Abstract
本発明の目的は、(S)n構造を有するポリスルフィドを効率的に標識するアルキル化剤を提供することである。 前記課題は、本発明の下記式(1):(式中、R1及びR2は、それぞれ独立して水素原子、炭素数1~6のアルキル基、置換基を有する炭素数1~6のアルキル基、炭素数3~7のシクロアルキル基、置換基を有する炭素数3~7のシクロアルキル基、炭素数1~6のアルコキシ基、又は置換基を有する炭素数1~6のアルコキシ基であり、R1及びR2は、R1及びR2が結合している炭素原子と一緒になって炭素数3~7のシクロアルキル基、置換基を有する炭素数3~7のシクロアルキル基、炭素数6~12のアリール基、置換基を有する炭素数6~12のアリール基、3~7員の複素環基、又は置換基を有する3~7員の複素環基であり、Xは、ハロゲン原子、メタンスルホニルオキシ基、トリフルオロメタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基であり、R1及びR2がいずれも水素原子の場合、Xはヨウ素原子以外のハロゲン原子、メタンスルホニルオキシ基、トリフルオロメタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基である)で表されるポリスルフィド標識用官能基によって解決することができる。
Description
本発明は、ポリスルフィド標識用アルキル化剤及びそれを用いたポリスルフィドの標識方法に関する。本発明によれば、ポリスルフィド構造を正確に標識および検出することができる。
近年、硫黄原子が連結したパースルフィド、トリスルフィド、又はテトラスルフィドなどのポリスルフィド構造を含む分子種が、生体内に多く存在していることが明らかになっている。例えば、チオール(-SH)基に複数の硫黄原子が付加したポリスルフィド構造(-(S)n-SH)を有するシステインやその酸化型構造(-S-(S)n-S-)を含むタンパク質が報告されており、これらのタンパク質は、高い求核性又は抗酸化活性を示すことが報告されている。しかしながら、複雑な化学特性を有するポリスルフィドは、検出が難しいことからその生理機能は十分に解明されていない。

ポリスルフィドの検出方法としては、チオール基を、例えばヨードアセトアミド(IAM)又はβ-(4-ヒドロキシフェニル)エチルヨードアセトアミド(HPE-IAM)で修飾して、質量分析により検出する方法が知られている(非特許文献1及び2)。しかしながら、従来のチオール基標識用のアルキル化剤は、十分にポリスルフィドを検出(標識)できなかった。
従って、本発明の目的は、(S)n構造を有するポリスルフィドを効率的に標識するアルキル化剤を提供することである。また、(S)n構造を有するポリスルフィドを効率的に標識するポリスルフィド標識方法及びポリスルフィドの検出方法を提供することである。

本発明者は、(S)n構造を有するポリスルフィドを効率的に標識するアルキル化剤について、鋭意研究した結果、驚くべきことに、アルキル化剤であるヨードアセトアミドを改変することにより、ポリスルフィドを効率的に標識できるアルキル化剤が得られることを見出した。
本発明は、こうした知見に基づくものである。
従って、本発明は、

本発明者らは、従来のアルキル化剤が、ポリスルフィドを十分に検出(標識)できない理由は、アルキル化剤の反応性が高く、ポリスルフィドを含まないチオール基にも十分反応してしまい、識別が困難となるため、あるいは還元型ポリスルフィド構造(-(S)n-SH)の(S)n構造を分解する可能性があるためであると考えている。本発明のポリスルフィド標識用アルキル化剤は、ポリスルフィドを含まないチオール基との反応を抑制しつつ、ポリスルフィド構造の分解を抑制することによって、ポリスルフィドを十分に標識できると考えられる。
本発明は、こうした知見に基づくものである。
従って、本発明は、

本発明者らは、従来のアルキル化剤が、ポリスルフィドを十分に検出(標識)できない理由は、アルキル化剤の反応性が高く、ポリスルフィドを含まないチオール基にも十分反応してしまい、識別が困難となるため、あるいは還元型ポリスルフィド構造(-(S)n-SH)の(S)n構造を分解する可能性があるためであると考えている。本発明のポリスルフィド標識用アルキル化剤は、ポリスルフィドを含まないチオール基との反応を抑制しつつ、ポリスルフィド構造の分解を抑制することによって、ポリスルフィドを十分に標識できると考えられる。
本発明のポリスルフィド標識用アルキル化剤によれば、従来のアルキル化剤と比較して、効率的にポリスルフィドを標識及び検出することができる。また、ポリスルフィドを検出することによって、ポリスルフィドタンパク質が関係する病態メカニズムの解明、並びに酸化ストレス研究、及びシグナル伝達研究などの様々な生化学研究に貢献することができる。
[1]ポリスルフィド標識用アルキル化剤
本発明のポリスルフィド標識用アルキル化剤は、ポリスルフィド構造の標識に適した官能基を有する。前記ポリスルフィド標識用官能基は、ポリスルフィドを含まないチオール基との反応を抑制しつつ、ポリスルフィド基を標識できる基であり、またポリスルフィド構造(-(S)n-SH)の(S)n構造の過剰な分解が抑制される基である。本発明のポリスルフィド標識用官能基を有することにより、ポリスルフィド標識用アルキル化剤は、効率的にポリスルフィドを標識し、そしてポリスルフィドを検出することができる。
本発明のポリスルフィド標識用アルキル化剤は、ポリスルフィド構造の標識に適した官能基を有する。前記ポリスルフィド標識用官能基は、ポリスルフィドを含まないチオール基との反応を抑制しつつ、ポリスルフィド基を標識できる基であり、またポリスルフィド構造(-(S)n-SH)の(S)n構造の過剰な分解が抑制される基である。本発明のポリスルフィド標識用官能基を有することにより、ポリスルフィド標識用アルキル化剤は、効率的にポリスルフィドを標識し、そしてポリスルフィドを検出することができる。
《ポリスルフィド標識用官能基》
本発明のポリスルフィド標識用官能基は、下記式(1):
で表される基である。本発明のポリスルフィド標識用官能基は、限定されるものではないが、還元型ポリスルフィド標識用又は酸化型ポリスルフィド標識用である。
R1及びR2は、Xと比較して、ポリスルフィド構造との反応性が高い官能基を有さない限りにおいて限定されるものではない。
R1及びR2は、例えば、それぞれ独立して水素原子、炭素数1~6のアルキル基、置換基を有する炭素数1~6のアルキル基、炭素数3~7のシクロアルキル基、置換基を有する炭素数3~7のシクロアルキル基、炭素数1~6のアルコキシ基、又は置換基を有する炭素数1~6のアルコキシ基である。
炭素数1~6のアルキル基は、好ましくは炭素数1~3のアルキル基であり、より好ましくは炭素数1~3の直鎖状のアルキル基である。炭素数1~6のアルキル基としては、具体的にはメチル基、エチル基、ノルマルプロピル基、イソプロピル基、ノルマルブチル基、イソブチル基、第二級ブチル基、第三級ブチル基、ノルマルペンチル基、イソペンチル基、ネオペンチル基、第三級ペンチル基、ノルマルへキシル基、及びイソへキシル基が挙げられる。
炭素数3~7のシクロアルキル基は、好ましくは炭素数4~6のシクロアルキル基である。炭素数3~7のシクロアルキル基としては、具体的にはシクロプロピル基、シクロブチル基、シクロペンチル基、シクロヘキシル基、又はシクロヘプチル基が挙げられる。

前記置換基を有する炭素数1~6のアルキル基、置換基を有する炭素数3~7のシクロアルキル基、又は置換基を有する炭素数1~6のアルコキシ基は、それぞれの基の水素原子が、置換基に置換された基である。なお、「置換基を有する炭素数1~6のアルキル基」の炭素数1~6には、置換基の炭素数は含まない。

3~7員の複素環基は、特に限定されるものではないが、3~6員の複素環が好ましい。複素環に含まれるヘテロ原子としては酸素原子(O)、窒素原子(N)、又は硫黄原子(S)が挙げられる。複素環に含まれるヘテロ原子の数も限定されないが、上限は好ましくは4以下であり、より好ましくは3以下であり、更に好ましくは2以下である。また、複素環基は、飽和の複素環基でもよく、不飽和の複素環基でもよい。3~7員の複素環としては、具体的には、オキセタン、オキセテン、ピロリジン、ピロール、テトラヒドロフラン、テトラヒドロチオフェン、イミダゾリジン、オキサゾリジン、ジオキソラン、ピペリジン、テトラヒドロピラン、ピラン、チアン、チオピラン、モルホリン、オキサジン、ジオキサン、ジオキシン、アゼパン、オキセパン、チエパンが挙げられる。
Xは、ハロゲン原子、メタンスルホニルオキシ基(OSO2CH3)、トリフルオロメタンスルホニルオキシ基(OSO2CF3)、又はパラトルエンスルホニルオキシ基(OSO2C6H4CH3)であり、好ましくはハロゲン原子である。ハロゲン原子としては、フッ素原子、塩素原子、臭素原子、又はヨウ素原子が挙げられるが、好ましくは、塩素原子、又は臭素原子である。
R1及びR2がいずれも水素原子の場合、Xはヨウ素原子以外のハロゲン原子、メタンスルホニルオキシ基、トリフルオロメタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基であり、好ましくはヨウ素原子以外のハロゲン原子、メタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基であり、より好ましくはヨウ素原子以外のハロゲン原子である。
本発明のポリスルフィド標識用官能基を含む化合物は、例えば下記式(2):

R3は、Xと比較して、ポリスルフィド構造との反応性が高い官能基を有さない限りにおいて限定されるものではない。また、R3は、ポリスルフィドとの反応基であるXから離れているため、本発明の官能基と、ポリスルフィド構造との反応へほとんど影響を与えない。従って、Xと比較して、ポリスルフィド構造との反応性が高い官能基を有さない限りにおいて限定されるものではないが、例えば、炭素数1~100のヘテロ原子を含むことのある炭化水素基(ヒドロカルビル基)が挙げられる。ヘテロ原子としては、硫黄原子、酸素原子、又は窒素原子が挙げられる。R3に含まれるヘテロ原子の数は限定されるものではないが、硫黄原子、酸素原子、又は窒素原子はそれぞれ独立して、0~20であり、好ましくは0~10であり、より好ましくは0~5である。

前記R3の基として、例えばビオチンを含む基を用いることができる。ビオチンは、アビジンと高い親和性を持つため、例えばアビジンビーズを用いることで、夾雑試料の中からビオチン構造を含む物質のみをアビジンビーズに結合させ、これを遊離させることにより、ビオチン構造を含む化合物を精製した状態で様々な分析に用いることができる。すなわち、R3の基としてビオチンを含む基を用いることによって、ポリスルフィド構造を含むタンパク質やそのペプチド消化物の精製が可能となり、分析感度を向上させることができる。
前記R3の基として、例えばアルキニル基を含む基を用いることができる。アルキニル基は、例えばアジド基と、銅触媒存在下でトリアゾール環化反応(ヒュスゲン環化付加反応、又はクリックケミストリー)を起こし、結合する。従って、アジド基を含む任意の化合物を容易に結合させることができる。すなわち、R3の基としてアルキニル基を含む基を用いることによって、さまざまな物質をアルキル化剤に結合させることができる。
また、R3の基として、安定同位体を含む基を用いることもできる。R3の基の一部の元素を安定同位体元素(重元素)に置き換えることで、安定同位体非含有標識試薬(軽元素)と同じ分子構造で異なる質量数のタグペアを調製することができる。本発明においても、ポリスルフィド標識分子の一部の元素を安定同位体元素に置き換えることで、特定のタンパク質やペプチド消化物のポリスルフィド含有率を相対的に定量することが可能となる。安定同位体元素としては重元素が用いられ、具体的には、2H、13C、15N、18O、又は34Sなどが挙げられるが、これらに限定されない。
本発明のアルキル化剤は、前記化合物の塩を含んでもよい。具体的な塩としては、無機塩基、有機塩基、又は金属アルコキシドとの塩が挙げられる。塩は、前記化合物と、無機塩基、有機塩基、又は金属アルコキシドとの混合により生成しうる。
塩を形成しうる無機塩基としては、アルカリ金属(例えば、リチウム、ナトリウム、又はカリウム等)の水酸化物、炭酸塩、炭酸水素塩、酢酸塩、又は水素化物;アルカリ土類金属(例えば、マグネシウム、カルシウム、又はバリウム)の水酸化物、又は水素化物等が挙げられる。

本発明のアルキル化剤は、タンパク質などの還元型ポリスルフィド構造(R4-(S)n-SH)のチオール基(-SH基)を修飾することができる。具体的には、本発明のポリスルフィド標識用官能基のXと、チオール基の水素原子が、HXとなって脱離し、チオール基がアルキル化剤によって修飾される。具体的な還元型ポリスルフィドとしては、ヒドロジスルフィド(ヒドロパースルフィド:R4-S-S-H)、ヒドロトリスルフィド(R4-S-S-S-H)、又はヒドロテトラスルフィド(R4-S-S-S-S-H)などが挙げられる。
更に、本発明のアルキル化剤は、タンパク質などの酸化型ポリスルフィド構造(R4-S-(S)n-S-R5)のポリスルフィド部位(-(S)n-)を修飾することができる。具体的な酸化型ポリスルフィドとしては、トリスルフィド(R4-S-S-S-R5)、又はテトラスルフィド(R4-S-S-S-S-R5)などが挙げられる。
更に、本発明のアルキル化剤は、タンパク質などの酸化型ポリスルフィド構造(R4-S-(S)n-S-R5)のポリスルフィド部位(-(S)n-)を修飾することができる。具体的な酸化型ポリスルフィドとしては、トリスルフィド(R4-S-S-S-R5)、又はテトラスルフィド(R4-S-S-S-S-R5)などが挙げられる。
《作用》
本発明のアルキル化剤が、従来のアルキル化剤と比較して、効率的にポリスルフィドを標識できる機構は、詳細に解明されたわけではないが、以下のように推定することができる。しかしながら、本発明は下記の推定によって、限定されるものではない。
本発明における基Xは、チオール基との結合の反応に関与する基である。従来のアルキル化剤であるヨードアセトアミド(IAM)又はβ-(4-ヒドロキシフェニル)エチルヨードアセトアミド(HPE-IAM)では、このXがヨウ素(I)である。
一般的に、ポリスルフィド構造を含むチオール基(R4-(S)n-SH)は、ポリスルフィドを含まないチオール基(R4-SH)よりも酸解離定数の負の常用対数であるpKaが低いため、R4-(S)n-SHのほうが反応性に富む。一方、Xがヨウ素(I)、R1、R2が水素のアルキル化剤は元来R4-SHに対する反応性が高いことが知られており、ポリスルフィド基の有無にかかわらずチオール基への高い反応性を示し、R4-(S)n-SHへの反応選択性が低い。このヨウ素(I)をR4-SHとの反応性の低いフッ素原子、塩素原子、臭素原子、メタンスルホニルオキシ基、トリフルオロメタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基に置換することによって、R4-(S)n-SHへの反応性を損なわずR4-SHとの反応性を落とすことで、ポリスルフィド構造への反応選択性を向上できると推定される。
一方、R1及びR2は、チオール基との反応に直接関与する基ではないが、Xの近くに位置する基である。従来のアルキル化剤であるヨードアセトアミド(IAM)又はβ-(4-ヒドロキシフェニル)エチルヨードアセトアミド(HPE-IAM)では、このR1及びR2が、水素原子(H)である。水素原子は分子量が小さいため、R4-SHとXとの反応を立体構造として阻害することがなく、R4-SHに対しても高い反応性を示す。この水素原子(H)を、分子量の大きな基に置換し、立体障害を生じさせることによって、R4-SHとXとの反応性を抑制することができると推定される。
また、ヨウ素(I)は、チオール基との反応性が高いが、反応性が高すぎるために、ポリスルフィドの-(S)n-構造を過剰に分解してしまうことがあった。このヨウ素(I)をチオール基との反応性の低いフッ素原子、塩素原子、臭素原子、メタンスルホニルオキシ基、トリフルオロメタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基に置換することによって、-(S)n-構造の分解を抑制し、ポリスルフィドを効率的に標識することができると推定される。

[2]ポリスルフィドの標識方法
本発明のポリスルフィドの標識方法は、ポリスルフィドに、本発明のアルキル化剤を接触させる工程を含む。
本発明の標識方法によって標識されるポリスルフィドは、例えば図1Bに示すポリスルフィド構造を有する還元型のチオール基(R4-(S)n-SH)及び酸化型のポリスルフィド構造(R4-S-(S)n-S-R5)を含む。還元型のポリスルフィドを例にとると、チオール基のHと本発明のアルキル化剤(X-Z)のXとが反応して、HXとなって脱離し、チオール基が修飾される(R4-(S)n-S-Z)。従来のタンパク質のシステイン残基は、図1Aの還元型チオール基(Cys-SH)に示されているように、1つの硫黄原子のみ有するチオール基であると考えられていた。最近、生体内で図1Bに示されるポリスルフィド構造が存在することが示された。本発明の標識方法は、これらのポリスルフィド構造を標識することができる。また本発明のアルキル化剤は酸化型システイン形成箇所に含まれるポリスルフィドとも反応し、アルキル化剤(X-Z)のXが離脱して反応することで、R4-(S)n-S-ZないしR5-(S)n-S-Zとして標識されると推定される。
図2に示すように、従来のアルキル化剤は反応性が高いため、パースルフィド構造、又はトリスルフィド構造の-(S)n-構造を過剰に分解することがあった。そのため、例えばチオール基と、そこに硫黄原子がさらに付加したポリスルフィド基(パースルフィド構造やトリスルフィド構造)を区別することができなった。
本発明のポリスルフィドの標識方法によれば、過剰な反応を抑制したアルキル化剤を用いることにより、効率的にポリスルフィドを標識することができる。
本発明のポリスルフィドの標識方法は、ポリスルフィドに、本発明のアルキル化剤を接触させる工程を含む。

図2に示すように、従来のアルキル化剤は反応性が高いため、パースルフィド構造、又はトリスルフィド構造の-(S)n-構造を過剰に分解することがあった。そのため、例えばチオール基と、そこに硫黄原子がさらに付加したポリスルフィド基(パースルフィド構造やトリスルフィド構造)を区別することができなった。
本発明のポリスルフィドの標識方法によれば、過剰な反応を抑制したアルキル化剤を用いることにより、効率的にポリスルフィドを標識することができる。
本発明のポリスルフィドの標識方法における反応温度は、特に限定されるものではないが、例えば4~37℃であり、好ましくは25~37℃である。
本発明のポリスルフィドの標識方法における反応時間は、特に限定されるものではないが、例えば30分~48時間であり、好ましくは1~24時間である。
本発明のポリスルフィドの標識方法に用いる緩衝液は、標識反応が進行する限りにおいて、特に限定されるものではないが、例えばリン酸緩衝液、トリス-塩酸緩衝液、HEPES緩衝液、又は炭酸アンモニウム緩衝液を用いることができる。
本発明のポリスルフィドの標識方法における反応時間は、特に限定されるものではないが、例えば30分~48時間であり、好ましくは1~24時間である。
本発明のポリスルフィドの標識方法に用いる緩衝液は、標識反応が進行する限りにおいて、特に限定されるものではないが、例えばリン酸緩衝液、トリス-塩酸緩衝液、HEPES緩衝液、又は炭酸アンモニウム緩衝液を用いることができる。
本発明のポリスルフィドの標識方法によって標識される物質は、ポリスルフィドを有する物質である限りにおいて、特に限定されないが、例えばタンパク質、又はペプチドホルモンが挙げられる。具体的な検体としては、ポリスルフィドを有する物質を含む検体である限りにおいて限定されるものではないが、細胞、細胞抽出液、細胞上清、バイオプシー検体、臓器抽出液、血液、血漿、血清、唾液、髄液、精液、又は尿などが挙げられる。
本発明のポリスルフィドの検出方法は、前記ポリスルフィドの標識方法によって標識されたポリスルフィドを含むタンパク質を質量分析法で分析する工程を含む。質量分析法は、ポリスルフィドの標識方法によって標識された検体を用いることを除いては、本分野で通常使用されている質量分析法を用いることができる。
例えば、標識されたポリスルフィドを含むタンパク質を酵素(例えば、トリプシン、エンドプロテイナーゼGlu-C、Lys-C、ペプシン、キモトリプシン、又はクロストリパイン)で消化する。消化されたタンパク質を質量分析装置で分析する。
例えば、標識されたポリスルフィドを含むタンパク質を酵素(例えば、トリプシン、エンドプロテイナーゼGlu-C、Lys-C、ペプシン、キモトリプシン、又はクロストリパイン)で消化する。消化されたタンパク質を質量分析装置で分析する。
以下、実施例によって本発明を具体的に説明するが、これらは本発明の範囲を限定するものではない。
《製造例1》
本製造例では、R1及びR2が水素原子、Xが塩素原子(Cl)のアルキル化剤1を製造した。
10mLフラスコに、クロロ酢酸(105mg、1.13mmol)、チラミン(151mg、1.10mmol)、N,N,N′,N′-テトラメチル-O-(N-スクシンイミジル)ウロニウムテトラフルオロボラート(350mg,11.6mmol)を加え、N,N-ジメチルホルムアミド(2mL)に溶解し、室温にて攪拌した。N,N-ジイソプロピルエチルアミン(0.28mL)を加え、室温にて1時間攪拌した。水を加えて反応を停止し、酢酸エチルにて抽出し、回収した酢酸エチル層を水で2回、飽和食塩水で1回洗浄し、硫酸ナトリウムにて乾燥させ、溶媒を減圧留去した。得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=50/50→25/75)、目的化合物を得た。(74.3mg、収率32.9%)

10mLフラスコに、クロロ酢酸(105mg、1.13mmol)、チラミン(151mg、1.10mmol)、N,N,N′,N′-テトラメチル-O-(N-スクシンイミジル)ウロニウムテトラフルオロボラート(350mg,11.6mmol)を加え、N,N-ジメチルホルムアミド(2mL)に溶解し、室温にて攪拌した。N,N-ジイソプロピルエチルアミン(0.28mL)を加え、室温にて1時間攪拌した。水を加えて反応を停止し、酢酸エチルにて抽出し、回収した酢酸エチル層を水で2回、飽和食塩水で1回洗浄し、硫酸ナトリウムにて乾燥させ、溶媒を減圧留去した。得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=50/50→25/75)、目的化合物を得た。(74.3mg、収率32.9%)

《製造例2》
本製造例では、R1が水素原子、R2がメチル基、そしてXが臭素原子(Br)のアルキル化剤2を製造した。
10mLフラスコに、2-ブロモプロピオン酸(100mg、0.66mmol)、チラミン(100mg、0.72mmol)、プロピルホスホン酸無水物(50%酢酸エチル溶液、500μL)を加え、N,N-ジメチルホルムアミド(2mL)に溶解し、室温にて攪拌した。N,N-ジイソプロピルエチルアミン(172μL)及び4-ジメチルアミノピリジン(22mg)を加え、室温にて2時間攪拌した。水を加えて反応を停止し、酢酸エチルにて抽出し、回収した酢酸エチル層を水で2回、飽和食塩水で1回洗浄し、硫酸ナトリウムにて乾燥させ、溶媒を減圧留去した。得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=70/30→30/70)、目的化合物を得た。(151mg、収率84.4%)

《比較例1》
本比較例では、アルキル化剤として、下記式(3):
のR1及びR2が水素原子、Xがヨウ素原子(I)のβ-(4-ヒドロキシフェニル)エチルヨードアセトアミド(HPE-IAM)(Toronto Research Chemicals社)を用い、ポリスルフィド化ヒト血清アルブミン(HSA)のポリスルフィドを分析した。
本比較例では、アルキル化剤として、下記式(3):
(ポリスルフィド化HSAの製造)
2mLマイクロチューブにHSA(4mg)を加え、超純水(100μL)を加えて溶解した。ここに、予め調製した150mM二硫化ナトリウム水溶液を2μL加え、37度にて1時間反応させた。この溶液をPD SpinTrap G-25カラム(Cytiva社製)を用いて、同社の推奨プロトコルに従って精製し、過剰な二硫化ナトリウムを除去し、超純水にて溶出した。この溶出液に更に超純水を加えて500μLに調製し、アミコンウルトラ-0.5mL遠心式フィルター(分画分子量:3kDa、メルクミリポア社製)を用いて遠心分離機にて限外濾過し(14,000xg、30分)、濃縮液に超純水を加えて500μLとし、限外濾過操作をもう一度繰り返し、ポリスルフィド化HSAを得た。

0.5mLマイクロチューブに、ポリスルフィド化HSAを10μg加え、50mMトリス-塩酸緩衝液(pH7.5)を8μL加えて希釈した。この溶液に、20mMアルキル化剤(エタノール溶液)を1μL添加し、次いで0.1μg/μLトリプシン溶液を1μL加え、37度にて16時間静置した。10%トリフルオロ酢酸水溶液を1μL加えて37度にて30分静置し、トリプシン消化を終了させたのち、ペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩し、80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル 0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。

(質量分析LC-MS/MSによる計測)
液体クロマトグラフィー(LC)にUltiMateTM 3000 TSLCnanoシステム(ThermoFisher Scientific社)を、質量分析部(MS/MS)にQ ExactiveTM ハイブリッド四重極-オービトラップ質量分析計(ThermoFisher Scientific社)を搭載したnano ESI LC-MS/MS装置を用いてペプチドの分析を行った。
LC分離のトラップカラムには、Acclaim PepMap100、C18(5μm、100Å、内径300μmx5mm,Thermo Fisher Scientific社)を用い、分析カラムにはNTCC-360/75-3-125(C18,粒子径0.3μm、内径0.075mmx125mm,日京テクノス社)を用いた。移動相は、0.1%ギ酸水溶液(Solvent A)及び0.1%ギ酸アセトニトリル溶液(Solvent B)を用い、流速0.3μL/minにて、以下の溶媒組成になるように送液した。

質量分析計のイオンソースにはnanoESIを用い、Spray voltage 2.00kV、キャピラリー温度275度、S-lens RF level 50.0Vにてイオン化を行った。MSスキャン範囲をm/z 350-1500、AGC(automatic gain control)targetを3x106Chargesとし、ポジティブモードにてMSデータを取得し、次いでNCE(Normalized Collision Energy)を27に設定し、AGC(automatic gain control)targetを1x105ChargesとしてMS/MSデータを取得した。取得したデータはPEAKSXPro(インフォコム社)ないしはProteome Discoverer 2.4 SP1(Thermo Fisher Scientific社)にて解析した。
HSAの34番、75番、101番、124番、177番、265番、392番、487番、567番のシステイン残基にアルキル化剤(Z)が標識されたペプチド(-S-Z、-S-S-Z、-S-S-S-Z)の検出強度の割合を表10に示す。

表10に示すように、Xをヨウ素原子(I)から塩素原子(Cl)に置換することによって、パースルフィド(-S-S-Z)及びトリスルフィド(-S-S-S-Z)などのポリスルフィドの検出率が向上した。また、R2を水素原子からメチル基に置換することによっても、ポリスルフィドの検出率が向上した。
また、質量分析測定により同定されたペプチドの例を図3に示す。HSAの99-106番目のペプチドであるNECFLQHKにおいて、ポリスルフィド化した101番目のシステイン残基に、アルキル化剤1が標識されていることが確認された。

(biotin-PEG3-amineの製造法)
30mLフラスコに、biotin-PEG3-azide(103mg)を加え、メタノール(10mL)に溶解した。減圧脱気及びアルゴン置換を行い、パラジウム炭素を触媒量加え、真空脱気及び水素ガス置換を行い、室温にて終夜攪拌した。反応液をセライト濾過し、濾液を減圧濃縮し、目的化合物を得た。(97.0mg、98.1%)
30mLフラスコに、biotin-PEG3-azide(103mg)を加え、メタノール(10mL)に溶解した。減圧脱気及びアルゴン置換を行い、パラジウム炭素を触媒量加え、真空脱気及び水素ガス置換を行い、室温にて終夜攪拌した。反応液をセライト濾過し、濾液を減圧濃縮し、目的化合物を得た。(97.0mg、98.1%)
(ビオチン化アルキル化剤の製造法)
10mLフラスコに、biotin-PEG3-amine(22.0mg、52.6mmol)、クロロ酢酸(8.6mg、113mmol)、N,N,N′,N′-テトラメチル-O-(N-スクシンイミジル)ウロニウムテトラフルオロボラート(25.2mg,83.8mmol)を加え、N,N-ジメチルホルムアミド(500μL)に溶解し、室温にて攪拌した。N,N-ジイソプロピルエチルアミン(14μL)を加え、更に室温にて2時間攪拌した。反応液を減圧濃縮し、得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ジクロロメタン/メタノール=99/1→80/20)、目的化合物を得た。(19.1mg、73.5%)

本実施例では、製造例4で得られたビオチン化アルキル化剤を用いて、ヒト血漿タンパク質のポリスルフィドを分析した。
本実施例では、製造例4で得られたビオチン化アルキル化剤を用いて、ヒト血漿タンパク質のポリスルフィドを分析した。
(ヒト血漿サンプルからのアルブミン及びIgGの除去)
ヒト血漿サンプル50μLをPBS 50μLに希釈し、Albumin&IgG Depletion SpinTrap(Cytiva社)を用いて、同社の推奨プロトコルに従ってアルブミン及びIgGを除去した。以後の分析実験においては、このアルブミン/IgGを除去したサンプル及び、これらを除去しないサンプルの両方を用いた。
ヒト血漿サンプル50μLをPBS 50μLに希釈し、Albumin&IgG Depletion SpinTrap(Cytiva社)を用いて、同社の推奨プロトコルに従ってアルブミン及びIgGを除去した。以後の分析実験においては、このアルブミン/IgGを除去したサンプル及び、これらを除去しないサンプルの両方を用いた。
(ヒト血漿タンパク質のビオチン化及びトリプシン消化)
ヒト血漿サンプル及び、アルブミン/IgG除去サンプルを、それぞれProteoExtract(登録商標) Protein Precipitation Kit(メルクミリポア社)を用いて、同社の推奨プロトコルに従ってタンパク沈殿処理を行った。沈殿タンパク質を、20mM酢酸ナトリウム水溶液(pH5.5、6M尿素、0.1%RapiGest(Waters社)含有)を用いて再溶解し、タンパク濃度を測定した。
この再溶解サンプルから、それぞれタンパク量が50μgになるように溶液を分取し、50mM トリス-塩酸水溶液(pH7.5、0.1%RapiGest(Waters社)含有)を加え、タンパク質の終濃度が0.5μg/μLに、尿素の終濃度が1M以下になるように調製した。この溶液に、2mMビオチン化アルキル化剤(ジメチルスルホキシド溶液)を1μL添加し、0.5μg/μLトリプシン溶液を2μL加え、37度にて16時間静置した。10%トリフルオロ酢酸水溶液を1μL加え、37度にて30分静置し、反応を終了させた。この溶液を、陽イオン交換カラム(GL-Tip SCX、GLサイエンス社)を用いて、同社の推奨プロトコルに従って精製し、未反応のビオチン化アルキル化剤を除去した。10mM KH2PO4水溶液(1M KCl、25%アセトニトリル、pH3.0)を100μL用いてペプチドの溶出を行い、アビジン精製用サンプルとして用いた。
ヒト血漿サンプル及び、アルブミン/IgG除去サンプルを、それぞれProteoExtract(登録商標) Protein Precipitation Kit(メルクミリポア社)を用いて、同社の推奨プロトコルに従ってタンパク沈殿処理を行った。沈殿タンパク質を、20mM酢酸ナトリウム水溶液(pH5.5、6M尿素、0.1%RapiGest(Waters社)含有)を用いて再溶解し、タンパク濃度を測定した。

(アビジン(NeutrAvidin)ビーズによるビオチン化ペプチドの濃縮回収)
ペプチド溶出液100μLを2mLマイクロチューブに移し、PBSを150μL加えて希釈し、次いでアビジンビーズ(NeutrAvidin(登録商標) Agarose HC(Thermo Fisher Scientific社))を10μL添加し、ローテーターを用いて室温にて2時間攪拌した。ビーズを遠心分離にて回収し、上清を除去後、ビーズをPBS(5%アセトニトリル)にて4回、PBSにて3回、超純水にて3回洗浄した。このビーズに80%アセトニトリル水溶液(0.2%ギ酸、0.1%トリフルオロ酢酸含有)を50μL加え、70度にて10分振盪攪拌してビオチン化ペプチドを溶出し、室温に戻した後遠心分離し、上清を回収した。残ったビーズに80%アセトニトリル水溶液(0.2%ギ酸、0.1%トリフルオロ酢酸含有)を50μL加え、上清を回収し、これらの上清を混合したのち遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。
ペプチド溶出液100μLを2mLマイクロチューブに移し、PBSを150μL加えて希釈し、次いでアビジンビーズ(NeutrAvidin(登録商標) Agarose HC(Thermo Fisher Scientific社))を10μL添加し、ローテーターを用いて室温にて2時間攪拌した。ビーズを遠心分離にて回収し、上清を除去後、ビーズをPBS(5%アセトニトリル)にて4回、PBSにて3回、超純水にて3回洗浄した。このビーズに80%アセトニトリル水溶液(0.2%ギ酸、0.1%トリフルオロ酢酸含有)を50μL加え、70度にて10分振盪攪拌してビオチン化ペプチドを溶出し、室温に戻した後遠心分離し、上清を回収した。残ったビーズに80%アセトニトリル水溶液(0.2%ギ酸、0.1%トリフルオロ酢酸含有)を50μL加え、上清を回収し、これらの上清を混合したのち遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。
質量分析装置及び測定条件は、移動相の混合比率を以下のように変更したこと以外は、比較例1の操作を繰り返した。結果を表11に示す。
血漿タンパク質を用いた解析結果を以下に示す。
血漿タンパク質において、63種類のタンパク質にパースルフィド部位が含まれ、うち7種類のタンパク質にトリスルフィド構造が含まれることが同定された。
すなわち、ビオチン化アルキル化剤を用いて、ポリスルフィド構造を濃縮することが可能であり、ポリスルフィドの検出感度が向上した。
血漿タンパク質において、63種類のタンパク質にパースルフィド部位が含まれ、うち7種類のタンパク質にトリスルフィド構造が含まれることが同定された。
すなわち、ビオチン化アルキル化剤を用いて、ポリスルフィド構造を濃縮することが可能であり、ポリスルフィドの検出感度が向上した。
(キモトリプシンを用いたポリスルフィド化HSAの消化及びポリスルフィド分析)
《実施例5~7、及び比較例2》
本実施例及び比較例では、製造例1~3のアルキル化剤1~3及び比較例1のアルキル化剤を用い、ポリスルフィド化ヒト血漿タンパク質(HSA)のポリスルフィドを分析した。
0.5mLマイクロチューブに、ポリスルフィド化HSAを10μg加え、1M尿素を含む50mMトリス-塩酸緩衝液(pH7.5)を15.5μL加えて希釈した。この溶液に、それぞれ10mM実施例1~3又は比較例1(ジメチルスルホキシド溶液)を2.0μL添加し、次いで100mM塩化カルシウム水溶液を2.0μL、消化酵素溶液(0.5μg/μLキモトリプシン溶液)を0.5μL加え、37度にて4時間静置し、次いで50mMトリス-塩酸緩衝液(pH7.5)を35μL加えて希釈し、37度にて17時間静置した。10%トリフルオロ酢酸水溶液を加えて酵素消化を終了させたのち、ペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩し、80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。比較例1の操作を繰り返して、ポリスルフィド化HSAのポリスルフィドを分析した。結果を表14に示す。
《実施例5~7、及び比較例2》
本実施例及び比較例では、製造例1~3のアルキル化剤1~3及び比較例1のアルキル化剤を用い、ポリスルフィド化ヒト血漿タンパク質(HSA)のポリスルフィドを分析した。
0.5mLマイクロチューブに、ポリスルフィド化HSAを10μg加え、1M尿素を含む50mMトリス-塩酸緩衝液(pH7.5)を15.5μL加えて希釈した。この溶液に、それぞれ10mM実施例1~3又は比較例1(ジメチルスルホキシド溶液)を2.0μL添加し、次いで100mM塩化カルシウム水溶液を2.0μL、消化酵素溶液(0.5μg/μLキモトリプシン溶液)を0.5μL加え、37度にて4時間静置し、次いで50mMトリス-塩酸緩衝液(pH7.5)を35μL加えて希釈し、37度にて17時間静置した。10%トリフルオロ酢酸水溶液を加えて酵素消化を終了させたのち、ペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩し、80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。比較例1の操作を繰り返して、ポリスルフィド化HSAのポリスルフィドを分析した。結果を表14に示す。
表14に示すように、消化酵素をキモトリプシンに変更しても、実施例5~7におけるパースルフィド(-S-S-Z)及びトリスルフィド(-S-S-S-Z)などのポリスルフィドの検出率が、比較例2に比べて向上した。また、同定されたこれらのシステイン残基のうち、34番目のシステイン残基(C34)は還元型システインとして存在することが報告されており、また75,253,265,316,392,487,514番目のシステイン残基は酸化型システイン(ジスルフィド結合)を形成する部位として報告されている(それぞれ、C75-C91、C245-C253、C265-C279、C316-C361、C392-C438、C476-C487、C514-C559の組み合わせでジスルフィド結合を形成する)。すなわち、本発明におけるアルキル化剤は、これらの個所に含まれるポリスルフィド構造をいずれも高効率に検出できる。
《製造例5》
本製造例では、下記式(5)のR1が水素原子、R2がエチル基、Xが臭素原子(Br)、及びR3にヒドロキシフェニルエチル基を含むアルキル化剤5を製造した。
2mLチューブに、2-ブロモ酪酸(79mg、0.47mmol)、チラミン(65mg、0.47mmol)、プロピルホスホン酸無水物(50%酢酸エチル溶液、450μL)を加え、N,N-ジメチルホルムアミド(500μL)に溶解し、室温にて攪拌した。N,N-ジイソプロピルエチルアミン(165μL)および4-ジメチルアミノピリジン(4mg)を加え、室温にて45分間攪拌した。反応終了後、シリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=80/20→50/50)、目的化合物を得た。(93.1mg、収率69.0%)
本製造例では、下記式(5)のR1が水素原子、R2がエチル基、Xが臭素原子(Br)、及びR3にヒドロキシフェニルエチル基を含むアルキル化剤5を製造した。
《製造比較例1》
本製造比較例では、下記式(6)のR1及びR2が水素原子、Xがヨウ素原子(I)、及びR3にアルキニル基を含む比較アルキル化剤1を製造した。
10mLナスフラスコに、ヨード酢酸(145mg、0.78mmol)、プロパルギルアミン(43mg、0.47mmol)を加え、N,N-ジメチルホルムアミド(1.5mL)に溶解し、室温にて攪拌した。N,N-ジイソプロピルエチルアミン(200μL)およびN,N,N′,N′-Tetramethyl-O-(N-succinimidyl)uronium tetrafluoroborate(252mg、0.84mmol)を加え、室温にて1時間半攪拌した。反応終了後、溶媒を減圧留去し、得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=70/30→50/50)、目的化合物を得た。(107mg、収率61.5%)

10mLナスフラスコに、クロロ酢酸(77mg、0.79mmol)、プロパルギルアミン(43mg、0.47mmol)を加え、N,N-ジメチルホルムアミド(1.5mL)に溶解し、室温にて攪拌した。N,N-ジイソプロピルエチルアミン(200μL)およびN,N,N′,N′-Tetramethyl-O-(N-succinimidyl)uronium tetrafluoroborate(247mg、0.88 mmol)を加え、室温にて1時間攪拌した。反応終了後、溶媒を減圧留去し、得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=80/20→50/50)、目的化合物を得た。(105mg、収率98.0%)

10mLナスフラスコに、L-フェニルアラニン-N-FMOC(13C9,99%;15N,99%,CNLM-4362-H-PK,Cambridge Isotope Laboratories社)(28.1mg、0.084mmol)、Tetramethyl-O-(N-succinimidyl)uronium tetrafluoroborate(35.3mg,0.117mmol)を加え、よく乾燥させた後、N,N-ジメチルホルムアミド(300μL)に溶解し、プロパルギルアミン(8.6mg、0.47mmol)、N,N-ジイソプロピルエチルアミン(30μL)を加え、室温にて1時間攪拌した。反応溶媒を減圧留去し、得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=70/30→30/70)、目的化合物Fmoc-Phe-alkyne(13C9,15N)を得た。(27.7mg、収率90.3%)
次に、下記式(11)の化合物を合成した。
10mLナスフラスコに、Fmoc-Phe-alkyne(13C9,15N)(27.7mg、0.064mmol)、N,N-ジメチルホルムアミド(600μL)、ピペリジン(150μL)を加え、室温にて30分間攪拌した。反応溶媒を減圧留去し、得られた残渣をシリカゲルクロマトグラフィーにて精製し(充填剤:アミノ修飾型、溶離液:ヘキサン/酢酸エチル=25/75→0/100)、目的化合物H-Phe-alkyne(13C9,15N)を得た。(13.6mg、収率100%)
次に、下記の操作を行い、前記式(8)のアルキル化剤8を得た。10mLナスフラスコに、H-Phe-alkyne(13C9,15N)(13.6mg、0.064mmol)、Tetramethyl-O-(N-succinimidyl)uronium tetrafluoroborate(38.6mg,0.128mmol)を加え、よく乾燥させた後、N,N-ジメチルホルムアミド(500μL)に溶解し、(R)-(+)-2-ブロモプロピオン酸(19.5mg、0.094mmol)、N,N-ジイソプロピルエチルアミン(28μL)を加え、室温にて2時間半攪拌した。反応溶媒を減圧留去し、得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=70/30→50/50)、目的化合物を得た。(19.7mg、収率89.1%)

次に、下記の操作を行い、前記式(12)のアルキル化剤9を得た。10mLナスフラスコに、H-Phe-alkyne(9.5mg、0.047mmol)、Tetramethyl-O-(N-succinimidyl)uronium tetrafluoroborate(30.0mg, 0.100mmol)を加え、よく乾燥させた後、N,N-ジメチルホルムアミド(200μL)に溶解し、(R)-(+)-2-ブロモプロピオン酸(14.3mg、0.094mmol)、N,N-ジイソプロピルエチルアミン(20μL)を加え、室温にて2時間半攪拌した。反応溶媒を減圧留去し、得られた残渣をシリカゲルクロマトグラフィーにて精製し(溶離液:ヘキサン/酢酸エチル=70/30→50/50)、目的化合物を得た。(15.1mg、収率95.6%)

《実施例8~12、比較例3》
本実施例及び比較例では、前記アルキル化剤5~9及び比較アルキル化剤1を用い、ヒト血漿タンパク質(HSA)のポリスルフィドを分析した。
0.5mLマイクロチューブに、ポリスルフィド化HSAを10μg加え、6M尿素を含む50mMトリス-塩酸緩衝液(pH7.5)を3.6μL加えて希釈した。この溶液に、それぞれ40mMアルキル化剤6~10又は比較アルキル化剤1(ジメチルスルホキシド溶液)を0.5μL添加し、次いで消化酵素溶液(0.5μg/μLトリプシン-LysC混合溶液)を0.4μL加え、37度にて4時間静置し、次いで50mMトリス-塩酸緩衝液(pH7.5)を35μL加えて希釈し、37度にて20時間静置した。50%トリフルオロ酢酸水溶液を1μL加えて酵素消化を終了させたのち、ペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩し、80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。
本実施例及び比較例では、前記アルキル化剤5~9及び比較アルキル化剤1を用い、ヒト血漿タンパク質(HSA)のポリスルフィドを分析した。

比較例1と同様に質量分析を行った。結果を表15に示す。
表15に示すように、R2がエチル基のアルキル化剤(実施例8)もパースルフィド(-S-S-Z)及びトリスルフィド(-S-S-S-Z)などのポリスルフィドの検出率が向上した。更に、R3にアルキニル基を含む化合物においても、Xをヨウ素原子(I)から塩素原子(Cl)に置換すること(実施例9)によってパースルフィド(-S-S-Z)及びトリスルフィド(-S-S-S-Z)などのポリスルフィドの検出率が向上した。また、R2を水素原子からメチル基に置換し、Xをヨウ素原子(I)から臭素原子(Br)に置換すること(実施例10)によっても、ポリスルフィドの検出率が向上した。さらに、実施例10、11、及び12を比較すると、アルキニル基および安定同位体を含む化合物においても、XおよびR1の構造が同じであれば、ポリスルフィド検出率に大きな差はないことが判明した。
《実施例13~15》
本実施例では、製造例7で得られたアルキル化剤7をポリスルフィド化HSAに反応させた後に、アルキル化剤のアルキニル基と、アジド基を有するビオチンとを結合させた。得られた産物を質量分析することによって、HSAのポリスルフィドを分析した。
0.5mLマイクロチューブに、ポリスルフィド化HSAを50μg加え、6M尿素を含む50mMトリス-塩酸緩衝液(pH7.5)を18μL加えて希釈した。この溶液に、製造例7で得られた40mMアルキル化剤8(ジメチルスルホキシド溶液)を2.5μL添加し、次いで消化酵素溶液(0.5μg/μLトリプシン-LysC混合溶液)を2μL加え、37度にて4時間静置し、次いで50mMトリス-塩酸緩衝液(pH7.5)を140μL加えて希釈し、37度にて18時間静置した。この溶液を32ulずつ2つの0.5mLマイクロチューブに分注し、一方には10mM biotin-PEG3-azide(実施例13;ジメチルスルホキシド溶液)を4ul、もう一方には10mM Biotin-PC-azide(実施例14;Sigma-Aldrich社)を4ul加え、それぞれのチューブに25mMトリス(3-ヒドロキシプロピルトリアゾリルメチル)アミン水溶液を1.6μL、25mM硫酸銅水溶液を1.6μL加え、次いで100mMアスコルビン酸ナトリウムを0.8μL加え、室温にて1時間振盪した。50%トリフルオロ酢酸水溶液を1μL加えたのち、ペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩した。80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル 0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。またこのうち、Biotin-PC-azideを結合させたサンプルに関しては、脱塩後のサンプルを一部分注し、紫外線(365nm)を1時間照射し、リンカー部分が光反応により開裂しているか(実施例15)を質量分析にて確認した。
以下に、前記の工程のスキームを示す。なお、biotin-PEG3-azideについては記載していない。
アルブミンのそれぞれ75、101、177、265、392、487番目のシステインを含むペプチド消化物において、ポリスルフィド基にアルキン含有タグ分子およびアジド化合物が結合した質量数を加味したペプチドがそれぞれ同定された。すなわち、アルキン含有標識分子のポリスルフィド基への選択的な標識、及びヒュスゲン環化付加反応が進行した。加えて、biotin-PC-azideを結合させたペプチドにおいては、光照射による分子構造変化を反映した質量数変化を質量分析測定にて確認したことから、刺激により開裂するリンカーを含むビオチン化合物をポリスルフィド標識試薬へ応用することが可能となった。
本実施例では、製造例7で得られたアルキル化剤7をポリスルフィド化HSAに反応させた後に、アルキル化剤のアルキニル基と、アジド基を有するビオチンとを結合させた。得られた産物を質量分析することによって、HSAのポリスルフィドを分析した。
0.5mLマイクロチューブに、ポリスルフィド化HSAを50μg加え、6M尿素を含む50mMトリス-塩酸緩衝液(pH7.5)を18μL加えて希釈した。この溶液に、製造例7で得られた40mMアルキル化剤8(ジメチルスルホキシド溶液)を2.5μL添加し、次いで消化酵素溶液(0.5μg/μLトリプシン-LysC混合溶液)を2μL加え、37度にて4時間静置し、次いで50mMトリス-塩酸緩衝液(pH7.5)を140μL加えて希釈し、37度にて18時間静置した。この溶液を32ulずつ2つの0.5mLマイクロチューブに分注し、一方には10mM biotin-PEG3-azide(実施例13;ジメチルスルホキシド溶液)を4ul、もう一方には10mM Biotin-PC-azide(実施例14;Sigma-Aldrich社)を4ul加え、それぞれのチューブに25mMトリス(3-ヒドロキシプロピルトリアゾリルメチル)アミン水溶液を1.6μL、25mM硫酸銅水溶液を1.6μL加え、次いで100mMアスコルビン酸ナトリウムを0.8μL加え、室温にて1時間振盪した。50%トリフルオロ酢酸水溶液を1μL加えたのち、ペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩した。80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル 0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。またこのうち、Biotin-PC-azideを結合させたサンプルに関しては、脱塩後のサンプルを一部分注し、紫外線(365nm)を1時間照射し、リンカー部分が光反応により開裂しているか(実施例15)を質量分析にて確認した。
以下に、前記の工程のスキームを示す。なお、biotin-PEG3-azideについては記載していない。
《実施例16》
(ヒト血漿タンパク質のビオチン化及びトリプシン消化)
ヒト血漿サンプル及び、アルブミン/IgG除去サンプルを、それぞれProteoExtract(登録商標) Protein Precipitation Kit(メルクミリポア社)を用いて、同社の推奨プロトコルに従ってタンパク沈殿処理を行った。沈殿タンパク質を、20mM酢酸ナトリウム水溶液(pH6.0、6M尿素)を用いて再溶解し、タンパク濃度を測定した。
この再溶解サンプルから、それぞれタンパク量が70μgになるように溶液を分取し、25mM トリス-塩酸水溶液(pH7.5、6M尿素)を加え、次いで製造例7で得られた40mMのアルキル化剤7(アセトニトリル溶液)を7μL添加し、消化酵素溶液(0.5μg/μLトリプシン-LysC混合溶液)を1.4μL加え、タンパク質の終濃度が1.0μg/μLになるように調製した。37度にて5時間静置し、次いで25mMトリス-塩酸緩衝液(pH7.5)を350μL加えて希釈し、37度にて14時間静置した。
この溶液に、10mM Biotin-PC-azide(実施例12;Sigma-Aldrich社)を42μL加え、超純水75.6μLで希釈し、100mMトリス(3-ヒドロキシプロピルトリアゾリルメチル)アミン水溶液を5.6μL、100mM硫酸銅水溶液を5.6μL加え、次いで100mMアスコルビン酸ナトリウムを11.2μL加え、室温にて1.5時間振盪した。反応液を30%アセトニトリル 0.1%トリフルオロ酢酸水溶液490μLで希釈し、50%トリフルオロ酢酸水溶液を1μL加えたのち陽イオン交換カラム(Monospin(登録商標) SCX、GLサイエンス社)を用いて、同社の推奨プロトコルに従って精製し、未反応のアルキル化剤およびビオチン化合物を除去した。500mM 酢酸アンモニウム水溶液(30%アセトニトリル 4%トリフルオロ酢酸)および500mM 酢酸アンモニウム水溶液(30%アセトニトリル)をそれぞれ100μL用いてペプチドの溶出を行い、遠心濃縮装置にて乾燥し、アビジン精製用サンプルとして用いた。
(ヒト血漿タンパク質のビオチン化及びトリプシン消化)

この溶液に、10mM Biotin-PC-azide(実施例12;Sigma-Aldrich社)を42μL加え、超純水75.6μLで希釈し、100mMトリス(3-ヒドロキシプロピルトリアゾリルメチル)アミン水溶液を5.6μL、100mM硫酸銅水溶液を5.6μL加え、次いで100mMアスコルビン酸ナトリウムを11.2μL加え、室温にて1.5時間振盪した。反応液を30%アセトニトリル 0.1%トリフルオロ酢酸水溶液490μLで希釈し、50%トリフルオロ酢酸水溶液を1μL加えたのち陽イオン交換カラム(Monospin(登録商標) SCX、GLサイエンス社)を用いて、同社の推奨プロトコルに従って精製し、未反応のアルキル化剤およびビオチン化合物を除去した。500mM 酢酸アンモニウム水溶液(30%アセトニトリル 4%トリフルオロ酢酸)および500mM 酢酸アンモニウム水溶液(30%アセトニトリル)をそれぞれ100μL用いてペプチドの溶出を行い、遠心濃縮装置にて乾燥し、アビジン精製用サンプルとして用いた。
(アビジン(NeutrAvidin)ビーズによるビオチン化ペプチドの濃縮回収)
ペプチド溶出乾固物をPBS(2.5%アセトニトリル)70μLに溶解し、1M トリス-塩酸水溶液(pH8.0)を用いてpHを7にし、PBS(2.5%アセトニトリル)を加えて140μLになるように調製した。これを120μL分注し、次いでアビジンビーズ(NeutrAvidin(登録商標) Agarose HC(Thermo Fisher Scientific社))を15μL添加し、PBS(2.5%アセトニトリル)を加えて300μLになるように調製し、ローテーターを用いて室温にて2時間攪拌した。ビーズを遠心分離にて回収し、上清を除去後、ビーズをPBS(2.5%アセトニトリル)にて3回、超純水にて2回洗浄した。このビーズを50mMトリス-塩酸水溶液(2.5%アセトニトリル、pH7.5)50μLに懸濁し、紫外線(365nm)を1時間照射し、照射後の懸濁液をフィルター濾過し、濾液をペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩し、80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。
ペプチド溶出乾固物をPBS(2.5%アセトニトリル)70μLに溶解し、1M トリス-塩酸水溶液(pH8.0)を用いてpHを7にし、PBS(2.5%アセトニトリル)を加えて140μLになるように調製した。これを120μL分注し、次いでアビジンビーズ(NeutrAvidin(登録商標) Agarose HC(Thermo Fisher Scientific社))を15μL添加し、PBS(2.5%アセトニトリル)を加えて300μLになるように調製し、ローテーターを用いて室温にて2時間攪拌した。ビーズを遠心分離にて回収し、上清を除去後、ビーズをPBS(2.5%アセトニトリル)にて3回、超純水にて2回洗浄した。このビーズを50mMトリス-塩酸水溶液(2.5%アセトニトリル、pH7.5)50μLに懸濁し、紫外線(365nm)を1時間照射し、照射後の懸濁液をフィルター濾過し、濾液をペプチド脱塩・濃縮用チップ(GL-Tip SDB、GLサイエンス社製)を用いて脱塩し、80%アセトニトリル、0.1%トリフルオロ酢酸水溶液にてチップから溶出したペプチド分画を遠心濃縮装置にて乾燥し、乾固物を2%アセトニトリル0.1%ギ酸水溶液に溶解し、これを質量分析計測用サンプルとした。
質量分析測定によるプロテオーム解析の結果、血漿タンパク質において、パースルフィド(-S-S-Z)、トリスルフィド(-S-S-S-Z)、テトラスルフィド(-S-S-S-S-Z)を含むペプチドをそれぞれ150個、25個、6個同定し、ポリスルフィドを含むタンパク質を50種類同定した。ここでしめすZとは、Biotin-PC-azideを結合させ紫外線照射により生じた分子構造を示す(実施例14)。すなわち、アルキン含有標識分子のポリスルフィド基への選択的な標識、biotin-PC-azideのヒュスゲン環化反応、アビジンビーズによる濃縮および紫外線照射によるリンカー開裂を経て、ポリスルフィド構造の濃縮が可能であり、ポリスルフィドの検出感度が向上した。
また、特定のタンパク質におけるポリスルフィド化部位の同定を行ったところ、例えばアルブミン(Protein accession:P02768、Albumin precursor)においては99、125、148、201、289、416、500、511、582、591番目のシステイン残基(シグナルペプチドおよびプロペプチド切断後の配列においては75、101、124、177、265、392、476、487、558、567番目のシステイン残基)にポリスルフィド構造が含まれることを検出した。これらのシステイン残基の多くは、酸化型構造(ジスルフィド結合)を形成する個所として知られている。また、α1-アンチトリプシン(Protein accession:P01009、Alpha-1-antitrypsin precursor)においては、256番目のシステイン残基にポリスルフィド構造が含まれることを検出した。このシステイン残基は、還元型構造として知られている。すなわち、内在性のタンパク質においても、上記構造中に含まれるポリスルフィド修飾構造の検出に成功した。

Claims (4)
- 下記式(1):
R1及びR2は、R1及びR2が結合している炭素原子と一緒になって炭素数3~7のシクロアルキル基、置換基を有する炭素数3~7のシクロアルキル基、炭素数6~12のアリール基、置換基を有する炭素数6~12のアリール基、3~7員の複素環基、又は置換基を有する3~7員の複素環基であり、
Xは、ハロゲン原子、メタンスルホニルオキシ基、トリフルオロメタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基であり、
R1及びR2がいずれも水素原子の場合、Xはヨウ素原子以外のハロゲン原子、メタンスルホニルオキシ基、トリフルオロメタンスルホニルオキシ基、又はパラトルエンスルホニルオキシ基である)で表されるポリスルフィド標識用官能基。 - 請求項1に記載の官能基を有する化合物又はその塩を含むポリスルフィド標識用アルキル化剤。
- ポリスルフィドに、請求項2に記載のアルキル化剤を接触させる工程を含む、ポリスルフィドの標識方法。
- 請求項3のポリスルフィドの標識方法によって、標識されたポリスルフィドを含むタンパク質を質量分析法で分析する工程を含む、ポリスルフィド構造の検出方法。
