본 발명은 해양생태 교란종 및 연중 생산가능하고 경제적이며 친환경적인 해조류에서 폴리데옥시리보뉴클레오타이드를 (PDRN)을 추출하고 피부 및 조직 침투율이 뛰어난 50kDa이하의 크기로 저분자 공정을 거쳐 최종적으로는 저분자의 PDRN을 제조하는 방법이다. 보다 상세하게는 인체에 유해한 페놀과 같은 물질을 사용하지 않고 식물성 친환경 및 안전한 물질을 이용하여 유기용매의 사용 및 공정과정을 최소화하여 경제적이고 안전하게 친환경 해조류 유래 저분자 PDRN 및 추출방법에 관한 것이다.
PDRN(Polydeoxyribonucleotide) 및 폴리뉴클레오타이드(polynucleotide; PN)는 고분자물의 특성인 점탄성을 가지는 동시에, 피내 주입시 조직수복 효과를 나타낸다. 즉, 피부조직을 구성하고 있는 fibroblast를 증식시키고 활성을 높여 collagen 같은 extracellular matrix 분비를 촉진함으로써 단순한 피부 충전제가 아닌 피부조직의 재생을 유도할 수 있는 새로운 개념의 조직수복의료기기의 주성분이 된다.

PDRN 및 PN은 인산 및 네 가지 염기 아데닌, 구아닌, 티민, 사이토신을 포함한 것으로 4종의 염기와 데옥시리보오스 그리고 인산이 결합된 구조가 반복단위를 구성하여 고분자량의 이중가닥 중합체를 형성한 것이며, 추가적으로 아데닌(A), 구아닌(G), 시토신(C), 우라실(U)의 4종의 염기와 당과 인산이 반복단위가 되어 단일 가닥의 중합체를 형성한 유전 물질(이하, 유전자로 약칭함)이다.
이들 DNA 또는 RNA와 같은 유전자 물질은 가수분해 될 수 있으며 모든 생명체에 단백질과 결합하거나 또는 유리 상태로 분포하고 있다. 유리 상태의 단일가닥 RNA는 상보성을 갖는 부분이 서로 수소 결합을 형성함으로써 헤어핀 구조를 이루면서 부분적으로 이중가닥을 형성하기도 한다.
동물의 정자나 난자 또는 식물의 생장점 부위는 다른 조직들에 비하여, PDRN 및 PN 함량이 다른 부위에 비하여 상대적으로 높으며 이러한 유전 물질은 저분자, 고분자, 추출방법에 따라 효율 및 효과가 나뉘어지며 최종 산물인 PDRN 및 PN은 안전성이 확보된 저분자공정화되어 아데노신 A2 Receptor(리셉터)와 복합체를 이루고 VEGF 분비를 촉진시켜 혈관생성을 유도하고, 섬유 아세포, 골아세포, 연골 세포들을 활성화시킨다는 사실이 알려져 있다.
국내 공개특허번호 제10-2019-0139633 및 국내 등록특허번호 제10-2031152호에는 피디알엔(PDRN)을 추출하는 방법에 관하여 게시하고 있으나 상기 선행문헌은 동물 유래의 피디알엔을 추출하는 방법으로 본원발명과는 차이가 있다.
현재 동물유래 PDRN의 경우 동물 조직 유래 바이러스 혼입 가능성 및 지속적인 원료 확보가 어려운 부분이 있다. 이에 해양생태 교란종 및 연중 생산가능하고 경제적이고 친환경적인 해조류를 이용함으로써 상기부분을 해결함으로써 안전성, 안정성이 모두 확보가 가능한 기능성 저분자 PDRN을 생산하고자 한다.
본 발명은 김(포괄적으로 해조류)에서 피디알엔(PDRN)을 추출하고 저분자공정 방법에 관한 것으로, 상세하게는 김(포괄적으로 해조류) 또는 해양생태교란종(괭생이모자반 등)로부터 페놀과 같은 유독성 물질을 사용하지 않고 PDRN을 최소공정으로 추출하여 경제적으로 비용절감의 피디알엔(PDRN)을 추출하는 방법을 제공하고자 한다.
또한, 친환경적인 방법과 해조류라는 친환경원료에서 세포 활성 효과가 있는 PDRN(폴리데옥시리보뉴클레오타이드) 또는 PN(폴리뉴클레오타이드)와 같은 유전물질을 추출하고 정제하는 방법을 제공한다. 특히, 본 발명은 김(포괄적으로 해조류)의 특정조직이 아닌 모든 조직에서 PDRN 및 PN과 같은 유전물질을 분리하고, 확보된 해조류 유래 유전자 혼합물로부터 아데노신 A2 수용체를 활성화시키는 저분자의 PDRN, PN 혼합물을 제조하는 방법을 제공한다.
기존 연어, 송어 등(동물성원료)에서의 바이러스 혼입 위험성 및 원료수급의 제한성 등을 해결하고자 원활한 원료수급이 가능한 매생이, 모자반, 다시마, 미역 등 포괄적으로는 해조류 및 해양생태교란종의 조직으로부터 콩, 율무, 녹차(green tea) 추출물, 콩(soybean) 지방산, 토코페롤 cocamidopropyl betain, Olive oil carboxylate 등을 첨가하여 50kDa-20000kDa 사이의 분자량을 갖는 유전물질 및 유전물질 추출 방법을 제공한다.
상기 언급한 바와 같이 동물유래가 아닌 바이러스 감염 위험에 안전한 천연해조류의 생장점 포함된 조직으로부터 유래된 유전물질을 분리하는데 기존에 사용되는 페놀, 수산화나트륨 등의 유해물질을 사용하지 않는 동시에 제조공정 및 저분자 공정을 단순화하여 가격경쟁력이 확보된 PDRN 및 PN을 제공하고자 한다.
본 발명은 해조류로부터 정제 추출된 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드를 제공한다. 상기 해조류는 매생이, 모자반, 다시마, 미역, 김 중에서 선택되는 하나 이상의 것일 수 있다.
본 발명의 해조류는 해조류를 건조하고 분쇄한 해조류 원료에 콩 10중량%, 율무 10중량%, 녹차(green tea) 추출물 20중량%, 콩(soybean) 지방산20중량%, 토코페롤 cocamidopropyl betain 20중량%, Olive oil carboxylate 20중량%로 이루어진 단백질 용해 혼합용액을 제조, 첨가하여 단백질을 용해시킨 후, 리보뉴클레아제를 추가하여 폴리뉴클레오타이드 및 폴리데옥시리보뉴클레오타이드 혼합물을 분리 반응시켜 얻을 수 있다. 또한 본 발명에서 추출된 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드는 분자량 5kDa-20000kDa인 것을 특징으로 한다.
본 발명의 실시예에 따른 신혈관생성 및 세포재생 효과의 해조류 유래 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드 추출 방법은 해조류를 건조하고 친환경물질을 이용하여 단백질 제거 및 분해하는 원료확보 단계(가); 콩 추출물 10중량%, 율무 추출물 10중량%, 녹차(green tea) 추출물 20중량%, 콩(soybean) 지방산20중량%, 토코페롤 cocamidopropyl betain 20중량%, Olive oil carboxylate 20중량% 로 이루어진 혼합용액을 제조하고, 상기 (가)단계를 거쳐 건조 및 분쇄한 해조류 원료에 첨가하여 단백질을 용해시킨 후, 리보뉴클레아제를 추가하여 폴리뉴클레오타이드 및 폴리데옥시리보뉴클레오타이드 혼합물을 분리반응 시키는 단계를 포함하는 단백질 제거 및 분해단계(나); 상기 (나)단계를 거친 혼합물을 원심분리하여 불순물을 제거하고 필터링 후 건조하여 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 순수 분리하는 불순물 제거 및 순수분리단계 (다); 상기 (다)단계를 통해 순수 분리된 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 세척 및 건조하는 세척 및 건조단계(라); 상기 (라)단계로 건조된 결과물을 1% 나트륨 정제수 1ml를 첨가하여 희석 후 발효 아세트산을 이용하여 10분간 처리 후 소니케이션 처리를 10분간 진행하며, 원심분리를 실시하여 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확보하는 저분자 공정단계(마); 상기 (마)단계를 거친 결과물의 DNA흡광도를 측정하여 그 값이 1.7 내지 2.0인 순도 99%의 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확인하고 분말화하는 품질검사 단계(바)로 이루어진 것일 수 있다.
상기 (나)단계에서 제조한 혼합용액은 상기 (가)단계의 해조류 원료 중량 1g 대비 40ml씩 첨가하고, 리보뉴클레아제는 (가)단계의 해조류 원료 중량 1g대비 0.1ml씩 첨가하는 것일 수 있다. 또한, 상기 해조류는 매생이, 모자반, 다시마, 미역, 김 중에서 선택되는 하나 이상의 것일 수 있다.
본 발명은 원료수급이 원활한 친환경의 해조류 또는 김을 이용하고 공정과정에 유해물질이 아닌 친환경의 원료를 이용하기 때문에 안전성이 확보되며 추가적으로는 추출 및 저분자공정과정에서 물리적인 방법을 이용하므로 기존의 방법보다 경제적이고 효과적인 PDRN을 이용할 수 있다.

본 발명에서 지칭하는 용어, 폴리디옥시리보뉴클레오티드 (PDRN; Polydeoxyribonucleotide) 및 폴리뉴클레오타이드(PN;Polynucleotide)은 인산 및 네 가지 염기 아데닌, 구아닌, 티민, 사이토신을 포함한 것으로 4종의 염기와 데옥시리보오스 그리고 인산이 결합된 구조가 반복단위를 구성하여 고분자량의 이중가닥 중합체 또는 저분자량의 이중가닥 중합체를 형성한 것이며, 추가적으로 아데닌(A), 구아닌(G), 시토신(C), 우라실(U)의 4종의 염기와 당과 인산이 반복단위가 되어 단일 가닥의 중합체를 형성한 유전 물질(이하, 유전자로 약칭함)을 지칭한다.
또한, 본 발명에서 제조되는 저분자된 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드의 분자량은 5kDa-20000kDa인 것일 수 있으며 더 자세하게는 저분자된 폴리디옥시리보뉴클레오티드는 5kDa 이하 일 수 있다.
본 발명은 동물성원료가 아닌 원료수급 및 바이러스의 혼입 위험성 문제를 개선하기 위해 매생이, 모자반, 다시마, 미역, 김 등을 포함하는 해조류를 이용하여 비페놀사용 및 유기용매 사용을 최소화하고 기존의 원심분리법에서 진공추출방식을 사용함으로써 공정과정 및 비용을 최소화하였다. 추가적으로는 수산화나트륨, 염산 등의 독극물을 사용하지 않고 비독성물질 및 물리적인 공정을 통해 저분자 공정을 진행하였다.
이하, 본 발명의 신혈관생성 및 세포재생 효과의 해조류 유래 폴리데옥시리보뉴클레오타이드를 비페놀법으로 추출하는 방법과 관련한 도면을 첨부하여 상세히 설명하면 다음과 같다.
도 1은 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드 추출방법 모식도를 나타낸다. 본 발명의 매생이, 모자반, 다시마, 미역 등 포괄적으로는 해조류 유래 본 발명의 해조류 유래 폴리데옥시리보뉴클레오타이드 추출방법은 해조류를 건조하고 분쇄하는 원료확보 단계(가); 콩 추출물 10중량%, 율무 추출물 10중량%, 녹차(green tea) 추출물 20중량%, 콩(soybean) 지방산20중량%, 토코페롤 cocamidopropyl betain 20중량%, Olive oil carboxylate 20중량% 로 이루어진 혼합용액을 제조하고, 상기 (가)단계를 거쳐 건조 및 분쇄한 해조류 원료에 첨가하여 단백질을 용해시킨 후, 리보뉴클레아제를 추가하여 폴리뉴클레오타이드 및 폴리데옥시리보뉴클레오타이드 혼합물을 분리반응 시키는 단계를 포함하는 단백질 제거 및 분해단계(나); 상기 (나)단계를 거친 혼합물을 원심분리하여 불순물을 제거하고 필터링 후 건조하여 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 순수 분리하는 불순물 제거 및 순수분리단계 (다); 상기 (다)단계를 통해 순수 분리된 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 세척 및 건조하는 세척 및 건조단계(라); 상기 (라)단계로 건조된 결과물을 1% 나트륨 정제수 1ml를 첨가하여 희석 후 발효 아세트산을 이용하여 10분간 처리 후 소니케이션 처리를 10분간 진행하며, 원심분리를 실시하여 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확보하는 저분자 공정단계(마); 상기 (마)단계를 거친 결과물의 DNA흡광도를 측정하여 그 값이 1.7 내지 2.0인 순도 99%의 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확인하고 분말화하는 품질검사 단계(바)로 이루어질 수 있다.
(가) 원료확보 단계
본 발명의 원료확보단계(가)는 해조류를 건조하고 단백질 제거 및 분해하는 단계를 포함한다. 본 발명에서 지칭하는 해조류는 매생이, 모자반, 다시마, 미역, 김 중에서 선택되는 하나 이상의 것일 수 있다. 상기 준비된 해조류는 건조 및 분쇄한다.
(나) 단백질 제거 및 분해단계
본 발명의 단백질 제거 및 분해단계(나)는 콩 추출물 10중량%, 율무 추출물 10중량%, 녹차(green tea) 추출물 20중량%, 콩(soybean) 지방산20중량%, 토코페롤 cocamidopropyl betain 20중량%, Olive oil carboxylate 20중량% 로 이루어진 혼합용액을 제조한다. 상기 (가)단계를 거쳐 건조 및 분쇄한 해조류 원료에 상기 제조된 혼합용액을 첨가하여 단백질을 용해시키고, 단백질 용해 후 리보뉴클레아제를 추가하여 폴리뉴클레오타이드 및 폴리데옥시리보뉴클레오타이드 혼합물을 분리반응 시키는 단계를 포함할 수 있다.
상기 혼합용액을 구성하는 콩추출물, 율무추출물, 녹차추출물은 각각의 재료와 증류수를 1 : 5 비로 첨가하여 100도로 2 내지 3시간 가열하여 열수추출한 용액을 동결건조하여 각각의 분말을 얻는다. 각각의 분말은 상기 혼합용액을 구성하는 각각의 혼합비율로 혼합하여 혼합된 분말과 동일한 중량의 증류수를 혼합하여 용액으로 제조한다. 여기에 콩(soybean) 지방산, 토코페롤 cocamidopropyl betain 및 Olive oil carboxylate를 각각의 비율로 혼합하여 혼합용액을 완성한다. 상기 sloybean) 지방산, 토코페롤 cocamidopropyl betain 및 Olive oil carboxylate는 시판제품을 사용하였다.
상기 폴리사카라이드를 제거와 세포 용해 및 단백질분해를 위해 제조한 혼합용액은 상기 (가)단계의 해조류 원료 중량 1g 대비 40ml씩 첨가하여 단백질을 용해시킬 수 있다.
상기 단백질의 용해단계 후 (가)단계의 해조류 원료 중량 1g대비 0.1ml의 리보뉴클레아제를 추가하여 PN(Polynucleotide) /PDRN (Polydeoxyribonucleotide) 혼합물을 분리 반응을 시킬 수 있다. 본 발명에 따른 (나)단계에서 상기 단백질 제거 및 분해에 첨가되는 물질은 친환경 분해효소로서 기존의 방법에 비해 안정성이 확보될 수 있다.
(다) 불순물 제거 및 순수 분리단계
불순물 제거 및 순수 분리단계(다)는 상기 (나)단계를 거친 혼합물을 원심분리하여 불순물을 제거하고 필터링 후 건조하여 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 순수 분리하는 단계를 포함한다.
상기 (나)단계를 거친 혼합물은 2000rpm에서 1시간동안 원심분리하여 상층액을 수득한다. 이후 수득한 상측액은 부피비를 기준으로 정제수와 1:1 볼륨으로 희석하여 5000rpm에서 30분간 원심 분리하여 추가 상층액 분리 및 잔류물 제거한다.
이후 정제수를 분리된 상층액 부피 볼륨의 2배 추가하고 마이크로 필터를 이용하여 500mbar의 진공압력펌프를 이용하여 30분 동안 필터링(filtering)을 실시한다. 필터링이 완료된 필터지는 상온에서 1시간 건조시킨다.
분리단계는 일반적으로 원심분리하여 상등액을 회수하는 방법으로 이루어지나, 본원발명에서는 진공펌프압력과 마이크로 필터를 병행하여 사용함으로서 순도가 높은 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드의 분리가 가능하였다.
(라) 세척 및 건조 단계
본 발명의 세척 및 건조 단계(라)는 상기 (다)단계를 통해 순수 분리된 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 세척 및 건조하는 단계를 포함한다. 건조된 필터지에 정제수 10ml을 넣고 상기 (다)단계를 통해 필터링된 산물을 30-60도씨에서 1시간 건조를 실시한다.
(마) 저분자 공정단계
본 발명의 저분자 공정단계(마)는 상기 (라)단계로 건조된 결과물을 1% 나트륨 정제수 1ml를 첨가하여 희석 후 발효 아세트산을 이용하여 10분간 처리 후 소니케이션 처리를 10분간 진행하며, 원심분리를 실시하여 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확보하는 단계를 포함한다.
상기 (라)단계로 건조된 결과물은 1% 나트륨 정제수 10ml를 첨가하여 희석하고 부피비를 기준으로 사과와 쌀을 1:1비율로 혼합한 발효 아세트산을 10ml 추가하여 30-60℃에서 30분간 처리한다.
이후 소니케이션(sonication) 처리를 10분간 진행하고 micro-filter column에 옮겨 8000rpm 10분간 원심분리를 실시한다. 이후 정제수를 5ml 넣고 12000rpm에서 2분간 원심분리를 실시하여 최종 저분자의 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확보한다.
(바) 품질검사 단계
본 발명의 품질검사 단계(바)는 상기 (마)단계를 거친 결과물의 DNA흡광도를 측정하여 그 값이 1.7 내지 2.0인 순도 99%의 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확인하고 분말화하는 단계를 포함한다. 본 발명에 따른 DNA흡광도는 Thermo 사의 nanodrop-1000장비를 이용하여 측정하고 260(DNA)/280nm(단백질) 값이 1.7 내지 2.0 사이일 경우 순도 99%로 판정한다.
DNA 원액의 농도는 DNA 원액을 분광광도계를 사용하여 260nm에서 흡광도(Absorbance, A)를 측정하고, 추출된 PDRN의 순도를 확인하기 위하여 260/280 nm에서 흡광도 측정하고 그 비율이 1.7∼2.0일 경우 순도 99% 이상으로 판정한다. 일반적인 PDRN의 순도 측정의 경우 DNA 흡광도를 활용한 함량 측정하며 그 값이 1.7∼2.0일 경우 평균 99% 이상 높은 순도라고 볼 수 있다.
본 발명에 따른 방법은 해조류를 원료로 하여 외부 점액질 및 폴리사카라이드의 생성으로 인해 이를 제거하는 과정이 반드시 필요하고 비페놀 및 수산화나트륨, 염산 등 유해물질을 사용하지 않고 분해하기 때문에 보다 친환경적이고 안정적인 효과가 있으며 원료의 경우 동물성이 아닌 바다식물을 이용하여 바이러스 유입가능성 및 경제적인 비용에서 효과가 있다.
또한 본 발명은 기존의 연어, 송어 등(동물성원료)에서의 바이러스 혼입 위험성 및 원료수급의 제한성 등을 해결하고자 원활한 원료 수급이 가능하고 경제적이고 친환경적인 매생이, 모자반, 다시마, 미역, 김 등의 해조류 및 해양생태교란종의 조직으로부터 친환경물질 및 공정과정을 축소화하여 5kDa-20000kDa 또는 5kDa 이하의 분자량을 갖는 유전물질을 추출이 가능하다.
상기와 같이 동물유래가 아닌 바이러스 감염 위험에 안전한 천연 해조류의 생장점 포함된 조직으로부터 유래된 유전물질을 분리하는데 기존에 사용되는 페놀, 수산화나트륨 등의 유해물질을 사용하지 않고 제조공정을 단순화하여 추출 및 저분자 공정 시간을 줄이고 가격경쟁력이 확보된 PDRN 및 PN을 제조가 가능하다.
본 발명의 실험예를 들어 상세하게 설명하면 다음과 같다.
실험예 1. 저분자 전기영동실험
본 발명의 실험예 1에 따르면 기존 의료용으로 사용되고 있는 고분자 PDRN과 본 발명에 따른 방법으로 제조한 해조류에서 유래된 저분자PDRN을 전기영동실험을 실시하였다.
도 2는 본 발명의 실험예 1에 따른 전기영동 결과를 나타낸다. 일반적으로 피부흡수, 조직 침투율, 흡수율 등을 효과적으로 하기 위해 저분자화 과정이 반드시 필요하다. 따라서 본 실험결과에서도 알 수 있듯이 5kDa이하의 저분자 공정으로 생산된 유전물질이 향후 조직, 피부 등에서 침투, 흡수율을 높일 수 있는 것으로 판단된다.
실험예 2. in vitro 신혈관생성 실험
본 발명의 실험예 2는 해조류(매생이, 모자반, 다시마, 미역, 김 등) 유래 저분자 공정된 PDRN과 기존 연어유래 고분자 PDRN, 대조구에 대한 in vitro 신혈관생성 실험 결과에 대한 사진으로 신생혈관의 형성은 창상의 치유 과정에서 영양분과 산소의 공급을 위해서 필수적인 과정으로 인간제대정맥내피세포를 이용한 관형성실험(tube formation)을 통하여 현미경으로 분석하였다.
EGM2 배지에 20% FBS, Growth factor, 1% 페니실린/스트렙토마이신 첨가 후 세포 배양하고 Plate에 1X Matrigel 50 ㎕ 코팅 후 HUVEC을 분주(3x105 cell/ml)하고 시약 분주(대조군, 실험군, VEH(실험군에서 사용한 용매만 처리한 대조군), 타사제품) 뒤 4 시간 후, tube formation 확인하였다.
도 3은 해조류(매생이, 모자반, 다시마, 미역, 김 등) 유래 저분자 공정된 PDRN과 기존 연어유래 고분자 PDRN, 대조구에 대한 in vitro 신혈관생성 실험 결과를 나타낸다. 그 결과 신생혈관의 형성은 창상의 치유 과정에서 영양분과 산소의 공급을 위해서 필수적인 과정으로 인간제대정맥내피세포를 이용한 관형성실험(tube formation)을 통하여 현미경으로 분석한 결과 본 발명의 해조류(매생이, 모자반, 다시마, 미역, 김 등) 저분자 PDRN 물질이 타사제품보다 Tube formation 효과가 30% 이상, 대조구 대비 100% 가량 증가된 것을 확인하였다.
Tube formation의 경우 신혈관형성 실험으로 창상의 치유 과정에서 영양분과 산소의 공급 및 노폐물의 제거를 위해 필수적이며 실험은 인간제대정맥내피세포(HUVEC)를 이용한 관형성(tube formation) 실험으로Migration assay를 통해 HUVEC 세포가 PDRN에 의해 이동하는가에 대한 검증하여 Matrigel 상에서 인간제대정맥내피세포의 관형성을 유도하는 PDRN의 효과를 확인하였다.
실험예 3. in vitro 세포재생 실험
본 발명의 실험예 3은 해조류(매생이, 모자반, 다시마, 미역, 김 등) 유래 저분자 공정된 PDRN과 기존 연어유래 고분자 PDRN, 대조구에 대한 in vitro 세포재생 실험을 실시하였다.
Wound healing 의 경우는 상처 조직의 연속성이 외부의 작용에 의해 그 본래의 연속성을 상태를 의미하고 조직이란 주로 피부를 의미하게 되는데, 피부는 우리 몸에서 가장 큰 장기이며, 외부의 자극 및 감염 위험으로부터 일차적인 방어역할을 담당한다. 피부는 표피와 진피로 이루어지고, 표피는 내부로부터 기저층, 유극층, 과립층, 각질층의 순서로 층형상을 이루고 있으며 표피를 구성하는 각화 세포 (keratinocyte)는 세포 분열에 의해서 생성된 기저 세포가 유극 세포, 과립세포, 각질 세포의 순서로 분화한 후, 표면으로부터 각편으로서 박리하는 신진대사(턴오버)를 반복하고 있으며 과정은 다음과 같다.
상처 치유의 단계는 사람의 피부조직이 어떤 원인으로든 손상을 입게 되면, 상처를 치유하기 위한 반응이 곧장 시작되는데, 이는 상처의 치유 형태와 상관없이 모든 상처에서 일어나는 과정으로, 각 반응을 단계별로 살펴보면 다음과 같다.
1) 염증단계: 상처가 생기면 상처 주위 손상된 혈관에 있는 세포들(혈소팜, 백혈구, 대식세포 등)이 활성화되어 세균과 이물질, 괴사조직등을 제거하고, 또 이들 세포에서 여러 가지 활성 물질들이 분비되어 상처 주위 피부 세포들을 자극하는 단계
2) 상피화단계: 상처면의 피부 세포(상피세포)가 세포분열을 동해 분화하고 이동하며, 상처 전체에 걸쳐 분열하는 상피 세포가 가득 채워지는 단계
3) 증식단계: 세포질과 세포의 기질들이 증식하는 단계로 주로 콜라겐 합성을 하는 단계
4) 성숙단계: 상피화 단계와 증식단계를 거쳐 형성된 반흔 조직 내에 교원질 생성과 분해 사이의 균형을 맞추어 가는 단계
상기 반응들은 명확한 구분 없이 중첩되어 일련의 단계를 거치게 되며 창상은 물리, 화학적 상처 또는 세균감염에 의하여 정상인 조직의 연속체가 파괴되는 것으로 다양한 세포, 분자적 후유증을 야기하게 되며, 창상 치유는 특별한 방식으로 상처입은 조직의 성장과 재생을 유도하는 체계적인 생화학적, 세포적 과정으로서 혈액세포, cytokines, 성장인자들 간의 상호작용을 수반하여 손상된 피부나 조직을 정상적인 상태로의 복원을 유도하는 복잡한 과정이다.
또한 창상은 발생 조기에 치유되도록 하는 것이 중요하며, 창상이 오래 열려있는 경우 감염 등 이차적인 합병증이 올 수 있고 치료기간 동안 통증 및 일상생활에서의 불편함이 동반되기 때문에 조기에 창상이 닫히도록 해야 한다.
그러나 현재 일부에서는 합성 의약품의 무분별한 사용으로 인하여 항생제에 대한 면역성을 가진 병원성 미생물의 출현이 증가하였으며, 합성 의약품의 비싼 가격과 부작용은 질병 치료의 주요 문제로 대두되고 있으며, 보다 나은 생체활성 잠재력과 부작용이 적은 의약품의 지속적 개발이 요구되고 있다.
이에 본 발명의 실험예 4는 DMEM 배지에 10% FBS, 1% 페니실린/스트렙토마이신 첨가 후 세포 배양 후 Plate에 HaCaT을 분주(2x105 cell/ml)하고 4 시간 incubation 시키고 무혈청 배지에 HaCaT starvation 후 16 시간 incubation 뒤 Plate에 스크래치를 낸 후, 시약 분주(대조군, 실험군, VEH(실험군에서 사용한 용매만 처리한 대조군), 타사제품)뒤 24 시간 후, wound healing 확인한 것이며 사람 상피세포주(HaCaT)-wound healing model: confluent한 cell에 스크래치를 내서 세포가 이동하는 정도를 현미경으로 분석한 결과로 스크래치의 범위가 좁아질수록 재생효과가 있는 것이며 그래프에서는 스크래치 면적을 수치화 한 것으로 값이 낮을수록 효과가 있는 것으로 볼 수 있다.
도 4는 해조류(매생이, 모자반, 다시마, 미역, 김 등) 유래 저분자 공정된 PDRN과 기존 연어유래 고분자 PDRN, 대조구에 대한 in vitro 세포재생 실험 결과를 나타낸다. Wound healing model을 이용하여 사람 상피세포주(HaCaT)에 scratch를 내서 세포가 이동하는 정도를 현미경으로 분석한 결과, 본 발명의 해조류(매생이, 모자반, 다시마, 미역, 김 등) 저분자 PDRN 물질이 타사제품보다 wound healing 효과가 5% 이상, 대조구 대비 25% 이상 증가된 것을 확인하였다.
본 발명은 김의 특정조직이 아닌 모든 조직에서 PDRN 및 PN과 같은 유전물질을 분리하고, 확보된 해조류 유래 유전자 혼합물로부터 아데노신 A2 수용체를 활성화시키는 저분자의 PDRN, PN 혼합물을 제조하는 방법을 제공함으로써 신혈관생성 및 세포재생 효과가 있어 관계되는 산업상 이용가능성이 있다.
Claims (7)
- 해조류로부터 추출된 것을 특징으로 하는 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드
- 청구항 1에 있어서, 상기 해조류는 매생이, 모자반, 다시마, 미역, 김 중에서 선택되는 하나 이상의 것인 것을 특징으로 하는 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드
- 청구항 1에 있어서, 추출된 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드는 분자량 5kDa-20000kDa인 것을 것을 특징으로 하는 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드
- 해조류를 건조하고 친환경물질을 이용하여 단백질 제거 및 분해하는 원료확보 단계(가);상기 (가)단계를 거쳐 건조 및 분쇄한 해조류 원료에 단백질 용해 혼합용액을 첨가하여 단백질을 용해시킨 후, 리보뉴클레아제를 추가하여 폴리뉴클레오타이드 및 폴리데옥시리보뉴클레오타이드 혼합물을 분리 반응시키는 단계를 포함하는 단백질 제거 및 분해단계(나);상기 (나)단계를 거친 혼합물을 원심분리하여 불순물을 제거하고 필터링 후 건조하여 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 순수 분리하는 불순물 제거 및 순수분리단계 (다);상기 (다)단계를 통해 순수 분리된 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 세척 및 건조하는 세척 및 건조단계(라);상기 (라)단계로 건조된 결과물을 원심분리하여 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확보하는 저분자 공정단계(마);상기 (마)단계를 거친 결과물의 DNA흡광도를 측정하여 그 값이 1.7 내지 2.0인 순도 99%의 저분자 폴리디옥시리보뉴클레오티드 및 폴리뉴클레오타이드를 확인하고 분말화하는 품질검사 단계(바)로 이루어진 것을 특징으로 하는 신혈관생성 및 세포재생 효과를 갖는 해조류 유래 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드 추출방법
- 제4항에 있어서, 상기 단백질 용해 혼합용액은 콩 10중량%, 율무 10중량%, 녹차(green tea) 추출물 20중량%, 콩(soybean) 지방산20중량%, 토코페롤 cocamidopropyl betain 20중량%, Olive oil carboxylate 20중량% 로 이루어진 것을 특징으로 하는 신혈관생성 및 세포재생 효과를 갖는 해조류 유래 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드 추출방법
- 제4항에 있어서, 상기 해조류는 매생이, 모자반, 다시마, 미역, 김 중에서 선택되는 하나 이상의 것인 것을 특징으로 하는 신혈관생성 및 세포재생 효과를 갖는 해조류 유래 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드 추출방법
- 제5항에 있어서, 상기 (나)단계에서 제조한 단백질 용해 혼합용액은 상기 (가)단계의 해조류 원료 중량 1g 대비 40ml씩 첨가하고, 리보뉴클레아제는 (가)단계의 해조류 원료 중량 1g대비 0.1ml씩 첨가하는 것을 특징으로 하는 신혈관생성 및 세포재생 효과를 갖는 해조류 유래 폴리데옥시리보뉴클레오타이드 및 폴리뉴클레오타이드 추출방법
