Abstract

PURPOSE:To efficiently separate and purify a biomembrane without deforming spherical particles even under pressure, by passing a solution containing the biomembrane through a column packed with spherical particles of crosslinked glucomannan. CONSTITUTION:Glucomannan is swollen with formamide, treated with acetic acid, etc., in the presence of an esterifying catalyst to give a glucomannan ester, which is dissolved in a solvent such as dichloromethane. In the operation, a simultaneous addition of 100-500wt.% based on the glucomannan of a diluent (e.g. THF) can provide a gel having >=1,000,000 exclusion limit mol.wt. Then the solution is stirred in an aqueous medium to give spherical particles having 1-500mum particle diameter. The particles are packed into a column, a solution containing a biomembrane (e.g. natural liposome, synthetic bimolecular membrane or multi-layer liposome) is passed through the column and separated.
