分子量標準試薬を所定の濃度で純水に溶解したものを用いる。

 Kav値は、下記の式(1)により算出する。
 Kav= (V- V)/(V- V)・・・(1)
 ここで、VはDextran2000の溶出体積、Vはカラムベッドの体積(カラム断面積×ゲル高さ)、Vは分子量標準の溶出体積である。
 分子量標準の分子量値の対数をX軸、Kav値をY軸にプロットして、高分子量側で直線領域を示すデータポイントについて、最小二乗法で一次式の当てはめを行い、下記の一次式(2)を得る。
 Kav= a・Log MW + b・・・(2)
 X軸と上記一次式(2)の直線が交わる点が排除限界分子量を指す。すなわち、排除限界分子量は式(2)においてKav=0の時のLog MW値であるため、下記式(3)から算出される。
 排除限界分子量の対数値 =b/-a・・・(3)
 本発明の磁性粒子の形状は、球状、略球状、立方体、円柱、多角柱、不定形のものなど特に限定されないが、球状又は略球状であることが好ましい。
 また、磁性粒子の粒径は、0.1μm以上が好ましく、10μm以上がより好ましく、20μm以上がさらに好ましく、40μm以上が特に好ましい。また、粒径の上限は、1000μm以下が好ましく、500μm以下が好ましく、350μm以下がさらに好ましく、200μm以下が特に好ましい。なお、磁性被覆層により粒子に磁性が付与される限りにおいて磁性被覆層の厚さは特に限定されず、コア粒子の粒径と磁性粒子の粒径との差がごく小さくても構わない。
 磁性粒子の大きさをこのような範囲とすることにより、磁力による磁性粒子の液体中からの回収(磁気分離)を効率的に行うことができる。特に、磁性粒子の粒径が0.1μm未満の場合は、回収に長時間を要するため、磁気分離に供する磁性粒子の大きさとして好ましくない。
 また、本発明においてコア粒子の粒径は、0.1μm以上が好ましく、10μm以上がより好ましく、20μm以上がさらに好ましく、40μm以上が特に好ましい。また、粒径の上限は、1000μm以下が好ましく、500μm以下がより好ましく、350μm以下がさらに好ましく、200μm以下が特に好ましい。
 なお、上記コア粒子の粒径は、湿潤状態で測定された値である。
 磁性粒子又はコア粒子の粒子径は、電気抵抗法を用いて測定された粒子径(算術径)から算出することができる。電気抵抗法は、粒子が感応領域を通過する際に生じる2電極間の電気抵抗の変化を利用する方法である。電気抵抗は粒子の体積に比例することから、電気抵抗の変化を測定し、これを粒子径に換算することによって、粒子径を測定することができる。また、平均粒子径は、前述の電気抵抗法による「算術径」値の全データを平均して求めることができる。測定装置としては、ベックマンコールター株式会社の精密粒度分布測定装置(製品名「Multisizer3」)などを用いることができる。
 あるいは、粒子を光学顕微鏡で撮影した画像において、ノギスなどを用いて粒子径を計測して、撮影倍率から元の粒子径を求めることもできる。そして、光学顕微鏡写真から求めたそれぞれの粒子径の値から、下記の式によって平均粒子径を算出することができる。
 個数平均粒子径(M) = Σ(nd)/Σ(n)

(式中、ndは光学顕微鏡写真から求めたそれぞれの粒子径の値を表し、nは、測定した粒子の個数を表す。)
 なお、本発明において、粒子径は電気測定法を用いて測定した。
 本発明における磁性被覆層は、磁気誘導により容易に磁化されうる材料を含み、かかる材料としては特に限定されないが、例えば、酸化鉄(四酸化三鉄、三酸化二鉄等)、鉄、マンガン、ニッケル、酸化ニッケル、各種フェライト、コバルト、コバルト鉄酸化物、クロム、バリウムフェライト、炭素鋼、タングステン鋼、KS鋼、希土類コバルト磁石、ヘマタイト等が挙げられる。これらのうち酸化鉄を含むことがより好ましく、四酸化三鉄を含むことがさらに好ましい。
 磁性被覆層は、コア粒子の表面に形成されており、被覆の態様はコア粒子に対する物理吸着でも化学結合のいずれでも構わない。
 前述のとおり本発明におけるコア粒子は多孔質体であるため、通常、磁性被覆層はコア粒子の孔に入り込むようにコア粒子の表面の凹凸を覆う形態である。それにより磁性被覆層はコア粒子に強固に嵌合し剥がれにくくなり、また粒子に強い磁性が付与される。
 本発明の磁性粒子における磁性被覆層は、コア粒子の表面全体を覆っていてもよく、コア粒子の表面の一部を覆っていてもよい。
 本発明の粒子における磁性被覆層による被覆量は特に規定されないが、磁性の確保の観点から、体積磁化率として好ましくは0.1emu/cm以上、より好ましくは0.3emu/cm以上、さらに好ましくは0.5emu/cm以上の磁性を示す被覆量である。
 本発明の磁性粒子は、任意の方法で製造することができる。
 例えば、コア粒子表面に、前述の磁気誘導により容易に磁化されうる材料を析出させることにより磁性被覆層を形成させる方法によって製造する方法が挙げられる。
 以下に、磁性被覆層が酸化鉄を含む場合の製造方法について例示するが、本発明の磁性粒子の製造方法はこれに限定されない。
 本発明の磁性粒子は、コア粒子と水溶性鉄塩とを含有する水溶液にアルカリを添加する工程を含む、磁性粒子の方法により製造することができる。
 本方法において、水溶性鉄塩としては、塩化鉄、硫酸鉄、硝酸鉄等が挙げられる。
 前記水溶液のアルカリ添加前のpHは、通常6以下であることが好ましい。
 また、前記水溶液の温度は特に限定されないが60~90℃とすることが好ましい。なお、かかる温度に調整することは、アルカリを添加する前でも後でもよい。
 アルカリは、例えば水酸化ナトリウム水溶液を用いることができ、アルカリの添加により水溶液のpHを12以上とすることが好ましい。
 本方法におけるコア粒子や多孔質体についての説明は前述のとおりである。
 かかる工程により、コア粒子上で酸化鉄が析出し、コア粒子表面に磁性被覆層が形成される。
 その後、精製水等によって粒子を洗浄してもよい。
 本発明の磁性粒子は、磁性被覆層の上にさらに任意の他の被覆層を設けたり、官能基やリガンド等の任意の修飾を施してもよい。
 以下、具体的な実験例をあげて、本発明をさらに詳細に説明するが、本発明は以下の態様にのみ限定されない。
<実施例1>
 500mLのフラスコにMilli-Q水を100mL、コア粒子として湿潤状態で平均粒径が100μmであり、排除限界分子量が300,000である多孔性セルロースA(商品名:セルファイン、JNC株式会社製)を5.0g入れた。次に、塩化第二鉄・六水和物(2.0mol/L)及び塩化第一鉄・四水和物(1.0mol/L)混合水溶液を5.0mL入れ、メカニカルスターラー(LABORATORYHIGHT POWER MIXER、ASONE社製)で攪拌した。この混合溶液を70℃に昇温した後、水酸化ナトリウム水溶液(1.0mol/L)を40.0mL滴下し、1時間程度反応させた後撹拌を停止し、磁性粒子Aの粗分散液を得た。
 なお、「Milli-Q水」は、メルク社製超純水製造装置「IQ7003」によって精製された、導電率18MΩcmの精製水を指す。
 得られた磁性粒子Aの粗分散液を50μmメッシュの金網にて篩い、多孔質セルロースAに吸着していない余分な磁性粒子を十分なMilli-Q水にて洗い流し、洗浄液が目視にて透明になった時点で、金網上に残った粒子を磁性粒子Aとして回収した。回収した磁性粒子Aは、乾燥を防ぐためにMilli-Q水に保存し、磁性粒子A分散液とした。
 得られた磁性粒子A分散液から、磁気天秤(商品名:AUTO、SherwoodScientific社製)付属のサンプル管(商品名:太め管、Sherwood Scientific社製)に、粒子の高さが5.0mmになるように、ネオジム磁石(株式会社二六製作所、NE038)を用いて余分な水を抜きながら詰め込んだ。その後、同磁気天秤にて体積磁化率を測定したところ、多孔質セルロースAをコアに使用した磁性粒子Aは、0.89emu/cmを示した。ネオジム磁石への集積、磁気天秤での定量化から、磁性粒子Aが磁化されていることを確認した。
<実施例2>
 コア粒子として、多孔性セルロースAの代わりに、湿潤状態で平均粒径が70μmであり、排除限界分子量が900である多孔性セルロースB(商品名セルファイン、JNC株式会社製)を使用した以外は、実施例1と同様の方法にて磁性粒子Bを作製・洗浄し、磁性粒子B分散液を得た。
 実施例1と同様の方法にて磁性粒子Bの体積磁化率を測定したところ、0.57emu/cmを示した。ネオジム磁石への集積、磁気天秤での定量化から、磁性粒子Bが磁化されていることを確認した。
<実施例3>
 コア粒子として、多孔性セルロースAの代わりに、湿潤状態で平均粒径が230μmであり、排除限界分子量が300,000である多孔性セルロースC(商品名:セルファイン、JNC株式会社製)を使用した以外は、実施例1と同様の方法にて磁性粒子Cを作製・洗浄し、磁性粒子C分散液を得た。
 実施例1と同様の方法にて磁性粒子Cの体積磁化率を測定したところ、0.61emu/cmを示した。ネオジム磁石への集積、磁気天秤での定量化から、磁性粒子Cが磁化されていることを確認した。
<実施例4>
 コア粒子として、多孔性セルロースAの代わりに、湿潤状態で平均粒径が50μmであり、排除限界分子量が250,000である多孔性アクリルアミド(商品名:セファクリル S-200HR、グローバルライフサイエンステクノロジーズジャパン株式会社製)を使用し、50μmメッシュの金網による篩いをろ紙(商品名:定量濾紙No.5A、アドバンテック東洋株式会社製)を用いた吸引ろ過に変えた以外は、実施例1と同様の方法にて磁性粒子Dを作製・洗浄し、磁性粒子D分散液を得た。
 実施例1と同様の方法にて磁性粒子Dの体積磁化率を測定したところ、1.58emu/cmを示した。ネオジム磁石への集積、磁気天秤での定量化から、磁性粒子Dが磁化されていることを確認した。
<実施例5>
 コア粒子として、多孔性アクリルアミドDの代わりに、湿潤状態で平均粒径が45μmであり、排除限界分子量が500,000である多孔性ビニルポリマー(商品名:トヨパール HW-65F、東ソー株式会社製)を使用した以外は、実施例4と同様の方法にて磁性粒子Eを作製・洗浄し、磁性粒子E分散液を得た。
 実施例1と同様の方法にて磁性粒子Eの体積磁化率を測定したところ、0.70emu/cmを示した。ネオジム磁石への集積、磁気天秤での定量化から、磁性粒子Eが磁化されていることを確認した。
<比較例1>
 コア粒子として多孔性セルロースAの代わりに、湿潤状態で平均粒径が80μmであり、排除限界分子量が300である多孔性セルロースF(商品名セルファイン、JNC株式会社製)を使用した以外は、実施例1と同様の方法にて磁性粒子Fを作製・洗浄し、磁性粒子E分散液を得た。しかし、分散液取得72時間経過後に再び磁性粒子F分散液を観察したところ、上澄み液が褐変していた。これは、表面の磁性被覆層が剥離しその断片が上澄み液に溶出したためと予想された。
 実施例1と同様の方法にて、磁性粒子Fの体積磁化率測定を試みたところ、ネオジウム磁石を近づけても、磁性粒子Fはほとんど反応を示さず、余分な水を抜けないため、磁気天秤付属のサンプル管に粒子を詰め込めなかった。この結果から、磁性粒子Fは、ネオジム磁石で集積可能な一般的な磁性粒子として扱えるほど磁化されていないと判断した。
<比較例2>
 コア粒子として多孔性セルロースAの代わりに、湿潤状態で平均粒径が80μmであり、排除限界分子量が220未満であるポリエチレンG(商品名白色ポリエチレン粒子、Cospheric LLC製)を使用した以外は、実施例1と同様の方法にて磁性粒子Gを作製・洗浄し、磁性粒子F分散液を得た。しかし、分散液を観察したところ、磁性粒子Gは一部灰色を示すものの、大部分はポリエチレンGとほぼ同等の白色を示し、これは、表面の磁性被覆層自体が十分に形成されていないと予想された。
 実施例1と同様の方法にて、磁性粒子Gの体積磁化率測定を試みたところ、ネオジウム磁石を近づけても、磁性粒子Gはほとんど反応を示さず、余分な水を抜けないため、磁気天秤付属のサンプル管に粒子を詰め込めなかった。この結果から、磁性粒子Gは、ネオジム磁石で集積可能な一般的な磁性粒子として扱えるほど磁化されていないと判断した。

Claims (9)

  1.  コア粒子と前記コア粒子の表面に形成された磁性被覆層とを有する磁性粒子であって、 前記コア粒子が排除限界分子量500以上の多孔質体である、磁性粒子。
  2.  前記磁性被覆層が酸化鉄を含む、請求項1に記載の磁性粒子。
  3.  前記磁性粒子の粒径が0.1μm以上1000μm以下である、請求項1又は2に記載の磁性粒子。
  4.  前記多孔質体が多糖類又はその誘導体、アクリルアミド系重合体、及びビニル系重合体から選択される、請求項1~3の何れか一項に記載の磁性粒子。
  5.  前記多糖類がセルロースである、請求項4に記載の磁性粒子。
  6.  コア粒子と水溶性鉄塩とを含有する水溶液にアルカリを添加する工程を含む、磁性粒子の製造方法であって、
     前記コア粒子が排除限界分子量500以上の多孔質体である、製造方法。
  7.  前記コア粒子の粒径が、0.1μm以上1000μm以下である、請求項6に記載の製造方法。
  8.  前記多孔質体が多糖類又はその誘導体、アクリルアミド系重合体、及びビニル系重合体から選択される、請求項6又は7に記載の製造方法。
  9.  前記多糖類がセルロースである、請求項8に記載の製造方法。
