Abstract
清涼化剤が有する細胞傷害性の低減及び/又は、清涼化剤が有する清涼感の増強を目的とする。清涼化剤であるメントールを含有する眼科用製剤において、ブリモニジンを配合することにより、清涼化剤であるメントールが有する細胞傷害性を低減すること、並びにメントールが有する清涼感を増強することを見出した。さらに、ブリモニジンと、ベルガモット油、ゲラニオール及びボルネオールからなる群から選ばれる清涼化剤とを配合した眼科用製剤でも同様に、清涼化剤による細胞傷害性を、ブリモニジンを配合することで低減することが示された。以上の結果に基づき、清涼化剤及びブリモニジンを含有する眼科用製剤を提供する。
Description
本発明は、低濃度のブリモニジン及び清涼化剤を含有する眼科用液体製剤に関する。
ブリモニジン及びその塩は、α-2アドレナリン受容体アゴニストとして知られている。ヒトの眼は、多くのα-2アドレナリン受容体(以下、α-2受容体と略す場合がある)を有しており、α-2受容体のアゴニストは、眼房水産生抑制と共にぶどう膜強膜流出路を介した眼房水の流出を促進することによって、眼圧を低下させる作用を有する。この作用に基づき、α-2受容体アゴニストは、従来、緑内障や高眼圧症の治療に使用されている。また、α-2受容体のアゴニストは、α-2受容体集中細動脈、特に終末細動脈の内腔サイズの低減を引き起こす作用を有する。この作用により、血管収縮が生じ、目の赤みを低減し、そして白みを増加させ、眼の審美的外観を改善することができる(特許文献1:特許第5671459号公報;特許文献2:特許第5738890号公報)。
眼科用液体製剤には目の不快な症状・疲れ目の緩和を目的として清涼感や爽快感を付与するため清涼化剤を配合することがある。清涼化剤の一例としてメントールが挙げられる。
一方、メントールは、清涼感を発揮する成分として、化粧料、医薬部外品、及び医薬品に広く配合されている。なかでも、口腔洗浄剤又は点眼剤には、メントールが配合される場合が多い。メントールは、眼科用製剤に配合されることで、すっきりとした指し心地を付与することができる。メントールによる清涼感は、主に冷感受容体を活性化することにより引き起こされると考えられている。経皮投与での傷害性は低いと考えられており、粘膜に使用されることのない化粧料については最大配合量7%と定められている。一方、点眼剤については通常0.0005%~0.05%の範囲で使用される。
メントール等の清涼化剤はすっきりとした指し心地を付与することができるため、多くの眼科用液体製剤に配合されている。一方で、化学物質の多くは細胞傷害性を有するものが多い。本発明者らが行った培養角膜上皮細胞に対してメントール等の清涼化剤を含む試験点眼液を添加した試験において、メントール等の清涼化剤が細胞傷害性を示すことがあることが判った。そこで、メントール等の清涼化剤を含有する眼科用液体製剤において、メントール等の清涼化剤の細胞傷害性を低減可能な成分を一緒に配合することが求められる。
そこで、本発明者らが、メントール等の清涼化剤の細胞傷害性を低減する成分について探索したところ、ブリモニジン及び/又はその塩が、メントール等の清涼化剤による細胞傷害性を低減することができることを見出した。さらに、本発明者らは、ブリモニジン及び/又はその塩はメントール等の清涼化剤含有眼科用液体製剤において、メントール等の清涼化剤による清涼感を増強することができることも見出した。メントール等の清涼化剤の清涼感を増強することができれば、同量でより高い清涼化効果を発揮でき、またメントール等の清涼化剤の配合量を低減することも可能になる。
本発明の一実施形態として、下記に示す眼科用液体製剤を提供する。
[1] 0.01w/v%~0.05w/v%のブリモニジン及び/又はその塩と、清涼化剤とを含有する眼科用液体製剤。
[2] 前記清涼化剤が、メントール、ベルガモット油、ゲラニオール及びボルネオールからなる群から選ばれる、項目1に記載の眼科用液体製剤。
[3] 前記メントールが、l-メントールである、項目2に記載の眼科用液体製剤。
[4] 前記メントールが、0.001%~0.05%で配合される、項目2又は3に記載の眼科用液体製剤。

本発明によれば、メントール等の清涼化剤を含む眼科用液体製剤に対し、ブリモニジン及び/又はその塩を配合することで、細胞傷害性を低減する効果及び/又はメントール等の清涼化剤の清涼感を増強する効果を発揮する。
本明細書において使用される用語は、特に言及しない限り、当該分野で通常用いられる意味で用いられることが理解される。従って、他に定義されない限り、本明細書中で使用されるすべての専門用語及び科学技術用語は、本発明の属する分野の当業者によって一般的に理解されるのと同じ意味を有する。本明細書で特定される数値範囲は、その下限値及び上限値を含むものとする。
(定義)
本明細書において、「ブリモニジン」は、IUPAC名:5-ブロモ-N-(4,5-ジヒドロ-1H-イミダゾール-2-イル)キノキサリン-6‐アミン(5-Bromo-N-(4,5-dihydro-1H-imidazol-2-yl)quinoxalin-6-amine)の化合物を指す。また、本明細書において、ブリモニジン及び/又はその塩の濃度は、特に明記しない限り、ブリモニジン酒石酸塩に換算された濃度である。
本明細書において、「ブリモニジン」は、IUPAC名:5-ブロモ-N-(4,5-ジヒドロ-1H-イミダゾール-2-イル)キノキサリン-6‐アミン(5-Bromo-N-(4,5-dihydro-1H-imidazol-2-yl)quinoxalin-6-amine)の化合物を指す。また、本明細書において、ブリモニジン及び/又はその塩の濃度は、特に明記しない限り、ブリモニジン酒石酸塩に換算された濃度である。
本明細書において、「低濃度のブリモニジン」とは、0.05w/v%以下の濃度のブリモニジンを指す。
本明細書において、「眼科用液体製剤」とは、水を基剤とする水性液体製剤であり、眼科用製剤、特に点眼に用いられる製剤を指す。
メントールとは、IUPAC名:2-イソプロピル-5-メチルシクロヘキサノールの化合物を指し、複数の立体異性体を含む。ハッカやペパーミントに含まれる天然のメントールとして、一般的にはdl-メントール又はl-メントールを指すことが多い。メントールはハッカ臭を有するとともに、冷感受容体を刺激し、清涼感(冷感ともいう)を引き起こす。
本明細書において、「細胞傷害性」は、培養細胞、特に培養上皮細胞、より具体的には培養角膜上皮細胞に対する傷害性として計測される。具体的に、細胞傷害性は、試験点眼液を培養細胞に添加し、一定期間培養後の生細胞数及び/又は死細胞数に基づいて決定される。こうして決定された試験点眼液の細胞傷害性は、生体の細胞や眼組織、特に上皮細胞、より具体的に角膜上皮細胞でも同様に発揮されうる。
本明細書において、「細胞傷害性低減」とは、清涼化剤配合の眼科用液体製剤による細胞傷害性を低減することをいう。具体的に、清涼化剤配合の眼科用液体製剤にブリモニジン及び/又はその塩を配合することで、清涼化剤の細胞傷害性を低減することができる。
本明細書において、「細胞傷害性低減剤」とは、清涼化剤配合の眼科用液体製剤を眼に点眼した際の細胞傷害性による影響を抑制し、細胞が本来持つ機能を維持し続けるために配合される剤のことをいう。具体的には、清涼化剤配合の眼科用液体製剤に、細胞傷害性低減剤としてブリモニジン及び/又はその塩を配合することで、清涼化剤配合の眼科用液体製剤の細胞傷害性を低減することができる。ブリモニジン及び/又はその塩は、他の用途を目的として配合されてもよい。
本明細書において、「清涼感」とは、清涼化剤に起因する冷感のことをいう。特に清涼化剤が眼科用液体製剤に配合されると、眼科用液体製剤を眼に点眼した際に、すっきりとした指し心地を与えることができる。清涼感は、官能試験により評価することができ、主に清涼感の強さと清涼感の持続性の観点で評価される。
本明細書において、「清涼感増強」とは、眼科用液体製剤を眼に点眼した際の清涼感の強さ及び持続性のうちのいずれか、又は両方を増強することをいう。したがって、清涼感増強剤は、清涼感の強さの増強剤又は清涼感持続性増強剤ともいうことができる。
本明細書において、「清涼感増強剤」とは、眼科用液体製剤を眼に点眼した際の清涼感の強さ及び持続性のうちのいずれか、又は両方を増強することを目的として配合される添加剤のことをいう。具体的に、清涼化剤配合の眼科用液体製剤に、清涼感増強剤としてブリモニジン及び/又はその塩を配合することで、眼科用液体製剤の清涼感を増強することができる。ブリモニジン及び/又はその塩は、他の用途を目的として配合されてもよい。

以下に本発明の好ましい実施形態を説明する。以下に提供される実施形態は、本発明のよりよい理解のために提供されるものであり、本発明の範囲は以下の記載に限定されるべきでないことが理解される。従って、当業者は、本明細書中の記載を参酌して、本発明の範囲内で適宜改変を行うことができることは明らかである。また、以下の実施形態は単独でも使用されあるいはそれらを組み合わせて使用することができることが理解される。
以下に本発明の好ましい実施形態を説明する。以下に提供される実施形態は、本発明のよりよい理解のために提供されるものであり、本発明の範囲は以下の記載に限定されるべきでないことが理解される。従って、当業者は、本明細書中の記載を参酌して、本発明の範囲内で適宜改変を行うことができることは明らかである。また、以下の実施形態は単独でも使用されあるいはそれらを組み合わせて使用することができることが理解される。
本発明の眼科用液体製剤は、清涼化剤及びブリモニジン及び/又はその塩を含有する。
ブリモニジンの塩としては、医薬として許容される塩であれば任意の塩が挙げられる。ブリモニジンの医薬として許容される塩として、塩酸塩、硫酸塩、リン酸塩、酢酸塩、クエン酸塩、シュウ酸塩、マロン酸塩、サリチル酸塩、リンゴ酸塩、フマル酸塩、コハク酸塩、アスコルビン酸塩、マレイン酸塩、メタンスルホン酸塩、酒石酸塩および当業者に周知の他の無機カルボン酸塩が挙げられ、好ましくは酒石酸塩である。
本発明において、低濃度のブリモニジンとは、0.05w/v%以下の濃度のブリモニジンを指す。一例として、濃度の上限は、副作用を予防する観点から0.04w/v%、又は0.03w/v%であってもよい。濃度の下限は、ブリモニジンが含有される限り限定されないが、0.01w/v%が用いられてもよく、一例として本剤の効果を鑑み、0.015w/v%、又は0.02w/v%が用いられてもよい。
本発明において、清涼化剤は、眼科用液体製剤に清涼感を付与する目的で配合される。
本発明において、眼科用液体製剤に用いることができる清涼化剤としては、テルペノイド、クローブ香料、ペパーミント香料、ユーカリ香料、レモン香料、ローレル香料、ラベンダー香料、タラゴン香料、カルダモン香料、シダー香料、グレープフルーツ香料、オレンジ香料、ショウガ香料、ベルガモット香料、コリアンダー香料、シナモン香料、ジャスミン香料、ローズマリー香料、ローズ香料、パイン香料、カルダモン香料、ベンゾイン香料、ゼラニウム香料、カモミール香料、マージョラム香料、スペアミント香料、フェンネル香料、バニラフレーバー、ハッカ油、ハッカ水及びベルガモット油等を挙げることができる。テルペノイドとしては、例えば、メントール、メントン、カンフル、ボルネオール、ゲラニオール、シネオール、リモネン、オイゲノール、シトラール、ピネン、リナロール、フェンチルアルコール、ヨノン、サフラナール、テルピネン等のモノテルペンを挙げることができる。その中でも特に、ヒト角膜細胞に対する細胞傷害性の低さの観点から、特にメントール、ベルガモット油、ゲラニオール、及びボルネオールを用いるのが好ましい。さらに、特にメントールを用いるのが好ましい。これらの清涼化剤は1種単独で、又は2種以上を適宜組み合わせて、使用することができる。
メントールとしては、清涼感を発揮できる任意の立体異性体であってもよい。メントールは、(+)-メントール、(+)-イソメントール、(+)-ネオメントール、(+)-ネオイソメントール、(-)-メントール、(-)-イソメントール、(-)-ネオメントール、(-)-ネオイソメントールが挙げられる。ハッカやペパーミントから得られる観点から、(-)-メントールが好ましい。(-)-メントールは、l-メントールとも呼ばれる。メントールは、天然エキス又は天然油として配合されていてもよく、例えばハッカエキス、ハッカ油、ミントエキス、ミント油等が配合された眼科用液体製剤も、メントールを含む眼科用液体製剤に該当する。
本発明において清涼化剤の濃度は、眼科用液体製剤に使用する観点から、清涼化剤の種類に応じて適宜決定することができる。清涼化剤としてメントールを使用する場合、例えば0.001%~0.1%の濃度で配合されうる。清涼感を高める観点から、メントールは0.002%以上で配合されることが好ましく、0.01%以上で配合されることがさらに好ましい。適切な使用感を得る観点から、0.05%以下で配合されることが好ましく、0.03%以下で配合されることがさらに好ましい。
清涼化剤としてハッカ油を用いる場合、例えば0.0002%~0.1%の濃度で配合されうる。清涼感を高める観点から、ハッカ油は0.001%以上で配合されることが好ましく、0.01%以上で配合されることがさらに好ましい。適切な使用感を得る観点から、0.05%以下で配合されることが好ましく、0.02%以下で配合されることがさらに好ましい。
清涼化剤としてユーカリ油を用いる場合、例えば0.00005%~0.05%の濃度で配合されうる。清涼感を高める観点から、ユーカリ油は0.0005%以上で配合されることが好ましく、0.001%以上で配合されることがさらに好ましい。適切な使用感を得る観点から、0.01%以下で配合されることが好ましく、0.005%以下で配合されることがさらに好ましい。
清涼化剤としてベルガモット油を用いる場合、例えば0.00002%~0.02%の濃度で配合されうる。清涼感を高める観点から、ベルガモット油は0.0002%以上で配合されることが好ましく、0.0005%以上で配合されることがさらに好ましい。適切な使用感を得る観点から、0.005%以下で配合されることが好ましく、0.002%以下で配合されることがさらに好ましい。
清涼化剤としてゲラニオールを用いる場合、例えば0.0001%~0.1%の濃度で配合されうる。清涼感を高める観点から、ゲラニオールは0.0005%以上で配合されることが好ましく、0.001%以上で配合されることがさらに好ましい。適切な使用感を得る観点から、0.03%以下で配合されることが好ましく、0.01%以下で配合されることがさらに好ましい。
清涼化剤としてカンフルを用いる場合、例えば0.0001%~0.1%の濃度で配合されうる。清涼感を高める観点から、カンフルは0.0005%以上で配合されることが好ましく、0.001%以上で配合されることがさらに好ましい。適切な使用感を得る観点から、0.03%以下で配合されることが好ましく、0.01%以下で配合されることがさらに好ましい。
清涼化剤としてボルネオールを用いる場合、例えば0.0001%~5%の濃度で配合されうる。清涼感を高める観点から、ボルネオールは0.0005%以上で配合されることが好ましく、0.005%以上で配合されることがさらに好ましい。適切な使用感を得る観点から、0.5%以下で配合されることが好ましく、0.02%以下で配合されることがさらに好ましい。
本発明において眼科用液体製剤は、水を基剤とする水性液体製剤であるが、点眼薬に使用しうる任意の液体基剤をさらに含んでいてもよい。本発明の眼科用液体製剤は、点眼薬として許容されるpH、浸透圧を有するように調製される。眼科用液体製剤のpHは、pH調整剤を使用して5.0~9.0に調整することができ、一例として5.5~8.5である。一例として6.0~8.0にも調整することもできる。眼科用液体製剤の浸透圧比の一例として、好ましくは0.5~2.5、より好ましくは0.7~1.5になるように眼科用液体製剤に添加する成分量を調節される。
本発明の眼科用液体製剤は、好ましくは点眼剤、より好ましくは一般用点眼剤であり、清涼感を与える観点からメントール、ベルガモット油、ゲラニオール、及びボルネオールからなる群から選ばれる清涼化剤が配合される。本発明の眼科用液体製剤にはブリモニジン及び/又はその塩がさらに配合される。低濃度のブリモニジン及び/又はその塩は目の充血を緩和するかまたは抑制する薬効を有する。目の充血を緩和するかまたは抑制するとは、白目部分の白みを増加させることを言い、目のホワイトニングともいうことができる。目の充血を緩和するかまたは抑制する観点から、本発明の眼科用液体製剤は、低濃度のブリモニジン及び/又はその塩を含むことが好ましい。
白目の表面は、強膜上を覆う結膜から構成されており、結膜はさらに瞼の内側を覆っている。結膜表面は、結膜上皮細胞から主に構成される。結膜の上皮層には血管、繊維組織、リンパ管が含まれる。結膜は白目と黒目の境界で角膜と接しており、角膜と結膜は外界にさらされる目の最外層を構成する。結膜は外界にさらされていることから、細菌やウイルスなどに侵されやすく、炎症が生じやすい。また炎症が起こっていない場合であっても、寝不足や目の酷使により、目に酸素や栄養を供給するために血流量が増えて充血する。本発明においてブリモニジン及び/又はその塩は、結膜内の毛細血管に到達し、毛細血管の血流量を低減することで、充血緩和、目の赤みの低減又は白化という作用を発揮する。
本発明の眼科用液体製剤は、点眼薬に使用しうる任意の成分を、本発明の効果を損なわない範囲で含有することができる。本発明の眼科用液体製剤に含まれる清涼化剤及びブリモニジン酒石酸塩とは別に、任意の有効成分及び添加物などの任意的成分を含んでもよい。このような任意的成分としては、充血除去剤、ピント調節機能改善剤、消炎・収れん剤、抗ヒスタミン剤、ビタミン類、保水・栄養成分、サルファ剤、防腐剤、pH調整剤、等張化剤、増粘剤、抗酸化剤、溶解補助剤、安定化剤、界面活性剤などが挙げられるが、これらに限定されることを意図するものではない。これらの有効成分及び添加剤は、それぞれ各カテゴリーの中から1種類のみを用いてもよいし、複数の種類を組み合わせて使用することができる。以下に、任意的成分を説明するが、列挙された作用以外の作用を目的として使用されてもよい。
ビタミン類とは、生物の生存又は生育に微量に必要な栄養素のうち、その生物の体内で十分な量を合成できない炭水化物・タンパク質・脂質以外の有機化合物の総称である。ビタミン類としては、水溶性ビタミン類と、脂溶性ビタミン類に大別される。水溶性ビタミン類としては、ビタミンB類及びビタミンC(アスコルビン酸)が挙げられる。脂溶性ビタミンとしては、ビタミンA類、ビタミンD類、ビタミンE類、ビタミンK類が挙げられる。一般用医薬品製造(輸入)承認基準では、点眼薬に配合されるビタミン類が規定されており、この観点から、特にビタミンA類、ビタミンB類、ビタミンE類が好ましい。
ビタミンA類としては、レチノール、及びその関連物質が使用されうる。レチノール関連物質として、レチナール、レチノイン酸、パルミチン酸レチノールやその他のレチノイド、例えばイソトレチノイン、アリトレチノイン、アシトレチン、エトレチナート、アダパレン、タザロテン、ベキサロテン等も挙げられる。点眼薬として配合する観点からパルミチン酸レチノール、酢酸レチノールが好ましい。ビタミンA類は上皮細胞に作用し増殖を誘導することから、角膜や結膜保護等を目的として点眼薬に配合されうる。夜盲症、結膜乾燥症、角膜乾燥症、角膜軟化症等の眼科系疾患の治療のためにも点眼薬に配合されうる。
ビタミンB類としては、ビタミンB1(チアミン等)、ビタミンB2、ビタミンB3(ナイアシン等)、ビタミンB5、ビタミンB6、ビタミンB7(ビオチン等)、ビタミンB9(葉酸等)、ビタミンB12が使用されうる。ビタミン類には、誘導体であるプロビタミンや医薬として許容される塩を包含する。ビタミンB類の中でも、特に点眼薬に配合される観点から、ビタミンB2、B5、B6、B12が好ましい。
ビタミンB2としてはリボフラビン、リン酸リボフラビン、酪酸リボフラビン、酢酸リボフラビン、フラビンアデニンジヌクレオチド、フラビンモノヌクレオチド、又は薬学的に許容されるそれらの塩等が使用されうる。塩としては、ナトリウム塩、カリウム塩等が挙げられる。点眼薬として配合される観点からフラビンアデニンジヌクレイオチドナトリウムが好ましい。ビタミンB2は、酸化還元に直接関与し、点眼薬として使用されると角膜・結膜細胞の酵素呼吸代謝を促進し、角膜・結膜の保護作用を有する。またビタミンB2は、ビタミンB2欠乏または代謝障害が関与すると推定される角膜炎に対して治療のために点眼薬に配合されうる。
ビタミンB5としてはパンテノール、パントテン酸又はそれらの誘導体又はそれらの塩が使用されうる。一例として、パンテノール、パントテン酸の他に、誘導体又はそれらの塩として、パンテチン、パンテテイン、パントテニールアルコール、パントテニールエチルエーテル、パンテテインパントテニールアルコール、パントテン酸カルシウム、パントテン酸ナトリウム等が挙げられる。点眼薬として使用される観点から、ビタミンB5としてはパンテノール、パントテン酸カルシウム、パントテン酸ナトリウムが好ましい。
ビタミンB6としては、ピリドキサール、ピリドキサミン、ピリドキシン又はそれらの医薬として許容される塩が使用されうる。点眼薬として使用される観点から、ピリドキシン塩酸塩が好ましい。ビタミンB6は、生体内でアミノ酸脱炭酸酵素及びアミノ基転移酵素の補酵素として、タンパク質の代謝に関わっており、眼精疲労の抑制のために点眼薬に配合されうる。
ビタミンB12はコリン環にコバルトが配位した構造を有する化合物であり、具体的には、シアノコバラミン、メコバラミン(メチルコバラミン)、ヒドロキソコバラミン、アデノシルコバラミン、塩酸ヒドロキソコバラミン、酢酸ヒドロキソコバラミン等が挙げられる。ビタミンB12は疲れ目や眼精疲労の改善等の薬理作用のために点眼薬に配合されうる。
ビタミンE類としては、トコフェロール、トコトリエノール、トコフェルソラン、又はそれらの誘導体が使用されうる。トコフェロール、トコトリエノールは、α-、β-、γ-、δ-のいずれであってもよく、また、d体、dl体のいずれであってもよい。点眼薬として使用される観点から、一例として酢酸d-α-トコフェロールが挙げられる。
ビタミンCとしては、アスコルビン酸又はその塩が挙げられる。ビタミンD類としては、ビタミンD2(エルゴステロール、エルゴカルシフェロール)、D3(7-デヒドロコレステロール)、プレビタミンD3(コレカルシフェロール、25-ヒドロキシコレカルシフェロール、カルシトリオール(1,25-ジヒドロキシコレカルシフェロール)、カルシトロン酸)、ビタミンD4(ジヒドロエルゴカルシフェロール)、ビタミンD5(ジヒドロタキステロール・カルシポトリオール・タカルシトール・パリカルシトール)等が挙げられる。ビタミンK類としては、フィロキノン(K1)、メナキノン(K2)、メナジオン(K3)が挙げられる。
充血除去剤としては、エピネフリン、エフェドリン、テトラヒドロゾリン、ナファゾリン、フェニレフリン、メチルエフェドリン、又はそれらの塩が使用されうる。
ピント調節機能改善剤としては、メチル硫酸ネオスチグミンが使用されうる。
消炎・収れん剤としては、ε-アミノカプロン酸、アラントイン、ベルベリン又はその塩、アズレンスルホン酸又はその塩、グリチルリチン酸又はその塩、硫酸亜鉛、乳酸亜鉛、塩化リゾチームが使用されうる。
抗ヒスタミン剤としては、塩酸ジフェンヒドラミン、マレイン酸クロルフェニラミンが使用されうる。
保水・栄養成分としては、アミノ酸類又はその塩、コンドロイチン硫酸エステルナトリウムが使用されうる。本発明においてアミノ酸類は、安定性の向上及び結膜組織移行性の向上を目的として眼科用液体製剤に配合されるが、保水・栄養成分として同一又は異なるアミノ酸類を配合することもできる。アミノ酸類又はその塩としては、アミノ酸類とは、アミノ酸以外に、アミノ酸のカルボキシル基の代わりに硫酸基を有する物質、例えばタウリンにも関する。アミノ酸としては、一例として、グリシン、アラニン、メチオニン、バリン、トレオニン、グルタミン、グルタミン酸、アスパラギン、アスパラギン酸、システイン、ヒスチジン、イソロイシン、ロイシン、リシン、フェニルアラニン、トリプトファン、アルギニン、プロリン、チロシン、セリンが挙げられる。アミノ酸としては、好ましくはアスパラギン酸、メチオニン、グリシンが挙げられる。グリシンを除くアミノ酸は、L体のアミノ酸であってもよいし、D体のアミノ酸であってもよいし、DL体のアミノ酸であってもよい。
サルファ剤としては、スルファメトキサゾール、スルファメトキサゾールナトリウム、スルフイソキサゾール、スルフイソミジンナトリウムが使用されうる。
防腐剤としては、パラオキシ安息香酸メチル、パラオキシ安息香酸エチル、パラオキシ安息香酸プロピル、パラオキシ安息香酸ブチル、硫酸オキシキノリン、ベンザルコニウム塩化物、クロロブタノール、亜塩素酸ナトリウム、ベンゾドデシニウム臭化物、クロルヘキシジングルコン酸塩、ソルビン酸塩、デヒドロ酢酸ナトリウム、安息香酸塩、ベンジルアルコール、アルキルポリアミノエチルグリシン塩酸塩、ポリヘキサエチレンビグアニド、ホウ酸、ホウ砂などが使用されうる。
pH調整剤としては、クエン酸緩衝剤、酢酸緩衝剤、炭酸緩衝剤、ホウ酸緩衝剤、リン酸緩衝剤などの緩衝剤とともに、例えば、塩酸、酢酸、ホウ酸、炭酸、硫酸、リン酸、クエン酸、酒石酸などの酸や、水酸化ナトリウム、炭酸水素ナトリウム、炭酸ナトリウム、トリエタノールアミン、モノエタノールアミンなどの塩基が使用されうる。
等張化剤としては、糖類や塩類が挙げられ、塩類としては、亜硫酸水素ナトリウム、亜硫酸ナトリウム、塩化カリウム、塩化カルシウム、塩化ナトリウム、塩化マグネシウム、酢酸カリウム、酢酸ナトリウム、炭酸水素ナトリウム、炭酸ナトリウム、チオ硫酸ナトリウム、硫酸マグネシウム、リン酸水素二ナトリウム、リン酸二水素ナトリウム、リン酸二水素カリウムなどが用いられうる。糖類としては、任意の単糖類又は多糖類を使用することができ、一例としてグルコース、シクロデキストリン、キシリトール、ソルビトール、マンニトール等を使用することができる。
増粘剤としては、コンドロイチン硫酸ナトリウム、ポリビニルアルコール、カルボキシビニルポリマー、ヒドロキシエチルセルロース、ヒドロキシプロピルメチルセルロース、メチルセルロース、アルギン酸、ヒアルロン酸、ポリビニルピロリドンなど又はそれらの塩が使用されうる。
溶解補助剤としては、ポリオキシエチレンソルビタンモノオレエート、ポリオキシエチレン硬化ヒマシ油、チロキサポール、プルロニック等の非イオン性界面活性剤;グリセリン、マクロゴール等の多価アルコール等が使用されうる。
安定化剤としては、例えば、ポリビニルピロリドン、亜硫酸塩、モノエタノールアミン、グリセリン、プロピレングリコール、ポリエチレングリコール、シクロデキストリン、デキストラン、アスコルビン酸、エデト酸塩、タウリン、トコフェロール等が使用されうる。
界面活性剤としては、例えば、チロキサポール、ポリオキシエチレン硬化ヒマシ油、ポリオキシエチレンポリオキシプロピレンブロックコポリマー、ポリオキシエチレンソルビタン脂肪酸エステル、オクトキシノール等の非イオン性界面活性剤;アルキルジアミノエチルグリシン、ラウリルジメチルアミノ酢酸ベタイン等の両性界面活性剤;アルキル硫酸塩、N-アシルタウリン塩、ポリオキシエチレンアルキルエーテルリン酸塩、ポリオキシエチレンアルキルエーテル硫酸塩等の陰イオン界面活性剤;アルキルピリジニウム塩、アルキルアミン塩等の陽イオン界面活性剤等が使用されうる。

以下に説明する本発明の実施例は例示のみを目的とし、本発明の技術的範囲を限定するものではない。本発明の技術的範囲は特許請求の範囲の記載によってのみ限定される。本発明の趣旨を逸脱しないことを条件として、本発明の変更、例えば、本発明の構成要件の追加、削除及び置換を行うことができる。
[試験例1]細胞傷害性試験
1.試験点眼液の調製
精製水にl-メントール(鈴木薄荷社製)を添加して攪拌し、溶解を確認したのち、ホウ酸(BORAX社製)及びブリモニジン酒石酸塩(HANGZHOU DELI CHEMICAL社製)を加えた。次いで、pH調整剤(1mol/L 塩酸又は0.1N NaOH)によりpHを調製して塩化ナトリウム(富田製薬社製)を添加し、精製水で100mlにメスアップすることで、表1の比較例1~3及び実施例1の試験点眼液を得た。処方に応じてl-メントール、ブリモニジン酒石酸塩は適宜添加された。なお、表中の数値は、pHを除きw/v%である。

2.細胞傷害性試験
96ウェルプレート(Falcon)に、1.5×104個の不死化ヒト角膜上皮細胞(HCE-t)を増殖用細胞培養培地(血清、上皮成長因子、抗生物質、及びインスリンを添加したDMEM/F12)100μLと共に播種し、37℃5%CO2インキュベーターで16時間以上培養して定着させた。
顕微鏡でウェル内に十分な細胞が存在しウェルに定着していることを確認し、ウェル内の増殖用細胞培養培地を吸引した。各ウェルをDPBS(Thermo Fisher Scientific社製)100μLずつで洗浄した。各ウェルに調製された試験点眼液100μLを添加した。37℃5%CO2のインキュベーター内で約30分インキュベートした。インキュベート後、各ウェルの試験溶液を吸引し、DPBS100μLで洗浄後、DMEM/F12(Thermo Fisher Scientific社製)90%、Cell counting kit-8(同仁化学研究所社製)10%の割合の溶液を100μL添加し、37℃5%CO2のインキュベーターで約1時間以上インキュベートした。インキュベート後、マイクロプレートリーダーで450nmの吸光度を測定した。細胞を含まないウェルでの試験をローコントロールとし、試験溶液の代わりに増殖用細胞培養培地を用いた試験をハイコントロールとした。結果を図1に示す。メントールを添加することで、細胞生存率が低下する一方、メントールとブリモニジンとを併用することで、細胞生存率の低下が抑制された。

[試験例2]メントール濃度を変化させた場合のブリモニジンによる細胞傷害性の低減効果
1.試験点眼液の調製

顕微鏡でウェル内に十分な細胞が存在しウェルに定着していることを確認し、ウェル内の増殖細胞培養培地を吸引した。各ウェルをDPBS100μLずつで洗浄した。各ウェルに調製された試験点眼液100μLを添加した。37℃5%CO2のインキュベーターで約30分インキュベートした。インキュベート後、各ウェルの試験溶液を吸引し、DPBS100μLで洗浄後、DMEM/F12 90%、Cell counting kit-8 10%の割合の溶液を100μL添加し、37℃5%CO2のインキュベーターで約1時間以上インキュベートした。インキュベート後、マイクロプレートリーダーで450nmの吸光度を測定した。細胞を含まないウェルでの試験をローコントロールとし、試験溶液の代わりに増殖細胞培養培地を用いた試験をハイコントロールとした。各試験点眼液の吸光度(ABS)とローコントロール及びハイコントロールの吸光度(ABS)から、下記の式に基づき細胞死滅抑制率(%)を求め、図2に示した。0.025w/v%ブリモニジンによるメントールの細胞傷害性抑制効果は、メントール濃度が0.002w/v%~0.03w/v%の範囲で見られ、なかでも0.01w/v%時に最も効果が大きかった。

[試験例3]充血除去成分についての細胞傷害性低減効果
1.試験点眼液の調製
精製水にl-メントール(鈴木薄荷社製)及びポリソルベート80(販売元日油社製)を添加して攪拌し、溶解を確認したのち、ホウ酸(BORAX社製)及び、充血除去成分としてブリモニジン酒石酸塩(HANGZHOU DELI CHEMICAL社製)、テトラヒドロゾリン塩酸塩(販売元東京化成工業社製)、又はナファゾリン塩酸塩(販売元東京化成工業社製)を加えた。次いでpH調整剤(1mol/L 塩酸又は0.1N NaOH)によりpHを調製して塩化ナトリウム(富田製薬社製)を添加し、精製水で100mlにメスアップすることで、表3の比較例6’、比較例7~8、及び実施例6の試験点眼液を得た。処方に応じて、ブリモニジン酒石酸塩、テトラヒドロゾリン塩酸塩、又はナファゾリン塩酸塩は適宜添加された。なお、表中の数値は、pHを除きw/v%である。
1.試験点眼液の調製

2.細胞傷害性試験
96ウェルプレート(販売元Falcon)に、1.5×104個の不死化ヒト角膜上皮細胞(HCE-t)を増殖用細胞培養培地(血清、上皮成長因子、抗生物質、及びインスリンを添加したDMEM/F12)100μLと共に播種し、37℃5%CO2インキュベーターで16時間以上培養して定着させた。
顕微鏡でウェル内に十分な細胞が存在しウェルに定着していることを確認し、ウェル内の細胞培養培地を吸引した。各ウェルをDPBS100μLずつで洗浄した。各ウェルに調製された試験点眼液100μLを添加した。37℃5%CO2のインキュベーターで約60分インキュベートした。インキュベート後、各ウェルの試験溶液を吸引し、DPBS100μLで洗浄後、DMEM/F12 90%、Cell counting kit-8 10%の割合の溶液を100μL添加し、37℃5%CO2のインキュベーターで約1時間以上インキュベートした。インキュベート後、マイクロプレートリーダーで450nmの吸光度を測定した。細胞を含まないウェルでの試験をローコントロールとし、試験溶液の代わりに細胞培養培地を用いた試験をハイコントロールとした。結果を図3に示す。ブリモニジン酒石酸塩と同じ充血除去作用があるテトラヒドロゾリン及びナファゾリンではl-メントールの細胞傷害性が低減されず、l-メントールの細胞傷害性を低減する効果は、充血除去成分のなかでもブリモニジン酒石酸塩に特徴的な効果であることがわかった。

[試験例4]官能評価試験
1.試験点眼液の調製
精製水にl-メントール(鈴木薄荷社製)を添加して攪拌し、溶解を確認したのち、ホウ酸(BORAX社製)、塩化ナトリウム(富田製薬社製)、ベンザルコニウム塩化物(岡見化学工業社製)及びブリモニジン酒石酸塩(HANGZHOU DELI CHEMICAL社製)を加え、pH調整剤(1mol/L 塩酸又は0.1N NaOH)によりpHを調製し、精製水で100mlにメスアップすることで、表4の実施例7(B-06とラベル)及び比較例9(B-05とラベル)の試験点眼液を調製した。調製された試験点眼液を0.22μmのフィルターに通してろ過し、滅菌済みの点眼容器に充てんして試験に供した。なお、調製後及び使用感試験実施期間中にl-メントール及びブリモニジン酒石酸塩の含量分析を行い、所定量含有されていることを確認した。

精製水にl-メントール(鈴木薄荷社製)を添加して攪拌し、溶解を確認したのち、ホウ酸(BORAX社製)、塩化ナトリウム(富田製薬社製)、ベンザルコニウム塩化物(岡見化学工業社製)及びブリモニジン酒石酸塩(HANGZHOU DELI CHEMICAL社製)を加え、pH調整剤(1mol/L 塩酸又は0.1N NaOH)によりpHを調製し、精製水で100mlにメスアップすることで、表4の実施例7(B-06とラベル)及び比較例9(B-05とラベル)の試験点眼液を調製した。調製された試験点眼液を0.22μmのフィルターに通してろ過し、滅菌済みの点眼容器に充てんして試験に供した。なお、調製後及び使用感試験実施期間中にl-メントール及びブリモニジン酒石酸塩の含量分析を行い、所定量含有されていることを確認した。
事前に眼に疾患がなく、臨床試験の倫理審査委員会(外部機関)で承認された同意説明文書に対して同意を得た被験者4名(被験者No.1~4)が点眼試験に参加した。被験者は、各検体を点眼したのち、表5の基準に基づき、それぞれの検体についてアンケートに回答した。実施した試験方法の詳細を下記に示した。
1)コンタクトレンズ装用者は点眼する10分以上前にあらかじめコンタクトレンズを外した。
2)被験者は容器にB-05及びB-06とラベリングされた試験点眼液を受け取った。
3)左眼にB-05を1回1滴点眼し、アンケートの表5 1)「清涼感評価基準」の項に回答した。
4)右眼にB-06を1回1滴点眼し、3)と同様に、アンケートの表5 1)「清涼感評価基準」の項に回答した。
5)B-05及びB-06の「清涼感の強さ」及び「清涼感の持続」を比較する目的で、アンケートの表5 2)「B-05及びB-06の清涼感比較」及び3)「B-05及びB-06の「清涼感の持続の比較」の項に回答した。
6)このアンケートを提出するまで、点眼液の処方は被験者に開示しなかった。

6)このアンケートを提出するまで、点眼液の処方は被験者に開示しなかった。
B-06(実施例7)はB-05(比較例9)に比べて、清涼感の平均評価点数が高く、使用感の比較において清涼感を強く感じ、且つその清涼感が持続することが確認できた。
以上のことから、l-メントール含有眼科用剤に0.025%ブリモニジニン酒石酸塩を配合することによって清涼感を増幅し、また、その清涼感を持続させることができることがわかった。

[試験例5]清涼化剤による細胞毒性試験
1.試験点眼液の調製
精製水に清涼化剤(ベルガモット油(富士フイルム和光純薬)、ゲラニオール(富士フイルム和光純薬)、ボルネオール(富士フイルム和光純薬))及び及びポリソルベート80(日油社製)を添加して攪拌し、溶解を確認したのち、ホウ酸(小堺製薬社製)及びブリモニジン酒石酸塩(HANGZHOU DELI CHEMICAL社製)を加えた。次いで、pH調整剤(1mol/L 塩酸又は0.1N NaOH)によりpHを6.5に調整して塩化ナトリウム(富田製薬社製)を添加し、精製水で100mlにメスアップすることで、表8の実施例8~10及び比較例10~14の試験点眼液を得た。なお、表中の数値は、w/v%である。

精製水に清涼化剤(ベルガモット油(富士フイルム和光純薬)、ゲラニオール(富士フイルム和光純薬)、ボルネオール(富士フイルム和光純薬))及び及びポリソルベート80(日油社製)を添加して攪拌し、溶解を確認したのち、ホウ酸(小堺製薬社製)及びブリモニジン酒石酸塩(HANGZHOU DELI CHEMICAL社製)を加えた。次いで、pH調整剤(1mol/L 塩酸又は0.1N NaOH)によりpHを6.5に調整して塩化ナトリウム(富田製薬社製)を添加し、精製水で100mlにメスアップすることで、表8の実施例8~10及び比較例10~14の試験点眼液を得た。なお、表中の数値は、w/v%である。
2.細胞傷害性試験

顕微鏡でウェル内に十分な細胞が存在しウェルに定着していることを確認し、ウェル内の増殖用細胞培養培地を吸引した。各ウェルをDPBS(Thermo Fisher Scientific社製)100μLずつで洗浄した。各ウェルに調製された試験点眼液100μLを添加した。37℃5%CO2のインキュベーター内で約60分インキュベートした。インキュベート後、各ウェルの試験溶液を吸引し、DPBS100μLで洗浄後、DMEM/F12(Thermo Fisher Scientific社製)90%、Cell counting kit-8(同仁化学研究所社製)10%の割合の溶液を100μL添加し、37℃5%CO2のインキュベーターで約1時間以上インキュベートした。インキュベート後、マイクロプレートリーダーで450nmの吸光度を測定した(図5)。細胞を含まないウェルでの試験をローコントロールとし、試験溶液の代わりに増殖用細胞培養培地を用いた試験をハイコントロールとした。
下記の数式に基づいて、細胞死滅抑制率(%)を計算した:
細胞死滅抑制率(%)についての結果を図6に示す。

下記の数式に基づいて、細胞死滅抑制率(%)を計算した:
試験例5の細胞傷害性試験により、清涼化剤であるベルガモット油、ボルネオール、及びゲラニオールが細胞傷害性を有しており、ブリモニジン酒石酸塩を併用することでその細胞傷害性が軽減することが示された。
製剤例
本発明のブリモニジン及び/又はその塩と、清涼化剤とを含有する医薬は、例えば、次のような処方によって製造することができる。製剤例を挙げて本発明の薬剤をさらに具体的に説明するが、本発明はこれらの製剤例にのみ限定されるものではない。

(6)pH調整剤 適量
(7)滅菌精製水 適量
(1)~(6)を、(7)滅菌精製水に溶解して、pHを6.5に調整し、点眼液を調製する。

(1)ブリモニジン酒石酸塩 0.025g

Claims (8)
- 0.01w/v%~0.05w/v%のブリモニジン及び/又はその塩と、清涼化剤とを含有する眼科用液体製剤。
- 前記清涼化剤が、メントール、ベルガモット油、ゲラニオール及びボルネオールからなる群から選ばれる、請求項1に記載の眼科用液体製剤。
- 前記メントールが、l-メントールである、請求項2に記載の眼科用液体製剤。
- 前記メントールが、0.001%~0.05%で配合される、請求項2又は3に記載の眼科用液体製剤。
- 前記清涼化剤が、以下の:
0.00002%~0.02%のベルガモット油;
0.0001%~0.1%のゲラニオール;及び
0.0001%~5%のボルネオール
から選ばれる、請求項1又は2に記載の眼科用液体製剤。 - 0.01w/v%~0.05w/v%のブリモニジン及び/又はその塩を含む、清涼化剤を含有する眼科用液体製剤についての清涼感増強剤。
- 前記清涼化剤が、メントール、ベルガモット油、ゲラニオール、及びボルネオールからなる群から選ばれる、請求項6に記載の清涼感増強剤。
- 0.01w/v%~0.05w/v%のブリモニジン及び/又はその塩を含む、清涼化剤を含有する眼科用液体製剤についての細胞傷害性低減剤。
