Description

각각의 치료제 개발 현황을 살펴보면, P2X7 antagonist는 화이자, 아스트라제네카, 얀센과 같은 글로벌 제약사에서 류마티스 관절염, 크론병, 우울증 등을 적응증으로 하여 개발에 착수하였으나 임상에서 효능이 미달되어 대부분 개발이 중단되었다. 이후 벤처기업에서 개발을 이어갔으나 2020년 기준 P2X7을 타겟으로 하는 치료제 개발 파이프라인은 10개 미만에 불과하며 현재 임상단계에 접어든 신약은 얀센에서 우울증 치료제로서 개발중인 18F-JNJ-64413739으로 임상 1상을 마친 상태이며 다른 파이프라인들은 전임상 혹은 개발단계에 머물러 있다.
Caspase-1 inhibitor는 임상에서 유효성 목표 미달 및 안전성 이슈로 2020년 중단됨에 따라 글로벌 개발 파이프라인이 없는 실정이다.
NLRP3 inhibitor는 OLATEC, INFLAZOME, IFM THERAPEUTICS, NODTHERA, AC Immune과 같은 벤처기업에서 골관절염, 수축기 심부전, 파킨슨병, 염증성 장 질환, 알츠하이머병 등을 적응증으로 하여 개발 중에 있다.
전술한 바와 같이 인플라마좀-관련 signal을 타겟으로 한 약물은 2020년 기준 전 세계에서 20개 미만의 회사에서 개발을 시도하는 초기개발 단계에 머물러 있다. 2020년 기준 염증성 질환을 치료하기 위한 목적으로 임상 실험을 진행하고 있는 신약 파이프라인은 3종, 전임상 단계에서 개발되고 있는 신약 파이프 라인은 5종이 있다. 하지만 현재까지 알려진 신약 후보물질은 IL-1β, IL-18와 같은 염증활성으로 촉발된 염증성 사이토카인은 선택적으로 억제하나 염증개시단계에서 촉발된 TNFα 염증성 사이토카인은 억제하지 못하는 항염 효능의 한계가 있다. 이는 NLRP3, P2X7를 타겟으로 하는 신약이 가지고 있는 기본적인 한계로 염증 활성단계만을 선택적으로 차단하는 약리기전을 지니고 있는 약물은 다양한 염증 우회경로를 통한 염증반응을 억제하지 못하는 점에서 기인한다. 또한 NLRP3와 P2X7에 내재해 있는 redundancy와 유전적 다형성 때문에 실제 임상 약효 반응률이 낮다는 단점도 있다.
한편, GPCR-gated ion channel은 현재까지 GIRK1/2/3/4의 4 종류가 알려져 있으며 심근계 이온조절, 신경통증 신호, 알코올 중독 등의 생리현상에 연관되어 있다고 알려져 있으나 정확한 기작과 상호연관 수용체의 관계는 자세히 알려져 있지 않은 실정이다.
이에, 본 발명자들은 인플라마좀-관련 signal을 타겟으로 하는 약물들의 한계를 극복하기 위한 방법으로 GPCR-gated ion channel을 타겟으로 하는 약리 기전을 연구하였고, 그 결과 P2X7이 GPCR19-regulated ion channel임을 새로이 밝힘으로써, 본 출원에 이르렀다. 구체적으로, 본 발명자들은 특정 GPCR19 작용제가 NLRP3 inflammasomal activation에 중요한 역할을 하는 P2X7 ion channel을 조절한다는 점을 새로이 밝혔다. 즉, GPCR19가 P2X7 수용체와 상호 결합하여 P2X7 이온 채널을 상위 조절하며 이를 통해 NLRP3 inflammasome을 조절하는 기전을 밝혔다. 또한 GPCR19 및 P2X7 수용체 복합체의 상호작용을 조절하는 물질이 GPCR19-cAMP 신호전달 활성을 통해 염증개시유래 TNFα를 억제함과 동시에 GPCR19 매개 P2X7 신호전달억제를 통해 염증활성유래 IL-1β를 억제하는 약리기전을 밝힘으로써, 본 출원에 이르게 되었다.
본 발명의 목적은 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 조절하는 물질을 스크리닝하는 방법을 제공하는 것이다.
본 발명의 다른 목적은 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 이용한 NLRP3 인플라마좀-관련 질환 예방 또는 치료를 위한 물질을 스크리닝하는 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 약학적으로 유효한 양의 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질을 개체에 투여하는 단계를 포함하는, NLRP3 인플라마좀-관련 질환 예방 또는 치료 방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은
1) GPCR19 및 P2Xn 수용체를 발현하는 세포에 후보물질 및 제1물질을 처리하는 단계;
2) 상기 단계 1)의 세포에 제2물질을 처리하는 단계;
3) 상기 단계 2)의 세포에서 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 측정하는 단계; 및
4) 상기 단계 3)의 상호작용 측정 결과, 후보물질 무처리 대조군과 비교하여 GPCR19 및 P2Xn 수용체 복합체의 상호작용에 차이가 나는 후보물질을 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 조절하는 물질로 판단하는 단계를 포함하는,
GPCR19 및 P2Xn 수용체 복합체의 상호작용을 조절하는 물질을 스크리닝하는 방법을 제공한다.
또한, 본 발명은
1) GPCR19 및 P2Xn 수용체를 발현하는 세포에 후보물질 및 제1물질을 처리하는 단계;
2) 상기 단계 1)의 세포에 제2물질을 처리하는 단계;

아울러, 본 발명은 약학적으로 유효한 양의 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질을 개체에 투여하는 단계를 포함하는, NLRP3 인플라마좀-관련 질환 예방 또는 치료 방법을 제공한다.
본 발명에서는 GPCR19 및 P2X7 수용체가 상호 결합하여 상호작용하는 것을 확인하였다. 또한, 미생물로 인한 PAMP, ATP와 같은 생체 물질로 인한 DAMP 스트레스 염증 유발 상황에서 P2X7을 매개로 하는 NLRP3 inflammasomal activation pathway가 GPCR19에 의해서 조절되는 생리기전을 확인하였다. 아울러, GPCR19와 P2X7 수용체 간 상호 결합을 유도하는 물질을 이용하여 염증반응 중에 GPCR19와 P2X7 수용체 간 상호 결합을 유도하면 P2X7으로부터 시작되는 염증 반응을 예방 또는 완화할 수 있음을 확인하였다. 이에, 상기 GPCR19 및 P2X7 수용체 간 상호작용을 조절하는 물질을 스크리닝하고, 상기 스크리닝한 물질을 이용하여 염증성 질환을 포함한 NLRP3 인플라마좀-관련 질환(NLRP3 inflammasome-associated disease) 예방 또는 치료를 위한 제제를 개발할 수 있다.

본 발명은 GPCR19-P2Xn 수용체 복합체 및 이의 용도에 관한 것으로, 구체적으로 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 조절하는 물질을 스크리닝하는 방법; GPCR19 및 P2Xn 수용체 복합체의 상호작용을 이용한 NLRP3 인플라마좀-관련 질환 예방 또는 치료를 위한 물질을 스크리닝하는 방법; 및 약학적으로 유효한 양의 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질을 개체에 투여하는 단계를 포함하는, NLRP3 인플라마좀-관련 질환 예방 또는 치료 방법에 관한 것이다.
이하, 본 발명은 하기의 실시예에 의해 상세히 설명한다.
본 발명은 GPCR19(G-protein coupled receptor 19) 및 P2Xn 수용체를 코딩하는 유전자를 포함하는 벡터를 제공한다.
또한, 본 발명은 상기 벡터로 형질전환된 세포를 제공한다.
또한, 본 발명은 상기 세포로부터 분리된 GPCR19 및 P2Xn 수용체 복합체를 제공한다.
본 발명에서, 상기 GPCR19는 인간 GPBAR1 유전자에 의해 코딩되는 단백질로, G-protein coupled receptor superfamily 중 하나이다. GPCR19는 담즙산에 대한 세포 표면 수용체 역할을 한다. 구체적으로, 담즙산에 의해 GPCR19가 활성화되면 cAMP를 생산하고, cAMP에 의해 MAP kinase signal pathway가 활성화되어, NF-κB action을 조절한다.
본 발명에서, 상기 P2Xn 수용체는 2-transmembrane family 중 하나이다. P2Xn 수용체는 비-선택적 양이온 채널을 포함하여 빠른 시냅스 전달에 역할을 할 수 있다. 또한 P2Xn 수용체는 7개의 subtype, 보다 구체적으로 P2X1, P2X2, P2X3, P2X4, P2X5, P2X6, P2X7 수용체가 알려져 있으며, 본 발명에서는 P2X7이 바람직하나 이에 제한되는 것은 아니다.
본 발명에서, 상기 GPCR19 및 P2Xn을 코딩하는 유전자는 전장 및/또는 단편의 형태로 사용될 수 있다. 구체적으로, 본 발명에 개시된 단백질을 코딩하는 야생형 유전자 서열 및 그 단편은 물론 세포 내에서의 발현 또는 단백질의 안정성 등과 같은 특징이 유리하도록 염기서열의 일부가 인위적으로 변형된 유전자, 및 자연적으로 발견되는 염기서열 일부가 변형된 유전자 또는 이들의 모두의 단편을 포함하는 것이다. 유전자 염기서열의 변형은 상응하는 아미노산의 변형을 수반하거나 하지 않을 수 있으며, 아미노산의 변형을 수반하는 경우에는 이러한 변형이 유발된 유전자는 이에 의해 코딩되는 단백질에서 하나 이상의 아미노산이 치환, 결실, 부가 및/또는 삽입된 아미노산 서열로 이루어지는 단백질을 암호화하는 것으로, 돌연변이체(mutants), 유도체(derivatives), 대립유전자(alleles), 변형체(variants) 및 동질체 (homologues)를 포함하는 것이다. 유전자 염기서열의 변이가 단백질 중의 아미노산의 변형을 수반하지 않는 경우는 예를 들어 축중변이가 있고, 이러한 축중변이체(degeneracy mutants)도 본 발명의 상기 유전자에 포함된다.
인위적 유전자 염기서열의 변형은 당업자에게 잘 알려져 있는 방법, 예를 들어, 부위-유도성 돌연변이법 (site-directed mutagenesis, Kramer et al, 1987), 에러유발 PCR (Cadwell, R.C. and G.F. Joyce. 1992. PCR methods Appl., 2:28-33.), 점돌연변이법 (Sambrook and Russel, Molecular Cloning: A Laboratory Manual, 3rd Ed. 2001, Cold Spring Harbor Laboratory Press.) 등에 의해 제조될 수 있다.
본 발명에서, 상기 벡터는 숙주 세포에서 목적 유전자를 발현시키기 위한 수단을 의미한다. 상기 벡터는 목적 유전자 발현을 위한 요소(elements)를 포함하는 것으로, 복제원점(replication origin), 프로모터, 작동 유전자(operator), 전사 종결 서열(terminator) 등을 포함할 수 있고, 숙주 세포의 게놈 내로의 도입을 위한 적절한 효소 부위(예컨대, 제한 효소 부위) 및/또는 숙주 세포 내로의 성공적인 도입을 확인하기 위한 선별 마커 및/또는 단백질로의 번역을 위한 리보좀 결합 부위(ribosome binding site; RBS), IRES(Internal Ribosome Entry Site) 등을 추가로 포함할 수 있다. 상기 벡터는 상기 프로모터 이외의 전사 조절 서열(예를 들어, 인핸서 등)을 추가로 포함할 수 있다.
또한, 상기 벡터는 당업계에 공지된 플라스미드 DNA, 재조합 벡터 또는 기타 매개체일 수 있고, 구체적으로 선형 DNA 플라스미드 DNA, 재조합 비바이러스성 벡터, 재조합 바이러스성 벡터 또는 유전자 발현 유도 벡터 시스템(inducible gene expression vector system)일 수 있으며, 상기 재조합 바이러스성 벡터는 레트로바이러스, 아데노바이러스, 아데노 부속 바이러스, 헬퍼-의존형 아데노바이러스, 헤르페스 심플렉스 바이러스, 렌티바이러스 벡터 또는 백시니아바이러스일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서, 상기 용어 "형질전환"은 외부로부터 주어진 DNA에 의하여 생물의 유전적인 성질이 변하는 것으로, 즉 생물의 어떤 계통의 세포에서 추출된 핵산의 일종인 DNA를 다른 계통의 살아있는 세포에 도입했을 때 DNA가 그 세포에 들어가서 유전형질이 변화하는 현상을 의미한다.
본 발명에서, 상기 GPCR19 및 P2Xn 수용체를 코딩하는 유전자는 전술한 바와 같이 공지된 서열로부터 이를 특이적으로 인식할 수 있는 프라이머를 제작하고, 이를 이용하여 중합효소연쇄증폭반응(Polymerase Chain Reaction)을 통해 상기 유전자를 증폭하고 이를 상술한 바와 같은 발현 벡터에 도입한 후 세포에 도입할 수 있다. 도입 방법은 공지되어 있으며 예를 들면 리포좀 매개 전달이입, 칼슘포스페이트법, DEAE-덱스트란 매개 전달이입, 양전하 지질 매개 전달이입법, 전기천공, 파아지 시스템을 이용한 트랜스덕션 또는 바이러스를 이용한 감염 방법 등을 포함하나 이에 제한되는 것은 아니다.

본 발명에 따른 방법에 있어서, 상기 후보물질은 통상적인 선정 방식에 따라 GPCR19 및 P2Xn 수용체 복합체의 상호작용의 조절 가능성을 지닌 것으로 추정되거나 또는 무작위적으로 선정된 개별적인 펩티드, 앱타머, 항체, 단백질, 비펩티드성 화합물, 활성 화합물, 발효 생산물, 세포 추출액, 식물 추출액 또는 동물조직 추출액일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 따른 방법에 있어서, 상기 제1물질은 염증 유도제(inflammatory inducer) 또는 NLRP3 인플라마좀(inflammasome) activator 일 수 있다.
또한, 상기 염증 유도제는 TLR(Toll-like receptor) ligand 또는 cytokine일 수 있고, 예컨대 LPS(lipopolysaccharide), peptidoglycan, TNF-α, IL-1β 또는 IL-17일 수 있으나, 이에 제한되는 것은 아니며, NF-κB signal transduction pathway를 통해 염증을 유도하는 것이면 제한없이 이용할 수 있다.
또한, 상기 NLRP3 인플라마좀 activator는 아밀로이드-베타(amyloid-β, Aβ)일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 따른 방법에 있어서, 상기 제2물질은 P2Xn 수용체 agonist일 수 있고, 예컨대 ATP, BzATP 또는 nigericin일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 따른 방법에 있어서, 상기 P2Xn 수용체는 P2X1, P2X2, P2X3, P2X4, P2X5, P2X6 또는 P2X7 수용체일 수 있고, 구체적으로 P2X7 수용체일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 따른 방법에 있어서, 상기 세포는 줄기세포, 동물세포, 곤충세포 또는 식물세포일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 줄기세포는 구체적으로 배아줄기세포, 성체줄기세포, 유도만능줄기세포(induced pluripotent stem cell, iPS)일 수 있고, 보다 구체적으로 성체줄기세포(중간엽줄기세포)일 수 있으나, 이에 제한하는 것은 아니다. 상기 성체줄기세포는, 골수, 혈액, 뇌, 피부, 지방, 골격근, 탯줄, 제대혈 등의 다양한 성체 세포로부터 유래할 수 있다. 구체예로, 중간엽줄기세포(Mesenchymal Stem Cell; MSC), 골격근줄기세포(skeletal muscle stem cell), 조혈줄기세포(hematopoietic stem cell), 신경줄기세포(neural stem cell), 간줄기세포(hepatic stem cell), 지방유래줄기세포(adipose-derived stem cell), 지방유래전구세포(adipose-derived progenitor cell), 혈관내피전구세포(vascular endothelial progenitor cell) 등을 들 수 있으나, 이에 제한되는 것은 아니다.

또한, 상기 세포는 GPCR19 및 P2Xn 수용체를 heterologously 또는 endogenously 발현할 수 있고, 상기 heterologously 발현을 위해 상기에서 전술한 바와 같이 세포에 도입할 수 있다.
본 발명에 따른 방법에 있어서, 상기 단계 3)에서 GPCR19 및 P2Xn 수용체 복합체의 상호작용은 다음 중 어느 하나 이상의 특성을 분석하여 측정할 수 있다:
i) GPCR19 및 P2Xn 수용체의 세포 표면, 세포질 또는 세포핵 내 colocalization 변화;
ii) GPCR19-mediated signal transduction pathway 활성 변화;
iii) P2Xn 수용체-mediated signal transduction pathway 활성 변화; 및
iv) 염증성 사이토카인 수준 변화.
구체적으로, 상기 GPCR19-mediated signal transduction pathway 활성 변화는 cAMP 수준 또는 PKA 활성 변화일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 P2Xn 수용체-mediated signal transduction pathway 활성 변화는 Ca++ mobilization 또는 inflammasomal activation 변화일 수 있고, 상기 inflammasomal activation 변화는 NLRP3, ASC, Pro-IL-1β, IL-1β, Pro-IL-18, IL-18, Pro-Caspase-1, Caspase-1 또는 gasdermin D 수준 변화, NLRP3 inflammasome oligomerization 변화, 또는 IL-1β, IL-18 또는 Caspase-1 immature form의 mature form으로의 성숙 변화일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 염증성 사이토카인 수준 변화는 TNF-α, IL-1β, IL-18, RANTES 또는 MCP-1 수준 변화일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에 따른 방법에 있어서, 상기 특성은 예를 들어 칼슘이온측정법, 면역형광법, 면역침강법, 단백질 칩 분석, 웨스턴 블럿팅(Western blotting), 효소면역측정법(ELISA), RT-PCR(Reverse transcription polymerase chain reaction), 실시간 RT-PCR, 노던 블럿팅(Northern blotting), DNA 칩 분석, 리간드 바인딩 분석법(ligand binding assay), 방사선 면역분석법, 조직면역염색법 또는 면역측정법 등의 방법으로 측정할 수 있으나, 이에 제한되는 것은 아니며, 상기 특성을 분석하기 위해 당업계에 알려진 방법은 제한없이 사용할 수 있다.
또한, 본 발명은
1) GPCR19 및 P2Xn 수용체를 발현하는 세포에 후보물질 및 제1물질을 처리하는 단계;
2) 상기 단계 1)의 세포에 제2물질을 처리하는 단계;
3) 상기 단계 2)의 세포에서 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 측정하는 단계; 및
4) 상기 단계 3)의 상호작용 측정 결과, 후보물질 무처리 대조군과 비교하여 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 증가시키는 후보물질을 NLRP3 인플라마좀-관련 질환(NLRP3 inflammasome-associated disease) 예방 또는 치료를 위한 물질로 판단하는 단계를 포함하는,
NLRP3 인플라마좀-관련 질환 예방 또는 치료를 위한 물질을 스크리닝하는 방법을 제공한다.
본 발명에서, 상기 후보물질, 제1물질, 제2물질, 세포, 상호작용 측정 방법 등에 대해서는 상기 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 조절하는 물질을 스크리닝하는 방법에 대한 설명과 동일한 바, 구체적은 설명은 상기 내용을 원용하고, 이하에서는 NLRP3 인플라마좀-관련 질환 예방 또는 치료를 위한 물질을 스크리닝하는 방법의 특유한 구성에 대해서만 설명하도록 한다.
본 발명에 따른 방법에 있어서, 상기 단계 4)에서 후보물질 무처리 대조군과 비교하여 다음 중 어느 하나 이상의 특성이 나타나면 GPCR19 및 P2Xn 수용체 복합체의 상호작용이 증가된 것으로 판단할 수 있다:
i) GPCR19 및 P2Xn 수용체의 세포 표면, 세포질 또는 세포핵 내 colocalization 증가를 나타내는 경우;
ii) GPCR19-mediated signal transduction pathway 활성 증가를 나타내는 경우, 보다 구체적으로 cAMP 수준 또는 PKA 활성 증가를 나타내는 경우;
iii) P2Xn 수용체-mediated signal transduction pathway 활성 억제를 나타내는 경우, 보다 구체적으로 Ca++ mobilization 또는 inflammasomal activation 감소를 나타내는 경우, 보다 더 구체적으로 Ca++ mobilization 감소 또는 NLRP3, ASC, IL-1β, IL-18, Caspase-1 또는 gasdermin D 수준 감소를 나타내는 경우, NLRP3 inflammasome oligomerization 감소를 나타내는 경우, 또는 IL-1β, IL-18 또는 Caspase-1 immature form의 mature form으로의 성숙 감소를 나타내는 경우; 및
iv) 염증성 사이토카인 수준 감소를 나타내는 경우, 보다 구체적으로 TNF-α, IL-1β, IL-18, RANTES 또는 MCP-1 수준 감소를 나타내는 경우.
본 발명에 따른 방법에 있어서, 상기 NLRP3 인플라마좀-관련 질환은 염증성 질환, 퇴행성 질환, 대사성 질환, 신경계 질환 또는 암일 수 있고, 보다 구체적으로 암, 낭창, 통풍, 패혈증, 류마티스 관절염, 골관절염, 소아 특발성 관절염, 허혈성 망막변증, 나이관련황반변성, 이식 만성거부반응, 건선, 건선성 관절염, 죽상경화증, 심방세동(Atrial fibrillation), 재협착증, 비만, 폐 고혈압, 만성 호흡기 질환, 뇌경색, 협심증, 관상 동맥 질환, 고혈압, 뇌졸중, 빈혈, 편두통, 신경통증, 부정맥, 혈관종, 고지혈증, 말초 혈관 질환, 혈관 기형, 치매, 염증성 장 질환, 골다공증, 골흡수, 궤양성 대장염, 호흡 곤란 증후군, 당뇨병, 비알콜성 지방간염(NASH), 아토피성 피부염, 광선 각화증, 피부 지연형 과민증 장애, 알츠하이머병, 파킨슨병 다발성 경화증, 다발성 골수종, 천식, 비염, 간염, 각막염, 위염, 장염, 신장염, 기관지염, 흉막염, 복막염, 척추염, 췌장염, 염증성 통증, 요도염, 방광염, 화상 염증, 피부염, 치주염, 치은염, 표피용해성 각화증(epidermolytic ichthyosis), 퇴행성 신경병증, 만성 폐쇄성 폐질환, 폐섬유증, 크리요피린 관련 주기적 증후군(Cryopyrin-Associated Periodic Syndromes) 또는 내독소 유도성 질환일 수 있으나, 이에 제한되는 것은 아니다.
아울러, 본 발명은 약학적으로 유효한 양의 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질을 개체에 투여하는 단계를 포함하는, NLRP3 인플라마좀-관련 질환 예방 또는 치료 방법을 제공한다.
본 발명에서, 상기 P2Xn 수용체는 P2X1, P2X2, P2X3, P2X4, P2X5, P2X6 또는 P2X7 수용체일 수 있고, 구체적으로 P2X7 수용체일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서, 상기 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질은 GPCR19 및 P2Xn 수용체 간 상호 결합을 유도한다. 그 결과, GPCR19 및 P2Xn 수용체 간 복합체, 보다 구체적으로 heterooligomeric 복합체를 형성하여 서로 상호작용한다. 상기 GPCR19 및 P2Xn 수용체 복합체의 상호작용은 구체적으로 GPCR19가 활성화되어 P2Xn 수용체의 활성을 저해하는 것으로, 이로 인해 GPCR19-mediated signal transduction pathway는 활성화되고, P2Xn 수용체-mediated signal transduction pathway는 비활성화된다(도 6 참조).
이에 상기 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질을 개체에 투여하여 다음 중 어느 하나 이상의 특성을 나타낼 수 있다:
i) GPCR19 및 P2Xn 수용체의 세포 표면, 세포질 또는 세포핵 내 colocalization 증가;
ii) GPCR19-mediated signal transduction pathway 활성 증가, 보다 구체적으로 cAMP 수준 또는 PKA 활성 증가 ;
iii) P2Xn 수용체-mediated signal transduction pathway 활성 억제, 보다 구체적으로 Ca++ mobilization 또는 inflammasomal activation 감소, 보다 더 구체적으로 Ca++ mobilization 감소, NLRP3, ASC, IL-1β, IL-18, Caspase-1 또는 gasdermin D 수준 감소, NLRP3 inflammasome oligomerization 감소, 또는 IL-1β, IL-18 또는 Caspase-1 immature form의 mature form으로의 성숙 감소; 및

본 발명에 따른 유효 성분, 구체적으로 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서, "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 배합된 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명에 따른 유효 성분은 필요에 따라 기타 성분을 포함할 수 있다. 기타 성분으로서는, 특별히 이에 제한되지 않지만, 안정제, 계면활성제, 가소제, 활택제, 가용화제, 완충제, 감미제, 기재, 흡착제, 교미제, 결합제, 현탁화제, 항산화제, 광택화제, 코팅제, 착향제, 향료, 습윤제, 습윤 조정제, 소포제, 저작제, 청량화제, 착색제, 당의제, 등장화제, pH 조절제, 연화제, 유화제, 점착제, 점착증강제, 점조제, 점조화제, 발포제, 부형제, 분산제, 분사제, 붕괴제, 붕괴보조제, 방향제, 방습제, 방부제, 보존제, 무통화제, 용제, 용해제, 용해보조제, 유동화제 등의 의약품 첨가제를 들 수 있다.
본 발명의 유효 성분은 개체에 투여할 수 있고, 상기 개체는 포유동물, 구체적으로 인간, 인간이 아닌 영장류와 같은 비인간 포유동물, 모델 시스템에 사용된 동물(예를 들어, 약제의 스크리닝, 특징화 및 평가에 사용되는 마우스 및 랫트) 및 그 밖의 포유동물, 예를 들어 토끼, 기니아피그, 햄스터, 개, 고양이, 침팬지, 고릴라, 원숭이와 같은 유인원류 동물일 수 있다.
본 발명의 유효 성분은 경구 또는 비경구로 투여할 수 있고, 비경구로 투여되는 경우, 경피, 정맥, 근육, 비강 및 직장으로 이루어진 군으로부터 선택되는 어느 하나 이상으로 투여될 수 있으나 이에 제한되는 것은 아니다.
본 발명에 따른 유효 성분의 투여 형태는, 제제화 방법, 투여 방식, 환자의 연령, 체중, 질환, 증상 및 그 정도 등에 따라 적절히 선택하면 되며, 특별히 제한되지 않는다. 예를 들면 정제(설하정, 구강내 붕괴정을 포함한다), 과립제, 산제, 액제, 시럽제(드라이시럽제를 포함한다.), 젤리제, 캡슐제(소프트 캡슐, 마이크로 캡슐을 포함한다.)등에 의한 경구 투여; 주사제(피하주사제, 정맥내 주사제, 근육내 주사제, 복강내 주사제 등), 질 정제, 질 연고/크림, 질 링, 질 겔 또는 기포제, 질 삽입제, 좌제(직장 좌제, 질 좌제를 포함한다.), 흡입제, 경피흡수제, 점안제, 점비제 등에 의한 비경구 투여를 들 수 있다.
본 발명에 따른 유효 성분의 투여량은, 환자의 연령, 성별, 체중, 질환, 증상 및 그 정도 등에 따라 적절히 결정된다.
본 발명에 따른 유효 성분의 제제화에 있어서, 당업계에서 공지된 첨가제를 배합할 수 있다. 예를 들면, 경구용 고형 제제를 조제하는 경우, 유효 성분에 부형제, 결합제, 붕괴제, 활택제, 착색제, 교미제, 교취제 등을 가한 후, 통상적인 방법에 따라 성형, 조립, 캡슐화 등 함으로써, 피복정제, 과립제, 산제, 캡슐제 등을 제조할 수 있다. 또한, 경구용 액상 제제를 조제하는 경우에는, 유효 성분에 정제수나 에탄올 등의 용제, 용해보조제, 현탁화제, 등장화제, 교미제, 완충제, 안정화제, 교취제 등을 가한 후, 통상적인 방법에 따라 조액 분포 등을 함으로써, 내복액제, 시럽제 등을 제조할 수 있다. 또한, 주사제를 조제하는 경우에는, 유효 성분에 pH 조정제, 완충제, 안정화제, 등장화제, 국소 마취제 등을 첨가한 후, 통상적인 방법에 따라 용기 내에 무균 봉입 등을 함으로써, 피하, 근육내, 정맥내용 주사제 등을 제조할 수 있다. 또한, 직장 좌제를 조제하는 경우에는, 유효 성분에 부형제, 계면활성제 등을 가한 후, 통상적인 방법에 따라 혼합 및 성형 등을 함으로써, 제제화할 수 있다.
본 발명에 따른 유효 성분의 투여용량은, 바람직한 효과를 위하여 본 발명의 유효 성분은 성인의 경우, 1일 1회 내지 수회 투여 시, 0.0001 내지 50 mg/kg 또는 0.001 내지 50 mg/kg의 용량으로 투여될 수 있다. 상기 투여용량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
이하, 본 발명을 실시예 및 실험예에 의하여 상세히 설명한다.
단, 하기 실시예 및 실험예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예 및 실험예에 의하여 한정되는 것은 아니다.
<실시예 1> GPCR19 및 P2X7 수용체의 colocalization 확인
다양한 세포에서 G-protein coupled receptor 19 (GPCR19)와 P2X7 수용체의 colocalization을 확인하였다.
<1-1> Keratinocyte에서 GPCR19 및 P2X7 수용체의 colocalization 확인
Keratinocyte에서 GPCR19와 P2X7 수용체의 colocalization을 확인하였다.
구체적으로, Keratinocyte로 HaCaT 세포 표면에서 GPCR19 및 P2X7 수용체의 colocalization을 DNCB(2,4-Dinitrochlorobenzene) + TNF-α ± 타우로데옥시콜린산나트륨(sodium taurodeoxycholate, 이하, 'HY209'라 명명함)로 자극한 후 공초점 현미경으로 분석하였다. 이때, DNCB는 NLRP3 inflammasome을 자극하기 위해 처리하였고, TNF-α는 염증 유도제(inflammatory inducer)로서 NF-κB signal을 활성화하여 염증 반응을 자극하기 위해 처리하였다. P2X7 (녹색 채널), GPCR19 (빨간색 채널) 및 colocalization 된 P2X7/GPCR19 (노란색 채널)의 MFI를 분석하였다. 각 실험 세트에서 4 ~ 5 ROI (x 600)의 MFI를 분석하였다. GPCR19 및 P2X7 수용체를 염색하기 위해, HaCaT 세포를 TNF-α (20 ng/ml) + DNCB (5 ㎍/ml) ± HY209 (400 ng/ml)로 4 시간 동안 처리하였다. 이후 HaCaT 세포를 커버글라스 (Deckglaser, Luda-Konlgshofen, Germany)에 분주하고 4% paraformaldehyde 로 10 분 동안 고정시키고 0.3% Triton X-100으로 10 분 동안 투과시켰다. 1 % BSA 및 10 % 정상 염소 혈청을 포함하는 PBS로 blocking한 후, 항-P2X7 폴리클로날 Ab (Alomone labs, Jerusalem, Israel) 또는 항-GPCR19 폴리클로날 Ab (R & D, Systems Minneapolis, MN, USA)로 30 분 동안 실온에서 염색한 후 세포를 Fluorochrome 표지된 항-HOST IgG로 염색하였다. 형광 항체 염색이 끝난 세포는 DAPI (Vector laboratories, Burlingam, CA, USA)를 포함하는 마운팅 배지로 마운팅하고, 공초점 형광 현미경 (Nikon, ECLIPSE Ti, New York, USA)을 사용하여 관찰하였다.
그 결과, 도 1A에 나타낸 바와 같이, 휴지기 상태에서 GPCR19와 P2X7 수용체가 HaCaT 세포 표면에서 함께 colocalization 되는 반면, TNF-α + DNCB 처리군의 경우 P2X7 발현이 유의하게 증가하고 GPCR19의 발현이 억제됨을 확인하였다. 그러나, TNF-α + DNCB와 함께 HY209를 처리한 군의 경우 GPCR19의 발현이 증가하고 P2X7의 발현이 감소하였으며, 휴지기 상태와 같이 GPCR19와 P2X7 수용체가 colocalization 됨을 확인하였다.
<1-2> Microglia에서 GPCR19 및 P2X7 수용체의 colocalization 확인
Microglia에서 GPCR19와 P2X7 수용체의 colocalization을 확인하였다.
구체적으로, Microglia로 BV2 세포 표면에서 GPCR19 및 P2X7 수용체의 colocalization을 Aβ± ATP ± HY209로 자극한 후 공초점 현미경으로 분석하였다. 이때, Aβ는 BV2 세포에서 NF-κB signal을 통한 염증반응을 자극하기 위해 처리하였고, ATP는 P2X7 수용체를 활성화하기 위해 처리하였다. GPCR19 및 P2X7 수용체를 염색하기 위해, BV2 세포에 Aβ (2 μM) ± HY209 (400 ng/ml)를 1 시간 동안 처리하였다. ATP (1 mM)는 샘플을 회수하기 전에 추가로 1 시간 동안 처리하였다. 처리 후 세포를 상기 실시예 <1-1>에 기재된 방법과 동일한 방법으로 세포 표면 GPCR19 은 항-GPCR19 항체와 Alex488-표지 2차 항체 (초록색)로 염색하고, P2X7R은 항-P2X7R 항체와 Alex446-표지 2차 항체 (빨간색)로 면역-염색한 다음 DAPI로 핵을 염색하고 마운팅 하였다. 이후 공초점 형광 현미경 (Nikon, ECLIPSE Ti, New York, USA)을 사용하여 관찰하였다.
그 결과, 도 1B에 나타낸 바와 같이, 휴지기 상태에서 GPCR19와 P2X7 수용체가 BV2 세포 표면에서 함께 colocalization 되는 반면, Aβ 단독 처리군 및 Aβ+ ATP 처리군의 경우 P2X7 발현이 유의하게 증가하고 GPCR19의 발현이 억제됨을 확인하였다. 그러나, Aβ+ HY209 처리군 및 Aβ+ ATP + HY209 처리군의 경우 GPCR19의 발현이 증가하고, P2X7의 발현이 감소하는 것을 확인하였다. 특히, Aβ+ ATP + HY209 처리군의 경우 휴지기 상태와 같이 GPCR19와 P2X7 수용체가 colocalization 됨을 확인하였다.

구체적으로, Macrophage로 BMDM(bone marrow-derived macrophage) 세포 표면 및 세포질에서 GPCR19 및 P2X7 수용체의 colocalization을 LPS + BzATP ± HY209로 자극한 후 공초점 현미경으로 분석하였다. 이때, LPS는 BMDM 세포에서 염증 유도제로서 NF-κB signal을 활성화하여 염증 반응을 자극하기 위해 처리하였고, BzATP는 P2X7 수용체를 활성화하기 위해 처리하였다. GPCR19 및 P2X7 수용체를 염색하기 위해, BMDM 세포에 LPS (10 ng/ml) ± HY209 (400 ng/ml)를 1 시간 동안 처리하였다. BzATP (300 μm)는 샘플을 회수하기 전에 추가로 1 시간 동안 처리하였다. 처리 후 세포를 상기 실시예 <1-1>에 기재된 방법과 동일한 방법으로 세포 표면 GPCR19 은 항-GPCR19 항체와 Alex488-표지 2차 항체 (초록색)로 염색하고, P2X7R은 항-P2X7R 항체와 Alex446-표지 2차 항체 (빨간색)로 면역-염색한 다음 DAPI로 핵을 염색하고 마운팅 하였다. 이후 공초점 형광 현미경 (Nikon, ECLIPSE Ti, New York, USA)을 사용하여 관찰하였다.
그 결과, 도 1C에 나타낸 바와 같이, 휴지기 상태에서 GPCR19와 P2X7 수용체가 BMDM 세포 표면 및 세포질에서 함께 colocalization 되는 반면, LPS + BzATP 처리군의 경우 P2X7 발현이 유의하게 증가하고 GPCR19의 발현이 억제됨을 확인하였다. 그러나, LPS + BzATP와 함께 HY209를 처리한 군의 경우 GPCR19의 발현이 증가하고 P2X7의 발현이 다소 감소하였으며, 휴지기 상태와 같이 GPCR19와 P2X7 수용체가 colocalization 됨을 확인하였다.
상기 결과를 통해 GPCR19 및 P2X7 수용체는 세포내-외부에서 상호결합 및 간섭을 한다는 것을 알 수 있다. 또한 염증 유도 상황에서는 GPCR19의 발현이 감소하고 P2X7의 발현히 증가하여 상호간의 결합 작용이 관측되지 않으나, HY209가 존재하면 GPCR19의 발현량과 P2X7 수용체 간 상호 결합을 증가시킬 수 있음을 알 수 있다.

구체적으로, 각각 3마리의 정상(wild-type, WT), GPCR19 KO 또는 P2X7 KO 마우스로부터 BMDM을 획득하였다. 그 다음, 획득한 BMDM에 ATP 20 uM을 처리하고 글라스 커버슬립(class coverslip)에 부착한 후, 세포 내 Ca++ 이동을 관찰하기 위해 NaCl 138 mM, KCl 5.6 mM, MgCl2 2 mM, HEPES 10 mM 및 pH 7.4 글루코스(glucose) 10 mM로 이루어진 생리적 외부 용액(physiological external solution) 에서 2 μM Fluo-4/AM으로 37℃ 배양기에서 30분간 배양하였다. 이후 개방형 관류 챔버 (open perfusion chamber)로 옮겨 잔여 Fluo-4/AM을 제거하고 여기(excitation) 494 ㎚ 및 방출(emission) 506 ㎚ 조건에서 형광 현미경(fluorescence microscope, Nikon, Tokyo, Japan)을 이용하여 형광준위를 측정하였다. 현미경에는 LED 램프(Andover, UK), 인터그레이티드 셔터(integrated shutter) 및 냉각 EM-CCD 카메라(cooled EM-CCD camera)가 장착되어 있으며, 셔터 및 카메라는 메타모프 소프트웨어(MetaMorph software, Molecular Devices, US)를 이용하여 제어되었다. 또한, 단일세포를 관심영역(region of interest, ROI)으로 설정하였고, 1×1의 비닝을 가지는 16 비트 그레이 스케일 이미지(16-bit grayscale image)를 노출 시간 1초로 설정하여 촬영하였다.
형광 현미경으로 측정한 형광신호 강도를 그래프화 하였다. 결과에서, 그래프의 각 막대는 SEM을 나타내고 굵은 선은 평균 강도를 나타낸다(도 2A, 왼쪽 패널). 세 번의 독립적인 실험을 진행한 후, 측정한 데이터의 합을 표시하였다(도 2A, 오른쪽 패널).
그 결과, 도 2A에 나타낸 바와 같이, 정상(WT) 마우스의 BMDM을 ATP로 자극하자 세포 내 Ca++ 이동이 증가하는 것을 확인하였다. 이에 비하여 GPCR19 KO 및 P2X7 KO 마우스의 BMDM의 경우에는 ATP로 자극하여도 세포 내 Ca++ 이동이 크지 않은 것으로 나타났다. 또한, 정상 마우스와 GPCR19 KO 및 P2X7 KO 마우스 사이에 유의한 차이가 있는 것을 확인하였다.
<2-2> GPCR19 결핍 또는 P2X7 결핍된 Microglia에서 Ca
++
이동 확인
GPCR19 KO 마우스 또는 P2X7 KO 마우스의 Microglia에서 P2X7 수용체에 의한 Ca++ 이동을 확인하였다.
구체적으로, 각각 6마리의 WT (B6), GPCR19 KO 또는 P2X7 KO 마우스로부터 Microglia를 획득하였다. 그 다음, 획득한 Microglia에 ATP 40 μM(왼쪽) 또는 BzATP 40 μM(오른쪽)을 처리하고 글라스 커버슬립에 부착한 후, 세포 내 Ca++ 이동을 상기 실시예 <2-1>에 기재된 방법과 동일한 방법으로 Fura-2/AM을 이용하여 형광 현미경으로 측정하고, 그래프화하였다. 측정된 결과에서, 그래프의 각 막대는 SEM을 나타내고 굵은 선은 평균 강도를 나타낸다(1열 패널들). 세 번의 독립적인 실험을 진행한 후, 측정한 데이터의 합을 표시하였다(2열 패널들).
그 결과, 도 2B에 나타낸 바와 같이, 정상(WT) 마우스의 Microglia을 ATP 또는 BzATP로 자극하자 세포 내 Ca++ 이동이 증가하는 것을 확인하였다. 이에 비하여 GPCR19 KO 및 P2X7 KO 마우스의 BMDM의 경우에는 ATP로 자극하여도 세포 내 Ca++ 이동이 크지 않은 것으로 나타났다. 또한, 정상 마우스와 GPCR19 KO 및 P2X7 KO 마우스 사이에 유의한 차이가 있는 것을 확인하였다.
<2-3> Keratinocyte에서 염증 유도제 및 HY209 처리에 따른 Ca
++
이동 확인
상기 <실시예 1>을 통해 HY209가 GPCR19 및 P2X7 수용체 간 상호 결합 유도제임을 확인하였다. 이에 염증 유도제로 염증성 사이토카인를 처리하고, HY209를 처리한 Keratinocyte에서 P2X7 수용체에 의한 Ca++ 이동을 확인하였다.
구체적으로, HaCaT 세포에 IL-1β 10 ng/㎖ 및 TNF-α 10 ng/ml를 처리하고 4 시간 뒤 HY209를 200, 400 및 800 ng/ml로 각각 처리하였다. HY209 200 ng/㎖ 처리 후부터 Ca++ 이동 측정을 시작하였고, 측정 시작 후 50초에 BzATP 또는 ATP 20 μM를 처리하였다. 세포 내 Ca++ 이동은 상기 실시예 <2-1>에 기재된 방법과 동일한 방법으로 Fura-2/AM을 이용하여 형광 현미경으로 측정하고, 그래프화하였다(도 2C, 1열 및 2열 왼쪽 패널). 세 번의 독립적인 실험을 진행한 후, 측정한 데이터의 합을 표시하였다(도 2C, 1열 및 2열 오른쪽 패널).
그 결과, 도 2C에 나타낸 바와 같이, 염증성 사이토카인 IL-1β및 TNF-α가 HaCaT 세포의 P2X7 수용체에 의한 Ca++ 이동을 증가시키는 반면, HY209를 함께 처리한 경우 HY209 농도가 증가함에 따라 IL-1β및 TNF-α의 영향으로 증가한 Ca++ 이동이 현저히 감소하는 것을 확인하였다.
<2-4> Macrophage에서 염증 유도제 및 HY209 처리에 따른 Ca
++
이동 확인
염증 유도제로 LPS(lipopolysaccharide)를 처리하고, HY209를 처리한 Macrophage에서 P2X7 수용체에 의한 Ca++ 이동을 확인하였다.
구체적으로, BMDM 세포에 LPS 100 ng/ml ± HY209 400 ng/ml를 1 시간 동안 처리하였다. 처리 후부터 Ca++ 이동 측정을 시작하였고, 측정 시작 후 50초에 2 mM CaCl2 및 0.5 mM MgCl2 존재 하에서 ATP 20 um 또는 BzATP 40 um을 처리하였다. 세포 내 Ca++ 이동은 상기 실시예 <2-1>에 기재된 방법과 동일한 방법으로 Fura-2/AM을 이용하여 형광 현미경으로 측정하고, 그래프화하였다(도 2D, 1열 및 2열 왼쪽 패널). 세 번의 독립적인 실험을 진행한 후, 측정한 데이터의 합을 표시하였다(도 2D, 1열 및 2열 오른쪽 패널 패널).
그 결과, 도 2D에 나타낸 바와 같이, LPS가 BMDM 세포의 P2X7 수용체에 의한 Ca++ 이동을 증가시키는 반면, HY209를 함께 처리한 경우 LPS의 영향으로 증가한 Ca++ 이동이 현저히 감소하는 것을 확인하였다.
<2-5> Microglia에서 NLRP3 inflammasome activator 및 HY209 처리에 따른 Ca
++
이동 확인
NLRP3 inflammasom activator로 Aβ를 처리하고, HY209를 처리한 Microglia에서 P2X7 수용체에 의한 Ca++ 이동을 확인하였다.
구체적으로, BV2 세포에 Aβ 2 μM ± HY209 400 ng/ml를 1 시간 동안 처리하였다. 처리 후부터 Ca++ 이동 측정을 시작하였고, 측정 시작 후 50초에 2 mM CaCl2 및 0.5 mM MgCl2 존재 하에서 ATP 20 um 또는 BzATP 40 um을 처리하였다. 세포 내 Ca++ 이동은 상기 실시예 <2-1>에 기재된 방법과 동일한 방법으로 Fura-2/AM을 이용하여 형광 현미경으로 측정하고, 그래프화하였다(도 2E, 1열 및 2열 왼쪽 패널). 세 번의 독립적인 실험을 진행한 후, 측정한 데이터의 합을 표시하였다(도 2E, 1열 및 2열 오른쪽 패널 패널).
그 결과, 도 2E에 나타낸 바와 같이, Aβ가 BMDM 세포의 P2X7 수용체에 의한 Ca++ 이동을 증가시키는 반면, HY209를 함께 처리한 경우 Aβ의 영향으로 증가한 Ca++ 이동이 현저히 감소하는 것을 확인하였다.
상기 결과들을 통해 우선 P2X7을 통한 칼슘이온 이동은 GPCR19 수용체에 의존적임을 확인하였다. 그리고 염증 유발 상황에서 GPCR19 수용체를 매개로 한 칼슘이온 이동을 관찰하고, HY209로 GPCR19과 P2X7 수용체 간 상호 결합을 유도하면 세포내 칼슘이온의 축적이 억제되고 염증성 인자들이 감소함을 알 수 있다.
<실시예 3> 염증 유도 세포에서 GPCR19 및 P2X7 수용체 간 상호 결합의 역할 확인
염증 유도된 세포에서 GPCR19 및 P2X7 수용체 간 상호 결합의 역할을 알아보기 위하여, DNCB/TNFα를 처리하고 HY209를 처리한 세포에서 inflammasomal 성분 및 IL-1β 발현 변화를 확인하였다.
구체적으로, HaCaT 세포에 TNF-α (20 ng/ml) ± HY209 (400 ng/ml)를 3 시간 동안 처리한 후 DNCB (5 μg/ml)를 추가로 24 시간 동안 처리하였다. 그 다음, 면역염색을 위해 세포를 커버 글라스 (Deckglaser, Luda-Konlgshofen, Germany)에 분주한 후, 4% 파라포름알데히드로 10 분 동안 고정시키고 0.3% Triton X-100으로 10 분 동안 투과시키고, 1% BSA 및 10 % 정상 염소 혈청을 포함하는 PBS로 1시간 blocking 하였다. 이후 inflammasomal 성분을 염색하기 위하여, 세포를 밤새 4℃에서 항-NLRP3 복합항체 (Abcam, Cambridge, UK) 및 항-ASC Ab (Clone B-3, Santa Cruz Biotechnology, Inc. Dallas, Texas, USA)로 염색한 다음 Alexa Flour 488-표지 또는 Alexa Flour 532-표지 이차 복합항체 (Invitrogen, Carlsbad, CA, USA)로 염색하였다. 슬라이드 상의 세포를 DAPI (Vector laboratories, Burlingam, CA, USA)가 포함된 장착 배지에 mounting 하고 공초점 형광 현미경 (Nikon, ECLIPSE Ti, 미국 뉴욕)을 사용하여 관찰하고, 이를 그래프화하였다(도 3A).
또한, HaCaT 세포에 DNCB (5 μg/ml) ± TNF-α (20 ng/ml) ± HY209 (400 ng/ml)를 24 시간 동안 처리한 후 ATP를 추가로 3 시간 동안 처리한 후 배양 상층액을 회수하였다. 그 다음, IL-1β ELISA kit (R&D Systems Minneapolis, MN, USA)를 이용하여 제조사의 절차에 따라 IL-1β 농도를 측정하였다(도 3B).
그 결과, 도 3A에 나타낸 바와 같이, TNF-α + DNCB 처리군의 경우 hNLRP3 및 hASC 발현, 및 hNLRP3-ASC oligomerization이 유의하게 증가하는 것을 확인하였다. 반면, TNF-α + DNCB와 함께 HY209를 처리한 군의 경우 hNLRP3 및 hASC 발현, 및 hNLRP3-ASC oligomerization의 증가가 억제되는 것을 확인하였다.
또한, 도 3B에 나타낸 바와 같이, TNF-α + DNCB 처리군의 경우 IL-1β의 생성이 유의하게 증가하는 반면, TNF-α + DNCB와 함께 HY209를 처리한 군의 경우 IL-1β의 생성 증가가 억제되는 것을 확인하였다. 또한, TNF-α + DNCB와 함께 ATP를 처리한 군의 경우 TNF-α + DNCB 처리군보다 IL-1β의 생성이 증가하였고, HY209에 의해 상기 IL-1β의 생성 증가가 억제되는 것을 확인하였다.
상기 결과를 통해 GPCR19 및 P2X7 수용체 간 상호 결합이 저해되면, GPCR19의 비활성화로 cAMP-매개 NF-κB pathway가, P2X7 수용체의 활성화로 NLRP3 inflammasomal activation pathway가 활성화되어 염증 반응이 촉진되는 것을 알 수 있다. 반면, GPCR19 및 P2X7 수용체 간 상호 결합을 유도하는 물질은 상기 GPCR19 및 P2X7 수용체 간 상호 결합 저해를 억제하여 상기 pathway를 비활성화함으로써, 염증 반응을 완화할 수 있음을 알 수 있다.
<실시예 4> 염증 유도 세포에서 HY209 처리에 따른 염증 반응 완화 확인
염증 유도된 세포에서 GPCR19 및 P2X7 수용체 간 상호 결합을 유도하는 물질이 GPCR19 및 P2X7 수용체 간 상호 결합 저해를 억제하여 염증 반응을 완화하는지 알아보기 위하여, LPS를 처리하고 HY209를 처리한 세포에 ATP를 추가로 처리한 후 inflammasomal 성분 및 IL-1β발현 변화를 확인하였다.
구체적으로, HY209의 농도별 처리에 따른 효과를 알아보기 위하여, BMDM 세포에 LPS (10 ng/ml) ± HY209 (0, 25, 100, 400 ng/ml)를 1 시간 동안 처리하였다. ATP (500 μm) 또는 BzATP (300 μm)는 샘플의 배양 상층액을 회수하기 전에 추가로 1 시간 동안 처리하였다. IL-1β ELISA kit (R & D Systems Minneapolis, MN, USA)를 이용하여 제조사의 절차에 따라 회수한 배양 상층액에서 IL-1β 농도를 측정하였다(도 4A).
또한, HY209의 전처리/후처리에 따른 효과를 알아보기 위하여, BMDM 세포에 LPS (10 ng/ml) 처리 1 시간 전 또는 처리 3 시간 후에 HY209 (400 ng/ml)를 처리하였다. 그 다음, BzATP (300 μm)를 추가로 1 시간 동안 처리한 후 샘플의 배양 상층액을 회수하였다. IL-1β ELISA kit (R&D Systems Minneapolis, MN, USA)를 이용하여 제조사의 절차에 따라 회수한 배양 상층액에서 IL-1β 농도를 측정하였다(도 4B).
또한, BMDM 세포에 LPS (10 ng/ml) ± HY209 (400 ng/ml)를 1 시간 동안 처리하고, BzATP (300 μm)를 추가로 1 시간 동안 처리한 후, 상기 <실시예 3>에 기재된 방법과 동일한 방법으로 inflammasomal 성분을 염색하고 공초점 형광 현미경 (Nikon, ECLIPSE Ti, US)을 사용하여 관찰하였다(도 4C).
그 결과, 도 4A에 나타낸 바와 같이, LPS + ATP와 함께 HY209를 처리한 군 및 LPS + BzATP와 함께 HY209를 처리한 군 모두 HY209 처리 농도 의존적으로 IL-1β의 생성 증가가 억제되는 것을 확인하였다.
또한, 도 4B에 나타낸 바와 같이, LPS + ATP와 함께 HY209를 전처리한 군 및 LPS + BzATP와 함께 HY209를 후처리한 군 모두 HY209에 의해 IL-1β의 생성 증가가 억제되는 것을 확인하였다.
아울러, 도 4C에 나타낸 바와 같이, LPS + BzATP 처리군의 경우 hNLRP3 및 hASC 발현, 및 hNLRP3-ASC oligomerization이 유의하게 증가하는 것을 확인하였다. 반면, LPS + BzATP와 함께 HY209를 처리한 군의 경우 hNLRP3 및 hASC 발현, 및 hNLRP3-ASC oligomerization의 증가가 억제되는 것을 확인하였다.
상기 결과를 통해 반면, GPCR19 및 P2X7 수용체 간 상호 결합을 유도하는 물질은 GPCR19 및 P2X7 수용체 간 상호 결합 저해를 억제하고, GPCR19 및 P2X7 수용체 간 상호 결합을 유도하여 cAMP-매개 NF-κB pathway 및 NLRP3 inflammasomal activation pathway를 비활성화함으로써, 염증 반응을 예방 및 완화할 수 있음을 알 수 있다.
<실시예 5> 유세포 계측 비드 분석법(cytometric bead array, CBA)을 이용한 사이토카인 측정
염증 유도된 세포에서 GPCR19 및 P2X7 수용체 간 상호 결합을 유도하는 물질과 기존에 알려진 염증 복합체 억제제 간 염증 반응 완화 효과를 비교 분석하였다.
구체적으로, 확인하고자 하는 5종의 염증성 사이토카인, TNF-α, RANTES, MCP-1, IL-1β및 IL-8에 대한 항체가 결합된 형광 비드(bead)를 섞어 칵테일 비드를 만들었다. 그다음, 제조한 칵테일 비드 50 ㎕, 준비된 시료, 및 표준 시료를 상온의 어두운 환경에서 1시간 동안 반응시켰다. 이때, 시료는 GPCR19 및 P2X7 수용체 간 상호 결합을 유도하는 물질로 상기 실시예에서 확인한 HY209, GPCR19 agonist로 INT777(Cat. No. HY-15677, MedChemExpress), PDE4 inhibitor로 crisaborole(Eucrisa, Pfizer), NLRP3 inhibitor로 MCC950(CAS No. 256373-96-3, Calbiochem), P2X7 antagonist로 A740003(CAS No. 861393-28-4, Sigma-Aldrich) 및 GW791343 (CAS No. 309712-55-8, Tocris), JAK inhibitor로 Tofacitinib (CAS No. 540737-29-9, Sigma-Aldrich)를 각 1 μM씩 사용하였고, 양성대조군으로 corticosteroid인 Prednisolone을 1 μM 사용하였다. 그리고 피코에리트린 검출 시약(phycoerythrin(PE) detection reagent)이 1 ㎕/시료(sample)가 되도록 필요한 양을 새로운 튜브(tube)로 옮기고, 세척 완충액을 이용하여 워싱한 뒤 각 시료 당 50 ㎕가 들어갈 수 있는 부피를 계산하여 캡쳐 비드 희석제(capture bead diluent)를 넣어주고, 상온의 어두운 환경에서 15분 동안 반응시켰다. 그다음, 준비된 PE 검출 시약을 칵테일 비즈, 준비된 시료 및 표준 시료가 반응하고 있는 튜브에 50 ㎕ 넣어주고, 잘 섞은 후 2시간 더 반응시켰다. 이후 모든 반응이 완료되면 1 ㎖의 세척 완충액을 넣어주고 200 g으로 5분 동안 원심분리한 뒤 상층액을 제거하고 300 ㎕의 세척 완충액을 넣은 후 유세포 분석 분리기(CANTO II FACS, BD Biosciences, US)를 이용하여 5종의 염증성 사이토카인에 대한 정량을 측정하였다.
그 결과, 도 5에 나타낸 바와 같이, inflammasome 자극 signal에 의해 생성되는 5종의 염증성 사이토카인에 대해 HY207의 억제 효과가 가장 우수한 것을 확인하였다.
<실시예 6> Potassium ion channel assay 분석법을 이용한 ion channel의 조절 확인
염증 유도된 세포에서 GPCR19 agonist인 HY209에 의해서 ion channel의 활성도 변화를 비교 분석하였다.
구체적으로, GPCR19의 조절에 의해서 ion channel의 활성이 변화하는 지 확인하기 위해서 U937세포를 이용하였다. U937세포에 PMA 25nM을 처리하여 분화를 시켰다. 이후에 형광 probe를 넣어 1시간 반응시켰다. 이후에 BzATP 600uM를 처리하여 ion channel이 활성화되는 것을 ELISA기기를 이용하여 누적 형광값을 측정하였다. 상기 실시예에서 확인한 결과 BzATP 600uM를 PMA에 의해서 분화된 U937세포에 처리하면 ion channel이 활성화되는 것을 누적 형광의 증가로 확인하였다. 이때, HY209 1uM를 1시간 전에 처리한 후, BzATP를 처리하였을 때, BzATP에 의해서 증가된 누적 형광값이 100% 감소되는 확인하였다.
이상의 결과를 통해서 HY209가 GPCR19을 조절하여 염증 반응에서 중요한 ion channel인 P2X7을 효과적으로 조절하는 것을 확인하였다.
상기 <실시예 1> 내지 <실시예 6>의 결과를 통해 도 7의 모식도와 같이 미생물로 인한 PAMP, ATP와 같은 생체 물질로 인한 DAMP 스트레스 염증 유발 상황에서 P2X7을 매개로 하는 NLRP3 inflammasomal activation pathway가 GPCR19에 의해서 조절되는 생리기전을 확인하였다. 또한, GPCR19와 P2X7 수용체 간 상호 결합을 유도하는 물질을 이용하여 염증반응 중에 GPCR19와 P2X7 수용체 간 상호 결합을 유도하면 P2X7으로부터 시작되는 염증 반응을 예방 또는 완화할 수 있음을 확인하였다. 아울러, 종래 염증 복합제 억제제 및 GPCR19 작용제와 달리 HY209는 GPCR19와 P2X7 수용체 간 상호 결합을 유도하여 염증 반응 예방 또는 완화에 있어 보다 우수한 효과를 나타냄을 확인하였다.
GPCR19 및 P2Xn 수용체 복합체와 그 용도에 따라, 물질 스크리닝, NLRP3 인플라좀-관련 질환 예방 또는 치료가 가능하여, 의약, 제약 분야 등에서 유용하게 이용될 수 있다.

ii) GPCR19-mediated signal transduction pathway 활성 변화;
iii) P2Xn 수용체-mediated signal transduction pathway 활성 변화; 및
iv) 염증성 사이토카인 수준 변화.
제13항에 있어서,
상기 GPCR19-mediated signal transduction pathway 활성 변화는 cAMP 수준 또는 PKA 활성 변화인 것인, 방법.
제13항에 있어서,
상기 P2Xn 수용체-mediated signal transduction pathway 활성 변화는 Ca++ mobilization 또는 inflammasomal activation 변화인 것인, 방법.
제15항에 있어서,
상기 inflammasomal activation 변화는 NLRP3, ASC, Pro-IL-1β, IL-1β, Pro-IL-18, IL-18, Pro-Caspase-1, Caspase-1 또는 gasdermin D 수준 변화, NLRP3 inflammasome oligomerization 변화, 또는 IL-1β, IL-18 또는 Caspase-1 immature form의 mature form으로의 성숙 변화인 것인, 방법.
제13항에 있어서,
상기 염증성 사이토카인은 TNF-α, IL-1β, IL-18, RANTES 및 MCP-1로 이루어진 군으로부터 선택되는 것인, 방법.
1) GPCR19 및 P2Xn 수용체를 발현하는 세포에 후보물질 및 제1물질을 처리하는 단계;
2) 상기 단계 1)의 세포에 제2물질을 처리하는 단계;
3) 상기 단계 2)의 세포에서 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 측정하는 단계; 및
4) 상기 단계 3)의 상호작용 측정 결과, 후보물질 무처리 대조군과 비교하여 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 증가시키는 후보물질을 NLRP3 인플라마좀-관련 질환(NLRP3 inflammasome-associated disease) 예방 또는 치료를 위한 물질로 판단하는 단계를 포함하는,
NLRP3 인플라마좀-관련 질환 예방 또는 치료를 위한 물질을 스크리닝하는 방법.
제18항에 있어서,
상기 제1물질은 염증 유도제 또는 NLRP3 인플라마좀 activator 인 것인, 방법.
제18항에 있어서,
상기 제2물질은 P2Xn 수용체 agonist인, 방법.
제18항에 있어서,
상기 단계 4)에서 후보물질 무처리 대조군과 비교하여 다음 중 어느 하나 이상의 특성이 나타나면 GPCR19 및 P2Xn 수용체 복합체의 상호작용이 증가된 것으로 판단하는 것인, 방법:
i) GPCR19 및 P2Xn 수용체의 세포 표면, 세포질 또는 세포핵 내 colocalization 증가;
ii) GPCR19-mediated signal transduction pathway 활성 증가;
iii) P2Xn 수용체-mediated signal transduction pathway 활성 억제; 및
iv) 염증성 사이토카인 수준 감소.
제21항에 있어서,
상기 ii) 특성은 cAMP 수준 또는 PKA 활성 증가를 나타내는 경우인, 방법.
제21항에 있어서,
상기 iii) 특성은 Ca++ mobilization 또는 inflammasomal activation 감소를 나타내는 경우인, 방법.
제23항에 있어서,
상기 inflammasomal activation 감소를 나타내는 경우는 NLRP3, ASC, IL-1β, IL-18, Caspase-1 또는 gasdermin D 수준 감소를 나타내는 경우, NLRP3 inflammasome oligomerization 감소를 나타내는 경우, 또는 IL-1β, IL-18 또는 Caspase-1 immature form의 mature form으로의 성숙 감소를 나타내는 경우인, 방법.

상기 NLRP3 인플라마좀-관련 질환은 암, 낭창, 통풍, 패혈증, 류마티스 관절염, 골관절염, 소아 특발성 관절염, 허혈성 망막변증, 나이관련황반변성, 이식 만성거부반응, 건선, 건선성 관절염, 죽상경화증, 심방세동(Atrial fibrillation), 재협착증, 비만, 폐 고혈압, 만성 호흡기 질환, 뇌경색, 협심증, 관상 동맥 질환, 고혈압, 뇌졸중, 빈혈, 편두통, 신경통증, 부정맥, 혈관종, 고지혈증, 말초 혈관 질환, 혈관 기형, 치매, 염증성 장 질환, 골다공증, 골흡수, 궤양성 대장염, 호흡 곤란 증후군, 당뇨병, 비알콜성 지방간염(NASH), 아토피성 피부염, 광선 각화증, 피부 지연형 과민증 장애, 알츠하이머병, 파킨슨병 다발성 경화증, 다발성 골수종, 천식, 비염, 간염, 각막염, 위염, 장염, 신장염, 기관지염, 흉막염, 복막염, 척추염, 췌장염, 염증성 통증, 요도염, 방광염, 화상 염증, 피부염, 치주염, 치은염, 표피용해성 각화증(epidermolytic ichthyosis), 퇴행성 신경병증, 만성 폐쇄성 폐질환, 폐섬유증, 크리요피린 관련 주기적 증후군(Cryopyrin-Associated Periodic Syndromes) 및 내독소 유도성 질환으로 이루어진 군으로부터 선택되는 것인, 방법.
NLRP3 인플라마좀-관련 질환 예방 또는 치료를 위한 물질을 스크리닝하기 위한, 분리된 GPCR19 및 P2Xn 수용체 복합체.
약학적으로 유효한 양의 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질을 개체에 투여하는 단계를 포함하는, NLRP3 인플라마좀-관련 질환 예방 또는 치료 방법.
제29항에 있어서,
상기 GPCR19 및 P2Xn 수용체 복합체의 상호작용을 유도하는 물질은 개체에 투여하여 다음 중 어느 하나 이상의 특성을 나타내는 것인, 방법:
i) GPCR19 및 P2Xn 수용체의 세포 표면, 세포질 또는 세포핵 내 colocalization 증가;
ii) GPCR19-mediated signal transduction pathway 활성 증가;
iii) P2Xn 수용체-mediated signal transduction pathway 활성 억제; 및
iv) 염증성 사이토카인 수준 감소.
